Abstract
【解決手段】本発明は、少なくとも1つの標的遺伝子の2つまたはそれ以上の配列と実質的に相同かつ相補的である、2つまたはそれ以上の異なる二本鎖RNA配列を含む、複数標的の部分的に二本鎖のRNA分子を用いることで上述した課題を解決するものである。

Description
当該分野で周知の問題の宿主は、有用な医薬品として広く受け入れられるようになってきた多数のポリヌクレオチド組成物を妨害してきた。従って、哺乳類における疾患の治療のために医学界ですでに受け入れられているかかるDNAワクチンまたは治療剤はあまりない。
さらにさらなる態様において、本発明は、標的ポリヌクレオチドが感染哺乳類細胞内でウイルスの複製および/または発生病理に必要なウイルスポリヌクレオチド配列である、1つ以上の前記記載の組成物を、医薬上許容される担体中、細胞内のポリヌクレオチド取り込みを促進する所望の第二物質と共に哺乳類に投与することによって、哺乳類のウイルス感染を予防する方法を提供する。本組成物は、その後のウイルスの哺乳類細胞への導入におけるウイルス配列の機能を減少あるいは阻害するのに有効な量で投与される。
もう1つの態様において、本発明は、細胞内病原体による哺乳類の感染の治療または予防方法を提供する。哺乳類には、標的ポリヌクレオチドが、感染哺乳類細胞内で病原体の複製および/または発生病理に必要な細胞内病原体のポリヌクレオチド配列である、1つ以上の本明細書中に記載の組成物を投与する。本組成物は、哺乳類において配列の機能を減少あるいは阻害するのに有効な量で、医薬上許容される担体中、細胞内ポリヌクレオチド取り込みを促進する所望の第二物質と共に投与される。
さらにもう一つの態様において、本発明は哺乳類の癌の治療または予防方法を提供する。哺乳類に、標的ポリヌクレオチドが、また遺伝子または制御配列の正常なコピーを有する、哺乳類における異常な癌を引き起こす遺伝子または非−発現制御配列である1つ以上の前記記載の組成物を投与する。この態様によれば、異常な配列および正常な配列間の差異はポリヌクレオチドの差異である。本組成物は、哺乳類において異常な配列の機能を減少あるいは阻害するのに有効な量で、医薬上許容される担体中、細胞内ポリヌクレオチド取り込みを促進する所望の第二物質と共に投与される。
本発明の他の態様は、以下のその好ましい具体例の詳細な記載に記載される。
本発明によって、哺乳類細胞の標的ポリヌクレオチド配列の機能を阻害する組成物は、少なくとも部分的二本鎖のRNA分子を哺乳類細胞に供する物質を含む。一般に、「RNA」なる語はまた、他に記載がなければ、例えば、リボースの2'−OH基が独特の結合に必要であるといったRNA−DNAハイブリッドも含む。RNA分子は分子中に少なくとも長さ約200ヌクレオチドのポリヌクレオチド配列を含む。重要なことには、RNA分子のこのポリヌクレオチド配列は、実質的に標的ポリヌクレオチド配列と相同である。このポリヌクレオチド配列はまた、好ましくは、エクソン配列またはその部分を含む。望ましくは、ポリヌクレオチド配列はイントロン配列を含まない。好ましくは、RNA分子は機能タンパク質を産生せず、より好ましくは、翻訳されない。RNA分子のポリヌクレオチド配列は、好ましくは実質的に、いずれの天然に存在し、正常に機能し、必須の哺乳類ポリヌクレオチド配列とも非−相同である。本明細書中に記載のポリヌクレオチド配列は、複数標的またはポリエピトープ方法、例えば、単一の標的病原体の、1つ以上の標的病原体に対する、またはサイレンシングしようと望まれる他の種類の標的の1つ以上の遺伝子の配列をコードすることを使用する。
さらにもう1つの具体例において、RNAポリヌクレオチド配列は望ましくは、標的配列に対し少なくとも10%の全体的相同性を有し、その領域で標的配列の同様の30nts領域に対して少なくとも50%の相同性を有する30の連続したヌクレオチド領域を少なくとも2つ含む。この式の他の具体例は当業者であれば発展させることができる。
相同性を算出するのに必要な相同性の選択、プログラムのためのデフォルトの選択および用いられるプログラムの選択は、本明細書のおしえ、科学論文中の知識があるとすれば当該分野の範囲内であると理解されるべきである。

