Description
- Die vorliegende Erfindung betrifft neue Aryl- und Heteroarylethylen- Derivate, ein Verfahren zu deren Herstellung, pharmazeutische Zusammensetzungen die diese enthalten und deren Verwendung als therapeutische Mittel.
- Shiraishi et al. berichteten in Chem. Pharm. Bull. 36(3) 974 - 981 (1988) von Untersuchungen zur inhibitorischen Aktivität einer Reihe von α- Cyanocinnamamid-Derivaten gegen Tyrosin-spezifische Proteinkinase. Untersuchung zur Blockierung der epidermalen Wachstumstaktoren (EGF) Rezeptorkinase durch eine Reihe von strukturell verwandten Verbindungen wurde von Yaish etc al. in Science 242, 933 (1988) berichtet. In Yakugaku Zasshi, Band 97(9), 1033 - 1039 (1977) beschreiben Kobayashi et al. Untersuchungen zur Antitumor-Aktivität von Indolyl-Derivaten von Oxindol. Strukturell verwandte Indolyl-Derivate von Oxindol werden in Chem. Ber. 102, 1347 - 1356 (1969) offenbart, wobei diese Veröffentlichung nicht aut pharmazeutische Verwendungen hinweist.
- Die vorliegende Erfindung stellt Verbindungen mit der allgemeinen Formel (I)
- bereit, worin Y ein bicyclisches Ringsystem ist, ausgewählt aus (A) und (B)
- R eine Gruppe mit der Formel (a), (b) oder (c) ist
- worin R&sub3; -OH ist;
- R&sub1; ein Wasserstoffatom, eine C&sub1;-C&sub6;-Alkyl- oder eine C&sub2;-C&sub6;-Alkanoyl- Gruppe ist; R&sub2; ein Wasserstoffatom, ein Halogenatom, eine Cyano- oder eine C&sub1;-C&sub6;-Alkyl-Gruppe ist;
- n Null oder eine ganze Zahl von 1 bis 3 ist, wenii Y das Ringsystem (A) ist, aber n Null, 1 oder 2 ist wenn Y das Ringsystem (B) ist; oder deren pharmazeutisch verträglichen Salze; wobei jeder der Substituenten R, OR&sub1; und R&sub2; unabhängig voneinander an jede der beiden Aryl-Gruppen des bicyclischen Ringsystems (A) gebunden sein kann, wohingegen im bicyclischen Ringsystem (B) nur der Benzolrest substituiert sein kann und worin, wenn Y das Ringsystem (A) ist, R von den obigen Gruppen (a) und (b) verschieden ist.
- Die Erfindung schließt alle mögiichen Isomere, Stereoisomere, insbesondere Z und E-Isomere und deren Mischungen sowie die Metaboliten und die Metabolit-Vorstufen oder Bio-Vorstufen (auch als pro-drugs bekannt) der Verbindung mit der Formel (I) ein.
- Der Substituent R ist vorzugsweise in 1- oder 2-Position der Ringsysteme (A) und (B) gebunden. Der Substituent R&sub2; kann unabhängig davon an jeden der Ringe in den bicyclischen Ringsystemen (A) und (B) gebunden sein.
- Wenn Y das bicyclische Ringsystem (A) ist, sind die -OR&sub1;-Gruppen bevorzugt an den gleichen Benzolringen gebunden wie die R-Gruppe. Im Ringsystem (A) ist der Substituent R&sub2; bevorzugt an den gleichen 6- gliedrigen Ring gebunden wie der Substituent -OR&sub1;, d.h. in ortho-, meta- oder para-Position zu -OR&sub1;. Vorzugsweise ist R&sub2; in ortho- oder para- Position zu OR&sub1;.
- Der Substituent -OR&sub1; ist vorzugsweise in 1-, 2-, 3-, 4-, 5- oder 8- und besonders bevorzugt in 1-, 2-, 3 oder 4-Position in den Ringsystemen (A) und (B) gebunden. Natürlich kann nur einer der Substituenten R, -OR&sub1; und R&sub2; an die gleiche Position in den Ringsystemen (A) und (B) gebunden sein.
- Wenn n 2 oder 3 ist, können die -OR&sub1;-Gruppen gleich oder verschieden sein.
- Die Alkyl-Gruppen und die Alkyl-Reste in den Alkanoyl-Gruppen können verzweigt oder geradkettige Alkyl-Ketten sein. Eine C&sub1;-C&sub6;-Alkyl-Gruppe ist bevorzugt eine C&sub1;-C&sub4;-Alkyl-Gruppe, z.R. Methyl, Ethyl Propyl, Isopropyl, Butyl, sek-Butyl oder tert-Butyl, insbesondere Methyl oder Ethyl. Eine C&sub2;-C&sub6;-Alkanoyl-Gruppe ist bevorzugt eine C&sub2;-C&sub4;-Alkanoyl-Gruppe, insbesondere Acetyl, Propionyl oder Butyryl.
