Description
- Die Erfindung betrifft ein Diphenylethylenderivat, ein Verfahren zur Herstellung desselben und eine pharmazeutische Verwendung hiervon.
- Die ernsthaftesten Krankheiten der Menschheit beinhalten im Augenblick akute vaskuläre Krankheiten, wie myokardialer Infarkt, zerebrale Apoplexie, zerebrale Thrombosis, zerebraler Infarkt, pulmonäre Embolie, tiefe Phlebothrombosis und periphere Arteriothrombosis.
- Seit kurzem haben Antiplättchenmittel öffentliche Aufmerksamkeit erweckt und werden klinisch zur Behandlung dieser Krankheiten eingesetzt. Jedoch wurde ihre Anwendbarkeit erst kürzlich erkannt, so dass zu erwarten steht, dass bessere Mittel in der Zukunft entwickelt werden.
- EP-A-0 124 791 offenbart Aralkanamido-Verbindungen, die als pharmazeutische Mittel zur Verbesserung von Arteriosklerosis geeignet sind.
- Das der vorliegenden Erfindung zugrundeliegende Problem besteht darin, Verbindungen zu schaffen, die eine höhere Wirksamkeit bei der Inhibierung der Agglutination von Plättchen und eine geringere Toxizität aufweisen.
- Die Erfindung schafft ein Diphenylethylenderivat der Formel (I)
- wobei R¹ und R² unabhängig voneinander Wasserstoff, Hydroxyl oder eine Niedrigalkoxygruppe bedeuten, X eine Cyanogruppe ist, Y eine Gruppe -R¹&sup0;-COOR³ bedeutet, wobei R³ Wasserstoff oder eine Niedrigalkylgruppe ist, und R¹&sup0; eine Alkylengruppe mit 1 bis 3 Kohlenstoffatomen, geradkettig oder verzweigt, bedeutet, oder ein pharmazeutisch verträgliches Salz hiervon.
- Bei der obigen Definition beinhaltet eine Niedrigalkylgruppe, wie sie mit Hinblick auf den Substituenten R³ genannt ist, geradkettige oder verzweigte Alkylgruppen, die 1 bis 6 Kohlenstoffatome tragen, wie z.B. Methyl, Ethyl, n-Propyl, n-Butyl, Isopropyl, Isobutyl, 1-Methylpropyl, t-Butyl, n-Pentyl, 1-Ethylpropyl, Isoamyl und n-Hexyl. Eine Alkoxygruppe, wie sie im Hinblick auf die Substituenten R¹ und R² genannt ist, beinhaltet jede Niedrigalkoxygruppe, die von den oben genannten Niedrigalkylgruppen abgeleitet ist. Unter diesen Gruppen sind Methyl- und Ethylgruppen die bevorzugtesten Niedrigalkylgruppen, während eine Methoxygruppe die bevorzugteste Niedrigalkoxygruppe ist.
- Ein pharmazeutisch verträgliches Salz der Zielverbindung, bei der R³ ein Wasserstoffatom ist, beinhaltet Metallsalze, wie Salze von Na, K, Ca und Mg.
- Gemäss einer bevorzugten Ausführungsform bedeuten die Substituenten R¹ und R² Niedrigalkoxygruppen, insbesondere Methoxygruppen.
- Besonders bevorzugte Verbindungen sind 4-Cyano-5,5-bis(4-methoxyphenyl)-4-pentensäure, Methyl-5-cyano-6,6-bis(4-methoxyphenyl)-5-hexenoat und Ethyl-4-cyano-5,5-bis(4-methoxyphenyl)-4-pentenoat.
- Darüber hinaus schafft die Erfindung ein Verfahren zur Herstellung des oben definierten Diphenylethylenderivates. Dieses Verfahren umfasst die Reaktionsbildung eines Diphenylethylenderivates der Formel (I) wobei R¹ und R² unabhängig voneinander Wasserstoff, Hydroxyl oder eine Niedrigalkoxygruppe bedeuten, X eine Cyanogruppe ist und Y eine Gruppe -R¹&sup0;-COOR³ bedeutet, wobei R³ Wasserstoff oder eine Niedrigalkylgruppe ist, und wobei R¹&sup0; eine Alkylengruppe mit 1 bis 3 Kohlenstoffatomen ist und eine verzweigte Alkylgruppe tragen kann, oder eines pharmazeutisch verträglichen Salzes hiervon, das die Umsetzung eines Ketons der allgemeinen Formel (VI) wobei R¹ und R² wie oben definiert sind, mit einem Halogenid der allgemeinen Formel (VII)
- wobei Hal ein Halogenatom bedeutet und wobei R³ und R¹&sup0; wie oben definiert sind, in Gegenwart von Zink in einer Reformatsky-Reaktion zur Herstellung einer Hydroxyverbindung der Formel (VIII)
- wobei R¹, R², R³ und R¹&sup0; wie oben definiert sind, und anschliessend die Dehydratisierung der so erhaltenen Hydroxyverbindung (VIII) auf übliche Weise und, falls notwendig, die Umwandlung des Produktes zu einem pharmazeutisch verträglichen Salz umfasst.
