Description
Foreliggende oppfinnelse angår området purin-L-nukleosider, farmasøytiske preparater omfattende dem og anvendelser derav.
I de siste tiår er det blitt gjort betydelige anstrengelser for å undersøke mulige anvendelser av D-nukleosid-analoger som antivirale midler. Noe av dette arbeidet har båret frukter, og flere nukleosid-analoger markedsføres fortiden som antivirale medikamenter, inkludert HIV-reverstranskriptase-inhibitorene (AZT, ddl, ddC, d4T og 3TC).
Forskjellige purin-D-nukleosid-analoger er også blitt undersøkt ved søken etter immunmodulatorer. Guanosinanaloger som har substituenter i 7- og/eller 8-stillingene er for eksempel blitt vist å stimulere immunsystemet (for en oversikt, se: Weigle, W.O. CRC Crit. Rev. Immunol. 1987, 7, 285; Lin et al. J. Med. Chem. 1985, 28,1194-1198; Reitz, et al. J. Med. Chem. 1994, 37, 3561-3578, Michael et al. J. Med. Chem. 1993, 36, 3431-3436). Visse 3-p-D-ribofuranosyltiazolo[4,5-d]-pyrimidiner har også demonstrert betydelig immunoaktivitet, omfattende murin miltcelle-proliferasjon og in vivo aktivitet mot Semliki Forest virus (Nagahara, et al. J. Med. Chem. 1990, 33, 407- 415; Robins et al. US-patent 5 041 426). I annen forskning er 7-deazaguanosin og analoger blitt vist å oppvise antiviral aktivitet i mus mot en rekke RNA-virus, selv om forbindelsen mangler antivirale egenskaper i cellekultur. 3-Deazaguanin-nukleosider og nukleotider har også vist betydelig bred-spektrum antiviral aktivitet mot visse DNA- og RNA-virus (Revankar et al. J. Med. Chem. 1984, 27,1389-1396). Visse 7- og 9-deazaguanin-C-nukleosider viser evne til å beskytte mus mot en dødelig utfordring av Semliki Forest virus (Girgis et al. J. Med. Chem. 1990, 33, 2750-2755). Visse 6-sulfenamid- og 6-sulfin-amid-purin-nukleosider har vist betydelig antitumoraktivitet (Robins et al. US-patent 4 328 336). Visse pyrimido[5,4-D]pyrimidin-nukleosider var effektive ved behandling mot L1210 i BDF1 mus (Robins et al. US-patent 5 041 542) og der ble antiviral- og antitumor-aktivitetene for de ovenfor nevnte nukleosider foreslått å være resultatene av deres rolle som immunomodulatorer (Bonnet et al. J med. Chem. 1993, 36, 635-653).
Ett mulig mål for immunomodulering involverer stimulering eller undertrykkelse av Th1- og Th2-lymfokiner. Type l (Th1) -celler produserer interleukin 2 (IL-2), tumor-nekrosefaktor (TNFa) og interferon gamma (IFNy), og de er primært an-svarlige for celle-formidlet immunitet så som hypersensitivitet av forsinket type og antiviral immunitet. Type 2 (Th2) -celler produserer interleukiner, IL4, IL-5, IL-6, IL-9, IL-10 og IL-13 og er først og fremst involvert i å assistere humorale immun-responser så som de sett i respons til allergener, f.eks. IgE og lgG4 antistoff isotyp omkopling (Mosmann, 1989, Annu Rev Immunol, 7:145-173). D-guanosin-analoger er vist å frembringe forskjellige virkninger på lymfokiner IL-1, IL-6, IFNa og TNFa (indirekte) in vitro (Goodman, 1988, Int J Immunopharmacol, 10, 579-88) og in vivo (Smee et al., 1991, Antiviral Res 15: 229). Evnen til D-guanosin-analogene så som 7- tio-8-oksoguanosin til å modulere type 1 - eller type 2-cytokiner direkte i T-celler var imidlertid ineffektiv eller er ikke blitt beskrevet.
