本发明揭示了一种三白保湿美白凝胶制备方法及其用途,包括如下步骤:步骤一、制作白芷提取原液、白术提取原液、白茯苓提取原液;步骤二、制作2%的卡波姆940凝胶基质,然后分别取白芷提取原液、白术提取原液、白茯苓提取原液、丙三醇、0.5%海藻糖、0.5%海藻糖、5%羟苯乙酯甲醇液与水混合,再加入PBS,然后添加三乙醇胺调节pH至6.8,继续搅拌至晶莹剔透状,即得到三白凝胶。本发明通过白芷、白术和白茯苓进行原液提取,并确定其最合适的配比,有效提高了三白保湿美白凝胶对于DPPH自由基的清除率,还提高了抗紫外线性能和保湿率的化妆品或药品。

一种三白保湿美白凝胶制备方法及其用途
技术领域
本发明涉及护肤品加工技术领域,尤其涉及一种三白保湿美白凝胶制备方法及其用途。
背景技术
白芷为伞形科植物白芷的干燥根,白茯苓为多孔菌科真菌茯苓的干燥菌核,白术为白术为菊科白术的干燥根茎。白芷作为宫廷美白保湿秘方成分之一成为研究者的青睐,白芷、白术作为“七白散”的主要成分,常用于面肤黯斑和皮肤增白,现代医学研究证明,白芷挥发油中的不饱和的烯类化合物具抗氧化活性和有抑制酪氨酸酶活性ER;白茯苓无水乙醇提取液对酪氨酸酶的抑制率为91.7%,具有较好的防止黑色素沉积作用[3]。白术中的挥发油具有抑制黑色素的作用,可以预防色素在皮肤表面沉积,从而达到祛斑美白,美容养颜功效[5]
凝胶剂系指药物与凝胶的辅料制成半固体经皮制剂。与乳膏剂相比凝胶剂外观晶莹剔透,易于涂布、制法简单、原料易得、更是由于其具有经皮制剂的缓释功效,而受到研究者和患者的青睐。凝胶剂的应用范围还在不断拓宽,前景十分广阔。
发明内容
为解决上述问题,本发明公开了一种三白保湿美白凝胶制备方法及其用途,本发明通过白芷、白术和白茯苓进行原液提取,并确定其最合适的配比,有效提高了三白保湿美白凝胶对于DPPH自由基的清除率,还提高了抗紫外线性能和保湿率的化妆品或药品。
为实现上述目的,本发明的技术方案如下:
一种三白保湿美白凝胶制备方法,包括如下步骤:
步骤一、制作白芷提取原液、白术提取原液、白茯苓提取原液;
步骤二、制作2%的卡波姆940凝胶基质,然后分别取白芷提取原液、白术提取原液、白茯苓提取原液、丙三醇、0.5%海藻糖、0.5%海藻糖、5%羟苯乙酯甲醇液与水混合,再加入PBS,然后添加三乙醇胺调节pH至6.8,继续搅拌至晶莹剔透状,即得到三白凝胶。
进一步的改进,所述步骤二中,制作2%的卡波姆940凝胶基质50g,然后分别取白芷提取原液2-7ml、白术提取原液2-7ml、白茯苓提取原液2-7ml、丙三醇10mL、海藻糖4mL、0.5%海藻糖2.0mL、5%羟苯乙酯甲醇液与水混合至总重100g,再加入PBS 4ml,然后添加三乙醇胺调节pH至6.8,继续搅拌至晶莹剔透状,即得到三白凝胶。
进一步的改进,所述白芷提取原液加入量为5.0ml、白术提取原液5.0mL、白茯苓提取原液5.0mL。
进一步的改进,所述白芷提取原液的制备方法如下:
称取白芷饮片,切碎后至圆底烧瓶中,加水蒸馏3次,每次至无油溜出为止,取流出液,流出液用甲醇溶解后加水稀释成为0.01mL·mL-1的白芷原液,其中蒸馏时,加水量与白芷饮片的体积质量比为15:1ml/g。
进一步的改进,所述白术提取原液的制作方法如下:
分别称取白术粉末50g,加入75%乙醇500mL,超声20min,60℃~70℃回流提取0.5h,连续2次,趁热减压过滤,合并滤液,回收溶剂至无醇味,继续浓缩至75%乙醇体积的十分之一,即为质量浓度为1.0g·mL-1原液供试液,即得到白术提取原液,4℃保存备用;75%乙醇与白术粉末的制剂质量比10:1ml/g。
如权利要求1所述的三白保湿美白凝胶制备方法,其特征在于,所述白茯苓提取原液的制作方法如下:
分别称取白茯苓粉末50g,加入75%乙醇500mL,超声20min,60℃~70℃回流提取0.5h,连续2次,趁热减压过滤,合并滤液,回收溶剂至无醇味,继续浓缩至75%乙醇体积的十分之一,即为质量浓度为1.0g·mL-1原液供试液,即得到白术提取原液,4℃保存备用;75%乙醇与白术粉末的制剂质量比10:1ml/g。
一种三白保湿美白凝胶的用途,所述三白保湿美白凝胶通过权利要求1-6任一所述三白保湿美白凝胶制备方法制得;所述三白保湿美白凝胶用作提高DPPH自由基的清除率、提高抗紫外线性能和保湿率的化妆品或药品。
本发明的有益效果在于:
通过白芷、白术和白茯苓进行原液提取,并确定其最合适的配比,有效提高了三白保湿美白凝胶对于DPPH自由基的清除率,还提高了抗紫外线性能和保湿率的化妆品或药品。

