本明細書では、治療薬の結合のオフターゲット結合を評価するために有用であるバキュロウイルス(BV)粒子への結合の検出に基づくアッセイを記述する。このアッセイは、とりわけ、適切な薬剤を得る確率を高めるために、抗体リード・ジェネレーション又は最適化中に使用することができる。
所定の実施態様において、本明細書において与えられる抗体は、解離定数(Kd)が、≦1μM、≦100nM、≦10nM、≦1nM、≦0.1nM、≦0.01nM、又は≦0.001nM(例えば、10−8M以下、例えば、10−8Mから10−13M、例えば、10−9Mから10−13M)。

ある実施態様において、本明細書で提供される抗体は、多重特異性抗体、例えば二重特異性抗体である。多重特異性抗体は、少なくとも二つの異なる部位に対して結合特異性を有するモノクローナル抗体である。ある実施態様において、結合特異性の一つは[[PRO]]に対してであり、他は、任意の他の抗原に対してである。ある実施態様において、二重特異性抗体は、[[PRO]]の2つの異なるエピトープに結合することができる。二重特異性抗体はまた[[PRO]]を発現する細胞に対して細胞傷害性薬物を局在化させるために用いることができる。二重特異性抗体は、全長抗体又は抗体断片として調製することができる。

ある実施態様において、一つ以上のアミノ酸置換を有する抗体変異体が提供される。置換型変異誘発の対象となる部位は、HVRとFRを含む。保存的置換は、表1の「保存的置換」の見出しの下に示されている。より実質的な変更が、表1の「典型的な置換」の見出しの下に与えられ、アミノ酸側鎖のクラスを参照して以下に更に説明される。アミノ酸置換は、目的の抗体に導入することができ、その産物は、所望の活性、例えば、抗原結合の保持/改善、免疫原性の減少、又はADCC又はCDCの改善についてスクリーニングされる。

ある実施態様において、1つ又は複数のアミノ酸改変を、本明細書で提供される抗体のFc領域に導入することができ、それによってFc領域変異体を生成する。Fc領域の変異体は、1つ又は複数のアミノ酸位置においてアミノ酸修飾(例えば置換)を含むヒトFc領域の配列(例えば、ヒトIgG1、IgG2、IgG3又はIgG4のFc領域)を含んでもよい。

カニクイザルにおけるクリアランスの決定
この実施例では、非ヒト霊長類における抗体のクリアランスを決定するために使用される標準的な方法を示す。
FcRnの親和性の決定
次の例では、平衡解離定数(KD)をどのように決定することができるかを詳述する。 固定化抗体へのカニクイザルFcRnの結合についての平衡解離定数(KD)は、ビアコア(登録商標)4000装置(GE Healthcare)による表面プラズモン共鳴(SPR)測定から決定した。前述のようにカニクイザルFcRnのを調製した(Yeung, Y.A.ら、Engineering human IgG1 affinity to human neonatal Fc receptor: impact of affinity improvement on pharmacokinetics in primates. J Immunol 182, 7663-7671 (2009))。シリーズSセンサーチップがドッキングされ、標準化され、製造業者によって供給されたプロトコルを使用して、水力学的アドレッシングのために調製された。4つのフローセルの各々の全5スポットはアミンカップリングのために、EDC/NHS溶液(0.2MのN−エチル−N’−(3−ジエチルアミノプロピル)−カルボジイミド(EDC)及び0.05MのN−ヒドロキシスクシンイミド(NHS)に10分間の曝露により活性化された。抗体は、2μg/mlの抗体及び10mMのNaOAc(pH5)を含む溶液への7分間の曝露により、スポット1、2、4及び5に結合された。未反応部位はその後、1Mのエタノールアミンへの全ての5スポットの7分間の曝露を通じてブロックした。この形式では、スポット3は基準セルであり、4つの異なる抗体がフローセルごとに結合され、センサーチップあたり合計16となる。カップリング密度は、抗体の200〜1000RUの範囲であった。別々の実験では、計算されたKDは、この範囲のカップリング密度では変化しなかったことが示された(データ非表示)。10μMから0.04μMの濃度で2倍刻みで変化させたカニクイザルFcRnの一連の溶液は、25mMのMES、25mMのHEPES、150mMのNaCl、0.05%ポリソルベート20、pH5.8を含むランニングバッファー中で調製した。センサーグラムは、30μL/分の流速でのセンサーチップ上で、これらの溶液及びバッファーのみのコントロールの60μLの注入について収集した。測定温度は25℃で、解離は180秒間モニターされ、次いで結合したままの任意FcRnは、50mMのトリス−HCl、150mMのNaCl、0.05%ポリソルベート20、pH8を含む溶液30μLを注入することにより溶出された。解離定数は、ビアコア4000評価ソフトウェア1.0を使用して定常状態の親和性解析から計算した。
ELISA用抗原の作製
この例では、本明細書に記載のアッセイで使用するためのマイクロタイタープレートをコーティングするために使用される基質の作製が記載されている。
ELISAアッセイ
抗原(例えば、バキュロウイルス粒子)を384ウェルのELISAプレート(Nunc Maxisorp)に、各ウェルに、50mMの炭酸ナトリウム緩衝液pH9.6中の1%バキュロウイルス懸濁液25μlを添加することにより、固定化し、粒子を4℃で一晩、プレートに吸着させた。ウェルをブロッキング緩衝液(0.5%BSAを含有するPBS)50μlで室温で1時間ブロックした。PBSでプレートを3回洗浄した後、精製した抗体はブロッキング緩衝液で段階希釈し。25μlをELISAウェルに二連で添加し、室温で1時間インキュベートした。プレートをPBSで6回洗浄し、西洋ワサビペルオキシダーゼ(Jackson)にコンジュゲートした10ng/mlのヤギ抗ヒトIgG(Fcガンマ断片特異的)25μlを各ウェルに加えた。プレートは、室温で1時間インキュベートし、PBS中で6回洗浄し、TMB基質25μlを各ウェルに加えた。反応は、15分後に各ウェルに1Mリン酸を25μl添加することによって停止され、吸光度を450nmで読み取り、620nmで参照された。BVスコアは、非被覆ウェルについて観察された平均シグナルで除算することにより各々が正規化されていた6つの光学密度測定の平均から計算した。Sf9細胞膜によるアッセイにおいて、バキュロウイルス粒子の代わりにELISAプレートを被覆するため粗製膜の2%懸濁液を使用した。界面活性剤は、任意の段階でバッファーに添加されなかった。