- Ein Halogenatom ist bevorzugt Chlor, Brom oder Fluor, insbesondere Brom.
- Pharmazeutisch verträgliche Salze der erfindungsgemäßen Verbindung schließen Säureadditionssalze mit anorganischen Säuren z.B. Salpetersäure, Salzsäure, Bromwasserstoff, Schwefelsäure, Perchlorsäure und Phosphorsäure oder mit organischen Säuren wie z.B. Essigsäure, Propionsäure, Glykolsäure, Milchsäure, Oxalsäure, Malonsäure, Äpfelsäure, Maleinsäure, Weinsäure, Zitronensäure, Benzoesäure, Zimtsäure, Mandelsäure und Salicylsäure und Salze mit anorganischen Materialien, z.B. Alkalimetallbasen, insbesondere Natrium oder Kalium oder Erdalkalimetallbasen, insbesondere Calcium oder Magnesium oder mit organischen Basen, z.B. Alkylamin, bevorzugt Triethylamin ein.
- Wie oben ausgeführt, sind erfindungsgemäß auch pharmazeutisch verträgliche Biovorstufen (auch als pro-drugs bekannt) der Verbindung mit der Formel (I) eingeschlossen, d.h. Verbindungen, die eine andere Formel als Formel (I) haben, die jedoch nach Verabreichung an einen Menschen direkt oder indirekt in vivo in eine Verbindung mit der Formel (I) umgewandelt werden. Bevorzugte erfindungsgemäße Verbindungen sind Verbindungen mit der Formel (I) worin
- R&sub1; ein Wasserstoffatom, eine C&sub1;-C&sub4;-Alkyl- oder C&sub2;-C&sub4;-Alkanoyl-Gruppe ist, und
- R&sub2; ein Wasserstoffatom ist;
- sowie deren pharmazeutisch verträglichen Salze. Besonders bevorzugte erfindungsgemäße Verbindungen sind Verbindungen mit der Formel (I), in denen
- R&sub1; und R&sub2; Wasserstoffatome sind;
- und deren pharmazeutisch verträglichen Salze.
- Erfindungsgemäß wird auch eine Verbindung zur Verwendung als Tyrosinkinase-inhibitor und eine pharmazeutische Zusammensetzung, umfassend ein pharmazeutisch verträgliches Adjuvanz oder Streckmittel und eine aktive Komponente, wobei die Verbindung und die aktive Komponente jeweils eine Verbindung mit der Formel (I)
- sind, worin Y ein bicyclisches Ringsystem ausgewählt aus (A) und (B)
- ist und R eine Gruppe mit der Formel (a), (b) oder (c) ist
- worin R&sub3; -OH ist;
- R&sub1; ein Wasserstoffatoin, eine C&sub1;-C&sub6;-Alkyl- oder C&sub2;-C&sub6;-Alkanoyl-Gruppe ist;
- R&sub2; ein Wasserstoffatom, ein Halogenatom, eine Cyano- oder eine C&sub1;-C&sub6;- Alkyl-Gruppe ist;
- n Null oder eine ganze Zahl von 1 bis 3 ist, wenn Y das Ringsystem (A) ist, wohingegen n Null, 1 oder 2 ist, wenn Y das Ringsystem (B) ist; oder deren pharmazeutisch verträglichen Salze;
- und worin jeder der Substituenten R, OR&sub1; und R&sub2; unabhängig an jede der beiden Aryl-Gruppen im bicyclischen Ringsystem (A) gebunden sein kann, wohingegen im bicyclischen Ringsystem (B) nur der Benzolrest substituiert sein kann und worin, wenn Y das Ringsystem (A) ist R von der oben definierten Gruppe (a) verschieden ist.
- Die vorliegende Erfindung schließt auch die Verwendung einer Verbindung mit der Formel (I)
- ein, worin Y ein bicyclisches Ringsystem ausgewählt aus (A) und (B) ist
- und R eine Gruppe mit der Formel (a), (b) oder (c) ist
- worin R&sub3; -OH ist;
- R&sub1; ein Wasserstoffatom, eine C&sub1;-C&sub6;-Alkyl- oder eine c&sub2;-C&sub6;-Alkanoyl- Gruppe ist;
- R&sub2; ein Wasserstoffatom, ein Halogenatom, eine Cyano-Gruppe oder eine C&sub1;-C&sub6;-Alkyl-Gruppe ist;
- n Null oder eine ganze Zahl von 1 bis 3 ist, wenn Y das Ringsystem (A) ist, n aber Null, 1 oder 2 ist, wenn Y das Ringsystem (B) ist; oder deren pharmazeutisch verträglichen Salze;
- und worin jeder der Substituenten R, OR&sub1; und R&sub2; unabhängig an jede der beiden Aryl-Gruppen im bicyclischen Ringsystem (A) gebunden sein kann, aber im bicyclischen (B) nur der Benzolrest substituiert sein kann bei der Herstellung eines Medikaments zur Verwendung als Tyrosinkinase-Inhibitor.
