De uitvinding heeft betrekking op ontstekingwerende middelen voor uitwendig gebruik bij mens en dier, door plaatselijke toediening van farmaceutische samenstellingen die deze middelen bevatten.
5 De bekende ontstekingwerende middelen omvatten steroide en niet-steroide geneesmiddelen. Veel corticosteroiden bezitten een krachtige ontstekingwerende werking, zowel systemisch als plaatselijk, doch hun toepassing in plaatselijk op te,brengen samenstellingen wordt beperkt door het feit dat ze intradermaal 10 kunnen worden geabsorbeerd en door de neveneffekten die bij langdurige toediening kunnen optreden.
De bekende niet-steroide ontstekingwerende geneesmiddelen omvatten een aantal verbindingen, bijvoorbeeld aspirine en andere salicylzuur verbindingen, alclofenac, bindazac, bufexamac, 15 fenoprofen, flufenaminezuur, ibuprofen, indomethacin, kefpsrofen, mefëinaminezuur, naproxen en fenylbutazon. Met uitzondering van een gering aantal zijn deze niet-steroide ontstekingwerende geneesmiddelen op systemische of plaatselijke wijze gebruikt, doch niet volgens beide toedieningswijzen. Aspirine en veel andere analgetische 20 middelen kunnen bijvoorbeeld systemisch worden toegediend, gewoonlijk op orale wijze, voor de behandeling van arthritis, rheumatiek en andere met ontsteking gepaard gaande aandoeningen, doch zij worden niet toegepast voor plaatselijke toediening, als uitwendige ontsteking-werende middelen. Anderzijds wordt methylsalicylaat, 25 dat uitwendig wordt toegepast voor rheumatische aandoeningen, te giftig beschouwd voor inwendige toediening. Evenzo worden verbindingen zoals bindazac en bufexamac gewoonlijk uitsluitend uitwendig toegepast. In het algemeen wordt er derhalve ten aanzien van de niet-steroide ontstekingwerende middelen een onderscheid aanwe-30 zig geacht tussen de middelen voor inwendig en die voor uitwendig f t gebruik.

De Amerikaanse octrooischriften 4.161.538 en 4.254.274 beschrijven fenylazijnduurderivaten die een analgetische, antipy-retische en ontstekingwerende werking bezitten. De voor deze verbindingen in deze octrooischriften beschreven werkzaamheden zijn 5 gebleken na inwendige toediening van de verbindingen volgens orale of intestinale wijze, doch deze octrooischriften bevatten geen aanwijzing dat deze verbindingen een werking bezitten, wanneer zij plaatselijk worden aangebracht op het menselijk of dierlijk lichaam, zoals bij uitwendige ontstekingwerende middelen. Verrassenderwijze 10 is nu gevonden dat een aantal van deze verbindingen en hun nauw verwante derivaten een bruikbare ontstekingwerende werking bezitten wanneer zij uitwendig worden aangebracht.
De uitvindingen heeft derhalve betrekking op een ont-stekingwerend middel voor uitwendig gebruik dat een fenylazijnzuor-15 derivaat met formule 1 van het formuleblad bevat waarin: n = 1 of 2; een waterstofatoom of een lagere alkylgroep; 2 R een waterstofatoom, een lagere alkylgroep, een aralkylgroep of een pyridylmethylgroep en 20 --B — een ^CH — CH2-of = CH — groep voorstelt.
Wanneer R^ in formule 1 een lagere alkylgroep is, kan deze recht of vertakt zijn en bezit op geschikte wijze 1 tot 4 koolstofatomen, bijvoorbeeld methyl, ethyl, propyl, isopropyl, 25 butyl of isobutyl.

