まず、式(I)、式(II)、式(III)等における置換基(例えば、Q、T、X等)について説明する。
Q中の酸素原子及び硫黄原子からなる群より選ばれる少なくとも1種のヘテロ原子を1~2含有する、5若しくは6員飽和ヘテロ環基としては、例えばテトラヒドロフラン-2-イル、テトラヒドロフラン-3-イル、ジオキソラン-2-イル、ジオキソラン-4-イル、テトラヒドロピラン-2-イル、1,4-ジオキサン-2-イル、1,3-ジオキサン-2-イル、1,3-ジオキサン-4-イル、テトラヒドロチオフェン-2-イル、テトラヒドロチオピラン-2-イル、1,3-ジチアン-2-イル、1,4-ジチアン-2-イル、1,4-チオキサン-2-イル、1,4-チオキサン-3-イルなどが挙げられる。
尚、当該ヘテロ環基はアルキルで置換されていてもよい。当該アルキルは炭素数1~6、望ましくは、炭素数1又は2であって、直鎖状又は分枝状のいずれでもよい。
T及びX中のハロゲン又は置換基としてのハロゲンとしては、弗素、塩素、臭素又は沃素の各原子が挙げられ、望ましくは弗素原子又は塩素原子である。置換基としてのハロゲンの置換数は1又は2以上であってよく、2以上の場合、各ハロゲンは同一でも相異なってもよい。また、ハロゲンの置換位置はいずれの位置でもよい。
前記式(I)の化合物は、以下の反応[A]のようにして製造することができる。
反応[A]中、一酸化炭素は、式(II)の化合物1モルに対して、通常1~10当量、望ましくは1~4当量使用することができる。また、圧力は通常0.1~10MPa、望ましくは0.5~5MPaの範囲から選ぶことができる。但し、反応条件によっては、これら範囲外の量や圧力を使用又は選択することもできる。
反応[A]中、式R-OHで表される化合物は、式(II)の化合物1モルに対して、少なくとも1当量以上の量で使用することができる。また、過剰量用いれば溶媒を兼ねることもできる。好ましくは、式(II)の化合物1モルに対して、1~30当量である。
但し、反応条件によっては、この範囲外の量を使用することもできる。
R-OHにおいて、Rとしては炭素数1~6のアルキルであるメチル、エチル、プロピル、イソプロピル、ブチル、tert-ブチル、ペンチル、ヘキシル等が挙げられ、メチルが望ましい。
塩基としては、例えば水酸化ナトリウム、水酸化カリウムのようなアルカリ金属水酸化物;水酸化カルシウム、水酸化マグネシウムのようなアルカリ土類金属水酸化物;炭酸ナトリウム、炭酸カリウムのようなアルカリ金属炭酸塩;重炭酸カルシウム、重炭酸マグネシウムのようなアルカリ土類金属重炭酸塩;重炭酸ナトリウム、重炭酸カリウムのようなアルカリ金属重炭酸塩;酢酸ナトリウム、酢酸カリウムのようなカルボン酸アルカリ金属塩;酢酸カルシウム、酢酸マグネシウムのようなカルボン酸アルカリ土類金属塩;トリメチルアミン、トリエチルアミン、ジイソプロピルアミン、トリイソプロピルアミン、ジイソプロピルエチルアミン、ピリジン、4-ジメチルアミノピリジン、2,6-ジメチルピリジン、4-ピロリジノピリジン、N-メチルモルホリン、N,N-ジメチルアニリン、N,N-ジエチルアニリン、N-エチル-N-メチルアニリン、1,8-ジアザビシクロ〔5.4.0〕-7-ウンデセン、1,4-ジアザビシクロ〔2.2.2〕オクタンのような第三級アミン類などを挙げることができる。なかでもアルカリ金属炭酸塩、アルカリ金属重炭酸塩、カルボン酸アルカリ金属塩、トリエチルアミンなどが望ましい。更にこれらのなかでも、アルカリ金属炭酸塩及びトリエチルアミンが、収率をより向上させる面等において望ましい。アルカリ金属炭酸塩としては、炭酸ナトリウム及び炭酸カリウムが望ましい。前述した塩基は、単独で使用しても、2種以上を混用してもよい。
またパラジウムを含む触媒としては、ホスフィンを配位子とするジクロロビス(トリフェニルホスフィン)パラジウム、ジクロロビス(トリイソプロピルホスフィン)パラジウム、テトラキス(トリフェニルホスフィン)パラジウム、ジクロロ(1,4-ビス(ジフェニルホスフィノ)ブタン)パラジウム、ジクロロビス(トリフェニルホスファイト)パラジウムなども挙げられる。
