様々な態様において、本発明は、MSを有する、MSを発症する危険性がある、及び/またはIL-17アンタゴニストでの治療に応答性である可能性が高い対象を同定する方法を提供する。本方法は、TIMP1、LRG1、CXCL1、CXCL5、CXCL10、IL8、NFκBIZ、YKL40(CHI3L1)、及びG-CSF(CSF3)の群から選択される1つ以上の遺伝子の発現レベルを、対象から取得した生体試料中で測定するステップを含む。本方法は、遺伝子(複数可)の発現レベルを対照遺伝子の発現レベルと比較することを更に含んでもよい。対照と比べて増加した発現レベルは、対象がMSを有する、MSを発症する危険性がある、及び/またはIL-17アンタゴニストでの治療に応答性である可能性が高いことを示す。あるいは、対照と比べて減少した発現レベル、または同じ発現レベルは、対象がMSを有しない、MSを発症する危険性がない、及び/またはIL-17アンタゴニストでの治療に応答性である可能性が低いことを示す。
RNA転写物(例えば、mRNA)またはタンパク質の発現レベルを決定する様々な方法としては、遺伝子発現プロファイリング、定量的リアルタイムPCR(qRT-PCR)を含むポリメラーゼ連鎖反応(PCR)、市販の機器により製造業者のプロトコルに従って、例えばAffymetrix GenChip技術を使用して行うことができるマイクロアレイ分析、遺伝子発現の逐次分析(SAGE)(Velculescu et al.,Science 270:484-487(1995)、及びVelculescu et al.,Cell 88:243-51(1997))、MassARRAY、Massively Parallel Signature Sequencing(MPSS)法による遺伝子発現分析(Brenner et al.,Nature Biotechnology 18:630-634(2000))、プロテオミクス、免疫組織化学(IHC)等が挙げられるが、これらに限定されない。本発明の一実施形態では、mRNAは定量化される。更なる実施形態では、かかるmRNA分析は、ポリメラーゼ連鎖反応(PCR)の技術を使用して、またはマイクロアレイ分析によって行われる。PCRを利用する場合、PCRの好ましい形態は、定量的リアルタイムPCR(qRT-PCR)である。

本発明に従ってIL-17、具体的にはIL-17AAと相関するバイオマーカーを発現すると同定された対象は、IL-17アンタゴニストでの治療に応答性である可能性が高い。よって、本発明は、上記のように、TIMP1、LRG1、CXCL1、CXCL5、CXCL10、IL8、NFκBIZ、YKL40(CHI3L1)、及びG-CSF(CSF3)の群から選択される1つ以上の遺伝子のRNA転写物またはそのタンパク質産物の発現レベルを、対象から取得した生体試料中で測定することにより、MSを有するか、MSを発症する危険性がある対象を治療する方法を提供する。本方法は、遺伝子(複数可)の発現レベルを対照遺伝子の発現レベルと比較することを更に含んでもよい。対象と比べて1つ以上の遺伝子の増加した発現レベルを有する対象は、有効量のIL-17アンタゴニストで治療される。

産生の例示的な技術について、本発明に従って使用される抗体が以下に提供される。抗体の産生に使用される抗原は、所望のエピトープを含有する全長、またはその一部であってもよい。あるいは、所望の抗原またはエピトープをそれらの細胞表面で発現する細胞を使用して、抗体を生成してもよい。抗体を生成するのに有用な抗原の他の形態は、当業者には明らかになるであろう。
ポリクローナル抗体は、関連抗原及びアジュバントを複数回、皮下(sc)または腹腔内(ip)注射することにより動物で産生するのが好ましい。二官能性剤または誘導化剤、例えば、マレイミドベンゾイルスルホスクシンイミドエステル(システイン残基を介したコンジュゲーション)、N-ヒドロキシスクシンイミド(リジン残基を介する)、グルタールアルデヒド、コハク酸無水物、SOCl2、またはR1N=C=NR(式中、R及びR1は異なるアルキル基である)を使用し、関連抗原を、免疫する種において免疫原性であるタンパク質、例えば、キーホールリンペットヘモシアニン、血清アルブミン、ウシサイログロブリン、またはダイズトリプシン阻害剤とコンジュゲートすることが有用であり得る。
本明細書におけるモノクローナル抗体を作製する様々な方法が当該技術分野で利用可能である。例えば、モノクローナル抗体は、Kohler et al.,Nature,256:495(1975)に最初に記載されたハイブリドーマ法を使用して、組み換えDNA法(米国特許第4,816,567号)によって作製することができる。

