Description

ノーザンブロット分析を、Human Multiple Tissue Northern Blots IおよびII(Clontech#7760−1および#7759−1)を使用して、実行した。膜を、ハイブリダイゼーション溶液中(50%ホルムアミド、2.5×デンハルト、0.2%SDS、10mM EDTA、2×SSC、50mM NaH2PO4、pH6.5、200μg/mlの超音波処理したサケ***DNA)で2時間、60℃でインキュベートした。hBAFFのアミノ酸136−285に対応するヌクレオチドを含むアンチセンスRNAプローブを、熱変性して、そして2×106cpm/mlにて、新鮮なハイブリダイゼーション溶液中に加えた。この膜を、62℃で16時間ハイブリダイズさせ、2×SSC、0.05%SDS(25℃で30分間)で1度洗浄し、0.1×SSC、0.1%SDS(65℃で20分間)洗浄し、−70℃にてX線フィルムに曝露した。

全長hBAFFを、オリゴJT1069(5’−GACAAGCTTGCCACCATGGATGACTCCACA−3’)およびJT637(5’−ACTAGTCACAGCAGTTTCAATGC−3’)を使用して、増幅させた。PCR産物をPCR−0ブラントにクローン化して、そしてHindIII/EcoRIフラグメントとしてPCR−3哺乳動物発現ベクター内に再度サブクローン化した。可溶性BAFFの短バージョン(version)(アミノ酸Q136−L285)を、オリゴJT636(5’−CTGCAGGGTCCAGAAGAAACAG−3’)およびJT637を使用して増幅した。可溶性BAFF(アミノ酸L83−L285)の長バージョンを、内部PstI部位を使用して全長BAFFから得た。可溶性BAFFを、PstI/EcoRIフラグメントとして、改変されたPCR−3ベクターのヘマグルチニン(haemaglutinin)シグナルペプチドおよびFlag配列の後ろに再度サブクローン化し、そしてPstI/SpeIフラグメントとして、N−末端Flag配列(14)を有する、改変されたpQE16細菌発現ベクターにインフレーム(in frame)で再度サブクローン化した。構築物を、両方の鎖について配列決定した。短い可溶性形態または全長BAFFを発現する安定の293細胞株の樹立、および細菌および哺乳動物293細胞に由来する組換え可溶性BAFFの発現および精製を、記載されるように実施した(14、15)。
T細胞、B細胞、インビトロ誘導された未熟な樹状細胞、293wtおよび293−BAFF(全長)細胞から抽出された総RNAを、製造業者の指示に従って、Ready to Go system(Pharmacia)を使用して逆転写した。BAFFおよびβアクチンcDNAを、特定のオリゴヌクレオチド(BAFFについて、JT1322 5’−GGAGAAGGCAACTCCAGTCAGAAC−3’およびJT1323 5’−CAATTCATCCCCAAAGACATGGAC−3’;IL−2レセプターα鎖について、JT1368 5’−TCGGAACACAACGAAACAAGTC−3’およびJT1369 5’−CTTCTCCTTCACCTGGAAACTGACTG−3’;βアクチンについて、5’GGCATGGTGATGGACTCCG−3’および5’−GCTGGAAGGTGGACAGCGA−3’)を使用して、TaqDNAポリメラーゼを用いてPCR増幅(94℃、55℃および72℃をそれぞれ1分間ずつ30サイクルの工程)によって検出した。
293T細胞を、可溶性BAFFの短い形態を用いて一過的にトランスフェクトし、そして無血清Optimen培地中で7時間、増殖させた。馴化上清を20×に濃縮し、内部標準のカタラーゼおよびオボアルブミンと混合し、そしてSuperdex−200 HR10/30カラム上にロードした。タンパク質を、0.5ml/分にてPBS中で溶出させ、そして画分(0.25ml)をトリクロロ酢酸で沈殿させ、そして抗Flag M2抗体を使用してウエスタンブロッティングによって、分析した。カラムを標準タンパク質(フェリチン(440kDa)、カタラーゼ(232kDa)、アルドラーゼ(158kDa)ウシ血清アルブミン(67kDa)、オボアルブミン(43kDa)、キモトリプシノーゲンA(25kDa)およびリボヌクレアーゼA(13.7kDa)で較正した。