- Beispiele für einzelne erfindungsgemäße Verbindungen sind die folgenden Verbindungen, die je nach Bedarf entweder die Z- oder E-Diasteroeomere oder eine Z-, E-Mischung der Diastereomere sind:
- 2-Cyano-3-(2-hydroxynaphth-1-yl)thioacrylamid;
- 2cyano-3-(3-hydroxynaphth-1-yl) thioacrylamid;
- 2-Cyano-3-(4-hydroxynaphth-1-yl)thioacrylamid;
- 2-Cyano-3-(1-hydroxynaphth-2-yl)thioacrylamid;
- 2-cyano-3-(3-hydroxynaphth-2-yl)thioacrylamid;
- 2-Cyano-3-(4-hydroxynaphth-2-yl)thioacrylamid;

- Die erfindungsgemäßen Verbindungen und deren pharmazeutisch verträglichen Salze können in einem Verfahren erhalten werden, das die Kondensation eines Aldehyds mit der Formel (II)
- worin
- Y, R&sub1;, R&sub2; und n wie oben definiert sind, mit einer Verbindung der Formel (a'), (b') bzw. (c')
- NC-CH&sub2;-COR&sub3; NC-CH&sub2;-CN NC-CH&sub2;-CSNH&sub2;
- (a') (b') (c')
- worin R&sub3; wie oben definiert ist, umfaßt und falls gewünscht, das Überführen einer Verbindung mit der Formel (I) in eine andere Verbindung mit der Formel (I) und/oder falls gewünscht das Überführen einer Formel (I) in ein pharmazeutisch verträgliches Salz davon und/oder falls gewünscht das Überführen eines Salzes in die freie Verbindung und/oder falls gewünscht das Trennen einer Mischung von Isomeren von einer Verbindung mit der Formel (I) in die einzelnen Isomere. Die Reaktion einer Verbindung mit der Formel (II) mit einer Verbindung mit der Formel (a'), (b') bzw. (c') ist ein analoges Verfahren, das wie im folgenden beschrieben nach bekannten Methoden ausgeführt werden kann, und zwar bevorzugt in Gegenwart eines basischen Katalysators, z.B. Pyridin, Piperidin, Dimethylamin oder eines geeignetes Alkalimetallhydroxids oder Alkoxids.
- Z.B. kann die Reaktion einer Verbindung mit der Formel (II) mit einer Verbindung mit der Formel (a'), (b') bzw. (c') unter den Bedingungen einer Knoevenagel-Reaktion, wie z.B. von G. Jones in Organic Reactions 15, 204 (1967) beschrieben, erfolgen. Geeignete Katalysatoren sind organische Basen wie Pyridin, Piperidin oder Diethylamin. Die Kondensation kann in einem inerten organischen Lösungsmittel, z.B. Pyridin, Ethanol, Methanol, Benzol oder Dioxan bei einer Temperatur zwischen 0ºC bis 100ºC ausgeführt werden. Bevorzugt wird die Reaktion in warmer ethanolischer Lösung in Gegenwart eines Piperidin-Katalysators ausgeführt.
- Eine Verbindung mit der Formel (I) kann in eine andere Verbindung mit der Formel (I) nach bekannten Methoden überführt werden. Z.B. kann eine Verbindung mit der Formel (I), worin einer oder mehrere R&sub1;-Substituenten C&sub1;-C&sub6;-Alkyl-Gruppen sind, nach in der organischen Chemie bekannten Methoden de-etherifiziert werden, um eine Verbindung mit der Formel (I) zu erhalten, worin einer oder mehrere R&sub1;-Substituenten Wasserstoffatome sind. Im Falle eines phenolischen Methylethers kann die Spaltung z.B. mit Bortribromid wie von J. F. N. McOmie in Tetrahedron 24, 2289 (1968) beschrieben, erfolgen. Es ist ratsam, etwa 1 mol Bortribormid pro Ether-Gruppe zusammen mit einem zusätzlichen Mol des Reagenzes für jede Gruppe mit einem potentiell basischen Stickstoff- oder Sauerstoffatom zu verwenden. Die Reaktion kann in einem inerten organischen Lösungsmittel wie Methylenchlorid, Pentan oder Benzol unter einer Inertgas-Atmosphäre z.B. Stickstoff, bei einer Temperatur zwischen -78ºC bis Raumtemperatur erfolgen.
