Description
A találmány tárgya új eljárás az I általános képletű fenil-etanol-aminok előállítására. Az I általános képletbenThe present invention relates to a novel process for the preparation of phenylethanolamines of formula I. In the general formula I
Rt klór- vagy brómatomot,R t is chlorine or bromine,
R2 trifluor-metil-csoportot, fluor- vagy klóratomot ésR 2 is trifluoromethyl, fluoro or chloro; and
Rj elágazó szénláncú 3-5 szénatomos alkilcsoportot vagy 3-5 szénatomos cikloalkilcsoportot jelent.R 1 is a branched C 3 -C 5 alkyl group or a C 3 -C 5 cycloalkyl group.
Az I általános képletű vegyületeknek értékes farmakológiai tulajdonságaik vannak, elsősorban broncholitikus hatásúak.The compounds of the formula I have valuable pharmacological properties, in particular their broncholytic activity.
Among the meanings given above for R 3 , these include, in particular, isopropyl, isobutyl, tert-butyl, isopentyl, neopentyl, tert-pentyl, cyclopropyl, cyclobutyl or cyclopentyl.
According to the present invention, the phenylethanolamines of formula I are prepared by reacting a 4-aminobenzaldehyde of formula II, R 1 and R 2 as defined above, with an isocyanide of formula III, R 3 having the same meaning as above, and carboxylic acid of formula IV, - R 4 represents a carboxylic acid acyl group - is reacted, the resulting V α-acyloxy-acetamide of the formula - R-R4 are as defined above - is reduced to a hydride such as a metal hydride or complex metal hydride.
The radicals defined above for the definition of R 4 are primarily acyl groups of aliphatic or aromatic carboxylic acids such as acetic acid, propionic acid, trimethylacetic acid, valeric acid, benzoic acid, nitrobenzoic acid or naphthalene-2-carboxylic acid.
The reaction of 4-aminobenzaldehyde II with isocyanide III and carboxylic acid IV is conveniently carried out in a suitable solvent such as methylene chloride, chloroform, benzene, ether, tetrahydrofuran or dioxane at low temperatures, for example, -20 to 75 ° C. , preferably at room temperature. However, the reaction is conveniently carried out by dropwise addition of a compound of Formula III and a carboxylic acid of Formula IV to a solution of an aldehyde of Formula II or a solution of an isocyanide of Formula III and an aldehyde of Formula II in a drip funnel at the same time. the carboxylic acid of formula IV is added dropwise over a period of several hours, for example 5-10 hours, followed by stirring for a further 10-165 hours.
The reduction of the resulting α-acyloxyacetamide V with a hydride is preferably carried out in a suitable solvent such as ether, tetrahydrofuran or dioxane at low to slightly higher temperatures, for example 0 to 100 ° C, preferably at the reflux temperature of the reaction mixture. It is particularly preferred to reduce with borane tetrahydrofuran and the boiling point of tetrahydrofuran as solvent.
A kívánt I általános képletű tennék elkülönítését célszerűen vizes bázissal, például vizes ammóniaoldattál végezzük, miközben az adott esetben még jelenlévő α-aciloxi-csoport lehasad. The desired product of formula I is conveniently isolated using an aqueous base such as an aqueous ammonia solution while the optional α-acyloxy group is cleaved.
A kapott I általános képletű fenil-etanol-amint kívánt esetben szervetlen vagy szerves savval fizio2 lógiailag elviselhető savaddíciós sójává alakíthatjuk. Savként például sósavat, hidrogénbromidot, kénsavat, foszforsavat, tejsavat, borkősavat, citromsavat, fumársavat vagy maleinsavat használhatunk. The resulting phenylethanolamine of formula I can be converted, if desired, into a physiologically acceptable acid addition salt of an inorganic or organic acid. Suitable acids are, for example, hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, lactic acid, tartaric acid, citric acid, fumaric acid or maleic acid.
A kiindulási anyagként használt III és IV általános képletű vegyületek a szakirodalomból ismertek. [G. Entenmann: Methodicum Chimicum, VI. kötet, 795. oldal; Georg Thieme Verlag, Stuttgart, 1974.].The starting compounds of formulas III and IV are known in the art. [G. Entenmann, Methodicum Chimicum, VI. 795, p. Georg Thieme Verlag, Stuttgart, 1974.].
The aldehyde of formula II used as starting material may be prepared, for example, by reduction of a suitable 4-aminobenzoic acid halide or ester and oxidation of the optionally obtained benzyl alcohol with manganese dioxide.
A process for the preparation of the phenylethanolamines of formula I is known from the literature (Hungarian Patent No. 161,551); the process involves the reduction of a glycolic acid amide of formula VI, R 1 , R 2 and R 3 as defined above, with a complex metal hydride.
A disadvantage of the known process is that the glycolic acid amide required as starting material can only be prepared under very conditions.
A találmány szerinti új eljárással elért jó kitermelések a szakember számára nem kézenfekvőek, mert a szakirodalomból ismert [J. Amer. Chem. The good yields obtained by the novel process of the present invention are not obvious to those skilled in the art because they are known in the art [J. Amer. Chem. Soc., 67, 1499-1500 (1945)] that the Passerini reaction is sterically inhibited and cannot be accomplished with α, β-unsaturated carbonyl compounds. The inability of α, β-unsaturated carbonyl compounds, such as croton aldehyde, to react with the Passerini reaction is explained by neutralizing the electron center of the carbonyl function based on the mesomeric boundary structures shown in Scheme [A], i.e. the electron transport effect of a neighboring group.
It is also known that an amino group at the o- or p-position of a phenyl ring exhibits a strong electron transfer.
One of ordinary skill in the art would therefore have expected that the electron center of the carbonyl function of the 4-aminobenzaldehyde of formula II would be deactivated based on the mesomeric boundary structure of formula IIa and thus the Passerini reaction would not take place. Surprisingly, the exact opposite has happened.
A találmány szerinti eljárást közelebbről mutatják be a következő példák.The following examples illustrate the process of the invention in more detail.
