Description
Die Erfindung betrifft ein neues Verfahren zur Herstellung
N-alkylsubstituierter Aminomethylphosphonsäurederivate der
Formel
worin R₁ für eine gegebenenfalls substituierte Alkyl-
oder Cycloalkylgruppe mit 1 bis 18 Kohlenstoffatom(en)
steht und R₂ ein Wasserstoffatom oder eine Methyl- oder
Ethylgruppe darstellt, insbesondere ein Verfahren zur
Herstellung von Methylaminomethylphosphorsäuredimethylestern
und -diethylestern und der entsprechenden Säuren
in hoher Ausbeute.
Die bekannte Methylaminomethylphosphonsäure der Formel
genießt als Mittel zum Feuerfestausrüsten und als Herbizid
große Aufmerksamkeit (vgl. SU-PS 10 74 886 = Chemical
Abstracts, 100, 19 347C (1984) und DE-OS 28 48 869 =
Chemical Abstracts, 93, 39 529Z (1980)). Es gibt zwar eine
Reihe von Verfahren zur Herstellung von N-alkylsubstituierten
Aminomethylphosphonsäuren, die einzelnen Verfahren
sind jedoch mit verschiedenen Nachteilen behaftet.
Gemäß der US-PS 23 28 358 erfolgt eine Umsetzung von
N-Methyl-N-hydroxymethylstearamid mit PCl₃ und anschließende
Behandlung des Reaktionsprodukts mit verdünnter Salzsäure
zur Gewinnung von N-Methylstearamidomethylphosphonsäure
der Formel
Das hierbei erhaltene Reaktionsprodukt wird schließlich
zu Methylaminomethylphosphonsäure hydrolysiert. Dieses
Verfahren dauert unangemessen lang. Das als Reaktionspartner
verwendete Stearamid erschwert einerseits die
Verfahrensdurchführung und ist andererseits teuer.
Gemäß der US-PS 39 07 652 gewinnt man die N-Methylamino-
di(methylphosphonsäure) der Formel
aus Methylamin, Formaldehyd und Phosphorsäure (H₃PO₃) (vgl.
"Journal of Organic Chemistry", 31, 1603 (1966)). Das Reaktionsprodukt
wird unter Entfernung einer Phosphonomethylgruppe
durch oxidative Elektrolyse in Methylaminomethylphosphonsäure
überführt. Dieses Verfahren erfordert Spezialanlagen
für letztere Reaktion, die in starker Säure durchgeführt
wird.
Aus der US-PS 43 51 779 sind die Kondensation von Trimethylhexahydro-
s-triazin mit überschüssigem sek.-Dialkylphosphit
durch Erwärmen auf 20-150°C und anschließende
Hydrolyse oder Pyrolyse des Kondensationsprodukts bei hoher
Temperatur (230-240°C) bekannt. Die bei diesem Verfahren
eingesetzte Phosphorquelle, nämlich das sek.-Dialkylphoshit,
ist für eine wirtschaftliche Herstellung des gewünschten
Reaktionsprodukts zu teuer.
Bei dem aus der US-PS 41 60 779 bekannten Verfahren erhält
man die Methylaminomethylphosphonsäure durch Umsetzung
von Bis(chlormethyl)phosphonsäure in wäßrigem Ammoniak bei
150°C und 80 bar Druck während 7 h. Dieses Verfahren erfordert
einen kostspieligen Hochdruckautoklaven. Darüber
hinaus benötigt man zur Reinigung des Rohprodukts einen
Säureionenaustauscher. Diese Erfordernisse sind ein Hinderungsgrund
zur Durchführung dieses Verfahrens in großtechnischem
Maßstab.
Bei zwei anderen Verfahren erfolgt eine Reaktion eines Amids
mit Phosphortrichlorid. Das von Tyka und Hagele in "Synthesis",
218-19 (1984) vorgeschlagene Verfahren ist ein Laborverfahren
für die Synthese von N-Alkylaminomethylphosphonsäure
aus N-Alkyl-N-hydroxymethylformamiden und Phosphortrichlorid.
Nachteilig an diesem Verfahren ist, daß die Herstellung und
Isolierung des Ausgangsprodukts, nämlich des N-Alkyl-N-
hydroxymethylformamids, nur in für eine großtechnische Durchführung
viel zu geringer Ausbeute möglich ist. Dieses Problem
soll gemäß der US-PS 48 30 788 dadurch gelöst werden, daß
man das Amid für die Herstellung von Methylaminomethylphosphonsäure,
nämlich das Amid der Formel
oder seinen niedrigmolekularen Carbonsäureester der Formel
in situ aus N-Methylacetamid, Essigsäure, Essigsäureanhydrid
und p-Formaldehyd durch Erwärmen auf 116°C herstellt, danach
das Reaktionsgemisch auf 25°C abkühlt und schließlich
tropfenweise mit Phosphortrichlorid versetzt. Nahe dem Ende
der PCl₃-Zugabe wird die Temperatur 45 min lang auf 59-70°C
gehalten und dann während 3 h schrittweise auf 130°C erhöht.
Dieses Verfahren erfordert die Zugabe zu vieler Reaktionsteilnehmer
und eine Temperatursteuerung, wodurch die Ausbeute