- Die Acylierung einer Verbindung mit der Formel (I), worin einer oder mehrere R&sub1;-Substituenten Wasserstoffatome sind, um die entsprechende Verbindung mit der Formel (I) zu erhalten, worin einer oder mehrere R&sub1;- Substituenten eine C&sub2;-C&sub6;-Alkanoyl-Gruppe ist, kann durch Reaktion mit dem entsprechenden reaktiven Derivat einer geeigneten Carbonsäure, wie einem Anhydrid oder einem Halogenid in Gegenwart einer Base erfolgen, wobei die Temperatur zwischen etwa 0ºC und etwa 50ºC liegt. Bevorzugt wird die Acylierung durch Reaktion mit dem entsprechenden Anhydrid in Gegenwart einer organischen Base wie Pyridin ausgeführt.
- Die mögliche Bildung eines Salzes einer Verbindung mit der Formel (I) sowie die Überführung eines Salzes in die freie Verbindung und die Trennung der Isomeren-Mischung in die einzelnen Isomere können nach herkömmlichen Methoden durchgeführt werden.
- Z.B. kann die Trennung einer Mischung der geometrischen Isomeren, z.B. der cis- und trans-Isomeren durch fraktionierte Kristallisation aus einem geeigneten Lösungsmittel oder durch Chromatographie, d.h. entweder Säulenchromatographie oder Hochdruck-Flüssigkeitschromatographie erfolgen.
- Die Verbindungen mit der Formel (II) können nach bekannten Methoden aus Verbindungen mit der Formel (III) erhalten werden
- worin Y, R&sub1;, R&sub2; und n wie oben definiert sind.
- Z.B. kann eine phenolische Verbindung mit der Formel (III) mit Chloroform und Alkalihydroxiden in einer wäßrigen oder einer wäßrigen alkoholischen Lösung nach der bekannten Reimer-Thiemann-Methode behandelt werden. Wenn ein aromatischer Methylether als Edukt verwendet wird, kann die von N. S. Narasimhan et al. in Tetrahedron 31, 1005 (1975) beschriebene Methode angewendet werden. Entsprechend wird der Methylether mit der Formel (III) mit Butyllithium unter Reflux in Ether lithiiert. Die Behandlung der organometallischen Verbindung mit N-Methylformanilid liefert das Foryml- Derivat. Die Verbindungen mit der Formel (III) sind bekannt, oder können nach bekannten Methoden aus bekannten Verbindungen erhalten werden.
- Die erfindungsgemäßen Verbindungen besitzen eine spezifische Tyrosinkinase-inhibierende Wirkung. Daher können sie in der Behandlung von Krebs und anderen pathologischen proliferativen Zuständen verwendet werden.
- Neuerliche Untersuchungen der molekularen Grundlage von neoplastischen Transformationen haben eine Gruppe von Genen identifiziert, Onkogene genannt, deren irrläuferhafte Expression zur Tumorneubildung führt.
- Z.B. besitzen RNA-Tumorviren solch eine onkogene Sequenz, deren Expression die neoplastischen Umwandlungen der infizierten Zellen bestimmt. Mehrere dieser onkogenen-endkodierten Proteine wie pp60v-src, p70gag-yes, p130gas-fps und p70gag-fgr zeigen Proteintyrosinkinase-Aktivität, da sie den Transfer des y-Phosphats von Adenosintriphosphat (ATP) auf Tyrosin- Reste in Protein-Substraten katalysieren. In normalen Zellen zeigen mehrere Wachstumsfaktor Rezeptoren z.B. die Rezeptoren für PDGF, EGF, α-TGF und Insulin Tyrosinkinase-Aktivität. Die Bindung des Wachstumsfaktors (GF) aktiviert den Tyrosinkinase-Rezeptor zur Autophosphorylierung und zur Phosphorylierung benachbarter Moleküle im Tyrosin.
- Es wird daher angenommen, daß die Phosphorylierung dieser Tyrosinkinase-Rezeptoren eine wichtige Rolle bei der Signalübertragung spielt und daß die Hauptfunktion der Tyrosinkinase-Aktivität in normalen Zellen die Regulierung des Zellwachstums ist. Die Störung dieser Aktivität durch onkogene Tyrosinkinase, die entweder zuviel produziert wurde und/oder eine veränderte Substratspezifität zeigt, kann zum Verlust der Wachstumskontrolle und/oder der neoplastischen Transformation führen. Entsprechend kann ein spezifischer Inhibitor der Tyrosinkinasen für die Untersuchung des Mechanismus in der Carcinogenese, der Zellproliferation und der Differenzierung nützlich sein und er kann wirksam in der Vorbeugung und Chemotherapie von Krebs und anderen pathologischen proliferativen Zuständen sein. Die Tyrosin-spezifische Proteinkinase-Aktivität dieser Verbindungen ist z.B. durch die Tatsache gezeigt daß sie in dem von B. Ferguson et al., in J. Biol. Chem. 1985, 260, 3652 beschriebenen in vitro-Test aktiv sind.