医薬的使用のための本発明の組成物は、望ましくは前記の合成RNA分子または医薬品送達のための付加的な所望の成分と共に医薬上許容される担体中、インビボで哺乳類細胞へそのRNA分子を供する物質を含む。医薬組成物の具体的処方は、RNA分子を送達する物質の形態に依存する。
適当な医薬上許容される担体は、本発明のポリヌクレオチド組成物の投与を促進するが、生理学的に不活性かつ/あるいは無害である。担体は当業者によって選択される。かかる単体は、滅菌生理食塩水、リン酸、緩衝生理食塩水、デキストロース、滅菌水、グリセロール、エタノール、乳糖、シュークロース、リン酸カルシウム、ゼラチン、デキストラン、カンテン、ペクチン、落花生油、オリーブ油、ゴマ油および水、およびその組み合わせを含むがそれに限定されない。さらに、担体または希釈剤は、単独であるいはロウと一緒にグリセロール、モノステアリン酸またはジステアリン酸グリセロールのごとき時間遅延物質を含んでもよい。さらに、徐放性ポリマー処方が使用できる。処方は、送達物質形態ばかりでなく、投与様式にも適さなければならない。投与様式による適当な担体の選択は、当業者によってルーチン的に行われる。
組成物の送達物質がポリヌクレオチド組成物とは別物、例えば、前記のトランスフェクトされた供与体または細菌である場合、組成物はまた、出典明示により本明細書の一部とされる米国特許第5,824,538; 5,643,771; 5,877, 159号に記載されるもののごとき他の添加物を含む。
ある好ましい具体例において、本発明の医薬組成物は、例えば、液剤、散剤、エアロゾル、錠剤、カプセル剤、腸溶性錠剤またはカプセル剤、または坐剤の形態で、哺乳類患者に投与されるために調製される。
本発明の教え、および本発明の組成物でトランスフェクトあるいは感染された細胞よりも多く増殖させられた本発明の方法および組成物の阻害効果の見かけの能力があるとすれば、前記の用量から低く適当に用量調整できる可能性がある。従って、前記用量範囲は単なるガイドラインである。一般に、医薬組成物は、かかる標的機能が疾病または疾病の原因のさらなる広がりに必要である、疾病、疾患または感染の治療または予防のために標的ポリヌクレオチド配列の機能を阻害あるいは減少するのに有効な量で投与される。使用される用量単位での医薬組成物の量は、本発明の組成物に対するインビトロでの細胞の応答および実験動物の応答に基づいて、経験的に決定される。最適用量は、各治療モダリティーおよび適応症のための標準法によって決定される。従って、用量、タイミング、投与経路、およびこれら組成物の再投与の必要性は、治療されるべき状態、その重症度、悪化する状態および、哺乳類患者の年齢および物理的状態などの因子、活性化合物の使用などを考慮に入れて当業者によって決定される。
本発明の方法は、前に詳細に記述した組成物および哺乳類細胞に部分的二本鎖のRNA分子を供する当該分野で最近使用されるあるいは他のポリヌクレオチド配列(例えば、機能的であってもなくてもタンパク質をコードするポリヌクレオチド分子またはリボザイムのごとき公知のRNA触媒配列)を使用できる。しかしながら、この方法の有効性は、タンパク質を産生しないRNA分子の使用によって促進される。これら方法は、哺乳類または哺乳類細胞において標的ポリヌクレオチド配列の機能を減少あるいは阻害する。前記の組成物、医薬組成物、投与の用量および様式は、細胞外または細胞内病原体のいずれかの異種病原体生物による感染を含む哺乳類を苦しめる種々の疾患の治療に特に望ましい。さらに、本発明の組成物は、病原体の哺乳類への感染を予防する上であるいは、潜在的病原体の再活性化により引き起こされる疾患の発生を予防する上で有用である。これら組成物はまた、病原体誘発癌の治療にも有用である。
