[0063] 遺伝子発現を促進するか又は阻害するために核酸(NA)ベース治療薬が開発されている。遺伝子中の突然変異とmiRNAプロフィール中の変化が癌や他の疾患の根本原因であると考えられているので、NAベースの薬剤は、根本的な病因に直接作用して、治療ポテンシャルを最大にする可能性がある。NAベース治療薬の非限定的な例には:プラスミドDNA(pDNA)、低分子干渉RNA(siRNA)、小ヘアピンRNA(shRNA)、マイクロRNA(miR)、ミミック(模倣体)、抗miR/アンタゴmiR/miR阻害剤、及びアンチセンスオリゴヌクレオチド(ASO)が含まれる。本明細書に記載するナノ粒子組成物の開発まで、NAベース治療薬の臨床転用は、NAをその細胞内標的へ輸送することが特に難題であること、そしてNAが相対的に不安定で血清や細胞ヌクレアーゼによる分解を受けやすいことから、その実行においていくつかの障害(obstacles)に直面した。さらに、NAの高い陰電荷は細胞膜を通過するその輸送を不可能にして、さらに有用性を制限した。
[0097] このLNの物理特性は、有窓性の腫瘍血管構造において亢進された透過性及び保持(EPR)を促進するが、慣用のLN製剤では、エンドソームからの脱出が依然として課題である。この目的を達成するために、核酸の複合化のために陽荷電の四級又は三級アミンベースのカチオン性脂質を含んでなる脂質ナノ粒子を開発した。四級アミンベースのカチオン性脂質は、永続的な陽電荷を担って、核酸との安定した静電複合体を生成することが可能である。しかしながら、三級アミン−カチオン性脂質は、条件付きでイオン化可能であって、その陽電荷は、大部分がpHによって調節される。本発明で提供するのは、四級アミン−カチオン性脂質と三級アミン−カチオン性脂質の組合せ(QTソーム)を含んでなるLNであって、これは、治療薬剤がエンドソームの内側でLNより放出され得る機序を提供する。QTソームは、条件付きでイオン化可能であって、エンドソームの酸性条件下で、脂質二重層の破壊とオリゴヌクレオチドのエンドソーム放出を促進する。四級アミン−カチオン性脂質は、永続的に帯電していて、脂質とオリゴヌクレオチドの間の強い相互作用を確実にして、それによって安定性を確実にする。三級カチオン性脂質と四級カチオン性脂質の組合せは、それぞれの脂質群が個別ではあり得ない最適のpH応答プロフィールを提供する。QTソームは、細胞をトランスフェクトする場合に、通常のカチオン性リポソームより有効である。
[00101] 本発明でさらに提供するのは、低分子ペプチド性の脂質ナノ粒子(SPLN)であるLNである。ある態様において、SPLNは、抗生物質のグラミシジンを含む。本明細書に記載するのは、これまでトランスフェクション剤として検討されてこなかったグラミシジンのいくつかの変異体(A、B、C、D)である。これらのグラミシジンサブタイプは、保存されたペプチド配列をグラミシジンSと共有するが、グラミシジンA、B、C、及びDは、グラミシジンSが環状構造を形成するのに対し、βヘリックス構造を形成する。故に、グラミシジンA〜Dは、グラミシジンSとは異なる。グラミシジン類は二量体化してイオノフォアを形成して膜融合を促進するが、それによりエンドソームとLNの脂質二重層の不安定化が促進される。従って、グラミシジンを含んでなるSPLNは、核酸ベース治療薬において理想的である。グラミシジンA、B、C、D、又はSのLN中への取込みは、ASOとsiRNAの細胞トランスフェクション効率を有意に高める。
