「エピトープ」という用語は、抗体に対する特異的結合が可能なタンパク質決定因子を意味する。エピトープは通常、アミノ酸又は糖側鎖などの化学的に活性な分子表面基で構成されており、通常、特定の3次元構造特性並びに特定の電荷特性を有する。構造的及び非構造的エピトープは、後者ではなく前者に対する結合が、変性溶媒の存在下で損失するという点で区別される。
特性
本発明は、10H10(Edgingtonら、米国特許第5,223,427号)として知られるヒトTFに結合する、凝固を阻害しないマウス抗体は、特定の細胞におけるTFのシグナル伝達を抑制することができるという予想外の発見に基づいている(Ahmedら、2006、先に引用、PCT国際公開特許第2007/056352A2号)。したがって、本発明の抗体は、マウス抗体10H10の結合エピトープを保持するものであり、このマウス抗体10H10は、FVIIa結合に関して組織因子と競合せず、TF−VIIa複合体の凝固を促進するアミド分解活性を実質的に阻害し、並びに、TF−VIIaによって媒介されるシグナル伝達、及びサイトカインIL−8の放出などの下流の発癌作用を阻害する。本発明の抗体は、IMGTデータベースに記載されているヒト生殖細胞系列IgG遺伝子に適合し、ヒト血漿中のカルシウムの存在下で凝固を開始させるTFの能力を妨げずに、ヒトTFに結合し続ける。
組換え方法によって生産された治療用モノクローナル抗体の使用が広がるにつれて、こうした複合体組成物の特徴及び特性が探求されてきた。免疫特異性及び抗原標的化の特徴は、一般に、CDRとしても知られる高頻度可変領域のループ末端などの可変ドメイン及びサブドメインに存在するが、複合体は、IgGのFc部分などの定常ドメインによって形成される構造体によって提供される他の受容体及び血清成分と相互作用する。
本出願に記載される抗体の特徴及び生物活性を有する抗体は、任意の哺乳類、例えば、非限定的にヒト、マウス、ウサギ、ラット、げっ歯類、霊長類、ヤギ、若しくはそれらの任意の組み合わせを含むか、又はそれらに由来することができ、単離されたヒト、霊長類、げっ歯類、哺乳類、キメラ、ヒト又は霊長類適応抗体、免疫グロブリン、開裂生成物、及び他の特定部分、並びにそれらの変異型を含む。モノクローナル抗体は、ハイブリドーマ技術(Kohler and Milstein,1975,Nature,256:495〜497)、及びB細胞と融合する不死化融合パートナーを用いる関連方法など、当該技術分野において周知の任意の方法によって調製され得る。本発明で用いる抗体はまた、例えば、Babcook,J.ら、1996,Proc.Natl.Acad.Sci.USA 93(15):7843〜78481、国際公開特許第92/02551号、同第2004/051268号、及びPCT国際特許出願第2004/10637号に記載される方法による特異抗体の生成に関して選択される1個のリンパ球から生成する免疫グロブリン可変領域cDNAsのクローニング及び発現に基づいた単一リンパ球抗体法を用いて産生することもできる。
本発明は更に、ヒトTFに結合するヒト免疫グロブリン(又は抗体)を提供する。これらの抗体はまた、遺伝子操作を受けた又は適応されたとして特徴付けされ得る。免疫グロブリンは、実質的にヒト生殖系列免疫グロブリンからの可変領域(複数可)を有し、抗原認識に関与することが知られる残基において、指向された変形、例えば、KabatのCDR又は構造的に画定されるような高頻度可変ループを含む。存在する場合、定常領域(1つ以上)も、実質的にヒト免疫グロブリンからである。ヒト抗体は、少なくとも約10−6M(1マイクロM)、約10−7M(100nM)、10−9M(1nM)以下のTFに対するKDを示す。親和性の変化に影響を及ぼす、例えば、TFに対するヒト抗体の親和性を改善する、又はそのKDを減少させるために、CDR残基又は他の残基のいずれかの置換が行われ得る。