本発明の組成物および先行技術のタンパク質をコードする可能性のある分子は、また本発明のもう1つの新規の方法で使用される。かかる組成物はまた、ある癌または遺伝疾患のごとき哺乳類のある非−病原性疾病または疾患の治療において有用である。本発明による治療または予防に特に敏感な症状の中には、その機能が疾患の開始または進行に必要である異常型の哺乳類ポリヌクレオチド配列の存在により特徴付けられる症状があるが、害を引き起こしたり、そうでなければ、哺乳類の健康に過度に悪影響を与えたりすることなしに阻害できる。換言すれば、この治療に適当な疾患の特徴は、哺乳類が遺伝子の機能なしで生き残るか、あるいは、遺伝子の機能が実質的に減少している場合に生き残っているということである。かかる場合には、異常型または病的なポリヌクレオチド配列の機能は、治療によって外来性に置換できる。別の場合には、疾病は、哺乳類がまた、ポリヌクレオチド配列または遺伝子の正常なコピーを有し、異常遺伝子および正常遺伝子間の差異がヌクレオチド配列の差異である哺乳類における異常ポリヌクレオチド配列または遺伝子の存在または機能によって引き起こされる。かかる場合、本発明の方法による異常ポリヌクレオチド配列の機能阻害は、外来性の治療なしで、正常ポリヌクレオチド配列を機能させるようである。
異常および正常ポリヌクレオチド配列の存在によって特徴付けられる哺乳類の癌は、慢性骨髄性白血病(CML)および異常配列が2つの遺伝子の融合体、すなわちbcr−ablである急性リンパ芽球性白血病(ALL)を含む。例えば、これら疾病の記載のため出典明示により本明細書の一部とされる、1994年6月23日発行の国際特許WO/13793におけるこれら癌の記載を参照のこと。CMLのごときかかる癌または疾病において、苦しんでいる患者はまた、ポリヌクレオチド配列または遺伝子の正常なコピーを有し、異常および正常配列または遺伝子間の差異はヌクレオチド配列の差異である。例えば、CMLでは、異常配列は、bcr−abl融合体であり、他方で、正常配列はbcrおよびablである。従って、前記方法では、融合体をスパンする配列である標的ポリヌクレオチド配列が用いられる。哺乳類のかかる癌の治療または予防方法は、標的ポリヌクレオチドが、遺伝子の正常コピーを有し、異常遺伝子および正常遺伝子間の差異がポリヌクレオチド配列の差異である哺乳類における異常な癌を引き起こす遺伝子のポリヌクレオチド配列である本発明の組成物を哺乳類へ投与することを含む。組成物は、哺乳類における異常配列の機能を減少あるいは阻害するのに有効な量で、医薬上許容される担体中に、細胞内ポリヌクレオチド取り込みを促進する所望の第二物質と共に投与される。
前記組成物およびこれら組成物を標的ポリヌクレオチド配列の機能を阻害あるいは減少するために使用する一般的方法は、また、種々の研究およびインビトロでの応用に応用できる。例えば、本発明の方法は、RNA分子ポリヌクレオチド配列が、選択される配列に実質的に相同であって、好ましくは、動物の他のポリヌクレオチド配列に実質的に非−相同である本発明の組成物を、組織培養細胞またはインビボで動物に投与することによって、細胞系または哺乳類実験動物の選択されるポリヌクレオチド配列の機能を決定する研究に応用できる。この標的配列の機能阻害は、動物の生物学および生理学に与える影響という研究を可能にする。
同様な方法で、選択される遺伝子の機能を永久に停止するのに十分な投与量に変化させることによって、「ノックアウト」された実験動物が調製できる。従って、前記の実験動物において「ノックアウト」されるのに選択される配列と実質的に相同なポリヌクレオチド配列を有するRNA分子を送達する本発明の方法は、製薬および遺伝的研究に有用な「ノックアウト」されるマウスおよび他の実験動物を開発するより簡単な技術を提供する。
本発明の組成物および方法を使用するさらに他の研究方法は、望ましいタンパク質の微生物製品のための研究試薬または産業製品株としての使用のための、真核および原核両者の微生物変異体の調製を含む。さらに他の使用は、本明細書中の教えがあるとすれば、当業者には明らかであると思われる。
実施例1: ウイルス感染細胞におけるHIVp24の機能の減少または阻害
HIV感染中、感染された***細胞の宿主染色体中に組み込まれる、ウイルスゲノムは、DNAテンプレートに逆転写される。組込みコピーは、それから、より多くのHIV粒子が作成される青写真である。、HIVの複製および/または発生病理に必須のポリヌクレオチド配列が減少あるいは阻害されるとすれば、本発明によって、ウイルス感染は治療できる。本実施例は、本発明の方法の具体例の1つの実施を示す。
