Description
Foreliggende oppfinnelse er rettet mot en fremgangsmåte
for debromering av 6,6-dibrompenicillansyre-l,1-dioksyd og derivater derav, ved hjelp av et bisulfitt-salt. De derved oppnådde 6-a-monobrom-forbindelser finner anvendelse til forskjellige formål, som p-laktamase-inhibitorer eller som mellomprodukter ved syntese av andre anvendelige P-laktam-forbindelser, omtalt nedenfor.
Lignende debromeringer er tidligere utført ved hjelp av trialkyltinn- eller triaryltinn-hydrider. EP patentsøknad, publ. nr. 0013617 beskriver for eksempel omdannelsen av benzyl-6-a-brompenicillanat til benzylpenicillanat ved hjelp av tributyltinnhydrid og azobisisobutyronitril i benzen under tilbakeløpskjøling; omdannelsen av forskjellige estere av 6,6-dibrom-penicillanat til de korresponderende 6-3-brompenicil-lanater med de samme reagenser; omdannelsen av benzyl-6,6-dibrompenicillanat-1,1-dioksyd til en blanding av benzyl-6-cx-
og 6-p<->brompenicillanater med de samme reagenser; og omdannelse av benzyl eller pivaloyloksymetyl-6,6-dibrompenicillanat til en blanding av de korresponderende 6-a- og 6-P-bromestere ved bruk av trifenyltinnhydrid i stedet for tributyltinnhydrid.
Den samme europeiske patentsøknad beskriver videre bruken av metyllitium ved -78°C, efterfulgt av reaksjonsavbrudd ved hjelp av eddiksyre, til omdannelse av pivaloyloksymetyl-6,6-dibrompenicillanat i lavt utbytte til den korresponderende 6-P-bromester.
Pratt, US-patent 4.180.506, beskriver katalytisk hydrogenering av 6,6-dibrompenicillansyre over Pd/C katalysator for fremstilling av en blanding av 6-a- og 6-e-brompenicillansyre. Clayton, J. Chem. Soc. (C) , s. 2123-2127 (1969) , beskriver hydrogeneringen av metyl-6,6-dibrompenicillanat over Pd/CaC03for fremstilling av metyl-6-a-brompenicillanat og metylpenicillanat, samt hydrogenering av 6,6-dibrompenicillansyre over den samme katalysator for fremstilling av penicillansyre.
GB patentsøknad 2.051.046 beskriver omdannelsen av 6,6-dibrompenicillansyre-salter til 6-P-brompenicillansyre og av pivaloyloksymetyl-6,6-dibrompenicillanat, med NaBH4eller NaCNEH3som reagens til pivaloyloksymetyl-6-P-brompenicillanat.
Foreliggende oppfinnelse angår en fremgangsmåte for fremstilling av en monobrom-forbindelse med formelen
hvor R<8>er hydrogen, en konvensjonell karboksy-beskyttende gruppe som kan fjernes ved hydrogenolyse eller et konvensjonelt esterdannende radikal som er hydrolyserbart under fysiologiske betingelser. Fremgangsmåten karakteriseres ved at en dibrom-forbindelse med formelen
hvorR<8>er som tidligere angitt, behandles med fra én til to molekvivalenter av et bisulfitt-salt i et reaksjonsinert,
vandig oppløsningsmiddel ved 0-100°C. Det foretrukne temperatur-område, spesielt når R<8>er en hydrolyserbar estergruppe, er 0-40°C. I dette lavere temperaturområdet kan et bisulfitt-overskudd benyttes for å øke reaksjonshastigheten uten vesentlig tap av det annet bromatom. Når R<8>i produktet med formel I er hydrogen, kan den eventuelt omdannes til en av de andre betydninger. Tilsvarende kan en av de andre betydninger for R<8>overføres til hydrogen.
Hvilket kation som er forbundet med bisulfitt-anionet er ikke av vesentlig betydning for foreliggende oppfinnelse, men et alkalimetall, spesielt natrium, er i almindelighet å foretrekke.
Bortsett fra når R<8>er et ester-radikal som er hydrolyserbart under fysiologiske betingelser, foretas prosessen i henhold til oppfinnelsen fortrinnsvis i nærvær av et svakt buffer-materiale. Én til tre molekvivalenter natriumbikarbonat er spesielt velegnet til formålet.
Ved omtale av estere som er hydrolyserbare under fysiologiske betingelser, skal det forstås estere som ofte betegnes "pro-drugs". Slike estere er nå like velkjente og vanlige innen penicillin-fagområdet som farmasøytisk akseptable salter. Estrene benyttes i almindelighet for å øke den perorale absorpsjon, men hydrolyseres alltid lett in vivo til modersyren. De foretrukne esterdannende radikaler er:

hvor R<2>er hydrogen eller metyl og R<3>er (C^-Cg)alkyl. De mest foretrukne radikaler er pivaloyloksymetyl og 1-etoksy-karbonyloksyetyl.