Det gjenstår således et behov for nye L-nukleosid-analoger, inkludert nye purin-L-nukleosid-analoger. Det er et spesielt behov for nye purin-L-nukleosider som har immunomodulerende aktivitet og spesielt for nye purin-L-nukleosider som modulerer Th1- og Th2-aktivitet.
Foreliggende oppfinnelse er rettet mot nye purin L-nukleosid-forbindelser og deres anvendelser for fremstilling av et farmasøytisk preparat, samt farmasøytisk preparat omfattende en forbindelse ifølge oppfinnelsen.
I ett aspekt av oppfinnelsen tilveiebringes purin L-nukleosid-analoger med formlene 1 og 3.
I et annet aspekt ved oppfinnelsen omfatter et farmasøytisk preparat en terapeutisk effektiv mengde av en forbindelse med formel 1 og 3 eller en farmasøytisk godtagbar ester eller salt derav, i blanding med minst én farmasøytisk godtagbar bærer.
I enda et annet aspekt ved oppfinnelsen anvendes en forbindelse i henhold til formlene 1 og 3 for fremstilling av et farmasøytisk preparat for behandling av en pasient som har en medisinsk betennelsestilstand som reagerer positivt på administrering av en effektiv dose av forbindelsen til pasienten, og kontroll av pasienten med henblikk på effektivitet og bivirkninger. Blant annet anses det at forbindelser med formel 1 kan anvendes for å behandle en infeksjon, en infestasjon, kreft, tumor eller annen neoplasme eller en autoimmunsykdom. Figurer 1-2 (skjemaer 1 og 2) viser syntetiske kjemiske trinn som kan anvendes for syntetisering av forbindelsene ifølge foreliggende oppfinnelse. Skjemaer som vedrører syntesen av et spesielt preparat er henvist til i eksemplene angitt heri. Figur 3 er en grafisk fremstilling av effekt av eksempler på L-guanosin-analoger på Th1 og Th2.
Der hvor de følgende betegnelser anvendes i denne beskrivelsen, anvendes de som definert nedenfor.
Betegnelsen "nukleosid" angir en forbindelse sammensatt av hvilken som helst pentose- eller modifisert pentosegruppe bundet til en spesifikk stilling i en heterosyklus eller til den naturlige stilling i et purin (9-stilling) eller pyrimidin (1-stilling) eller til den ekvivalente stilling i en analog.
Betegnelsen "nukleotid" angir en fosfatester substituert i 5'-stillingen på et nukleosid.
Betegnelsen "purin" angir nitrogenholdige bisykliske heterosykluser. Betegnelsen "pyrimidin" angir nitrogenholdige monosykliske heterosykluser.
Betegnelsen "D-nukleosider" som anvendes i foreliggende oppfinnelse beskriver de nukleoside forbindelser som har en D-ribose-sukkergruppe (f.eks., adenosin).
Betegnelsen "L-nukleosider" som anvendes i foreliggende oppfinnelse beskriver de nukleoside forbindelser som har en L-ribose-sukkergruppe.
Betegnelsen "L-konfigurasjon" anvendes i hele foreliggende oppfinnelse for å beskrive den kjemiske konfigurasjon av ribofuranosylgruppen i forbindelsene som er knyttet til nukleobasene. L-konfigurasjonen av sukkergruppen i forbindelser ifølge foreliggende oppfinnelse står i motsetning til D-konfigurasjonen av ribose-sukkergrupper i de naturlig forekommende nukleosider så som cytidin, adenosin, tymidin, guanosin og uridin.
Betegnelsen "immunomodulatorer" angir naturlige eller syntetiske produkter som er i stand til å modifisere det normale eller avvikende immunsystem ved stimulering eller undertrykkelse.
Betegnelsen "effektiv mengde" angir mengden av en forbindelse med formel (1) som vil gjenopprette immunfunksjon til normale nivåer eller øke immun-funksjonen til over normale nivåer for å eliminere infeksjon.
Forbindelsene med formel 1 og 3 kan ha flere asymmetriske sentre. Følgelig kan de fremstilles enten i optisk aktiv form eller som en racemisk blanding. Oppfinnelsens ramme som angitt i beskrivelse og patentkrav omfatter de enkelte optiske isomerer og ikke-racemiske blandinger derav, samt de racemiske former av forbindelsene med formel 1 og 3.