- Beispiele für das Lösungsmittel, das für die Reformatsky-Reaktion geeignet ist, sind Tetrahydrofuran, Benzol und Ether. Weiterhin kann eine Lösungsmittelmischung, die z.B. Trimethylborat oder Triethylborat und Tetrahydrofuran umfasst, eingesetzt werden.
- Diese Reaktion kann üblicherweise bei einer Temperatur von etwa -70 bis 150ºC durchgeführt werden.
- Beispiele für die Lösungsmittel, die für die Dehydratisierung geeignet sind, sind Benzol, Toluol, Tetrahydrofuran, Ether und Dioxan, während Beispiele des Katalysators hierfür p-Toluolsulfonsäure, Thionylchlorid, Phosphorpentaoxid, Jod und Salzsäure sind. Diese Reaktion kann bei einer Temperatur von etwa -70 bis 150ºC durchgeführt werden.
- Darüber hinaus liefert die Erfindung eine pharmazeutische Zusammensetzung, die eine pharmakologisch wirksame Menge des Diphenylethylenderivates, wie es oben definiert ist, oder eines pharmakologisch verträglichen Salzes hiervon und einen pharmakologisch verträglichen Träger umfasst.
- In diesem Zusammenhang schafft die Erfindung die Verwendung des oben definierten Diphenylethylenderivates oder eines pharmakologisch verträglichen Salzes hiervon zur Herstellung eines Arzneimittels zur Behandlung einer Krankheit, die durch eine Blutflussstörung verursacht ist.
- Um die vorliegende Erfindung weiter zu erläutern, werden typische Beispiele der Verbindung der vorliegenden Erfindung im folgenden angegeben. Alle diese Verbindungen sind in der freien Form aufgeführt.

- Das Diphenylethylenderivat der Erfindung weist eine ausgezeichnete Wirkung hinsichtlich der pharmakologischen Eigenschaften auf. Es inhibiert wirksam die Agglutination von Plättchen und ist als Antiplättchenund Antithrombosemittel geeignet. Insbesondere ist es zur Behandlung und/oder Vorbeugung zerebrovaskulärer Krankheiten, wie transiente ischämische Attacke (TIA), zerebraler Infarkt (Thrombus und Embolus) und zerebrale Arteriesklerosis; postopertiver Thrombus, Embolus und Blutflussstörungen, im Zusammenhang mit vaskulärer Operation und extrakorporaler Zirkulation; chronische arterielle Störungen, wie Bürger's Krankheit, obstruktive Arteriosklerosis, periphere Arteriosklerosis, SLE- und Raynaud's Krankheit; und ischämische, kardiale Krankheiten, wie Stenocardia und myokardialer Infarkt, geeignet. Weiterhin besteht eine Eignung zur Verhinderung des Wiederauftretens dieser Krankheiten und zur Verbesserung der Prognose hierfür.
- Die Wirkung des erfindingsgemässen Produktes wird durch die im folgenden gegebenen pharmakologischen Tests hinsichtlich des Diphenylethylenderivates unterstützt.
- Blut wurde aus einer menschlichen, kubitalen Vene auf solch eine Weise gesammelt, dass man eine 3,8 %-ige Lösung an Natriumcitrat in einer Menge von 1,10 mal dem Volumen des Blutes erhielt. Anschliessend wurde das plättchenreiche Plasma (PRP) hieraus gemäss dem Verfahren von Packham et al (cf. Packham, M.A., et al, J. Exp. Med. 126, 171-18g (1967)) hergestellt. Zu 0,2 ml des erhaltenen PRP wurden 25 ul Teile von Lösungen jeder Verbindung der vorliegenden Erfindung (A) bis (C) in verschiedenen Konzentrationen hinzugegeben und bei 37ºC 3 Minuten lang inkubiert. Anschliessend wurde die Agglutination der Plättchen mit Arachidonsäure, Kollagen, ADP und PAF eingeleitet. Die Agglutination der Plättchen wurde gemäss dem Verfahren von Mustard et al (cf. Mustard, J.F., et al, J. Lab. Clin. Med. 54, 548-559 (1964)) unter Einsatz eines von Schenko oder Niko Bioscience Co. verfügbaren Aggregometers bewertet. In anderen Worten, dieser Test wurde ausgeführt, um die Wirkung hinsichtlich der Plättchenaggregation (in vitro) zu bestimmen.