プラスミド、HIVgpt(AIDS Research and Reference Reagent Program Catalog)を使用して、欠失HIVゲノムの組込みコピー、HIVgptを含む安定な組込みラブドミオザルコーマ(Rhabdomyosarcoma)(RD)またはCOS7細胞系を得た。HIVgptゲノムは、エンベロープ遺伝子の場所にミコフェノール酸(MPA)耐性遺伝子をコードし、それによってMPA耐性を授与する。プラスミドで細胞をトランスフェクトし、ついでMPA中で細胞選択することによって細胞系を作成した。MPA耐性細胞は、クローンとして増殖した。培養した、クローンとして増殖した細胞の培地を、ついで、p24ELISAアッセイキット(Coulter Corporation)を使用して、p24(細胞外で分泌されるHIVgagポリペプチド)の存在についてアッセイした。全細胞がp24陽性であった。
本発明の試薬を生じるために、HIV HXB2株のgag遺伝子の同一の座標に位置づけ、キャップ、ポリアデニル化配列および本来の開始コドンを欠失する600ヌクレオチド(nt)センスRNA、600ntアンチセンスRNAおよび600bp二本鎖RNA(dsRNA)が細胞をトランスフェクトするのに使用される。RNA分子が、1つの末端にバクテリオファージT7ポリメラーゼプロモーターを有するPCR産物の転写を介して生じる(図1Aおよび1Bを参照)。プライマーの座標はHIV(HXB2)、Genbank 受け入れ番号K03455の完全ゲノムの地図由来だった[また、L. Ratnerら、AIDS Res.Hum.Retroviruses, 3(1):57−69 (1987)を参照]。前方gagプライマーは座標901−924に位置し、この配列は、T7前方gagプライマーのT7プロモーターの後に続く。逆転gagプライマーは、座標1476−1500に位置し、T7逆転gagプライマーのT7プロモーターの後に続く。

6穴プレート上5−6x105細胞/ウェルを約80−90%集密に達するまで培養し、トランスフェクト剤として10μlのリポフェクトアミン(Gibco−BRL)を用いて2−3μgの選択されるRNA分子または対照分子でトランスフェクトする。トランスフェクトされた細胞を、1ないし17時間の範囲でインキュベートする。別の細胞培養液を1μgないし500μgの範囲の量のRNAでトランスフェクトし、公知のトランスフェクト剤なしで送達し、0.5分から約2日間細胞上でインキュベートする。例えば、ある群の細胞は、センスgagRNAで、別の細胞は、アンチセンスgagRNAで、別の細胞はdsgagRNAで、別の細胞はセンスgDRNA対照で、別の細胞はアンチセンスgDRNA対照で、および別の細胞はdsgDRNA対照でトランスフェクトされる。また、さらなる陰性対照は、RNA分子を受け取らない細胞である。
本発明によれば、gD RNA分子はどれもp24合成を阻止あるいは阻害する能力を示さない。しかしながら、本発明によれば、dsgagRNA分子は、p24合成を阻害あるいはダウンレギュレートする。
これらRNAが、ある程度の二本鎖を形成できなければ、センスおよびアンチセンスRNA分子は、p24合成において、たとえあるとしても緩和な阻害効果のみを引き起こすと思われる。
ダウンレギュレートされたシグナルがダウンレギュレートされていない細胞に伝達されることを測定するために、本実施例は、減少/阻害効果(すなわち、p24合成の阻害または減少)が物質によってトランスフェクトされていない培養系の細胞に伝達されることを示す。
P24合成減少を示す実施例1の培養液の細胞は、予めいずれのRNA分子ともインキュベートされておらず、実際野生型レベルでp24を合成している対照細胞と一緒に培養する。本発明によれば、予めトランスフェクトされた細胞は、トランスフェクトされてない細胞に標的ポリヌクレオチド機能阻害を転移でき、共培養系の対照細胞はp24合成減少によって特徴付けられる。
