Den foreliggende opfindelse angår en særegen fremgangsmåde til fremstilling af 2-thiacephemderivater med den almene formel (I) >9^ - C00R2 10 hvor Ri betyder et hydrogenatom, en alkylgruppe med 1-12 carbonatomer eller en cycloalkylgruppe med 4-7 carbonatomer, hvilke alkyl- og cycloalkylgrupper eventuelt er substitueret med hydroxy, beskyttet hydroxy, amino, beskyttet amino, 15 mercapto, beskyttet mercapto eller cyano, R2 betyder en carboxybeskyttende gruppe valgt blandt alkylgrupper med 1-6 carbonatomer, halogensubstituerede alkylgrupper med 1-6 carbonatomer, eventuelt substituerede arylgrupper, aryl-substituerede alkylgrupper, hvis alkyldel har 1-6 carbon-20 atomer, og hvis aryldel eventuelt er substitueret, aryloxy-substituerede alkylgrupper, benzhydryl, o-nitrobenzhydryl, acetonyl, trimethylsilyl, diphenyl-t-butylsilyl, dimethyl-t-butylsilyl, acetoxymethyl, pivaloyloxymethyl og phthalidyl, og Y betyder et hydrogen- eller halogenatom.

De ved fremgangsmåden isolerede salte vil især være pharmaceutisk og/eller veterinært acceptable salte, og de kan både være salte med syrer, enten uorganiske syrer, såsom saltsyre eller svovlsyre, eller organiske syrer, såsom ci-5 tronsyre, vinsyre, myresyre eller methansulphonsyre, og salte med baser, enten uorganiske baser, såsom alkalimetal-eller jordalkalimetalhydroxider, især natrium- og kaliumhydroxider, eller organiske baser, såsom triethylamin, py-ridin, benzylamin eller collidin.
10 Forbindelser med formlen (IV), der kan anvendes som udgangsmaterialer, er kendte forbindelser eller kan fås ud fra kendte forbindelser ved i og for sig kendte metoder. Fremstillingen af visse repræsentative forbindelser er beskrevet i eksemplerne.
15 Forbindelsen med formlen (IV) ozonolyseres først, hvorved fås en forbindelse med formlen (VI). Hydroxygruppen omdannes derpå til en gruppe L, og den fremkomne forbindelse med formlen (VIII) cycliseres, hvorved fås en forbindelse med formlen (I), hvor Y er et hydrogenatom. Eventuelt kan 20 methylgruppen derpå halogeneres, således at der fås en forbindelse med formlen (I), hvor Y er et halogenatom.
Omdannelsen af hydroxygruppen til en gruppe L i enolen (VI) eller (VIII), der kan være i ligevægt med den tilsvarende ketotautomer, er fortrinsvis en mesylering. Det har 25 overraskende vist sig, at der, når denne reaktion udføres i tetrahydrofuran i stedet for de almindeligt anvendte halogenerede carbonhydrider, næsten udelukkende fås mesylater (VIII) med Z-alkengeometri, der er de mest egnede til den følgende cyclisering (lignende omdannelse udført i dichlor-30 methan giver i reglen en l:l-blanding af Ε,Ζ-isomere, jfr.

Chem. Soc., Chem. Comm. 1979, 663). Cyclisering af forbindelsen med formlen (VIII) kan udføres i et enkelt trin, ved 35 omsætning med et sulphid eller hydrosulphid, såsom Na2S,
NaHS, BU4NHS eller med H2S, i nærvær af en base, såsom tri-

Cyclisering af (VIII), hvor Z er en gruppe, der ikke er SR7, giver den åbenbare fordel, at der dannes let fra-skillelige, i reglen vandopløselige bi-produkter ZH (f.eks.
5 phenylsulphinsyre, succinimid) i stedet for biprodukter R3SH (f.eks. mercaptobenzthiazol), der i reglen kræver chro-matografisk fraskillelse eller fældning som tungmetalsalte (Ag+, Pb2+).
Stik mod enhver fornuftig forventning om, at mulighe-10 den for halogenering af 3-methylgruppen i forbindelserne med formlen (I) (Y = H) skulle være afhængig af tilstedeværelsen af disulphidmolekyldelen, er der nu fundet en fremgangsmåde til udførelse af en sådan omdannelse med stort udbytte. Derved kan der fremstilles forbindelser med formlen 15 (I) (Y = halogen), som er uvurderlige mellemprodukter ved syntesen af højaktive penemantibiotika. Et foretrukket ha-logeneringsreagens til en sådan omdannelse er N-bromsuc-cinimid, der bedst anvendes i nærværelse af en radikalinitiator, såsom azobisisobutyronitril eller benzoylperoxid, i 20 nærvær af syrefjernende midler, såsom epoxider (f.eks. propyl enoxid) , jordalkalimetaloxider (f.eks. calciumoxid) eller molekylsigter, i opløsningsmidler, såsom benzen eller car-bontetrachlorid, ethylformiat, ved en temperatur fra 20 til 130 °C.
25 Forbindelserne med formlen (I) (Y = halogen) kan omdannes til forbindelser med formlen (I) (Y = en organisk gruppe) ved i og for sig kendte metoder, f.eks.: 1) En forbindelse med formlen (I) (Y = Br eller Cl) kan omdannes til en forbindelse med formlen (I) (Y = fri 30 eller beskyttet OH) ved mild, alkalisk hydrolyse eller ved omsætning med cuprooxid/dimethylsulphoxid/vand eller ved omsætning med et salt af en stærk uorganisk syre, f.eks. et nitrat eller perchlorat, hvorved der fås en labil ester med den uorganiske syre, hvilken ester kan hydrolyseres senere 35 eller i samme reaktionsmedium til den ønskede stamalkohol. Foretrukne salte af denne type er AgN03, AgCl04 og NaN03.

