【発明の詳細な説明】
抗ウィルス及び抗菌組成物並びに使用方法発明の技術分野
本発明は、薬学的に許容される担体と共に重合された酵素を含有する新規な抗ウ
ィルス及び抗菌組成物、並びにそれらの使用方法に関する。
発明の背景
抗生物質に耐性のある菌株及びウィルス性疾患の増加により、新種の組成物を作
り出し人間や動物を治療することが必要となった。現在の多くの治療法及び薬剤
の中には、様々な疾患で苦しむ患者を治療するために酵素を単量体の形態で使用
することが知られている。酵素は、生体内でのほとんど全ての主要な生命現象を
遂行する触媒的に活性な蛋白質である。このように、多くの酵素は、個別的にあ
るいはある種の組合せで、それらの物理化学的、生理学的、もしくは生物学的作
用に従って分離されてきた。
リゾチーム及びリボヌクレアーゼはこれ迄に治療効果が明かにされた種々の酵素
中の1種である。リゾチームは、1922年フレミングによって発見された。し
かしリゾチームの酵素機能は、1950年以後になって始めて明らかにされた。
それ以来、リゾチームは物理化学的、生理学的及び臨床的研究の主題として大き
な価値を有するが、この化合物の生物学的意義の程度は未だ明らかにされていな
い。これまでは、リゾチームは、抗ウィルス、抗菌、抗炎症及び抗ヒスタミン特
性のような種々の治療特性を有すると見られてきた。抗菌作用は、細菌壁に含ま
れるN−アセチルオムラミニックアシッドとN−アセチルグルコサミンとの間の
β−1−4−グリコシド結合の加水分解に基づいて発現される。
食細胞中にリゾチームが存在することもまたよく知られている。この分野での研
究により、リゾチームに含まれる細胞内リゾチームが貧食された細菌を消化する
ことが明かにされた。人体中で、リゾチームは10〜400trrg / dと
いう生理学上の濃度で食作用を刺激することが観察されている。
リゾチームの他の特性もまた報告されている。例えば、リゾチームが感染過程の
間、毒素によって遊離された体内発熱性物質に対する応答である体温を下げる作
用を有することが示されている。リゾチームがγ−グロブリン、オプソニン、そ
の他の抗体の合成を刺激することにより免疫学的過程に介入することも示されて
いる。更に、リゾチームが強い抗炎症作用を有することが示唆されている。
リゾチームのこれら公知の有用な特性にもがかわらず、またリゾチームに基づく
多数の研究計画や薬学的製剤の生産にもかかわらず、治療目的のためのこの酵素
の使用は非常に制限されている。
様々な生物学的効果のために研究されてきたもう一つの酵素グループは、リボヌ
クレアーゼである。リボヌクレアーゼは、多くの動物や植物体中にまた細菌細胞
中によく認められる酵素グループである。その特性の研究及び単離方法の研究は
、シュミットとマクドナルドによって1955年に始められた。この酵素に基づ
く研究で、ガン組織中でリボヌクレアーゼ活性が相当低下することが見出された
。例えば、白血病性ウィルスがマウスの酸性リボヌクレアーゼの活性を急激に減
少させることが明らかにされた。また、ウィルス性白血病のマウスにおいて、牌
組織から得られたミトコンドリア及びミクロソーム分画中の酸性リボヌクレアー
ゼの活性の相当の減少が認められた。
上記研究は、リボヌクレアーゼの活性の減少がウィルスによる感染と密に関連し
ていることを示しており、従ってリボヌクレアーゼ酵素が抗ウィルス活性を有す
るということが示唆されている。しかしながら現在、抗ウイルス試薬としてこの
酵素を使用するについての報告は全くなされていない。
潜在的に有益な酵素が未だその治療効果に対して広く使用されていない主要な理
由の1つは、これら及び他の酵素の単量体の形態で認められる細胞障害作用にあ
る。
培養した繊維芽細胞を用いた試験で、リゾチーム及びリボヌクレアーゼの単量体
の何れもがごく少量で細胞障害作用を有することが認められた。従って、これら
及び他の酵素に潜在する有益な作用は、細胞障害作用を制御する効果的な方法が
開発されない限り、活用できないこととなる。それゆえ、酵素を単量体の形態で
用いても細胞障害作用を伴うことなく、ウィルス性もしくは細菌性の疾患或いは
