後天性免疫不全症候群(AIDS)は、現代の医学が直面している最も大きな健康の脅威の1つとして認識される。現在もなお、この疾患についての療法は存在しない。

本発明の1つの局面は、例えば、対応する野生型配列と比較して改善された発現を有する、HIV−1コード配列ならびに関連の配列の作製である。
第1に、得られる核酸コード配列が、高度に発現されるヒト遺伝子に見出されるコドン使用法に匹敵するように、HIV−1コドン使用法パターンを改変した。HIVコドン使用法は、コドントリプレットのヌクレオチドAまたはTの高い含量を反映する。HIV−1コドン使用法の効果は、DNA配列における高いAT含量であり、減少した翻訳能力およびmRNAの不安定性をもたらす。比較すると、高度に発現されるヒトコドンは、ヌクレオチドGまたはCを好む。HIVコード配列は、高度に発現されるヒト遺伝子において見出されるコドン使用法に匹敵するように改変された。

本発明の合成HIVコード配列(発現カセット)は、発現のレベルを評価するため、およびGag含有構築物の場合には、VLPの産生を評価するために、多数の異なる発現ベクターにクローニングされ得る。HIVポリペプチドに対する合成DNAフラグメントは、真核生物発現ベクター(一過性発現ベクター、CMVプロモーターに基づく哺乳動物ベクター、およびバキュロウイルス発現系において使用するためのシャトルベクターが挙げられる)にクローニングされ得る。対応する野生型配列もまた、同じベクターにクローニングされ得る。
ヒト免疫不全ウイルス1型(HIV−1)の群特異的抗原(Gag)は、発芽によって種々の真核生物細胞から放出される非感染性ウイルス様粒子(VLP)に自己構築する(Freed,E.O.,Virology 251:1−15,1998による総説)。本発明のGag含有合成発現カセットは、種々の異なる細胞型(哺乳動物細胞が挙げられるが、これに限定されない)を使用するHIV−Gagウイルス様粒子(VLP)の生成を提供する。

(合成コード配列についての発現アッセイ)
野生型配列は、合成HIV由来の配列がクローニングされるベクターと同じ特徴を有する発現ベクターにクローニングされる。
(発現のウエスタンブロット分析)
本明細書中に記載されるHIV発現カセットをトランスフェクトした細胞のウエスタンブロット分析を、共有に係るWO 00/39302に記載されるように本質的に実施する。ヒト293細胞を、実施例2に記載されるように、ネイティブまたは合成のHIV発現カセットを含む、pCMV6のベースのベクターでトランスフェクトする。細胞を、トランスフェクション後、60時間培養する。上澄みを記載したように調製する。細胞溶解物を、以下のように調製する。細胞を、リン酸緩衝生理食塩水で1回洗浄し、界面活性剤[0.1M Tris−HCl(pH7.5)中の1% NP40(Sigma Chemical Co.St.Louis、MO)]で溶解し、そして溶解物を新しいチューブに移す。SDS−ポリアクリルアミドゲル(前もって調製した(pre−cast)8〜16%;Novex、San Diego、CA)に、20μlの上清または12.5μlの細胞溶解液を充填する。タンパク質スタンダードもまた、充填する(5μl、広いサイズ範囲のスタンダード;BioRad Laboratories、Hercules、CA)。電気泳動を行い、BioRad Transfer Chamber(BioRad Laboratories、Hercules、CA)を用いて、製造業者(Millipore)によって推奨される転写緩衝液を用いて、100ボルトで90分間、転写を実施し、タンパク質をImmobilon P膜(Millipore Corp.、Bedford、MA)に転写させる。この膜を、HIV−1−ポジティブヒト患者血清にさらし、そしてo−フェニレンジアミンジヒドロクロリド(OPD;Sigma)を用いて免疫染色する。
合成HIV発現カセットのインビボ免疫原性
A.免疫
合成HIV発現カセットの免疫原性を評価するために、マウス研究を実施し得る。プラスミドDNA(例えば、本発明の合成配列を含む発現カセットを有するpCMVKM2)を、100μlの総注射容量において以下の最終濃度まで希釈する:20μg、2μg、0.2μgおよび0.02μg。希釈DNAの起こり得るネガティブな希釈効果を克服するために、各サンプル中の総DNA濃度を、ベクター(pCMVKM2)単独を使用して、20μgまで上昇させる。コントロールとして、ネイティブな対応するポリペプチドをコードする発現カセットを含むプラスミドDNAを、同様に扱う。4匹のBalb/cマウス(Charles River,Boston,MA)の12群に、種々の投薬量を使用して筋肉内免疫する(50μl/肢、前脛骨筋中に筋肉内注射)。
