Folglich hat die Duftindustrie ein großes Interesse an synthetischen Duftstoffen mit ambraähnlichen Eigenschaften. Naturambra und Verbindungen, die Amberdüfte besitzen, werden von G. Ohloff, Kapitel 15 in Fragrance Chemistry: The Science of the Sense of Smell, herausgegeben von E.T. Theimer, Academic Press, 1982 ausführlich diskutiert. Verbindungen, die starke Ambra- oder ambraähnliche Gerüche haben, haben im allgemeinen bizyklische oder trizyklische Strukturen. Z. B. besitzt Dodecahydro-3a,6,6,9-tetramethylnaphtho[2,1-b]furan (I) einen starken Ambrageruch (siehe M. Hinder und M. Stoll, Helv. Chim. Acta (1950)33 1308).
- α-Ambrinol (II) ist ein wichtiger synthetischer Ambraduftstoff (siehe M. Stoll und M. Hinder, Helv. Chim. Acta, (1955), 38. 1953). Die Verbindungen III und IV die von G. Ohloff et al., Helv. Chim. Acta, (1976). 59 75 und ebenda (1973), 56 1414 beschrieben wurden, besitzen ebenfalls Ambradüfte.
- Vor kurzem wurden im U.S.-Patent 4 162 226 (1979) von D. Helmlenger und P. Naegeli Verbindungen der Struktur V beschrieben, worin drei der R-Gruppen Methyl sind und eine Wasserstoff ist, und eines von R&sub1; Wasserstoff und das andere R&sub1; Acetyl ist.
- In DE-A-2 442 024 werden Ketone der allgemeinen Formel (I) offenbart
- worin 0, 1 oder 2 der Gruppen R¹ bis R&sup4; unabhängig voneinander Methyl darstellen, und der Rest für Wasserstoff steht.
- Diese ungesättigten Ketone besitzen wertvolle Dufteigenschaften und können als Duftstoffe verwendet werden. Einige von ihnen zeigen einen Duft, der dem der Ketone ähnelt, aus denen sie hergestellt wurden, andere riechen süß, trocken, mehr nach Ambra.
- Nichts was Stand der Technik ist, beschreibt oder deutet daraufhin, daß die neuartigen monozyklischen Verbindungen der Erfindung wertvolle Ambra oder holzig-ambraähnliche Gerüche besitzen könnten. Eine Ambraduftverbindung α-Ambrinol
- Die vorliegende Erfindung zielt auf Alkyltetramethylcyclohexan-Derivate und ihre Verwendung in Duftzubereitungen als Ambradüfte, holzige, würzige oder ambraartige Düfte.
- Die Alkyltetramethylcyclohexan-Derivate haben die Formel
- worin X für H, R¹CO- oder R¹CHOH- steht; Y für H oder R¹CO- steht; R¹ Wasserstoff oder eine Alkylgruppe mit 1 bis 4 Kohlenstoffatomen ist: R² bis R&sup6; für Methyl- oder Niederalkylgruppen oder für 1 bis 4 Kohlenstoffatome stehen: eine Einfach- oder Doppelbindung bedeutet; unter der Maßgabe, daß der Kohlenstoff in Position 4 und 4 nur eine Doppelbindung hat, daß mindestens vier von R² bis R&sup6; Methyl bedeuten und daß eines von X und Y, aber nicht beide, immer für H steht. Die Zahlen bezeichnen die Positionen der Kohlenstoffe der Cyclohexanderivate.
- Bevorzugte Derivate schließen folgende ein: jene, worin Y für H steht; jene, worin R², R³. R&sup5; und R&sup6; für Methyl stehen; jene, worin R¹ für Methyl, Ethyl oder Isopropyl steht; jene. worin X für Carbonyl steht und jene, worin die Bindungen zwischen den Kohlenstoffen 4' und 5' oder 3 und 4 doppelt sind.