他の有害状態の治療に有効に使用できるリゾチーム、リボヌクレアーゼ、もしく
は他の類似酵素を主成分とする組成物を開発することが望まれる。
発明の概要
本発明によれば、有効成分としてリゾチーム、リボヌクレアーゼ、もしくは他の
酵素を含有し、しかも細胞障害を有しない抗ウィルス又は抗菌組成物が当該酵素
を二量体の形態で使用することによって調製され得ることが見出された。有効成
分としてリゾチームニ量体又はリボヌクレアーゼ単量体及び薬学的に許容される
担体を用いて組成物を調製することにより、多くの伝染病を細胞障害作用を伴う
ことなくうまく治療することができる。
好ましい実施態様の詳細な説明
本発明の抗ウィルス及び抗菌組成物は、最初にリゾチーム及びリボヌクレアーゼ
を単量体の形態で得ることにより調製され得る。本発明に従い調製された化合物
の製剤に使用されたりゾチーム単量体(カタログ磁28260)及びリボヌクレ
アーゼ単量体(カタログ阻34388)は、ハイデルベルクD69−000のセ
ルバ ファイネ バイオケミ力、ゲーエムベーハー ランド コーポレーション
(Servx Fsine Biochemica 、 Gmbhund Co
orpera+ion )から入手された。これら酵素の単量体は、当分野で現
在用いられている通常の方法により二量体に重合できる。しかしながら、特に好
ましい方法は、カールソンら(Carlsson N !l)による酵素重合法
である[バイオケミストリー ジャーナル(BioCbsmistryJour
nal) 173 : 723〜737(1978)]。他の好ましい方法はソ
ルレンチノら(Sorr!ntino et !+)により明かにされた方法で
ある[ヨーロピアン ジャーナル オブ バイオケミストリー (Eur、 J
、Biocbsm、) 124 : 183〜9 (1982)コ。本発明の組
成物に使用することができる特に有用なりボヌクレアーゼの二量体は、動物の膵
臓組織から単離された膵リボヌクレアーゼAから調製された二量体である。
有効成分としてリゾチームもしくはりボヌクレアーゼの二量体及び薬学的に許容
される担体を含む本発明組成物は、好ましくない細胞障害作用を伴うことなく種
々の細菌性及びウィルス性疾患を治療するのに有効であることが見出された。酵
素の潜在的な有害作用は、リゾチーム及びリボヌクレアーゼの単量体と二量体の
両方を用いた比較研究で検討した。これらの研究は、緑ザルの腎臓(G M K
)の繊維芽細胞培養物にリゾチーム及びリボヌクレアーゼの単量体及び二量体
を種々の濃度で投与することにより行なわれた。リゾチームの単量体は、0.1
mg / xi及び1. Otag/xlの濃度で24時間後に繊維芽細胞に
細胞障害作用を与えることが観察された。インキュベーションの3日後には、繊
維芽細胞の細胞障害作用は、O,01mg/z/の濃度でさえも観察され、イン
キュベートされた細胞の50%に影響を及ぼした。5日後、培養された細胞の
75%が、1.0及び0. 1ag/zlの濃度でリゾチームの単量体の細胞障
害活性による影響を受けた。一方リゾチームの二量体は、これら試験においてい
かなる濃度においても、しかも7日間後であっても細胞障害作用を示さなかった
。これらの研究は、リゾチームの二量体は、単量体形態に比してGMK繊維芽細
胞に対する毒性がおよそ100倍小さいことを示している。
リボヌクレアーゼに関する研究でも、二量体形態のものは同様に細胞障害作用を
欠損することが認められた。
その研究で、リボヌクレアーゼは、0. 0001 mg/d程度の低濃度での
5日間培養でGMK繊維芽細胞に対して細胞障害があることが認められた。培1
7日後、0.01■/d及びそれ以上の濃度で、培養された細胞の100%が、
リボヌクレアーゼの細胞障害作用により死滅した。一方、リボヌクレアーゼの二
量体は、GMK繊維芽細胞に対して全ゆる濃度レベルで7日間インキュベージジ
ンした後でさえも、細胞障害作用を示さなかった。このように、リボヌクレアー
ゼの二量体形態は、単量体の形態に比して繊維芽細胞に対する毒性がおよそ1.
