Abstract
Description
[1]複数の多能性幹細胞由来軟骨パーティクルが合体してなる板状軟骨。

を含むことを特徴とする方法。

本発明は、多能性幹細胞由来の板状軟骨を提供する。本発明の板状軟骨は、複数の多能性幹細胞由来軟骨パーティクルが合体してなる板状軟骨(板状軟骨組織)である。本発明の板状軟骨は、複数の多能性幹細胞由来軟骨パーティクルが複層に合体してなる板状軟骨であってもよく、複数の多能性幹細胞由来軟骨パーティクルが単層に合体してなる板状軟骨であってもよい。
ES細胞は、ヒトやマウスなどの哺乳動物の初期胚(例えば胚盤胞)の内部細胞塊から樹立された、多能性と自己複製による増殖能を有する幹細胞である。
***幹細胞は、精巣由来の多能性幹細胞であり、***形成のための起源となる細胞である。この細胞は、ES細胞と同様に、種々の系列の細胞に分化誘導可能であり、例えばマウス胚盤胞に移植するとキメラマウスを作出できるなどの性質をもつ(M. Kanatsu-Shinohara et al. (2003) Biol. Reprod., 69:612-616; K. Shinohara et al. (2004), Cell, 119:1001-1012)。神経膠細胞系由来神経栄養因子(glial cell line-derived neurotrophic factor (GDNF))を含む培養液で自己複製可能であるし、またES細胞と同様の培養条件下で継代を繰り返すことによって、***幹細胞を得ることができる(竹林正則ら(2008),実験医学,26巻,5号(増刊),41〜46頁,羊土社(東京、日本))。
胚性生殖細胞は、胎生期の始原生殖細胞から樹立される、ES細胞と同様な多能性をもつ細胞であり、LIF、bFGF、幹細胞因子(stem cell factor)などの物質の存在下で始原生殖細胞を培養することによって樹立しうる(Y. Matsui et al. (1992), Cell, 70:841-847; J.L. Resnick et al. (1992), Nature, 359:550-551)。
人工多能性幹(iPS)細胞は、特定の初期化因子を、DNAまたはタンパク質の形態で体細胞に導入することによって作製することができる、ES細胞とほぼ同等の特性、例えば分化多能性と自己複製による増殖能、を有する体細胞由来の人工の幹細胞である(K. Takahashi and S. Yamanaka (2006) Cell, 126:663-676; K. Takahashi et al. (2007), Cell, 131:861-872; J. Yu et al. (2007), Science, 318:1917-1920; Nakagawa, M.ら,Nat. Biotechnol. 26:101-106 (2008);国際公開WO 2007/069666)。初期化因子は、ES細胞に特異的に発現している遺伝子、その遺伝子産物もしくはnon-cording RNAまたはES細胞の未分化維持に重要な役割を果たす遺伝子、その遺伝子産物もしくはnon-cording RNA、あるいは低分子化合物によって構成されてもよい。初期化因子に含まれる遺伝子として、例えば、Oct3/4、Sox2、Sox1、Sox3、Sox15、Sox17、Klf4、Klf2、c-Myc、N-Myc、L-Myc、Nanog、Lin28、Fbx15、ERas、ECAT15-2、Tcl1、beta-catenin、Lin28b、Sall1、Sall4、Esrrb、Nr5a2、Tbx3またはGlis1等が例示され、これらの初期化因子は、単独で用いても良く、組み合わせて用いても良い。