Description
- Die vorliegende Erfindung betrifft Verbindungen, die eine Bindungsaffinität für sowohl den ανβ3-Rezeptor und einen Neuropeptidrezeptor, insbesondere den Somatostatinrezeptor, aufweisen.
- Der Intergrin ανβ3-Rezeptor wird vorherrschend in wachsenden und wandernden Endothelzellen exprimiert und wurde als ein Marker des angionetischen Phänotyps von vaskulären Zellen während z. B. der Tumorangiogenese identifiziert. Das ανβ3-Integrin selbst wird ebenfalls durch Tumorzellen exprimiert. Der ανβ3-Rezeptor ist daher ein potentielles Target für Tumor-gerichtete Moleküle.
- Ein weiterer Typ von Rezeptor, der in Tumoren auftritt, sind die Somatostatinrezeptoren. Ihre Anwesenheit wurde in einer Vielzahl von Tumoren und auch auf Immunzellen durch klassische biochemische Bindungstechniken, Autoradiographie, in situ-Hybridisierung und RT-PCR gezeigt. Die Bindung eines Liganden an den Somatostatinrezeptor führt meistens zur Internalisierung des Liganden/Somatostatin-Rezeptorkomplexes.
- Der natürliche Ligand für die Somatostatinrezeptoren ist Somatostatin, ein 14- oder 28-Aminosäure-Neuropeptid, das mit hoher Affinität an alle 5 Somatostatinrezeptorsubtypen (SST) bindet. Somatostatin wird im Plasma schnell abgebaut, jedoch wurden enzymatischer Abbau-stabile Somatostatinanaloga entwickelt. Die klinisch am meisten verwendeten Analoga sind Octreotid und Lantreotid, diese Verbindungen binden mit hoher Affinität an die SST 2, 3 und 5.
- Es wurde gemäß der Erfindung vorgesehen, Liganden, die eine Affinität für beide Typen von Rezeptoren aufweisen, in einer Verbindung zu kombinieren, um die Gesamtaffinität der Verbindung für Tumore zu verbessern.
- Zu diesem Zweck betrifft die Erfindung Verbindungen, die eine Bindungsaffinität für sowohl den ανβ3-Rezeptor und einem Neuropeptidrezeptor, insbesondere den Somatostatinrezeptor aufweisen, wobei die Verbindung einen ersten Peptidteil umfaßt, der mindestens einmal die Aminosäuresequenz Arg-Gly-Asp umfaßt, und einen zweiten daran gekoppelten gegebenenfalls über einen Linker Peptidteil, wobei der zweite Peptidteil ein Neuropeptid ist.
- Sowohl
EP 0 385 659 (Fujita Health University) und ein Artikel von Vella et al. (J. Peptide Res. 54, 1999, 415–426) betreffend die Insertion der RGD-Sequenz in multifunktionelle Polypeptide, um neue Polypeptide zu produzieren, die zusätzliche Zell-adhesive Aktivität aufweisen. Daher unterscheiden sich sowohl der Zweck und die Ausgangssubstanzen fundamental von denjenigen der vorliegenden Erfindung, die als ihre Aufgabe „die Verbesserung der Gesamtaffinität für Tumore" aufweist und worin kleine Neuropeptide als das Ausgangsmaterial verwendet werden. Eine Publikation von De Jong et al. (Quart J. Nucl. Med. 43, 1999, 356–366) betrifft die Therapie von neuroendokrinen Tumoren mit radiomarkierten Somatostatinanaloga und macht keinerlei Vorschlag zur Kopplung eines RGD-umfassend Peptidteils an solch ein Neuropeptid. - Die vorliegende Erfindung, wie oben definiert, ist nicht nur auf Somatostatin anwendbar, sondern auch auf andere Neuropeptide. Konsequenter Weise ist der zweite Peptidteil ausgewählt aus der Gruppe bestehend aus CCK, Gasttin, Substanz P, Bombesin, VIP (Vasoaktives intestinales Peptid), PACAP (pituitäre Adenylatcyclase-aktivierendes Peptid), Somatostatin und Analoga von diesen. Die Analoga können modifizierte Versionen des Ausgangspeptids sein, um die Stabilität oder Aktivität zu verbessern oder können Teile der Peptide sein, die immer noch ihre biologische Aktivität beibehalten.