000〜10,000倍小さいことが認められた。
これらの試験は、リゾチーム及びリボヌクレアーゼのような酵素の単量体形態の
使用に通常伴う細胞障害作用を、それらの酵素を二量体形態で使用することによ
り実質上取り除くことができることを示ししている。
更に本発明者の研究によれば、明らかな細胞障害の作用の欠損にもかかわらず、
リゾチーム及びリボヌクレアーゼは二量体形態でウィルス性及び細菌性感染の治
療に非常に有効であることが見出された。メントリの受精卵及びセンダイウィル
ス株を用いた試験において、リゾチームの二量体を、種々の濃度で卵に羊膜内投
与した。各卵にはまた、センダイウィルスの2ユニツトを投与した。
インキュベーション後、感染させた卵及び対照卵から羊水及び尿膜膣液を採取し
て比較した。これらの試験は、リゾチームの二量体が10日齢のメントリ受精卵
中で培養されたセンダイウィルスの増殖を、0.01■/ xiの低濃度でさえ
も抑制することを示した。リゾチーム及びリボヌクレアーゼの二量体を用いた同
様の試験は、ストレプトコッカス菌株に対するこれら二量体酵素の静菌作用を示
した。
このように、リゾチームまたはりボヌクレアーゼの二量体から本発明に従って調
製された組成物は、種々のウィルス性および細菌性の感染を治療するのに使用で
きる。
本発明組成物は、様々な製剤形態に調製することができ、これら酵素−担体製剤
は、人間又は動物の患者に治療を必要とする疾患に応じて内科的にもしくは外的
に投与することができる。耳感染、乳腺炎、胃又は腟路感染のような内科的疾患
に対しては、適宜に調製された本発明の二量体組成物を経口的に、静脈内に、非
経口的に、座薬的に、もしくは二量体溶液を感染部に到達させることができる他
のどんな方法によっても投与することができる。
ウィルス性又は細菌性皮膚病、感染創傷、ヘルペス又は外的症状を有する他の性
病のような外的疾患に対しては、本発明の組成物は、種々の適当な形態で患者に
局所的に投与し得る。
このように治療される疾患又は感染の特定の性質に応じて、本発明の組成物の適
切な製剤形態を決定すればよい。外的治療では、本発明組成物は軟膏、ローショ
ン、溶液、オイルなどの様々な形態で投与され得る。内科的適用が必要なときは
、ドロップ、タブレット、溶液、カプセル、プントリフイスなどの多くの適当な
形態で適用することができる。患者に適用される組成物の特定の形態に応じて本
発明二量体酵素組成物に使用される薬学的に許容される担体を決定すればよい。
使用可能な多くの適切な担体には、親水性基剤、生理学的に許容される塩溶液、
水、軟膏、粉末などがある。
本発明は、上述した二量体組成物の有効量を人間又は動物の患者に投与すること
により細胞障害作用を伴わないウィルス性又は細菌性の感染の酵素治療法を提供
する。
「有効量」は、抗ウイルス性又は抗菌性作用を発現するに必要な量を意味する。
効果的な治療に必要な量は、治療された疾患の性質及び投与される二量体組成物
の形態により各ケース毎に変化する。一般に、本発明組成物は、0.01〜50
.Qmg/kg体重程度、特に好ましくは1゜0〜2. 0mg/kg体重の範
囲の投与規準で投与される。
局所的な治療の場合、たとえば水、パラフィン及びプロピレングリコールの溶液
のおよそ200z/中に二量体的4mgを使用して調製した軟膏では、1日に4
〜5回適用するのが効果的である。これらの範囲での投薬は、酵素単量体による
治療に伴う有害な細胞障害作用なしに多くのウィルス性又は細菌性の疾患を治療
するのに十分である。
次に実施例を挙げて本発明を説明するが、本発明は斯かる実施例に制限されない
。
実施例1
緑ザル腎臓(GMK)の繊維芽細胞の培養物に対するリゾチーム及び膵リボヌク
レアーゼAの単量体及び二量体の細胞障害作用を比較した。これらの試験は、単
量体及び二量体を0.0001から1. Owrg/xiの濃度で繊維芽細胞
の培養物に適用することにより行なった。培養物は、7日間インキニベートした
後に細胞障害を調べた。
これら試験の結果を第1表及び第2表に示す。第1表から明かなように、リゾチ
ーム単量体はO,1mg/z/及び1、 Owrg/dの濃度で24時間後繊維
芽細胞に細胞障害を与える。インキュベーションの3日後、単量体は0.01■
/ 2/の濃度でさえも繊維芽細胞に対して細胞障害作用を示し、培養された細
胞の50%に影響を及ぼした。5日後、培養された細胞の75%が1. 0mg
/z/及び0. 1rag/xiの濃度でリゾチーム単量体の細胞障害作用の影
響を受けた。
対照的に、二量体形態のリゾチームは、試験された全ての濃度において、7日間
のインキュベーションの完了後でも細胞障害作用を示さなかった。このようにリ
ゾチームニ量体はりゾチームの単量体に比してGMKへの毒性が約100借手さ
いということが明らかとなった。
第2表から明かなように、膵リボヌクレアーゼAの単量体は5日培養で、0.