初期化因子の組み合わせとしては、WO2007/069666、WO2008/118820、WO2009/007852、WO2009/032194、WO2009/058413、WO2009/057831、WO2009/075119、WO2009/079007、WO2009/091659、WO2009/101084、WO2009/101407、WO2009/102983、WO2009/114949、WO2009/117439、WO2009/126250、WO2009/126251、WO2009/126655、WO2009/157593、WO2010/009015、WO2010/033906、WO2010/033920、WO2010/042800、WO2010/050626、WO 2010/056831、WO2010/068955、WO2010/098419、WO2010/102267、WO 2010/111409、WO 2010/111422、WO2010/115050、WO2010/124290、WO2010/147395、WO2010/147612、Huangfu D, et al. ntES細胞は、核移植技術によって作製されたクローン胚由来のES細胞であり、受精卵由来のES細胞とほぼ同じ特性を有している(T. Wakayama et al. (2001), Science, 292:740-743; S. Wakayama et al. (2005), Biol. Reprod., 72:932-936; J. Byrne et al. (2007), Nature, 450:497-502)。すなわち、未受精卵の核を体細胞の核と置換することによって得られたクローン胚由来の胚盤胞の内部細胞塊から樹立されたES細胞がntES(nuclear transfer ES)細胞である。ntES細胞の作製のためには、核移植技術(J.B. Cibelli et al. (1998), Nature Biotechnol., 16:642-646)とES細胞作製技術(上記)との組み合わせが利用される(若山清香ら(2008),実験医学,26巻,5号(増刊), 47〜52頁)。核移植においては、哺乳動物の除核した未受精卵に、体細胞の核を注入し、数時間培養することで初期化することができる。
Muse細胞は、WO2011/007900に記載された方法にて製造された多能性幹細胞であり、詳細には、線維芽細胞または骨髄間質細胞を長時間トリプシン処理、好ましくは8時間または16時間トリプシン処理した後、浮遊培養することで得られる多能性を有した細胞であり、SSEA-3およびCD105が陽性である。
本発明は、上記板状軟骨の製造方法を提供する。本発明の製造方法は、以下の工程を含むものであればよい。
工程1:多能性幹細胞由来軟骨パーティクルを製造する工程
工程2:板状軟骨の形成に必要数の軟骨パーティクルを、隣接する軟骨パーティクルが相互に接触し得る状態で培養する工程
(i)多能性幹細胞を骨形成タンパク質(BMP)2、トランスフォーミング増殖因子(TGF)βおよび増殖分化因子(GDF)5から成る群より選択される1以上の物質ならびにヒドロキシメチルグルタリルCoA(HMG-CoA)還元酵素阻害薬を含む培養液中で接着培養する工程、および
(ii)前記工程(i)で得られた細胞をBMP2、TGFβおよびGDF5から成る群より選択される1以上の物質ならびにHMG-CoA還元酵素阻害薬を含む培養液中で浮遊培養する工程。
本発明は、上記本発明の板状軟骨を含む、損傷軟骨修復用の医薬組成物を提供する。本発明の医薬組成物はまた、有効量の本発明の板状軟骨を含みうる。ここで、医薬組成物には、医薬品、医療機器、および再生医療製品が含まれうる。本発明の医薬組成物の投与方法としては、損傷した軟骨における軟骨欠損部に板状軟骨を投与(移植)する方法が挙げられる。投与する板状軟骨は、軟骨欠損部の形状および大きさに適合するように形成あるいはトリミングして投与することが好ましい。軟骨欠損部と板状軟骨の接触部には、フィブリン糊、ゼラチンゲル、コラーゲンゲル、ヒアルロン酸ゲル等を塗布して板状軟骨の剥がれ落ちを防止してもよい。また、軟骨欠損部に投与(移植)した板状軟骨を骨膜等で固定してもよい。また、軟骨板に糸をかけて周囲軟骨あるいは骨に逢着してもよい。本発明の医薬組成物の投与対象は、例えば哺乳動物(例:マウス、ラット、ハムスター、ウサギ、ネコ、イヌ、ウシ、ヒツジ、サル、ヒト)であり、好ましくはヒトである。