サンプルを、20μlの0.5%SDS、1%2メルカプトエタノール中で3分間、95℃にて加熱し、次いで冷却して10%Nonidet P−40(2μl)、0.5Mリン酸ナトリウム、pH7.5(2μl)およびペプチドNグリカナーゼF(125units/μl、1μlまたはコントロールにおいては酵素なし)を補充した。サンプルを、ウエスタンブロット法による分析の前に37℃で3時間インキュベートした。
293T細胞を、一過的に、長い形態の可溶性BAFFを用いてトランスフェクトし、そして無血清Optimen培地中で7日間増殖させた。馴化上清を、20倍に濃縮して、SDS−PAGEによって分画し、これまでに記載されたようにポリビニリデンジフルオリド膜(BioRad Labs、Hercules、CA)上にブロットし(16)、次いで、フェニルチオヒダントインC18 2.1×250mmカラムを装備したアナライザー(ABI 120A、Perkin Elmer)と連結した気相シーケンサー(ABI 120A、Perkin Elmer、Foster City、CA)を使用して、配列決定した。データを、ソフトウェアABI610(Perkin Elmer)を使用して分析した。
ポリクローナル抗体を、組換え可溶性BAFFを用いて免疫化したウサギ(Eurogentec、Seraing、Belgium)によって生成した。同じ抗原で免疫化したラットの脾臓を、x63Ag8.653マウスミエローマ細胞と融合させ、そしてハイブリドーマを、BAFF特異的IgGについてスクリーニングした。これらのモノクローナル抗体の一つ(43.9)が、hBAFFを特異的に認識するIgG2aである。
末梢血白血球を、2μg/mlのヤギ抗ヒトμ鎖抗体(Sigma)またはコントロールF(ab’)2の存在下または非存在下で72時間、96ウェルプレート(10%FCSを補充した100μlのRPMI中で105細胞/ウェル)において、示された濃度のネイティブまたは煮沸された可溶性BAFF/長とともにインキュベートした。細胞を、[3H]チミジン(1μCi/ウェル)を用いてさらに6時間パルスし、そして収集した。[3H]チミジンの取り込みを、液体シンチレーション計数によってモニターした。いくつかの実施形態において、組換え可溶性BAFFを、1%パラホルムアミド中、25℃で5分間固定された全長BAFF(またはコントロールとして293wt)で安定にトランスフェクトされた293細胞によって置換した。アッセイを、記載されるように実施した(17)。さらなる実験において、CD19+細胞を、磁気ビーズを用いてPBLから単離し、そして残りのCD19-細胞を、CD19+細胞で再構築する前に、照射した(3000ラド)。次いでBAFFに対する増殖アッセイを上記のように実施した。
精製されたB細胞を、記載されるようにEL−4培養系において活性化した(13)。手短には、5×104個の照射されたマウスEL−4胸腺腫細胞(クローンB5)と混合された104個のB細胞を、PHA(1μg/ml)およびPMA(1ng/ml)で48時間活性化されたヒトT細胞(106/ml)に由来する、5%v/vの培養上清を含む200μl培地において5〜6日間培養した。次いでB細胞を、抗CD19ビーズを用いて再度単離して、さらに7日間(200μl中5×104個の細胞、平底96ウェルプレートにおける2連培養または3連培養)、培地単独または5%T細胞上清もしくは50ng/ml IL−2(先のGlaxo Institute for Molecular Biology、Genevaから恵与された)および各々10ng/mlのIL−4およびIL−10(Peprotech、London、UK)を補充した培地中で、sBAFFの存在下または非存在下において培養した。抗Flag M2抗体を、2μg/mlの濃度にて添加し、そして単独では効果がなかった。培養上清中のIgM、IgGおよびIgAを、記載されるように、ELISAアッセイによって定量した(13)。