- Das verwendete Enzym ist die Abelson-Tyrosinkinase p 60v-abl. Deren Herstellung und Isolierung erfolgt gemäß einer modifizierten Methode von B. Ferguson et al. (ibidem). Als Substrat wird α-Casein oder (Val&sup5;)- Angiotensin verwendet. Der Inhibitor wird mit dem Enzym 5 min bei 25ºC vorinkubiert. Die Reaktionsbedingungen sind: 100 mM MOPS-Puffer, 10 mM MgCl&sub2;, 2 uM (γ-³²P) ATG (6Ci/mmol), 1 mg/ml α-Casein [ein alternatives Substrat ist (Val&sup5;) Angiotensin II] und 7,5 ug/ml Enzym in einem Gesamtvolumen von 30 ul und bei einem pH von 7,0.
- Die Reaktion wird 10 min bei 25ºC inkubiert. An die Trichloressigsäure-Fällung des Proteins schließt sich die rasche Filtration und Quantifizierung des phosphorylierten Substrats durch einen Flüssigkeitsgeigerzähler an. Alternativ wird die Reaktionsmischung einer Natriumdodecylsulfatpolyacrylamid-Elektrophorese unterworfen und das phosphorylierte Substrat wird durch Autoradiographie oder P³²-Messung des ausgewählten Punktes bestimmt.
- Aufgrund der hohen Aktivität und geringen Toxizität der erfindungsgemäßen Verbindungen können diese sicher in der Medizin eingesetzt werden.
- Z.B. ist die ungefährer akute Toxizität (LD&sub5;&sub0;) der erfindungsgemäßen Verbindung in Mäusen nach einmaliger Verabreichung einer gesteigerten Dosis und Bestimmung am 7. Tag nach der Behandlung vernachlässigbar. Die erfindungsgemäßen Verbindungen können in einer Reihe von Darreichungsformen verabreicht werden, d.h. z.B. oral in Form von Tabletten, Kapseln, Zucker oder filmbeschichteten Tabletten, flüssigen Lösungen oder Suspensionen, rektal in Form von Zäpfchen, parenteral z.B. intramuskulär oder durch intravenöse Injektion oder Infusion oder topisch.
- Die Dosierung hängt vom Alter, vom Gewicht, vom Zustand des Patienten und vom Administrationsweg ab, .B. kann die orale Administration bei erwachsenen Menschen im Bereich von etwa 10 bis etwa 150 - 200 mg pro Dosis 1 bis 5mal täglich erfolgen.
- Natürlich kann die Darreichungsform zum Erreichen einer optimalen therapeutischen Wirkung angepaßt werden.
- Die erfindungsgemäßen pharmazeutischen Zusammensetzungen werden normalerweise nach herkömmlichen Methoden hergestellt und in einer pharmazeutisch geeigneten Form verabreicht.
- Z.B. können feste orale Formen neben der aktiven Verbindung Streckmittel, z.B. Lactose, Dextrose, Saccharose, Cellulose, Maisstärke oder Kartoffelstärke, Gleitmittel, z.B. Silica, Talk, Stearinsäure, Magnesium oder Calciumstearat und/oder Polyethylenglykole, Bindemittel, z.B. Stärken, Gummi arabicum, Gelatine, Methyicellulose, Carboxymethylcellulose oder Polyvinylpyrrolidon; aggregationshemmende Mittel, z .B. Stärke, Alginatsäuren, Alginate oder Natriumstärkeglykolate, Brausemischungen; Farbstoffe; Süßstoffe; Netzmittel wie Lecithin, Polysorbate, Laurylsulfate; und allgemein nicht-toxische und pharmakologisch inaktive Substanzen enthalten, die in pharmazeutischen Formulierungen Verwendung finden. Die pharmazeutischen Zubereitungen können auf bekannte Art hergestellt werden, z.B. durch Mischen, Granulieren, Tablettieren, Zuckerbeschichten oder Filmbeschichten.
- Die flüssigen Dispersionen zur oralen Verabreichung können z.B. sirupe, Emulsionen und Suspensionen sein.