- Besonders bevorzugte Spezies schließen folgende ein: 4-(2,2,4,6,6-Pentamethyl-3-cyclohexen-1-yl)-4-penten-2-on mit einem starken holzig-würzigen Ambrageruch mit pudrig-fruchtigen Duftnoten (E,Z)4-(2,2,4,6,6-Pentamethyl-3-cyclohexen-1-yl)-3-penten-2-on mit einem starken holzig-würzigen Ambrageruch mit pudrig-fruchtigen Duftnoten 5-(2,2,4,6,6-Pentamethyl-3-cyclohexen-1-yl)-5-hexen-3-on α-2,2,4,6,6-Hexamethyl-γ-methylen-3-cyclohexen-1-propanol mit einem starken, holzig-erdigen Ambrageruch (E,Z) 4-(2,2,4,6,6-Pentamethyl-3-cyclohexen-1-yl)-3-penten-2-ol mit einem holzigen Ambrageruch 2,2,4,6,6-Pentamethyl-γ-methylen-3-cyclohexen-1-propanol mit einem mäßig holzigen Ambrageruch mit einer würzigen Duftnote (E,Z) 3-(2,2,4,6,6-Pentamethyl-3-cyclo-hexen-1-yl)-2-buten-1-ol α-Ethyl-2,2,4,6,6-pentamethyl-γ-methylen-3-cyclohexen-1-propanol mit einem mäßig holzigen Ambrageruch mit grün-blumigen Duftnoten α(1-Methylethyl)-2,2,4,6,6-pentamethyl-γ-methylen-3-cyclohexen-1-propanol mit einem schwachen, holzigen Ambrageruch α,γ,2,2,4,6,6-Heptamethyl-3-cyclohexen-1-propanol mit einem schwachen, holzigen Geruch α,γ,2,2,4,6,6-Heptamethyl-cyclohexan-1-propanol mit einem sehr schwachen holzigen Geruch 4-(2,2,4,6,6-Pentamethyl-3-cyclohexen-1-yl)-2-pentanon 4-(2,2,4,6,6-Pentamethyl-1-cyclohexyl)-2-pentanon
- Ebenfalls in der Erfindung eingeschlossen sind parfümierte Zubereitungen der vorangehenden Cyclohexanderivate und ein Träger. Diese Zubereitungen umfassen ein Kölnischwasser, Toilettewasser, Parfüm, Öl, Lotion, Creme, Talk, Körperpuder oder ein Körperspray. Diese Zubereitungen werden durch Kombination des Trägers mit einem Duftöl aus dem vorangehenden Derivat und einem aromatischen Spiritus hergestellt.
- Zusätzlich können diese Zubereitungen als Lufterfrischer, Raumlufterfrischer und dergleichen verwendet werden.
- Die Derivate dieser Erfindung können einfach aus 4-(1-Methylethenyl)-1-cyclohexen- Derivat (VI) gemäß der Reaktion A wie folgt hergestellt werden. Reaktion A
- Das Startmaterial VI kann, wie von H.M.R. Hoffman und H. Vathke-Ernst, Chem. Ber., 114. 1981, 1182-1186 beschrieben. erhalten werden. Die Reaktion von VI mit Niederalkyl- Säureanhydriden in der Gegenwart von sauren Katalysatoren stellt eine Mischung der Verbindungen VIIa-c bereit.
- Die relativen Anteile von VIIa-c werden je nach angewandten Bedingungen und der Identität von R¹ variieren. Die Gegenwart der Verbindung VIIc hat keinen Einfluß auf den Geruch und die Stärke der Verbindungen VIIa und b. Unter typischen Reaktionsbedingungen wird die Verbindung VIIb in vorherrschendem Anteil hergestellt, während die Verbindung VIIa in geringerem Anteil hergestellt wird. (Siehe C.D.Nenitzescu und A.T. Balaban in "Friedel-Crafts" und Verwandte Reaktionen. G. Olah, Herausgeber, Band 3, Pt.
- 2, 1963, S. 1033-1152). Anstelle von Anhydriden können auch Säurechloride verwendet werden.
- Lewis-Säuren und protonische Säuren können beide als Katalysatoren verwendet werden. Protonische Säuren, wie Polyphosphorsäure, Schwefelsäure, Phosphorsäure, Phosphorsäureahhydrid, Methansulfonsäure und deren Mischungen können verwendet werden. Lewis-Säuren, wie Aluminiumchlorid, Eisen(III)-chlorid, Alkylaluminiumchlorid, Bortrifluorid-Etherat, Zinkchlorid, Zinnchlorid oder Titanchlorid, sind die bevorzugten Katalysatoren. Die am meisten vorzuziehenden Katalysatoren sind Zinkchlorid oder Bortrifluorid-Etherat. Die Menge des Katalysators kann von 5 bis 200 Mol-% relativ zur Verbindung VI betragen, wobei 25 bis 75 Prozent bevorzugt werden, und 35 bis 50 Molprozent besonders bevorzugt werden.
- Ein Vielzahl von Lösungsmitteln kann verwendet werden, wie Dichlormethan. Hexan, Chlorbenzol, Kohlenstofftetrachlorid. Tetrachlorethylen oder Kohlenstoffdisulfid. Die bevorzugten Lösungsmittel sind Dichlormethan oder Dichlorethan.