[00106] ヌクレオチドの投与後の血清中での分解は、脂質又はポリマー担体が関与するものであっても、核酸ベース治療薬にとって不断の懸案事項である。ヌクレオチドを分解に対してよりよく防護するために、しばしば、骨格や塩基対の修飾といった、ヌクレオチドへの改変が実行される。しかしながら、これらの修飾は、薬物の低下した活性又は標的外の活性をもたらす場合がある。この問題を克服するために、本発明で提供するのは、DNA分解酵素若しくはRNA分解酵素分解剤を含んでなる脂質ナノ粒子製剤である。特別な態様において、DNA分解酵素若しくはRNA分解酵素分解剤は、プロテイナーゼK(PrK)である。このプロテイナーゼKコーティングは、血清に存在するDNA分解酵素及びRNA分解酵素よりオリゴヌクレオチドを防護する。これを図23に図示する。プロテイナーゼKは、プロテイナーゼKの無い脂質ナノ粒子よりよくsiRNAを防護することができる。プロテイナーゼKの包含は、KB細胞において有意な細胞毒性を伴わずに、血清の存在下でトランスフェクション効率を高める。PrKソームは、きわめて多用途性であって、天然オリゴヌクレオチドと化学修飾オリゴヌクレオチドの両方へ適用可能である。
[00109] 応用に依存して、本明細書に開示する脂質ナノ粒子は、核酸との相互作用の増加、血清安定性の増加、RES媒介性の取込みの低下、標的指向性の送達、又はエンドソーム内部でのpH感受性の放出といった特性に有利になるように設計してよい。LN製剤の多様な性質の故に、特別な治療目標を達成するには、本発明で提供するいくつかの方法のいずれも適用してよい。具体的な目標に応じるために、カチオン性脂質、アニオン性脂質、PEG−脂質、中性脂質、膜融合性脂質、アミノ脂質、カチオン性ポリマー、アニオン性ポリマー、ポリマーコンジュゲート、ペプチド、標的化部分、並びにこれらの組合せを適用してよい。

[00138] 様々なSPLN−G製剤について、トランスフェクション効率を分析した:SPLN−G50(40:5:50:5のモル比でのDMHDA、DOTAP、GRAM、TPGS)、SPLN−G35(40:5:35:15:5のモル比でのDMHDA、DOTAP、GRAM、DOPE、TPGS)、SPLN−G30(40:5:30:20:5のモル比でのDMHDA、DOTAP、GRAM、DOPE、TPGS)、SPLN−G20(40:5:20:30:5のモル比でのDMHDA、DOTAP、GRAM、DOPE、TPGS)、SPLN−G10(40:5:10:40:5のモル比でのDMHDA、DOTAP、GRAM、DOPE、TPGSS)、及びSPLN−G0(40:5:50:5のモル比でのDMHDA、DOTAP、DOPE、TPGS)。いずれの脂質も Avanti Polar Lipids(アメリカ)又はシグマ・アルドリッチ(アメリカ)より購入して、さらに精製せずに使用した。脂質とペプチドをエタノールに溶かして、適正な比で合わせて、LNを生成した。追加のエタノールとクエン酸緩衝液を加えて、30%の最終エタノール含量とした。この2.0mg/mLのLN溶液をsiRNA又はODNと15:1のN:Pで合わせてそのまま室温で15分間反応させてから、さらに希釈した。この製剤の粒径は、100〜300nmの間に及んだ。脱イオン水で希釈してc−myb ODNと合わせたこの製剤のゼータ電位測定値(図3)は、グラミシジン含有製剤では、5〜15mVの間に及ぶゼータ電位を示した。これは、35mVを超えるゼータ電位を示した陽性対照のリポフェクタミン2000と対照的である。
[00142] EDC/NHS(1:5:10:5、DOPE:LA:EDC:NHS)を使用してラクトビオン酸をDOPEと架橋連結させることによって、ラクトシル化DOPE(L−DOPE)を生成した。脂質とペプチド(45:5:5:10:28:2:10のモル比でのDODAP:DOTAP:L−DOPE:DMG−PEG:グラミシジン)を1X PBS中で合わせた。L−DOPE(DOPEの代用)及び/又はグラミシジンを用いて、他の対照製剤を完成させた。この製剤化LNの粒径(図8)は、50nmと150nmの間へ低下した。この製剤は、30日間にわたってコロイド安定性を示した。このLNのゼータ電位(図8)は、−10mVと10mVの間に及んだ。ODNローディング効率の検討により、75%を超える濃縮が明らかになった。