第二のプロトコルでは、減少したp24産生を示す、実施例1のトランスフェクトされたRD細胞を組込みヒグロマイシン耐性遺伝子を含むように作成して、6穴プレート上総細胞数6−7x105まで1/1000ないし1/10(RD/対照RD)の範囲の割合という異なる細胞割合を用いて正常レベルのp24を発現する、トランスフェクトされていないRD細胞と共培養する。ヒグロマイシン耐性RD細胞は以下のごとく作成する:RD細胞(5−6x105細胞) を6穴プレート上80−90%コンフルエンスまで培養し、チミジンキナーゼ(TK)プロモーターの制御下、ヒグロマイシン耐性遺伝子を含むpCEP4(Invitrogen Corp.)のNru 1−Sal 1フラグメント、2.5μgでトランスフェクトする。トランスフェクションは、トランスフェクト剤、リポフェクトアミン(Gibco BRL)を用いて行われる。トランスフェクション2日後、400μg/mlヒグロマイシン存在下、細胞をインキュベートする。耐性細胞がクローン的に広がる。1つ以上のクローン的に広がった細胞系を対照として実験に用いる。
本発明によって、p24産生阻害は少なくとも対照細胞のサブセットで示される。
A.本発明の組成物としてのRNA分子の設計
この実験の全RNA分子は長さ600ntsに近く、全RNA分子は、IL−12のp40鎖を産生できないように設計する。分子はキャップおよびポリ−A配列を有さず;本来の開始コドンは存在せず、RNAは全長産物をコードしない。以下のRNA分子を設計する:
(1)IL−12p40ネズミ伝令RNA(mRNA)に相同な一本鎖(ss)センスRNAポリヌクレオチド配列;
(2)IL−12p40ネズミmRNAに相補的なssアンチセンスRNAポリヌクレオチド配列;
(3)センスおよびアンチセンス両者のp40IL−12ネズミmRNAポリヌクレオチド配列を含む二本鎖(ds)RNA;
(4)IL−12p40ネズミ異種RNA(hnRNA)に相同なssセンスRNAポリヌクレオチド配列;
(5)IL−12 p40ネズミhnRNAに相補的なssアンチセンスRNAポリヌクレオチド配列;
(6)センスおよびアンチセンス両者のIL−12 p40ネズミhnRNAポリヌクレオチド配列を含むdsRNA分子;
(7)IL−12p40プロモーターの上流鎖に相同なssネズミRNAポリヌクレオチド配列;
(8)IL−12p40プロモーターの下流鎖に相同なssネズミRNAポリヌクレオチド配列および
(9)IL−12p40プロモーターの上流および下流鎖に相同なネズミRNAポリヌクレオチド配列を含むdsRNA分子。
実施例1に記載のごとく、片側末端にT7プロモーターを有するPCR産物のT7RNAポリメラーゼ転写を介して前記(1)−(9)の種々のRNA分子が生じる。センスRNAが所望の場合は、T7プロモーターは前方PCRプライマーの5'末端に位置する。アンチセンスRNAが所望の場合は、T7プロモーターは逆転PCRプライマーの5'末端に位置する。dsRNAが所望の場合は、両タイプのPCR産物をT7転写反応中に含む。あるいは、転写後、センスおよびアンチセンスRNAを混合する。

前記のネズミIL−12 p40鎖特異的RNAまたは前記同定の対照を10μgおよび500μgの範囲の用量で、Balb/cネズミ(ネズミ5匹/群)に、筋肉内あるいは腹腔内注射する。3週間という期間4日ごとにネズミから血清を集め、Quantikine M−IL−12 p40 ELISA アッセイ(Genzyme)を用いてIL−12p40鎖のレベルにつきアッセイする。
本発明によれば、IL−12mRNA、IL−12hnRNA両者由来のdsRNA分子およびIL−12プロモーター由来のdsRNAを受けるネズミはIL−12産生の減少または阻害を示す。RNA分子があるレベルの二本鎖を形成する能力を有さなければ、一本鎖IL−12由来RNA分子を受けるネズミ血清中には、たとえあるとしても緩和な阻害効果が観察される。特定の方法では、HSV gD由来RNAはインビボでIL−12を減少あるいは阻害しないと期待される。
A.インビトロアッセイ