- Die Reaktion kann im Temperaturbereich von -10ºC bis 100ºC durchgeführt werden. Der bevorzugte Temperaturbereich liegt zwischen 0º und 50ºC. Der am meisten bevorzugte Temperaturbereich ist von 20ºC bis 45ºC. Die Zeit für die Durchführung der Reaktion kann von 1 bis 12 Stunden betragen. Vorzugsweise 1 bis 6 Stunden, am meisten vorzuziehen sind 3 bis 5 Stunden. Reaktion B
- Wie in der Reaktion B gezeigt können die vorweg genannten Alkohole VIIa und VIIb durch Reduktion der Ketone VIIIa und VIIIb mittels Standardverfahren, die Fachleuten geläufig sind, hergestellt werden. Siehe C.A. Buehler und D.E.Pearson, "Survey of Organic Synthesis". Wiley-Interscience, Band 1(1920), S. 193-207 und Band 2 (1977), S.
- 228-239. Reduktion mit Metallhydriden, wie Natriumborhydrid oder Lithiumaluminiumhydrid ist ein geeignetes Verfahren. Reaktion C
- Alternativ kann der Alkohol VIIIa, wie in der Reaktion C gezeigt, durch Kondensation von 1,3,3,5,5-Pentamethyl-4-(1-methylethenyl)-1-cyclohexen (VI) mit Aldehyden in der Gegenwart von Lewis-Säuren hergestellt werden. (Siehe B.B. Snider et al., Tetrahedron 37 3927-34 (1981)). Der Alkohol VIIIa (R = H) kann unter Verwendung jeder der voranstehenden Lewis-Säuren als Katalysatoren hergestellt werden, wobei Bortrifluorid- Etherat und Aluminiumchlorid bevorzugt werden. Die Alkohole VIIIa (R' = Niederalkyl) werden am besten hergestellt, indem VI mit einem Aldehyd in Anwesenheit von Dimethylaluminiumchlorid zur Reaktion gebracht wird. Viele inerte Lösungsmittel können bei diesen Reaktionen verwendet werden, aber Dichlormethan oder 1,2-Dichlorethan sind bevorzugt. Die Reaktionen werden im Temperaturbereich von -20ºC bis 75ºC durchgeführt. Der bevorzugte Temperaturbereich liegt zwischen -10ºC und 50&sup0;C, am meisten vorzuziehen sind 0ºC bis 40ºC. Nützliche Lösungsmittel schließen jene oben für Reaktion A beschriebenen ein. Die Reaktionszeit ist gleich der, die für die vorangehende Reaktion A beschrieben wurde. Reaktion D
- Die Oxidation des Alkohols Villa (Reaktion D) mittels Standardreagenzien, wie Chromtrioxid Aluminium-t-butoxid, n-Bromsuccinimid und dergleichen, liefert die Carbonylverbindungen VIIa und/oder VIIb (siehe C.A. Buehler und D.E. Pearson "Survey of Organic Synthesis", Wiley-Interscience, Band 1(1970), S. 625-630, S. 545-553 und Band 2 (1977), S.484-487, S. 536-540). Die Carbonylverbindungen VIIa können problemlos (manchmal mit einer Oxidation einhergehend) zu den Verbindungen VIIb isomerisiert werden. Pyridin-chlorchromat (siehe E.J. Corey und J.W.Suggs, Tetrahedron Lett, 1975, 2647-2650) ist ein besonders wirksames Reagenz für diese Reaktion. Durch Regulieren der Reaktionsbedingungen kann entweder VIIa oder VIIb in hoher Ausbeute hergestellt werden.
- Die Verbindung VIIa kann selektiv entweder zu der Verbindung IX oder X durch Anwendung der geeigneten Hydrierungsbedingungen reduziert werden, unter der Verwendung solcher Katalysatoren, wie Palladium, Platin, Raney-Nickel und dergleichen (siehe P.N. Rylander, "Catalytic Hydrogenation in Organic Synthesis", Academic Press (1979) S. 51-59).
- Die reduzierten Alkohole XI oder XII können durch Reduktion der zugehörigen Ketone IX oder X mittels solcher Reagenzien, wie Lithium-Aluminiumhydrid, Natriumborhydrid, Dialkyl-Aluminiumhydrid, Natriumcyanoborhydrid, Wasserstoff und Edelmetallkatalysatoren, sowie anderer, erhalten werden. Nützliche Lösungsmittel schließen Alkohole, Tetrahydrofuran, Wasser, Dioxan und Ether ein. Nützliche Temperaturen schließen 0ºC bis 80ºC ein, vorzugsweise 0ºC bis 60ºC. am meisten vorzuziehen sind 10ºC bis 60ºC.
- Die Alkohole XI können auch durch Reduktion der Ketone VIIa mit einem Alkalimetall in der Gegenwart eines Protonendonators hergestellt werden (siehe H.O. House "Modern Synthetic Reactions", The Benjamini Cummings Publishing Co. (1972) S. 145-205). Nützliche Alkalimetalle schließen Lithium, Natrium, Calcium und Kalium ein, wobei Lithium und Natrium besonders bevorzugt sind. Die Reduktion kann in Lösungsmitteln, wie Alkoholen, Ether, Ammoniak oder Niederalkylaminen durchgeführt werden. Besonders bevorzugt sind Mischungen aus Alkohol. Ether und Ammoniak.