Konvensjonelle karboksy-beskyttende grupper som lar seg fjerne ved hydrogenolyse, er også meget vanlige innen penicillinfeltet. I det foreliggende tilfellet er benzyl, benzhydryl og 2-naftylmetyl foretrukne eksempler på slike grupper, men oppfinnelsen er ikke begrenset til disse tre hydrogenolyserbare grupper.
I denne beskrivelse viser uttrykket "reaksjons-inert oppløsningsmiddel" til et oppløsningsmiddel som ikke innvirker på utgangsmaterialene, reagensene, mellomproduktene eller produktene på en måte som ugunstig påvirker utbyttet av det ønskede produkt. For foreliggende fremgangsmåte er vann velegnet, fortrinnsvis i kombinasjon med et relativt lavtkokende, organisk oppløsningsmiddel som er blandbart med vann, så som tetrahydrofuran.
Hovedanvendelsen av forbindelsene med formel (I) er som mellomprodukter ved syntese av e-laktamase-inhibitorer med formel hvor R er (C1-C4) alkyl eller benzyl; og R<1>er hydrogen eller et konvensjonelt esterdannende radikal som er hydrolyserbart under fysiologiske betingelser; de farmasøytisk akseptable kationiske salter derav nårR<1>er hydrogen; og farmasøytisk akseptable
syreaddisjonssalter derav.
Farmasøytisk akseptable kationiske salter innbefatter, men er ikke begrenset til, saltene av natrium, kalium, kalsium, N,N'-dibenzyletylendiamin, N-metylglukamin (meglumin) og dietanolamin. Farmasøytisk akseptable syreaddisjonssalter innbefatter, men er ikke begrenset til, salter dannet medHC1, H2S04, HN03, CH3S02H og pCH3C6H4S03H.
Forbindelsene med formel (IX) er gjenstand for US patent 4.452.796.
Fremgangsmåtene for fremstilling av forbindelsene med formel (VIII), hvor forbindelsene fremstilt ifølge foreliggende oppfinnelse benyttes som mellomprodukter, omfattes av US patent 4.499.017.
Fremgangsmåten i henhold til oppfinnelsen kan lett utføres etter de metoder som er sammenfattet ovenfor og utførlig beskrevet i efterfølgende eksempler.
Utgangsmaterialene for foreliggende fremgangsmåte er enten kjente forbindelser (hvorav flere er beskrevet i de ovenfor angitte referanser) eller lar seg fremstille ifølge de senere følgende, utførlig beskrevne, Fremstillinger.
Som nevnt kan P-laktamase-inhiberende forbindelser med formel (VIII) og (IX) lett syntetiseres fra debromerte forbindelser fremstilt i henhold til foreliggende oppfinnelse. Fra forbindelse (I) kan for eksempel R<8->gruppen, når den foreligger i syre- (eller salt-) form, omdannes til en hydrogeno-lyserbar eller hydrolyserbar ester, i begge tilfeller ved bruk av velkjente fremgangsmåter innen penicillinfeltet (se f.eks. US-patent 4.234.579, 4.287.181 og 4.348.264).
Eksempel 1
6- a- brompenicillansvre- l. 1- dioksvd
Til 6,6-dibrompenicillansyre-l,1-dioksyd (117,3 g,
0,3 mol) ble det under omrøring i en blanding av H2O (600 ml) og etylacetat (400 ml) porsjonsvis tilsatt NaHC03(75,6 g,
0,9 mol) og derefter NaHS03(37,5 g, 0,36 mol). Efter omrøring 1 1 time ble pH justert fra 3,7 til 1,5 med konsentrert HC1. Det vandige lag ble fraskilt og ekstrahert med 1 x 400 ml frisk etylacetat. De kombinerte organiske lag ble vasket med saltoppløsning, tørket og inndampet, hvorved tittelproduktet ble oppnådd som et fast stoff; 72 g (76,7%); smp. 136-137°; pnmr/D20-NaHC03/S; 1,48 (s,CH3); 1,62 (s, CH3); 4,28 (s, C.3-H); 5,12 (d, J=l,7, C.6-H); 5,37 (d, J=l,7, C.5-H).
Eksempel 2
Benzvl- 6- a- brompenicillanat- l. 1- dioksyd
Til tittelproduktet fra foregående eksempel (24,3 g,
0,0779 mol) i 75 ml DMF ble det tilsatt TEA (7,87 g, 0,0779
mol) og benzylbromid (13,32 g, 0,0779 mol). Blandingen ble omrørt i 16 timer, hellet over i 250 ml vann og ekstrahert med2x 200 ml etylacetat. De kombinerte organiske lag ble vasket med mettet NaHC03, H20 og saltoppløsning, tørket, inndampet til tørrhet, hvorpå residuet ble krystallisert fra etylacetat/- heksan; 28,8 g (92%); smp. 72-74°; pnmr/CDCl3/S (ppm): 1,27
(s, CH3); 1,53 (s, CH3); 4,53 (s, C.3H); 4,8 (d, J=l,7, C.6-H);5,27(d, J=l,7, C.5-H); 5,3 (d, CH2); 7,5 (s, C6H5).
