Description
本発明は、C型肝炎の治療および診断におけるCD81タンパク質およびこのタンパク質をコードする核酸の使用、ならびにそのような使用のための薬学的組成物、動物モデルおよび診断キットに関する。
総ての刊行物、説明書、特許、および本明細書に引用された特許出願は、全体が参考として援用される。HCV(以前は非A 非B肝炎−NANBVとして公知であった)は、約3000アミノ酸のポリタンパク質をコードする単一のオープンリーディングフレームを有する約10000のヌクレチドの+鎖RNAウイルスである。ウイルスの構造が、組換えDNA技術(ヨーロッパ特許出願EP−A−0318216およびヨーロッパ特許出願EP−A−0388232)によって解明されたにもかかわらず、ウイルスそれ自体は、単離されず、そしてポリタンパク質のタンパク質分解によって産生された様々なウイルスタンパク質は、類似のゲノム組織の他の類似のウイルスとの類似性によって推論されただけである(非特許文献1)。
本発明に従って、C型肝炎(HCV)の治療または診断に使用するためのCD81タンパク質、またはその機能的な等価物が提供される。本発明のさらなる局面に従って、HCVの治療または診断に使用するためのCD81タンパク質に特異的に結合する化合物が提供される。
・本発明はさらに、以下を提供し得る:
・(項目1) HCVの治療または診断に使用するためのCD81タンパク質、またはその機能的な等価物。
・(項目2) HCVの治療もしくは診断に使用するためのSWISSPROTデータベース(受託番号P18582)、もしくはEMBL/GENBANKデータベース(受託番号M33690)に記載されるヒトCD81配列、またはその機能的な等価物を含む、タンパク質。
・(項目3) SWISSPROTデータベース(受託番号P18582)、もしくはEMBL/GENBANKデータベース(受託番号M33690)に記載されるヒトCD81配列と少なくとも80%の相同性を有するアミノ酸配列を含むタンパク質であって、相同性がPileup配列解析ソフトウエアパッケージ(Wisconsin,1996)を用いて定義され、HCVの治療または診断に使用するための、タンパク質。
・(項目4) SWISSPROTデータベース(受託番号P18582)、もしくはEMBL/GENBANKデータベース(受託番号M33690)に記載されるヒトCD81配列のアミノ酸113〜201、またはその機能的な等価物からなる、タンパク質。
・(項目5) HCVの治療または診断に使用するための項目4に記載のタンパク質。
・(項目6) CD81タンパク質に特異的に結合する化合物であって、HCVの治療または診断に使用するための化合物。
・(項目7) HCVの感染を処置するための方法であって、治療的に効果的な量のCD81タンパク質もしくはその機能的な等価物を患者に投与する工程、またはウイルスの感染力を減少するために当該CD81タンパク質に特異的に結合する化合物を投与する工程を包含する、方法。
・(項目8) CD81タンパク質、またはその機能的な等価物、またはCD81タンパク質に特異的に結合する化合物を含む薬学的組成物であって、必要に応じて薬学的に受容可能な塩として薬学的に受容可能なキャリアーと組み合わせる、薬学的組成物。
・(項目9) 薬学的に受容可能なキャリアーと組み合わせる項目4に記載のタンパク質を含む、薬学的組成物。
・(項目10) HCVの治療または診断に使用するための、項目8または9のいずれかに記載の、薬学的組成物。
・(項目11) 項目8または9において定義された薬学的組成物を調製するためのプロセスであって、CD81タンパク質、またはその機能的な等価物、または項目4に記載のタンパク質、またはCD81タンパク質に特異的に結合する化合物が、薬学的に受容可能なキャリアーとの会合を起こさせる、プロセス。

(実施例1:本研究において使用される、組換えE2、細胞株、ベクターDNA、および抗体)