- Mischungen der gemäß der voranstehenden Verfahren hergestellten Derivate können unter Anwendung von Fachleuten geläufigen Techniken getrennt und gereinigt werden. Eingeschlossen sind die Vakuumdestillation. Säulenchromatographie, fraktionierte Kristallisation, Hochdruck-Flüssigchromatographie (HPLC) und dergleichen.
- Die aus den Derivaten gemäß der vorliegenden Erfindung hergestellten Duftkompositionen können gemäß in der Parfümtechnik bekannten Verfahren zubereitet werden. Das Derivat wird zuerst mit einem aromatischen Spiritus zur Bildung einer Ölessenz compoundiert. Nützliche Spiritsorten schließen Ethanol, Propanol, Ethylenglykol, Glycerin und dergleichen ein. Die Ölessenz wird sodann mit einem Träger, ausgewählt aus den typischerweise für Talks, Lotionen, Sprays, Kölnischwassern, Parfüms und dergleichen verwendeten, zubereitet. Diese Träger enthalten Inhaltsstoffe, wie Alkohole, Glycerine, Emulgatoren, Glykole, Wasser, Starken. Mineralöl, Wachs, Rohvaseline bzw. Petrolatium, Lanolinderivate, Fettsäuren, Fettalkohole, Salze, Kollagen, oberflächenaktive Stoffe, Talke, Metallchelate und dergleichen.
- Die folgenden Beispiele führen die Erfindung weiter aus. BEISPIEL 1 Cyclohexen-Butenol-Derivat
- Eine Lösung aus Bortrifluorid-Etherat (2 ml) in Dirchlormethan (30 ml) wurde einer Mischung aus 1,3,3,5,5-Pentamethyl-4-(1-methylethenyl)-1-cyclohexen (19,24 g, 0,1 Mol) und Paraformaldehyd (1,5 g, 0,05 Mol) bei 25ºC in Dichlormethan (200 ml) über 45 min zugegeben. Die Mischung wurde bei 25ºC während 5 Stunden gerührt. Daraufhin wurde eine 30%ige, wäßrige Kaliumcarbonatlösung (50 ml) tröpfchenweise zugegeben. Die wäßrige Schicht wurde mit Dichlormethan (50 ml) extrahiert. Die Dichlormethanlösung wurde mit 10%iger, wäßriger Natriumhydroxidlösung (100 ml), Wasser (100 ml), Salzlösung (100 ml) gewaschen und über Natriumsulfat getrocknet. Die Verdampfung des Lösungsmittels und die Destillation lieferten folgendes: 5,5 g an 2,2,4,6,6-Hexamethyl-γmethylen-3-cyclohexen-1-propanol, Sdp. 100-104ºC, 0,5 mm (GLC-Reinheit: 94%). ¹H- NMR (60 MHz, CDCl&sub3;) δ: 0 94 und 1.03 (12H, 2s), 1,63 (3H, s: breit), 1,6-2,7 (6H, m) 3,65-3,98 (2H, m), 4,93 (1H, s: breit). 4,95-5,13 (2H, m); IR (Film) vmax: 3300, 2950, 1640. 1465, 1440, 1380, 1360 cm&supmin;¹; MS m/e: 222, 207, 204, 189, 126, 111, 96. BEISPIEL 2 Cyclohexen-Pentenol-Derivat
- Dimethylaluminiumchlorid (300 ml einer 1 M Hexanlösung) wurde einer gekühlten Lösung (10 C) aus 1,3,3,5,5-Pentamethyl-4-(1-methylethenyl)-1-cyclohexen (46, 18 g, 0,24 Mol) in Dichlormethan (500 ml) zugesetzt. Die Mischung wurde bei 25ºC während 2 Stunden gerührt. Daraufhin wurde die Mischung durch Zugabe einer Phosphatpufferlösung (200 ml, hergestellt aus 100 ml 0,1 M Kaliumphosphat, einwertig, und 44,8 ml von auf 200 ml mit Wasser verdünntem 0, 1 M Natriumhydroxid) hydrolysiert, gefolgt von der Zugabe von ausreichend 1 N Salzsäure, um die Aluminiumsalze zu lösen. Die Schichten wurden getrennt, und die wäßrige Schicht mit Dichlormethan extrahiert (200 ml). Die vereinigte organische Schicht wurde mit Dichlormethan (200 