本発明の抗体分子が、本明細書に記載される構造及び機能特性に従って特定されたら、抗体鎖の所望の部分又は抗体鎖全体をコードする核酸配列は、クローン化、コピー、又は化学的に合成されることができ、かつ常法によって抗体を発現させるために単離及び使用されることができる。本発明の抗体は、免疫グロブリン分子の精製のための当該技術分野において周知の任意の方法によって、例えば、クロマトグラフィ(例えば、イオン交換、アフィニティー、及びサイズ排除カラムクロマトグラフィ)、遠心分離、溶解度差によって、又はタンパク質の精製のための任意の他の標準的な技術によって、精製され得る。更に、本発明の抗体及びその断片は、精製を容易にするために、本明細書に記載される、さもなければ当該技術分野において既知の異種ポリペプチド配列と融合することができる。
本明細書で説明されるように、組換えFc含有タンパク質又はモノクローナル抗体の発現のために選択される宿主細胞は、免疫グロブリンCH2ドメイン内でタンパク質を修飾するオリゴ糖部分の組成物における変化を含むが、これに限定されず、最終的な組成物にとって重要な寄与因子である。したがって、本発明の1つの態様は、所望の治療用タンパク質を発現する産生細胞の使用及び/又は開発のために、適切な宿主細胞の選択を含む。
上述された方法のうちのいずれかによって生成される組成物(抗体、抗体変異体、又は断片)は、ヒトの疾患、又は細胞、組織、器官、体液、若しくは、一般に宿主における特定の病理を、診断、治療、検出、又は調節するために使用され得る。本明細書において教示されるように、抗体が元のターゲッティング性を保持した状態で、標的結合後のエフェクター機能のより特異的に適した範囲をもたらすための、抗体のFc部分、Fc融合タンパク質、又はFc断片の修飾は、特定応用及び治療指標のための抗体の変異体を産生する。
タンパク質及び抗体スタンダード
ヒトTFの組み換え細胞外ドメイン(ECD)を、ELISA及びBiacoreを基礎とする直接的な結合アッセイ用に2つの形態で構築し、TF(配列番号1)の成熟鎖のアミノ酸1〜219を、C末端His6−tagペプチドを有する哺乳類系で発現させ、共結晶構造解析に関しては、配列番号1のアミノ酸5〜213を、C末端His6−tagペプチドを有する細菌系で発現させた。タンパク質のアミン残基を標的にするNHS−ester化学分析を用いて、ヒトTF1−219をビオチン化した。凝固アッセイでは、Innovin(登録商標)(Dade Behring Inc.カタログ番号B4212)(診断用途用の、リン脂質、カルシウム、緩衝液、及び安定剤と混合された凍結乾燥された組み換えヒト組織因子)を用いた。
常法を用いて、開示される抗体を発現及び生成した。一次スクリーニングでは、これら分子をコードするDNAを、96ウェルプレートの中のHEK 293E細胞の中で一時的に発現させ、トランスフェクションの後96時間後に、上清を活性(結合)に関して試験した。パイロットスケールでの発現及び精製の該当点を特定して選択した。パイロットスケールでの発現は、750mLの量のHEK 293F細胞又はCHO−S中で一時的に行われた。採取した上清をタンパク質Aクロマトグラフィで精製し、精製タンパク質を、それらの親和性及び機能活性について評価した。更に、精製タンパク質を、SDS−PAGE、SE−HPLC、及び相互作用クロマトグラフィ(CIC)による生物物理学的特徴付けに供した。理論的等電点(pI)を、各変異体についても計算した。パイロットスケールでの特徴付けから、最終的な有力候補の組をWAVE生物反応器にトランスフェクトし、タンパク質Aクロマトグラフィ(chromatorgraphy)で精製した。
ファージパニングラウンドからのグリセロールストックをミニプレップし、pIX遺伝子をNheI/SpeI消化により除去した。再連結の後、DNAをTG−1細胞に転換し、LB/寒天平板培地で一晩増殖させた。次の日、コロニーを採取し、一晩増殖させ、この培養物を、(i)V領域のコロニーPCR及びシークエンシング、並びに(ii)Fab産生の誘発で用いた。Fabを産生するため、一晩培養物を新しい培地で10〜100倍に希釈し、37℃で5〜6時間にわたって増殖させた。Fab産生は、IPTGを含有する新鮮培地を加えることによって誘発され、培養物を30℃にて一晩増殖させた。次の日、培養物を回転沈降させ、可溶性のFabタンパク質を含有する上清を、FabのELISで用いた。