- In einer bevorzugten Ausführungsform ist der zweite Peptidteil ein Somatostatinanalogon, bevorzugterweise ausgewählt aus der Gruppe bestehend aus Octreotat, Octreotid, Lantreotid, Vapreotid oder Derivaten davon.
- Der erste Peptidteil ist ein sogenanntes RGD-Peptid, was ein Peptid ist, das mindestens einmal das Arg-Gly-Asp-Motiv aufweist. Analoga dieses Ausgangs-RGD-Peptids können zusätzliche Aminosäuren umfassen, wie z. B. Tyrosin für die Jodierung. In einer bevorzugten Ausführungsform des RGD-Analogons ist ein zusätzliches Asp zwischen dem Tyr und dem Linker vorhanden. Dieses Asp dient für die Zyklisierung des RGD-Peptidteils, um es stabiler zu machen.
- Ein geeigneter Linker ist z. B. Lysin, das zwei NH2-Gruppen aufweist. Eine von diesen kann zur Kopplung an das RGD-Peptid verwendet werden, während die COOH-Gruppe für die Kopplung an das Neuropeptid verwendet wird. Die verbleibende NH2-Gruppe kann dann zur Kopplung an einen Chelator verwendet werden. Der Chelator wird zur Komplexierung eines (radioaktiven) Markers verwendet.
- Radiomarkieren dieser Neuropeptid-RGD-Verbindungen, entweder direkt oder über einen Chelatbildner (mit oder ohne Spacer) macht diese Verbindungen als Radiodiagnostika oder Radiopharmazeutika geeignet. Geeignete Isotope für die Radiomarkierung sind die folgenden: 213Bi, 186Re, 188Re, 77AS, 90Y, 66Ga, 67Cu, 169Er, 114mIn, 117mSn, 121Sn, 127Te, 142Pe, 143Pr, 198Au, 199Au, 149Tb, 109Pd, 165Dy, 149Pm, 151Pm, 153Sm, 157Gd, 159Gd, 166Ho, 172Tm, 169Yb, 175Yb, 177Lu, 105Rh, 103mRh, 195mPt, 111Ag, 124J, 131J und 211At, 99mTc, 203Pb, 67Ga, 68Ga, 72As, 111In, 113mIn, 97Ru, 62Cu, 64Cu, 52Fe, 52mMn, 51Cr, 123J, 131J, 75Br, 76Br, 77Br und 82Br. Ein Beispiel solch einer radiomarkierten Verbindung ist die 111In-DTPA-Somatostatinanalogon-RGD-Verbindung.
- An Somatostatinanaloga gekoppelte RGD-Peptide (oder andere Neuropeptide und deren Analoga) können anbinden an und in die Zelle eindringen über entweder den RGD-Rezeptor (ανβ3) oder einen der Somatostatinrezeptoren. Von der Verbindung, die an zwei verschiedene Rezeptoren bindet, nämlich dem Neuropeptidrezeptor und dem ανβ3-Integrin kann daher erwartet werden, diese Verbindung auf verschiedenen Zielzellen, wie z. B. Tumorzellen zu finden, sowie auf Zellen der Tumorvaskularisierung. Dieses kann zu einem höheren Ziel/Hintergrundverhältnis beitragen.
- Ein Prototyp für Verbindungen der Erfindung ist RGD-Octreotid. Octreotid ist ein stabiles Peptid (gegenüber Plasmaabbau resistent), das an den Somatostatinrezeptor (SST), Subtypen 2, 3 und 5, bindet. Andere Verbindungen sind wie in den Beispielen beschrieben.