- Der Sirup kann als Träger z.B. Saccharose oder Saccharose mit Glycerin und/oder Mannitol und/oder Sorbitol enthalten. Die Suspensionen und Emulsionen können als Träger z.B. natürliches Gummi, Agar, Natriumalginat, Pectin, Methylcellulose, Carboxymethylcellulose oder Polyvinylalkohol enthalten. Die Suspensionen oder Lösungen zur intramuskulären Injektion können zusammen mit der aktiven Verbindung einen pharmazeutisch verträglichen Träger, z.f3. steriles Wasser, Olivenöl, Ethyloleat, Glykole, z.B. Propylenglykol und, falls gewünscht eine geeignete Menge an Lidocainhydrochlorid enthalten. Die Lösungen zur intravenösen Injektion oder Infusion können als Träger z.B. steriles Wasser enthalten oder bevorzugt liegen sie in Form von sterilen wäßrigen isotonischen Salz läsungen vor.
- Die Zäpfchen können zusammen mit der aktiven Verbindung einen pharmazeutisch verträglichen Träger, z.B. Kakaobutter, Polyethylenglykol, ein Polyoxyethylensorbitanfettsäureester-Tensid oder Lecithin enthalten.
- Zusammensetzung zur topischen Verwendung, z.B. Cremes, Lotions oder Pasten können durch Mischen der aktiven Komponente mit einem herkömmlichen ölartigen oder emulsifizierenden Träger hergestellt werden. Die folgenden Beispiele illustrieren die Erfindung, beschränken sie jedoch nicht.
- Eine Lösung von 5-Formyl-8-hydroxychinolin (173 mg, 1 mmol), Cyanoacetamid (92 mg, 1,1 mmol) und Piperidin (60 mg, 0,7 mmol) in absolutem Ethanol (20 m1) wurde 4 h auf 50ºC erwärmt. Die Reaktionsmischung wurde auf 0 bis 5ºC abgekühlt, das Präzipitat abfiltriert, der Rückstand mit eisgekühltem Ethanol gewaschen und dann im Vakuum getrocknet.
- Die reine Titelverbindung wurde so in 70%iger Ausbeute (167 mg) erhalten. Verbindungen höherer Reinheit, Schmelzpunkt: 275ºC, wurde durch Kristaltisation aus Ethanol erhalten.

- Nach dem oben beschriebenen Verfahren kann die folgende Verbindung hergestellt werden:
- 2-Cyano-3-(1-hydroxy-5,6,7,8-tetrahydronaphth-2-yl)acrylamid.
- Eine Mischung von 2-Hydroxy-1-naphthylaldehyd (172 mg, 1 mmol), 2- Cyanothioacetamid (110 mg, 1 mmol), N,N-Diethylaminoethanol (23 mg, 0,2 mmol) und 15 ml Ethanol wurde 30 min unter Rückfluß und Stickstoff gerührt. Dann wurde die Mischung abgekühlt, das Präzipitat abfiltriert, mit eisgekühltem Ethanol gewaschen und im Vakuumofen getrocknet. So wurde die nahezu reine Titelverbindung in 85%iger Ausbeute (1080 mg) erhalten. Durch Umkristallisation aus Ethanol wurden sehr reine Proben erhalten.

- Nach dem oben beschriebenen Verfahren können die folgenden Verbindungen hergestellt werden:

- 2-Cyano-3-(4-hydroxynaphth-2-yl)thioacrylamid;
- 2-Cyano-3-(2-hydroxy-5,6,7,8-tetrahydronaphth-1-yl)thioacrylamid;
- 2-Cyano-3-(3-hydroxy-5,6,7,8-tetrahydronaphth-1-y1)thioacrylamid;
- 2-Cyano-3-(4-hydroxy-5,6,7,8-tetrahydronaphth-1-yl)thioacrylamid;

- Zu einer Mischung aus 1-Hydroxy-2-naphthylaldehyd (172 mg, 1 mmol) und Cyanoessigsäure (85 mg, 1 mmol) in trockenem Dioxan (2 ml) wurde bei 0 bis 5ºC tropfenweise Piperidin (42 mg, 0,5 mmol) zugegeben. Die Mischung wurde über Nacht bei Raumtemperatur gelagert. Die gebildeten Kristalle wurden abfiltriert und aus Chloroform umkristallisiert. So wurden 200 mg der reinen Titelverbindung erhalten, was 90 % Ausbeute entspricht.

- Nach der oben beschriebenen Methode können ausgehend von den entsprechenden Aldehyd-Derivaten die folgenden erfindungsgemäßen Verbindungen hergestellt werden:
- 2-Cyano-3-(2-hydroxy-5,6,7,8-tetrahydronaphth-1-yl)acrylsäure;
- 2-Cyano-3-(3-hydroxy-5,6,7,8-tetrahydronaphth-1-yl)acrylsäure;
- 2-Cyano-3-(4-hydroxy-5,6,7,8-tetrahydronaphth-1-yl)acrylsäure;
- 2-Cyano-3-(1-hydroxy-5,6,7,8-tetrahydronaphth-2-yl)acrylsäure;
- 2-Cyano-3-(3-hydroxy-5,6,7,8-tetrahydronaphth-2-yl)acrylsäure; und
- 2-Cyano-3-(4-hydroxy-5,6,7,8-tetrahydronaphth-2-yl)acrylsäure.