化学発光による検出を用いた溶液相直接TF結合ELISAを用いて、ヒトフフレームワーク適応ライブラリから最も良く結合するもののランク付けを行った。96ウェルblack maxisorpプレートを、炭酸塩−重炭酸塩緩衝液で希釈した100uLの4ug/mLヤギ抗ヒトIgG FCで、pH 9.4、4℃にて一晩コーティングした後、洗浄緩衝液(0.05%のTween−20溶液を有するPBS)で3回洗浄し、300μLの1% BSA/10mM PBS溶液で1時間にわたって阻害し、その後、先と同様に洗浄した。試料又は標準物質をアッセイ緩衝液(PBS+05% Tween中1% BSA)で50ng/mLに希釈し、100μLを室温にて1時間にわたり振盪させながらアッセイプレートに添加した。これらプレートを3回洗浄し、1ウェル当たり100uLの、Hisタグのついたヒト又はカニクイザルTF−ECDを、アッセイ緩衝液で希釈して100ng/mLで添加し、室温にて2時間インキュベートした。洗浄後、アッセイ緩衝液で1:2000に希釈したQiagenペルオキシダーゼ共役penta−hisを1ウェル当たり100uLで添加し、室温で1時間にわたり振盪させながらインキュベートした。BM ChemiLum Substrate(BM Chemilum,POD,Roche)を緩衝液中1:100の希釈で新たに調製し、最終洗浄後に100uLをプレートに添加した。10分後に、Perkin Elmer Envision Reader,BM ChemiLum programでプレートを読み取る。
このアッセイは、乳癌細胞に発現した内在性ヒトTFに対する抗体の直接結合を検出するために用いられる。FACS緩衝液(PBS中1% FBS)中の試験mAbの4点滴定を2組調製する。滴定を1:4希釈にて1,000ng/mLで開始する。親分子であるM1を陽性対照として用い、坑RSV mAbであるB37を陰性/アイソタイプ対照として用いる。無染色細胞及び二次抗体である、FACS緩衝液1:200中のCy−5共役ヤギ坑ヒトIgG Fc抗体を対照として用い、使用直前に調製した。
Thermofluor技術は、加熱されたときの分子アンフォールディングの反応速度測定である。分子が加熱されると、分子が展開するときに、色素(ANS)は分子に結合することができる。色素は分子に結合すると蛍光を発し、この蛍光が経時的に測定される。このアッセイでは、抗体の展開は37℃〜95℃で測定され、0.5℃毎に検出される。アッセイ対照として用いられる既知のTmsを有する2つのmAb(Emmp 4A5、及びEmmp 5F6)と同様に、マウス及びキメラ形態(10H10、M1、5G9、及びCNTO860)の両方の親分子のTmsも測定した。
種々の抗体と他のヒト抗体との相互作用を判定するために、ヒトIgG(Sigma Aldrich)結合カラムを用いてクロマトグラフィ実験を行った。簡潔に言うと、50mgのヒトIgGを、メーカーの説明書に従って、1mLのNHS−セファロースカラム(GE Healthcare)に結合させた。結合されなかったIgGを、0.1M Tris、pH8、0.5M NaClで洗浄することによって取り除き、非反応NHS群を同じ緩衝液によりブロックした。結合効率は、Pierce’s Coomassie Plusアッセイキット(Thermo Pierce)を使用して、非反応結合緩衝液及び洗浄液中に残っているタンパク質濃度を測定し、固定化の前のタンパク質の量から減算することによって決定された。タンパク質を樹脂に添加しなかったこと以外は、同じプロトコルを用いて対照カラムも調製した。