以下の実験手順を、実施例1〜6において使用した。
ヒトcDNA配列およびマウスcDNA配列は、両方とも以前に記載されている(Schneiderら、1999)。全長マウスBAFFをコードするPCRフラグメントを、RT−PCRにより作製した。第1鎖cDNAを、製造者のプロトコール(GibcoBRL、Grand Island、NY)に従う、オリゴdTを使用して、マウス肺ポリA+(Clontech、Palo、Alto、CA)から合成した。このPCR反応物は、1×Pfu緩衝液(Stratagene,La Jola,CA),0.2mM dNTP,10% DMSO,12.5pM プライマー,5ユニットのPfu酵素(Stratagene)およびNot1制限部位を有する以下のプライマー:5’−TAAGAATGCGGCCGCGGAATGGATGAGTCTGCAAA−3’および5’−TAAGAATGCGGCCGCGGGATCACGCACTCCAGCAA−3’を含んだ。鋳型を94℃1分間、54℃2分間および72℃3分間の30サイクル、続いて72℃で10分間の伸長の間に増幅した。この配列は、GenBankファイルAF119383のヌクレオチド214〜1171に対応する。このPCRフラグメントをNot1で消化し、次いで変形型pCEP4ベクター(Invitrogen、Carlsbad、CA)中にクローン化した。マウスBAFFを有するフラグメントを、SV40 ポリA付加部位の配列を含ませるためにXbaIで取り出した。このフラグメントをプロモーター配列を付加したpUCベースのベクター中にクローン化した。このプロモーター(ヒトApoEエンハンサーおよびAAT(α抗トリプシン)プロモーターを含む1Kbの平滑末端Bg12−Not1フラグメントを含む)を、プラスミドクローン540B(Katherine Parker Ponder博士(Washington University、St.Louis,MO)からの好意の提供)から精製した。EcoRV/Bgl2フラグメントを最終ベクターから精製し、そしてトランスジェニックマウスの作製に使用した。導入した結果である子孫C57BL/6J(雌性)×DBA/2J(雄性)F1(BDF1)マウスを、C57BL/6マウスと戻し交配した。マイクロインジェクションおよびトランスジェニックマウスの作製の技術は、以前に記載されている(Mcknightsら、1983)。
血清サンプルを、リニアーな(linear)12.5%ゲルを使用して還元型SDS−PAGE分析に供した。製造者の指針に従い、Promega(Madison、WI)製の単離キットを使用してマウス肝臓由来の総RNAを、調製し、そしてノーザンブロット分析のために処理した。BAFF導入遺伝子特異的mRNAを、その導入遺伝子構築物のSV40ポリAテールにまたがるプローブを使用して検出し、そしてXba1およびBamH1を用いて、改変型pCEP4ベクターの消化により得た。このプローブは、BAFF導入遺伝子由来のmRNAに対応する1.8〜2Kdのバンドを認識する。BAFF Tgマウス由来のテールDNAのPCR分析を、1×Taq ポリメラーゼ緩衝液(Stratagene)、0.2nMのdNTP、10% DMSOおよび5ユニットのTaqポリメラーゼ(Stratagene)を含む反応物中で、12.5pMの以下のプライマー:5’−GCAGTTTCACAGCGATGTCCT−3’および5’−GTCTCCGTTGCGTGAAATCTG−3’を使用して行った。719bpの導入遺伝子を、94℃にて30秒間、54℃にて1分間および72℃にて1.5分間の35サイクル、続いて72℃で10分間の伸長の間に増幅した。
