제3도는 전체 T-억압인자 아군집(t-억압인자)의 경우 FACS/본 발명에 따른 방법과 비교를 나타낸 도표.

Claims (13)

  1. 결정할 세포개체군과 샘플을 정량할 세포개체군의 특색적 표면항원에 특이적으로 대응하는 표지항체와 반응시키고, 하나 또는 그 이상의 표준물질과 알려진 다른 입자농도를 같은 방법으로 같은 표지항체와 반응시키고, 그 경우 표준용액은 침전능에서 결정할 세포와 비교되는 입자로 구성하고, 그 입자는 표지된 항체 또는 그의 일부분에 대응하는 분자로 채우고, 샘플용액의 세포 및 표준용액의 입자를 과량의 표지항체로 부터 분리하고, 표지물질의 양은 세로 및 입자에서도 측정되며, 표준용액으로 부터 측정값과 샘플로부터 측정값을 비교함으로써, 샘플에서 결정할 세포의 수가 확인되는 것을 특징으로 하는 세포개체군 및 아군집의 정량방법.
  2. 제1항에 있어서, 표지항체로서는 T-림프구 또는 T-헬퍼 또는 T-억제인자 아군집의 표면항원에 대응한 표지된 단일클론항체가 사용됨을 특징으로 하는 방법.
  3. 제1항 또는 제2항에 있어서, 표지는 방사선동위원소, 염료, 형광염료 또는 효소로 일어남을 특징으로 하는 방법.
  4. 제3항에 있어서, 표지효소로서는 퍼록시다제, 알칼라인 포스파리제, 글루코오스옥시다제 또는 β-갈락토시다제가 사용됨을 특징으로 하는 방법.
  5. 제1항에 있어서, 실제 앞서 제시하고 예시한 바와같이 세포 개체군 또는 아군집의 정량방법.
  6. 표지된 항체 또는 그의 일부분에 대응한 분자가 수행되고 결정된 세포로서 침전능이 같은 입자가 포함되어진 제1항 내지 5항중 어느 항에 따른 방법에 따라서 세포 개체군 또는 아군집의 정량을 위한 표준.
  7. 제6항에 있어서, 입자는 유리나 또는 합성수지로 구성되어 있음을 특징으로 하는 표준.
  8. 제7항에 있어서, 합성수지입자는 메타크릴레이트를 기본으로 한 라텍스입자로 이루어짐을 특징으로 하는 표준.
  9. 제6항 내지 8항중 어느 한 항에 있어서, 입자는 결정되어진 세포에 대해 표면항원으로써 같은 항원작용을 가지는 단백질, 특이적으로 표지물질을 인식하는 표지항체 또는 항체에 대응한 항-항체를 운반하는 것을 특징으로 하는 표준.
  10. 제6항에 있어서, 실제앞서 제시하고 예시한 바와같은 것을 특징으로 하는 표준.
  11. 결정되어진 세포개체군에 특이적으로 대응한 표지된 항체를 포함하고, 제6항 내지 10항중 어느 항에 따른 하나 또는 그 이상의 표준, 표지시약의 측정을 위한 적절한 검출시스템 뿐만 아니라 임의로 보조제를 더 필요로 하는, 제1항 내지 5항중 어느 항에 따른 방법을 수행하기 위한 시약혼합물.
  12. 제11항에 있어서, 항체를 β-갈락토시다제로 표지하고, 검출시스템은 β-갈락토시다제 검출을 위해 필요한 시약을 포함함을 특징으로 하는 시약.
  13. 제11항에 있어서, 실제 앞서 제시하고 예시한 바와 같은 것을 특징으로 하는 시약.
