Claims (42)
- 1.特定の刺激に応答して色素を分散もしくは凝集させる能力及びGPCリセプ ターをコードする外来クローンを発現する能力を有する色素細胞ラインの試験細 胞に、外来GPCリセプターの活性化が色素の分散を誘発する場合には該試験細 胞内で色素が凝集するような色素配置の初期状態をセットする刺激を導入し、あ るいは外来GPCリセプターの活性化が色素の凝集を誘発する場合には該試験細 胞内で該色素が分散するような色素配置の初期状態をセットする刺激を導入し該 初期色素配置状態にセットされた試験細胞を試験化学物質と接触させ;そして該 化学物質で処理された試験細胞内の色素配置が初期色素配置状態から変化したか 否かを調べ、外来GPCリセプターを発現する該試験細胞内に観察される色素配 置の変化を、該化学物質が該外来GPCリセプターのアゴニストであることを示 唆するものとすることよりなる;GPCリセプターに対するアゴニストとして作 用する化学物質を同定する方法。
- 2.試験細胞が、Xenopus1aevisのメラノフォアであり、そして刺 激が化学物質または光である、請求項1記載の方法。
- 3.外来リセプターの活性化が色素の分散を誘発する場合には刺激がメラトニン であり、または外来リセプターの活性化が色素の凝集を誘発する場合には刺激が 光、メラノサイト刺激ホルモンまたはベータ1−アドレナリン性リセプターアゴ ニストである、請求項1記載の方法。
- 4.特定の刺激に応答して色素を分散または凝集する能力を有し且つ該リセプタ ーをコードする外来クローンを発現しないコントロール細胞に、外来GPCリセ プターの活性化が色素の分散を誘発する場合には該コントロール細胞内で色素が 凝集するような色素配置の初期状態をセットする刺激を導入し、あるいは外来G PCリセプターの活性化が色素の凝集を誘発する場合には該コントロール細胞内 で該色素が分散するような色素配置の初期状態をセットする刺激を導入し;該初 期色素配置状態にセットされたコントロール細胞を試験化学物質と接触させ;そ して該化学物質で処理されたコントロール細胞内の色素配置が初期色素配置状態 から変化したか否かを調べ、該化学物質と接触したコントロール細胞内の色素配 置に変化が観察されず試験細胞には変化が見られるときは該化学物質が外来GP Cリセプターのアゴニストであるとみなす、工程からなるコントロール操作をさ らに含む、請求項1記載の方法。
- 5.特定の刺激に応答して色素を分散もしくは凝集させる能力及びGPCリセプ ターをコードする外来クローンを発現する能力を有する色素細胞ラインの試験細 胞に、外来GPCリセプターの活性化が色素の分散を誘発する場合には該試験細 胞内で色素が凝集するような色素配置の初期状態をセットする第一の刺激を導入 し、あるいは外来GPCリセプターの活性化が色素の凝集を誘発する場合には該 試験細胞内で該色素が分散するような色素配置の初期状態をセットする第一の刺 激を導入し;該初期色素配置状態にセットされた試験細胞を同定すべき化学物質 と接触させ;該細胞を観察して色素配置の状態が変化していないことを調べ;同 定すべき化学物質と接触させた該試験細胞に、外来GPCリセプターの活性化が 色素の分散を誘発する場合には外来GPCリセプターの活性化により色素の分散 が誘発されるような第二の刺激を加え、あるいは該外来GPCリセプターの活性 化が色素の凝集を誘発する場合には該外来GPCリセブターの活性化により色素 の凝集が誘発されるような第二の刺激を加え;そして第二の刺激を加えた試験細 胞内の色素配置が初期色素配置状態から変化したかどうかを調べることよりなる 、GPCリセプターのアンタゴニストとして作用する化学物質を同定する方法。
- 6.試験細胞が、Xenopus1aevisのメラノフォアであり、そして刺 激が化学物質または光である、請求項5記載の方法。