- Es wurde in Audioradiographieexperimenten mit Geweben, die entweder SST-Rezeptoren oder ανβ3-Rezeptoren aufweisen gefunden, daß, obwohl beide Ausgangspeptide in einer neuen Verbindung kombiniert sind, beide ihre Bindungsaffinität für ihre eigenen Rezeptoren beibehalten. Die Bindung der neuen Verbindung an die jeweiligen Rezeptoren könnte mit einem Überschuß der verschiedenen kompetetiven Analoga blockiert werden.
- Die vorliegende Erfindung wird in den folgenden Beispielen weiter verdeutlicht, und diese sind in keiner Weise dazu gedacht, die Erfindung zu begrenzen.
- Beispiele
- Beispiel 1
- Allgemeines Verfahren zur Synthese von RGD-Neuropeptidkonjugaten und deren entsprechenden DTPA- oder DOTA-Derivaten
- Das Folgende ist ein generelles Verfahren zur Herstellung von Verbindungen der Erfindung.
- Festphasenpeptidsynthese (SPPS) wird unter Verwendung eines PE-Biosystems „Pioneer"-Synthesizers unter der Verwendung der Fmoc-Strategie durchgeführt. Ein lineares Peptid, das aus allen Aminosäuren der Verbindung besteht, wird in einem 0,1 mmol Maßstab mit Fmoc-AA1 (OtBu)-PEG-PS (PE Biosystems, 0,18 mmol/g Beladung) präpariert, wobei AA1 die C-terminale Aminosäure ist, als das Ausgangsharz. Fmoc-geschützte Aminosäuren (0,4 mmol) werden mit N[(Dimethylamino)-1H-1,2r3-triazolo[4,5-b]pyridin-1-yl-methylen]-N-methylmethanaminiumhexafluorophosphat-N-oxid (HATU) aktiviert. Alle Aminosäuren und Peptidsynthesereagenzien wurden käuflich erworben.
- Die „on-board" Amidzyklisierung des Peptids wird unter Verwendung des „Allyl-Deblockierungs"-Protokolls (Pd(PPh3)4, N-Methylmorpholin, Essigsäure, Chloroform) erhalten, gefolgt von 7-Azabenzotriazol-1-yl-oxytris(pyrrolidino)-phosphoniumhexafluor-phosphat (PyAOP)-Aktivierung. Das Harz, das das geschützte, zyklisierte Peptid enthält, wird dann von dem Instrument entfernt.
- Das Harz wird in 8 ml an Dimethylformamid, enthaltend 92 mg Thalliumbifluoracetat, suspendiert. Das Gemisch wird für 2–3 Stunden geschüttelt, filtriert, nacheinander mit 10 ml DMF, 10 ml DMF-Wasser (1 : 1), 10 ml von DMF und THF gewaschen, um an das Harz geheftetes geschütztes Peptid (III) zu erhalten. Das Harz wird dann in zwei Teile aufgeteilt.
- Das Peptid wird von dem Harz abgespalten und unter der Verwendung von 85 TFA/5% Wasser/5% Phenol/5% Thianisol für 10 bis 12 Stunden entschützt. Das Rohpeptid wird durch Ausfällen mit t-Butylmethylether isoliert, gefolgt von Zentrifugieren und Reinigen durch reverse Phase HPLC unter der Verwendung von einem Acetonitril/Wasser-Gradienten, der 0,1% TFA enthielt (Lösungsmittel A: 0,1% TFA/H2O, Lösungsmittel B: 0,1% TFA/10% H2O/CH3CN; Gradient: Halten bei 95% A/5% B für 2,0 Minuten, gefolgt von Lösungsmittel A (100%) zu 50%A : 50%B über eine Zeitdauer von 20 Minuten hinweg).