0001a+g/j/という低濃度であってもGMK繊維芽細胞に細胞障害を与
える。
0.01++g/z/及びそれ以上の濃度での膵すボヌクレアーゼA細胞障害作
用は、非常に強く培養7日後、培養された細胞の100%を殺してしまう。
リゾチームニ量体の場合と同じように、二量体形態の膵リボヌクレアーゼAは、
7日間の期間中に試験されたいかなる濃度でもGMK繊維芽細胞に細胞障害作用
を示さなかった。この結果は、膵すボヌクレアーゼへ二量体が膵すボヌクレアー
ゼA単量体に比して約1000〜100’00倍小さいGMK繊維芽細胞への毒
性であるということを示している。
実施例2
リゾチームニ量体の抗ウイルス性作用を検討した。リゾチームニ量体を10日齢
受精鶏卵に10. Ovag/zl、1、 On+g/z/、 0. 1w
Ig/z/、 0. 01mg/z/及び0 、 001 mg/z/の濃度で
注射した。センダイウィルス株(血球凝集力価−1: 128HA)を、各濃度
の二量体に2血球凝集ユニットの量で加えた。リゾチームニ量体及びウィルスの
投与後、卵を37℃で72時間インキニベートした。インキニベーション終了後
、感染卵から羊水及び尿膜液を採取し、タカチーセット(Takalsyset
)及びメントリの血球の使用によるマイクロ法(miCromethod )
を用いて血球凝集試験に供した。実験を繰り返し行った。上記試験で得られた結
果を第3表に示す。第3表から明かな様にリゾチームニ量体は、10日齢受精鶏
卵中で培養されたセンダイウィルスの増殖を、0、 01mg/z/程度の低濃
度であっても抑制した。

1、試験は、センダイウィルス株を用いて行った。ウィルス血球凝集力価は、1
: 128HAであった。
2.10日齢の受精鶏卵を実験モデルとした。
3、センダイウィルス2血球凝集ユニットを、各濃度のりゾチームニ量体に加え
た。同時に、血球凝集特性を有するか否かを確認するためにリゾチームニ量体に
つき対照試験を開始した。結果は負であった。各濃度のりゾチームニ量体と22
HAユニツトのウィルスを4個の鶏卵の羊膜内に感染させるために投与した。卵
を37℃で72時間インキュベートした。
4、インキニベーション終了後、羊水及び尿膜液を感染卵から採取した。血球凝
集試験を、タカチーテストセット(TakajB +esl 5et)及びメン
トリの赤血球細胞を使用して行った(血球凝集マイクロ法)。
実施例3
乳腺炎の雌牛から採取した数種の病原性菌株を用いて細菌に対する二量体形態の
リゾチーム及び膵リボヌクレアーゼAの作用を試験した。3つの菌株[ストレプ
トコッカス アガラクティエ(SfreptococCus agalacji
ae)、ストレプトコッカス ディスガラクチイエ(SHepjococcus
dysgalactia+ )及びストレプトコッカス リベリス(Stre
ptococcus lib!r口)に対するリゾチームニ量体の各濃度での効
果を第4表に示す。この試験の結果から、3つ全てのストレプトコッカス株がリ
ゾチームニ量体の活性に感染性を有することがわかる。このことは、ストレプト
コッカス リベリスの場合に最も顕著であり、1.25@g/z/程度の低い濃
度で二量体の活性の影響を受けた。ストレプトコッカスの他の菌株に対する静菌
作用は、約10+ig/z/の濃度から観察された。
第5表に、人間患者から培養した病原性菌株に対する膵リボヌクレア・−ゼA二
量体及びリゾチームニ量体の作用を示す。この表から明かなように、膵すボヌク
レアーゼA二量体は、プソイドモナス アエルジノーサ(Pseudomona
s asruginosa) 、大腸菌及びプロテウスブルガルス(ProHu
s Vulgarus)の菌株に対し、特に約5〜10mg/z/の範囲の濃度
で最も効果的であった。スタフィロコッカス及びストレプトコッカスの菌株は約
5〜10■/ xi濃度のりゾチームニ量体に感受性であることがわかった。感