このスクリーニングにおいて用いた組換えE2を、CHO細胞において生成した(E2−CHO)(WO97/09349)。E2−CHOは、ヒトT細胞リンパ腫細胞株Molt−4に結合する。Molt−4亜株(A2A6と名づける)を、個々のMolt−4細胞コロニーを拡大すること、および細胞表面に結合したE2−CHOの量を試験することにより同定した。A2A6亜株は、その親株より多くのE2−CHO分子にその表面で結合することが見出され、従って、RNAの供給源として選択された。このことは、この亜株が、E2結合分子をコードする転写産物がより多量に示されていることを期待している。これらの細胞を、アッセイを用いて選択し、このアッセイによって、ヒトBリンパ腫細胞株およびTリンパ腫細胞株ならびに悪性肝細胞ガン細胞株を、D.Rosaら,Proc.Natl.Acad.Sci.USA 93,1759(1996)に記載のように哺乳動物細胞(CHO)において発現された組換えE2とともにインキュベートし、そしてRosaら(1996)に記載のようにビオチン標識抗E2抗体を用いて染色した。最大のE2結合能を有する細胞を、FacsVantage(Becton Dickinson)を用いて選別し、そして限界希釈によってサブクローニングした。増殖するクローンを、FacsにおいてE2結合についてスクリーニングし、そして最大の平均蛍光強度を有するクローンをさらに拡大させた。
WOPは、ポリオーマT抗原を発現するNIH3T3由来細胞である(L.DaileyおよびC.Basilico,J.Virol.54,739(1985))。この細胞株では、ポリオーマの複製起点を含有するプラスミドがエピソーム的に増幅し得る。pCDM8(Invitrogen)を用いて構築した組換えDNAを、選択したトランスフェクタントから回収し得る。このトランスフェクタントは、ポリオーマの複製起点を含み、そして真核生物細胞における発現ライブラリーの操作のために設計される。
総RNAを、A2A6細胞株から、ChomczinskyおよびSacchi(Chomczinsky,P.およびSacchi,N.(1987)Anal.Biochem.162:156−159)によって記載される方法に従って抽出した。ポリ(A)+を、オリゴ(dT)セルロースを用いて2回富化した。テンプレートとしての2μgのこのRNAから出発して、二本鎖の相補的なDNAを、Superscript II cDNA合成キット(Life Technologies)をオリゴ(dT)(100ng)およびランダムヘキサマープライマー(100ng)の存在下で用いて合成した。T4 DNAポリメラーゼを用いてこのcDNAを平滑末端化し、そしてBstXIリンカーと連結した。BstXIリンカーは、発現ベクターpCDM8のポリリンカー領域における同じ制限部位へのこのフラグメントの挿入を可能にする。リンカーに連結したcDNAをフェノール抽出し、そして硫酸アンモニウムを用いてエタノール沈殿して遊離のモノヌクレオチドを除去し、続いてSephacryl 500クロマトグラフィー(Lifetechnologies)を行ってcDNAをサイズ分画した。500bpを超える精製cDNAフラグメントをプールし、そしてBstXI消化pCDM8と分子比約1:1で連結した。この最終連結反応物を、E.coli MC1061/P3の形質転換から、Gene−Pulser(BIORAD)を用いるエレクトロポレーションにより用いた。計2×106cfuが増幅され、そしてcDNAライブラリーとして液体細菌培養物中にプールされた。
スクリーニング手順は、大部分はCampbellら(Campbell,I.G.,Jones,T.A.,Foulkes,W.D.およびTrowsdale,J.Cancer Res.51:5329−5338,1991)により記載された方法に基づいていた。富化を、磁性ビーズ(1回目〜3回目)(図1A)およびパンニング技術(4回目)(図1B)を用いて実施した。
