Fluoralkylphenyl-cycloamidine
Verfahren zu ihrer Herstellung und ihre Verwendung als Ekto-
parasiticide.
Die vorliegende Erfindung betrifft neue Fluoralkylphenylcycloamidine,
die eine Wirksamkeit gegen Ektoparasiten aufweisen. Verfahren zu ihrer Herstellung und ihre Verwendung als
Ektoparasitizide.
Es wurde gefunden, dass Fluoralkylphenyl-cycloamidine der Formel

Difluormethyl, R' Halogen wie Fluor, Chlor, Brom, Alkyl oder
Alkenyl mit jeweils 1-4 C-Atomen und R'' Alkyl oder Alkenyl mit 1 - 6 .C-Atomen bedeuten, zur
Bekämpfung tierischer Ektoparasiten aus der Klasse der Acariden hervorragend eignen.

Es sind bereits cyclische und nicht cyclische Phenylamidine
bekannt. Bisher wurde lediglich bei nicht cyclischen Phenylamidinen eine Zeckenwirksamkeit aufgefunden. Eine
dieser Verbindungen (I, Südafrikanische Patentschrift 66/4135) wurde wegen ihrer Instabilität zurückgezogen.
Von cyclischen Amidinen, von denen die aus der U.S.A. Patentschrift 3 189 698 bekannte Verbindung II, die den
erfindungsgemässen Wirkstoffen in der Konstitution am nächsten kommt, genannt sei, wurde bisher keine ektoparasitizide
Aktivität bekannt.

Durch die Einführung einer Trifluor- oder Difluormethylgruppe in die Phenylcycloamidine wurde nun überraschenderweise eine
viel stärkere Wirkung gegen Zecken, insbesondere gegen Zeckenstämme, die gegen Phosphorsäure resistent geworden sind, aufgefunden.
Die erfindungsgemässen Verbindungen sind darüberhinaus bei der Anwendung im "Rinderdip" beständig.
Die Herstellung der erfindungsgemässen Halogenalkenylphenyl-cycloamidine
erfolgt dadurch, dass man Anilinderivate der Formel

Diese Umsetzungen können so vorgenommen werden, dass man .aus den Lactamen und dem Säurehalogenid zunächst den
reaktiven Lactimester, gegebenenfalls in Anwesenheit von Lösungsmitteln wie Benzol, Toluol oder Tetramethylensulfon
herstellt, dann das substituierte Anilin zutropft und gegebenenfalls das Reaktionsgemisch eine Zeitlang nacherhitzt.
Man kann aber auch so vergehen, dass man in ein Gemisch von Lactam und substituiertem Anilin gegebenenfalls in
Anwesenheit von Lösungsmittel das Säurechlorid, beispielsweise Phosphoroxychlorid eintropft und das Reaktionsgemisch
eine Zeitlang nacherhitzt.

Die erfindungsgemäßen Wirkstoffe besitzen basischen Charakter.
Sie können als freie Basen oder in Form ihrer Salze, beispielsweise
Hydrochloride, Sulfate, Phosphate, Nitrate, Acetate oder Naphthalin-disulfonate angewandt werden.
Die freien Basen wie die Salze weisen starke akarizide Eigenschaften
auf, besonders gegen Akariden, die als tierische Ektoparasiten domestizierte Tiere wie Rinder und Schafe
befallen. Sie eignen sich deshalb gut für die Bekämpfung von tierischen Ektoparasiten aus der Ordnung der Akaride.
Als wirtschaftlich wichtige Ektoparasiten dieser Art, die besonders in tropischen und subtropischen Ländern eine
große Rolle spielen, seien beispielsweise genannt: die australische und südamerikanische Rinderzecke Boophilus
microplus, die südafrikanische Rinderzecke Boophilus decoloratus, beide aus der Familie der Ixodidae.
Im Laufe der Zeit sind in verschiedenen Gebieten die genannten Zecken gegen die als Bekämpfungsmittel bisher verwendeten
Phosphorsäureester und Carbamate resistent geworden, so daß der Bekämpfungserfolg dort in Frage gestellt wird. Zur Sicherung
einer wirtschaftlichen Viehhaltung in den Befallsgebieten besteht daher ein Bedarf an Mitteln, mit denen Zecken, auch
resistenter Stämme, beispielsweise des Genus Boophilus sicher bekämpft werden können. In hohem Maße gegen die bisherigen
Phosphorsäureestermittel und Carbamate resistent sind beispielsweise in Australien der Ridgeland-Stamm und der Biarra-Stamm
von Boophilus microplus. Die erfindungsgemäßen Wirkstoffe sind sowohl gegen die normalempfindlichen als auch die
resistenten Stämme, zum Beispiel von Boophilus gleich gut wirksam. Sie wirken beispielsweise stark eiablagehemmend auf
die adulten Formen.