ml) extrahiert. Die vereinigten organischen Schichten wurden mit Wasser (100 ml) und gesättigter Natriumbicarbonatlösung (2·100 ml) gewaschen. Die Lösungsmittel wurden verdampft, und der Rückstand wurde chromatographiert. Die Vereinigung von Fraktionen und der Destillation lieferten folgendes: 27,8 g an α,2,2,4,6,6-Hexamethyl-γ-methylen-3-cyclohexen-1-propanol, Sdp. 100-105ºC, 0,5 mm (GLC-Reinheit, zwei Isomere, 41,4% und 56,6%). ¹H-NMR (60 MHz, CDCl&sub3;) :6 0,92 und 0,98 (12H, 2s), 1,20 (3H, d, J = 6 Hz), 1,58 (3H, s) 1,2- 2,6 (5H, m) 3,8-4,2 (1M, m), 4,9-5,2 (3H, m); IR (Film) vmax: 3340, 2950, 1635, 1440, 1370. 1350 cm&supmin;¹; MS m/e: 236, 221, 218, 203, 140, 125, 107, 96. BEISPIEL 3 Cyclohexen-Heptenol-Derivat
- Dimethylaluminiumchlorid (60 ml einer 1 M Hexanlösung) wurde tröpfchenweise einer Lösung aus 1,3,3,5,5-Pentamethyl-4-(1-methylethenyl)-1-cyclohexen (7,70 g, 0,04 Mol) und Propionaldehyd (2,30 g, 0,04 Mol) in Dichlormethan (150 ml) bei 25-30ºC über eine 15 minütige Zeitdauer zugesetzt. Die Mischung wurde bei 25ºC während 16 Stunden gerührt. Die Aufarbeitung (wie in Beispiel 2 beschrieben) und die Chromatographie lieferten folgendes: 2,20 g an α-Ethyl-2,2,4,6,6-pentamethyl-γ-methylen-3-cyclohexen-1- propanol, Sdp. 125ºC (Kugelrohrbad), 0,5 mm, (GLC-Reinheit, zwei Isomere. 40.7% und 57.9%). ¹H-NMR (60 MHz, CDCl&sub3;) δ: 0,98 und 1,03 (12H, 2s), 1,02 (3H, t J = 4 Hz), 1.63 (3H, s), 1,1-2.6 (8H, m), 3,5-3.9 (1H, m), 5,0-5,3 (3H, s: breit); IR (Film) vmax: 3350, 2950, 1640, 1460, 1380. 1360 cm&supmin;¹: MS m/e: 250, 235, 217, 125, 107, 96. Die Massenspektren der Diastereomere waren fast identisch. BEISPIEL 4 Cyclohexen-Methyl-Heptenol-Derivat
- Dimethylaluminiumchlorid (3 ml einer 1 M Hexanlösung) wurde mit einer Lösung von 1,3,3,5,5-Pentamethyl-4-(1-methylethenyl)-1-cyclohexen (0,48 g, 0,0025 mol) und Isobutyraldehyd (0,18 g, 0,0025 Mol) in Dichlormethan (10 ml) gemäß dem in Beispiel 3 beschriebenen Verfahren zur Reaktion gebracht. Aufarbeitung und Chromatographie ergaben folgendes: 0.300 g (45% Ausbeute) an α-(1-Methylethyl)-2,2,4,6,6-pentamethyl-γ -methylen-3-cyclohexen-1-propanol, Sdp. 135ºC (Kugelrohrbad), 0,5 mm (GLC-Reinheit 94,4%, 2 : 1 Mischung der Diastereomere). ¹H-NMR (60 MHz, CDCl&sub3;) δ: 0,88-1,07 (18H, 1d und 1 breites s), 1,4-2,4 (10H, m) 3,2-3,7 (1H, m) 4,9-5,2 (3H, m); IR (Film) vmax: 3450, 2950, 1640, 1465, 1380, 1360 cm&supmin;¹; MS m/e: 264, 249, 246, 221, 203, 177, 107, 96. Die Massenspektren der Diastereomere waren fast identisch. BEISPIEL 5 Cyclohexen-Butenol-Derivat
- Eine Lösung von Essigsäureanhydrid (0,23 ml) in Dichlormethan (2 ml) wurde mit einer Mischung aus 2,2,4,6,6-Pentamethyl-γ-methylen-3-cyclohexen-1-propanol (0.56 g, 0,0025 mol). Triethylamin (0,41 ml), 4-Dimethylaminopyrimidin (0,031 g) und Dichlormethan bei 25ºC zur Reaktion gebracht. Aufarbeitung, Chromatographie und Destillation ergaben 0,60 g an 2,2,4,6,6-Pentamethyl-γ-methylen-3-cyclohexen-1-propanol-acetat. Sdp. 110- 120ºC (Kugelrohrbad), 0,5 mm (GLC-Reinheit: 88%). ¹H-NMR (60 MHz. CDCl&sub3;) δ: 0,93 (6H, s), 1,00 und 1,03 (6H, 2s), 1,60 (3H, s), 2,00 (3H, s), 0,9-2,6 (5H, m), 4,20 (1H, t, J = 7 Hz), 4,96-5,07 (3H, m); IR (Film) vmax: 2960, 1735, 1635, 1440, 1370, 1350 cm&supmin;¹; MS (m;e): 264, 249, 221, 204, 189, 108, 96. BEISPIEL 6 Cyclohexen-Butenol-Derivat