Wanneer R een lagere alkylgroep is kan deze recht of vertakt zijn en 1-6 koolstofatomen bezitten, bijvoorbeeld methyl, ethyl, propyl, isopropyl, butyl, isobutyl, pentyl, isopentyl, 2 hexyl of isohexyl. Wanneer R een aralkylgroep is bijvoorbeeld 30 benzyl of fenetyl, kan deze desgewenst -gesubstitueerd zijn in de aromatische ring met één of meer lagere alkylgroepen (bijvoorbeeld methyl, ethyl, propyl, isopropyl), lagere alkoxygroepen 8102775 -3- (bijvoorbeeld methoxy, ethoxy, propoxy of isopropoxy), halogeen- atomen, (bijvoorbeeld fluor, chloor of broom), of trifluormethyl- 2 groepen. Wanneer R een pyridylmethylgroep is, kan dit 2-, 3-of 4-pyridylmethyl zijn.
5 Bijzondere voorkeur verdienen de verbindingen met for mule 1, waarin : n = 1 of 2; R een methylgroep; 2 R een waterstofatoom, een alkylgroep met 1-6 koolstofatomen, een 10 benzylgroep of een pyridylmethylgroep, en - B— een ^XH — CH— of == CH— groep voorstelt.
De uitvinding heeft eveneens betrekking op de toepassing van de farmaceutisch aanvaardbare zouten van deze verbindin- 2 15 gen met formule 1 waarin R een waterstofatoom is. Dergelijke farmaceutisch aanvaardbare zouten omvatten bijvoorbeeld die welke met alkali-iof aardalkalimetalen (zoals natrium of calcium), aluminium, ammonium, organische basen (zoals triethylamine, dicyclo-hexylamine, dibenzylamine, morfoline of piperidine), en basische 20 aminozuren (zoals lycine of arginine) zijn gevormd.
De fenylazijizuurderivaten met formule 1, die worden toegepast in de onderhavige uitvinding omvatten zowel bekende als nieuwe verbindingen. Kenmerkende voorbeelden van de toegepaste nieuwe verbindingen zijn weergegeven in hierna volgende tabel A, waarin hun 10 struktuur met betrekking tot dezelfde symbolen als in formule 1, hun elementair analyse en hun massaspectrum zijn weergegeven.
In het bijzonder heeft de onderhavige uitvinding betrek- 2 king op de nieuwe verbindingen met formule 1, waarin R een aralkyΙοί pyridylgroep voorstelt en de andere symbolen dezelfde betekenis- 15 sen bezitten zoals reeds voor formule 1 is vermeld, evenals de ver- 2 bindingen waarin R een lagere alkylgroep en de groep % « ^A—B— een ^,CH-Cl·^— groep voorstelt.
De nieuwe verbindingen net formule 1, die in de onderhavige uitvinding kunnen worden toegepast, kunnen worden bereid 2 20 door een zuur met formule 1, waarin R een waterstofatoom voorstelt (of een reaktief derivaat.van een dergelijk zuur, zoals een 2 zuurhalogenide) te laten reageren met een alkanol met formule R OH, 2 waarin R een aralkyl of pyridylmethylgroep is. Deze reaktie kan uitgevoerd worden volgens verschillende werkwijzen, A, B en C 25 weergegeven op het formuleblad, waarin alle symbolen de voor formule 1 reeds bepaalde betekenissen bezitten, behalve dat X een halo-geenatoom zoals chloor of broom voorstekt.
Werkwijze A omvat dat men een zuur (formule 2) laat reageren met de alkanol met formule 3, door koken onder terugvloei-30 koeling, in tegenwoordigheid van een dehydratatiemiddel zoals zwavelzuur, een polyfosforzuur of p-tolueensulfonzuur..Ongeveer 2 tot 3 mol van de alkanol, (formule 3) kan worden toegepast per mol 8102775 TABEL A "5"