an Reaktionsprodukt zu wünschen übrig läßt. Darüber hinaus
entweicht mehrere h lang korrodierender gasförmiger Chlorwasserstoff.
Zur Reinigung des Reaktionsprodukts muß aus
der Hydrolyse von Amid und Acetylchlorid herrührende Essigsäure
abdestilliert werden, was das Verfahren aufwendig
macht.
Erfindungsgemäß wurde nun ein einfach und unter milden Bedingungen
durchführbares Verfahren zur Herstellung
N-alkylsubstituierter Aminomethylphosphonsäurederivate in
guter Ausbeute entwickelt. Hierbei werden N-Alkyl- bzw.
N-Cycloalkylhexahydro-s-triazine der Formel
worin R₁ für gegebenenfalls substituierte (z. B. cyclohexylsubstituierte)
Alkyl- und Cycloalkylgruppen mit 1-18 Kohlenstoffatom(en),
z. B. Methyl-, Ethyl, Isopropyl-, n-Butyl-,
n-Octyl-, n-Octadecyl- oder Cyclohexylgruppen steht, mit
Trialkylphosphiten der Formel
P(OR₂)₃ (III)
worin R₂ für ein Wasserstoffatom oder eine Methyl- oder
Ethylgruppe steht, in Gegenwart von Titantetrachlorid
(TiCl₄) bei -20°C bis +20°C, vorzugsweise +10°C, zu
N-alkyl- bzw. N-cycloalkylsubstituierten Aminomethylphosphonsäuredialkylestern
umgesetzt. Diese können dann zur
Gewinnung der entsprechenden kristallinen festen
N-alkyl- bzw. N-cycloalkylsubstituierten Aminomethylphosphonsäuren
hydrolysiert werden.
Bei der Zugabe von Titantetrachlorid zu einem N-Alkyl-
oder N-Cycloalkylhexahydro-s-triazin in Methylenchlorid
erhält man eine dunkelrote Lösung. Dieser Lösung wird ein
Trialkylphosphit einverleibt, worauf das erhaltene
Reaktionsgemisch bei -20°C bis +10°C gerührt wird. Nach
beendeter Umsetzung wird das Reaktionsgemisch langsam auf
Eis gegossen, damit die Reaktionstemperatur nicht über 20°C
steigt. Anschließend wird die Lösung mit einer gesättigten
NaHCO₃-Lösung neutralisiert, worauf das Reaktionsprodukt
mit Methylenchlorid extrahiert wird. Die organische Schicht
wird mit Wasser und Salzlake gewaschen und eingeengt, wobei
der jeweilige N-Alkyl- bzw. N-Cycloalkylaminomethylphosphonsäuredialkylester
in guter Ausbeute erhalten wird. Gegebenenfalls
erhält man die entsprechende Säure durch
Hydrolyse ohne Isolieren des Esters. Das ölige Reaktionsprodukt,
nämlich der N-substituierte Aminomethylphosphonsäuredialkylester,
wird mit konzentrierter Salzsäure unter
Rückflußtemperatur hydrolysiert. Eine einfache Hydrolyse
erreicht man auch mit 1,5-Moläquivalenten Bromtrimethylsilan
bei Raumtemperatur (vgl. C. E. McKenna und Mitarbeiter in
"Tetrahydron Letters", 155 (1977)). Nach beendeter Hydrolyse
werden sämtliche flüchtigen Bestandteile (Lösungsmittel und
Nebenprodukte) abdestilliert, worauf zur Kristallisation
der jeweiligen N-substituierten Aminomethylphosphonsäure
ein niedrigmolekularer Alkohol, wie Methanol, Ethanol oder
Isopropanol, zugegeben wird. Das erhaltene Reaktionsprodukt
wird abfiltriert und mit einem niedrigmolekularen Alkohol
gewaschen, wobei die jeweils gewünschte N-substituierte
Aminomethylphosphonsäure in hoher Ausbeute und hoher Reinheit
in kristalliner Form erhalten wird.
Die folgenden Beispiele sollen das Verfahren gemäß der Erfindung
näher veranschaulichen.
12,9 g (0,1 Mol) Trimethylhexahydro-s-triazin in 150 ml
Methylenchlorid werden bei -10°C mit 56,7 g (0,3 Mol)
Titantetrachlorid versetzt, worauf das Gemisch 10 min
lang bei -10°C verrührt wird. Die hierbei gebildete dunkelrote
Lösung wird mit 37,3 g (0,3 Mol) Trimethylphosphit bzw.
49,8 g (0,3 Mol) Triethylphosphit versetzt. Danach wird das
Reaktionsgemisch 1,5 h lang bei -10°C bis +10°C verrührt.
Nach beendeter Umsetzung wird das Reaktionsprodukt langsam
auf 200 g Eis gegossen, damit die Temperatur nicht über 20°C
steigt. Anschließend wird die Lösung mit gesättigter NaHCO₃-
Lösung neutralisiert, worauf das Reaktionsprodukt mit
Methylenchlorid extrahiert wird. Nach Entfernung des Lösungsmittels
wird der Methylaminomethylphosphonsäuredimethylester
bzw. -diethylester in 74- bzw. 80%iger Ausbeute
isoliert. Die NMR-Spektren des gereinigten Dimethylesters
bzw. Diethylesters der Methylaminomethylphosphonsäure entsprechen
ihren Strukturen.