- Natriumhydrid (0,6 g einer 60%igen Öldispersion, 0,015 Mol) wurde mit Hexan (2·3 ml) gewaschen und in THF (10 ml) suspendiert. Eine Lösung aus 2,2,4,6,6-Pentamethyl-γmethylen-3-cyclohexen-1-propanol (1,11 g, 0,005 Mol) in THF (5 ml) wurde zugegeben, gefolgt von Methyliodid (1,4 g, 0,001 Mol). Die Mischung wurde unter Rückfluß während 4 Stunden erhitzt. Daraufhin ergaben Verarbeitung, Chromatographie und Kugelrohrdestillation folgendes: 1,10 g an 2,2,4,6,6-Pentamethyl-γ-methylen-3-cyclohexen- 1-propanolmethylether, ¹H-NMR (60 MHz, CDCl&sub3;) 6 : 0,95 (6H, s), 1,03 (6H, s: breit), 1,63 (3H, s: breit), 1,5-2,6 (5H, m), 3,33 (3H, s) 3,58 (2H, t, J = 7 Hz), 4,85-5,15 (3H, m); IR (Film) vmax: 2960, 1640, 1440, 1380, 1360 cm&supmin;¹; MS m/e: 236, 221, 204, 189, 140, 125, 96. BEISPIEL 7 Cyclohexen-Pentenon-Derivat
- Eine Lösung aus α,2,2,4,6,6-Hexamethyl-γ-methylen-3-cyclohexen-1-propanol (2,36 g, 0.01 Mol) in Dichlormethan (10 ml) wurde während einer 30 minütigen Zeitdauer einer Mischung von Pyridiniumchlorchromat (3,25 g, 0,015 Mol), Natriumacetat (2,30 g, 0,03 Mol) und Dichlormethan (15 ml) bei 25ºC zugegeben. Die Mischung wurde filtriert und das Filtrat mit Wasser (100 ml) und 5%iger Natriumcarbonatlösung (100 ml) gewaschen. Die Lösungsmittel wurden verdampft, und der Rückstand wurde chromatographiert, wodurch nach Kugelrohrdestillation folgendes erhalten wurde: 1,75 g (75% Ausbeute) an 4-(2,2,4,6,6-Pentamethyl-3-cyclohexen-1-yl)-4-penten-2-on (GLC-Reinheit: 94%). ¹H-NMR δ: 0,93 (3H, s) 0,95 (3H, s), 1,02 (6H, s), 1,62 (3H, s) 1,6-1.8 (2H, m), 1.97 (1H, s), 2,18 (3H, s), 3,15 (2H, s), 5,05 (2H, s: breit); IR (Film) vmax: 2950, 1710, 1630, 1430, 1380, 1350 cm&supmin;¹; MS m/e: 234, 219, 201, 191, 176, 161, 149, 123, 96. BEISPIEL 8 Cyclohexen-Pentenon-Derivat
- Eine Mischung aus 4-(2,2,4,6,6-Pentamethyl-3-cyclohexen-1-yl)-4-penten-2-on (1 ,50 g, 0,0063 Mol), Methanol (30 ml) und Natriummethoxid (0,02 g) wurde bei 25ºC während 18 Stunden verrührt. Die Mischung wurde dann unter Rückfluß während 3 Stunden erhitzt. Die Mischung wurde auf 25ºC abgekühlt, und das meiste Methanol wurde unter verringertem Druck verdampft. Der Rückstand wurde zwischen Ether (75 ml) und Wasser (15 ml) aufgeteilt. Die wäßrige Schicht wurde mit Ether (50 ml) extrahiert. Die Etherextrakte wurden mit gesättigter Natriumbicarbonatlösung sowie einer Salzlösung gewaschen und getrocknet. Verdampfung der Lösungsmittel und die Kugelrohrdestillation des Rückstandes lieferten folgendes: 1,28 g (85% Ausbeute) an 4-(2,2,4,6,6-Pentamethyl-3-cyclohexen-1- yl)-3-penten-2-on (GLC-Reinheit: 93,5%). ¹H-NMR (60 MHz) δ: 0,93 (12h, s: breit), 1.57 (3H, s), 1,5-1,9 (2H, m), 1,88 (1H, s: breit), 3,42 (3H, s), 5,01(1H, s: breit), 6,02 (1H, s: breit): IR (Film) vmax: 2950, 1685, 1600, 1440, 1380, 1365 cm&supmin;¹; MS m/e: 234, 216, 191, 149, 135, 121. BEISPIEL 9 Cyclohexen-Pentenon-Derivat