20−30%コンフルエンス密度で接種されたベロおよび/またはBHK細胞を、10%FBSを含むDMEM中、37℃にて6穴プレート上で培養する。細胞が80−90%集密に達したら、トランスフェクト剤としてリポフェクトアミン(Gibco−BRL)を用いて実施例1に記載の2−3μgのHIVgag−およびHSVgD−特異的RNA分子でトランスフェクトする。RNA分子はまた、5および100μg間で変化する量のいずれの公知のトランスフェクト剤の非存在下送達される。別の群の細胞はRNAを受けない。
トランスフェクトされた細胞を、10%FBSを含むDMEM中、37℃にて培養する。トランスフェクション後1、2、4および7日に、DMEM250μlの接種原中0.1の感染多重度(MOI)にてHSV2で細胞を感染する。接種原は、ウェル毎DMEM(10%FBS)2mlを添加した1時間後吸着される。トランスフェクション後4および7日に感染したこれら細胞につき、細胞が感染時間で全面成長しているので、新しい6穴プレートに移動する。細胞が移動されない場合、過密になる。
HSVgDタンパク質を作る能力および対照としてHIVgag特異的RNA分子を有さない実施例1記載のHSV−gD特異的RNA分子を用いて、注入された本発明のHSV2gD特異的RNA分子の使用を介して、ネズミをHSV攻撃からの保護につき評価する。
Balb/cマウス(マウス5匹/群)を、10および500μgRNAの範囲の用量で記載RNA分子で筋肉内または腹腔内に免疫処理する。RNA注入後1、2、4および7日にマウスを経膣接種によってHSV−2(30μl中105pfu)で攻撃する。HSV−2接種後毎日、マウスを感染の徴候につき観察し、0−4の等級で等級付けする。ゼロは感染の徴候なし;1は赤みを示す;2は小胞および赤みを示す;3は小胞、赤みおよび失禁を示す;4は麻痺を示す。
Claims (67)
- 少なくとも1つの標的遺伝子の2つまたはそれ以上の配列と実質的に相同かつ相補的である、2つまたはそれ以上の異なる二本鎖RNA配列を含む、複数標的の部分的に二本鎖のRNA分子。
- 2つまたはそれ以上の異なる二本鎖RNA配列が複数の標的遺伝子の2つまたはそれ以上の配列と実質的に相同または相補的である、ところの請求項1記載の複数標的の部分的に二本鎖のRNA分子。
- 複数標的の部分的に二本鎖のRNA分子の少なくとも11ないし30個のヌクレオチドが異なる二本鎖配列の各々に含まれる、ところの請求項1記載の複数標的の部分的に二本鎖のRNA分子。
- 異なる二本鎖RNA配列の各々が標的配列の30個のヌクレオチド領域と類似する少なくとも50%の相同性を有する連続する30個のヌクレオチドの少なくとも1つのセグメントを含み、その連続する30個のヌクレオチドのセグメントも天然に存する必須のポリヌクレオチド配列と非相同である、ところの請求項1記載の複数標的の部分的に二本鎖のRNA分子。
- 複数標的の部分的に二本鎖のRNA分子が長さが約100個と10000個の間のポリヌクレオチドである、ところの請求項1記載の複数標的の部分的に二本鎖のRNA分子。
- 複数標的の部分的に二本鎖のRNA分子が長さが少なくとも約200個のヌクレオチドである、ところの請求項1記載の複数標的の部分的に二本鎖のRNA分子。
- 異なる二本鎖RNA配列の1つまたは複数が、非塩基対ポリヌクレオチド配列により分離されている、センスポリヌクレオチドおよびアンチセンスポリヌクレオチドを含む、ところの請求項1記載の複数標的の部分的に二本鎖のRNA分子。
- センスおよびアンチセンスポリヌクレオチドがヘアピンを形成する、ところの請求項7記載の複数標的の部分的に二本鎖のRNA分子。
- 2つまたはそれ以上の異なる二本鎖RNA配列が開裂配列により分離されている、ところの請求項1記載の複数標的の部分的に二本鎖のRNA分子。
- 開裂配列が自己触媒配列またはスプライス部位である、ところの請求項9記載の複数標的の部分的に二本鎖のRNA分子。
- 少なくとも1つの標的遺伝子が単一の標的病原体から由来する、ところの請求項1記載の複数標的の部分的に二本鎖のRNA分子。
- 複数の標的遺伝子が複数の標的病原体から由来する、ところの請求項2記載の複数標的の部分的に二本鎖のRNA分子。
- 標的病原体がウイルスである、ところの請求項11記載の複数標的の部分的に二本鎖のRNA分子。