EDTAで抗凝固化された新鮮な全血の示差的なWBC計数を、Abbott Cell Dyne 3500装置(Chicago、IL)を用いて行った。FACS分析のために、フルオレセイン(FITC)標識したラット抗マウス抗体、Cy−クロム−標識したラット抗マウス抗体、およびフィコエリトリン(PE)標識したラット抗マウス抗体:抗B220、抗CD4、抗CD8、抗CD43、抗IgM、抗CD5、抗CD25、抗CD24、抗CD38、抗CD21、抗CD44、抗L−セレクチンおよびハムスター抗Bcl−2/コントロールハムスターIgキットをPharmingen(San Diego、CA)から購入した。組換えE.coliならびに哺乳動物細胞由来のヒトFlagタグ化BAFFおよびマウスFlagタグ化BAFFの生成は、以前に記載された(Schneiderら、1999)。全ての抗体を製造者の仕様書に従い使用した。PBLを以下のようにマウスの血液から精製した:マウスの血液をEDTAの入ったマイクロチューブに収集し、そしてPBSで1/2に希釈した。500μlの希釈した血液を4mlのガラス試験管中の1mlのフィコール(Celardane,Hornby,Ontario,Canada)の上にアプライし、勾配遠心を2000rpmで30分間室温にて行い、そしてリンパ球を含む界面を収集し、そしてFACS染色の前にPBS中で2回洗浄した。脾臓、骨髄および腸間膜リンパ節を粉砕して、RPMI培地(Life Technologies,Inc.、Grand Island、NY)中で単一細胞懸濁液にし、そしてFACS緩衝液(2%のウシ胎仔血清(JRH Biosciences、Lenexa、KS)を補充したPBS)中で洗浄した。最初に細胞をブロック試薬(10μg/mlのヒトIg(Sandoz、Basel、Switzerland)および10μg/mlの抗マウスFc遮断抗体(Pharmingen))を補充したFACS緩衝液に懸濁し、そして蛍光色素標識抗体で染色する前に氷上で30分間インキュベートした。全ての抗体を上記のブロック試薬を含むFACS緩衝液中に希釈した。サンプルを、FACScan細胞蛍光測定器(Becthon Dickinson)を使用して分析した。
ELISAプレート(Corning glass works、Corning,NY)を、50mMの炭酸水素ナトリウム緩衝液(pH9.6)中の10μg/mlのヤギ抗総マウスIg(Soutern Biotechnology Associates,Inc.Birmingham,AL)の溶液を用いて一晩4℃にてコーティングした。プレートをPBS/0.1% Tweenで3回洗浄し、そしてPBS中で1%ゼラチンで一晩ブロックした。100μl/ウェルの血清段階希釈物または標準の希釈物を、30分間37℃にてプレートに添加した。マウスIgを、30分間37℃にて、アルカリホスファターゼ(AP)標識ヤギ抗総マウスIg(Southern Biothechnology Associates)の1μg/mlの溶液を100μl/ウェル使用して検出した。最終洗浄(PBS/0.1% Tweenの3回)後、酵素反応を、10%のジエタノールアミン中の10μg/mlのp−ニトロフェニルホスフェート(Boehringer Mannheim、Indianapolis、IN)の溶液を使用して発色させた。この反応を100μl 3N NaOH/ウェルを添加することにより停止した。光学密度(O.D.)を、Molecular Devices(Sunnyvale、CA)からの分光光度計を使用して405nmで測定した。標準曲線を、Southern Biothechnology Associatesから購入した精製されたマウスIgを使用して得た。リウマチ因子(RF)の検出の場合、このプレートを、ヤギ抗マウスIgの代わりに正常ヤギIg(Jackson ImmunoResearch laboratories,Inc.(West Grove、PA))を用いてコートし、マウスIgの検出を上記のように行った。RFアッセイにおけるマウスアイソタイプの検出を、AP標識ヤギ抗マウスIgA、IgM、IgG2a、IgG2bおよびIgG3を使用して行い、ならびに標準曲線のために精製マウスIgA、IgM、IgG2a、IgG2bおよびIgG3(Southern Biothecnology Associates Inc.)を使用して行った。全ての統計学的比較を分散分析により実行した。
