20Lのジャケット付きガラス反応容器に、イサト酸無水物(500g、3.06mol)およびエタノール(2500mL)を入れた。この後、30wt%メチルアミンのエタノール溶液(378.5g、3.98mol)を、20±5℃でさらにエタノール(330mL)で滴下漏斗から70分かけて希釈し、調節しながら滴下した。得られた混合物を20±5℃で60分間攪拌し、反応の完了をHPLCで確認した(<1A% SM)。反応が完了したら、13% NaCl(2610mLのDI水に390gのNaClを溶解して調整した)を20.0分かけて加えた。得られた混合物を10分間20±5℃で攪拌し、10分間放置した。底部の水層を除いた。有機層を(2210mLのDI水にNaCl 792gを溶解して調整した)26% NaClで洗った。合わせた水層洗浄液を酢酸エチル(5100mL)で抽出した。前記バッチと酢酸エチル抽出物を合わせ、バッチ容積が約1.5L(イサト酸無水物重量の3倍)になるまで12Lの丸底フラスコで30〜40℃で減圧(50〜80mmHg)濃縮した。前記残渣に酢酸エチル(5100mL)を追加し、次に2回目は約1.5L(イサト酸無水物重量の3倍)になるまでで減圧(50〜80mmHg)濃縮した。この濃縮バッチにアセトニトリル5.0Lを追加した。得られた混合物を室温で60分間攪拌し、空隙率が細かい焼結ガラス漏斗にセライトのパッドを載せてろ過した。前記パッドをアセトニトリル(500mL)で洗い、洗浄液を前記バッチと合わせた。透明なろ液を20Lのジャケット付きガラス反応容器に移した。Hunigの塩基(712.6g)と2,4,5−トリクロロピリミジン(657.3g)を加えた。工程内試験で示されるとおり、前記反応が完了するまで、得られた溶液を73±3℃に加熱し、73±3℃で攪拌した。前記反応混合物を30分で0±5℃に冷却し、この温度で1〜2時間攪拌した。前記生成物を焼結ガラス漏斗で減圧ろ過により回収した。前記ケーキをアセトニトリル(1240mL)で洗い流し、残った水およびアセトニトリルの内容量がNMR解析およびKFによる用量調節で1wt%未満となるまで、窒素ブリードにより減圧吸引乾燥した。合計746.3g(全体の収率81.9%)のCEP−19036を、次の品質特性で淡黄褐色の固体として得た:99.8A%、98.99wt%、0.1wt% NaCl、0.1wt% H2O、0.1wt%アセトニトリル。
オーバーヘッドスターラーと滴下漏斗を取り付けた5Lの三口丸底フラスコに遊離塩基(145.83g、251.4mmol)を合わせた。これらの固体にnBuOHを加え、40分間攪拌後、黄色の混濁溶液(大きな固体なし)が得られた。直ちに沈殿が起こり、23℃までわずかな発熱が認められた。得られたスラリーは45分間で85℃に加熱した。60℃付近で固体は溶解した。加熱が80℃に達したら、HCl(1:1:1 MeOH/EtOH/水で2.5M、307mL、767mmol)を4分かけて追加し、種結晶(CEP−37440 H2A3、1.7g)を加えた。85℃では1時間で種結晶はそのまま残り、さらに個体が形成した。この溶液を1時間かけて室温まで冷却し、さらに30分間2℃まで冷却した。このスラリーを0〜5℃で1時間かけて攪拌してから、ろ過し、0℃のnBuOH(400mL)で洗った。最初のケーキの寸法(シリンダー)は13.5cmの直径で6.0cmの高さであった。洗浄後、圧縮したケーキは4.3cmの高さとなった。母液の損失は約0.5%であった。得られた固体を60℃で24時間減圧乾燥し(最初の16時間後、N2をパージ)、望みの生成物162gが得られた(98.8HPLC純度、出発原料を100wt%と想定し単離収率88%、NMR、XRPD、およびm.p.で残りのnBuOH 0.5%、H2A3塩形態として確認)。

Claims (43)
- 請求項1記載の方法において、前記アルコールはメタノールである方法。
- 請求項1または2記載の方法において、前記化合物IAのL−酒石酸塩は、少なくとも85%のジアステレオマー過剰率で生成される方法。
- 前述の請求項のいずれか1つに記載の方法において、前記分割する工程は約15℃〜約40℃の温度で実施される方法。
- 前述の請求項のいずれか1つに記載の方法において、前記溶媒系における化合物IAおよびIBの混合物の濃度は、約0.02g/mL〜約1g/mLである方法。
- 請求項6記載の方法において、前記第1の水素化触媒は、イリジウム触媒、ロジウム触媒、またはルテニウム触媒である方法。
- 請求項7記載の方法において、前記第1の水素化触媒は、ビス((μ−クロロ)ビス(シクロオクテン)イリジウムである方法。
- 請求項6〜8のいずれか1つに記載の方法において、前記リガンドはMETAMORPhosである方法。
- 請求項6〜9のいずれか1つに記載の方法において、前記添加物はホウ酸塩である方法。
- 請求項10記載の方法において、前記ホウ酸塩がテトラキス[3,5−ビス(トリフルオロメチル)フェニル]ホウ酸ナトリウムである方法。
- 請求項6〜11のいずれか1つに記載の方法において、前記水素は大気圧以上の圧力で存在する方法。
- 請求項6〜12のいずれか1つに記載の方法において、前記水素は約40〜60atmの圧力で存在する方法。