- Tabelle 1 zeigt die erhaltenen Ergebnisse. - Die Verbindungen (A) bis (C), wie sie oben gezeigt sind, entspechenden den Zielverbindungen, die in den Beispielen 1 bis 3 erhalten wurden.
- Die Verbindungen (A) bis (C), die typische Beispiele der Verbindung der vorliegenden Erfindung darstellen, wurden oral Meerschweinchen verabreicht. Nach 2 Stunden wurde das Blut eines jeden Tieres aus der abdominalen Aorta unter Ethernarkose gesammelt. Anschliessend wurde die Wirkung jeder Verbindung hinsichtlich der Agglutination der Plättchen, induziert durch Kollagen (3 ug/ml) und Arachidonsäure (50/uM), bestimmt. Tabelle 2 zeigt die 50 %-Wirksamkeitsdosen, bestimmt aus den Lösungsmittel- Verabreichungsverhältnissen. In anderen Worten, dieser Test wurde durchgeführt, um die Wirkung hinsichtlich der Plättchenaggregation (ex vivo) zu prüfen. - Die akute Toxizität der Verbindungen (A) bis (C), die typische Beispiele der Verbindung der vorliegenden Erfindung darstellen, wurde geprüft, indem man diese Verbindung männlichen Wistar-Ratten mit 300 bis 400 g Körpergewicht verabreichte. Als Ergebnis wurde gefunden, dass der LD&sub5;&sub0;-Wert jeder Verbindung höher als 500 mg/kg war.
- Wenn die Verbindung der vorliegenden Erfindung als Antiplättchenoder antithrombotisches Mittel verwendet wird, dann kann sie oral oder parenteral, z.B. intramuskulär, subkutan oder intravenös, verabreicht werden. Die Dosis hierfür kann z.B. in Abhängigkeit von der Krankheit, dem Zustand und dem Alter des Patienten variieren. Wenn keine besondere Begrenzung besteht, dann kann sie in einer Dosis von 0,1 bis 300 mg, vorzugsweise von 0,1 bis 60 mg, und insbesondere bevorzugt von 0,3 bis 30 mg, und ganz besonders bevorzugt von 0,6 bis 10 mg, einer Person pro Tag verabreicht werden.
- Die Verbindung der vorliegenden Erfindung kann in z.B. Tabletten, Granulen, Pulver, Kapseln, Injektionen oder Zäpfchen auf übliche Weise formuliert werden.
- Wenn sie in feste Zubereitungen für die orale Verabreichung formuliert werden soll, dann werden Trägerstoffe und, falls gewünscht, andere Additive, wie Bindemittel, Trennmittel, Schmiermittel, Farbmittel und Korrigentien, zu dem Grundstoff hinzugegeben und die erhaltene Mischung wird dann z.B. zu Tabletten, beschichteten Tabletten, Granulen, Pulver oder Kapseln auf übliche Weise formuliert.
- Beispiele für die Trägerstoffe sind Lactose, Maisstärke, weisser Zucker, Glucose, Sorbit und kristalline Zellulose. Beispiele für das Bindemittel sind Polyvinylalkohol, Polyvinylether, Ethylzellulose, Methylzellulose, Gummiarabikum, Tragacanth, Gelatine, Shellack, Hydroxypropylzellulose, Hydroxypropylstärke und Polyvinylpyrrolidon. Beispiele des Trennmittels sind Stärke, Agar, pulverförmige Gelatine, kristalline Zellulose, Kalziumcarbonat, Kalziumhydrogencarbonat, Kalziumcitrat, Dextrin und Pectin. Beispiele für die Schmiermittel sind Magnesiumstearat, Talk, Polyethylenglykol, Silica, und gehärtete pflanzliche Öle. Beispiele für die Färbemittel sind solche, die sich als Additive für Arzneimittel als geeignet erwiesen haben. Beispiele für Korrigentien sind Kakaopulver, Menthol, aromatische Säuren, Pfefferminzöl, Borneokampfer und Cinnamonpulver. Diese Tabletten und Granulen können selbstverständlich mit z.B. Zucker oder Gelatine, falls notwendig, beschichtet sein.