度試験は、WHOによって推薦され一般的に受は入れられている国際規定に従っ
て行われた。
実施例4
に−562赤血白血病細胞ラインの増殖に対するリゾチームニ量体及び膵すボヌ
クレアーゼA二量体の作用を、上記ラインからの細胞に種々の濃度の二量体を適
用することにより確認した。上記試験の結果を第6表及び第7表に示す。簡潔に
は試験したりゾチームニ量体は全ての濃度においてに一562細胞に対して強い
細胞変性作用を示した。また第7表から明かなように、膵すボヌクレアーゼA二
量体も、赤面白血病細胞ラインに対して同様の効果を示したが、それは1.0m
g/dの濃度のときのみであった。
実施例5
犬の化膿性中耳炎に対するリゾチームニ量体の作用を試験した。様々な疾患をも
つ異なる種の大19頭を使用して試験した。疾患は、炎症の進行により特徴づけ
られ、平均約7〜14日間炎症が持続したが、9ケ月続く場合が1例あった。7
頭の犬につき、炎症した耳からの化膿排出物を治療にかかる前に検査して菌株を
同定した。
培養試験は、スタフィロコッカス細菌、ブルー バスバシリス(blue pu
sbaCillis) 、緑膿菌、原虫類及び様々な細菌種の存在を示した。
罹病大は、頭を振ったり、炎症した耳に足を達せしめようと試みる等積々の苦痛
の徴候を表わした。犬は、一般的に食欲がなくなり、体温が上昇した(39.2
〜41.2℃)。18頭の供試大には薬理学的方法を用いた予備的治療を全くし
なかった。炎症が9ケ月間持続した1頭の犬は、数種の抗生物質を与えてきたが
、これらは疾患の治療に効果はなかった。
リゾチームニ量体20+egと生理学的塩251/との溶液からなる本発明組成
物を用いて犬を治療した。組成物をドロップの形態で適用し、1日に4〜5回炎
症した耳にドロップ10個を適用した。治療初日をすぎるとすでに著しい改善が
見られた。即ち、体温が下がり、犬は明かにこれまでより元気そうで食欲が出て
きた。化膿性炎症の症候は、治療の3日目から6日目の間に完全に後退した。不
成功であったが前もって9ケ月間抗生物質で治療されてきた犬でも、10日後に
疾患は後退した。このようにリゾチームニ量体は、犬の化膿性中耳炎の治療に効
果的であることを示した。
実施例6
乳腺炎の雌牛に対するリゾチームニ量体の作用を試験した。6頭の乳腺炎の雌牛
を試験に供した。これらの雌牛は40.5℃以上の体温及び食欲減退などの疾患
の症候を示した。6頭全てに、治療を発病後2日目に始めた。
組成物を投与する前に、ミルクを細菌試験の試料として採取した。培養物には、
スタフィロコッカス アガラクナイエ及びストレプトコッカス アガラクティエ
のような微生物が含まれていた。
リゾチームニ量体を、生理学的塩50xlの溶液中40111gの投与量で1日
に2回注射器を通して各雌牛の感染した乳首に投与した。僅か24時間後、体温
は再び平常となり、食欲が回復したことが認められた。3日後、治療中の全ての
雌牛は、疾患の症候を表わさなくなった。従って治療は、4日目まで続けただけ
で、その後のミルクの検査は、治療前に見られた病原性微生物が消滅しているこ
とを示した。更に、ミルクには無症状の乳腺炎を示唆する変化は見られなかった
。治療された雌牛にミルク収量の減少したものは見られず、乳首の開放性もまた
損じられなかった。リゾチームニ曾体治療の24時間後、治療された雌牛のミル
ク中に何等の遮断物質も見出せなかった。このように疾病症候の早期消失及びミ
ルク産生能の完全な保持は本発明のりゾチームニ量体組成物が牛の乳腺炎の治療
に効果的に使用され得ることを示している。この治療は、現在酪農産業で治療に
1年問おおよそ54億ドルかかるスタフィロコッカス及びストレプトコッカス菌
株による乳腺炎感染を防ぐのに特に経済的重要性を有する。
実施例7
犬パルボウイルス(CPV)感染を、リゾチームニ量体の経口投与により治療し