k’の計算は、タンパク質誘導体化カラム(IgG結合カラム)での保持時間(tR)と、タンパク質が結合していないカラムでの保持時間(t0)の差である。計算は、カラムを標準化するために、両方のカラムでのアセトンの保持時間も考慮に入れる。k’の許容値は0.3未満である。
種々の抗体の室温での溶解性を判定するため、遠心濾過装置を使用して濃度実験を行った。簡潔に言うと、PBS中の抗体調製物を、室温にてVivaspin−15(15mL)遠心濾過装置(30,000MWCO、Sartorius,Goettingen,Germany)に加えた。フィルタを、Beckman Allegra X15−R遠心機の中でスインギングバケットローターを使用して、3000×gで20分間隔で回転させた。体積が約2mLまで減少したら、上清をVivaspin−4(4mL)濾過装置(30,000MWCO)に移し、20分間隔で4,000×gで遠心分離した。体積が500Lまで減少したら、サンプルをVivaspin−500濾過装置に移し、15分間Eppendorf 5424遠心機で15,000×gにて遠心分離した。タンパク質濃度が100mg/mL以上に達するまでこれを繰り返した。タンパク質濃度は、適切な希釈を用い、BioTek SynergyHT(商標)分光光度計上での280nm及び310nmにおける吸光度により決定された。この時点で、遠心分離を停止させ、試料を一晩室温で保持して、平衡に達した。次の朝、試料の沈殿の兆候をチェックした。濃度が100mg/mL超であった場合、プロセスを中止した。
このアッセイを用いて、TF結合抗体が、TFを発現するヒト細胞からのFVIIa誘発IL−8放出をなくすかどうかを試験した。DMEM及び10% FBS(Gibco:カタログ番号11995及びカタログ番号16140)中で成長するように適合されたヒト乳腺癌細胞(MDA−MB−231)(ATCC:HTB−26)を、96ウェルの細胞培養プレート(Nunc:カタログ番号167008)に、1ウェル当たり20000個の細胞(100,000個の細胞/mL)の密度で、標準的細胞培養技術を用いて蒔いた。2μg/mLで開始し、FBSを有さないDMEMで1:2又は1:4のいずれかで連続希釈される抗体処理の前に、細胞を2日にわたって回収した。抗体は、FBSを有さないDMEM中最終濃度50nMのヒトFVIIa(Innovative Research:カタログ番号IHFVIIa、ロット:2824)で処理する1時間前に加えられた。細胞をインキュベータに24時間入れた。処理の後、上清を収集し、IL−8の量を、メーカーのプロトコル(R&D Systems:カタログ番号D8000C)にしたがってELISAによって検出した。簡単に言えば、各処理試料の光学密度(OD)を450nm及び540nmにて読み取った。アッセイプレートの光学的不完全性を補正するために、540nmにおける読み取り値を用いたが、一方で、メーカーのプロトコルに従って作製したIL−8標準曲線を用いて処理試料中のIL−8含有量を計算するために、450nmにおける補正された読み取り値(OD 450マイナスOD 540)を用いた。抗体及びFVIIa処理を受けていない細胞を有するウェルを用いて、内因性IL−8レベルを決定する一方で、FVIIaのみを受けた細胞を有するウェルを用いて、「非阻害」IL−8レベルを決定し、それによって、最小及び最大IL−8レベルをそれぞれ定義した。MAb滴定処理試料を、上で定義された最大及び最小IL−8レベルに対して正規化し、阻害百分率で表した。正規化データは、棒グラフで表されるか、又は各MAbのEC50値を抽出するようにフィットする4パラメータのロジスティック曲線に当て嵌められるかのいずれかであった。
