2-(氨基)乙基甲基砜盐酸盐制备抗肿瘤药物的重要中间体。目前原有的合成方法,由含砜基的氰基化合物经乙硼烷选择性还原,再与氯化氢成盐得到。用乙硼烷还原成本高,不适合规模化生产。本发明专利涉及一种2-(N,N-二苄基氨基乙基)甲基砜及其盐,用其制备2-(氨基)乙基甲基砜及其盐操作简便,适合于工业生产。

一种2-(N,N-二苄基氨基乙基)甲基砜及其盐
技术领域
本发明涉及一种2-(N,N-二苄基氨基乙基)甲基砜及其盐,用于规模化制备2-(氨基)乙基甲基砜及其盐。
背景技术
2-(氨基)乙基甲基砜是合成某些具有抗肿瘤药物活性成分的重要中间体。文献报道2-(氨基)乙基甲基砜合成路线如下:
制备得到盐酸盐。制备过程规模化工业生产操作困难,成本高。
发明内容
针对以上路线制备的2-(氨基)乙基甲基砜盐酸盐难以规模化生产的缺点,本发明涉及一种2-(N,N-二苄基氨基乙基)甲基砜及其盐,由该类化合物经催化加氢,可方便地制备2-(氨基)乙基甲基砜及其盐。2-(N,N-二苄基氨基乙基)甲基砜及其盐结构如下:
在醇、酸、醚等有机溶剂的碱溶液,加入2-(N,N-二苄基氨基乙基)甲基砜或它的盐,加入Raney镍或钯碳,在一定压力下氢化还原,即得到2-(氨基)乙基甲基砜或它的盐。反应式如下:
从而分别得到2-(氨基)乙基甲基砜不同的盐。
实施例:
2-(氨基)乙基甲基砜的制备
1升压力釜中加入400毫升乙醇,加入60克2-(N,N-二苄基氨基乙基)甲基砜,15克Raney镍,通氢气至压力20公斤,加热至50度,搅拌还原,TCL跟踪反应至原料点小时。冷却,放出氢气,过滤,浓缩,得2-(氨基)乙基甲基砜油状物。
2-(氨基)乙基甲基砜盐酸盐
将上述得到的2-(氨基)乙基甲基砜溶于100毫升无水乙醇,加入氯化氢的乙醇溶液,至pH2,搅拌1小时,过滤,滤饼用乙醇洗涤,干燥,得11克2-(氨基)乙基甲基砜盐酸盐。

3.一种2-(氨基)乙基甲基砜及其盐的制备方法,为将权利要求1所述的2-(N,N-二苄基氨基)乙基甲基砜及其盐经催化加氢制得,所使用的催化剂为Raney镍、钯碳或二氧化铂。