た。種及び重量の異なる3ケ月から6歳迄の27頭の犬に、パルボウイルスの典
型的な微症候群を認めた。全ての供試動物には、高い体温(40〜41.6℃)
、頻度の大量嘔吐の発作、おびただしい特徴的な悪臭の下痢糞、並びに脱水症及
び感受性の症状があった。また動物は、激しい苦痛を訴えた。治療は、飼主がベ
ットを獣医にどのくらい早く連れて行くかによるが、平均して感染の3〜5日目
に開始した。感染した犬には、1日に2回、体重当り1〜2mgの投与量でリゾ
チームニ量体を投与した。まだ飲むことのできる動物には、リゾチームニ量体を
飲料水の形態で投与した。
飲むことのできない動物には、生理学的塩溶液の形態でプローブを通して投与し
た。治療した27頭の犬のうち25頭は、治療3〜5日後に完全に肉体的健康状
態を回復した。治療初日の間でさえも糞の回数及び嘔吐発作の症状の顕著な減少
が観察され、大多数の犬は治療2日後にこれら症状が完全になくなった。少数の
犬は、これら症状が組成物の最初の投与後になくなった。リゾチーム二量体組成
物の投与に関連する副作用は、どの動物にも観察されなかった。これらの臨床試
験は、本発明のりゾチームニ量体組成物がパルボウイルス感染の犬に好適に使用
され得ることを示している。
実施例8
人間の皮膚病に対する本発明リゾチーム二量体組成物の作用を試験した。従来の
方法で治療してきたがうまく治療できなかった種々の皮膚病にかかつている15
〜35才の数ダースのヒト患者に対して試験を行なった。
疾患別に次のグループに分類した:

患者において、細菌学的培養試験を予め行った。殆んどのケースでスタフィロコ
ッカス アウレウスが採取された物質から培養された。治療は、リゾチームニ量
体の4mgを含む軟膏を一日に4回投与して行った。特に好ましい軟膏の処方は
、次のとおりである:

すべて患者において、種々の皮膚症状は10〜12日以内になくなり、3日後に
はっきりと観察されたものも数例あった。治療は、皮膚症状の重症度、及び治療
前に疾患がどのくらい進行したかに依存するが、慢性ホルンカロシス(Foru
nculosis)の患者で回復に4〜5週間がかり、静脈瘤潰瘍の患者で2〜
12週間かかった。この研究の結果は、リゾチーム二量体が様々な皮膚病に好適
に使用され得ることを示している。
実施例9
生殖部位の種々の感染病を本発明のりゾチームニ量体組成物を使用して治療した
。25〜49才の9人の女性がこの試験で治療され、そのうち7人は慢性膣炎、
1人はダグラス膿瘍、1人はバルトリン腺炎であった。慢性膣炎の患者には、親
水性基剤2ccm中にリゾチーム二量体10mgを含む腟内生薬の形態で投与し
た。生薬は、1日に2回、7日間投与された。全ての患者に生殖部の炎症の総合
的な後転が見られた。更にロイコルヒア(1eucorrhed)及び他の症状
もなくなった。ダグラス膿瘍の患者には、0.9%NaC/溶液52/中に溶解
したりゾチームニ量体20mgを1日に2回4日間投与した。
溶液は、ダグラス空洞中に直接投与された。リゾチーム二量体の投与前に、化膿
性内容物をダグラス空洞から吸い出した。これらの培養物にはストレプトコッカ
ス へモリティカス(sHeptococcus hamo17jicus )
及び大腸菌が含まれていた。患者の以上に高い体温は最初のりゾチームニ量体投
与後24時間以内に平常値に下がった。
その上苦痛の症状もこの間になくなった。リゾチーム二量体組成物の二回目の投
与後4日目にはダグラス空洞に膿はなかった。この場合、疾患の再発が3週間後
にあったが、ダグラス空洞中へ別にリゾチーム二量体の投与を2回行った場合に
は、疾患の進行を制御することができた。
バルトリン腺炎の患者は、患部からの化膿性内容物の吸引後、直ちに0.9%N
aC/のlccmに溶解したりゾチームニ量体20IIIgを化膿した線中へ1
回の投与により治療した。4日後、この患者は完全に治殖した。その後の4ケ月