- Wenn eine Injektion zubereitet werden soll, dann werden verschiedene Additive, wie pH-Einstellmittel, Puffer, Stabilisatoren und Konservierungsmittel zu dem Grundstoff hinzugegeben und die erhaltene Mischung wird zu einer Injektion für die subkutane, intramuskuläre oder intravenöse Verabreichung formuliert.
- Die Erfindung wird mit Hilfe der folgenden Beispiele, die nicht begrenzend sind, weiter verdeutlicht.
- 2,42 g (0,01 M) 4,4'-Dimethoxybenzophenon, 1 g Zink und 2,1 g Trimethylborat wurden in 15 ml Tetrahydrofuran suspendiert. Zu der erhaltenen Suspension wurden 2,2 g Ethyl-4-Bromo-4-cyanobutyrat und eine katalytische Menge an Jod hinzugegeben, und die Mischung liess man bei Raumtemperatur 5 Stunden lang reagieren. Nach Beendigung der Reaktion wurden 10 ml einer gesättigten wässrigen Lösung von Ammoniumchlorid hinzugegeben und die Mischung wurde 1 Stunde lang gerührt. Nach Abfiltrieren des Zinks wurde das Filtrat mit Ethylacetat extrahiert. Das erhaltene Rohprodukt wurde mittels Silicagel-Chromatografie gereinigt, wobei man 1,5 g Kristalle erhielt. Die Kristalle wurden in 10 ml Benzol gelöst und 1 ml Thionylchlorid wurde zu der erhaltenen Lösung hinzugegeben. Nach Rühren bei Raumtemperatur während 1 Stunde wurde die Reaktionsmischung im Vakuum eingeengt und in Eis/Wasser dispergiert. Anschliessend wurde sie mit Benzol extrahiert, mit Wasser gewaschen und eingeengt. Hierbei wurden 1,3 g der Titelverbindung in Form eines farblosen, öligen Produktes erhalten.
- NMR (CDCl&sub3;) delta: 6,7-7,3 (8H), 4,1 (2H), 3,8 (6H), 2,7 (4H) und 1,3 (3H).
- 3,6 g Ethyl-4-cyano-5,5-bis(4-methoxyphenyl)-4-pentenoat wurden in 10 ml Dioxan gelöst und es wuden 3 ml einer 5 N wässrigen Lösung von NaOH hierzu hinzugegeben. Die erhaltene Mischung wurde bei 60ºC 5 Stunden lang gerührt. Nach Beendigung der Reaktion wurde die Reaktionsmischung angesäuert und mit Ethylacetat extrahiert. Hierbei wurden 3,2 g der Titelverbindung mit den folgenden physikalisch-chemischen Eigenschaften erhalten. Das Produkt konnte weiterhin durch Umkristallisieren aus Ethylacetat/Hexan gereinigt werden.
- Schmelzpunkt: l24-125ºC
- NMR (CDCl&sub3;) delta: 9,5 (1H), 6,8-7,4 (8H), 3,8 (6H), 2,7 (4H).
- Das Verfahren von Beispiel 1 wurde wiederholt, mit der Ausnahme, dass das Ethyl-4-bromo-4-cyanobutyrat durch 2,2 g Methyl-5-bromo-5-cyano- pentanoat ersetzt wurde. Dabei wurde die Titelverbindung mit den folgenden physikalisch-chemischen Eigenschaften erhalten.
- NMR (CDCl&sub3;) delta: 6,7-7,3 (8H), 3,8 (6H), 3,6 (3H) und 1,8-2,6 (6H).
- Beispiel 3 wurde wiederholt, mit der Ausnahme, dass 4,4'-Diethoxy- benzophenon und Tetrahydrofuran als Lösungsmittel verwendet wurden. Die Produktmischung wurde mit Thionylchlorid entwässert und auf die gleiche Weise wie in Beispiel 2 hydrolysiert.
- Beispiel 1 wurde wiederholt, mit der Ausnahme, dass 4-Methoxy-4'- methoxymethoxybenzophenon eingesetzt wurde. Die Produktmischung wurde mit Salzsäure behandelt und auf die gleiche Weise wie in Beispiel 2 hydrolysiert. Die Elution der Produktmischung mit einer Mischung aus Methanol und Chloroform bei einem Verhältnis von 5:95 bei der Chromatografie mit Silicagel lieferte die (Z)-Verbindung. Die Elution mit der obigen Mischung bei einem Verhältnis von 10:90 lieferte die (E)-Verbindung.