Eine Verseifung erreicht man durch 4stündiges Erwärmen einer
Lösung des Diethylesters der Methylaminomethylphosphonsäure
mit 100 ml konzentrierter Salzsäure auf Rückflußtemperatur
und anschließendes Einengen des Reaktionsgemischs zur
Trockene. Der ölige Rückstand wird aus Ethanol/Wasser umkristallisiert,
wobei 21,3 g (57%) reine Methylaminomethylphosphonsäure
eines Fp von 272-274°C erhalten werden. Die
Hydrolyse des Methylaminophosphonsäuredimethylesters
liefert dasselbe Produkt in 51%iger Gesamtausbeute.
Beispiel 1 wird wiederholt, wobei jedoch das Trimethylhexahydro-
s-triazin durch eine äquimolare Menge Triethylhexahydro-
s-triazin ersetzt wird. Letztlich erhält man in 84%iger
Ausbeute den Ethylaminomethylphosphonsäurediethylester.

Die Hydrolyse des Esters nach dem in Beispiel 1 beschriebenen
Verfahren liefert in 58%iger Gesamtausbeute kristalline
feste Ethylaminomethylphosphonsäure eines Fp von 276-279°C.
Beispiel 1 wird wiederholt, wobei jedoch das Trimethylhexahydro-
s-triazin durch eine äquimolare Menge Triisopropylhexahydro-
s-triazin ersetzt wird. Letztlich erhält man in
68%iger bzw. 71%iger Ausbeute den Isopropylaminomethyl-
phosphonsäuredimethylester bzw. -diethylester.

Eine Hydrolyse des Diethylesters nach dem in Beispiel 1
beschriebenen Verfahren liefert in 59%iger Gesamtausbeute
kristalline feste Ethylaminomethylphosphonsäure eines
Fp von 267-269°C.
Beispiel 1 wird wiederholt, wobei jedoch das Trimethylhexahydro-
s-triazin durch eine äquimolare Menge Tri-n-butyl-
hexahydro-s-triazin ersetzt wird. Letztlich erhält man
in 86%iger Ausbeute n-Butylaminomethylphosphonsäurediethylester.