- Das Edukt für dieses De-etherifizierungs-Beispiel ist 2-Cyano-3-(1- methoxy-5,6,7,8-tetrahydronaphth-2-yl)acrylamid, das nach dem in Referenzbeispiel 1 beschriebenen Verfahren erhalten werden kann.
- Unter Rühren gab man zu einer Lösung aus 2-Cyano-3-(1-methoxy- 5,6,7,8-tetrahydronaphth-2-yl)acrylamid (256 mg, 1 mmol) in wäßrigem Dichlormethan (10 ml) bei -78ºC unter Stickstoff innerhalb von 10 min eine 1,0 M Lösung von Bortribromid in Dichlormethan (3 ml, 3 mmol). Die resultierende Mischung wurde eine weitere Stunde bei -78ºC gerührt und dann ließ man auf Raumtemperatur kommen. Nach 1 1/2 h Rühren bei 20 - 25ºC wurde die Mischung auf -10ºC abgekühlt und dann durch 10 min tropfenweise Zugabe von Wasser (10 ml) queched. Nach Zugabe von Ethylacetat (10 ml) wurde die organische Phase abgetrennt, mit Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuum zur Trockne eingeengt. Der Rückstand wurde aus Ethanol kristallisiert, wobei 169 mg der reinen Titelverbindung erhalten wurden (Ausbeute 70 %).

- Nach dem oben beschriebenen Verfahren können ausgehend von den entsprechenden phenolischen Methylethern die in den Beispielen 1 und 2 erwähnten erfindungsgemäßen Verbindungen erhalten werden.
- Das Edukt für dieses Acylierungsbeispiel ist 2-Cyano-3-(1-hydroxy- 5,6,7,8-tetrahydronaphth-2-yl)acrylamid, das nach dem in Referenzbeispiel 1 beschriebenen Verfahren erhalten werden kann.
- Zu einer gekühlten Lösung von 2-Cyano-3-(1-hydroxy-5,6,7,8-tetrahydronaphth-2-yl)acrylamid (242 mg, 1 mmol) in trockenem Pyridin (0,5 ml) wurde Essigsäureanhydrid (204 mg, 2 mmol) gegeben und die Mischung über Nacht bei 0 bis 5ºC gehalten. Anschließend wurde die Mischung im Vakuum aufkonzentriert, der Rückstand in Dichlormethan gelöst, die organische Phase mit Wasser gewaschen und dann unter verminderten Druck eingedampft. Das Rohprodukt wurde aus Chloroform/Methanol kristallisiert, um die reine Titelverbindung in 90%iger Ausbeute (256 mg) zu erhalten.

- Nach dem oben beschriebenen Verfahren können die in den Beispielen 1 und 2 erhaltenen Phenole in die entsprechenden C&sub2;-C&sub6;-Alkanoyl-Derivate überführt werden.
- 0,150 g schwere Tabletten, die 25 mg aktive Substanz enthalten, können wie folgt hergestellt werden:
- 2-Cyano-3-(1-hydroxynaphth-2-yl)acrylamid 250 g
- Lactose 800 g
- Maisstärke 415 g
- Talkpulver 30 g
- Magnesiumstearat 5 g
- Das 2-Cyano-3-(1-hydroxynaphth-2-yl)acrylamid, die Lactose und die Hälfte der Maisstärke wurden gemischt und die Mischung dann durch ein Sieb mit 0,5 mm Maschenweite gedrückt. Die Maisstärke (10 g) wurde in warmem Wasser (90 ml) suspendiert und die resultierende Paste zur Granulierung des Pulvers verwendet. Das Granulat wurde getrocknet, durch ein Sieb mit 1,4 mm Maschenweite gegeben und dann die restliche Stärke, der Talk und das Magnesiumstearat zugegeben, vorsichtig gemischt und zu Tabletten verarbeitet.
- Kapseln von jeweils 0,200 g, die 20 mg an aktiver Substanz enthalten, können hergestellt werden.