具体实施方式
下面对本发明的具体实施例作进一步详细描述。
(一)活性物质的提取
①白芷提取原液的制备
采用水蒸气蒸馏法提取。称取白芷饮片50g,切碎后至圆底烧瓶中,加水750mL蒸馏3次,每次至无油溜出为止。取流出液0.5mL,甲醇溶解后加水50mL稀释成为0.01mL·mL-1的白芷原液。
②白术、白茯苓提取原液的制备
分别称取白术、白茯苓各粉末50g,加入75%乙醇500mL,超声(50HZ,100W)20min,60℃~70℃回流提取0.5h,连续2次,趁热减压过滤,合并滤液,回收溶剂至无醇味,继续浓缩至50mL,即为质量浓度为1.0g·mL-1原液供试液,4℃保存备用。
(二)“三白”保凝胶的制备
称取卡波姆940、水,自然溶胀8h以上,搅拌均匀至晶莹剔透状,即得到2%的卡波姆940凝胶基质50g。另取处方量的白芷提取原液、白术、白茯苓提取原液各适量,丙三醇10mL、0.5%海藻糖4mL、5%羟苯乙酯甲醇液2.0mL和适量的水搅拌均匀后与2%卡波姆940基质50g混合得到100g混合物,加PBS(pH6.8)4mL,逐滴添加三乙醇胺调pH6.8,继续搅拌至晶莹剔透状,即得到“三白”凝胶,常温保存备用。
(三)凝胶处方的优化
①单因素分析
初步选定A、B、C三个影响因素六水平,以综合指数值(对DPPH自由基的去除率的50%+对酪氨酸酶活性抑制率的50%)为考察指标,单因素分析确定因素的响应值。因素-水平表见表1,各因素对综合评价值影响结果见图1。

图1-图3分贝显示,白芷、白术、白茯苓提取液均在5.0mL达到满意综合效果,增加剂量并不增效,这说明已经达到白芷、白术、白茯苓活性成分的清除极限,从而提示,白芷、白术、白茯苓活性成分的抗氧化活性存在饱和性。
②正交设计实验

通过直观分析最优处方方案为C2A3B2,其主次影响因素的顺序为C>A>B;正交设计结果与直观分析相符,主体间效应检验差异显著(P<0.05)。白芷提取原液体积5.0mL、白术提取原液体积5.0mL白茯苓提取原液体积5.0mL时,为最优制备工艺参数。
③验证性实验
按照C2A3B2方案,即取处方量的白芷提取原液、白术、白茯苓提取原液各5.0mL,平行制备3份,综合指数结果见表5。

(五)“三白”保湿美白凝胶的功能性测定
样品供试液的配制
分别称取“三白”凝胶、市售凝胶各5g,高速离心(10000r·min-1)后吸出上清液5mL,蒸馏水稀释定容至100mL。
①对DPPH自由基的清除率测定
精密移取样品供试液2mL、浓度为20μg·mL-1二苯基苦基苯肼(DPPH)溶液2mL,先后加入同一具塞试管中摇匀,在室温暗室处静置30min后于517nm波长下测定样品的吸光度A。按以下公式(1)计算样品对DPPH自由基的清除率(K)