HEK293細胞を、DMEM−10%ウシ胎児血清中に5×106細胞/mlで再懸濁し、100μl/ウェルでU底96ウェルプレートに分注した。PBS(150mMのNaCl、10mMリン酸ナトリウム、pH7.4)で希釈したIgGの等量(30〜50nMの最終濃度)を細胞に添加し、4℃で1時間インキュベートした。次いで、細胞を、氷冷PBSで3回洗浄し、抗ヒトIgG Fc−R−フィコエリトリンコンジュゲート(Jackson Immunoresearch)又は抗ヒトIgG−Alexa488(Molecular Probes)コンジュゲートと共に4℃で30分間インキュベートし、氷冷したPBSで2回洗浄し、0.1%パラホルムアルデヒドを含むPBS中で固定した。細胞は、ハイスループットサンプラーを装備したFACSCaliburフローサイトメーター(BD Biosciences)で分析した。
統計的分析
全ての統計分析は、Rを用いて行った(R Development Core Team (2011)。R:統計計算のための言語と環境。統計コンピューティングのためのR財団、ウィーン、オーストリア。ISBN 3-900051-07-0,R-project[dot]org)で、オンラインで入手可能。.図5の説明に記載のようにオッズ比を使用して急速及び低速でクリアする抗体を正しく同定することにおけるBVスコアの性能の評価は、BVのビニング閾値=5を決定するために使用される同じデータセットを用いて行ったので楽観的である。5分割交差検定及びBVスコア閾値を決定する回帰モデリングを使用する10mL/kg/日のクリアランス閾値に対してインビトロアッセイの有用性のより保守的な評価が得られた;このアプローチは、14.1のより控えめなオッズ比推定値を与えた(95%信頼区間(2.4,113.3))。
サルのクリアランスからのヒトのクリアランスの予測的スケーリング
以前に利用可能な8つよりもわずかに大きいデータセットを使用して、我々は、カニクイザル及びヒトにおいて測定されたクリアランス値の間に強い相関関係を確認している(図1、スピアマンの相関係数(ρ)=0.74)。ヒトで測定されたクリアランスはカニクイザルにおいて測定されたクリアランスより約2倍遅いとする単純なスケーリングガイドラインが得られ、異常値として抗NRP1抗体を含む。
治療用抗体のパネルのためのカニクイザルにおける構造変異体とクリアランスの特徴付け
前臨床開発の通常の過程において、カニクイザルにおける薬物動態データが、52のヒト化及びヒト抗体用に収集された。これらの抗体の優勢なアイソタイプは、IgG1、カッパ(n=46)である。この抗体パネルはまた4つのIgG1、ラムダ、及び2つのIgG4、カッパ抗体を含む。IgG4抗体は、S228Pヒンジ安定化置換を組み込んでいる17。52抗体のうち5つは、エフェクター機能を低減するためにアグリコシル化(aglycosylated)され、1つはADCC活性を増加させるためにアフコシル化され、及び3つはFcガンマR結合を切断又は増加させるFcのアミノ酸置換を有する。これらの抗体の可変ドメインは、ヒト化(n=32)、合成ヒト抗体ファージライブラリー18由来(n=16)、ヒト(n=3)、又はマウス(n=1;リツキシマブ)の何れかである。これらの抗体は、広範囲のクリアランス値を示し(図2)、15(29%)の抗体が10mL/kg/日より速いクリアランスを有する。
カニクイザルへの結合親和性
FcRn結合における変化が観察されたクリアランス値の範囲を説明することができるかどうかを試験するために、我々は、pH5.8で精製したカニクイザルのFcRnに対するこれらの抗体の結合についてのKD値を決定するために、表面プラズモン共鳴(SPR)法を用いた。IgG−FcRn結合は比較的弱い相互作用であることから、我々は、結合定数を決定するために、以前に説明した定常状態のアプローチ11を使用した。2:1のFcRn:IgG相互作用から生じる結合活性効果は、分析用に利用可能である各々の抗体(n=44)を固定することによって回避された。