(i)本発明の板状軟骨を軟骨欠損部位に移植する工程を含む、軟骨修復方法。
(ii)軟骨修復のために使用する、本発明の板状軟骨。
(iii)損傷軟骨修復用医薬組成物を製造するための、本発明の板状軟骨の使用。
(1)多能性幹細胞由来軟骨同士および/または多能性幹細胞由来軟骨と成体軟骨との融合を促進する方法であって、FGF受容体アゴニストの存在下で、多能性幹細胞由来軟骨同士および/または多能性幹細胞由来軟骨と成体軟骨とを接触させることを特徴とする方法。
(2)FGF受容体アゴニストがFGF18である前記(1)に記載の方法。
(3)FGF受容体アゴニストを含む培地を用いて、複数の多能性幹細胞由来軟骨を接触させながら培養する工程を含む、移植用軟骨塊の製造方法。
(4)FGF受容体アゴニストがFGF18である前記(3)に記載の方法。
(5)複数の多能性幹細胞由来軟骨が融合(integrate)してなる移植用軟骨塊。

1.材料および方法
(1)細胞
京都大学iPS細胞研究所の、沖田博士、中川博士および山中博士から分与された4系統のヒトiPS細胞株(409B2、604B1、1231A3およびQHJI)を、ヒトiPS細胞として使用した。409B2はヒト皮膚線維芽細胞から作製され、604B1、1231A3およびQHJIはヒト末梢単核球から作製された。エピソームプラスミドベクター(pCXLE-hOCT3/4-shp53-F, hSK, hUL, EBNA1)がエレクトロポレーションされ、全ての細胞はゲノム組み込みについて陰性であった。
非特許文献1に記載の方法を一部改変した方法で、ヒトiPS細胞由来軟骨パーティクル(以下、「軟骨パーティクル」と記す)を作製した。具体的には、以下の方法で軟骨パーティクルを作製した。
培養中のヒトiPS細胞に、0.5×TrypLE Selectを添加し、インキュベーションの後、セルスクレーパーを用いて細胞を剥離させた。細胞を計数し、0.5-1.0×107個を 100 mLバイオリアクター(BWV-S10A、エイブル)へ移し、10 nM Y-27632(Wako)を添加したStemFit AK03N(Ajinomoto)を100 mLを加えて、マグネチックスターラー(BWS-S03NOS-6、エイブル)により60 rpmで回転させ、37 ℃、CO2 5%の条件下で、4-7日間培養した。その結果、直径50 μmから300 μmのiPS細胞塊が得られた。
上記の方法で得られたiPS細胞塊を回収し、軟骨分化培地を5 mLを入れた10 cm suspension culture dish(sumitomo)4-12 dishesへ iPS細胞塊を播種した。なお、軟骨分化培地としては、0.2% FBS(Invitrogen)、1% ITS-X(Invitrogen)、50 μg/mL ascorbic acid(Nakalai)、1×10-4 M nonessential amino acids(Invitrogen)、1 mM sodium pyruvate(Invitrogen)、10ng/mL BMP2(PeproTech)、10ng/mL TGF-β3(Wako)、10ng/mL GDF5(Biovision)、1μM rosuvastatin(Biovision)、を添加したDMEM(SIGMA)を用いた。播種後、37 ℃、CO2 5%条件下で培養した。1-3日後に、新しい軟骨分化培地へ交換し、以後、2-5日の間隔で培地交換を行い、2-3週間培養を継続した。すると、iPS細胞塊は次第にdishへ接着して結節(nodule)が形成された。得られた結節は自然にdishからはがれて浮遊するか、あるいは結節をセルスクレーパーで剥がし、6 cm suspension culture dish(sumitomo)へ移し、37 ℃、CO2 5%条件下で培養した。1-5日後に、新しい軟骨分化培地へ交換した。以後、2-7日ごとに培地交換を行った。なお、培地交換時に、dishに貼付いた結節があればセルスクレーパーで剥がして浮遊させた。分化誘導開始から12週間後の軟骨パーティクルを以下の実験に使用した。
