Abstract

本发明提供了一种同种异体骨的化学处理方法,包括异体骨的加工切割和冷冻干燥。该方法是将异体骨材料加工切割后用纯化水清洗干净,除去骨髓,再依次用氢氧化钠、甲醇、三氯甲烷、十二烷基磺酸钠、磷酸盐、酒精处理后,在‑70℃下的环境中深低温冷冻16‑25小时,然后将其放入冷冻干燥机中,按照设定好的冻干曲线进行冷冻干燥。辐照灭菌后得到同种异体骨成品。本发明使用氢氧化钠、甲醇、三氯甲烷、十二烷基磺酸钠、磷酸盐、酒精联合处理异体骨原材料,可以有效地去除骨髓以及其中细胞、蛋白和脂肪成分,同时通过冷冻干燥处理可以进一步的降低同种异体骨的免疫原性,同时又能保持骨组织的三维结构和生物力学性能。另外,本发明设计的化学处理方法简单易行,方便规模化生产操作。

Description

一种同种异体骨的化学处理方法
技术领域
本发明设涉及生物材料领域,具体设计一种同种异体植入性生物材料的化学处理方法。
背景技术
骨折外伤、肿瘤等会导致骨的缺损和结构破坏。已有的技术中常常应用自体骨或人工骨材料等来修复骨的缺损。但是,自体骨的取材有限,同时取材时也加重了病人的创伤并增加了并发症的风险。而陶瓷等人工骨材料等用于骨的修复时,其生物力学性能较差,骨生物活性也有待提高。同种异体骨技术是采用合法捐献的人类合格供体骨组织,经加工处理后,作为充填材料植入体内,是自体骨移植的代用材料。
同种异体骨是目前临床上应用最为广泛的骨替代材料。它具有来源丰富、不受形态、大小限制、具有较好的骨生物活性等优点,在临床上有较好的应用前景。新鲜的同种异体骨具有较强的免疫原性,植入体内后会诱发排斥反应。虽然同种异体骨移植诱发的宿主免疫排异反应一般不会引起危及生命的严重后果,但是免疫排异反应往往干扰移植骨的愈合,并可导致移植骨的过度吸收或骨不连、深部感染等,影响治疗效果。骨组织由矿物质、胶原、非胶原蛋白和细胞成分组成。骨骼中的矿物质不具有抗原性,胶原和非胶原蛋白仅是弱抗原,同种异体骨移植的抗原刺激主要来自其骨髓内的造血细胞、血管内皮细胞、树突细胞、巨噬细胞和骨系细胞。为了降低或消除异体骨的排斥反应,目前国际上普遍采用的是冷冻干燥或深度冷冻的处理方法,消毒则利用γ 射线照线或环氧乙烷来进行。这些处理方法已经成为国际上的一些权威机构,如美国组织库和欧洲组织库的产品生产标准。
目前临床上使用最多的同种异体骨是以下两类:深低温冷冻辐照灭菌骨及冷冻干燥辐照灭菌骨。但单纯采用深低温冷冻及冷冻干燥的方法并不能完全消除同种异体骨的免疫原性方面,需要采用其它的方法进一步降低骨的免疫原性。
发明内容
本发明的目的在于提供一种可以有效地除去异体骨材料中的免疫原成分,同时也可用于规模化生产的同种异体骨的化学处理方法。
本发明的技术方案是:一种同种异体骨的化学处理方法,包括异体骨的加工切割和冷冻干燥,其中,该方法按以下步骤进行:
(1)将加工切割后的异体骨材料用纯化水清洗干净,去除骨髓;
(2)用分析纯的氢氧化钠配置氢氧化钠溶液,将氢氧化钠溶液与异体骨原材料按体积:质量为5-2:1的比例混合,水浴恒温震荡脱去脂肪成分,直至溶液中无明显漂浮油脂为止;
(3)将步骤(2)所得的异体骨材料用甲醇、三氯甲烷体积比为3-1:1的混合溶液浸泡14-18小时;
(4)将步骤(3)所得的异体骨材料用十二烷基磺酸钠溶液浸泡16-20小时;
(5)将步骤(4)所得的异体骨材料用磷酸盐缓冲液浸泡16-20小时;
(6)将步骤(5)所得的异体骨材料用酒精浸泡16-20小时;
(7)将步骤(6)所得的异体骨材料用大量纯化水反复清洗至末道清洗液pH值为6.5-7.5;
(8)将步骤(7)所得的异体骨材料放入-70℃下的环境中冷冻16-25小时;
(9)将步骤(8)所得的异体骨材料放入冷冻干燥机中进行冷冻干燥;
(10)将步骤(9)所得的异体骨材料进行辐照灭菌。
根据本发明的同种异体骨的化学处理方法,较好的是,所述氢氧化钠溶液的质量分数为2%。
根据本发明的同种异体骨的化学处理方法,在一个优选的实施方案中,所述水浴恒温震荡的温度为25-37℃。
步骤(4)中所述十二烷基磺酸钠的质量分数为0.25%。
根据本发明的同种异体骨的化学处理方法,较好的是,所述磷酸盐缓冲液的浓度为0.1-0.2 mol/L,pH值为7.0-7.4 。