このアッセイを、以下の改変を伴って、以前に記載された(Juneら、1979;SinghおよびTingle、1982)ように実施した:ELISAプレート(Corning glass works)を、50mMの炭酸水素ナトリウム緩衝液(pH9.6)中の5μg/mlのヒトClq(Quidel、San Diego、CA)を用いて4℃にて一晩コートした。このプレートをPBS/0.1% Tweenで3回洗浄した。50μl/ウェルの0.3M EDTAを、50μl/ウェルの血清の段階希釈物または既知濃度の標準の免疫複合体(DAKO(Carpinteria、CA)製のペルオキシダーゼ−マウス抗ペルオキシダーゼ(PAP))の溶液を加えたプレートに添加した。このプレートを30分間37℃にてインキュベートした。このプレートをPBS/0.1% Tweenで3回洗浄した。この免疫複合体中のマウスIgを、ELISAアッセイについて上記するように、100μl/ウェルの、Ap標識ヤギ抗マウスIg(Southern Biotechnology Associates,Inc.)の1μg/ml溶液を使用して検出した。クリオグロブリンを水で1/15に希釈したマウス血清を4℃で一晩インキュベートすることにより検出し、そして沈降物を視覚的に評価した。
抗ssDNAをNUNC−immuno Plate MaxiSorpプレート(NUNC A/S、Denmark)を使用して行った。初めに、プレートを100μg/mlのメチル化BSA(Calbochem Corp.、La Jolla、CA)を用いて、次いで50μg/mlのグレードI仔ウシ胸腺DNA(Sigma,St.Louis,MO)を用いて4℃にて一晩コートした。この仔ウシ胸腺DNAは、使用前に超音波処理により剪断され、次いでS1ヌクレアーゼで消化した。抗ssDNAアッセイのために、使用前にこのDNAを10分間ボイルし、そして氷上で冷却した。ブロッキング後、血清サンプルの段階希釈物を添加し、そして室温にて2時間インキュベートした。自己抗体をヤギ抗マウスIgG−AP(Sigma)を用いて検出し、そしてELISAアッセイについて上記に記載するように発色させた。標準曲線を既知量の抗DNA mAb 205を使用して得た。この抗DNA mAb 205は、ssDNAおよびdsDNAの両方に特異的である(Dattaら、1987)。
脾臓およびリンパ節をO.T.C.包埋培地(Miles、Elkhart、IN)中で凍結し、そして低温槽切片化のために封入した。7〜10μm厚の切片を乾燥し、そしてアセトン中で固定した。全てのAb(10μg/ml)を、Tris−緩衝化生理食塩水A(TBS−A、0.05M Tris、0.15M NaCl、0.05% Tween−20(v/v)、0.25% BSA)に希釈した後、室温にて、1時間加湿ボックス中でインキュベーションし、TBS−B(0.05M Tris、0.15M NaCl、0.05% Tween−20)中でリンスし、そして酵素反応を誘発する前に、メタノール中で1分間固定した。西洋ワサビペルオキシダーゼ(HRP)活性およびアルカリホスファターゼ(AP)活性を、各々、ジアミノベンジジン(DAB)錠剤基質キット(Sigma)および5−ブロモ−4−クロロ−3−インドリルホスフェート/ニトロブルーテトラゾリウム(BCIP/NBT、Pierce、Rockford、IL)を使用して発色させた。染色した組織切片を最終的に5分間メタノール中で固定し、そしてGiemsa(Fluka、Buchs、Switzerland)で対比染色した。ビオチン標識したラット抗B220抗体、抗CD11c抗体、抗syndecan−1抗体ならびに非標識のラット抗CD4抗体、抗CD8μ抗体および抗CD8μ抗体をPharmingenから購入した。ビオチン標識したピーナッツ凝縮素(PNA)をVector laboratories(Burlingame、CA)から得た。(HPR)標識マウス抗ラットIgおよび(HRP)−ストレプトアビジンをJackson ImmunoResearch laboratories,Inc.から、そしてAp標識ストレプトアビジンをSouthern Biotechnology Associates,Inc.から購入した。Ig沈着を検出するための腎組織についての免疫組織化学の場合、パラフィン切片を用い、パラフィンを取り除き、そしてVector(Burlingame、CA)からの希釈したウマ血清を使用してブロックし、その後Jackson Immunoresearch製のHPR−ヤギ抗マウスIgで染色した。上記のように検出を行った。
