Abstract
La presente invención se refiere a un método para capturar, aislar y analizar células circulantes de mieloma múltiple en una muestra de sangre previamente obtenida de un sujeto de prueba, el método caracterizado porque comprende: a) poner en contacto dicha muestra con partículas magnéticas coloidales que se conjugan con un primer ligando, que es anti-CD138, y un segundo ligando, que es anti CD38; (b) someter la muestra de la etapa (a) a un campo magnético para producir una fracción separada de células circulantes de mieloma múltiple unidas a la partícula magnética; (c) tratar la muestra de la etapa (b) con un primer marcador adicional; y (d) analizar las células circulantes de mieloma múltiple. Un método in vitro para determinar si el paciente es un candidato probable para la intervención terapéutica para enfermedades asociadas con las células plasmáticas anormales, el método caracterizado porque comprende (a) procesar dicha sangre del paciente para determinar cuantas células de mieloma múltiples circulantes (CMMC) hay en la muestra, en donde el procesamiento comprende el método como el que se reclama (b) determinar mediante recuento si el número de CMMC presentes en dicha muestra es igual o mayor que o igual que el intervalo normal. Un método in vitro para determinar si un paciente sometido a intervención terapéutica está reduciendo el número de CMMC o para determinar si un paciente que tuvo una enfermedad de células plasmáticas anormales y se trató exitosamente por tal enfermedad, permanece en remisión, el método caracterizado porque comprende (a) procesar dicha sangre del paciente para determinar cuantas CMMC hay en la muestra en un primer punto en el tiempo, en donde el procesamiento comprende el método, (b) determinar mediante recuento si el número de CMMC presentes en dicha muestra es igual o mayor que o igual que el intervalo normal; (c) procesar dicha muestra de sangre del paciente para determinar cuántas CMMC hay en la muestra en un segundo punto en el tiempo, en donde el procesamiento comprende el método (d) determinar mediante recuento si el número de CMMC presente en dicha muestra es igual o mayor que o igual que el intervalo normal; y ( e) comparar los números de las etapas (b) y (d).