- Bortrifluoridetherat 9,3 ml, 0,07 Mol) wurde tröpfchenweise während einer Zeitdauer von 1 Stunde einer Mischung aus 1,3,3,5,5-Pentamethyl-4-(1-methylethenyl)-1-cyclohexen (28,83 g, 0,15 Mol), Essigsäureanhydrid (123,5 ml) und Dichlormethan (6,5 ml) unter Wahrung der Temperatur bei 40º-45ºC zugegeben. Die Mischung wurde bei 40ºC während 3 Stunden gerührt. Das meiste des überschüssigen Essigsäureanhydrids wurde unter verringertem Druck entfernt. Der Rückstand wurde in Dichlormethan (200 ml) gebracht und mit einer gesättigten Natriumcarbonatlösung während 0,5 Stunden verrührt. Die wäßrige Schicht wurde mit Dichlormethan (50 ml) extrahiert. Die kombinierten organischen Extrakte wurden mit 5%iger Natriumhydroxidlösung (3·100 ml) sowie Salzlösung (100 ml) gewaschen und getrocknet. Entfernung des Lösungsmittels und Destillation des Rückstands lieferten folgendes: 14,4 g (41% Ausbeute) an Ketonen, Sdp. 100-105ºC, 0,5 mm. Die GLC-Analyse zeigt, daß das Produkt 76,4% an 4-(2,2,4,6,6-Pentamethyl-3- cyclohexen-1-yl)-3-penten-2-on und 14,6% einer Komponente enthält, die mittels Chromatographie gereinigt wurde und die, wie mittels Spektroskopie gezeigt wurde, aus 1- [2,2,4,4,6-Pentamethyl-3-(1-methylethenyl)-5-cyclohexen-1-yl]-ethano-n bestand. ¹H-NMR (60 MHz, CDCl&sub3;) δ: 0,92 (3H, s), 1,01(6H, s), 1,13 (3H, s), 1,60 (3H, s) 1,82 (3H, s) 2,20 (3H, s), 2,57 (1H, s), 2,77 (1H, s), 4,75 (1H, s: breit), 4,98 (1H, s: breit), 5,27 (1H, s: breit); IR (Film)vmax: 2950, 1716, 1630, 1440, 1380, 1350 cm&supmin;¹; MS m/e: 234, 219, 216, 201, 191, 149, 135.
- Ein Lithiumgranulat (0,5 g, 0,071 Mol) wurde portionsweise während einer 40 minütigen Zeitdauer einer kalten (-30 bis -40ºC) Mischung aus 4-(2,2,4,6,6-Pentamethyl-3-cyclohexen-1-yl)-3-penten-2-on (1,08 g, 0.0046 Mol), Ether (20 ml), Ethanol (20 ml) und Ammoniak (80 ml) zugegeben. Ammoniumchlorid (8,0 g) wurde der blaugefärbten Mischung zugesetzt. Den Ammoniak ließ man verdampfen. Ether (100 ml) und Wasser (200 ml) wurden dem Rückstand zugesetzt. Die wäßrige Schicht wurde mit Ether (100 ml) extrahiert. Die kombinierten organischen Extrakte wurden mit 0,5 N HCl (2·100 ml) sowie gesättigter Natriumbicarbonatlösung (100 ml) gewaschen und getrocknet. Verdampfung der Lösungsmittel. Chromatographie des Rückstands und Kugelrohrdestillation lieferten folgendes: 0,850 g (78% Ausbeute) an α,γ,2,2,4,6,6-Heptamethyl-3-cyclohexen-1-propanol. Die GLC-Analyse zeigt 4 Isomere mit 4,1%, 12,8% und 82.3% (zwei Peaks wurden nicht vollständig aufgelöst). ¹H-NMR (60 MHz, CDCl&sub3;) δ: 0,98 und 1,05 (12H, 2s), 1,12 (3H, d. J = 6 Hz), 1,25 (3H, d, J = 4 Hz), 1,60 (3H, s), 1,0-2,5 (7H, m), 3,6-4,1(1H, m), 5,00 (1H, s: breit), IR (Film) vmax: 3320, 2950, 1640, 1470, 1450, 1380, 1360 cm&supmin;¹; MS m/e: 238, 223, 180, 151, 109, 96.
- Das folgende zeigt die Anwendbarkeit einer Mischung aus 4-(2,2,4,6,6-Pentamethyl-3- cyclohexen-1-yl)-3-penten-2-on und 1-[2,2,4,6,6-Pentamethyl-3-(1-methylethenyl)-5-cyclohexen-1-yl]ethanon, die Verbindungen aus Beispiel 9, in einer Duftzubereitung vom Chypre-Typ.