- 7.外来リセプターの活性化が色素の分数を誘発する場合には第一の刺激がメラ トニンであり、または外来リセプターの活性化が色素の凝集を誘発する場合には 第一の刺激が光、メラノサイト刺激ホルモンまたはベータ1−アドレナリン性リ セプターアゴニストである、請求項5記載の方法。
- 8.特定の刺激に応答して色素を分散もしくは凝集させる能力及びGPCリセプ ターをコードする外来クローンを発現する能力を有する色素細胞ラインのコント ロール細胞に、外来GPCリセプターの活性化が色素の分散を誘発する場合には 該コントロール細胞内で色素が凝集するような色素配置の初期状態をセットする 第一の刺激を導入し、あるいは外来GPCリセプターの活性化が色素の凝集を誘 発する場合には該コントロール細胞内で該色素が分散するような色素配置の初期 状態をセットする第一の刺激を導入し;該初期色素配置状態にセットされたコン トロール細胞を同定すべき化学物質と接触させ;該細胞を観察して色素配置の状 態が変化していないことを調べ;同定すべき化学物質と接触させた該コントロー ル細胞に、外来GPCリセブターの活性化が色素の分散を誘発する場合には内因 GPCリセプターの活性化により色素の分散が誘発されるような第二の刺激を加 え、あるいは該外来GPCリセプターの活性化が色素の凝集を誘発する場合には 内因GPCリセプターの活性化により色素の凝集が誘発されるような第二の刺激 を加え;そして第二の刺激を加えたコントロール細胞内の色素配置が初期色素配 置状態から変化したか否かを調べ、コントロール細胞内の色素配置が変化し試験 細胞に変化が見られない場合は第一の刺激は該外来リセプターに対する特異的ア ンタゴニストであるとみなす、工程からなるコントロール操作をさらに含む、請 求項5記載の方法。
- 9.特定の刺激に応答して色素を分散もしくは凝集させる能力及びPPG蛋白質 をコードする外来クローンを発現する能力を有する色素細胞ラインの試験細胞に 、外来PPG蛋白質の活性化が色素の分散を誘発する場合には該試験細胞内で色 素が凝集するような色素配置の初期状態をセットする刺激を導入し、あるいは外 来PPG蛋白質の活性化が色素の凝集を誘発する場合には該試験細胞内で該色素 が分散するような色素配置の初期状態をセットする刺激を導入し;該初期色素配 置状態にセットされた試験細胞を試験化学物質と接触させ;そして該化学物質で 処理された試験細胞内の色素配置が初期色素配置状態から変化したか否かを調べ 、外来GPCリセプターを発現する該試験細胞内に観察される色素配置の変化を 、該化学物質が該外来GPCリセプターのアゴニストであることを示唆するもの とすることよりなる;PPG蛋白質に対するアゴニストとして作用する化学物質 を同定する方法。
- 10.試験細胞が、Xenops1aevisのメラノフォアであり、そして刺 激が化学物質または光である、請求項9記載の方法。
- 11.外来PPG蛋白質の活性化が色素の分散を誘発する場合には刺激がメラト ニンであり、または外来PPG蛋白質の活性化が色素の凝集を誘発する場合には 刺激が光、メラノサイト刺激ホルモンまたはベータ1−アドレナリン性リセプタ ーアゴニストである、請求項9記載の方法。
- 12.特定の刺激に応答して色素を分散または凝集する能力を有し且つ該PPG 蛋白質をコードする外来クローンを発現しないコントロール細胞に、外来PPG 蛋白質の活性化が色素の分散を誘発する場合には該コントロール細胞内で色素が 凝集するような色素配置の初期状態をセットする刺激を導入し、あるいは外来P PG蛋白質の活性化が色素の凝集を誘発する場合には該コントロール細胞内で該 色素が分散するような色素配置の初期状態をセットする刺激を導入し;該初期色 素配置状態にセットされたコントロール細胞を試験化学物質と接触させ;そして 該化学物質で処理されたコントロール細胞内の色素配置が初期色素配置状態から 変化したか否かを調べ、核化学物質と接触したコントロール細胞内の色素配置に 変化が観察されず試験細胞には変化が見られるときは該化学物質が外来PPG蛋 白質のアゴニストであるとみなす、工程からなるコントロール操作をさらに含む 、請求項9記載の方法。