- ウイルスがHBV、HIV、HSV、CMV、HPV、HTLVおよびEBVからなる群より選択される、ところの請求項13記載の複数標的の部分的に二本鎖のRNA分子。
- 複数の標的病原体がウイルスである、ところの請求項12記載の複数標的の部分的に二本鎖のRNA分子。
- 複数のウイルスがHBV、HIV、HSV、CMV、HPV、HTLVおよびEBVからなる群より選択される、ところの請求項15記載の複数標的の部分的に二本鎖のRNA分子。
- 少なくとも1つの標的遺伝子が哺乳動物における疾患または障害と関連付けられる、ところの請求項1記載の複数標的の部分的に二本鎖のRNA分子。
- 少なくとも1つの標的遺伝子が癌関連遺伝子である、ところの請求項17記載の複数標的の部分的に二本鎖のRNA分子。
- 複数の標的遺伝子が哺乳動物における疾患または障害と関連付けられる、ところの請求項2記載の複数標的の部分的に二本鎖のRNA分子。
- 複数の標的遺伝子が癌関連遺伝子である、ところの請求項19記載の複数標的の部分的に二本鎖のRNA分子。
- 少なくとも1つの標的遺伝子の2つまたはそれ以上の配列が、転写配列、非転写配列、コード化配列、非コード化配列、エクソン含有配列、制御配列およびプロモーター配列からなる群より選択される、ところの請求項1記載の複数標的の部分的に二本鎖のRNA分子。
- 複数標的の部分的に二本鎖のRNA分子がポリアデニル化信号を欠く、ところの請求項1記載の複数標的の部分的に二本鎖のRNA分子。
- 請求項1に記載の複数標的の部分的に二本鎖のRNA分子を含む、組成物。
- 細胞によるポリヌクレオチド吸収を容易にする物質をさらに含む、請求項23記載の組成物。
- 請求項1に記載の複数標的の部分的に二本鎖のRNA分子をコード化するDNA分子。
- 請求項1に記載の複数標的の部分的に二本鎖のRNA分子をコード化する発現ベクター。
- 複数標的の部分的に二本鎖のRNA分子が、ミトコンドリア・プロモーター、RNApolIプロモーター、RNApolIIプロモーター、RNApolIIIプロモーター、ウイルス性プロモーター、細菌性プロモーターおよびバクテリオファージ・プロモーターからなる群より選択されるプロモーターを用いて発現される、ところの請求項26記載の発現ベクター。
- 複数標的の部分的に二本鎖のRNA分子がRNApolIIIプロモーターを用いて発現される、ところの請求項27記載の発現ベクター。
- ベクターがプラスミド、ファージまたは組換えウイルスである、ところの請求項26記載の発現ベクター。
- コード化された複数標的の部分的に二本鎖のRNA分子がポリアデニル化信号を欠く、ところの請求項26記載の発現ベクター。
- 細胞にて少なくとも1つの標的遺伝子の機能を軽減または阻害するための発現ベクターであって、少なくとも1つの標的遺伝子の2つまたはそれ以上の配列と相同かつ相補的である、2つまたはそれ以上の異なる二本鎖RNA配列をコードする、発現ベクター。
- 2つまたはそれ以上の異なる二本鎖RNA配列が複数の標的遺伝子の2つまたはそれ以上の配列と相同または相補的である、ところの請求項31記載の発現ベクター。
- 異なる二本鎖RNA配列の各々が二本鎖配列に含まれる少なくとも11ないし30個のヌクレオチドを含む、ところの請求項31記載の発現ベクター。
- 異なる二本鎖RNA配列の各々が標的配列の30個のヌクレオチド領域と類似する少なくとも50%の相同性を有する連続する30個のヌクレオチドの少なくとも1つのセグメントを含み、その連続する30個のヌクレオチドのセグメントも該細胞にて天然に存する必須のポリヌクレオチド配列と非相同である、ところの請求項31記載の発現ベクター。
- 2つまたはそれ以上の異なる二本鎖RNA配列を含む複数標的の部分的に二本鎖のRNA分子をコード化する、請求項31記載の発現ベクター。
- 複数標的の部分的に二本鎖のRNA分子が長さが約100個と10000個の間のポリヌクレオチドである、ところの請求項35記載の発現ベクター。
- 複数標的の部分的に二本鎖のRNA分子が長さが少なくとも約200個のヌクレオチドである、ところの請求項35記載の発現ベクター。