このアッセイは、カルシウム及び添加された組み換えヒトTF調製物(Innovin,Dade Behring Inc)の存在下でヒト血漿を使用して、坑ヒトTF抗体が生体外での凝固を阻害するかどうかを判定するために用いた。坑ヒトTF抗体をHBSS(Gibco、カタログ番号14175)で2mg/mL抗体まで希釈する。クエン酸ナトリウムを有するプールされたヒト血漿(George King Biomedical,Novi,MI)を、1000rpmにて5分にわたり回転沈降させ、透明な血漿を新しい管に移す。透明な96ウェルアッセイプレート(NUNC、カタログ番号439454)の各ウェルの中で、25uLの希釈した抗体を100uLのヒト血漿に加える。22mMのCaCL2を有するHBSSで1:500に希釈した125μLのInnovin(Dade Behring Inc.、カタログ番号B4212)を、抗体を有する又は有する血漿を収容しているウェルに添加することによって、反応を開始させる。凝固反応は、SpectraMax M2e読み取り機(Molecular Devices,Sunnyvale,CA)を使用して、反応開始直後及び30分にわたり37℃にて、OD 405で速度論的に監視する。各抗体のT1/2 Maxは、最大光学密度の50%に達するまでにかかる時間(秒)として、Softmax Proソフトウェアを用いて決定される。試料の時間(秒)は各プレートの参照に対して正規化されたが、統計的には、抗体を有さない試料、10H10、及び全て10H10由来でヒトに適合した変異体の平均時間である150秒及び200秒と記載している試料の間に差はなかった。
(実施例)
The Scripps Research Institute in La Jolla,CA(米国特許第5223427号、Morriseyら、1988 Thromb Res.52(3):247〜261)で産生された10H10として知られるマウス抗体を、ハイブリドーマTF9.10H10−3.2.2によって産生する。10H10ハイブリドーマクローンからの抗体の配列は、以前に報告されていない。
この配列は、2つの抗体鎖、VH及びVLを、5’GeneRacer(商標)(InVitrogen)、プライマー、並びにマウスIgG1定常領域及びマウスKappa定常領域内の配列にそれぞれ相補的な3’コンセンサスプライマーを用いて増殖せる、5’RACE法(Focus 25(2):25〜27,2003;Maruyama 1994 Gene 138,171〜174)を用いて特定された。配列解析により適したVL生成物を生成するために、5’GeneRacer Nestedプライマー及び3’コンセンサスプライマーを用いるNested PCR増幅を用いた。
ヒトTF ECDと、対応するFab断片との間の複合体の結晶構造決定により、10H10のエピトープマッピングを行った。Hisタグ付きヒトTF ECD(配列番号1の残基5〜213)を大腸菌内で発現させ、HisTrap HPカラム(GE Healthcare)及びQ HPカラム(GE Healthcare)をそれぞれ用いて、親和性及びイオン交換クロマトグラフィによって精製した。10H10 FabのHisタグ付きキメラバージョン(マウスV領域、ヒト定常ドメイン)をHEK細胞内で発現させ、親和性(TALONカラム、GE Healthcare)及びサイズ排除(HiLoad Superdex 200カラム、GE Healthcare)クロマトグラフィを用いて精製した。
10H10は、ECDのN末端ドメインとC末端ドメインとの間の接点でTFに結合する。TFの凸面は、抗体の凹状CDR表面に合致する。複合体形成の際に覆われる総面積は、相互作用分子のそれぞれの上で1,100Å2を超える。6つのCDRの全てが、TFとの直接接触に関与する(接触は4Å原子間距離と定義される)。合計で、24個のエピトープ残基及び25個のパラトープ残基が存在する。CDR L1、H1、及びH3は、接触の大部分を形成する。10H10:TF複合体のエピトープ及びパラトープを形成する残基は、図1に概略的に示されている。
ヒト、カニクイザル(配列番号2)、及びマウスTF ECD(配列番号3)のアミノ酸配列が、図2に整列配置されている。ヒトの配列とカニクイザルTF ECDの配列の類似性は高く、これら2つが10H10接触残基において異なるのは、配列番号1の位置197の1つ残基だけであり、この位置はカニクイザル配列ではR(Arg)である。ヒト配列のT197は、単一H−CDR2残基によって接触されるので、10H10由来の抗体の現在の組で見られる高レベルの交差反応性の説明がつく。