体液性免疫応答を、マウス血清の適切な抗HIV抗体ELISA(酵素結合免疫吸着アッセイ)を用いて、免疫後0および4週目(群5〜12)、そしてさらにそれぞれ免疫後6および8週目、2回目の免役後2および4週目(群1〜4)に検査する。
特異的細胞傷害性Tリンパ球(CTL)の頻度を、ペプチドをパルスしたBalb/cマウスCD4細胞の標準的なクロム放出アッセイにより評価する。HIVタンパク質発現ワクシニアウイルスに感染したCD−8細胞を、ポジティブコントロールとして使用する(vv−タンパク質)。簡潔には、脾臓細胞(エフェクター細胞、E)を、(上記のように免役した)BALB/cマウスから得る。細胞を培養し、再刺激し、そして例えば(Doe,B.およびWalker,C.M.,AIDA 10(7):793−794,1996に)記載されたGagペプチドでパルスした標的細胞に対するCTL活性についてアッセイする。細胞傷害活性を、標準的な51Cr放出アッセイにおいて測定する。標的(T)細胞をエフェクター(E)細胞とともに種々のE:T比で4時間培養し、そして複製ウェルからの平均cpmを使用して%特異的51Cr放出(%)を算出する。
合成HIV発現カセットのインビボ免疫原性
A.一般的な免疫方法
合成HIV発現カセットの免疫原性を評価するために、モルモット、ウサギ、マウス、マカクアカゲサル、およびヒヒを使用する研究を行う。研究を代表的には以下のように構築する:DNA免役単独(単回または複数回);DNA免役に続いてタンパク質免役(ブースト);DNA免役に続いてシンドビス粒子免役;シンドビス粒子単独による免役。
実験を、以下のように、モルモットを使用して実施し得る。各6匹のモルモットを含む群を、0、4および12週目に、本明細書中に記載される1以上の配列を含む発現カセットをコードするプラスミドDNAで、筋肉内または粘膜にて免疫する。続いて、約18週目に、単一用量の、プラスミドブーストの配列によってコードされるHIVタンパク質および/または他のHIVタンパク質で、動物をブーストする(筋肉内、皮内または粘膜)。抗体力価(相乗平均力価)を、3回目のDNA免役の2週間後およびタンパク質ブーストの2週間後に測定する。これらの結果を使用して、免役応答の生成のための合成構築物の有用性、およびDNA免役後の免役応答を増強するためにタンパク質ブーストを提供することの利点を証明する。
実験を、以下のように、ウサギを使用して実施し得る。ウサギを、本明細書中に記載されるHIVタンパク質をコードするプラスミドDNAで、筋肉内、粘膜、または皮内にて(Biojectニードルレスシリンジを使用して)免疫する。核酸免役に、初回免役後のタンパク質ブーストが続く。代表的には、本発明の合成HIVポリペプチドコードポリヌクレオチドを含む構築物は、ウサギにおいて、高度に免疫原性であり、そしてわずか2回の免役後に実質的な抗原結合抗体応答を生成する。
任意の免役動物モデルにおいて、体液性免役応答を、免役動物由来の血清標本において、免役後の種々の時間で、抗HIV抗体ELISA(酵素結合免役吸着アッセイ)を用いて検査する。血清の抗体力価を、上記のように抗HIV抗体ELISAによって決定する。簡潔には、免役動物由来の血清を、その動物を免疫するために使用したDNAおよび/またはポリペプチドによってコードされるHIVポリペプチド/タンパク質に対する抗体についてスクリーニングする。ELISAマイクロタイタープレートのウェルを、選択したHIVポリペプチド/タンパク質で一晩コートし、4回洗浄する;その後、ブロッキングを、PBS−0.2%Tween(Sigma)で2時間行う。ブロッキング溶液の除去後、100μlの希釈マウス血清を加える。血清を、1/25希釈率で試験し、そしてその後3倍系列希釈により試験する。マイクロタイタープレートを4回洗浄し、そしてペルオキシダーゼ結合抗マウスIgG二次抗体(Pierce,Rockford,IL)とともにインキュベートする。ELISAプレートを洗浄し、そして100μlの3,3’,5,5’−テトラメチルベンジジン(TMB;Pierce)を1ウェルあたりに加えた。各ウェルの光学密度を15分後に測定する。力価を、最大半値光学密度(O.D.)を与えた血清希釈率の逆数として代表的に報告した
細胞性免疫応答もまた評価し得る。
本発明の合成HIVポリヌクレオチドを含む発現カセットを使用する、ヒヒおよびマカクアカゲサルのDNA免役
A.ヒヒ