Men kan de twee verbindingen eveneens 5 met elkaar laten reageren in tegenwoordigheid van een dehydratatie-middel, zoals dicyclohexylcarbodiimide.
Werkwijze B omvat dat men eerste het zuur (formule 2) laat reageren met een halogenerend middel (zoals thionylchloride, fosforoxychloride of fosforpentachloridej^ ter vorming van eenzuur-10 halogenide (formule 4), en daarna het zuurhalogenide (formule 4) laat reageren met 1 of 2 mol van een alkanol (formule 3) in een inert organisch oplosmiddel (zoals tetrahydrofuran, dioxan, benzeen of tolueen), in tegenwoordigheid van een zuurbindend middel, zoals pyridine, triethylamine, trimethylamine, kaliumcarbonaat of 15 natriumcarbonaat.
Werkwijze C omvat dat men eerst onder koelen met ijs een alkanol (formule 3) mengt met een oplossing van het zuur (formule 2) in een inert organisch oplosmiddel (zoals tetrahydrofuran, dioxan, benzeen of tolueen), trifenylfosfine en pyridinedisulfide, 20 en daarna het mengsel door koken onder terugvloeikoeling laat reageren.
Onafhankelijk van de toggepaste werkwi^e kan het produkt uit het reaktiemengsel worden afgescheiden en op bekende wijze worden gezuiverd, zoals extraktie met een geschikt organisch oplos-25 middel en vacuumdestillatie.
De verbindingen volgens de uitvinding worden gemakkelijk percutaan geabsorbeerd en bezitten een grote ontstekingweren-de werking, wanneer zij plaatselijk op het menselijke of dierlijke lichaam worden aangebacht, opgelost of gedispergeerd in een ge-30 schikte farmaceutische drager. Deze werking blijkt uit de resultaten van de hierna volgende farmacologische proeven, onder toepassing van de werkwijze van Jonelli et al (G. Jonelli, L. Thibault, I.

Beproevingsmethode
Men past 3-4 weken oude vrouwelijke SD ratten met een lichaamsgewicht van 60-70 g toe alsmede een 4$'s crotonolie oplossing in diethylether die men verder verdunt met een oplosmiddel 10 (pyridine: water: diethylether = 4:1:5) om uiteindelijk een 2$2s oplossing te verkrijgen.
Men verdooft elke rat enigszins met diethylether. Men bestrijkt de binnenzijde van elke rechter oorschelp met^liethyl-ether bevochtigde watten, laat de ether verdampen, en brengt daar-15 na druppelsgewijze met een spuit 0,1 ml van de 2$'s crotonolie-oplossing aan, en droogt met een haardroger. Een kunststofkraag met een diameter van 16-19 mm is aangebracht om het hoofd van de rat, die in een kooi is geplaatst en op kamertemperatuur wordt gehouden (23-25°C).
20 Elke proefverbinding wordt opgelost in crotonolie en aangebracht op het oor van de rat. Daarnaast is de proefverbinding samengesteld tot een zalf,(onder toepassing van een lipofiele plastische base omvattende 95$ vloeibare paraffine en 5$ polyetheen). en de zalf wordt na aabbrengen van de crotonolie, grondig uitges 25 smeerd op het oor van de rat met een microspatel.
Zesïruren na de behandeling veegt men de zalf van het oor af, verdooft de rat licht met diethylether, en doorboort de oorschelp van beide oren met een leertang met een diameter van 7 mm. Men weegt elk uitgeboorde gedeelte van het oor en leidt het gewicht 30 van het uitgeboorde gedeelte van het niet behandelde linkeroor af uit het gewicht van het uitgeboorde gedeelte van het rechter oor, .r; dat behandeld is met de crotonolie en de zalf, om het percentage 8102775 n' -> van de gewichtstoename van het rechteroor te berekenen. Evenzo wordt een percentage van de onderdrukkingssnelheid berekend voor elke proefverbinding bij de toegepaste concentraties, ten opzichte van een controlegroep ratten waaraan alleen crotonolie is toege-5 diend.
De resultaten«zijn weergegeven in de hierna volgende tabellen B en C, waarin "n" het aantal ratten in elke testgroep weergeeft.
De bovenstaande proeven tonen duidelijk dat de in de 10 onderhavige uitvinding toegepaste verbindingen, weergegeven door de proefverbindingen A - E, een krachtige ontstekingwerende werking vertonen, wanneer zij uitwendig worden toegediend. De verbindingen met formule 1 en hun farmaceutisch aanvaardbare zouten zijn derhalve bruikbaar als uitwendige ontstekingswerende middelen voor 15 traumata, brandwonden, acute of chronische eïièmen, kontakt- dermatitis, zoster en andere ontstekingsziekten van de huid, veroorzaakt door verschillende factoren.
De uitvinding heeft derhalve betrekking op een ontste-kingwerende farmaceutische samenstelling voor uitwendig gebruik die 20 een verbinding met formule 1 of een farmaceutisch aanvaardbaar zout daarvan, opgelost of gedispergeerd in een voor plaatselijke toediening geschikt farmaceutisch middel bevatten, Hetavoor plaatselijke toediening geschikte farmaceutische middel kan een bekend middel zijn zoals wordt toegepast in bekende farmaceutische samenstellin-25 gen voor plaatselijke toediening, gekozen afhankelijk voor de te formuleren samenstelling, en de samenstelling kan evenzo tevens één of meer farmaceutische toevoegsels bevatten die voor die bijzondere samenstelling geschikt zijn zoals verzachtende middelen, bevochtigingsmiddelen, dispergeermiddelen, emulgeermiddelen, emul-30 siestabilisatoren, geleermiddelen, suspendeermiddelen, conserveringsmiddelen, percutane absorptie bevorderende middelen en aerosoldrijf-middelen. Kenmerkende bestanddelen voor dergelijke 10 formuleringen worden hierna gegeven. In elk geval kan de formulering op geschikte wijze 0,1-10$, bij/oorkeur 1-5% van het werkzame bestanddeel met formule 1 of het farmaceutisch aanvaardbare zout daarvan (berekend op een gewicht/gewicht of gewicht/volume-basis) bfevatten* Water in olie-emulsies vormen de samenstelling die verre-15 weg de voorkeur verdient, en die bij voorkeur het natriumzout of een organisch amine (bijvoorbeeld triethanolamine of benzylamine) zout van de verbinding met formule 1 bevat.