2個の軟骨パーティクルを96ウェル丸底プレートに入れ(図1(A)、(B)参照)、軟骨形成培地(以下を含むDMEM (Sigma):1% ITS, 1% FBS, 2 mM L-glutamine (Thermo), 1 x 10-4 M nonessential amino acids (Thermo), 1 mM sodium pyruvate (Thermo), 50 units penicillin, 50 mg/mL streptomycin, 50 μg/mL ascorbic acid (Nacalai), 10 ng/mL BMP2 (Peprotech), 10 ng/mL TGFβ1 (Peprotech), および10 ng/mL GDF5 (PTT))で、37 ℃、CO2 5%条件下で培養した。各1対の軟骨パーティクルを含むウェルを、合計84個準備した。ウェルを7つの群(1群あたり12対)に分け、実験開始後0、3、7、14、28、56日目に、肉眼的および組織学的分析を行った。培地交換は、軟骨パーティクルの位置を変えないように注意して、毎日穏やかに行った。
AAVS1遺伝子座を標的とするCAG-EGFP(317-12)またはCAG-mCherry(511-5B)のいずれかのトランスジーンを保持するヒトiPS細胞株201B7から、軟骨パーティクルを作製した。EGFP発現軟骨パーティクルとmCherry発現軟骨パーティクルの合体を、多光子レーザー顕微鏡(Nikon A1R MP +)と解析ソフトウェア(Nikon NIS Elements)を用いてコマ撮り観察を行った。蛍光画像を連続11.5日にわたって1時間ごとに記録した。ムービーの各画像は100ミリ秒を表す。したがって、24時間は2.4秒に相当する。iPS-Cartが動いて視野から外れると、コマ撮り画像が数回中断された。
組換えヒトFGF18(PeproTech)をリン酸緩衝生理食塩水(PBS)に溶解してストック溶液(100 μg/mL)を調製した。合計90対の軟骨パーティクルを、上記(3)の条件で培養した。1ウェルあたり1対の軟骨パーティクルを0.3 mLの培地中で培養した。45対の軟骨パーティクルを、溶媒(0.3 μlのPBS)を添加した培地中で培養し、他の45対を0.3μlのFGF18ストック溶液を添加した培地中(最終濃度、100 ng/mL)で培養した。各群の45対をさらに3つの群に分け、組織学的分析に供した(実験開始後3、7または14日)。
軟骨パーティクルの対を4%パラホルムアルデヒドで固定し、処理してパラフィン中に包埋した。2個の軟骨パーティクル間の最も接触する領域をカバーする切片を確保するために、サンプルの合体部の周囲に20〜150の連続切片を作製した(図1(C)参照)。最大の接触部分の連続切片を選択し、さらなる分析に使用した。切片をヘマトキシリン−エオジンおよびサフラニンO-ファストグリーン-鉄ヘマトキシリンで染色した。また、切片をヤギ抗I型コラーゲン抗体(Souther Biotech)および抗II型コラーゲン抗体(Thermo)で免疫染色した。
ヒトiPS細胞株QHJIから作製した軟骨パーティクルの軟骨膜様膜を中心軟骨から分離するために、実体顕微鏡下でピンセットを用いて軟骨膜様膜を剥がした。軟骨膜様膜および中心軟骨から別々にRNAを抽出した。試料を液体窒素中で凍結させ、Multi Beads Shocker(安井機械)で粉砕し、ISOGEN(登録商標)(Nippon Gene)を用いて全RNAを抽出し、RNeasy(登録商標)(Qiagen)で精製した。