Eksempel 3
PivaloYloksYmetyl- 6- a- brompenicillanat- l. 1- dioksyd
Pivaloyloksymetyl-6,6-dibrompenicillanat-l,1-dioksyd (505 mg, 1 mmol) og NaHS03(208 mg, 2 mmol) ble blandet i 15 ml THF:H20 (2:1). Efter 1 time ble THF fjernet ved fordampning og det vandige residuum ekstrahert med CH2CI2• Ekstrakten ble tørket og inndampet til tittelproduktet, 300 mg; pnmr-topper identiske med de i produktet fra det følgende Eksempel 4, som imidlertid oppviste forurensning med utgangsmaterialet. Denne forurensning kan unngås ved å forlenge reaksjonstiden til ca.
4 til 6 timer.
Eksempel 4
Pivaloyloksymetyl- 6- a- brompenicillanat- l. 1- dioksyd
6-a-brompenicillansyre-l,1-dioksyd (30 g, 0,096 mol) ble løst opp i DMF (100 ml). Trietylamin (9,68 g, 0,096 mol) og klormetylpivalat (14,57 g, 0,096 mol) ble tilsatt og blandingen omrørt en dag, derefter fortynnet med 400 ml H20 og 140 ml etylacetat, hvorpå pH ble justert fra 3,4 til 1,5 med fortynnet HC1. Det vandige lag ble ekstrahert med 2 x 140 ml ny etylacetat. De organiske lag ble kombinert, vasket med 1 x 100 ml mettet NaHC03, 1 x 100 ml H20 og 1 x 100 ml saltoppløsning og inndampet. Den gjenværende olje ble utgnidd med heksan, tatt opp i CH2C12og inndampet på ny, hvorved tittelproduktet ble oppnådd som et fast stoff; 10,5 g; smp. 94-97°; pnmr/CDCl3/TMS/S (ppm): 1,25 (s, 9H); 1,45 (s, 3H); 1,62 (s, 3H); 4,57 (s, 1H); 4,85 (d, 1H, J=l,7 Hz); 5,3 (d, 1H, J=l,7 Hz); 6,0 (q, 2H).
Fremstilling 1
Benzvl- 6. 6- dibrompenicillanat- l. 1- dioksyd
Efter fremgangsmåten i eksempel 2 ble 6,6-dibrompenicillansyre-1,1-dioksyd (39,2 g) omdannet til foreliggende tittelprodukt; 37 g (77%) ; smp. (rå) 134-136° (omkrystallisert) 146-148°; pnmr/CDCl3/S (ppm): 1,27 (s, CH3); 1,55 (s, CH3); 4,62 (s, C.3-H); 5,13 (s, C.5-H); 5,3 (d, CH2); 7,46 (s, C6H5).
Fremstilling 2
Pivaloyloksymetvl- 6. 6- dibrompenicillanat- l. 1- dioksyd
Efter fremgangsmåten fra foregående fremstilling, ble 6,6-dibrompencillansyre-1,1-dioksyd (98 g, 0,025 mol) omdannet til foreliggende tittelprodukt. Det først isolerte produkt ble renset videre ved kromatografi på silikagel med heksan:etylacetat (9:1) som eluent og ga tittelproduktet som et gummiaktig materiale som gikk over i fast form ved henstand; 25 g; pnmr/CDCl3/TMS/S (ppm): 1,23 (s, 9H); 1,43 (s, 3H); 1,6 (s,
3H); 4,63 (s, 1H); 5,13 (s, 1H); 5,93 (q, 2H).
Claims (3)
1. Fremgangsmåte for fremstilling av 6-a-monobrom-penicillansyre-forbindelser med formel
hvorR<8>er hydrogen, en konvensjonell karboksy-beskyttende gruppe som kan fjernes ved hydrogenolyse, eller et konvensjonelt esterdannende radikal som er hydrolyserbart under fysiologiske betingelser,
karakterisert vedat en dibrom-forbindelse med formel
hvorR<8>er som tidligere angitt, behandles med fra én til to molekvivalenter av et bisulfitt-salt i et reaksjons-inert vandig oppløsningsmiddel ved 0-100°C, og når R<8>er hydrogen, omdannes den eventuelt til en av de andre betydninger, eller eventuelt overføres en av de andre betydninger forR<8>til hydrogen•
2. Fremgangsmåte i henhold til krav 1,karakterisert vedat natriumbisulfitt anvendes som bisulfitt-salt ved 0-40°C.
3. Fremgangsmåte i henhold til krav 1,karakterisert vedat behandlingen foretas ved 0-40'C i nærvær av 1-3 ekvivalenter natriumbikarbonat som buffermiddel, og med natriumbisulfitt som bisulfitt-salt.