全長マウスBAFFを、APO Eエンハンサーを伴う肝臓特異的α-1抗トリプシンプロモーターを使用して、トランスジェニックマウスにおいて発現させた。全長バージョンは、BAFFが切断されそして全身的に作用するか、または膜結合性形態で保たれるかのどちらかであり、膜結合形態で保たれる場合に局所的な肝臓特異的な異常が観察されて、おそらく機能的な端緒を提供するという予想のもと、選択した。本発明者らは、BAFF導入遺伝子について陽性である13匹の創始マウスを得た(表2)。これらのマウスのうちの4匹を若齢で屠殺した。慣用的な病理学をマウス811およびマウス816に対して行った(表2)。これらのマウスにおいて明らかな感染はなかったが;しかし心血管異常および腎臓異常が明確であり、そして重篤な高血圧症について記載される異常(Fu、1995)と類似していた(表2)。創始816のヘマトキシリンおよびエオシン(HおよびE)で染色された腎組織切片は、そのマウスにおける糸球体の形態が異常であり、一方腎組織の残りは、正常のようであったことを示した(データは示さず)。多くのBAFFトランスジェニック創始マウスは、タンパク尿を有した(表2)。マウス816からの脾臓凍結組織切片の免疫組織化学は、異常かつ広範なB細胞染色およびT細胞についての染色の減少を明らかにし、そしてこの観察は、その子孫において確認された(以下の図12を参照のこと)。
コントロールのネガティブ同腹子と比較した場合、トランスジェニックマウス集団が血液中でより多くのリンパ球を有することを見出し、13000リンパ球/□l血液程度の高い値に達した(図7A)。対照的に、BAFF Tgマウスおよびコントロールマウスの両方において血液□lあたりの顆粒球の数は、通常の範囲内のままであった(図7A)。6匹の異なる創始マウスから生まれた18匹のBAFF Tgマウス由来の末梢血細胞(PBL)の抗CD4抗体および抗B220抗体を用いるFACS分析が、増加したB/T比を示したので(図7Bおよび7C)、上昇したリンパ球レベルは、増殖したB細胞部分集合に起因した。同様に、この方法を用いて、CD4循環T細胞の絶対数の計算は、コントロールマウスと比較した場合、BAFF Tgマウス中のこのT細胞部分集合の50%の減少を示し、そしてこの同様の観察が、CD8 T細胞部分集合についてなされた(データは示さず)。BAFF TgマウスのPBL由来の全てのB細胞を、コントロールマウス由来のB細胞と比較した場合、MHCクラスII発現およびBcl−2発現が増加し(それぞれ、図7Dおよび図7E)、BAFF TgマウスのPBLにおいていくらかのレベルのB細胞活性化を示した。BAFF Tgマウスの血液中のT細胞は、早期活性化マーカーCD69またはCD25を発現しなかった;しかし、40〜56%のCD4T細胞またはCD8T細胞は、コントロール同腹子のわずか8%〜12%のみに対して、CD44hi、L−セレクチンlo表現型を有する活性化エフェクターT細胞であった(図7F)。従って、BAFF Tgマウスは、明らかにT細胞改変とともにB細胞リンパ球増加症および全B細胞活性化の徴候を示す。