- 13.特定の刺激に応答して色素を分散もしくは凝集させる能力及びPPG蛋白 質をコードする外来クローンを発現する能力を有する色素細胞ラインの試験細胞 に、外来PPG蛋白質の活性化が色素の分散を誘発する場合には該試験細胞内で 色素が凝集するような色素配置の初期状態をセットする第一の刺激を導入し、あ るいは外来PPG蛋白質の活性化が色素の凝集を誘発する場合には該試験細胞内 で該色素が分散するような色素配置の初期状態をセットする第一の刺激を導入し ;該初期色素配置状態にセットされた試験細胞を同定すべき化学物質と接触させ :該細胞を観察して色素配置の状態が変化していないことを調べ;同定すべき化 学物質と接触させた談試験細胞に、外来PPG蛋白質の活性化が色素の分散を誘 発する場合には外来PPG蛋白質の活性化により色素の分散が誘発されるような 第二の刺激を加え、あるいは該外来PPG蛋白質の活性化が色素の凝集を誘発す る場合には該外来PPG蛋白質の活性化により色素の凝集が誘発されるような第 二の刺激を加え:そして第二の刺激を加えた試験細胞内の色素配置が初期色素配 置状態から変化したか否かを調べることよりなる、PPG蛋白質のアンタゴニス トとして作用する化学物質を同定する方法。
- 14.試験細胞が、Xenopus1aevisのメラノフォアであり、そして 刺激が化学物質または光である、請求項13記載の方法。
- 15.外来PPG蛋白質の活性化が色素の分散を誘発する場合には第ーの刺激が メラトニンであり、または外来PPG蛋白質の活性化が色素の凝集を誘発する場 合には第一の刺激が光、メラノサイト刺激ホルモンまたはベータ1−アドレナリ ン性リセプターアゴニストである、請求項13記載の方法。
- 16.特定の刺激に応答して色素を分散もしくは凝集させる能力及びPPG蛋白 質をコードする外来クローンを発現する能力を有する色素細胞ラインのコントロ ール細胞に、外来PPG蛋白質の活性化が色素の分散を誘発する場合には該コン トロール細胞内で色素が凝集するような色素配置の初期状態をセットする第一の 刺激を導入し、あるいは外来PPG蛋白質の活性化が色素の凝集を誘発する場合 には該コントロール細胞内で該色素が分散するような色素配置の初期状態をセッ トする第一の刺激を導入し;該初期色素配置状態にセットされたコントロール細 胞を同定すべき化学物質と接触させ;該細胞を観察して色素配置の状態が変化し ていないことを調べ;同定すべき化学物質と接触させた該コントロール細胞に、 外来PPG蛋白質の活性化が色素の分散を誘発する場合には内因PPG蛋白質の 活性化により色素の分散が誘発されるような第二の刺激を加え、あるいは該外来 PPG蛋白質の活性化が色素の凝集を誘発する場合には内因PPG蛋白質の活性 化により色素の凝集が誘発されるような第二の刺激を加え;そして第二の刺激を 加えたコントロール細胞内の色素配置が初期色素配置状態から変化したか否かを 調べ、コントロール細胞内の色素配置が変化した場合は第一の刺激は該外来PP G蛋白質に対する特異的アンタゴニストであるとみなす、工程からなるコントロ ール操作をさらに含む、請求項13記載の方法。
- 17.特定の刺激に応答して色素を分散または凝集する能力を有し、そして内在 セロトニンリセプターを発現する能力を有する色素細胞ラインの試験細胞に刺激 を導入し;色素凝集の初期状態にセットした該試験細胞を化学物質と接触させ該 化学物質で処理した該試験細胞内で色素が分散したか否かを調べ、適当なコント ロールと比べて色素が分散したときは該化学物質はセロトニンリセプターのアゴ ニストであると判断することよりなる、メラノフォアに関して内在性のセロトニ ンリセプターのアゴニストとして作用する化学物質を同定する方法。
- 18.刺激がメラトニンである、請求項17記載の方法。