- Die Mtt-Schutzgruppe des Lysins wird durch Behandlung mit 5% TFA/5% Triisopropylsilan (TIPS)/90% Dichlormethan (2 × 30 Minuten) entfernt. Das Harz wird mit Dichlormethan und Tetrahydrofuran gewaschen und in DMF (2,5 ml) resuspendiert, das DIEA (35 μl, 0,2 mmol) enthielt. In einen getrennten Gefäß wird Tris-t-butyl-DTPA-anhydrid oder DOTA (112 mg, 4,2 mmol) in DMF, enthaltend HBTU/HOBt (0,2 mmol, 1,0 ml einer 0,2 mmol/ml Lösung) und DIEA (35 μl, 0,2 mmol) gelöst, um eine 5 ml Lösung zu ergeben. Nach Schütteln für eine Stunde wird das aktivierte DTPA-Derivat zu dem vorhergehend suspendierten Harz hinzugefügt.
- Der Reaktion wird es ermöglicht über Nacht fortzufahren, bevor das Harz mit DMF und THF gewaschen wird.
- Das Peptid wurde von dem Harz abgespalten und t unter der Verwendung von 85% TFA/5% Wasser/5% Phenol/5% Thianisol für 10 bis 12 Stunden entschützt. Das Rohpeptid wird durch Ausfällen mit t-Butylmethylether isoliert, gefolgt von Zentrifugation, und gereinigt durch reverse Phase HPLC unter der Verwendung eines Azetonitril/Wasser-Gradienten, der 0,1% TFA enthielt (Lösungsmittel A: 0,1% TFA/H2O, Lösungsmittel B: 0,1% TFA/10% H2O/CH3CN; Gradient: Halten bei 95% A/5% B für 2,0 Minuten, gefolgt von Lösungsmittel A zu B über eine Zeitdauer von 20 Minuten hinweg).
- Beispiel 2
- Synthese von RGD-Octreotat (IV) und dem entsprechenden DTPA-Derivat (V)
- In Übereinstimmung mit dem wie in Beispiel 1 beschriebenen Verfahren wurden ein RGD-Octreotat und sein entsprechendes DTPA-Derivat gemäß dem folgenden Reaktionsschema präpariert:
- Die Massenspektrometerdaten für das RGD-Octreotat-Konjugat (IV) sind wie folgt: Berechnet 1766,6, Gefunden: 884,8 ((M + 2)/2).
- Die Massenspektrometerdaten für das DTPA-Derivat des RGD-Octreotat-Konjugats (V) sind wie folgt: Berechnet: 2139,9, Gefunden: 1071,2 ((M + 2)/2).

- In Übereinstimmung mit den Verfahren wie in Beispiel 1 beschreiben wurden ein RGD-Octreotid und sein entsprechendes DTPA-Derivat gemäß dem folgenden Reaktionsschema präpariert:
Claims (7)
- Verbindungen, die eine Bindungsaffinität für sowohl den ανβ3-Rezeptor als auch einen Neuropeptidrezeptor, insbesondere den Somatostatinrezeptor, aufweisen und konsequenterweise eine verbesserte Gesamtaffinität für Tumore aufweisen, wobei die Verbindungen einen ersten Peptidteil, der mindestens einmal die Aminosäuresequenz Arg-Gly-Asp umfaßt und einen daran, gegebenenfalls über einen Linker, gekoppelten zweiten Peptidteil umfassen, wobei der zweite Peptidteil ein Neuropeptid ist, das ausgewählt ist aus der Gruppe bestehend aus CCK, Gastrin, Substanz P, Bombesin, VIP (vasoaktives intestinales Peptid), PACAP (pituitäres Adenylatcyklase-aktivierendes Peptid), Somatostatin und Analoga von diesen.
- Verbindungen wie beansprucht in Anspruch 1, wobei der zweite Peptidteil ein Somatostatin-Analogon ist.
- Verbindungen wie beansprucht in Anspruch 2, wobei das Somatostatin-Analogon ausgewählt ist aus der Gruppe bestehend aus Octreotat, Octreotid, Lantreotid, Vapreotid oder Derivate davon.
- Verbindungen wie beansprucht in den Ansprüchen 1–3, wobei der Linker mindestens ein Lysin umfaßt.
- Verbindungen wie beansprucht in den Ansprüchen 1–4, wobei die Verbindung radiomarkiert ist.