トランスジェニックマウスにおけるBAFFの過剰発現が、骨髄における中枢B細胞画分に、そして二次リンパ器官における末梢B細胞に影響するか否かを調査するために、本発明者らは、4匹の異なる創始マウス由来の全7匹のBAFF Tgマウスおよび7匹のコントロール同腹子由来の脾臓、骨髄および腸間膜リンパ節をFACSにより試験した。成熟B細胞画分を、抗B220抗体および抗IgM抗体の両方で染色することにより分析した。2匹の代表のBAFF Tgマウスおよび1匹の代表のコントロール同腹子を図8に示す。成熟B細胞画分(IgM+。B220+)は、脾臓および腸間膜リンパ節の両方において上昇した(図8A、それぞれ、上パネルおよび下パネル)。骨髄におけるB220+/IgM+B細胞(図7A、中パネル)画分またはプロB細胞(CD43+/B220+)画分およびプレB細胞(CD43−/B220+)画分の分析(図8)は、BAFF Tgマウスおよびコントロール同腹子が、類似であることを示した。これらのデータは、BAFFの過剰発現が、末梢における成熟B細胞の増殖に影響しているが、骨髄における前駆B細胞の増殖には影響していないことを示す。脾臓におけるB細胞部分集団のFACSによる分析は、コントロールマウスと比較した場合、BAFF Tgマウスにおける辺縁層(MZ)B細胞の上昇した割合を示した(表3)。新しく形成されたB細胞の画分が、BAFF Tgマウスにおいてわずかに減少するのに対して、小胞B細胞の集団は、BAFF Tgマウスおよびコントロールマウスの両方において釣り合ったままであった(表3)。この結果はまた、B220+脾臓B細胞において、抗CD38対抗CD24抗体および抗IgM対抗IgD抗体を使用して、そして先に記載された(Oliverら、1997)ようにMZ B細胞集団についてCD38hi/CD24+およびIgMhi/IgDloのそれぞれで分析して確認した(データは示さず)。抗マウスIgM抗体を使用する免疫組織化学分析は、コントロールマウスと比較した場合、BAFF Tgマウスの脾臓におけるIgMブライト(bright)MZ B細胞領域の拡大を示した(データは示さず)。全てのBAFF Tg B220+脾臓B細胞はまた、コントロールマウス由来の脾臓B細胞と比較して、より高レベルのMHCクラスII(表3)およびBcl−2(データは示さず)を発現し、このことは、脾臓B細胞およびPBL由来のB細胞は、活性化状態であることを示す。
BAFF Tgマウスにおける上昇したB細胞画分は、これらの動物の血液中の全Igのレベルをまた、上昇し得ることを示唆した。BAFF Tgマウスおよびコントロール同腹子由来の血清のSDS−PAGE分析は、重鎖IgGバンドおよび軽鎖IgGバンドが、コントロール血清と比較して、4匹のBAFF Tgマウスのうち3匹において、少なくとも10倍よりも多いことを示した(図9A)。同様に、BAFF Tgマウス由来の血清に対するELISA測定は、コントロールマウスのELISA測定と比較した場合、有意に高い全Igレベルを示す(図9B)。
最初に、本発明者らは、本発明者らの創始マウスのうち2匹において狼瘡様疾患の腎臓異常レミニッセントを観察した(表II)。抗DNA自己抗体の存在はまた、SLE患者またはSLC様(SWR×NZB)F1(SNF1)マウスにおいて、記載されている(Dattaら、1987)。抗ssDNA自己抗体レベルを、最も高いレベルの全血清Igを有することが先に示されたBAFF Tgマウスにおいて、検出した(図10A)。本発明者らは、5匹のコントロール同腹子と平行して、ssDNAに対する抗体に対して陰性の2匹のBAFF Tgマウス(697−5および816−1−1)および抗dsDNA抗体の存在に対して抗ssDNA抗体を分泌する3匹のトランスジェニックマウス(820−14、816−8−3および820−7)の血清を分析した。BAFF Tgマウスはまた、抗dsDNAを分泌したが、分泌のレベルは、抗ssDNA抗体の分泌のレベルと必ずしも相関していたわけではなかった。なぜなら、検出可能なレベルの抗ssDNA抗体を含まないBAFF Tgマウス697−5由来の血清が、抗dsDNAの存在に対して明らかに陽性であったからである(図10B)。従って、最も重篤な高グロブリン血症を示すBAFF Tgマウスは、病理学的レベルの抗DNA自己抗体を分泌する。さらに、そしてこれらのマウスにおける狼瘡様問題のレミニッセントもまた示し、本発明者らは、分析した6匹のBAFF Tgマウスの腎臓において、免疫グロブリン沈着を検出した(図10C)。これらのマウスのうち3匹は、検出可能なレベルの抗DNA抗体を分泌しなかった(データは示さず)。

(c)BAFFリガンドまたはその活性なフラグメントに特異的な抗体;および
