技术领域
本发明属于食品技术领域,具体涉及一种促进儿童身高发育的营养组合物。
背景技术
白春利在中国专利201811388303.X中公布了一种帮助儿童身高发育的配方奶粉及生产方法,包含生牛乳或全脂奶粉、脱盐乳清粉、脱脂奶粉、固体玉米糖浆(或麦芽糊精)、乳清蛋白粉、结晶果糖、植物油、低聚果糖、无水奶油、低聚半乳糖、二十二碳六烯酸、牛初乳粉、乳矿物盐、水解蛋黄粉、乳铁蛋白、磷脂、维生素A、维生素D、维生素E、维生素K、维生素B1、维生素B2、维生素B6、维生素B12、维生素C、镁、钙、铁、锌、硒等成分。该专利营养配比均衡,同时强化了某些营养素(如水解蛋黄粉、钙、锌等)以帮助儿童身高发育。然而,影响儿童身高发育的因素有很多,除了保证科学的营养供应之外,生长激素本身也对儿童身高发育起主导作用,现有的儿童奶粉中均未添加生长激素促进剂和慢波睡眠促进剂,主要是依靠营养强化剂强壮骨骼和优质蛋白质帮助生长发育,本发明为一种营养组合物,可以添加至配方奶粉中,在满足儿童营养所需的同时,促进生长激素分泌。多维度帮助儿童身高发育。

下面对本发明作进一步说明:
本发明所述营养组合物中,生长激素促进剂可直接促进生长激素的分泌,从而引起骨骼生长。由于生长激素分泌旺盛的时期是在慢波睡眠阶段,慢波睡眠促进剂可延长慢波睡眠时间,间接促进生长激素的分泌。骨稳态调节剂可提高钙的吸收率,抑制骨吸收,增强骨密度。其中膳食丁酸可以有多种提供方式:①丁酸②丁酸盐(丁酸钠、丁酸钾、丁酸钙或丁酸镁)③包含丁酸的烷基,和/或甘油酯④丁酸-环糊精络合物。研究表明,膳食丁酸可以抑制破骨细胞生成,降低骨吸收,防止骨质流失,增加骨形成。益生元和菊粉属于内源性可产生内源性丁酸的组分,可刺激肠道中产生丁酸和其他短链脂肪酸,从而酸化肠腔,增加矿物质在肠道中的溶解度,吸收表面的扩大,增加长细胞的钙摄取。由于普通调制乳粉中某些微量元素和维生素含量不足,添加营养强化剂可以均衡调制乳粉的营养,提升其营养价值。
本发明所述的营养组合物从帮助生长激素分泌、调节骨稳态方面促进身高发育,与现有促进骨骼发育的营养组合物或奶粉相比,不仅含有骨稳态调节剂帮助提高钙的生物利用率、抑制骨吸收、增强骨密度,还包含生长激素促进剂和慢波睡眠促进剂,这两组成分都可以促使生物体自发的分泌生长激素,从而使骨头两端的骺软骨不断***出软骨细胞,这些软骨细胞通过钙化沉积下来形成骨头的生长。
总之,本发明所述促进儿童身高发育的营养组合物,可以添加至配方奶粉中,在满足儿童营养所需的同时,促进生长激素分泌,多维度帮助儿童身高发育。

各受试组组分配比(重量份)(表1):
空白组(普通调制羊乳粉):脱脂羊奶粉43,全脂羊奶粉5,浓缩羊乳清蛋白粉6,乳糖22,植物油14,葡萄糖浆固体1。
对照组1:脱脂羊奶粉43,全脂羊奶粉5,浓缩羊乳清蛋白粉6,乳糖22,植物油14,葡萄糖浆固体1,生长激素促进剂1.8。
对照组2:脱脂羊奶粉43,全脂羊奶粉5,浓缩羊乳清蛋白粉6,乳糖22,植物油14,葡萄糖浆固体1,慢波睡眠促进剂0.7。
对照组3:脱脂羊奶粉43,全脂羊奶粉5,浓缩羊乳清蛋白粉6,乳糖22,植物油14,葡萄糖浆固体1,骨稳态调节剂7。

制备工艺:
湿法工艺
(1)羊乳标准化:
生羊乳预杀菌:原料生羊乳验收合格后,进行巴氏杀菌,80~85℃,15~30s。
羊乳标准化:将预定重量配比的羊乳、脱脂羊奶粉、浓缩羊乳清蛋白粉、植物油、乳糖、葡萄糖浆混合均匀,得到混合溶液,放入暂存罐中暂存;
(2)高压均质:在高压均质机内进行均质,均质压力为12-15Mpa;
(3)杀菌:92-96℃,1min;
(4)浓缩:蒸发浓缩;
(5)杀菌:二次杀菌,72-75℃,1min;
(6)均质:二次均质混合,均质压力为12-15Mpa;
(7)干燥:喷雾干燥得到普通调制羊乳粉。
干法工艺
(1)干混:普通调制羊乳粉中添加该营养组合物;
(2)抽真空,充入惰性气体和氮气,包装即成。
实验方法:本研究采用雄性C57BL/6小鼠,6周龄,100只,适应性喂养一周后,随机分为3组,每组10只,按照下表饲喂各组小鼠,饲喂剂量每日5g/只,饲喂5周,进行相关指标检测。