Betegnelsene "a" og "p" indikerer den spesifikke stereokjemiske konfigurasjon av en substituent på et asymmetrisk karbonatom i en kjemisk struktur som tegnet. Forbindelsene beskrevet her har alle i L-furanosyl-konfigurasjonen.
Betegnelsen "enantiomerer" angir et par stereoisomerer som ikke er speil-bilder som kan legges på hverandre. En blanding av et par av enantiomerer, i et 1:1-forhold, er en "racemisk" blanding.
Betegnelsen "isomerer" angir forskjellige forbindelser som har samme formel. "Stereoisomerer" er isomerer som bare er forskjellige i måten atomene er anordnet i rommet.
Et "farmasøytisk godtagbart salt" kan være hvilket som helst salt avledet fra uorganiske og organiske syrer eller baser.

Forbindelsene ifølge foreliggende oppfinnelse er generelt beskrevet ved formel 1 og 3 som har strukturen:
Anvendelser
Forbindelsene ifølge foreliggende oppfinnelse i henhold til formlene 1 og 3 er tiltenkt anvendelse for å behandle en rekke tilstander og faktisk hvilken som helst tilstand som reagerer positivt på administrering av én eller flere av forbindelsene. Blant annet er forbindelsene ifølge foreliggende oppfinnelse spesielt tiltenkt å kunne anvendes for å behandle en infeksjon, infestasjon, en kreft-type eller en tumor eller en autoimmun sykdom.
Infeksjoner som kan tenkes behandlet med forbindelsene ifølge foreliggende oppfinnelse omfatter åndedretts-syncytialvirus (RSV), hepatitt B-virus (HBV), hepatitt C-virus (HCV), herpes simplex type 1 og 2, herpes genitalis, herpes keratitt, herpes encefalitt, herpes zoster, humant immunsvikt-virus (HIV), influensa A-virus, hantann-virus (hemoragisk feber), humant papillomvirus (HPV), meslinger og sopp.
Infestasjoner som kan tenkes behandlet med forbindelsene ifølge foreliggende oppfinnelse omfatter protozo-infestasjoner, så vel som invollsorm og andre parasittiske infestasjoner.
Kreft-typer eller tumorer som kan tenkes behandlet omfatter slike som er forårsaket av virus, og virkningen kan involvere inhibering av omdannelse av virus-infiserte celler til en neoplastisk tilstand, inhibering av spredningen av virus fra transformerte celler til andre normale celler og/eller stansing av veksten av virus-transformerte celler.
Autoimmune og andre sykdommer som kan tenkes behandlet omfatter artritt, psoriasis, tarmsykdom, ungdomsdiabetes, lupus, multippel sklerose, gikt og giktaktig artritt, reumatoid artritt, avvisning av transplantasjon, allergi og astma.
Enda ytterligere tiltenkte anvendelser av forbindelsene ifølge foreliggende oppfinnelse omfatter anvendelse som mellomprodukter i den kjemiske syntese av andre nukleosid- eller nukleotid-analoger som i sin tur er nyttige som terapeutiske midler eller for andre formål.
For behandling av et pattedyr, kan effekten av et farmasøytisk produkt inneholdende en terapeutisk og/eller profylaktisk effektiv mengde av en forbindelse ifølge foreliggende oppfinnelse, være relatert til modulering av en eller annen del av pattedyrets immunsystem, spesielt modulering av lymfokin-profiler for Th1 og Th2. Der hvor modulering av Th1- og Th2-lymfokiner finner sted, kan det tenkes at moduleringen kan omfatte stimulering av både Th1 og Th2, undertrykkelse av både Th1 og Th2, stimulering av enten Th1 eller Th2 og undertrykkelse av den andre, eller en bimodal modulering hvor én effekt på Th1/Th2-nivåer (så som generalisert undertrykkelse) opptrer ved en lav konsentrasjon, mens en annen effekt (så som stimulering av enten Th1 eller Th2 og undertrykkelse av den andre) opptrer ved en høyere konsentrasjon.