4匹のヒヒを、本明細書中に記載の遺伝子送達ビヒクルを使用して、3回(0、4および8週目)、四頭筋への筋肉内にて両側性に、または粘膜にて免役する。各免役の2週間後に動物から採血し、そしてHIV抗体ELISAを単離した血漿で実施する。ELISAを、二次抗体結合体が、1:500の希釈率で使用される抗ヒトIgG、g−鎖特異的ペルオキシダーゼ結合体(Sigma Chemical Co.,St.Louis,MD 63178)であることを除き、本質的には上記のように実施する。50μg/mlの酵母抽出物を、ヒヒの既存の酵母抗体に起因する非特異的バックグランドを低減させるために、血漿サンプルおよび抗体結合体の希釈物に加え得る。4回目の免役の2週間後(14週目)に、ヒヒにおいて、リンパ球増殖応答が観察され、そしてHIVポリペプチドでのブースト後(44週目および76週目)に実質的に増強される。このような増殖結果はT−ヘルパー細胞機能の誘導を示す。
本発明の合成コドン改変HIVポリペプチドコードポリヌクレオチドの向上した効力は、発現プラスミド中に構築される場合、マカクアカゲサルにおいて確証され得る。代表的には、マカクは、2または3回の1mg用量の改変HIVポリヌクレオチドの後、検出可能なHIV特異的CTLを有する。まとめると、これらの結果は、合成HIV DNAが非ヒト霊長類において免疫原性であることを証明する。中和抗体もまた検出され得る。
(モノシストロン性およびマルチシストロン性構築物の同時トランスフェクション)
本発明は、マルチ、モノシストロン性発現カセットでの同時トランスフェクション、ならびに1以上のマルチシストロン性発現カセットでの同時トランスフェクション、またはその組合せを包含する。
(HIV−1ワクチンの付属遺伝子成分:変異型Tat、RevおよびNefのC型抗原の機能分析)
HIV−1調節および付属遺伝子は、ウイルスの病原性における役割、高度に保存されたCTLエピトープの高い割合、およびウイルスのライフサイクルにおける早期の発現に起因して、HIVワクチンの成分として大きな注目を受けた。これらの遺伝子の様々な所望されない特性に起因して、ワクチン成分としてのそれらの安全性および安定性に関する疑問が生じる。本発明に支持して実行された実験は、効率的な発現および潜在的な有害な機能の不活性化について、候補HIV−1サブタイプC tat変異体、rev変異体およびnef変異体を分析した。他のHIVサブタイプの付属遺伝子は、同様に評価され得る。
(プラスミドをコードする様々なHIVポリペプチドの免疫原性の評価)
上記のように、本明細書中に記載される任意のポリヌクレオチドまたは発現カセットの免疫原性を、容易に評価する。以下の表(表D)は、ウサギ中で、エレクトロポレーションを使用して、独立して、そして混合サブタイプ(mixed−subtype)ワクチンとしての両方でサブタイプBエンベローププラスミドおよびサブタイプCエンベローププラスミドの免疫原性の比較に関する例示的手順である。このような方法は、任意の他のHIVポリペプチドに等しく適用可能であることが明らかである。
(GagおよびPol構築物でのマウス免疫化研究)
マウスにおいて、以下の構築物:Gag、Gagプロテアーゼ(+FS)(GP1、プロテアーゼ、コドン至適化およびINSの不活性化;GP2、プロテアーゼ、INSの不活性化のみ)、フレームシフトを有するGagPolΔインテグラーゼ(gagFSpol)、およびGagPolΔインテグラーゼインフレーム(GagPol)(図118を参照のこと)について、細胞性免疫反応および体液性免疫反応を評価した(実施例4に実質的に記載される)。弱力化プロテアーゼ(GagPolAtt)または非機能性プロテアーゼ(GagPolIna)を有するGagPolΔインテグラーゼインフレームの変形もまた、設計した。
(C型由来エンベロープポリペプチドを使用したタンパク質発現、免疫原性、および中和抗体の産生)
南アフリカ人の個体から回収されたC亜型初代単離体、TV1に由来するエンベロープ(Env)ワクチンを、以下のようにウサギで試験した。遺伝子カセットを、可変ループ領域、V2およびV1V2の欠失があるHIV−1エンベロープおよび欠失がないHIV−1エンベロープのポリタンパク質のgp120形態(表面抗原)、gp140形態(表面抗原+膜貫通タンパク質、gp41のエクトドメイン)、および全長(gp120+gp41)gp160形態を発現するように設計した。全ての遺伝子を配列改変して、Rev非依存様式での、コードされるEnv糖タンパク質の発現を増強し、そしてそれらを、続いて上記のDNAワクチンおよびタンパク質産生適用のためのpCMVベースのプラスミドベクターにクローニングした。この配列を、本明細書中に記載されるようにコドン至適化した。簡潔には、すべての改変エンベロープ遺伝子を、Chiron pCMVlinkプラスミドベクターに(好ましくは、EcoRI/XhoI部位に)クローニングした。