計375μgの増幅されたDNA(これは、独立した2×106個のcDNAクローンを示す)を調製した。各々のトランスフェクションでは、25μgのDNAを、107個のWOP細胞と、300V/500μFの条件下でGene−Pulserエレクトロポレーター(BIORAD)を用いて混合した。15セットのトランスフェクションを行った。トランスフェクション後、細胞を、37℃にて2日間インキュベートし、次いでトリプシン処理により細胞を脱離させ、そして360×gにて10分間、4℃の遠心分離により5% FCSおよび0.5mM EDTAを補充したPBSで2回洗浄した。細胞ペレットを、5%FCSおよび0.5mM EDTAを補充したPBS中に再懸濁し(107細胞/ml)、次いでE2−CHOを、10μg/mlの濃度で細胞懸濁物に添加した。細胞を、氷上で60分間インキュベートした。5%FCSおよび0.5mM EDTAを補充したPBSで2回洗浄した後、細胞懸濁物を、291A2抗体とともに氷上で30分間インキュベートした。5%FCSおよび0.5mM EDTAを補充したPBSで2回洗浄した後、10μlのヤギ抗マウスIGカップリングDynabeads(DYNAL)を細胞懸濁物に添加した。混合物を、Coulter Mixer(Coulter)を60分間、4℃にて用いて穏やかに攪拌した。結合した細胞を、Magnetic Particle Concentrator(DYNAL)を製造業者の使用説明書に従って用いて、非バインダーから分離し、このようにしてE2結合トランスフェクタントを富化した。プラスミドDNAを、結合したトランスフェクト細胞から、Campbellら(Campbell,I.G.,Jones,T.A.,Foulkes,W.D.およびTrowsdale,J.Cancer Res.51:5329−5338,1991)により記載されたプロトコルを用いて回収した。E.coli MC1061/P3を、エレクトロポレーションによりこのプラスミドを用いて形質転換した。このDNAプールを、最初の富化プール(1°EP)と呼ぶ。
1°EP由来の計150μgの増幅したDNAを調製し、そして6セットのトランスフェクションを行い、そしてトランスフェクタントを、1回目のスクリーニングと同じ条件を用いて富化した。このDNAプールを2°EPと呼ぶ。
2°EP由来の計25μgの増幅したDNAを調製し、そして1セットのトランスフェクションを行った。トランスフェクタントを、1回目のスクリーニングと同じ条件を用いて富化した。この分離工程の間にトランスフェクタントは凝集体を形成した。この凝集は、無関係な接着分子の発現により生じ得る。これは、富化の効率を減少させ得る。なぜなら、これらの凝集体は、非特異的に磁性ビーズを含んでいたからである。この潜在的な問題を回避するために、Magnetic Particle Concentratorによる2回目の分離後のトランスフェクタントを希釈し、そしてTerasakiプレートにプレーティングした。顕微鏡下で同定された約100個の単一細胞をプールし、そしてプラスミドDNAをそれらから抽出した。この工程から調製したDNAプールを、3°EPと呼ぶ。
291A1モノクローナル抗体を、ペトリ皿(90mm)中で10μg/mlの濃度で4℃にて一晩インキュベートした。
DNAを、4°EPに由来する単一コロニーから単離した。単一トランスフェクションを、各々のプラスミド調製物について、先のスクリーニング工程について用いたのと同じ条件を用いて行った。形質転換体のE2結合を、抗体結合体化Dynabeadsの代わりにモノクローナルヤギ抗マウスIgのフィコエリトリン結合体化Fabフラグメントを用いて検出した。3°EPおよび4°EPのトランスフェクタントをまた、同様にして分析した。E2に結合した細胞を、FACScan(Becton Dickinson)で検出し、そしてLYSIS IIプログラム(Becton Dickinson)を用いて分析した(図2)。E2−CHOは、精製工程が進むにつれてますます結合する。単一クローンP3は、強力なE2結合を示した。
P3は、約1kbのインサートを含む。トランスフェクションの際にWOPへのE2結合を付与するcDNAクローンのインサートのDNA配列を、T7プライマーを使用して、自動化配列決定システムによって決定した。T7プライマー配列は、pCDM8のクローニング部位に隣接して位置する。この配列は、UNIXコンピューター上でGCGプログラムを使用して、GenBankデータベースを介してスクリーニングした。この分析によって、P3インサートの5’部分がヒトCD81(TAPA−1)と同一であることが明らかになった。3つの酵素(BstXI、HincIIおよびNcoI)を使用する、P3の制限分析もまた、ヒトCD81 cDNAの制限マップと一致した。