其中Ai为DPPH溶液和样品供试液混合液的吸光度,Aj为样品供试液的吸光度,Ac为DPPH溶液的吸光度,并作F经验。实验结果见表1。
②对酪氨酸酶抑制活性测定
按照步骤依次加入PBS(pH=6.8)、样品供试液、酪氨酸溶液(1.0mg·mL-1),35℃水浴10min后,加入1.0mL的酪氨酸酶溶液,摇匀,快速转移至比色管中,于475nm处测定吸光度A。

③抗紫外线性能测定
按照QB/T 2410-1998规定的防晒霜UVB区防晒效果的评定方法[11]。分别称取“三白”保湿美白凝胶、市售芦荟美白祛斑保湿凝胶各2g均匀涂抹在3M医用透气胶带上,贴于比色皿内壁,以样品的医用透气胶带为参比,测定波长在在280~360nm波长范围内,每隔10nm扫描一次记录吸光度A,以吸光度平均值来判断样品在UVB区(280~320nm)的防晒效果[12]。结果见表8。

注:
表3显示,“三白”凝胶与市售凝胶抗紫外线性能存在显著性差异。可以用于中等紫外线照射的防护。
④保湿率测定
室温条件下,将2.0g凝胶直接均匀涂布在医用透气胶带(模仿真人皮肤)上,精确记录质量W0,然后将其置于相对湿度45%和80%的湿度干燥器内,分别于4、8、12和24h后,取出透气胶带,立即精确称取各样品质量,记录为Wi,按以下公式(3)计算保湿率

表4显示,8~24h内,在湿度45%和80%的环境下,“三白”凝胶与市售凝胶的保湿率无显著差异。
⑤性能表征考察
外观性状本品为白色凝胶,质地细腻均匀、晶莹剔透、粘稠度适宜、涂展性良好。pH值检查取本品供试液,pH值测定值为6.8。
冷热实验将本品封装于透明化妆品瓶中,分别置于4、25、50℃下,于7、30、180d取样观察,凝胶外观无分层、结块、霉变、变色现象。
离心实验将本品封装于离心试管中,2000r·min-1,于5、10、15min取样观察,凝胶外观无分层、结块现象。
强光照射实验将本品敞开口放在装有照度为4500Lx的日光灯的光橱内放置15天,第7、15天取样检测,凝胶的透明度、酸碱度、粘稠度均无变化。
留样观察将样品装入包装盒内加盖密闭,于室温下放置1个月,按质量标准检测,结果无明显变化。
最后应当说明的是,以上实施例仅用以说明本发明的技术方案,而非对本发明保护范围的限制,尽管参照较佳实施例对本发明作了详细地说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的实质和范围,都应涵盖在本发明的保护范围之内。因此,本发明的保护范围应该以权利要求所界定的保护范围为准。

1.一种三白保湿美白凝胶制备方法,其特征在于,包括如下步骤:
步骤一、制作白芷提取原液、白术提取原液、白茯苓提取原液;
步骤二、制作2%的卡波姆940凝胶基质,然后分别取白芷提取原液、白术提取原液、白茯苓提取原液、丙三醇、0.5%海藻糖、0.5%海藻糖、5%羟苯乙酯甲醇液与水混合,再加入PBS,然后添加三乙醇胺调节pH至6.8,继续搅拌至晶莹剔透状,即得到三白凝胶。
2.如权利要求1所述的三白保湿美白凝胶制备方法,其特征在于,所述步骤二中,制作2%的卡波姆940凝胶基质50g,然后分别取白芷提取原液2-7ml、白术提取原液2-7ml、白茯苓提取原液2-7ml、丙三醇10mL、海藻糖4mL、0.5%海藻糖2.0mL、5%羟苯乙酯甲醇液与水混合至总重100g,再加入PBS 4ml,然后添加三乙醇胺调节pH至6.8,继续搅拌至晶莹剔透状,即得到三白凝胶。

7.一种三白保湿美白凝胶的用途,其特征在于,所述三白保湿美白凝胶通过权利要求1-6任一所述三白保湿美白凝胶制备方法制得;所述三白保湿美白凝胶用作提高DPPH自由基的清除率、提高抗紫外线性能和保湿率的化妆品或药品。