7 2) En forbindelse med formlen (I) (Y = Br eller Cl) kan omdannes til en forbindelse med formlen (I) (Y = en usubstitueret eller N-alkylsubstitueret carbamoyloxygruppe) ved omdannelse til en forbindelse med formlen (I) (Y = OH) 5 som beskrevet ovenfor efterfulgt af omsætning med et hensigtsmæssigt isocyanat, f.eks. er trichloracetylisocyanat et foretrukket reagens til fremstilling af forbindelser med formlen (I) (Y = OCONH2), efterfulgt af fjernelse af beskyttelsen fra trichloracetylmolekyldelen i det først dannede 10 urethanadditionsprodukt.
3) En forbindelse med formlen (I) (Y = Br eller Cl) kan omdannes til en forbindelse med formlen (I) (Y = acyloxy) ved omsætning med et hensigtsmæssigt salt af den tilsvarende carboxylsyre i hensigtsmæssige opløsningsmidler eller under 15 faseoverføringskatalyse eller ved omdannelse til en forbindelse med formlen (I) (Y = OH) efterfulgt af gængs acylering.
4) En forbindelse med formlen (I) (Y = Br eller Cl) kan omdannes til en forbindelse med formlen (I) (Y = S-Het) ved omsætning med det tilsvarende HS-Het i nærvær af en 20 base, eller med et i forvejen dannet salt af HS-Het med en base, i et passende opløsningsmiddel, såsom tetrahydrofuran, acetone, acetonitril eller dimethylformamid. En hensigtsmæssig base er triethylamin, et hensigtsmæssigt, i forvejen dannet salt er et natriumsalt, f.eks. natrium-l-methyl-25 1,2,3,4-tetrazol-5-yl-mercaptid.
På grund af 3-hydroxymethyl-2-thiacephem-4-carboxyla-ternes udtalt tilbøjelighed til lactondannelse foretrækkes det i fremgangsmåde 1) ovenfor, at R2 repræsenterer en noget omfangsrig gruppe, således at der med den bundne carboxymo-30 lekyldel dannes en ester, der er forholdsvis inaktiv over for nukleofilt angreb på den ved siden af liggende hydroxy-gruppe, f.eks. en tert.butylester. Alternativt kan det være hensigtsmæssigt at fjerne beskyttelsen fra hydroxygruppen efter ringsammentrækningstrinnet til den tilsvarende penem, 35 eftersom 2-hydroxymethylpenemcarboxylater ikke så let danner lactoner. Således kan en forbindelse med formlen (I) (Y = 8

Br) omdannes til en forbindelse med formlen (I) (Y = 0N02), der let kan isoleres, renses om nødvendigt, afsvovles til den tilsvarende penem, hvis reduktive hydrolyse (f.eks. Zn/CHjCOOH) giver det frie hydroxyderivat uden problemer.
5 På grund af penem- og 2-thiacephemkernens forskellige stabilitet over for de betingelser, der er nødvendige til -COOR2-esterhydrolyse, er en af opfindelsens klare fordele, at esterhydrolyser, der ikke er forligelige med en penem, kan foregå på 2-thiacephemforstadiet, og ringsammentrækningen 10 kan ske på den frie syre eller på et salt med en organisk eller uorganisk base eller på en anden labil ester, der om ønsket kan fremstilles in situ, f.eks. en trimethyl-, t-bu-tyldimethylsilyl- eller t-butyldiphenylsilylester.
Opfindelsen vil i det følgende blive nærmere forklaret 15 ved hjælp af eksempler. Forkortelserne Me, Bu^, Ph, Ms, pNB, THF, EtOAc, DMSO, MeCN står hhv. for methyl, tert.butyl, phenyl, methansulphonyl, p-nitrobenzyl, tetrahydrofuran, e-thylacetat, dimethylsulphoxid og acetonitril. NMR-spektre måles på enten et Hitachi-Perkin Elmer 60 MHz-apparat eller 20 på en Brucker 90 MHzj adskillelsen af indre linier i AB-kvar-tetter refererer til spektre taget med sidstnævnte.