抗CD81抗体を使用して、E2とCD81との間の相互作用を評価した。EBV B細胞を、漸増濃度の組換えE2とともに4℃で1時間インキュベートし、次いで、抗CD81モノクローナル抗体(クローンJS−81,Pharmingen)を用いて染色した。図3に示されるように、組換えE2は、EBV形質転換B細胞(EBV−B細胞)への抗CD81抗体の結合を競合的に阻害することが見出された。このデータは、平均蛍光強度の阻害%として表される(Rosaら、1996)。
E2タンパク質に結合するCD81ドメインをマップするために、本発明者らの取り組みは、このタンパク質のEC2親水性細胞外ループに焦点を当てた。このフラグメントを、チオレドキシン−EC2融合タンパク質(これは、チオレドキシンとEC2との間にエンテロキナーゼ部位を有する)およびGST−EC2融合タンパク質(これは、GSTとEC2との間にトロンビン部位を有する)としてE.coliにおいて発現させ、そして6ヒスチジンタグをこのタンパク質のカルボキシ末端に付加した。本発明者らは、両方のタンパク質が発現されかつHCV E2に結合し得ることを示す。競合実験において、本発明者らはまた、精製された融合タンパク質およびGST−トロンビン−EC2−(His)6から切り出されたEC2−HisフラグメントがCD81発現細胞の表面上のE2の結合を阻害し得ることを示す。
図5は、pThio−His CにクローニングされたEC2フラグメントのヌクレオチドおよび推定アミノ酸配列ならびにチオレドキシンのカルボキシ末端およびエンテロキナーゼ切断部位をコードする上流プラスミド配列を示す。示されるように、EC2を、エンテロキナーゼ部位によってチオレドキシンとインフレームで融合する。このエンテロキナーゼ部位は、融合タンパク質からチオレドキシン部位を除去するために使用され得る。
標準的なクローニング技術(Sambrookら、1989)を使用して、PCR産物をXhoIおよびHindIIIで二重消化し、同じ制限酵素で消化したpThio−His C(Invitrogen)に連結し、そしてTop10 E.coli細胞に形質転換した。形質転換体を制限酵素分析およびプラスミドのDNA決定によって選択した後、予測されるチオレドキシンエンテロキナーゼ部位−EC2融合タンパク質をコードする正しい構築物を同定した。選択されたクローンのグリセロールバッチを−80℃で保存した。
チオレドキシン−EC2の精製について、以下の手順を開発した:
1)細胞の浸透圧ショック、2)30%飽和硫酸アンモニウムでのタンパク質沈降、および3)IMAC。浸透圧ショックののち、融合タンパク質の約50%を細胞から夾雑タンパク質とともに放出させた。硫酸アンモニウム沈澱は、大量の夾雑タンパク質を欠失したチオレドキシン−EC2を含んだペレットを生じた。再懸濁した沈澱物のIMACは、融合タンパク質を生じ、これは、SDS−PAGEによって評価されたように約85%の純度であった。この手順を用いて、本発明者らは、1リットルの培養物から5mgのチオレドキシン−EC2を精製した。この手順は、以下で詳細に記載される。
図8は、pGEX−KGにおいてクローニングされたEC2−(His)6フラグメントのヌクレオチドおよび推定アミノ酸配列、ならびにGSTのカルボキシ末端、トロンビン切断部位、および小さなグリシンスペーサーをコードする上流プラスミド配列を示す。示されるように、EC2を、トロンビン部位を介してGSTとインフレームで融合する。このトロンビン部位は、融合タンパク質からGSTを除去するために使用され得る。トロンビン部位とEC2との間に位置するグリシンリッチスペーサーは、融合タンパク質のトロンビンによる切断を容易にする(Guan,K.L.およびDixon,J.E.(1991)Anal.Biochem.192,262−267)。
PCR産物をXhoIおよびHindIIIで消化し、同じ制限酵素で消化したpGEX−KG(Guan,K.L.,およびDixon,J.E.(1991)Anal.Biochem.192,262−267)に連結し、そしてTOP10 E.coli細胞に形質転換した。形質転換体を制限酵素分析およびプラスミドのヌクレオチド配列決定によって選択した後、予測されるサイズのインサートを有するプラスミドが、上流トロンビンおよびGSTコード配列とインフレームで正しいEC2−(His)6配列を有することを見出した。次いで、選択されたTOP10クローンから調製されたプラスミドを、BL21細胞に形質転換した。選択されたクローンのグリセロールバッチを−80℃で保存した。