この方法で決定されたKD値は、約250nMから1500nMの範囲であった。各抗体は、アミノ基を介するランダムなカップリングを用いて別々に固定化し、センサーチップ表面上の配向に付随する不確実性を有するため、KD値の範囲は予想外ではなかった。ペルツズマブのKDの複数回(n=7)の測定では、940±140(S.D.)nMの平均値を示した。我々は、FcRnの親和性を増加させることが知られたFc変異体の結合作用(データ非表示)を再現できたので、本測定方法は、FcRnの親和性の変化を検出することができる19。幾つかの抗体について観察された速いクリアランスは、カニクイザルFcRnに対する低pH結合親和性の改変に起因するものではない(図4)。実際に、ほぼ同一のFcRn結合親和性を持つ複数の抗体の比較では、クリアランスの値に約30倍の値域を与えた。別々の実験(データ非表示)では、pH7.4でこれらの抗体からのカニクイザルのFcRnの解離速度は高速であり、試験されたパネル全体で同等であることを示した。これらの実験は、観察された高速クリアランスについての説明として、幾つかの抗体のリサイクル経路の変化を除外することは不十分であるが、pH5.8での結合親和性又は中性pHでの解離動態における有意な変化が高速クリアランスに大きな役割を果たしているという可能性は低いように見える。
高速のクリアランスのリスク軽減のためのインビトロアッセイの開発
少数の抗体について観察された高速クリアランスは、特異的14,15又は非特異的13、16オフターゲット結合に関連している。抗体の産生及び最適化の最中に、我々のパネル中の抗体のほとんどを、密接に関連する抗原ホモログへの結合の欠如についてスクリーニングした。加えて、ファージディスプレイを介して由来する抗体の多くは、非特異的結合の濃縮を防ぐために添加剤の存在下で選択した20。Wuら13によって示されるように、限られた種の抗原によるELISAによるスクリーニングは、インビボでの挙動に影響し得る特異的又は非特異的オフターゲット結合の種類を検出できない場合がある。従って、我々は、高速クリアランスを示す可能性が高い候補抗体を同定するために有用なインビトロアッセイを開発しようとした。ELISAアッセイにおいて抗原として特異的膜結合型標的を有するインタクトなBV粒子を用いる実験の過程で21、我々は、幾つかの抗体は、標的を発現しないバキュロウイルス粒子に結合することを述べた。これらの抗体の多くはまた、FACSにより、非発現性ヒト293細胞に非特異的に結合することが示された(データ非表示)。しかし、293細胞上の幾つかの同族抗原の発現は、非特異的結合から特異的結合を区別することを困難にした。
抗体のこのパネルについて測定されたカニクイザルにおけるクリアランス値の広範な分布、並びに15の抗体で観察された高速クリアランス(>10mL/kg/日)は予想外であった。52の抗体のうち40が同一のヒトIgG1のFc配列を有しているため、より高速なクリアランスはFcRnとの相互作用の変化とは関連されないことが予想された。実際、pH5.8でのFcRn結合について測定されたKDは7倍の値域にわたって変化するが、これはクリアランス値の傾向とは関連していない。pH5.8でのFcRnの親和性の比較的大きな増加がクリアランスの緩やかな改善へと導くとすれば19、ここで測定されるFcRnの親和性の僅かな相違がクリアランスに影響を与えないことは驚くべきことではない。試験した抗体の全ては、中性pHで、FcRnから同等の迅速な放出を示した。Wangらによって報告された結果とは対照的に22、我々は、中性pHでのFcRnの解離動態において抗体のFab部分への影響とクリアランスとの間の関連を検出していない。我々の研究は、結合に対する結合活性効果を最小にするために、より大きな抗体のパネルとSPR実験の異なる配向を使用した。
抗体クリアランスの最適化
この実施例において、我々は、更なる操作を通じて、候補選択の補助を支援するために、BV ELISAが将来を見越して使用することができるかどうかを試験した。