试验指标:
1)生长期小鼠体长:麻醉空白组,对照组,实验组的小鼠,用直尺测量其体长,从头部最顶端到小鼠臀围交接处即为体长,精确到1mm。
2)骨吸收降低程度(CTX-I标记物检测):腹腔注射麻醉小鼠,心脏穿刺吸取适量血液注入1.5mlEP管中。4℃离心机3000r/min,20min离心2次,每次取上清液,得到血清标本,按照ELISA试剂盒说明书检测破骨标志物。
3)血清中的生长激素:各组小鼠测定体长和骨吸收标记物后,在麻醉状态下,利用眼球取血法取小鼠血液,于室温下待血液自然凝固20min,以3000r/min离心20min,收集上清液,采用ELISA试剂盒测定GH水平。
4)小鼠骨密度:用脊椎脱位法迅速将小鼠处死,剥离小鼠双侧股骨,注意保持股骨头完整性,剔除其肉、筋膜等组织后,用浸有生理盐水的纱布包裹,于-20℃冰箱中保存,用于检测股骨骨密度。骨密度检测用双能X射线骨密度仪检测。
5)血钙浓度:对小鼠利用眼球取血法取血后,血液样品用2ml的离心管进行收集,4℃下4000r/min离心15min后,获得的血清与-25℃冰箱内保存,血清中血钙的测定参照试剂盒说明进行操作。
实验结果(图1至图5):
图1表明,添加了9%营养组合物的普通调制羊乳粉(实验组1)和对照组、空白组相比,对于小鼠的体长增长具有明显的促进作用,实验组2(营养组合物添加量5%)对于体长增加效果不及大部分对照组的效果,说明低剂量的添加该营养组合物不能产生良好的效果,实验组3(营养组合物添加量12%)与实验组1(营养组合物添加量9%)相比,小鼠体长增长的效果没有更进一步的增加,这可能是营养过量导致小鼠产生不耐受反应所致。
综上所述,该营养组合物各组分之间在促进小鼠体长增长方面有协同作用,且添加量在推荐范围内,才能达到理想效果。
骨吸收(Boneresorption)是指在较低的应力水平下,骨组织的体积和密度逐渐发生下降的生理行为。骨吸收是构成骨重建的重要功能,体现了骨组织对力学环境的适应能力。异常的骨重建现象可能会导致骨质疏松症状,从而成为骨折的诱因。骨转换生化标志物不仅可以反映骨吸收与骨形成的动态平衡状况,而且早于骨密度的改变,对骨质疏松症的早期诊断、代谢性骨病的鉴别诊断、抗骨质疏松药物疗效判定有重要价值。
Ⅰ型胶原末端肽是Ⅰ型胶原末端交叉连接的非螺旋区域肽链,是破骨细胞在骨吸收过程中降解产生的特异性产物,其在血液中以氨基末端肽(NTX)/羧基末端肽(CTX)形式存在,能直观的反映骨吸收情况。
图2表明,含有骨稳态调节剂的奶粉都具有抑制骨吸收的作用,但同时含有骨稳态调节剂和营养强化剂的奶粉抑制骨吸收作用更显著(实验组1,实验组2,实验组3)。这可能是由于骨稳态调节剂与营养强化剂中的钙和维生素D(D-泛酸钙),维生素K1(植物甲萘醌)产生了协同作用,共同抑制骨吸收。实验组2(营养组合物添加量5%)的抑制骨吸收作用逊于实验组1(营养组合物添加量9%)和实验组3(营养组合物添加量12%),说明营养组合物剂量未达到效果,而实验组3的效果并未优于实验组1,说明继续增大剂量不能显著提升效果,故没有实际意义。
图3表明,所有对照组和实验组的小鼠与空白组相比,体内的生长激素有增加,说明在影响小鼠生长激素方面,添加生长激素促进剂、慢波睡眠促进剂、骨稳态调节剂、营养强化剂中的任意一种或几种成分,都比不添加的效果好。
在对照组中添加了生长激素促进剂和慢波睡眠促进剂组别的小鼠(对照组1,对照组2,对照组5,对照组6)的生长激素要比不添加组别(对照组3,对照组4)高,说明生长激素促进剂和慢波睡眠促进剂对小鼠体内生长激素的分泌起重要作用。
实验组1(营养组合物添加量9%)的效果优于实验组1(营养组合物添加量5%)和实验组3(营养组合物添加量12%),说明营养组合物添加剂量不足不会产生理想效果,而添加剂量过大,也不会显著提升促进效果,反而有所下降,这可能是由于大剂量的营养物导致小鼠身体负担,对代谢产生了不利影响。
图4表明,骨密度可以反映骨骼的强壮程度,由上表可知,在对照组中添加了营养强化剂和骨稳态调节剂组别的小鼠(对照组3,对照组4,对照组5,对照组6)的生长激素要比不添加组别(对照组1,对照组2)高,实验组1和实验组3小鼠的骨密度显著高于其他组,说明生长激素促进剂,慢波睡眠促进剂与营养强化剂和骨稳态调节剂有协同作用,帮助增强骨骼强壮。实验组3强壮骨骼的效果并未显著优于实验组1,且过量的营养会导致小鼠身体负担,故没有意义增加添加量。
图5可知,骨稳态调节剂和营养强化剂有助于提升血钙浓度,而骨稳态调节剂与生长激素调节剂/慢波睡眠促进机联合,可以强化血钙浓度升高,更有助于钙离子吸收,加大剂量的营养组合物并未进一步提升血钙浓度,且过量的营养会导致小鼠身体负担,故不建议过量使用营养组合物。
综上所述,向普通配方羊奶粉中,按照7—10%的比例添加本营养组合物具有明显的提高身高的作用。

所述慢波睡眠促进剂由如下重量份的组分组成:γ-氨基丁酸1—2,L-色氨酸1.5—2.5;