Claims (8)
1. Verbindung mit der allgemeinen Formel (I)
worin
Y ein bicyclisches Ringsystem ausgewahlt aus (A) und (B) ist:
R eine Gruppe mit der Formel (a), (b) oder (c) ist;
worin R&sub3; -OH ist;
R&sub1; ein Wasserstoffatom, eine C&sub1;-C&sub6;-Alkyl- oder eine C&sub2;-C&sub6;-Alkanoyl-
Gruppe ist;
R&sub2; ein Wasserstoffatom, ein Halogenatom, eine Cyano-Gruppe oder eine
C&sub1;-C&sub6;-Alkyl-Gruppe ist;
n Null oder eine ganze Zahl von 1 bis 3 ist, wenn Y das Ringsystem
(A) ist, aber n Null, 1 oder 2 ist, wenn Y das Ringsystem (B) ist; oder
deren pharmazeutisch verträglichen Salze;
und worin jeder der Substituenten R, OR&sub1; und R&sub2; unabhängig
voneinander an jeden der beiden Ringe des bicyclischen Ringsystems (A)
gebunden sein kann, wohingegen im bicyclischen Ringsystem (B) nur der
Benzolrest substituiert sein kann und worin, wenn Y das Ringsystem (A) ist,
R von den obigen Gruppen (a) oder (b) verschieden ist.
2. Verbindung mit der Formel (I) gemäß Anspruch 1, worin R&sub1; ein
Wasserstoffatom, eine C&sub1;-C&sub4;-Alkyl- oder C&sub2;-C&sub4;-Alkanoyl-Gruppe ist und R&sub2;
ein Wasserstoffatom ist, oder deren pharmazeutisch verträglichen Salze.
3. Verbindung mit der Formel (I) gemäß Anspruch 1, worin R&sub1; und R&sub2;
Wasserstoffatome sind, oder deren pharmazeutisch verträgliche Salze.
4. Verbindung ausgewählt aus der Gruppe bestehend aus den folgenden
Verbindungen, wobei diese je nach dem entweder das Z- oder E-Diastereomere
oder Z,E-Mischungen der Diastereomeren sind:
2-Cyano-3-(2-hydroxynaphth-1-yl)thioacrylamid;
2-Cyano-3-(3-hydroxynaphth-1-yl)thioacrylamid;
2-Cyano-3-(4-hydroxynaphth-1-yl)thioacrylamid;

5. Verfahren zur Herstellung von Verbindungen mit der Formel (I) oder
deren pharmazeutisch vertragliche Salze, gemaß Anspruch 1, umfassend das
Kondensieren eines Aldehyds mit der Formel (II)
worin Y, R&sub1;, R&sub2; und n wie in Anspruch 1 definiert sind, mit einer
Verbindung mit der Formel (a'), (b') bzw. (c'):
und falls gewünscht, Überführen der Verbindung mit der Formel (I) in
ein pharmazeutisch verträgliches Salz davon und/oder falls gewünscht,
Überführen eines Salzes in die freie Verbindung und/oder falls gewünscht
Trennen der Isomeren-Mischung einer Verbindung mit Formel (I) in die
einzelnen Isomere.
6. Pharmazeutische Zusammensetzung, umfassend ein pharmazeutisch
verträgliches Adjuvanz oder Streckmittel und als aktive Komponente eine
Verbindung mit der allgemeinen Formel (I)
worin Y ein bicyclisches Ringsystem ausgewählt aus (A) und (B)
und R eine Gruppe mit der Formel (a), (b) oder (c) ist
worin R&sub3; -OH ist;
R&sub1; ein Wasserstoffatom, eine C&sub1;-C&sub6;-Alkyl-oder C&sub2;-C&sub6;-Alkanoyl-Gruppe
ist;
R&sub2; ein Wasserstoffatom, ein Halogenatom, eine Cyano-oder eine C&sub1;-C&sub6;-
Alkyl-Gruppe ist;
n Null oder eine ganze Zahl von 1 bis 3 ist, wenn Y das Ringsystem
(A) ist,
aber n Null, 1 oder 2 ist, wenn Y das Ringsystem (B) ist;
oder deren pharmazeutisch verträglichen Salze;
und worin jeder der Substituenten R, OR&sub1; und R&sub2; unabhängig an jeden
der beiden Ringe des bicyclischen Ringsystems (A) gebunden sein kann,
wohingegen im bicyclischen Ringsystem (B) nur der Benzol-Rest substituiert
sein kann und worin wenn Y das Ringsystem (A) ist, R von der oben
definierten Gruppe (a) verschieden ist.

R&sub1; ein Wasserstoffatom, eine C&sub1;-C&sub6;-Alkyl-oder C&sub2;-C&sub6;-Alkanoyl-Gruppe

R&sub2; ein Wasserstoffatom, ein Halogenatom, eine Cyano-oder eine C&sub1;-C&sub6;-
Alkyl-Gruppe ist;
n Null oder eine ganze Zahl von 1 bis 3 ist, wenn Y das Ringsystem
(A) ist,
aber n Null, 1 oder 2 ist, wenn Y das Ringsystem (B) ist;
oder deren pharmazeutisch verträgliche Salze;
und worin jeder der Substituenten R, OR&sub1; und R&sub2; unabhängig an jeden
der beiden Ringe des bicyclischen Ringsystems (A) gebunden sein kann,