- 19.特定の刺激に応答して色素を分散または凝集する能力を有し、そして内在 性セロトニンリセプターを発現する能力を有する色素細胞ラインの試験細胞に第 一の刺激を導入して、該試験細胞内で色素が凝集するような初期色素配置状態に セットし:該色素凝集初期状態にセットされた試験細胞を同定すべき化学物質と 接触させ;該細胞を観察してその色素配置状態が変化していないことを調べ;該 同定すべき化学物質と接触させた試験細胞に第二の刺激を与えて内在性セロトニ ンリセプターを活性化して色素分散を誘発させ;第二の刺激を加えた試験細胞内 の色素が分散したかを調べ、色素分散の欠如を試験化学物質がセロトニンリセプ ターのアンタゴニストであることの示唆とすることよりなる、メラノフォアに関 して内在性のセロトニンリセプターのアンタゴニストとして作用する化学物質を 同定する方法。
- 20.第一刺激がメラトニンであり、そしてコントロール操作として次の工程即 ち、試験細胞に、内在性セロトニンリセブターを活性化する前記第二刺激の代わ りに、セロトニンリセプターと同様に活性化された時色素分散を誘発する別の内 在性GPCリセプターを活性化する第二刺激を導入し、試験細胞に色素分散が見 られずコントロール細胞が色素分散を生じた時は、同定すべき化学物質は該内在 性セロトニンリセプターのアンタゴニストであるとする工程、をさらに含む、請 求項19記載の方法。
- 21.GPCリセプターがMSHリセプターである請求項20記載の方法。
- 22.特定の刺激に応答して色素を分散または凝集する能力を有し、そして内在 性PPG蛋白質を発現する能力を有する色素細胞ラインの試験細胞に刺激を導入 し;色素凝集の初期状態にセットした該試験細胞を化学物質と接触させ;該化学 物質で処理した該試験細胞内で色素が分散したか否かを調べ、適当なコントロー ルと比べて色素の配置が変化したときは該化学物質は内在性PPG蛋白質のアゴ ニストであると判断することよりなる、メラノフォアに関して内在性のPPG蛋 白質のアゴニストとして作用する化学物質を同定する方法。
- 23.PPG蛋白質が、キナーゼA、キナーゼC、ホスホリパーゼC、ホスホリ バーゼA2及びG−蛋白質からなる群から選択され、そして刺激がメラトニンで ある、請求項22記載の方法。
- 24.特定の刺激に応答して色素を分散または凝集する能力を有し、そして内在 性PPG蛋白質を発現する能力を有する色素細胞ラインの試験細胞に、PPG蛋 白質の活性化が色素の分散を誘発する場合には該試験細胞内で色素が凝集するよ うな色素配置の初期状態をセットする第一の刺激を導入し、あるいは内在PPG 蛋白質の活性化が色素の凝集を誘発する場合には該試験細胞内で該色素が分散す るような色素配置の初期状態をセットする第一の刺激を導入し;該初期色素配置 状態にセットされた試験細胞を同定すべき化学物質と接触させ;該細胞を観察し て色素配置の状態が変化していないことを調べ;同定すべき化学物質と接触させ た該試験細胞に、内在PPG蛋白質の活性化が色素の分散を誘発する場合には内 在PPG蛋白質の活性化により色素の分散が誘発されるような第二の刺激を加え 、あるいは該内在PPG蛋白質の活性化が色素の凝集を誘発する場合には該内在 PPG蛋白質の活性化により色素の凝集が誘発されるような第二の刺激を加えそ して第二の刺激を加えた試験細胞内の色素配置が初期色素配置状態から変化した か否かを調べることよりなる、メラノフォアに関して内在性のPPG蛋白質のア ンタゴニストとして作用する化学物質を同定する方法。
- 25.PPG蛋白質が、プロテインキナーゼA、プロテインキナーゼC、ホスホ リバーゼC、ホスホリバーゼA2及びG−蛋白質からなる群から選択される、請 求項24記載の方法。