Generelt er de mest foretrukne anvendelser i henhold til foreliggende oppfinnelse de hvor de aktive forbindelsene er relativt mindre cytotoksisk til ikke-mål verts-cellene og relativt mer aktive mot målet. I denne sammenheng kan det også være fordelaktig at L-nukleosider kan ha øket stabilitet i forhold til D-nukleosider, noe som kan føre til bedre farmakokinetikk. Dette resultat kan oppnås fordi L-nukleosider muligens ikke gjenkjennes av enzymer og derfor kan ha lengre halveringstider.
Det anses at forbindelser ifølge foreliggende oppfinnelse kan administreres i hvilken som helst passende farmasøytisk formulering og under hvilken som helst passende protokoll. Administrering kan således finne sted oralt, parenteralt (omfattende subkutane injeksjoner, intravenøse, intramuskulære, intrasternale injeksjons- eller infusjons-teknikker), ved inhaleringsspray, eller rektalt, topisk og så videre, og i doseenhetsformuleringer inneholdende konvensjonelle ikke-toksiske farmasøytisk godtagbare bærere, hjelpestoffer og bindemidler.
Som eksempel kan det tenkes at forbindelser ifølge foreliggende oppfinnelse kan formuleres i blanding med en farmasøytisk godtagbar bærer. Forbindelsene ifølge foreliggende oppfinnelse kan for eksempel administreres oralt som farmakologisk godtagbare salter. Fordi forbindelsene ifølge foreliggende oppfinnelse stort sett er vannløselige, kan de administreres intravenøst i fysiologisk saltvannsløsning (f.eks., bufret til en pH på omtrent 7,2 til 7,5). Konvensjonelle buffere så som fosfater, bikarbonater eller citrater kan anvendes for dette formål. Selvfølgelig kan en fagmann på området modifisere formuleringene innenfor læren i beskrivelsen for å tilveiebringe en rekke preparater for en spesiell administreringsvei uten å gjøre preparatene ifølge foreliggende oppfinnelse ustabile eller bringe deres terapeutiske aktivitet i fare. Spesielt kan modifiseringen av foreliggende forbindelser for å gjøre dem mer oppløselige i vann eller andre bærere, for eksempel lett gjennomføres ved mindre modifikasjoner (saltdannelse, forestering, etc.) som er godt innenfor vanlig fagkunnskap på området. Det er også godt innenfor vanlig fagkunnskap på området å modifisere administreringsveien og doseregimet for en spesiell forbindelse for å styre farmakokinetikken for foreliggende forbindelser for maksimal fordelaktig virkning i pasienter.
I visse farmasøytiske doseformer foretrekkes pro-medikamentformen av forbindelsene, spesielt omfattende acylerte (acetylert eller annet) derivater, idet pyridinestere og forskjellige saltformer av foreliggende forbindelser er foretrukket. En fagmann på området vil forstå hvordan foreliggende forbindelser skal modifi-seres til pro-medikamentformer for å lette levering av aktive forbindelser tii et målsted i vertsorganismen eller pasienten. En fagmann på området vil også dra fordel av pro-medikamentformenes fordelaktige farmakokinetiske parametere, slik de er anvendbare for levering av foreliggende forbindelser til et målgitt sted i vertsorganismen eller pasienten, for å maksimere forbindelsens tiltenkte virkning.
I tillegg kan forbindelser ifølge foreliggende oppfinnelse administreres alene eller i kombinasjon med andre midler for behandlingen av de ovenfor angitte infeksjoner eller tilstander. Kombinasjonsterapier omfatter administrering av minst én forbindelse ifølge foreliggende oppfinnelse eller et funksjonelt derivat derav og minst én andre farmasøytisk aktiv bestanddel. Den/de aktive bestanddel(er) og farmasøytisk aktive midler kan administreres separat eller sammen, og ved separat administrering kan denne finne sted samtidig eller separat i hvilken som helst rekkefølge. Mengdene av den/de aktive bestanddel(er) og det/de farmasøytisk aktive middel/midler og de relative tider for administrering vil bli valgt for å oppnå den ønskede kombinerte terapeutiske virkning. Fortrinnsvis omfatter kombinasjonsterapien administrering av én forbindelse ifølge foreliggende oppfinnelse eller et fysiologisk funksjonelt derivat derav og ett av midlene nevnt i det følgende.