- Beispiel 5 wurde wiederholt, mit der Ausnahme, dass 4-Ethoxy-4'- methoxymethoxybenzophenon eingesetzt wurde.
- Beispiel 5 wurde wiederholt, mit der Ausnahme, dass 4,4'-Dimethoxy- methoxybenzophenon und 4,4'-Dimethoxybenzophenon verwendet wurden. Das Hydrolyseprodukt stellte die obige Zielverbindung dar.
- 260 mg des in Beispiel 3 erhaltenen Esters wurden auf die gleiche Weise wie in Beispiel 2 behandelt, wobei man 250 mg der obigen Zielverbindung erhielt.
- Die Daten der NMR-Analyse der in den Beispielen 4 bis 9 erhaltenen Verbindungen sind im folgenden aufgeführt. Die Analyse wurde mit CDCl&sub3;, delta, durchgeführt. Beispiel: Analyse:
Claims (9)
1. Diphenylethylenderivat der Formel (I)
wobei R&sub1; und R&sub2; unabhängig voneinander Wasserstoff, Hydroxyl oder
eine Niedrigalkoxygruppe sind, X eine Cyanogruppe darstellt, Y eine
Gruppe -R¹&sup0;-COOR³ bedeutet, wobei R³ Wasserstoff oder eine
Niedrigalkylgruppe ist, und wobei R¹&sup0; eine Alkylengruppe mit 1 bis 3
Kohlenstoffatomen, geradkettig oder verzweigt, ist, oder ein pharmazeutisch
verträgliches Salz hiervon.
2. Diphenylethylenderivat nach Anspruch 1, wobei sowohl R¹ als
auch R² Niedrigalkoxygruppen sind oder ein pharmazeutisch verträgliches
Salz hiervon.
3. Diphenylethylenderivat nach Anspruch 2, wobei sowohl R¹ und
R² Methoxygruppen sind oder ein pharmazeutisch verträgliches Salz
hiervon.
4. Diphenylethylenderivat nach Anspruch 1, das 4-Cyano-5,5-bis(4-
methoxyphenyl)-4-pentensäure oder ein pharmazeutisch verträgliches Salz
hiervon ist.
5. Diphenylethylenderivat nach Anspruch 1, das Methyl-5-cyano-6-6-
bis(4-methoxyphenyl)-5-hexenoat oder ein pharmazeutisch verträgliches
Salz hiervon ist.
6. Diphenylethylenderivat nach Anspruch l, das Ethyl-4-cyano-5,5-
bis(4-methoxyphenyl)-4-pentenoat oder ein pharmazeutisch verträgliches
Salz hiervon ist.
7. Verfahren zur Herstellung eines Diphenylethylenderivates der
Formel (I)
wobei R¹ und R² unabhängig voneinander Wasserstoff, Hydroxyl oder
eine Niedrigalkoxygruppe sind, X eine Cyanogruppe ist, Y eine Gruppe
-R¹&sup0;-COOR³ darstellt, wobei R³ Wasserstoff oder eine
Niedrigalkylgruppe ist, und wobei R¹&sup0; eine Alkylengruppe mit 1 bis 3
Kohlenstoffatomen, geradkettig oder verzweigt, ist, oder ein pharmazeutisch
verträgliches Salz hiervon, das die Reaktion eines Ketons der allgemeinen Formel
(VI)
wobei R¹ und R² wie oben definiert sind, mit einem Halogenid der
allgemeinen Formel (VII)
wobei Hal ein Halogenatom darstellt und R³ und R¹&sup0; wie oben definiert
sind, in Gegenwart von Zink in einer Reformatsky-Reaktion unter Bildung
einer Hydroxyverbindung der Formel (VIII)
wobei R¹, R², R³ und R¹&sup0; wie oben definiert sind, und die
anschliessende Entwässerung der so erhaltenen Hydroxyverbindung (VIII) auf
eine übliche Weise, und, falls notwendig, die Umwandlung des Produktes in
ein pharmazeutisch verträgliches Salz umfasst.
8. Pharmazeutische Zusammensetzung, die eine pharmakologisch
wirksame Menge des Diphenylethylenderivates gemäss einem der Ansprüche 1 bis
6 oder eines pharmakologisch verträglichen Salzes hiervon und einen
pharmakologisch verträglichen Träger umfasst.
9. Verwendung der Verbindungen gemäss einem der Ansprüche 1 bis 6
zur Herstellung eines Arzneimittels zur Behandlung von durch
Blutflussstörungen verursachten Krankheiten.