- 異なる二本鎖RNA配列の1つまたは複数が、非塩基対ポリヌクレオチド配列により分離されている、センスポリヌクレオチドおよびアンチセンスポリヌクレオチドを含む、ところの請求項31記載の発現ベクター。
- センスおよびアンチセンスポリヌクレオチドがヘアピンを形成する、ところの請求項38記載の発現ベクター。
- 2つまたはそれ以上の異なる二本鎖RNA配列が開裂配列により分離されている、ところの請求項35記載の発現ベクター。
- 開裂配列が自己触媒配列またはスプライス部位である、ところの請求項40記載の発現ベクター。
- 2つまたはそれ以上の異なる二本鎖RNA配列が、ミトコンドリア・プロモーター、RNApolIプロモーター、RNApolIIプロモーター、RNApolIIIプロモーター、ウイルス性プロモーター、細菌性プロモーターおよびバクテリオファージ・プロモーターからなる群より選択されるプロモーターを用いて発現される、ところの請求項31記載の発現ベクター。
- 1つまたは複数のプロモーターがRNApolIIIプロモーターである、ところの請求項42記載の発現ベクター。
- 2つまたはそれ以上の異なる二本鎖RNA配列が2つまたはそれ以上のプロモーターを用いて発現される、ところの請求項31記載の発現ベクター。
- 2つまたはそれ以上の異なる二本鎖RNA配列が2つまたはそれ以上のプロモーターを用いて発現される、ところの請求項38記載の発現ベクター。
- 2つまたはそれ以上のプロモーターが、ミトコンドリア・プロモーター、RNApolIプロモーター、RNApolIIプロモーター、RNApolIIIプロモーター、ウイルス性プロモーター、細菌性プロモーターおよびバクテリオファージ・プロモーターからなる群より選択される、ところの請求項44記載の発現ベクター。
- 2つまたはそれ以上のプロモーターがRNApolIIIプロモーターである、ところの請求項46記載の発現ベクター。
- 2つまたはそれ以上のプロモーターが、ミトコンドリア・プロモーター、RNApolIプロモーター、RNApolIIプロモーター、RNApolIIIプロモーター、ウイルス性プロモーター、細菌性プロモーターおよびバクテリオファージ・プロモーターからなる群より選択される、ところの請求項45記載の発現ベクター。
- 2つまたはそれ以上のプロモーターがRNApolIIIプロモーターである、ところの請求項48記載の発現ベクター。
- ベクターがプラスミド、ファージまたは組換えウイルスである、ところの請求項31記載の発現ベクター。
- 少なくとも1つの標的遺伝子が単一の標的病原体から由来する、ところの請求項31記載の発現ベクター。
- 複数の標的遺伝子が複数の標的病原体から由来する、ところの請求項32記載の発現ベクター。
- 標的病原体がウイルスである、ところの請求項50記載の発現ベクター。
- ウイルスがHBV、HIV、HSV、CMV、HPV、HTLVおよびEBVからなる群より選択される、ところの請求項53記載の発現ベクター。
- 複数の標的病原体がウイルスである、ところの請求項52記載の発現ベクター。
- 複数のウイルスがHBV、HIV、HSV、CMV、HPV、HTLVおよびEBVからなる群より選択される、ところの請求項55記載の発現ベクター。
- 少なくとも1つの標的遺伝子が哺乳動物における疾患または障害と関連付けられる、ところの請求項31記載の発現ベクター。
- 少なくとも1つの標的遺伝子が癌関連遺伝子である、ところの請求項57記載の発現ベクター。
- 複数の標的遺伝子が哺乳動物における疾患または障害と関連付けられる、ところの請求項32記載の発現ベクター。
- 複数の標的遺伝子が癌関連遺伝子である、ところの請求項59記載の発現ベクター。
- 少なくとも1つの標的遺伝子の2つまたはそれ以上の配列が、転写配列、非転写配列、コード化配列、非コード化配列、エクソン含有配列、制御配列およびプロモーター配列からなる群より選択される、ところの請求項31記載の発現ベクター。
- 2つまたはそれ以上の異なる二本鎖RNA配列がポリアデニル化信号を欠く、ところの請求項31記載の発現ベクター 2つまたはそれ以上のプロモーターがRNApolIIIプロモーターである、ところの請求項46記載の発現ベクター。