Eine Hydrolyse des Esters nach dem in Beispiel 1 beschriebenen
Verfahren liefert in 51%iger Gesamtausbeute kristalline
feste n-Butylaminomethylphosphonsäure eines Fp von
236-240°C.
Beispiel 1 wird wiederholt, wobei jedoch das Trimethylhexahydro-
s-triazin durch 13,9 g (30 mMol) Tri-n-octyl-
hexahydro-5-triazin ersetzt und 17,1 g (90 mMol) Titantetrachlorid
und 11,2 g (90 mMol) Trimethylphosphon verwendet
werden. Letztlich erhält man in 91%iger Ausbeute
den n-Octylaminomethylphosphonsäuredimethylester.

Durch Verseifung des Dimethylesters mit 40 ml konzentrierter
Salzsäure unter Rückflußtemperatur und anschließendes
Umkristallisieren in Isopropanol/Wasser erhält man die
n-Octylaminomethylphosphonsäure eines Fp von 262-265°C
in 66%iger Gesamtausbeute.
Beispiel 1 wird wiederholt, wobei jedoch 8,4 g (10 mMol)
Tri-n-Octadecylhexahydro-s-triazin, 5,7 g (30 mMol) Titantetrachlorid
und 3,7 g (30 mMol) Trimethylphosphin verwendet
werden. Letztlich erhält man in 85%iger Ausbeute
den n-Octadecylaminomethylphosphonsäuredimethylester.

Durch Verseifen des Dimethylesters mit 20 ml konzentrierter
Salzsäure unter Rückflußtemperatur und anschließendes Umkristallisieren
in Isopropanol/Wasser erhält man die
kristalline feste n-Octadecylaminomethylphosphonsäure eines
Fp von 110-112°C in 49%iger Gesamtausbeute.
Beispiel 1 wird wiederholt, wobei jedoch das Trimethylhexahydro-
s-triazin ersetzt wird. Letztlich erhält man in
78%iger bzw. 81%iger Ausbeute den Cyclohexylaminomethylphosphonsäuredimethylester
bzw. -diethylester.

Eine Hydrolyse des Diethylesters nach dem in Beispiel 1 beschriebenen
Verfahren liefert in 61%iger Gesamtausbeute
kristalline feste Cyclohexylaminomethylphosphonsäure eines
Fp von 281-284°C.
Claims (3)
1. Verfahren zur Herstellung von N-alkylsubstituierten
Aminomethylphosphonsäurederivaten der Formel
worin bedeuten:
R₁ eine gegebenenfalls substituierte Alkyl- oder Cycloalkylgruppe mit 1 bis 18 Kohlenstoffatomen und
R₂ ein Wasserstoffatom oder eine Methyl- oder Ethylgruppe,
dadurch gekennzeichnet, daß man ein Triazin der Formel worin R₁ die angegebene Bedeutung besitzt, in Gegenwart von Methylenchlorid mit einem Trialkylphosphit der FormelP(OR₂)₃ (III)worin R₂ für eine Methyl- oder Ethylgruppe steht, und Titantetrachlorid umsetzt und das Reaktionsprodukt zur Herstellung einer Verbindung der Formel (I) mit R₂ gleich einem Wasserstoffatom
R₁ eine gegebenenfalls substituierte Alkyl- oder Cycloalkylgruppe mit 1 bis 18 Kohlenstoffatomen und
R₂ ein Wasserstoffatom oder eine Methyl- oder Ethylgruppe,
dadurch gekennzeichnet, daß man ein Triazin der Formel worin R₁ die angegebene Bedeutung besitzt, in Gegenwart von Methylenchlorid mit einem Trialkylphosphit der FormelP(OR₂)₃ (III)worin R₂ für eine Methyl- oder Ethylgruppe steht, und Titantetrachlorid umsetzt und das Reaktionsprodukt zur Herstellung einer Verbindung der Formel (I) mit R₂ gleich einem Wasserstoffatom
- a) mit konzentrierter Salzsäure versetzt und 4 h lang auf Rückflußtemperatur erhitzt oder
- b) mit 1,5 Molen Bromtrimethylsilan versetzt und bei 20°C stehen läßt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß
man das Triazin der Formel (II), das Titantetrachlorid
und das Trialkylphosphit der Formel (III) miteinander
im Molverhältnis 1 : 3 : 3 umsetzt.
3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß
man die Umsetzung bei -20°C bis +20°C durchführt.