Die Applikation dieser Wirkstoffe erfolgt in üblicher
V/eise zum Beispiel durch Besprühen, Begießen, Vernebeln oder als Bad (Dip). Den Formulierungen oder den anwendungsfertigen
Lösungen können auch sonstige Hilf- oder Wirkstoffe, wie Desinfektionsmittel, zugemischt werden.
In vitro-Test auf eiablagehemmende Wirkung an Zecken.
3 Teile Wirkstoff werden mit 7 Teilen eines Gemisches aus gleichen Gewichtsteilen Äthylenglykolmonomethyläther und
Nonylphenolpolyglykoläther vermischt. Das so erhaltene Emulsionskonzentrat wird mit Wasser auf die jeweils gewünschte
Anwendungskonzentration verdünnt.
In diese Wirkstoffzubereitung werden adulte vollgesogene
Zeckenweibchen der Art Boophilus microplus (resistent) eine Minute lang getaucht. Nach dem Tauchen von je 10
weiblichen Exemplaren der verschiedenen Zeckenstämme überführt man die einzelnen Zecken in Kunststoffschalen, deren
Boden mit einer Pilterpapierscheibe belegt ist. Nach gen wird die Wirksamkeit der Wirkstoffzubereitung bestimmt durch Ermittlung der Hemmung der Ablage von fertilen
Eiern gegenüber der Eiablage von unbehandelten Kontrollzecken. Die Wirkung wird in # angegeben, wobei
100 fo bedeutet, daß keine fertilen Eier mehr abgelegt
wurden und 0 56 bedeutet, daß die Zecken in normaler Weise wie die unbehandelten Kontrollzecken
Eier abgelegt haben.

Zu einer Lösung von I65 g (1,0 Mol) 4-Chlor-3-difluormethyianilin
und 155,3 g (1,0 Mol) N-Butyl-pyrrolidon in 1000 ml
Toluol tropft man bei 200C 154 g (1,0 Mol) Phosphoroxychlorid,
rührt 90 Minuten bei Raumtemperatur und erhitzt 4 Stunden unter Rückfluss bis die HCl-Abspaltung beendet ist.
Nach dem Abkühlen dekantiert man die Toluollösung ab, nimmt
den Rückstand in Wasser auf, macht ihn mit Natronlauge alkalisch und schüttelt das Reaktionsprodukt mit einem Gemisch
von Chloroform und Aether aus. Nach dem Abdampfen des Lösungsmittels und Destillation unter vermindertem Druck erhält man
166 g l-Butyl-2-(4-chlor-3-difluormethyl-phenyl)iminopyrrolidin vom Kp.Q 2 154-158°.
Bei entsprechender Arbeitsweise werden folgende Phenylcycloamidine