[00146] 脂質(60:35:5のモル比でのDDAB、CHOL、Tween80)を100%エタノールに溶かした。この溶液の100μLを900μLの1X PBSに希釈した。ゲル移動度シフト分析(図13)のためにLNを様々なw/w比でLOR−1284(Dharmacon より購入したsiRNA)(0.1μg)と合わせた。1:8(siRNA:LN)で遅滞が生じた。下方調節試験のためにLNを0.1μM siRNAと合わせた。LN−siRNA LOR−1281製剤によるRNR R2の下方調節について、対照としてのアクチンと1:20及び1:30(siRNA:LN)のw/w比を使用するRT−PCRによって評価した。RPMI 1640培地において5% CO2雰囲気下に37℃で増殖させたKB細胞を、トランスフェクションに先立つ24時間前に、6ウェルプレートにおいてウェルにつき3.0x105個の細胞密度で蒔いた。細胞をほぼ80%の集密度まで増殖させて、血清含有培地を取り出した。細胞を1000μLのトランスフェクション培地でトランスフェクトして、4時間処理した。10%血清含有RPMI 1640培地の存在下でトランスフェクションが生じた。実験は、同一3検体で実施した。処理が完了した後で、細胞を1X PBSで洗浄し、血清含有RPMI 1640を戻した。処理が完了してから48時間後、細胞について、アクチンをハウスキーピング遺伝子とするRT−PCRによって、RNR R2発現レベルを分析した。結果を図14に示す。1:30(siRNA:LN)の製剤で、有意な下方調節が生じた。
[00150] SPLN−G20について、MDA−MB−468細胞中への抗miR送達を試験した。SPLN−G20を上記のように調製した。MDA−MB−468(トリプルネガティブ乳癌)細胞を、トランスフェクションに先立つ24時間前に、6ウェルプレートにおいて、1%ペニシリン/ストレプトマイシンと10% FBSを補充したDMEM/F12培地中2x104個/cm2の細胞密度で蒔いた。SPLN−G20を抗miR−221と合わせて、miR−221の下流標的であるp27/Kip1(腫瘍サプレッサー)を上方調節するその能力を評価した。抗miR−221の配列は、以下の通りであった:5’−gsasaacccagcagacaaugusasgscsu−Chol−3’[配列番号1]。この配列には、アンチセンスオリゴヌクレオチドのヌクレアーゼ安定性に役立つ2’−O−メチル−修飾化オリゴヌクレオチド(小文字)とホスホロチオエート連結(s下付き文字)が含まれた。さらに、疎水性部分(コレステロール(Chol))の3’端への付加部分を加えて、脂質ナノ粒子製剤との会合をさらに促進した。20%血清の存在下にSPLN−G20を50、100、及び250nMの抗miR−221とともに使用してMDA−MB−468細胞をトランスフェクトした。処理をそのまま4時間進行させて、この時点でトランスフェクション培地を取り出して、新鮮な培地(10% FBSを補充した)と交換した。細胞をそのままさらに44時間増殖させた後で、RT−PCRを開始した。細胞からのRNAを TRIzol 試薬(Life Technologies)によって抽出して、製造業者の説明書に従ってSuperScript(登録商標)III First-Strang Synthesis System(Life Technologies)によって、cDNAを産生した。次いで、SYBRグリーン(Life Technologies)とp27/kip1用のプライマー(Alpha DNA)を使用して、RT−PCRを実施した。

- 三級アミン−カチオン性脂質が、DODAP、DODMA、DC−CHOL、N,N−ジメチルヘキサデシルアミン、又はこれらの組合せより選択される、請求項1の脂質ナノ粒子。
- 四級アミン−カチオン性脂質が、DOTAP、DOTMA、DDAB、又はこれらの組合せより選択される、請求項1の脂質ナノ粒子。