治療用タンパク質の有効性は、望ましくない免疫反応によって制限され得る。非ヒトモノクローナル抗体は、ヒトでの免疫応答を誘発することができる、かなりの線形アミノ酸配列鎖と、局所的な構造コンフォメーションを有し得る。非ヒトMAbの標的結合の免疫特異性に関与している残基のヒト抗体骨格への移動は、しばしば、標的抗原に対する結合親和性の大きな損失をもたらす。したがって、ヒトに注入されたときに、親非ヒト分子の結合及び生物物理学的プロファイルを保持すると同時に、最小免疫原性反応を誘発する抗体分子を作成するための、しっかりした設計原理を用いることが極めて重要である。
抗原結合部位に含まれないV領域内の領域であると定義されるヒトFR領域は、機能的ヒト生殖細胞系IGHV及びIGHJ遺伝子のレパートリーから選択された。ヒト生殖系列遺伝子配列のレパートリーは、IMGTデータベース(Lefranc 2005)を検索し、「01」対立遺伝子を一括してコンパイルすることによって得た。このコンパイルから、重複遺伝子(アミノ酸レベルで100%同一)及び対を成さないシステイン残基を有する遺伝子はコンパイルから除外された。遺伝子コンパイルの最終更新は、2007年10月1日に行われた。
ヒト及びカニクイザルTFの両方に対する親キメラ抗体、M1、及びHFA変異体の結合は、化学発光による検出を用いる直接ELISA形式として行われた。粗上清中のライブラリ変異体の一次スクリーニングでは、試料又は対照は、消費FreeStyle 293 HEK培地(Gibco)内で50ng/mLに正規化され、単一濃度測定で検定された。本発明のアッセイでは、抗体の濃度は5ng(0.1mL使用)であり、TF ECD及び抗原は、His6−TF−ECD1−219であり、最終濃度10ng/ウェルで用いられた。
Fabライブラリは、米国特許第6,472,147号(Scripps)及び、国際公開特許第2009/085462号として発行された本出願人による同時継続中の出願に記載されている、pIXファージディスプレイシステムにおいて、制限酵素部位に若干の変更を加えて構築された。
単一LC可変領域(L3、配列番号23)及び単一HC可変領域(H22、配列番号19)を含み、いくつかのCDR残基に異なる残基を有する、M59に基づく変異体の、ヒトフフレームワーク適応及びライブラリ再選択の後、この新規Mabを、生物物理学的及び生理活性アッセイに供し、また、異なるパラトープ残基を有する一対(paring)、M1587を用いて、TF−ECDに対する10H10 Fabの結合に関して初めに特徴付けされたエピトープ(実施例2)が変わったどうかを再度調べた。
10H10 CDR、M1587(L3及びH116)に基づいたヒトに適合しかつ親和性の成熟抗体と、TF ECDとの共結晶化を、M1587−Fab:TF複合体が、16%のPEG 3350、0.2Mの酢酸アンモニウム、0.1Mの酢酸ナトリウム、pH 4.5を含有する溶液から結晶化されたことを以外は、10H10(実施例3)と同様の方法で行った。
これら抗体の、BiacoreによるKD分析(表11)、ヒトTFによるヒト血漿の凝固時間(表12)、及びFVIIaで刺激されたMD−MB−231細胞からのIL8放出に関するEC50(表13、14、及び図5)に関する要約データが、以下に示されている。
新規Mabは、カルシウム及び外から加えられたヒトTFの存在下で、生体外で測定した場合に、ヒト血漿の凝固を阻害せずにヒトTFに結合する能力によって特徴付けられる(表12)。17個のHFA(M数は100未満)変異体及び38個の親和性成熟変異体(M1583以上)を検定し、T1/2最大(最大光学密度の50%に達する時間(秒))を報告した。