Eksempler på slike ytterligere terapeutiske midler omfatter midler som er effektive for moduleringen av immunsystemet eller assosierte tilstander, så som AZT, 3TC, 8-substituerte guanosin-analoger, 2',3'-dideoksynukleosider, interteukin II, interferoner så som a-interferon, tucaresol, levamisol, isoprinosin og syklolignaner. Visse forbindelser ifølge foreliggende oppfinnelse kan være effektive for forbedring av den biologiske aktivitet for visse midler ifølge foreliggende oppfinnelse ved reduksjon av metabolismen eller inaktivering av andre forbindelser og ko-administreres som sådanne for denne tiltenkte virkning.
Når det gjelder dosering vil en fagmann på området forstå at en terapeutisk effektiv mengde vil variere med infeksjonen eller tilstanden som skal behandles, dens alvorlighetsgrad, behandlingsregimet som skal anvendes, farmakokinetikken for det anvendte middel, så vel som pasienten (dyr eller menneske) som behandles. Effektive doseringer kan være fra 1 mg/kg kroppsvekt eller mindre, til 25 mg/kg kroppsvekt eller mer. Generelt vil en terapeutisk effektiv mengde av foreliggende forbindelse i doseform vanligvis være fra litt mindre enn omtrent 1 mg/kg til omtrent 25 mg/kg pasientvekt, avhengig av forbindelsen som anvendes, tilstanden eller infeksjonen som behandles og administreringsveien. Dette doseområde gir generelt effektive blodnivå-konsentrasjoner av aktiv forbindelse fra omtrent 0,04 til omtrent 100 mikrogram/ml blod i pasienten. Det anses imidlertid at et passende regime vil utvikles ved å administrere en liten mengde, og deretter øke mengden inntil enten bivirkningene blir urimelig negative eller den tiltenkte effekt oppnås.
Administrering av den aktive forbindelse kan strekke seg fra kontinuerlig (intravenøs drypping) til flere orale administreringer pr. dag (for eksempel Q.I.D.) og kan omfatte oral, topisk, parenteral, intramuskulær, intravenøs, subkutan, transdermal (som kan omfatte et penetrasjonsforbedringsmiddel), bukkal administrering og suppositorie-administrering, blant andre administreringsveier.
For å fremstille de farmasøytiske preparatene ifølge foreliggende oppfinnelse blandes fortrinnsvis en terapeutisk effektive mengde av én eller flere av forbindelsene ifølge foreliggende oppfinnelse grundig med en farmasøytisk godtagbar bærer i henhold til konvensjonelle farmasøytiske kompounderings-teknikkerfor å produsere en dose. En bærer kan ha vidt forskjellige former avhengig av den preparatform som kreves for administrering, f.eks., oral eller parenteral. Ved fremstilling av farmasøytiske preparater i oral doseringsform, kan hvilke som helst av de vanlige farmasøytiske medier anvendes. For flytende orale preparater så som suspensjoner, eliksirer og løsninger, kan det således anvendes egnede bærere og additiver omfattende vann, glykoler, oljer, alkoholer, smaks-midler, konserveringsmidler, farvemidlerog lignende. Forfaste, orale preparater så som pulvere, tabletter, kapsler, og for faststoffpreparater så som suppositorier, kan det anvendes egnede bærere og additiver omfattende stivelser, sukker-bærere så som dekstrose, mannitol, laktose og lignende bærere, fortynnings-midler, granuleringsmidler, smøremidler, bindemidler, desintegreringsmidler og lignende. Om ønsket kan tablettene eller kapslene ved hjelp av standard teknikker belegges for tarm-frigivning eller forsinket frigivning.
