本发明公开了一种降糖颗粒及其制备方法,由以下重量份比的原料药制成:沙棘200-300份、葛根200-300份、黄精180-220份、阿胶130-170份、桔梗130-170份、栀子80-120份、山药180-220份、枸杞子80-120份、罗汉果180-220份、苦瓜干片130-170份、番石榴干片130-170份、桑叶80-120份。本发明组方严谨,具有降糖降脂、生津止渴、滋阴补肾等功效,可明显改善高血糖伴随高血脂、多饮、多尿、多食、消瘦、体倦无力、脾胃虚弱患者等症状,修复胰岛功能。原料药均取自“药食同源”品种及食品,使用安全。

技术领域
本发明属于保健食品领域,涉及一种降糖颗粒及其制备方法。
背景技术
糖尿病是一种全身慢性进行性疾病,是由于胰岛β细胞不能正常分泌胰岛素,胰岛素相对或绝对不足,以及靶细胞对胰岛素敏感性的降低,而引起糖、蛋白、脂肪和水、电解质代谢紊乱,使肝糖原和肌糖原不能合成。临床表现为血糖升高、尿糖阳性及糖耐量降低,典型症状为“三多一少”,即多饮、多尿、多食和体重减少。随着病程延长,容易并发全身神经、大血管、微血管病变,并可导致心、脑、肾、神经及眼等组织器官的慢性进行性病变,诸多并发症日趋增多,程度加重,是严重危害患者健康和生命的内分泌代谢性疾病。糖尿病是世界公认的四大顽症之一,并以难治愈、复发率高、终身服药、并发症多、病死率高等特点而困扰着广大患者。现代医学对糖尿病的治疗主要是降糖治疗,即应用药物把糖从血管移到肝、肌肉,变成肝糖原或转变为脂肪储藏起来。治疗糖尿病的药物有(1)胰岛素;(2)口服降糖药物-磺脲类及双胍类衍生物;(3)中草药等。中药及中药活性成分具有疗效稳定、毒性小、不良反应少和可长期使用的特点,在糖尿病及其并发症的防治方面日益引起人们的重视。
本发明特别针对糖尿病患者,研究出一种安全有效的降糖颗粒,由12味中草药制成,其中沙棘、葛根、黄精、阿胶、桔梗、栀子、山药、枸杞子、罗汉果、桑叶为***2002年公布的《关于进一步规范保健食品原料管理的通知》中既是食品又是药品名单,苦瓜为常食用蔬菜,番石榴为常食用水果,该制剂兼有营养和治疗的双向作用,目前,尚未见相关报道。
(1)沙棘,为胡颓子科植物沙棘Hippophae thamnoides L.的干燥成熟果实。《中国药典》2010年版记载:【性味与归经】酸、涩,温,归脾、胃、肺、心经;【功能与主治】健脾消食,止咳祛痰,活血散瘀。用于脾虚食少,食积腹痛,咳嗽痰多,胸痹心痛,瘀血经闭,跌扑瘀胂。
(2)葛根,为豆科植物野葛Pueraria lobata(Willd.)ohwi的干燥根。《中国药典》2010年版记载:【性味与归经】甘、辛,凉,归脾、胃、肺经;【功能与主治】解肌退热,生津止渴,透疹,升阳止泻,通经活络,解酒毒。用于外感发热头痛,项背强痛,口渴,消渴,麻疹不透,热痢,泄泻,眩晕头痛,中风偏瘫,胸痹心痛,酒毒伤中。
(3)黄精,为百合科植物滇黄精Polygonatum kingianum coll.et Hemsl.、黄精Polygonatum sibirifum Red.或多花黄精Polygonatum cyrtonema Hua的干燥根茎。《中国药典》2010年版记载:【性味与归经】甘,平,归脾、肺、肾经;【功能与主治】补气养阴,健脾,润肺,益肾。用于脾胃气虚,体倦乏力,胃阴不足,口干食少,肺虚燥咳,劳嗽咳血,精血不足,腰膝酸软,须发早白,内热消渴。
(4)阿胶,为马科动物驴Equus asinus L.的干燥皮或鲜皮经煎煮、浓缩制成的固体胶。《中国药典》2010年版记载:【性味与归经】甘,平,归肺、肝、肾经;【功能与主治】补血滋阴,润燥,止血。用于血虚萎黄,眩晕心悸,肌痿无力,心烦不眠,虚风内动,肺燥咳嗽,劳嗽咯血,吐血尿血,便血崩漏,妊娠胎漏。