Zu einer Lösung von 16,5 g 4-Chlor-3-difluormethylanilin und
15,5 g N-Buty!-pyrrolidon in 25 ml Toluol tropft man eine
Lösung von 9,9 g Phosgen in 75 ml Toluol, rührt 90 Minuten bei 20° und erhitzt dann 4 Stunden unter Rückfluss. Nach der
in Beispiel 1 beschriebenen Aufarbeitung erhält man 13*2 g l-Butyl-2-(4-chlor-3-difluormethyl-phenyl)-imino pyrrolidin
vom kP Q ρ 154-158°.
F Eine Lösung von 31,4 g N-Butyl-thiopyrrolidon und 40 g
4-Chlor-3-difluormethylanilin in 400 ml Aethanol wird unter Zusatz von 75 g Quecksilberoxid 8 Stunden bei 0° und dann 15
Stunden bei £J0° kräftig gerührt. Man saugt vom Niederschlag
ab, destilliert den Rückstand in Vakuum und erhält 12,4 g l-Butyl-2-(4-chlor-3-difluormethyl-phenyl)-imino pyrrolidin
vom kg 0 2 154-158°.
Zu 24,5 g (0,1 Mol) 4-Chlor-3-difluormethyliminopyrrolidin gelöst in 100 ml Tetrahydrofuran tropft man bei 20° 8,5 g
(0,11 Mol) Allylchlorid und erhitzt anschliessend über Nacht unter Rückfluss. Man saugt vom ausgefallenen Niederschlag ab.
Das Filtrat wird mit v/asser mehreremale gewaschen. Nach dem
Abtrennen der organischen Phase dampft man ein und erhält nach Destillation bei kp n o 130-160° ein Gemisch der Formeln

aus dem nach Kolonnendestillation das 1-Butyl-2-(4-chlor-3-difluormethyl-phenyl)-iminopyrroli'din,
kpn n(- 146 - 147
rein erhalten werden konnte. Ausbeute 6,8 g.
18,5 g 4-Chlor-3-trifluormethylanilin und 21,5 g N-Butylpyrrolidon-diäthylacetal
(hergestellt aus Triäthyloxoniumfluoroborat und Butyl-pyrrolidon) werden auf 110-140 erhitzt,
wobei innerhalb von 30 Minuten der bei der Reaktion frei gewordene Alkohol abdestilliert. Der Rückstand ergibt
nach Destillation in Vakuum 21,3 g 1-Butyl-2-(4-chlor-3-trifluormethylphenyl)-imino-pyrrolidin,
kp „ „, 150 - 155°.
50 ml n-Butylcaprolactam und 25 ml 4-Chlor-3-trifluorinethylisocyanat
werden solange unter Rückfluss erhitzt, bis die COp-Entwicklung beendet ist. Man destilliert das
Reaktionsprodukt im Vakuum und erhält 12,5 g 1-Butyl-2-(4-chlor-3-trifluormethyl-phenyl)-inino-azacycloheptan,
kp 0 2 172-176°.
Zu einer Lösung von 37 g (0,2 Mol) 4-Chlor~3-trifluoranilin
in 350 ml Benzol fügt nan 31,4 g (0,2 Mol) Jf-Chlorbuttersäurechlorid,
erhitzt 2 Stunden unter Rückfluss, kühlt ab, fügt 48,7 g Phosphorpentachlorid hinzu und erhitzt allmählich
auf Rückfluss. Nach Beendigung der Chlorwasserstoffentwicklung destilliert man das gebildete Phosphoroxychlorid
in Vakuum ab und erhält das 1 ,4-Dichlor-1-(4-chlor-3-trif'luorphenyliinino)-butan
als schwer flüchtiges Öl. Dieses nimmt man in 100 ml Benzol auf, tropft es bei 20° zu 33 g (0,45 Hol)
Butylainin in 200 ml Benzol

und erhitzt 2 Stunden unter Rückfluss. Man giesst in Eiswasser,
macht mit Natronlauge alkalisch und arbeitet analag Beispiel 1 auf. Man erhält 43 g l-Butyl-2-(4-chlor-3-trifluorme
thy !phenyl )imino-pyrrolidin, kp ß QC- 150-155°.