根据本发明的同种异体骨的化学处理方法,较好的是,所述酒精的质量分数为75-85%。
根据本发明的同种异体骨的化学处理方法,在一个优选的实施方案中,所述冷冻干燥时间为24小时。
根据本发明的同种异体骨的化学处理方法,较好的是,所述辐照灭菌的剂量为20-30kGy。
本发明的技术原理:通过化学处理的方法,可以使异体骨细胞失活,细胞表面蛋白成分分解,达到消除或减弱抗原性的目的,从而减弱异体骨移植时引起的免疫排斥反应。本发明通过除去骨髓、脱细胞、脱脂、脱蛋白等化学处理的方法,能够有效地消除异体骨材料中残存的细胞膜、完全去除细胞基质,消除免疫原,使植入的同种异体骨能够被宿主细胞所适应,并使免疫原性大大降低,免疫排斥基本消失。
本发明的有益效果在于将异体骨材料加工切割后用纯化水清洗干净,除去骨髓,再依次用氢氧化钠、甲醇、三氯甲烷、十二烷基磺酸钠、磷酸盐、酒精处理,能够有效地去除异体骨材料中的细胞膜和细胞基质等免疫原,大大降低了同种异体骨的免疫原性,同时通过深低温冷冻和冷冻干燥处理,可以进一步的降低同种异体骨的免疫原性,同时一方面保留了骨组织的三维缝隙结构,有利于成骨细胞的爬行,两一方面也保持了较好的生物力学性能。此外,本发明设计的化学处理方法步骤简单移行,有利于实现规模化生产操作。
具体实施方式
为使本领域技术人员更好地理解本发明的技术方案,下面结合实施例对本发明作进一步详细描述。
实施例1
将同种异体骨材料加工切割成颗粒状,然后用纯化水清洗除去骨髓,然后用分析纯的氢氧化钠配置乘2%氢氧化钠溶液,将氢氧化钠溶液与异体骨原材料按体积:质量为2:1的比例混合,37℃水浴恒温震荡脱去脂肪成分,直至溶液中无明显漂浮油脂为止。
将上述所得的异体骨材料用甲醇、三氯甲烷体积比为1:1的混合溶液浸泡16小时;接着用0.1mol/L pH7.2的磷酸盐缓冲液浸泡上一步得到的异体骨材料,时间为16小时;然后用质量分数为0.15%的十二烷基磺酸钠溶液浸泡上一步得到的异体骨材料,时间为16小时;再用质量分数为75%的酒精浸泡上一步得到的异体骨材料,脱水16小时后,用大量的纯化水反复清洗至末道清洗液pH值为7.0;将清洗后的异体骨材料放入-70℃下的环境中冷冻20小时后,放入冷冻干燥机中按照设定好的曲线进行冷冻干燥24小时。最后将冷冻干燥完成的异体骨在25kGy剂量的Co-60下辐照灭菌。
实施例2
将同种异体骨材料加工切割成长条状,然后用纯化水清洗除去骨髓,然后用分析纯的氢氧化钠配置乘2%氢氧化钠溶液,将氢氧化钠溶液与异体骨原材料按体积:质量为2.5:1的比例混合,37℃水浴恒温震荡脱去脂肪成分,直至溶液中无明显漂浮油脂为止。
将上述所得的异体骨材料用甲醇、三氯甲烷体积比为1:1的混合溶液浸泡16小时;接着用0.2mol/L pH7.2的磷酸盐缓冲液浸泡上一步得到的异体骨材料,时间为18小时;然后用质量分数为0.25%的十二烷基磺酸钠溶液浸泡上一步得到的异体骨材料,时间为18小时;再用质量分数为75%的酒精浸泡上一步得到的异体骨材料,脱水19小时后,用大量的纯化水反复清洗至末道清洗液pH值为7.0;将清洗后的异体骨材料放入-70℃下的环境中冷冻20小时后,放入冷冻干燥机中按照设定好的曲线进行冷冻干燥24小时。最后将冷冻干燥完成的异体骨在25kGy剂量的Co-60下辐照灭菌。
实施例3
将同种异体骨材料加工切割成长条状,然后用纯化水清洗除去骨髓,然后用分析纯的氢氧化钠配置乘3%氢氧化钠溶液,将氢氧化钠溶液与异体骨原材料按体积:质量为3:1的比例混合,37℃水浴恒温震荡脱去脂肪成分,直至溶液中无明显漂浮油脂为止。
将上述所得的异体骨材料用甲醇、三氯甲烷体积比为1:1的混合溶液浸泡16小时;接着用0.2mol/L pH7.2的磷酸盐缓冲液浸泡上一步得到的异体骨材料,时间为18小时;然后用质量分数为0.3%的十二烷基磺酸钠溶液浸泡上一步得到的异体骨材料,时间为18小时;再用质量分数为75%的酒精浸泡上一步得到的异体骨材料,脱水19小时后,用大量的纯化水反复清洗至末道清洗液pH值为7.0;将清洗后的异体骨材料放入-70℃下的环境中冷冻20小时后,放入冷冻干燥机中按照设定好的曲线进行冷冻干燥24小时。最后将冷冻干燥完成的异体骨在25kGy剂量的Co-60下辐照灭菌。