For parenterale preparater vil bæreren vanligvis omfatte sterilt vann eller vandig natriumkloirdløsning, selv om andre bestanddeler omfattende de som understøtter dispersjon kan inkluderes. Dersom sterilt vann skal anvendes og holdes sterilt, må preparatene og bærere selvsagt også steriliseres. Injiserbare suspensjoner kan også fremstilles, og i slike tilfeller kan det anvendes passende flytende bærere, suspenderingsmidler og lignende.
Testresultater
In wtøo-tester ble utført på en forbindelse ifølge formel 1 (5-amino-3-p-L-ribofuranosyltiazoio[4,5-d]pyrimidin-2,7(6H)-dion) og resultatene er beskrevet nedenfor.
Perifere mononukleære blodceller (PBMCs ) ble isolert fra "buffy coat" etter Ficoll-Hypaque densitets-gradient sentrifugering av 60 ml blod fra friske givere. T-celler ble deretter renset fra PBMC-ene ved anvendelse av Lymfokwik lymfocytt isoleringsreagens spesifikk for T-celler (LK-25T, One Lambda, Canoga Park CA). Et midlere utbytte på 40 - 60 x 10<6> T-celler ble deretter inkubert natten over ved 37 °C i 20 - 30 ml RPMI-AP5 (RPMl-1640 medium (ICN, Costa Mesa, CA) inneholdende 20 mM HEPES buffer, pH 7,4, 5 % autologt plasma, 1 % L-glutamin, 1 % penicillin/streptomycin og 0,05 % 2-merkaptoetanol) for å fjerne eventuelle kontaminerende vedhengende celler. I alle forsøk ble T-celler vasket med RPMI-AP5 og deretter anbrakt på mikrotiterplater med 96 fordypninger ved en celle-konsentrasjon på 1 x 10<6> celler/ml.
T-cellene ble aktivert ved tilsetning av 500 ng ionomycin og 10 ng phorbol-12-myristat-13-acetat (PMA) (Calbiochem, La Jolla, CA) og inkubert i 48 -72 timer ved 37 °C. PMA/ionomycin-aktiverte T-celler ble behandlet med 0,5 - 50 uM av forbindelsen som ble testet, eller med 250 -10000 U/ml av en kontroll-antiviral, interferon-alfa (Accurate, Westbury, NY), straks etterfølgende aktivering og re-behandlet 24 timer senere. T-celler fra hver plate ble anvendt for immuno-fluorescensanalyse og supernatantene anvendt for ekstracellulære cytokin-målinger. Etter aktivering ble 900 pl celle-supernatant fra hver mikroplate overført til en annen mikroplate for analyse av celle-avledet cytokinproduksjon. Cellene blir deretter anvendt i immunofluorescens-analyse med henblikk på intracellulære cytokinnivåer og cytokin-reseptor-ekspresjon.
Celle-avledede humane cytokin-konsentrasjoner ble bestemt i celle-super-natanterfra hver mikroplate. Aktiveringsbevirkede endringer i interteukin-2 (IL-2) - nivåer ble bestemt ved anvendelse av et kommersielt tilgjengelig ELISA sett (R & D systems Quantikine kit, Minneapolis, MN) eller ved bioundersøkelse ved anvendelse av den IL-2-avhengige cellelinje, CTLL-2 (ATCC, Rockville, MD). Aktiveringsbevirkede endringer i nivåer av interleukin-4 (IL-4), tumor-nekrosefaktor (TNFct) interieukin-8 (IL-8) (R & D systems (Quantikin kit, Minneapolis, MN) og interferon-gamma (IFN-y) (Endogen, Cambridge, MA) ble bestemt ved anvendelse av ELISA sett. Alle ELISA-resultater ble uttrykt som pg/ml og CTLL-2-biounder-søkelsen som tellinger pr. minutt representerende den IL-2-avhengige cellulære inkorporering av 3H-thymidin (ICN, Costa Mesa, CA) av CTLL-2 celler.
Resultatene for forbindelsen i henhold til formel 1 på nivåene av IL-2 TNFa, IFN-y, IL-4 og IL-5 er vist på figur 3, i kolonne 17323.