De bovengenoemde samenstellingen kunnen eveneens percu-i 20 tane absorptie bevorderende middelen bevatten, zoals dimethylsul- foxyde, propylgallaat, diisopropyladipaat, diethylsebacaat en ethyl-caproaat.
De uitvinding wordt nader toegelicht door de hierna volgende niet beperkende voorbeelden. De voorbeelden I-V beschrij-25 ven de bereiding van verbindingen met formule 1. De voorbeelden VI-IX tonen enkele kenmerkende farmaceutische samen&llingen volgens de uitvinding die de kemerkendeverbindingen met formule 1 bevatten. De in deze voorbeelden toegepaste verbindingen A, B en D zijn aangeduid met dezelfde letters die zijn toegepast bij de |30 hierboven beschreven farmacologische proeven.
VOORBEELD I

2- Pyridylmethyl 2-(4-(2-oxocylcopentaan-1-ylmethyl)fenyl)propionqgt
Men laat een mengsel van 4,93 g (2-(4-(2-oxycylcopen-taan-1-ylmethyl)fenyl]propionzuur, 4,8 g thionylchloride en 40 ml benzeen reageren door 2 uren koken onder terugvbeikoeling. Na af-5 loop van de reaktie verwijdert men het oplosmiddel en de thionylchloride door destillatie onder verminderde druk. Aan het verkregen residu voegt men 40 ml tetrahydrofuran en 3,3 g 2-pyridinemetha-nol toe en daarna, onder koelen met ijs, 3,0 g triethylamine, en roert het mengsel 2 uren bij kamertemperatuur. Na afloop van de 10 reaktie verwijdert men het oplosmiddel door destillatie onder verminderde druk. Aan het verkregen residu voegt men water toe en extraheert het mengsel met ethylacetaat. De ethylacetaatlaag wast men met achtereenvolgens een 5%' s waterige natriumcarbonaatoplos-sing en water en droogt boven natriumsulfaat.
15 Nadat het oplosmiddel door destillatie is verwijderd zuivert men het verkregen residu door destillatie onder verminderde druk bij 230-235°C/Q,3 mm kwik (badtemperatuur), ter verkrijging van 2,0g van de gewenste verbinding.