- 26.カエルのような下等動物由来のinvitroで連続増殖可能な色素細胞 に外来核酸クローンを導入し;該細胞に内在GPCリセプターを活性化して該細 胞内の色素配置を初期状態にセットする刺激を導入し:該色素の初期配置状態に セットされた細胞を該外来性リセプターを活性化する化学物質に接触させ;そし て該化学物質で処理された細胞のうち色素配置が初期状態から変化したものを同 定し、色素配置の変化を該リセプターをコードする外来クローンを発現する細胞 の指標とする、ことよりなるGPCリセプターのクローニング方法。
- 27.外来核酸クローンが、プラスミドベクター内に造成されたcDNAライブ ラリーからのものであり、そして色素細胞にエレクトロポレーションにより請人 される請求項26記載の方法。
- 28.一つまたはそれ以上の容器に、 下記リセプターをコードする外来クローンを発現する下等動物色素試験細胞;及 び該外来リセプターの活性化が色素の分散を誘発する場合には内在性リセブター の活性化により色素の凝集を誘発する刺激剤、および/または該外来リセブター の活性化が色素の凝集を誘発する場合には内在性リセプターの活性化により色素 の分散を誘発する刺激剤、 を含有してなる、化学物質が外来GPCリセプターのアゴニストとして作用する かどうかを決定するためのキット。
- 29.試験色素細胞がXenopus1aevisのメラノフォアであり、外来 リセプターの活性化が色素の分散を誘発する場合には刺激剤がメラトニンであり 、そして外来リセプターの活性化が色素の凝集を誘発する場合には刺激剤がメラ ノサイト刺激ホルモンまたはイソプロテレノールである、請求項28記載のキッ ト。
- 30.一つまたはそれ以上の容器に、 下記リセプターをコードする外来クローンを発現する下等動物色素試験細胞;外 来リセプターの活性化が色素の分散を誘発する場合には内在性リセプターを活性 化して色素の凝集を誘発する第一の刺激剤、および/または外来リセプターの活 性化が色素の凝集を誘発する場合には内在性リセプターを活性化して色素の分散 を誘発する第一の刺激剤;及び 外来リセプターの活性化が色素の分散を誘発する場合には該外来リセプターを活 性化して色素の分散を誘発する第二の刺激剤、および/または外来リセプターの 活性化が色素の凝集を誘発する場合には該外来リセプターを活性化して色素の凝 集を誘発する第二の刺激剤、 を含有してなる、化学物質が外来GPCリセプターのアンタゴニストとして作用 するかどうかを決定するためのキット。

- 32.一つまたはそれ以上の容器に、 下記蛋白質をコードする外来クローンを発現する下等動物色素試験細胞;及び該 外来蛋白質の活性化が色素の分散を誘発する場合には内在性リセプターの活性化 により色素の凝集を誘発する刺激剤、および/または該外来蛋白質の活性化が色 素の凝集を誘発する場合には内在性リセプターの活性化により色素の分散を誘発 する刺激剤、 を含有してなる、化学物質が外来PPG蛋白質のアゴニストとして作用するかど うかを決定するためのキット。
- 33.外来蛋白質の活性化が色素の分散を誘発する場合には刺激剤がメラトニン であり、そして外来リセプターの活性化が色素の凝集を誘発する場合には刺激剤 がメラノサイト刺激ホルモンまたはイソプロテレノールである、請求項32記載 のキット。
- 34.一つまたはそれ以上の容器に、 下記蛋白質をコードする外来クローンを発現する下等動物色素試験細胞;外来蛋 白質の活性化が色素の分散を誘発する場合には内在性リセプターを活性化して色 素の凝集を誘発する第一の刺激剤、および/または外来蛋白質の活性化が色素の 凝集を誘発する場合には内在性リセプターを活性化して色素の分散を誘発する第 一の刺激剤;及び 外来蛋白質の溶性化が色素の分散を誘発する場合には該外来蛋白質を活性化して 色素の分散を誘発する第二の刺激剤、および/または外来蛋白質の活性化が色素 の凝集を誘発する場合には該外来蛋白質を活性化して色素の凝集を誘発する第二 の刺激剤、 を含有してなる、化学物質が外来PPG蛋白質のアンタゴニストとして作用する かどうかを決定するためのキット。