Syntese
Forbindelsene ifølge foreliggende oppfinnelse kan fremstilles i henhold til syntetiske metoder som hver for seg allerede er kjente av fagfolk på området. Generelt syntetiseres forbindelser ifølge foreliggende oppfinnelse ved konden-sasjon av passende nukleosid-base med det nødvendige sukker-synthon for å gi det beskyttede L-nukleosid, som ved videre manipulering og avbeskyttelse av sukker-hydroksyl-beskyttelsesgruppene til sist vil føre til nukleosid-analog som har den ønskede ribofuranosyl-gruppe med L-konfigurasjon.
Skjema 1 viser syntesen av N<2->acetyl-3-deaza-L-guanosin. 3-Deazaguanin 11 (Cook et al. J. Med. Chem. 1976, 27,1389) ble behandlet med eddiksyreanhydrid i pyridin, hvilket ga N<2->acetyl-3-deazaguanin 12, som ble silylert og koblet med l-acetyl^.S.S-O-tribenzoyl-L-ribose, hvilket ga forbindelse 13. Fjerning av benzoylgruppen med ammoniakk/metanol ga N<2->acetyl-3-deaza-L-guanosin 14.
Skjema 2 viser syntesen av 5-amino-3-p-L-ribofuranosyltiazolo[4,5-d]pyrimidin-2,7(6H)-dion og analoger. 5-Aminotiazolo[4,5-d]pyrimidin-2,7(3H, 6H)-dion 32 (Baker et al. J. Chem. Soc. C 1970, 2478) ble koblet med den avbeskyttede ribose, hvilket ga nukleosidet 33, som ble avbeskyttet, hvilket ga 5-amino-3-p-L-ribofuranosyltiazolo[4,5-d]pyrimidin-2,7(6H)-dion 34. Forbindelse 33 kan være beskyttet med en nitrofenetylgruppe og deretter behandlet med butylnitritt og hydrogenfluorid i pyridin, hvilket ga fluoridderivatet 35. Behandling av 33 med t-butylnitritt (Nagahara et al. J. Med. Chem. 1990, 33,407) i THF kan erstatte aminogruppen med hydrogen for å få 36.
Forbindelsene beskrevet i skjemaene er p-L-guanosin-analoger. De korresponderende ot-L-analoger kan fremstilles på lignende måte, men med L-ribose som har andre beskyttelsesgaipper. 1-Acetyl-2,3,5-0-tribenzoyl-L-ribo-furanose kan erstattes med 1-brom-p-L-ribose-derivater som reagens, noe som ville gi cc-L-nukleosider som hovedprodukter.
Eksempler
Den følgende del viser de eksperimentelle prøver utført i søkerens labora-torium. Eksemplene er forsøkt gjort brede, men ikke for omfattende. Det utførte arbeid omfatter alle prøver beskrevet nedenfor og er ment å illustrere foreliggende oppfinnelse.
Eksempel 1
N<2->Acetyl-3-deazaguanin 12
Til en suspensjon av 3-deazaguanin 11 (2,0 g) i vannfritt pyridin (30 ml) ble det tilsatt eddiksyreanhydrid (5 ml) og den resulterende reaksjonsblandingen ble oppvarmet til 90 °C . Fast stoff ble oppløst litt etter litt og det dannet seg en brun løsning. Etter 10 minutter dukket fellingene igjen opp. Blandingen ble omrørt ved 90 °C i ytterligere 90 minutter og avkjølt til 50 °C . Fellingene ble filtrert og vasket med acetonitril, vann og acetonitril igjen, hvilket ga 1,79 g N<2->acetyl-3-deazaguanin 12 som et lysebrunt fast stoff.