- Das folgende stellt die Anwendbarkeit von α,2,2,4,6,6-Hexamethyl-γ-methylen-3- cyclohexen-1-propanol in einer Duftzubereitung vom Muguet-Typ dar.

Claims (31)
1. Alkyltetramethylcyclohexan-Derivat der Formel:
in der X für H, R¹CO- oder R¹CHOH-, Y für H oder R¹CO-; R¹ für Wasserstoff oder eine Alkylgruppe mit 1 bis 4 Kohlenstoffatomen; R², R³,, R&sup4;,, R&sup5; und R&sup6; für Nieder-
Alkylgruppen mit 1 bis 4 Kohlenstoffatomen und für eine Einfach- oder
Doppelbindung stehen: unter der Maßgabe, daß der Kohlenstoff in der Stellung 4 und 4' nur
eine Doppelbindung aufweist, daß mindestens vier von R², R³, R&sup4;, R&sup5; und R&sup6; Methyl
bedeuten, und daß eines von X und Y -aber nicht beide- immer für Wasserstoff steht.
2. Derivat gemäß Anspruch 1, worin R², R³, R&sup4;, R&sup5; und R&sup6; Methyl bedeuten.
3. Derivat gemäß Anspruch 1, worin R¹ Methyl, Ethyl oder Isopropyl bedeutet.
4. Derivat gemaß Anspruch 1, worin X R¹CO bedeutet.
5. Derivat gemäß Anspruch 4, worin die Bindung zwischen den Kohlenstoffatomen in
Stellung 4' und 5' eine Doppelbindung ist und Y für H steht.
6. Derivat gemäß Anspruch 5. worin die Bindung zwischen den Kohlenstoffatomen in
Stellung 3 und 4 eine Doppelbindung ist und Y für H steht.
7. Derivat gemäß Anspruch 5, worin die Bindung zwischen den Kohlenstoffatomen in
Stellung 4 und 5 eine Doppelbindung ist und Y für H steht.
8. Derivat gemäß Anspruch I, worin X R¹CO- oder R¹CHOH- ist und Y für H steht.
9. Derivat gemäß Anspruch 1, worin X H und Y R¹CO- bedeuten, die Bindung
zwischen den Kohlenstoffatomen 3' und 4' eine Doppelbindung ist und die Bindung
zwischen den Kohlenstoffatomen 4 und 5 eine Doppelbindung ist.
10. Mischung eines Derivats gemäß Anspruch 1, worin X R¹CO- und Y H bedeuten
und eines Derivats gemäß Anspruch 1, worin X H und Y R¹CO- bedeuten.
11. 4-(2,2,4,6,6-Pentarnethyl-3-cyclohexen-1-yl)-4-penten-2-on gemäß Anspruch 1.
12. (E,Z) 4-(2,2,4,6,6-Pentamethyl-3-cyclohexen-1-yl)-3-penten-2-on gemäß Anspruch 1.
13. 5-(2,24,6,6-Pentamethyl-3-cyclohexen-1-yl)-5-hexen-3-on gemäß Anspruch 1.
14. 5-(2,2,4,6,6-Pentamethyl-3cyclohexen-1-yl)-2-methyl-5-hexen-3-on gemäß Anspruch 1.
15. (E,Z) 5-(2,2,4,6,6-Pentamethyl-3-cyclohexen-1-yl)-2-methyl-4-hexen-3-on gemäß
Anspruch 1.
16. 3-(2,2,4,6,6-Pentamethyl-3cyclohexen-1-yl)-3-butenal gemäß Anspruch 1.
17. 3-(2,2,4,6,6-Pentamethyl-3cyclohexen-1-yl)-2-butenal gemäß Anspruch 1.
18. α,2,2,4,6,6-Mexamethyl-γ-methylen-3-cyclohexen-1-propanol gemäß Anspruch 1.
19. (E,Z) 4-(2,2,4,6,6-Pentamethyl-3-cyclohexen-1-yl)-3-penten-2-ol gemäß Anspruch 1.
20. 2,2,4,6,6-Pentamethyl-γ-methylen-3-cyclohexen-1-propanol gemäß Anspruch 1.

28. Duftöl, umfassend einen aromatischen Spirirus und ein Alkyltetramethylcyclohexan-
Derivat gemäß Anspruch 1.
29. Öl gemäß Anspruch 28, worin der aromatische Spiritus eine Alkohol- und Glycerin-
Mischung ist.
30. Parfümierte Zubereitung, umfassend das Duftöl gemäß Anspruch 28, kombiniert mit
einem aus der aus Kölnischwasser. Toilettewasser, Parfüm, Lotion, Creme, Talk,
Körperpulver und Körperspray bestehenden Gruppe ausgewählten Träger.
31. Lufterfrischer, umfassend einen Träger und ein Duftöl gemäß Anspruch 28.