TV1 Env抗原の全長(gp160)、短縮gp140(Envエクトドメインのみ)およびgp120のネイティブ型を、本明細書中に記載される発現カセットから産生した。gp140をコードする配列を、293T細胞に一過性にトランスフェクトした。遺伝子産物の発現レベルを、組織内(in−house)抗原捕捉ELISAによって評価した。TV001c8.2、TV001c8.5およびTV002c12.1のネイティブ配列から構築したエンベロープ遺伝子は、インビトロで正確なタンパク質を発現した。gp140TV001c8.2が、最高レベルの発現を示した。さらに、TV1由来クローン8.2から発現されたEnvタンパク質が、CD4レセプタータンパク質に結合すること(このことは、発現されたタンパク質のこの特性が、機能的配置で維持されていることを示す)を見出した。発現されたTV1 gp160タンパク質果のレセプター結合特性/機能性の結果もまた、細胞融合アッセイによって確認した。
一過性発現実験は、TV001ウイルス単離体およびTV002ウイルス単離体由来のエンベロープ遺伝子が所望のタンパク質産物を発現することを示した。これらの2つのウイルス株の、18人の感染した南アフリカ人(B亜型およびC亜型)からの血清を使用した相対的中和感度は、以下のとおりである。TV2株は、1:10血清希釈において、18血清中18血清で;1:50血清希釈において、18血清中16血清で;1:250血清希釈において、18血清中15血清で中和された。それに比べて、TV1単離体は、1:10血清希釈において、18血清中15血清で;1:50血清希釈において、18血清中6血清のみで中和され;そして1:250血清希釈において、いずれの試料によっても中和されなかった。さらに、TV001患者血清は、試験したすべての希釈で、TV002単離体に対する中和活性を示した。対照的に、TV002は、1:10血清希釈においてのみ、TV001の中和を示した。これらの結果は、TV001単離体が、感染した宿主において、TV002よりもより広範かつより強力な中和抗体反応を誘導し得ることを示唆する。
TV1 Env DNA(合成発現カセットを含む)およびタンパク質ワクチンを、下の表Hに示すように投与した。
DNA免疫には、C亜型配列改変遺伝子(TV1)−−gp160、gp160dV2、gp160dV1V2、gp140、gp140dV2およびgp140dV1V2−−ならびにB亜型SF162配列改変gp140dV2を使用した。DNA免疫を、0週目、4週目および20週目に針注射により、筋細胞および常在の抗原提示細胞のトランスフェクションを促進するためにエレクトロポレーションを使用して筋肉内経路によって実施した。
PBMC−増殖R5 HIV−1株に対する中和抗体反応を、2つの異なる研究所における2つの異なるアッセイ(Chironにおける5.25レポーター細胞株ベースのアッセイおよびDuke大学におけるDavid MontefioriのPBMCベースのアッセイ)を使用して、免疫したマウスから回収した血清においてモニターした。結果を、図121、図122、および図123に示す。Chironアッセイを、基本的に以下のように実施した。PBMC−増殖C亜型TV001株およびPBMC−増殖C亜型TV002株に対する中和抗体反応を、5.25細胞株を用いる組織内レポーター細胞株アッセイを使用して測定した。この細胞は、その細胞膜上にCD4、CCR5、CXCR4およびBONZOレセプター/コレセプターを有する。親の(parental)CEM細胞株は、急性リンパ芽球性白血病に罹患した4歳の白人の(Caucasian)女性に由来し、この細胞をヒトB細胞株721.174と融合して、CEMx174を作製した。CCR遺伝子(約1.1kb)を、pBABEpuroレトロウイルスベクターのBamH−I(5’)およびSal−I(3’)にクローニングし、それに次いでCEMx174に導入した後、LTR−GFPをこの細胞にトランスフェクトした。この細胞の緑色蛍光タンパク質(GFP)を、フローサイトメータ(FACScan)によって検出した。ウイルス中和アッセイについて、50ulの力価決定した(titrate)ウイルスおよび50ulの希釈免疫血清または免疫前血清を、室温で1時間インキュベートした。24ウエルプレートにプレートした、104細胞/ml細胞を有するウエルに、この混合物を添加し、そして37℃で5〜7日間インキュベートした。次いで、この細胞を、PBSで洗浄した後、2%ホルムアルデヒドで固定した。Cellquestソフトウエアを使用して、Becton Dickinson FACScanで各サンプルについて15,000事象(細胞)を収集した。示したデータは、3連のウエルの平均であった。以下の式:%ウイルス阻害=(ウイルスコントロール−実験)/(ウイルスコントロール−細胞コントロール)×100、を使用して、ウイルスコントロールと比較して、%中和を算出した。採血前に観察された任意のウイルス阻害を、各個々の動物について差し引いている。>50%の値を陽性と見なし、グレーで強調する。