また、第3級ホスフィンや、第3級ホスファイトを配位子として含まないパラジウム錯体と、第3級ホスフィンや第3級ホスファイトとを反応系中で反応させ、第3級ホスフィンや、第3級ホスファイトを配位子とするパラジウム錯体を発生させて、そのまま触媒として用いることも可能である。第3級ホスフィンや第3級ホスファイトとしては、トリフェニルホスフィン、フェニルジメチルホスフィン、トリ-o-トリルホスフィン、トリ-p-トリルホスフィン、1,2-ビス(ジフェニルホスフィノ)エタン、1,3-ビス(ジフェニルホスフィノ)プロパン、1,4-ビス(ジフェニルホスフィノ)ブタン、トリフェニルホスファイトなどが挙げられる。これらのなかでも、第3級ホスフィンを配位子とするパラジウム錯体を遷移金属触媒として用いることは、収率をより向上させる面等において望ましい。当該遷移金属触媒としては、予め第3級ホスフィンがパラジウムに配位した触媒を用いてもよく、また、第3級ホスフィンを配位子として含まないパラジウム錯体と第3級ホスフィンとを反応系中で反応させ、触媒として用いてもよい。当該第3級ホスフィンとしては、1,2-ビス(ジフェニルホスフィノ)エタン、1,3-ビス(ジフェニルホスフィノ)プロパン及び1,4-ビス(ジフェニルホスフィノ)ブタンが望ましい。
溶媒としては、反応に不活性である限り特に制限はなく、例えばメタノール、エタノール、プロパノール、n-ブタノール、tert-ブタノールのようなアルコール類;ペンタン、ヘキサン、ヘプタン、オクタン、シクロヘキサンのような脂肪族炭化水素類;ベンゼン、トルエン、キシレンのような芳香族炭化水素類;クロロベンゼン、ジクロロベンゼン、ジクロロメタン、ジクロロエタンのようなハロゲン化炭化水素類;ジエチルエーテル、ブチルエチルエーテル、メチルtert-ブチルエーテル、ジメトキシエタンのようなエーテル類;アセトニトリル、プロピオニトリル、N,N-ジメチルホルムアミド、ジメチルスルホキシド、スルホラン、N,N-ジメチルアセトアミド、N-メチルピロリドン、1,3-ジメチル-2-イミダゾリジノンのような極性非プロトン性溶媒;酢酸メチル、酢酸エチル、酢酸プロピルのようなエステル類;アセトン、メチルエチルケトン、メチルイソブチルケトンのようなケトン類;テトラヒドロフラン、ジオキサン、ピリジン、チオフェンのようなヘテロ環式化合物類;水などを挙げることができる。なかでもアルコール類、芳香族炭化水素類、エーテル類、ケトン類、水などが望ましい。前述した溶媒は、単独で使用しても、2種以上を混用してもよい。

酸性化に用いる酸としては、例えば塩酸、硫酸、リン酸、硝酸のような無機酸などが挙げられ、塩酸又は硫酸などが望ましい。
酸の使用量は、反応系内のpHが1.5以下となる添加量であれば任意に設定できる。例えば、式(II)の化合物1モルに対して、通常1~15当量、望ましくは1~6当量使用することができる。但し、反応条件によっては、この範囲外の量を使用することもできる。

反応[A]中、第2工程の加水分解工程の反応温度は、通常0~150℃、望ましくは20~100℃であり、反応時間は、通常0.1~10時間である。また、第2工程の酸性化工程の反応温度は、通常-20~100℃、望ましくは0~50℃であり、反応時間は、通常0.1~5時間である。
反応[A]の第1工程と第2工程は、式(III)の化合物を単離または精製することなく、続けて反応を行うことができる。
前記式(II)の化合物は新規化合物を含み、反応[B]又は反応[C]のようにして製造することができる。
Lは塩素原子、臭素原子又は沃素原子であり、塩素原子又は臭素原子が望ましい。
反応[B]中、式(V)の化合物は、式(IV)の化合物1モルに対して、通常1~5当量、望ましくは1~1.5当量使用することができる。但し、反応条件によっては、この範囲外の量を使用することもできる。
塩基としては、例えば前記反応[A]第2工程で挙げたものと同様のものなどを挙げることができる。なかでもアルカリ金属水素化物、アルカリ金属水酸化物、アルカリ金属炭酸塩、アルカリ土類金属重炭酸塩、トリエチルアミンなどが望ましい。更にこれらのなかでも、アルカリ金属炭酸塩及びトリエチルアミンが、収率をより向上させる面等において望ましい。当該アルカリ金属炭酸塩としては、炭酸ナトリウム及び炭酸カリウムが望ましい。前述した塩基は、単独で使用しても、2種以上を混用してもよい。
溶媒は、式(IV)の化合物に対し、通常0.1~10倍容量使用することができる。但し、反応条件によっては、この範囲外の量を使用することもできる。