本発明の方法での使用のための材料は、キットの調製に適している。本発明のキットは、本明細書に開示される遺伝子のRNA転写物の発現レベルを定量化するための、1つ以上の遺伝子特異的または遺伝子選択的プローブ及び/またはプライマーを含んでもよい。かかるキットは、任意で、生体試料からのRNAの抽出のための1つ以上の試薬を含有する。キットは、例えば、クロマトグラフカラム、既製マイクロアレイ、緩衝剤、適切なヌクレオチドトリホスページ(triphospage)、逆転写酵素、DNAポリメラーゼ、RNAポリメラーゼを含む方法に利用されるそれぞれ様々な材料または試薬のうちの1つ以上を有する1つ以上の容器と、本明細書に開示されるRNA転写物の発現レベルを定量化するための1つ以上のプローブ及びプライマーとを含んでもよい。
CSF IL17レベル及び他の炎症性メディエーターが、RRMS患者において上昇している
IL-17AA、IL-17FF、及び様々な他のタンパク質を、標準治療の療法を受けている50人のRRMS患者、及び20人の健常ドナー(HD)からの脳脊髄液(CSF)及び血清中で、IL-17A及びIL-17Fの場合にはErenna(登録商標)免疫測定プラットフォーム(Singulex,Inc.)に基づく免疫測定法、または標準サンドイッチ免疫測定法を使用して測定した。IL-17A免疫測定法は、およそ30%のIL-17AFヘテロ二量体との交差反応性を有するIL-17AAホモ二量体サイトカインを明確に測定した一方、IL-17Fアッセイは、IL-17AFまたはAAとの検出可能な交差反応性を有しないIL-17FFホモ二量体のみを測定した。全ての免疫測定法は、モノクローナル捕捉及び検出抗体を用いた。
ヒト脳内皮細胞及びミクログリアにおける生体内でのIL-17応答性遺伝子の上方制御
hCMEC/D3不死化ヒト脳内皮細胞系及びSV40形質転換ヒトミクログリア細胞系を、標準細胞培養プロトコルを使用して増殖し、次いで、TNFa、IL-6プラスsIL-6R、IL-17AA、またはこれらのサイトカインの組み合わせで6時間、刺激した。RNAを次いで細胞から単離し、マイクロアレイ分析を行い、追加のサイトカインを伴って、または伴わずにIL-17によって誘発された遺伝子発現を比較した。
EAEモデルにおける生体内でのRRMSバイオマーカーの同定

RRMSの血清マーカーとしてのG-CSF
G-CSF(CSF3)は、複数の細胞系譜中でIL-17によって調節されるサイトカインであり、好中球増殖の主要因である。G-CSFを、CSFでは検出できなかったが、血清中では検出することができた。よって、G-CSF血清レベルのIL-17AAとの相関を、EAEモデル及びヒト血清試料の両方において調査した。

- 再発寛解型多発性硬化症(RRMS)を有することが疑われるか、またはRRMSを発症する危険性がある哺乳動物対象中で、1つ以上の遺伝子の発現レベルを検出する方法であって、
前記対象から生体試料を取得することと、
TIMP1、LRG1、CXCL1、CXCL5、CXCL10、IL-8、NFκBIZ、YKL40、及びG-CSFの群から選択される1つ以上の遺伝子の発現レベルを、前記生体試料中で測定することと、を含む、前記方法。 - 前記発現レベルが対照と比べて増加している場合、前記対象がRRMSを有するか、またはそれを発症する危険性があると決定することを更に含む、請求項96に記載の方法。
- 前記哺乳動物対象が、ヒト患者である、請求項96または97に記載の方法。
- 前記RRMSが、IL-17の増加したレベルを特徴とする、請求項96~98のいずれか1項に記載の方法。

TIMP1、LRG1、CXCL1、CXCL5、CXCL10、IL-8、NFκBIZ、YKL40、及びG-CSFの群から選択される1つ以上の遺伝子の発現レベルを検出するための1つ以上の試薬を含む少なくとも1つの容器を含む、前記キット。 - 前記1つ以上の試薬が、前記1つ以上の遺伝子のRNA転写物の発現レベル及び/またはタンパク質産物の発現レベルを検出するための試薬を含む、請求項136に記載のキット。
- 前記試薬が、前記1つ以上の遺伝子の遺伝子特異的プライマーまたはプローブを1つ以上含む、請求項137に記載のキット。
- 前記1つ以上の試薬が、前記1つ以上の遺伝子のタンパク質産物の発現レベルを検出するための試薬を含む、請求項136に記載のキット。
- 前記試薬が、前記1つ以上の遺伝子のタンパク質産物に結合する、1つ以上の抗体またはその抗原結合断片を含む、請求項139に記載のキット。
- IL-17アンタゴニストを含む容器と、前記IL-17アンタゴニストを前記対象に投与するためのラベルまたは指示書と、を更に含む、請求項136~140のいずれか1項に記載のキット。