GST−EC2−(His)6融合タンパク質をグルタチオンセファロースカラムで精製し、そしてトロンビンで消化した(図10)。消化後、EC2−(His)6を部分を、GSTフラグメントを除去するためのグルタチオンセファロースカラムおよびIMACクロマトグラフィーから構成されているさらなる2つのクロマトグラフィー工程によってさらに精製した。この手順を以下で詳細に示す。
グルタチオンセファロースカラムから回収された9.6mgのタンパク質を、22ユニットのトロンビン(Pharmacia)で8時間室温で消化し、次いで、この酵素を0.13mM PMSF(Sigma)を使用して不活化させた。次いで、反応混合物をPBSに対して透析し、そしてPBSで平衡化した0.5mlのGST−セファロースカラムにロードした。カラムを1mlのPBSで洗浄した。流出および洗浄をプールし、そして0.250mlのニッケル活性化したキレート化セファロースカラムにロードした。EC2−(His)6を、1mlの20mM リン酸緩衝液、500mM NaCl、400mM イミダゾール、pH 7.8で溶出してカラムから回収した。次いで、透析をPBSに対して行った。
ヒト(マウスではない)のCD81のEC2ループを含むタンパク質をウェスタンブロットにおいてE2と結合させ(データは示さず)、そしてこれは、E2がヒト細胞へ結合することを阻害した(図11)。キメラタンパク質をポリスチレンビーズにコーティングして、既知量のウイルスRNA分子を含む感染血漿とともにインキュベートした。洗浄後、ビーズに結合したウイルスを、結合したHCV RNAの量について定量的RT−PCRによって評価した。この実験を以下で記載のように行った。
以下の構築物を設計し、そしてヒトCD81についてのマウストランスジェニックを生成するために作製した。
pSR1PインサートのSalI−BamHIフラグメントを、pCAGGS(条件つきでJ.Miyazaki博士(Osaka University,Japan)から贈与された)の改変されたプラスミドであるpCAGmcsの適合性の制限部位に挿入した。pCAGmcsは、ニワトリβアクチンプロモーターおよびヒトサイトメガロウイルスエンハンサーを含む(Niwa,H.ら、Gene 108,p193(1991))(図12におけるpCAGSR1Pp)。3.8kbのEcoRI−BamHIフラグメントを接合子注入に供した。
pSR1PのSalI部位を、E.coli DNAポリメラーゼIのクレノウフラグメントを用いた平滑末端形成の後に、BamHIリンカー連結によってBamHI部位に変更した。このBamHIフラグメントを、ALB e/p プラスミド(マウスアルブミンプロモーターおよびエンハンサーを有する(Pinkert,C.A.ら、Genes Dev.1,p268(1987)))(F.Chisari(Scripps Research Institute,La Jolla,San Diego)から受け取った)のBamHI部位に挿入した(図13のpAIbSRIP)。4.5kbのNotI−EcoRVフラグメントを、接合子注入に供した。
(4.ヒトCD81のBリンパ球特異的発現のためのトランスジーンの作製)
マウスイムノグロブリン重鎖エンハンサー(A.Kudo博士(Basel Institute for Immunology)から贈与された)の700bp BamHIフラグメントおよびマウスκ軽鎖プロモーターの2.3kb XbaI−SacIフラグメントを、pBluescript KS II(+)ベクターにサブクローニングした。SacI部位を、上記のHindIIIリンカーライゲーションによって、HindIII部位に変更した。pCAGSR1PのBamHI部位を、まずNotI部位に変更した。次いで、改変されたpCAGSR1P構築物のプロモーター領域を、EcoRI−HindIII制限消化によって除去し、そしてイムノグロブリンプロモーター−エンハンサーフラグメント(図15におけるpEhKpSR1P)と置き換えた。5.2kbのEcoRI−BamHIフラグメントを接合子注入に供した。