Claims (8)

1.一种同种异体骨的化学处理方法,包括异体骨的加工切割和冷冻干燥,其特征在于,该方法按以下步骤进行:
(1)将加工切割后的异体骨材料用纯化水清洗干净,去除骨髓;
(2)用分析纯的氢氧化钠配置氢氧化钠溶液,将氢氧化钠溶液与异体骨原材料按体积:质量为5-2:1的比例混合,水浴恒温震荡脱去脂肪成分,直至溶液中无明显漂浮油脂为止;
(3)将步骤(2)所得的异体骨材料用甲醇、三氯甲烷体积比为3-1:1的混合溶液浸泡14-18小时;
(4)将步骤(3)所得的异体骨材料用十二烷基磺酸钠溶液浸泡16-20小时;
(5)将步骤(4)所得的异体骨材料用磷酸盐缓冲液浸泡16-20小时;
(6)将步骤(5)所得的异体骨材料用酒精浸泡16-20小时;
(7)将步骤(6)所得的异体骨材料用大量纯化水反复清洗至末道清洗液pH值为6.5-7.5;
(8)将步骤(7)所得的异体骨材料放入-70℃下的环境中冷冻16-25小时;
(9)将步骤(8)所得的异体骨材料放入冷冻干燥机中进行冷冻干燥;
(10)将步骤(9)所得的异体骨材料进行辐照灭菌。
2.根据权利要求所述的同种异体骨的化学处理方法,其特征在于,所述氢氧化钠溶液的质量分数为2%-5%。
3.根据权利要求所述的同种异体骨的化学处理方法,其特征在于,所述水浴恒温震荡的温度为25-37℃。
4.根据权利要求所述的同种异体骨的化学处理方法,其特征在于,所述十二烷基磺酸钠的质量分数为0.1%-0.3%。
5.根据权利要求所述的同种异体骨的化学处理方法,其特征在于,所述磷酸盐缓冲液的浓度为0.1-0.2 mol/L,pH值为7.0-7.4 。
6.根据权利要求所述的同种异体骨的化学处理方法,其特征在于,所述酒精的质量分数为75-85%。