添加物としてはヨウ化カリウム、ヨウ化ナトリウム、臭化カリウムのようなアルカリ金属ハロゲン化物;テトラブチルアンモニウムブロミド、テトラエチルアンモニウムヨージド、テトラメチルアンモニウムヒドロキシドのような第4級アンモニウム化合物;18-クラウン-6、15-クラウン-5、ジベンゾ-18-クラウン-6のようなクラウンエーテル類などを挙げることができる。前述した添加物は、単独で使用しても、2種以上を混用してもよい。
反応[B]の反応温度は、通常0~200℃、望ましくは50~150℃であり、反応時間は、通常1~30時間である。
Zは塩素原子、臭素原子又は沃素原子であり、塩素原子又は臭素原子が望ましい。
反応[C]中、式(VII)の化合物は、式(VI)の化合物1モルに対して、通常1~5当量、望ましくは1~1.5当量使用することができる。但し、反応条件によっては、この範囲外の量を使用することもできる。

(1)反応[A]中、Qがジオキソラン-2-イルであり、Tがメチルスルホニルであり、Xが塩素原子であり、Yが臭素原子であり、nが2である、前記式(I)の化合物の製造方法。
(2)反応[B]中、Qがジオキソラン-2-イルであり、Tがメチルスルホニルであり、Xが塩素原子であり、Yが臭素原子であり、nが2であり、Lが臭素原子である、前記式(II)の化合物の製造方法。
(3)反応[C]中、Qがジオキソラン-2-イルであり、Tがメチルスルホニルであり、Xが塩素原子であり、Yが臭素原子であり、nが2であり、Zが塩素原子である、前記式(II)の化合物の製造方法。
(5)反応[E]中、Tがメチルスルホニルであり、Xが塩素原子であり、Yが臭素原子であり、Zが塩素原子である、前記式(IV)の化合物の製造方法。
(6)Qがジオキソラン-2-イルであり、Tがメチルスルホニルであり、Xが塩素原子であり、Yが臭素原子であり、nが2である、前記式(II)の化合物。
(7)Tがメチルスルホニルであり、Xが塩素原子であり、Yが臭素原子である、前記式(IV)の化合物。
これらの化合物の物性(融点、及び1H-NMRの測定値)は、第2表及び第3表に示す通りである。
当該化合物は除草活性を示す有用な化合物である。
第1表中、Phはフェニル基を示す。
製造例1
(1) 1-ブロモ-2,3-ジクロロ-4-(メチルスルホニル)ベンゼン448.9 g(1.41 mol)、トルエン1716 mL、N-メチルピロリドン42.9 mL及び水酸化ナトリウム282 gを混合し、還流下で7時間反応させた。反応液に40~50℃で水2571 mLを投入し、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール ナトリウム塩を完全に溶解させた。その後、トルエンを分液除去し、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール ナトリウム塩水溶液を得た。得られたナトリウム塩水溶液を、40質量%硫酸(760 g)にて酸性化させ、結晶を析出させた。析出した結晶をろ過、水洗、乾燥し、融点157~160℃の3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール401.4 g(純度95.2%, 収率94.9%)を得た。このものの1H-NMRスペクトルデータは以下の通りである。

得られた混合液に水酸化ナトリウム13.6 gを投入し、50℃で1時間反応させた。反応終了後、トルエン層を分液除去し、3-(2-(1,3-ジオキソラン-2-イル)エトキシ)-2-クロロ-4-(メチルスルホニル)安息香酸ナトリウム水溶液を得た。
得られた水溶液を、5~10℃にて、濃塩酸を用いて酸性化し、結晶を析出させた。析出した結晶をろ過し、水洗し、減圧乾燥して、融点142~144℃の3-(2-(1,3-ジオキソラン-2-イル)エトキシ)-2-クロロ-4-(メチルスルホニル)安息香酸81 g(純度89.9%,収率89.3%)を得た。このものの1H-NMRスペクトルデータは以下の通りである。

1-ブロモ-2,3-ジクロロ-4-(メチルスルホニル)ベンゼン31 g(0.1 mol)、トルエン121 mL、ジメチルスルホキシド3 mL及び水酸化ナトリウム20.3 gを混合し、還流下で5時間反応させた。反応後、反応液に水225 mLを投入し、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール ナトリウム塩を完全に溶解させた後、トルエンを分液除去し、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール ナトリウム塩の水溶液を得た。得られたナトリウム塩の水溶液を、濃塩酸にてpH1以下に酸性化し、結晶を析出させた。析出した結晶をろ過し、水洗し、乾燥して、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール27.97g(純度90.75%,収率90.08%)を得た。
