Abstract

"ENSAIOS DE CICLIZAçãO ENZIMáTICA PARA HOMOCISTEìNA E CISTATIONINA". A presente invenção refere-se a um ensaio de ciclização enzimática para avaliação da quantidade de homocisteína e/ou cistationina em uma solução tal como sangue, derivados de sangue ou urina. O ensaio compreende as etapas de contato da solução contendo homocisteína e/ou cistationina para formar uma mistura de reação, com CBS, ou um seu derivado, L-serina, e CBL, ou um seu derivado, por um período de tempo suficiente para catalisar a conversão clínica da forma homocisteína em cistationina e a reconversão de cistationina em homocisteína com a produção de piruvato e amónia; determinação da quantidade de homocisteína e/ou amónia presente na mistura de reação; e determinação da quantidade de homocisteína e/ou cistationina presente na solução baseada na quantidade de piruvato e/ou amónia formada. Vetores de expressão e procedimentos de isolamento para CBS, ou seus derivados, e CBI, ou seus derivados, são também providos, bem como kits de teste para realizar o ensaio. Em modalidades preferidas, os ensaios podem ser realizados em 15 minutos ou menos, com um mínimo de uso de enzima.