抽出したRNAの品質は、Bioanalyzer 2100(Agilent Technologies)で評価した。1 μgの全RNAをTruSeq(登録商標) Stranded mRNA Library Prep Kit(Illumina)を用いて、製造業者の指示に従ってライブラリーを作製した。構築したライブラリーの品質および量は、Bioanalyzer 2100およびQubit(登録商標) dsDNA HS assay kit(Thermo)で評価した。ライブラリーは、NextSeq(登録商標) 500(Illumina)の75サイクルのSingle-Read modeで配列決定した。全ての読み出された配列は、BCL2FASTQ Conversion Software(v2.17.1.14)を用いてFASTQフォーマットで抽出された。アダプター、ポリA配列、および3'読み取り末端の低品質塩基は、cutadapt-1.12によってトリミングされた。トリミングされていない読み出し配列およびトリミングされた読み出し配列を、TopHat-2.1.1を用いてヒトゲノムhg38にマッピングした。GENCODE release v25からのヒト遺伝子アノテーションを利用した。遺伝子発現分析のために、Cufflinks-2.1.1を使用して、各遺伝子の発現レベルを100万読み出し配列当たりのエクソン1kb当たりの読み出し(RPKM)に対して正規化した。
3つの軟骨膜様膜サンプル、3つの中心軟骨サンプル、および3つの軟骨パーティクル全体から、別々にトータルRNAを抽出した。500 ngの全RNAを、ReverTra Ace(登録商標)(東洋紡、東京、日本)およびオリゴ(dT)20プライマーを用いてfirst-strand cDNAに逆転写した。KAPA PROBE FAST qPCR kitまたはKAPA SYBR(登録商標) FAST qPCR kit Master Mix ABI prism(KAPA Biosystems、MA、USA)を使用してPCR増幅を行った。使用したPCRプライマーを表1に示した。RNA発現レベルをGAPDH発現のレベルに対して正規化した。増幅産物を用いて、リアルタイム定量RT-PCR用の標準曲線を導いた。
データは平均および標準偏差として示される。この研究では、両側スチューデントのt検定またはSteel-Dwass検定を使用した。P値が0.05未満のとき、統計的に有意であるとみなした。
(1)ヒトiPS細胞株QHJIから作製した軟骨パーティクル間のインビトロ合体過程の分析
2個の軟骨パーティクルは7日後に結合し始め、その後しっかりした結合を形成し始めた(図2、左列)。組織学的には、1つの軟骨パーティクルは、中心部の軟骨と軟骨を包む軟骨膜様の膜組織から構成されていた(図2、第2列、第3列、右列)。軟骨パーティクルが合体を開始した7日目では、軟骨膜様膜のみが合体し、軟骨パーティクルの軟骨は膜によって分離されていた。合体部における軟骨膜様膜の一部は、14、28および56日目に厚くなった(図2、右列)。これは軟骨膜様膜の細胞が増殖したことを示唆している。14日目に、2つの軟骨パーティクルの軟骨の一部が接触しているのがわかった。接触は28日目に実質的になり、軟骨は56日目および84日目までに合体した(図2、第2列、第3列)。
統計解析を行うために、各タイムポイントにおける軟骨パーティクルの合体の程度にグレードを付けた。合体なしをグレード0、軟骨膜様膜の合体をグレード1、軟骨の合体をグレード2とした(表2)。グレードはノンパラメトリックデータであるため、Steel-Dwass検定を使用して統計分析を行った。全てのサンプルは、0日目にグレード0(合体なし)、56日目にグレード2(軟骨の合体)であった。7日目以降のタイムポイントにおけるグレードは、0日目のグレードと有意に異なっていた(表3)。14日目以前のタイムポイントにおけるグレードは、56日目のグレードと有意に異なっていた(表3)。