前記製造例2において、ジメチルスルホキシドの代わりに1,3-ジメチル-2-イミダゾリジノンを3 mL使用した以外は同様に反応を行った。反応は11時間で完了させ、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール28.13g(純度92.95%,収率92.8%)を得た。
前記製造例2において、トルエン及びジメチルスルホキシドの代わりにtert-ブチルアルコール151 mLを使用し、水酸化ナトリウム使用量を16.3 gとした以外は同様に反応を行った。反応は9時間で完了し、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール29.73g(純度91.84%,収率は95.6%)を得た。
1-ブロモ-2,3-ジクロロ-4-(メチルスルホニル)ベンゼン51 g(0.16 mol)、ピリジン150 mL及び水酸化ナトリウム14.9 gを混合し、還流下で7時間反応させた。反応液に水357 mLを投入し、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール ナトリウム塩を完全に溶解させた後、濃塩酸にてpH1以下に酸性化し、結晶を析出させた。析出した結晶をろ過し、水洗し、乾燥して、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール42.56g(純度94.46%,収率85.6%)を得た。
1-ブロモ-2,3-ジクロロ-4-(メチルスルホニル)ベンゼン5.8 g(0.016 mol)、tert-ブチルアルコール15 mL及び水酸化カリウム2.7 gを混合し、還流下で7時間反応させた。反応液に水34.2mLを投入し、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール カリウム塩を完全に溶解させた後、濃塩酸にてpH1以下に酸性化し、結晶を析出させた。析出した結晶をろ過し、水洗し、乾燥して、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール4.99g(純度73.6%,収率77.6%)を得た。
前記製造例6において、水酸化カリウムの代わりにカリウム‐tert-ブトキシド4.7 gを使用した以外は同様に反応を行った。反応は1時間で完了し、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール4.84g(純度63.28%,収率64.76%)を得た。
tert-ブチルアルコール25 mL及び水素化ナトリウム(60% oil suspension)1.7 gを混合し、70 ℃で1時間反応させた後、1-ブロモ-2,3-ジクロロ-4-(メチルスルホニル)ベンゼン5.8 g(0.016 mol)を投入し、還流下で20時間反応させた。反応液に水50mLを投入し、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール ナトリウム塩を完全に溶解させた後、濃塩酸にてpH1以下に酸性化し、結晶を析出させた。析出した結晶をろ過し、水洗し、乾燥して、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール5.63g(純度76.22%,収率90.7%)を得た。
炭酸カリウム12.6 g及びN,N'-ジメチルホルムアミド20 mLを混合し、攪拌下、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール21 g(0.07 mol, 純度91.5%)を投入し、次いで、2-(2-ブロモエチル)-1,3-ジオキソラン17 g(純度96%)を投入し、85~105℃で6時間反応させた。反応後、反応液を20℃まで冷却し、水60 mLを加えて、撹拌しながら種結晶を添加し、結晶を析出させた。析出した結晶をろ過し、水洗し、乾燥して、融点74~75℃の2-(2-(3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノキシ)エチル)-1,3-ジオキソラン29.2 g(純度88.9%, 収率96.0%)を得た。このものの1H-NMRスペクトルデータは以下の通りである。