の観察の間、この疾患の再発はなかった。
これらの臨床試験は、リゾチーム二量体が女性の生殖部位の感染疾患に極めて有
益な治療効果を有することを示す。更に、化膿性内容物のある空洞中へ直接リゾ
チーム二量体を投与することにより局部的な膿瘍を治療することが可能となる。
実施例10
感染した創傷に対するリゾチーム二量体溶液の作用を試験した。このグループに
は、感染した術後傷のある4人の患者、即ち開腹術後の女性2人、糖尿病性脈管
障害によるヌクロシス(nucrosis)のための足祉切断術後の女性1人、
及びバーガー病(Burger’s disease)のための下肢切断術後の
女性1人がいた。全症例において、0.9%NaC152/に溶解したりゾチー
ムニ量体20mgの溶液である湿潤投与及び洗浄投与の形態で1日4回投与され
た。開腹術後化膿瘍のある患者では4日及び6日後に完全治癒し、他の患者では
それぞれ21日及び5ケ月後に治癒した。これらの試験は、リゾチーム二量体が
感染した術後傷の治療に副作用を伴うことなく使用し得ることを示している。
実施例11
臨床的観察は、リボヌクレアーゼニ量体組成物で治療された陰部庖疹の患者に対
し臨床観察を行った。試験グループは、23〜36オの女性5人の患者を含む。
そのうち4人の患者は、始めて疾患が現れたもので、もう1人の女性は3回目で
あった。全ての患者は発病3〜5日目の水痘期間中で、特に放尿中、会陰部に非
常に強い痛みを訴えた。全患者に唇腫脹及び唇感染と共に、唇粘膜及び縫部と肛
門部の外皮に混濁状液体で満たされた多数の水痘が見られた。全患者は鼠経部の
リンパ節が拡張し、苦痛を訴えた。
リボヌクレアーゼA二量体を含む軟膏を毎日4〜5回水庖水痘粘膜の感染部に適
用して治療した。適用された軟膏は、次の処方のものである:
リボヌクレアーゼニ量体 4.0■アセチルステアロイルオキシアル
コール 25.CI+g流動パラフィン
10.0■スパン60 5.0エツイーン6
0 8.0゜2プロピレングリコール 10.
Ot+gアセプチナM O,3mgアセプチナP
O,16■総量20011となる量の蒸留水
二量体治療を受けた患者は、最初の適用から数分後及び少なくとも一時間以内に
、痛みが実質上弱まり、次の10〜20時間の間にすっかりなくなったと報告し
た。
臨床試験は、4人の患者において、病理学上の変化が治療3日後にすっかり退い
たことを示した。他の患者において、病理学的症状は5日後にすっかり除かれた
。これらの患者で、膵すボヌクレアーゼA二量体を含む軟膏の適用後新しい水痘
が現れた者はいなかった。以上の試験は、本発明の二量体を含む軟膏が陰部庖疹
の治療に好適に使用され得ることを示す。
実施例12
本発明の組成物を用いて***庖疹の治療を様々な年齢の100Å以上の患者に対
し行った。全患者は上唇がはれ、赤くなり、そして混濁状の液体で満たされた多
数の水痘があるという症状があった。全ての患者は、疾患により影響を受けた部
分の皮膚に痛みを訴え、更に組織に突張り感があった。治療は、毎日4〜5回膵
リボすクレアーゼA二量体を含む軟膏の局部的な適用により行った。
治療された全ての患者は例外なく痛み及び組織の突張り感が急速に退いたと述べ
た。陰部庖疹に関する症例で見られたように、痛みは次の数時間以内に全てなく
なった。臨床試験は、浮腫や水痘が2〜3日以内になくなることを示した。個々
の症例において、症状が完全になくなり、完全な治癒が達せられるのに5日かか
った。さらにまた疾患の初日に治療を始めた患者では、浮腫、刺激及び丘疹のよ
うな皮膚症状は24時間後にすっかりなくなった。もう一つの観察は、この疾患
を頻繁に再発している人の再発するまでの期間が延長し、再発の症状がその都度
和らぐというものであった。全症例において、いかなる副作用も見られなかった
。
実施例13
臨床試験を、本発明の二量体組成物を用いて帯状庖疹の6人の患者を治療して行
