Claims (15)

  1. 植物細胞のゲノムにおいて予め定められた部位に外来DNA分子を導入するための方法であって、
    a.前記の予め定められた部位において二本鎖DNA切断を誘導する工程;
    b.前記の植物細胞に前記の外来DNA分子を導入する工程;および
    c.前記の外来DNAが前記の予め定められた部位に導入されている植物細胞を選択する工程
    を含み、前記の予め定められた部位は、ストレプトミセス・ハイグロスコピクス(Streptomyces hygroscopicus)によってコードされるホスフィノトリシンアセチルトランスフェラーゼをコードするDNA領域内(barコード領域)に含まれ、そして前記の二本鎖DNA切断は、前記の予め定められた部位を認識しそして前記の二本鎖切断を誘導する一本鎖メガヌクレアーゼの導入か、または前記の予め定められた部位を協調して認識しそして前記の二本鎖切断を誘導するメガヌクレアーゼの対の導入かによって誘導されることを特徴とする、前記方法。
  2. 植物細胞のゲノムにおいて予め定められた部位に外来DNA分子を導入するための方法
    であって、
    a.前記の予め定められた部位において二本鎖DNA切断を誘導する工程;
    b.前記の植物細胞に前記の外来DNA分子を導入する工程;および
    c.前記の外来DNAが前記の予め定められた部位に導入されている植物細胞を選択す
    る工程
    を含み、前記の予め定められた部位は、配列番号1または配列番号2のヌクレオチド配列
    を含み、そして前記の二本鎖DNA切断は、前記の予め定められた部位を認識しそして前
    記の二本鎖切断を誘導する一本鎖メガヌクレアーゼの導入か、または前記の予め定められ
    た部位を協調して認識しそして前記の二本鎖切断を誘導するメガヌクレアーゼの対の導入
    かによって誘導されることを特徴とする、前記方法。
  3. 前記のメガヌクレアーゼまたは前記のメガヌクレアーゼの対がI−CreIに由来し、そして以下のアミノ酸がメガヌクレアーゼユニット1に存在し:
    a.32位におけるS;
    b.33位におけるY;
    c.38位におけるE;
    d.40位におけるR;
    e.66位におけるK;
    f.80位におけるQ;
    g.42位におけるT;

  4. 前記のメガヌクレアーゼの対は義務的にヘテロダイマーを形成するか、または前記メガヌクレアーゼは、リンカーによって共有結合的に接続されたI−CreIに由来する2つのドメインを含む一本鎖メガヌクレアーゼである、請求項1〜のいずれか1項記載の方法。
  5. 前記のメガヌクレアーゼの対がそれぞれ配列番号5および配列番号6のアミノ酸配列を含むか、前記のメガヌクレアーゼの対が配列番号4のヌクレオチド2004位からヌクレオチド2525位までまたは2522位までのヌクレオチド配列、および配列番号4のヌクレオチド4885位からヌクレオチド5405位までまたは5403位までのヌクレオチド配列を含む核酸によってコードされるか、または前記の一本鎖のメガヌクレアーゼが、配列番号18の1位から167位までおよび206位から362位までのアミノ酸配列を含むか、または前記の一本鎖メガヌクレアーゼが、配列番号17のヌクレオチド1267位から1605位までおよび1795位から1956位までおよび2071位から2541位までのヌクレオチド配列を含む核酸分子によってコードされるか、あるいは、前記の一本鎖メガヌクレアーゼが、配列番号17の1267位から1605位までおよび1795位から2541位までのヌクレオチド配列を含む核酸分子によってコードされる、請求項1〜のいずれか1項記載の方法。
  6. 前記の外来DNAが、対象の植物で発現可能な遺伝子を含み、前記の対象の植物で発現可能な遺伝子が、除草剤耐性遺伝子、昆虫耐性遺伝子、疾病耐性遺伝子、非生物的ストレス耐性遺伝子、油生合成、炭水化物生合成に関与する酵素、線維の強度または線維の長さに関与する酵素、二次代謝物の生合成に関与する酵素の群から選択される、請求項1〜のいずれか1項記載の方法。
  7. 前記の植物細胞がさらに植物へと再生される、請求項1〜のいずれか1項記載の方法。
  8. 請求項1〜のいずれか1項記載の方法によって得られた、前記の外来DNAが前記の予め定められた部位に導入されている植物細胞、植物、またはその繁殖材料
  9. 化学物質を前記植物に、または前記植物が成長している基質に適用する工程を含む、請求項記載の植物を成長させる方法。
  10. 請求項の植物細胞から実質的になる植物を、別の植物またはそれ自体と交雑させ、そして場合により種子を収穫する工程を含む、barコード領域に組み込まれた外来DNAを含む植物を産生するための方法
  11. 化合物を、請求項記載の植物の種子に適用する工程を含む、処理された種子を産生する方法
  12. 外来DNAをbarコード領域に導入するための請求項1〜のいずれか1項に記載の通りのメガヌクレアーゼまたはメガヌクレアーゼの対の使用。
  13. 対象の外来DNAを植物細胞のゲノムにおいて予め定められた部位に導入するためのカスタムメイドのメガヌクレアーゼの使用。
  14. 植物細胞のゲノムにおいて予め定められた部位に外来DNA分子を導入するための方法であって、
    a.前記の予め定められた部位に二本鎖DNA切断を誘導する工程;
    b.前記の植物細胞に前記の外来DNA分子を導入する工程;および
    c.前記の外来DNAが前記の予め定められた部位に導入されている植物細胞を選択する工程
    を含み、前記の二本鎖DNA切断は、協調して前記の予め定められた部位を認識し、そして前記の二本鎖切断を誘導する天然には存在しない一本鎖メガヌクレアーゼの導入か、または天然には存在しないメガヌクレアーゼの対の導入かによって誘導されることを特徴とする、前記方法。
  15. トランスジェニック植物細胞のゲノムにおいて予め定められた部位に外来DNA分子を導入するための方法であって、
    a.前記の予め定められた部位に二本鎖DNA切断を導入する工程;
    b.前記の植物細胞に前記の外来DNA分子を導入する工程;および
    c.前記の外来DNAが前記の予め定められた部位に導入されている植物細胞を選択する工程
    を含み、前記の予め定められた部位は、天然に存在するメガヌクレアーゼのための認識部位とは異なるヌクレオチド配列であり、前記の予め定められた部位は、トランスジェニック植物において導入遺伝子の一部として一般的に導入されるヌクレオチド配列であり、そして二本鎖DNA切断は、前記の予め定められた部位を認識しそして前記の二本鎖切断を誘導する天然には存在しない一本鎖メガヌクレアーゼの導入か、または前記の予め定められた部位を協調して認識しそして前記の二本鎖切断を誘導する天然には存在しないメガヌクレアーゼの対の導入かによって誘導されることを特徴とする、前記方法。