全て、10H10(表12)で観察されたのと同様の応答を示し、T1/2最大の値は205秒未満であり、このことは、159±17(n=14)であった抗体のない溶媒対照群と比較した場合に、これら抗体が凝固時間を長引かせないことを示している。前述したヒトTF結合抗体であり(米国特許第7605235 B2号)、TFに対するFXの結合を阻害するマウス抗体5G9由来のCNTO860は、凝固を長引かせ、同じアッセイで1800秒以内に決して凝固に達しない。実施例4において変化したCDRを有すると記載された43個のMAbのうち5個は、2mg/mL未満の開始濃度を有したので、凝固アッセイで試験しなかった。M1、M59、及びCNTO860の値は、複数回の試験にわたって平均化された。
新規MAbは、TF/FVIIa複合体を介してシグナル伝達を阻害する能力の観点からも説明され得る。乳癌細胞のTF/VIIa/PAR2シグナル伝達は、VEGF25、Cyr61、VEGF−C、CTGF、CXCL1、及びIL−8などの血管新生促進因子の広範なレパートリーを誘導する。FVIIaが、TFを発現するヒト乳癌細胞株であるMDA−MB−231???において検出可能なIL−8を誘導することは、既に報告されている(Albrektsenら、J Thromb Haemost 5:1588〜1597,2007)。したがって、このアッセイをバイオアッセイとして用いて、TF/VIIaの誘導によるIL−8の産生を阻害する変異体抗体の能力を評価した。
MDA−MB−231を有するマウス異種移植モデル
MDA−MB−231ヒト乳癌細胞を、10%のFBS及び1%のLNNを有するDMEM培地で培養し、トリプシン処理によって対数期に採取し、滅菌無血清DMEM培地に5×107細胞/mLで再懸濁させた。20匹の雌SCIDベージュ(C.B−17/IcrCrl−scid−bgBR)マウスをCharles River Laboratoriesから入手し、実験に先立って14日間順応させた。生後約8週目に、マウスの右腋窩乳腺脂肪体に2.5×106 MDA−MB−231細胞を埋め込んだ。腫瘍のサイズが約100mm3になったとき、マウスを腫瘍サイズによって治療群に階層化した(N=10/群)。階層化した日に、10mg/体重1kgのDulbeccoリン酸緩衝生理食塩水(DPBS)又はM1593による腹腔内投与治療を開始し、週1回投与を続けて合計6回投与した。腫瘍及び体重を週1回記録した。各群の平均腫瘍容積が1500mm3に達した時点で研究を終了した。適用した統計検定は、反復測定二元配置分散分析(PRIZM 4.0、GraphPad)であった。
A431ヒト扁平上皮癌細胞を、10%のFBS及び1%のLNNを有するDMEM培地で培養し、トリプシン処理によって対数期に採取し、滅菌HBSSに1×107細胞/mLで再懸濁させた。20匹の雌SCIDベージュ(C.B−17/IcrCrl−scid−bgBR)マウスをCharles River Laboratoriesから入手し、実験に先立って14日間順応させた。生後約8週目に、マウスの右脇腹に2×106 A431細胞を埋め込んだ。腫瘍のサイズが約118mm3になったとき、マウスを腫瘍サイズによって治療群に階層化した(N=10/群)。階層化した日に、10mg/体重1kgのDPBS又はM1593による腹腔内投与治療を開始し、週1回投与を続けて合計6回投与した。腫瘍及び体重を週2回記録した。各群の平均腫瘍容積が1000mm3に達した時点で研究を終了した。適用した統計検定は、反復測定二元配置分散分析(PRIZM 4.0、GraphPad)でった。
天然ヒトFc受容体変異体は、ヒト抗体のFcタンパク質に対して実質的に異なる親和性を有する。更に、臨床研究で、Fc操作されたmAbで治療された後、強力結合Fc遺伝子型を有する患者の奏効率及び生存率が改善したことが立証されている(Musolinoら2008 J Clin Oncol 26:1789〜1796(2008);Bibeauら2009 J Clin Oncol 27:1122〜1129)。