(d)BAFFリガンドレセプターまたはそのエピトープに特異的な抗体、

(項目13) 動物において樹状細胞誘導性B細胞増殖および成熟を阻害する方法であって、以下:
(a)抗BAFFリガンド分子またはその活性なフラグメント;
(b)組換え無効性BAFFリガンド分子またはその活性なフラグメント;
(c)BAFFリガンドまたはその活性なフラグメントに特異的な抗体;および

(c)BAFFリガンドまたはその活性なフラグメントおよびCD40リガンド;
(d)BAFFリガンドまたはその活性なフラグメントおよび抗CD40リガンド分子;
(e)抗BAFFリガンド分子またはその活性なフラグメント;
(f)組換え無効性BAFFリガンド分子またはその活性なフラグメント;
(g)BAFFリガンドまたはその活性なフラグメントに特異的な抗体;ならびに
(h)BAFFリガンドレセプターまたはそのエピトープに特異的な抗体、
からなる群より選択される、治療有効量の組成物を投与する工程を包含する、方法。
(項目19) BAFFリガンドに関連する障害を処置する方法であって、以下:
(a)BAFF関連分子をコードする遺伝子を含む、治療有効量のベクターを、所望の細胞中に導入する工程;および
(b)該細胞中に該遺伝子を発現させる工程、
を包含する、方法。
(項目20) 項目19に記載の方法であって、前記BAFF関連分子が、以下:
(a)BAFFリガンドまたはその活性なフラグメント;
(b)BAFFリガンドまたはその活性なフラグメントおよび抗μ抗体;

(項目28) 細胞死を誘導する方法であって、レセプターに対するBAFFリガンドの結合を妨害し得る薬剤を投与する工程を包含する、方法。
(項目29) BAFFリガンドとそのレセプターとの間のシグナル伝達経路に関与する免疫応答を処置、抑制または変更する方法であって、該BAFFリガンドとそのレセプターとの間の結合を妨害し得る、有効量の薬剤を投与する

(項目30) 炎症を阻害する方法であって、BAFFリガンドまたはその活性なフラグメントに特異的な、治療有効量の抗体を投与する工程を包含する、方法。

(項目33) BAFFリガンドとそのレセプターとの間のシグナル伝達経路に関与する免疫応答を処置、抑制または変更する方法であって、該BAFFリガンドとそのレセプターとの間の結合を妨害し得る、有効量の薬剤を投与する工程を包含する、方法。
(項目34) 動物において高血圧を処置する方法であって、治療有効量のB細胞増殖インヒビターを投与する工程を包含する、方法。

Claims (12)
- 免疫抑制疾患の処置のための薬学的組成物の調製のための、可溶性BAFFリガンドまたはその活性なフラグメントの、使用。
- 前記免疫抑制疾患がHIVによって引き起こされる、請求項1に記載の使用。
- 患者におけるB細胞増殖を刺激することによって免疫抑制疾患を処置するための薬学的組成物の調製のための、可溶性BAFFリガンドまたはその活性なフラグメントの、使用。
- 患者における免疫グロブリンの産生を刺激することによって免疫抑制疾患を処置するための薬学的組成物の調製のための、可溶性BAFFリガンドまたはその活性なフラグメントの、使用。
- 患者におけるB細胞増殖および免疫グロブリンの産生を刺激することによって免疫抑制疾患を処置するための薬学的組成物の調製のための、可溶性BAFFリガンドまたはその活性なフラグメントの、使用。
- 前記免疫抑制疾患がHIVによって引き起こされる、請求項3〜5のいずれか1項に記載の使用。
- 免疫学的疾患を処置するために免疫系を操作するための薬学的組成物の調製のための、可溶性BAFFリガンドまたはその活性なフラグメントの、使用であって、該操作は、患者におけるB細胞増殖の刺激である、使用。
- 免疫学的疾患を処置するために免疫系を操作するための薬学的組成物の調製のための、可溶性BAFFリガンドまたはその活性なフラグメントの、使用であって、該操作は、患者における免疫グロブリンの産生の刺激である、使用。
- 前記操作が、患者におけるB細胞増殖および免疫グロブリンの産生の刺激である、請求項7または8に記載の使用。
