Abstract
La presente se refiere a moléculas de unión a antígeno biespecíficas con unión trivalente a CD40 y unión monovalente a un antígeno de célula diana y a métodos para producir estas moléculas y a métodos para usar las mismas. Reivindicación 1: Una molécula de unión a antígeno biespecífica, que consiste en (a) un primer fragmento Fab capaz de unirse de manera específica a CD40, (b) un segundo fragmento Fab capaz de unirse específicamente a CD40, (c) un tercer fragmento Fab capaz de unirse específicamente a CD40, (d) un dominio Fc compuesto de una primera y una segunda subunidad capaces de asociarse de manera estable, en donde el segundo fragmento Fab (b) está fusionado en el extremo C-terminal de la cadena VH-CH1 al extremo N-terminal de la cadena VH-CH1 del primer fragmento Fab (a), que a su vez está fusionado en su extremo C-terminal al extremo N-terminal de la primera subunidad del dominio Fc y el tercer fragmento Fab (c) está fusionado en el extremo C-terminal de la cadena pesada del Fab al extremo N-terminal de la segunda subunidad del dominio Fc y (e) un fragmento cross-Fab capaz de unirse específicamente a un antígeno de célula diana, en donde el fragmento cross-Fab está fusionado al extremo C-terminal de una de las subunidades del dominio Fc.