VOORBEELD II
3- Pyridylmethyl 2-(4-(2-«oxocylcopentaan-1-ylmethyl)fenyl)propionaat 25 Men kookt een mengsel van 4,93 g 2-(4-(2-oxocyclopen- taan-1-ylmethyl)-fenyl)propionzuur, 4,8 g thionylchloride en 40 ml benzeen 2 uren onder terugvloeikoeling.
Na afkoelen verwijdert men het oplosmiddel en het thionylchloride door destillatie. Aan het verkregen residu voegt 30 men 40 ml tetrahydrofuran en 3,3 g 3-pyridinemethanol toe en daarna, onder koelen met ijs, 3,0 g triethylamine, en roert het mengsel 2 uren bij kamertemperatuur. Na afloop van de reaktie verwijdert men i 8102775 -17- het oplosmiddel door destillatie onder verminderde druk. Men voegt aan het verkregen residu water toe en extraheert het mengsel daarna met ethylacetaat. De thylacetaatlaag wast men met achtereenvol-! gens een 5$*s waterige natriumcarbonaatoplossing en water en droogt 5 daarna boven natriumsulfaat.
Men verwijdert het oplosmiddel door destillatie en zuivert het verkregen residu door destillatie onder verminderde druk bij 215-220°C/0,3 mm kwik (badtemperatuur) ter vorming van 1,8 g van de gewenste verbinding.

VOORBEELD III
15 4-Pvridylmethyl 2(4-(2-oxocylcopentaan-1-ylmethvl)fenyl)propionaat Aan een oplossing van 4,93 g 2-(4-(2-oxocyclopentaan- 1- ylmethyl)-fenyl)propionzuur en 3,3 g 4-pyridinemethanol in 50 ml tetrahydrofuran voegt men 4,3 g dicyclohexylcarbodiimide toe.
Na 5 uren roeren bij kamertemperatuur voegt men aan het mengsel een 20 kleine hoeveelheid azijnzuur toe en verwijdert het onoplosbare ureum door filtratie. Men voegt ethylacetaat toe aan het filtraat en wast het mengsel met water en droogt boven natriumsulfaat. Na verwijderen van het oplosmiddel verkrijgt men een olieachtige stof, die men zuivert door destillatie onder verminderde druk bij 225-25 233°C/0,2 mm kwik (badtemperatuur), ter vorming van 1,8 g van de gewenste verbinding.

2- Pyridylmethyl 2-(4-(2-oxo-1-cyclohexylideenmethyl)fenyl)propionaat 81 02 7 75 -18- Ααη een oplossing van 2,58 g 2(4-(2-oxo-1-cyclohexyli-deenmethyl)-fenyl)propionzuur in 50 ml tetrahydrofuran voegt men, onder koelen met ijs, 2,62 g trifenylfosfine en 2,2 g pyridine-sulfide toe en roert het mengsel 30 minuten bij kamertemperatuur.
5 Aan het mengsel voegt men een oplossing van 2,73 g 2-pyridine- methanol in 5 ml tetrahydrofuran toe en kookt het verkregen mengsel 3 uren onder terugvloeikoeling.
Na afkoelen giet men het mengsel uit in ijswater en extraheert met ethylacetaat. Het extrakt wast men met water en 10 droogt boven natriumsulfaat. Men verwijdert het oplosmiddel door destillatie en zuivert het residu door silicagel kolomchromatogra-fie waarbij men 1,71 g van *een lichtgele olie verkrijgt.