Eksempel 2
N<2->Acetyl-3-deaza-p-L-guanosin 14
En suspensjon av N<2->acetyl-3-deazaguanin 12 (576 mg, 3,0 mmol), heksametyldisilazan (HMDS, 15 ml), pyridin (2 ml) og ammoniumsulfat (10 mg), ble om-rørt under tilbakeløp og utelukkelse av fuktighet i 2,5 timer. Løsemidler ble fordampet og resten tørket under vakuum i 2 timer hvilket ga en skum-sirup. Resten ble oppløst i metylenklorid (vannfritt, 30 ml) og 1-acetyl-2,3,5-tribenzoyl-L-ribose (1,51 g, 3,0 mmol) ble tilsatt, fulgt av langsom tilsetning av trimetylsilyltriflat (4,5 mmol, 0,81 ml). Den resulterende løsning ble tilbakeløpskokt i 20 timer. Løsemid-del ble fordampet og resten oppløst i etylacetat, vasket med 5% NaHCC>3, tørket (Na2S04) og konsentrert. Kromatografi på silika med 5%Et3N og 2-10% etanol i metylenklorid ga tre hovedprodukter: 340 mg produkt med høyere Rf, 368 mg produkt med medium Rf og 335 mg produkt med lavere Rf, alle som et lett gult fast stoff.
En oppløsning av produktet med medium Rf 13 (350 mg) i mettet ammoniakk/metanol sto ved romtemperatur i to dager. Ammoniakk og metanol ble avdam-pet og resten ble kromatografert på silika med 5%Et3N og 20% etanol i metylenklorid, hvilket ga 114 mg N<2->acetyl-3-deaza-p-L-guanosin som 14 som et hvitt, fast stoff.
Eksempel 3
5-Amino-3-B-L-ribofuranosyltiazolo[4,5-d]pyrimidin-2,7(6H)-dioh 34 5-Aminotiazolo[4,5-d]pyrimidin-2,7(6H)-dion 32 (400 mg, 2,71 mmol) ble suspendert i acetonitril (16 ml) og heksametyldisilazan (0,96 ml) og trimetylklor-silan (0,55 ml), og trimetylsilyltriflat (0,9 ml) ble tilsatt. Blandingen ble omrørt under tilbakeløp i 3,5 timer. En oppløsning av trimetylsilyltriflat (0,45 ml) i acetonitril (1,0 ml) ble tilsatt dråpevis og omrøring og oppvarming ble fortsatt i ytterligere 30 min. En oppslemning av 1-0-acetyl-2,3,5-0-tribenzoyl-L-ribofuranose (1,22 g, 2,28 mmol) i acetonitril (4,1 ml) ble tilsatt og blandingen omrørt under tilbakeløp i 30 min. Reaksjonsblandingen ble avkjølt og langsomt helt i en kraftig omrørt blanding
av natriumbikarbonat (2,81 g) og vann (96 ml), noe som ga et klebrig, fast stoff. Etylacetat ble tilsatt og blandingen omrørt inntil det faste stoffet var oppløst. Det vandige laget ble ekstrahert med etylacetat to ganger og de samlede organiske lag vasket med natriumbikarbonat, tørket (Na2S04) og konsentrert. Råproduktet ble renset ved kromatografi på silika med 5% Et3N og 5% etanol i metylenklorid, hvilket ga 1,10 g 5-amino-3-(2,,3,,5'-0-tribenzoyl-p-L-ribofuranosyl)tiazolo[4,5-d]pyrimidin-2,7(6H)-dion 33 som et hvitt, fast stoff.
5-Amino-3-(2,,3,,5,-0-tribenzoyl-p-L-ribofuranosyl)tiazolo[4,5-d]pyrimidin-2,7(6H)-dion 33 (1,09 g, 1,717 mmol) ble oppløst i metanol (25 ml), og natrium-metoksyd (5,4 M i metanol, 0,64 ml) ble tilsatt. Løsningen sto ved romtemperatur i 64 timer. Mesteparten av metanolen ble fordampet og vann (20 ml) og Amberite H-Form (1,0 g) tilsatt. Suspensjonen ble omrørt forsiktig i 20 min. og harpiksen filtrert fra ved suging, og vasket med vann (2x10 ml). Filtratet ble konsentrert og råproduktet ble renset ved krystallisering fra metanol, hvilket ga 368 mg 5-amino-3-p-L-ribofuranosyltiazolo[4,5-d]pyrimidin-2,7(6H)-dion 34 som et farveløst fast stoff.