ヒトiPS細胞株QHJIと異なるヒトiPS細胞株で作製した軟骨パーティクルも同様に合体することを確認するために、ヒトiPS細胞株409B2、604B1および1231A3から軟骨パーティクルを作製した。2個の軟骨パーティクルを培養ディッシュ内で接触した状態にして培養すすると、それらは徐々に自発的に合体し、60日目の組織切片のサフラニンO-ファストグリーン-鉄ヘマトキシリン染色標本では、組織学的にも合体が確認された(図4)。
取得した軟骨パーティクル合体のコマ撮り画像は、軟骨膜様膜の細胞が他の軟骨パーティクルの軟骨膜様膜に移動したことを示した。この観察結果は、軟骨膜様膜が合体において実質的な役割を果たすことを示唆している。
軟骨膜様膜を剥がした軟骨パーティクルの組織学的分析により、軟骨膜様膜と中心軟骨の分離が確認された(図5)。RNAを軟骨膜様膜および中心軟骨から別々に抽出し、RNA配列分析に供した。COL2A1およびSOX9のRPKM値は、軟骨膜様膜よりも中心軟骨の方が高かったが、COL1A1のRPKM値はその逆であった(表4)。この結果は、サンプリングとRNAシーケンス実験がうまく行われたことを示している。発現量が異なる遺伝子の中で、FGF18 mRNAは中心軟骨より軟骨膜様膜においてより高発現していた(表4、5、図6)。リアルタイムRT-PCR発現解析により、これらの遺伝子の発現が軟骨膜様膜と中心軟骨とで異なることを確認した(図7)。
培地へのFGF18の添加は軟骨パーティクルの合体を促進した。14日目において、溶媒群(Vehicle)では最低限の軟骨パーティクルの合体を示したが、FGF18群では実質的な合体を示した(図8)。組織学的には、FGF18群では軟骨膜様膜による厚い結合を示した。14日目において、溶媒群とFGF18群の軟骨パーティクルの合体グレードに有意差が見られた(表6)。
実施例1の材料および方法(2)と同じ方法でヒトiPS細胞株QHJIから軟骨パーティクルを作製し、分化誘導開始から4週間後の軟骨パーティクルを使用した。生検サンプル保存用のメッシュバッグ(栄研化学、商品名:サンプルバッグ、品番:KA1000、45 mm×74 mm)に軟骨パーティクルを入れた。軟骨パーティクル量は、軟骨パーティクルをメッシュバッグの片隅に寄せ、メッシュバッグの面積(30 mm×50 mm)の30%〜40%になる量とした。実施例1の材料および方法(3)に記載の軟骨形成培地を入れたバイオリアクター(BWV-S03A、エイブル)中に軟骨パーティクルを収容したメッシュバッグを投入し、37 ℃、CO2 5%条件下で、培養液を攪拌しながら4週間培養を行った。メッシュバッグから軟骨パーティクルを取り出したところ、板状に合体した軟骨が形成されていた(図10)。図11は、得られた板状軟骨の組織切片をサフラニンO-ファストグリーン-鉄ヘマトキシリンで染色した図である。サフラニンOで染色される軟骨ECM中に細胞が散在する軟骨の組織像を呈していた。
実施例1の材料および方法(2)と同様の方法でヒトiPS細胞株409B2から軟骨パーティクルを作製した。ただし、本実施例ではrosuvastatinは添加しなかった。分化誘導開始から8週間後の軟骨パーティクルを使用した。培養用3.5 cmディッシュに実施例1の材料および方法(3)に記載の軟骨形成培地を入れ、その中に星形の枠を置き、枠内に軟骨パーティクルをすき間なく置き、培養用ディッシュを静置して、37 ℃、CO2 5%条件下で16週間培養を行った。枠内の軟骨パーティクルは合体して星形の板状軟骨が形成されていた(図12)。
実施例1の材料および方法(2)と同じ方法でヒトiPS細胞株QHJIから軟骨パーティクルを作製し、分化誘導開始から4週間後の軟骨パーティクルを使用した。ポリカーボネート製トランスウェルおよびポリエステル製トランスウェルを用いた。24ウェルプレート(Corning)の各ウェルに実施例1の材料および方法(3)に記載の軟骨形成培地を入れ、トランスウェル(メンブレン径6.5 mm)を各ウェルに設置し、その中に軟骨パーティクルをすき間なく置き、プレートを静置して、37 ℃、CO2 5%条件下で5週間培養を行った。枠内の軟骨パーティクルは合体して板状軟骨が形成されていた(図13)。