1H NMR (500 MHz, CDCl3) δ :7.75 (d, J = 8.5 Hz, 1H), 7.60 (d, J = 8.5 Hz, 1H), 5.16 (t, J = 4.5 Hz, 1H), 4.39 (t, J = 6.0 Hz, 2H), 4.01 (m, 2H), 3.89 (m, 2H), 3.26 (s, 3H), 2.30 (m, 2H)
3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール50 g(0.159 mol, 純度90.9%)、トルエン25 mL及びトリエチルアミン20.9 gを混合し、100℃に加熱した。次いで、2-(2-ブロモエチル)-1,3-ジオキソラン41.6 g(純度90%)を投入し、100℃で3時間反応させた。反応後、反応液を20℃まで冷却し、トルエン175 mL及び水200 mLを加えて抽出した後、分液した。有機層に5質量%水酸化ナトリウム水溶液200 mLを加えて、洗浄し、分液した。得られた有機層を、減圧下で濃縮し、2-(2-(3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノキシ)エチル)-1,3-ジオキソラントルエン溶液101.8 g(純度54.2%, 収率89.9%)を得た。
炭酸カリウム17.9 g及びN-メチルピロリドン29 mLを混合し、攪拌下、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール31.5 g(0.1 mol, 純度90.7%)を投入し、次いで、2-(2-クロロエチル)-1,3-ジオキソラン18.5 g(純度96%)及びヨウ化カリウム1.66 gを投入し、120℃で5時間反応させた。反応後、反応液を20℃まで冷却し、トルエン103 mL及び水103 mLを加えて抽出した後、分液した。有機層を5質量%水酸化ナトリウム水溶液で洗浄し、分液した。得られた有機層を、減圧下で濃縮し、2-(2-(3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノキシ)エチル)-1,3-ジオキソラントルエン溶液95.3 g(純度39.4%, 収率97.4%)を得た。
炭酸カリウム17.9 g及びN-メチルピロリドン29 mLを混合し、攪拌下、3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノール31.5 g(0.1 mol, 純度90.7%)を投入し、次いで、2-(2-クロロエチル)-1,3-ジオキソラン18.5 g(純度96%)を投入し、120℃で23時間反応させた。反応後、反応液を20℃まで冷却し、トルエン103 mL及び水103 mLを加えて抽出した後、分液した。有機層を5質量%水酸化ナトリウム水溶液で洗浄し、分液した。得られた有機層を、減圧下で濃縮し、2-(2-(3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノキシ)エチル)-1,3-ジオキソラントルエン溶液95.0 g(純度37.5%, 収率92.5%)を得た。
2-(2-(3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノキシ)エチル)-1,3-ジオキソラントルエン溶液46.7 g(純度42.8%, 0.05 mol)、メタノール23.5 g、トリエチルアミン13.6 g、水10 g、5%-パラジウム炭素0.4 g(50%wet)及び1,4-ビス(ジフェニルホスフィノ)ブタン0.1 gを、200 mLオートクレーブに仕込み、反応器内を一酸化炭素ガスにて二回置換した。置換後、一酸化炭素ガスを2.0 MPa導入し、120℃で5時間反応させた。次いで、反応液を20℃に冷却した後、反応器内を常圧に戻し、水40 g及びトルエン40 mLを加えて反応液を希釈した。その後、パラジウム炭素をセライトろ過にて除去し、3-(2-(1,3-ジオキソラン-2-イル)エトキシ)-2-クロロ-4-(メチルスルホニル)安息香酸メチルのトルエン-水混合液を得た。