- 35.外来蛋白質の活性化が色素の分散を誘発する場合には第一の刺激剤がメラ トニンであり、そして外来蛋白質の活性化が色素の凝集を誘発する場合には第一 の刺激剤がメラノサイト刺激ホルモンまたはイソプロテレノールである、請求項 34記載のキット。
- 36.一つまたはそれ以上の容器に、 下記リセプターをコードする外来クローンを発現する下等動物色素試験細胞;及 び下記化学物質の榛的である下記内在性リセプターの活性化が色素の分散を誘発 する場合には内在性リセプターの活性化により色素の凝集を誘発する刺激剤、お よび/または下記化学物質の標的である該内在性リセプターの活性化が色素の凝 集を誘発する場合には内在住リセブターの活性化により色素の分散を誘発する刺 激剤、 を含有してなる、化学物質が内在GPCリセプターのアゴニストとして作用する かどうかを決定するためのキット。
- 37.内在性蛋白質の活性化が色素の分散を誘発する場合には刺激剤がメラトニ ンであり、または内在性蛋白質の活性化が色素の凝集を誘発する場合には刺激剤 がメラノサイト刺激ホルモンまたはイソプロテレノールである、請求項36記載 のキット。
- 38.一つまたはそれ以上の容器に、 下記リセプターをコードする外来クローンを発現する下等動物色素試験細胞;化 学物質の標的となる内在リセプターの活性化が色素の分散を誘発する場合には内 在性リセプターを活性化して色素の凝集を誘発する第一の刺激剤、及び/または 化学物質の標的となる内在リセプターの活性化が色素の凝集を誘発する場合には 内在性リセプターを活性化して色素の分散を誘発する第一の刺激剤;及び化学物 質の標的となる内在リセプターの活性化が色素の分散を誘発する場合には核化学 物質の標的となる内在リセプターを活性化して色素の分散を誘発する第二の刺激 剤、および/または化学物質の標的となる内在リセプターの活性化が色素の凝集 を誘発する場合には核化学物質の標的となる内在リセプターを活性化して色素の 凝集を誘発する第二の刺激剤、を含有してなる、化学物質が内在GPCリセプタ ーのアンタゴニストとして作用するかどうかを決定するためのキット。
- 39.内在性リセプターの活性化が色素の分散を誘発する場合には刺激剤がメラ トニンであり、または内在性リセプターの活性化が色素の凝集を誘発する場合に は刺激剤がメラノサイト刺激ホルモンまたはイソプロテレノールである、請求項 38記載のキット。
- 40.一つまたはそれ以上の容器に、 下記蛋白質をコードする外来クローンを発現する下等動物色素試験細胞;及び下 記内在性蛋白質の活性化が色素の分散を誘発する場合には内在性リセブターの活 性化により色素の凝集を誘発する刺激剤、および/または該内在性蛋白質の活性 化が色素の凝集を誘発する場合には内在性リセプターの活性化により色素の分散 を誘発する刺激剤、 を含有してなる、化学物質が内在PPG蛋白質のアゴニストとして作用するかど うかを決定するためのキット。
- 41.内在性PPG蛋白質の活性化が色素の分散を誘発する場合には刺激剤がメ ラトニンであり、または内在性PPG蛋白質の活性化が色素の凝集を誘発する場 合には刺激剤がメラノサイト刺激ホルモンまたはイソプロテレノールである、請 求項40記載のキット。
- 42.一つまたはそれ以上の容器に、 下記蛋白質をコードする外来クローンを発現する下等動物色素試験細胞:内在P PG蛋白質の活性化が色素の分散を誘発する場合には内在性リセプターを活性化 して色素の凝集を誘発する第一の刺激剤、及び/または内在PPG蛋白質の活性 化が色素の凝集を誘発する場合には内在性リセプターを活性化して色素の分散を 誘発する第一の刺激剤一及び 内在PPG蛋白質の活性化が色素の分散を誘発する場合には該内在PPG蛋自賛 を活性化して色素の分散を誘発する第二の刺激剤、および/または内在PPG蛋 白質の活性化が色素の凝集を誘発する場合には該内在PPG蛋白質を活性化して 色素の凝築を誘発する第二の刺激剤、を含有してなる、化学物質が内在PPG蛋 白質のアンタゴニストとして作用するかどうかを決定するためのキット。