った。5つの症例では疾患は典型的な症状で現われ、6番目の症例は下記で述べ
る特殊な合併症であった。これらの患者は膵すボヌクレアーゼへ二量体を含む軟
膏によって治療され、治療は疾患の3日目又は4日目に始められた。軟膏は、毎
日4〜5回適用された。
全症例において、痛みは初めの24〜48時間以内にすっかり退いた。水痘は、
3〜4日後に乾燥したことが観察され、従ってこの期間の後に治療を終えること
に決めた。数日以内に皮膚症状は完治した。最初のおよそ24〜48時間後に痛
みが存続している患者はおらず、1年以上もの間観察を続けた3人の患者におい
て症状は再発しなかった。前述した1人の患者には、疾患の通常ではない臨床的
進行があった。42才の女性は、彼女の免疫系をひどく害するコバルト療法によ
って肺ガンの治療を受けてきた。帯状庖疹の通常の症状の他に、彼女の体中に水
痘の播腫が生じていた。本発明のりボヌクレアーゼニ量体を用いた治療は、免疫
不全患者に極めて有効で、彼女は数日後水痘症状の完全な後転を示した。この患
者及びそのグループの他のメンバーにおける成功した結果は、水痘ウィルスによ
る帯状庖疹に対して膵すボヌクレアーゼA二世体の特別な効力を示す。

Claims (1)
- 【特許請求の範囲】 1 リゾチーム二量体及びリボヌクレアーゼ二量体からなる群から選ばれた二量 体を有効成分として薬学的に許容される担体と共に含有することを特徴とする抗 ウイルスもしくは抗菌組成物。 2 二量体がリゾチーム二量体である請求項1に記載の組成物。 3 二量体がリボヌクレアーゼ二量体である請求項1に記載の組成物。 4 二量体が膵リボヌクレアーゼA二量体である請求項3に記載の組成物。 5 薬学的に許容される担体が親水性基剤である請求項1に記載の組成物。 6 薬学的に許容される担体が0.5〜1.5%NaCl溶液である請求項1に 記載の組成物。 7 請求項1に記載の組成物の有効量を人もしくは動物患者に投与するウイルス 又は細菌感染の治療方法。 8 請求項1に記載の組成物の有効量を人もしくは動物患者に投与する細胞障害 作用を伴わないウイルス又は細菌感染の酵素的治療方法。 9 組成物を体重kg当り約0.01〜50.0mgの投与量で投与する請求項 7に記載の方法。 10 組成物を経口投与する請求項7に記載の方法。 11 組成物を静脈内投与する請求項7に記載の方法。 12 請求項1に記載の組成物の有効量を人もしくは動物患者に局所的に投与す るウイルス性又は細菌性皮膚病の治療方法。 13 組成物を軟膏の形態で患者に投与する請求項12に記載の方法。 14 組成物を生理学的に許容される塩溶液の形態で投与する請求項12に記載 の方法。 15 塩溶液が0.5〜1.5%NaCl溶液である請求項14に記載の方法。 16 請求項1に記載の組成物の有効量を感染部に局所的に適用してなるヘルペ スの治療方法。 17 請求項1に記載の組成物の有効量を感染部に局所的に適用してなる感染傷 の治療方法。 18 請求項1に記載の組成物の有効量を感染部に局所的に適用してなる膣感染 の治療方法。 19 感染が膣炎である請求項18に記載の膣感染の治療方法。 20 組成物を坐薬によって適用する請求項19に記載の膣感染の治療方法。 21 請求項1に記載の組成物の有効量を感染部に適用する耳炎の治療方法。 22 組成物をドロップの形態で感染患者の耳に適用する請求項21に記載の耳 炎の治療方法。 23 請求項1に記載の組成物の有効量を人もしくは動物患者に投与する乳腺炎 の治療方法。 24 組成物を注射器注入によって感染部に適用する請求項23に記載の乳腺炎 の治療方法。 25 リゾチーム二量体及びリボヌクレアーゼ二量体からなる群から選ばれた二 量体と薬剤として使用される薬学的に許容される担体とを含有する組成物。 26 酵素の二量体の形態及び薬学的に許容される担体を含有する抗ウイルス又 は抗菌組成物。