VOORBEELD V
Methyl 2-(4-(2-oxocylcopentaan-1-ylmethyl)fenyl)propionaat
Men laat een mengsel van 5,0 g 2(4-(2-oxocylcopentaan-20 1-ylmethyl)-fenyl)propionzuur, 50 ml methanol en 1,0 g geconcentreerd zwavelzuur 6 uren reageren door koken onder terugvloeikoeling. Na afloop van de reaktie verwijdert men de overmaat methanol door destillatie onder verminderde druk. Men voegt aan het residu water toe en extraheert het mengsel met diethylether. De 25 diethyletherlaag wast men achtereenvolgens met water, een 5$'s waterige natriumcarbonaatoplossing en water. Na drogen verwijdert ^ men de diethylether door afdampen. Men zuivert het verkregen residu door destillatie onder verminderde druk bij 195-200°C/0,1 mm kwik (bddtemperatuur) waarbij men 4,8 g van de gewenste verbinding 30 verkrijgt in de vorm van een kleurloze olie.

Claims (9)
1. Ontstekingwerende samenstelling voor uitwendig:?gebruik bij mens en dier, met het kenmerk, dat deze een verbinding met formule 1 van het formuleblad, waarin n = 1 of 3, een waterstof- 2 atoom of een lagere alkylgroep, R een waterstofatoom, lagere 5 alkyl-, aralkyl- of pyridylmethylgroep; en -B - eer^CH — CH^- of = CH — groep of een farmaceutisch aanvaardbaar zout van een dergelijke verbinding, opgenomen in een voor plaatselijke toediening geschikt farmaceutisch middel, bevat.
2. Samenstelling volgens conclusie 1, met het kenmerk, dat de samenstelling een verbinding met formule 1, waarin R een 2 methylgroep en R een waterstofatoom, een alkylgroep met 1-6 kool-stofatomen, een benzylgroep of een pyridylmethylgroep voorstelt, of een farmaceutisch aanvaardbaar zout daarvan, omvat.
3. Samenstelling volgens conclusie 1 of 2, met het ken merk, dat de samenstelling het natriumzout of een orgnisch amine-zout van een verbinding met formule 1 bevat.
4. Samenstelling volgens conclusie 1-3, met het kenmerk, dat deze geformuleerd is tot een crème, zalf, lotion, smeersel, 20 emulsie, pleister, vloeistof of aerosol met 0, \% tot 1 Q$ van de verbinding met formule 1 of een farmaceutisch aanvaardbaar zout daarvan.
5. Samenstelling volgens conclusie 4, met het kenmerk, dat deze 1-5% van de verbinding met formule 1 of het farmaceutisch 25 aanvaardbare zout daarvan bevat.
6. Samenstelling volgens conclusie 1-5, met het kenmerk, dat deze één of meer van de volgende farmaceutische toevoegsels^ bevat: verzachtende middelen, bevochtigende middelen, dispergeer-middelen, emulgeermiddelen, emulsiestabilisatoren, geleermiddelen, 30 suspendeermiddelen, conserveringsmiddelen, percutane absorptie-bevorderende middelen en aerosoldrijfmiddelen. 8102775 V A* -22-

Samenstelling volgens conclusie 1 tot 6, met het kenmerk , dat de samenstelling één of meer van de volgende bestanddelen bevat, natuurlijke of synthetische oliën, wassen, vetten, paraffinen, mengsels van zachte paraffine en zware oliën, hogere 5 vetzuuralcoholen, wolalcoholen, hogere vetzuren, glyceriden, zepen, sorbitanesters, -polysorbaten, macrogolesters en cellulose-ethers.
8. Verbindingen met formule 1 van het formuleblad^ waarin n = 1 of 2, een waterstofatoom of een lagere alkylgroep 2 10 en R ofwel een aralkylgroep of een pyridylmethylgroep en ^ A - B — een ^CH — CH^- of = CH— groep voorstelt ofwel een lagere alkylgroep voorstelt en -B- een ^CH — groep. 15 9. 2-Pyridylmethyl 2-(4-(2-oxocylcopentaan-1-ylmethyl) fenyl)propionaat. 10. 3-Pyridylmethyl 2(4(2-oxocylcopentaan-1-ylmethyl)fenyl) propionaat. ) 11. 4-Pyridylmethyl 2(4-(2-oxocyclopentaan-1-ylmethyl) 20 fenyl)propionaat. 12. 2-Pyridylmethyl 2(4-(2-oxo-1-cyclohexylideenmethyl) feoylèpropionaat.