Claims (8)
- Patentansprüche;uormethyl-phenyl-cycloamidine der FormelH"in der η 3, 4 oder 5, in null, 1 oder 2 ist; R Trifluormethyl oder Bifluormethyl, R1 Halogen, vorzugsweise Chlor, Fluor oder Brom, ferner Alkyl oder Alkenyl mit jeweils 1 - 4 C-Ato men und R" Alkyl oder Alkenyl mit 1 - 6 C-Atomen bedeutet.
- 2) Verfahren zur Herstellung von Fluoralkyl-phenyl-cycloamidinen und deren Salzen, dadurch gekennzeichnet, daß mana) Anilinderivate der Formelin der R, R? und ui die in Anspruch 1 angegebene Bedeutung besitzen, mit Lactamen der FormelCH2 )nR" -NLe A 12 803 - 21 -in der R" und η die in Anspruch 1 angegebene Bedeutung besitzen und W Sauerstoff oder Schv/efel bedeutet, bzw. deren Salzen oder reaktionsfähigen Derivaten, gegebenenfalls in Anwesenheit von Kondensationsm.itteln umsetztoder daß man b) cyclische Amidine der Formelin der R, R1 und m die in Anspruch 1 angegebene Bedeutung besitzt, mit alkylierenden Mitteln der FormelB - R"in der R die in Anspruch 1 angegebene Bedeutung besitzt und B eine reaktive Estergruppe, Halogen, Arylsulfonyloxy oder Alkylsulfonyloxy, zur Reaktion bringtoder daß man c) substituierte Aniline der Formel(R' )min der R, R1 und m die in Anspruch 1 angegebene Bedeutung besitzen, mit ^-Halogenalkansäurechloriden acyliert, die Acylierungsprodukte mit Phosphorhalogeniden in die entsprechenden Imidhalogenide der FormelLe A 12 803 - 22 -109837/16832UÜ9019 53Ί? V N=C- ( CH, ) „-Hal Clüberführt und diese mit Aminen der FormelR"-IHi2in der R" die in Anspruch 1 angegebene Bedeutung besitzt, umsetztdaß man d) Arylisocyanate der allgemeinen Formelbzw. - 3) Verfahren nach Anspruch 2, dadurch gekennzeichnet,
daß man Thiolactame der Formelin der R" die in Anspruch 1 angegebene Bedeutung besitzt, mit den substituierten Anilinen in Gegenwart von entschwefelnden Mitteln, wie HgO, AgpO oder Hg(GlOp umsetzt. - 4) Verfahren nach Anspruch 2, dadurch gekennzeichnet, daß man Acetale der FormelR" -N
Alkyl O ■ Ö Alkylin der R" die in Anspruch 1 angegebene Bedeutung besitzt, mit den substituierten Anilinen umsetzt. - 5) Ektoparasitizide Mittel,gekennzeichnet durch einen Gehalt an Fluoralkyl-phe:
gemäß Anspruch 1.* an Fluoralkyl-phenyl-cycloamidinen oder deren Salzen - 6) Verfahren zur 3ekämpfung von Ektoparasiten, dadurch gekennzeichnet, daß man Fluoralkyl-phenyl-cycloanidine oder deren Salze gemäß Anspruch 1 auf Ektoparasiten oder deren Lebensraum einwirken läßt.Le A 12 805 - 24 -109837/1683
- 7) Verwendung von Fluoralkyl-phenyl-cycloamidinen oder deren Salzen gemäß Anspruch 1 zur Bekämpfung von Ektoparasiten.
- 8) Verfahren zur Herstellung von ektoparasitiziden Mitteln, dadurch gekennzeichnet, daß man Fluoralkyl-phenyl-cycloamidine oder Salze gemäß Anspruch 1 mit Streckirtitteln und/oder oberflächenaktiven Mitteln mischt.Le A 12 80> - 25 -109837/1683