得られた混合液に水酸化ナトリウム3 gを投入し、50℃で1時間反応させた。反応終了後、トルエン層を分液除去し、3-(2-(1,3-ジオキソラン-2-イル)エトキシ)-2-クロロ-4-(メチルスルホニル)安息香酸ナトリウム水溶液を得た。
得られた水溶液を、5~10℃にて、濃塩酸を用いて酸性化し、結晶を析出させた。析出した結晶をろ過し、水洗し、減圧乾燥して、3-(2-(1,3-ジオキソラン-2-イル)エトキシ)-2-クロロ-4-(メチルスルホニル)安息香酸16.4 g(純度84.8%,収率76.7%)を得た。
2-(2-(3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノキシ)エチル)-1,3-ジオキソラン21.5 g(0.052 mol)、トリエチルアミン11 g、5%-パラジウム炭素 0.4 g(50%wet)、1,4-ビス(ジフェニルホスフィノ)ブタン0.1 g及びメタノール60 mLを、200 mLオートクレーブに仕込み、反応器内を一酸化炭素ガスにて二回置換した。置換後、一酸化炭素ガスを2.0 MPa導入し、110℃で3時間反応させた。次いで、反応液を20℃に冷却した後、反応器内を常圧に戻し、水100mL及びトルエン200mLを加え、トルエンにて抽出を行った。次いで、トルエン層を5質量%水酸化ナトリウム水溶液、水でそれぞれ洗浄し、硫酸マグネシウムで乾燥した。その後、トルエン層を濃縮した。得られた残渣を、シリカゲルカラムクロマトグラフィー (溶出液として、ヘキサン:酢酸エチル=3:2(容積比)を使用。) にて精製し、3-(2-(1,3-ジオキソラン-2-イル)エトキシ)-2-クロロ-4-(メチルスルホニル)安息香酸メチル14.3 g(純度99%、収率75.5%)を得た。このものの1H-NMRスペクトルデータは以下の通りである。
1H NMR (500 MHz, CDCl3) δ : 7.92 (d, J = 8.5 Hz, 1H), 7.63 (d, J = 8.5 Hz, 1H), 5.16 (t, J = 4.5 Hz, 1H), 4.40 (t, J = 6.5 Hz, 2H), 4.00 (m, 2H), 3.97 (s, 3H), 3.89 (m, 2H), 3.28 (s, 3H), 2.31 (m, 2H)
2-(2-(3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノキシ)エチル)-1,3-ジオキソラン1 g(2.59 mmol)、酢酸パラジウム 58 mg、トリフェニルホスフィン 203 mg、酢酸ナトリウム 425 mg、メタノール 10 mL及び1,3-ジオキサン 10 mLを、100 mLオートクレーブに仕込み、反応器内を一酸化炭素ガスにて二回置換した。置換後、一酸化炭素ガスを2.5 MPa導入し、150℃で19.5時間反応させた。反応後、反応液を室温に冷却した後、反応器内を常圧に戻し、飽和食塩水20mLを投入した。反応液を酢酸エチルにて抽出し、抽出液は硫酸ナトリウムで乾燥した。その後、酢酸エチル層は濃縮した。得られた残渣を、シリカゲルカラムクロマトグラフィー (溶出液として、ヘキサン:酢酸エチル=7:3(容積比)を使用。) にて精製し、3-(2-(1,3-ジオキソラン-2-イル)エトキシ)-2-クロロ-4-(メチルスルホニル)安息香酸メチル 766.5 mg(収率81.1%)を得た。
1-ブロモ-2,3-ジクロロ-4-(メチルスルホニル)ベンゼン1.0 g(3.68 mmol)、2-(1,3-ジオキソラン-2-イル)エタノール 0.52 g、水酸化ナトリウム 0.29 g及びトルエン 5 mLを混合し、60℃で23時間反応させた。反応後、反応液に氷冷下、水10mLを加え、さらに酢酸エチルを加えて抽出した。その後、硫酸ナトリウムで乾燥し、酢酸エチル溶液は濃縮した。得られた残渣を、シリカゲルカラムクロマトグラフィー(溶出液として、ヘキサン:酢酸エチル=75:25(容積比)を使用。)にて精製し、2-(2-(3-ブロモ-2-クロロ-6-(メチルスルホニル)フェノキシ)エチル)-1,3-ジオキソラン 1.058 g(収率74.6%)を得た。
