Abstract
a presente invenção divulga um polipeptídeo heterodimérico compreendendo um primeiro polipeptídeo que compreende, na direção n-terminal para a c-terminal, pelo menos uma porção de uma região de dobradiça de imunoglobulina que compreende um ou mais resíduos de cisteína, um domínio ch2 de imunoglobulina e um domínio ch3 de imunoglobulina, e um segundo polipeptídeo que compreende, na direção n-terminal para a c-terminal, pelo menos uma porção de uma região de dobradiça de imunoglobulina que compreende um ou mais resíduos de cisteína, um domínio ch2 de imunoglobulina e um domínio ch3 de imunoglobulina, em que o primeiro polipeptídeo compreende as mutações y349c, t366s, l368a e y407v (?cadeia-hole?) e o segundo polipeptídeo compreende as mutações s354c e t366w (?cadeia-knob?); em que o primeiro polipeptídeo (cadeia-hole) compreende as mutações i) i253a ou i253g, e ii)l314a ou l314g ou l314d; e em que o primeiro polipeptídeo e o segundo polipeptídeo estão conectados por uma ou mais ligações de bissulfeto, e em que o domínio ch3 do primeiro polipeptídeo e o domínio ch3 do segundo polipeptídeo se ligam ou não se ligam à proteína a (numeração de acordo com o índice eu de kabat).