(Rhesus Macaquesにおける免疫学的応答)
表Iに示すように構成された免疫化研究において、細胞性免疫反応および体液性免疫反応を、3群のrhesus macaques(各群は、4匹の動物で構成された)で評価した。免疫化組成物についての投与経路は、いずれの場合においてもエレクトロポレーションであった。抗体価を、2回目の免疫の2週間後について表Iに示す。
Claims (47)
- 第二可変領域(V2)を欠失したHIV Envポリペプチドを含むポリペプチドをコードするポリヌクレオチド配列を含む発現カセットであって、該Envポリペプチドをコードする該ポリヌクレオチド配列が、配列番号26に示される配列を含む、発現カセット。
- 請求項1に記載の発現カセットを含む、選択された宿主細胞における使用のための組換え発現系であって、前記ポリヌクレオチド配列が、該選択された宿主細胞における発現と適合する制御エレメントに作動可能に連結されている、組換え発現系。
- 請求項2に記載の組換え発現系であって、前記制御エレメントが、転写プロモーター、転写エンハンサーエレメント、転写終結シグナル、ポリアデニル化配列、翻訳の開始の最適化のための配列、および翻訳終結配列からなる群より選択される、組換え発現系。
- 請求項2に記載の組換え発現系であって、前記転写プロモーターが、CMV、CMVおよびイントロンA、SV40、RSV、HIV−Ltr、MMLV−ltr、ならびにメタロチオネインからなる群より選択される、組換え発現系。
- 請求項1に記載の発現カセットを含む細胞であって、前記ポリヌクレオチド配列が、前記選択された細胞における発現に適合する制御エレメントに作動可能に連結されている、細胞。
- 前記細胞が哺乳動物細胞である、請求項5に記載の細胞。
- 前記細胞が、BHK細胞、VERO細胞、HT1080細胞、293細胞、RD細胞、COS−7細胞、およびCHO細胞からなる群より選択される、請求項6に記載の細胞。
- 前記細胞が、CHO細胞である、請求項7に記載の細胞。
- 前記細胞が、昆虫細胞である、請求項5に記載の細胞。
- 前記細胞が、Trichoplusia ni(Tn5)昆虫細胞またはSf9昆虫細胞のいずれかである、請求項9に記載の細胞。
- 前記細胞が、細菌細胞である、請求項5に記載の細胞。
- 前記細胞が、酵母細胞である、請求項5に記載の細胞。
- 前記細胞が、植物細胞である、請求項5に記載の細胞。
- 前記細胞が、抗原提示細胞である、請求項5に記載の細胞。
- 請求項14に記載の細胞であって、前記抗原提示細胞が、リンパ細胞であり、該リンパ細胞が、マクロファージ、単球、樹状細胞、B細胞、T細胞、幹細胞、およびその始原細胞からなる群より選択される、細胞。
- 請求項5に記載の細胞であって、前記細胞が、初代細胞である、細胞。
- 請求項5に記載の細胞であって、前記細胞が、不死化細胞である、細胞。
- 請求項5に記載の細胞であって、前記細胞が、腫瘍由来細胞である、細胞。
- HIV Envポリペプチド配列を含むポリペプチドを産生するための方法であって、該方法が、以下:

を包含する、方法。 - 哺乳動物被験体における使用のための遺伝子送達ベクターであって、
該被験体における使用のための適切な遺伝子送達ベクターを含み、該ベクターが、請求項1に記載の発現カセットを含み、そして前記ポリヌクレオチド配列が、該被験体における発現と適合する制御エレメントに作動可能に連結されている、遺伝子送達ベクター。 - 請求項20に記載の遺伝子送達ベクターを含む、被験体のDNA免疫化のための組成物であって、該組成物は、該被験体における前記発現カセットの発現と適合する条件下で、該被験体に導入されるように調製されたものである、組成物。
- 前記遺伝子送達ベクターが、非ウイルスベクターである、請求項21に記載の組成物。
- 前記ベクターが粒子性キャリアーを使用して送達されるように調製されたものである、請求項21に記載の組成物。