En opløsning af 130 mg (0,3 irtmol) 3 (S) ~[i(R) -hydroxy-ethyl]-4(R)-benzthiazolyldithio-1-(l-methoxycarbonyl-2-hy-droxy-l-prop-l-enyl)-azetidin-2-on i 8 ml vandfrit dichlor-methan behandles ved -40°C først med 0,043 ml (0,3 mmol) 15 triethylamin og derefter med 0,024 ml (0,31 mmol) methansul-phonylchlorid. Reaktionen kvæles efter 5 minutter med koldt, vandigt 2%’s NaHC03. Fjernelse af opløsningsmidlet fra det organiske lag giver den rå, i overskriften nævnte forbindelse (kvantitativt udbytte), der anvendes som sådan til næste 20 trin.

Diphenylmethyl-(7S,6R)-7-[1(R)-methylsulphonyloxyethyl]-20 -3-methvl~2-thiacephem-4-carboxylat opløses i kold trifluoreddi-kesyre (0°C, ren). Efter 15 minutters omrøring ved samme temperatur tilsættes carbontetrachlorid, og opløsningen inddampes omhyggeligt under vakuum, uden ydre opvarmning. Remanensen udrives i CC14 og opsamles, hvorved fås den i overskriften nævnte 25 forbindelse.

En opløsning af 200 mg diphenylmethyl-(7S,6R)-7-[l(R)--p-nitrobenzyloxycarbonyloxyethyl]-3-methyl-2-thiacephem-4-carb-oxylat i 25 ml dichlormethan behandles i 30 minutter ved 0°C med 0/4 ml trifluoreddikesyre. Inddampning under vakuum i kulde ef-30 terlader den rå 2-thiacephem-4-carboxylsvre, der opløses i 10 ml acetonitril/dimethvlformamid = 2;1 og behandles med 0,050 ml triethylamin og 100 mg p-nitrobenzylbromid. Efter en time ved 25°C deles blandingen mellem ethylacetat og vandigt NaHCO^.

En opløsning af 310 mg diphenylmethyl-(7S,6R)-7-[l(R)--tert.butyldimethylsilvloxyethyl]-3-brommethyl-2-thiacephem-4--carboxylat i 15 ml tør acetone behandles med 0,4 ml pyridin.

Efter inddampning i vakuum optages remanensen i en lille smule chloroform. Der tilsættes ethylether under omrøring og derefter afdekanteres, hvilket efterlader den rå, 25 i overskriften nævnte forbindelse.

En opløsning af 460 mg p-nitrobenzyl-(7S,6R)-7-[l(R)--p-nitrobenzyloxycarbonyloxyethyl]-3-brommethyl-2-thiacephem-4--carboxylat i 5 ml DMF omrøres natten over i mørke i nærvær af 200 mg nicotinamid. Det meste af opløsningsmidlet afdestille- 15 res, og remanensen tages op i 150 ml tetrahydrofuran. Denne opløsning vaskes gentagne gange med en opløsning af NaCl i 0,1N HC1 (2 x 50 ml), med 2 x 50 ml saltvandsopløsning, tørres (NaS04) og inddampes. Remanensen fyldes på toppen af en kolonne pakket med silaniseret silicagel (Merck, art.
20 7719). Overskydende nicotinamid og urenheder elueres med
EtOAc, derpå opsamles produktet ved eluering med EtOAc/HOAc = 9:1. Inddampning i vakuum giver den i overskriften nævnte forbindelse.

Claims (3)
1. Fremgangsmåde til fremstilling af et 2-thiacephem-derivat med den almene formel I : Rlsv-^> (I) C00R2 10 hvor R^ betyder et hydrogenatom, en alkylgruppe med 1-12 carbonatomer eller en cycloalkylgruppe med 4-7 carbonatomer, hvilke alkyl- og cycloalkylgrupper eventuelt er substitueret med hydroxy, beskyttet hydroxy, amino, beskyttet amino, 15 mercapto, beskyttet mercapto eller cyano, R2 betyder en carboxybeskyttende gruppe valgt blandt alkylgrupper med 1-6 carbonatomer, halogensubstituerede alkylgrupper med 1-6 carbonatomer, eventuelt substituerede arylgrupper, aryl-substituerede alkylgrupper, hvis alkyldel har 1-6 carbon-20 atomer, og hvis aryldel eventuelt er substitueret, aryloxy-substituerede alkylgrupper, benzhydryl, o-nitrobenzhydryl, acetonyl, trimethylsilyl, diphenyl-t-butylsilyl, dimethyl-t-butylsilyl, acetoxymethyl, pivaloyloxymethyl og phthalidyl, og Y betyder et hydrogen- eller halogenatom, kende-25 tegnet ved, at en forbindelse med den almene formel (IV) R. 2. Fremgangsmåde ifølge krav l,kendeteg-15 net ved, at omdannelsen af hydroxygruppen til gruppen L fortrinsvis er en mesyleringsreaktion gennemført i tetrahy-drofuran.
3. Fremgangsmåde ifølge krav 1, kendetegnet ved, at en 2-thiacephemforbindelse med formlen (I), 20 hvor Y betyder hydrogen, om ønsket halogeneres med et halogener ingsmiddel i nærvær af en radikalinitiator og et syre-bindende middel i opløsningsmiddel ved en temperatur fra 20°C til 130°C.