フコース含有量が少ない抗体(M1593−LF)は、シグナルペプチドを有する以下に示されるM1593(IgG1/Kappa)鎖をコードするベクターを、CHO宿主細胞株からタンパク質の低フコシル化のために選択されたCHO宿主細胞亜系統中に電気穿孔することによって産生された。配列番号165は、可変ドメイン、残基1〜113(配列番号23及びFR4、配列番号61、下線)、及びヒトkappa定常軽鎖ドメインを含む完全な軽鎖を示す。可変ドメイン残基1〜120(配列番号139及びFR4、配列番号60を含む、下線)、並びにKabat位置239及び332(配列番号167の242及び335)である野生型ヒトIgGアイソタイプ1 CH1、CH2、及びCH3を含む重鎖は、変異体M1593−DEを産生するために、野生型残基S及びDからそれぞれ、D及びEに変更された。

Claims (31)
- ヒト組織因子への結合に関してマウス抗体10H10と競合する単離された抗体であって、前記抗体結合ドメインが、ヒトフレームワーク(FR)領域に適合され、かつFR1−CDR1−FR2−CDR2−FR3−CDR3の構造を有し、前記FRアミノ酸配列が、IMGTデータベースで認証されているヒト生殖系列遺伝子配列によってコードされたアミノ酸配列から変更されず、CDR配列が、配列番号6〜11及び27で表されるマウス10H10 CDRと50%以上の配列同一性を有する、単離された抗体。
- 前記抗体が、組織因子結合に関してFVIIaと競合せず、かつ、MDA−MB−231細胞からのサイトカインIL−8放出によって測定すると、TF−VIIa複合体の凝固促進活性、アミド分解活性を実質的に阻害しないが、TF−VIIaによって媒介されるシグナル伝達を阻害する、請求項1に記載の単離された抗体。

- 前記疾患が癌である、請求項21に記載の方法。
- 前記癌が、原発固形腫瘍、転移癌、細胞腫、腺癌、メラノーマ、液性腫瘍、リンパ腫、白血病、骨髄腫、軟部組織癌、悪性腫瘍、骨肉腫、胸腺腫、リンパ肉腫、線維肉腫、平滑筋肉腫、脂肪腫、神経膠芽腫、星状細胞腫(astrosarcoma)、前立腺、乳腺、卵巣、胃、膵臓、喉頭、食道、精巣、肝臓、耳下腺、胆道、結腸、直腸、頸、子宮、子宮内膜、甲状腺、肺、腎臓、又は膀胱の癌から選択される、請求項22に記載の方法。
- 前記疾患が、良性腫瘍、血管腫、聴神経腫瘍、神経線維腫、トラコーマ、及び化膿性肉芽腫;動脈硬化斑;眼球血管新生疾患、糖尿病性網膜症、未熟児網膜症、黄斑変性症、角膜移植片拒絶反応、血管新生緑内障、水晶体後線維増殖、ルベオーシス、網膜芽細胞腫、ブドウ膜炎及び眼の翼状片(異常な血管成長);リウマチ性関節炎;乾癬;創傷治癒遅延;子宮内膜症;脈管形成;肉芽形成;肥厚性瘢痕(ケロイド);癒着不能骨折;強皮症;トラコーマ;血管接着;心筋血管新生;冠側副;脳側副;動静脈奇形;虚血肢血管新生;オースラー・ウェーバー症候群;プラーク血管新生;毛細血管拡張症;血友病関節症;血管線維腫;線維筋性形成異常;創傷肉芽形成;クローン病;及びアテローム性動脈硬化症から選択される、請求項21に記載の方法。
- 薬学的に許容可能な調製物中に請求項1、3、19、又は20に記載の抗体を含む、被験者を治療するのに有用な医薬組成物。
- 安定な形態の請求項1、3、19、又は20に記載の抗体と、使用説明書と、を含む、キット。
- 請求項1、3、19、又は20に記載の抗体の前記抗体結合ドメインの1つ以上をコードする、単離された核酸。
- 請求項27に記載の少なくとも1つのポリヌクレオチドを含む、ベクター。
- 請求項28に記載のベクターを含む、宿主細胞。
- IgG1又はIgG4アイソタイプを有する、請求項1、3、19、又は20に記載の単離された抗体又は断片。
- Fc領域がヒトIgG1アイソタイプを含み、Kabat位置239及び332(これらは配列番号167の242及び335である)が、該Fc領域の変異により、野生型残基S及びDから、D及びEに改変されている、請求項30に記載の単離された抗体又は断片。