(5)桔梗,为桔梗科植物桔梗Platycodon grandiflorum(Jacq.)A.Dc.的干燥根。《中国药典》2010年版记载:【性味与归经】苦、辛,平,归肺经;【功能与主治】宣肺,利咽,祛痰,排脓。用于咳嗽痰多,胸闷不畅,咽痛音哑,肺痈吐脓。
(6)栀子,为茜草科植物栀子Gardenia jasminoides Ellis的干燥成熟果实。《中国药典》2010年版记载:【性味与归经】苦、寒,归心、肺、三焦经;【功能与主治】泻火除烦,清热利湿,凉血解毒;外用消肿止痛。用于热病心烦,湿热黄疸,淋证涩痛,血热吐衄,目赤肿痛,火毒疮疡;外治扭挫伤痛。
(7)山药,为薯蓣科植物薯蓣Dioscorea opposita Thunb.的干燥根茎。《中国药典》2010年版记载:【性味与归经】甘,平,归脾、肺、肾经;【功能与主治】补脾养胃,生津益肺,补肾涩精。用于脾虚食少,久泻不止,肺虚喘咳,肾虚遗精,带下,尿频,虚热消渴。麸炒山药补脾健胃。用于脾虚食少,泄泻便溏,白带过多。
(8)枸杞子,为茄科植物宁夏枸杞Lycium barbarum L.的干燥成熟果实。《中国药典》2010年版记载:【性味与归经】甘,平,归肝、肾经;【功能与主治】滋补肝肾,益精明目。用于虚劳精亏,腰膝酸痛,眩晕耳鸣,阳痿遗精,内热消渴,血虚萎黄,目昏不明。枸杞子为***2002年公布的《关于进一步规范保健食品原料管理的通知》中既是食品又是药品名单之一。
(9)罗汉果,为葫芦科植物罗汉果Siraiti grosvenorii(Swin gle)C.Jeffrey ex A.M.Lu et Z.Y.Zhang的干燥果实。《中国药典》2010年版记载:【性味与归经】甘,凉。归肺、大肠经;【功能与主治】清热润肺,利咽开音,滑肠通便。用于肺热燥咳,咽痛失音,肠燥便秘。
(10)苦瓜干片,为葫芦科植物苦瓜Momordica charantia L.的果实,切片晒干。《中华本草》记载:【性味与归经】味苦,性寒,归心、脾、肺经;【功能与主治】清暑涤热;明目;解毒。主暑热烦渴;消渴;赤眼疼痛;痢疾;疮痈肿毒。
(11)番石榴干片,为桃金娘科番石榴属植物番石榴Psidium guajava L.的果实,切片晒干。《全国中草药汇编》记载:【性味】甘、涩,平;【功能与主治】收敛止泻,消炎止血。
(12)桑叶,为桑科植物桑Morus alba L.的干燥叶。《中国药典》2010年版记载:【性味与归经】甘、苦,寒,归肺、肝经;【功能与主治】疏散风热,清肺润燥,清肝明目。用于风热感冒,肺热燥咳,头晕头痛,目赤昏花。
发明内容
本发明的目的是针对高血糖及并发症患者,提供一种降糖颗粒及其制备方法。
本发明人从“药食同源”类中药及食品中选择合适的原料,通过合理配制,制成一种降糖颗粒,含有丰富的营养物质及治疗疾病和改善人体健康的有效成分,口感好,具有降糖降脂、生津止渴、滋阴补肾等功效,可明显改善高血糖伴随高血脂、多饮、多尿、多食、消瘦、体倦无力、脾胃虚弱患者等症状,修复胰岛功能,同时还可以补充人体所需的营养。
本发明所采用的技术方案如下:
一种降糖颗粒,由以下重量份比的原料药制成:沙棘200-300份、葛根200-300份、黄精180-220份、阿胶130-170份、桔梗130-170份、栀子80-120份、山药180-220份、枸杞子80-120份、罗汉果180-220份、苦瓜干片130-170份、番石榴干片130-170份、桑叶80-120份,生产中加入允许的颗粒剂辅料制备而成。
以上所述的降糖颗粒,优选由以下重量份比的原料药制成:沙棘250份、葛根250份、黄精200份、阿胶150份、桔梗150份、栀子100份、山药200份、枸杞子100份、罗汉果200份、苦瓜干片150份、番石榴干片150份、桑叶100份。
以上所述降糖颗粒的制备方法,包括以下步骤:
1.按重量份比分别称取沙棘、葛根、黄精、阿胶、桔梗、栀子、山药、枸杞子、罗汉果、苦瓜干片、番石榴干片、桑叶;
2.将阿胶粉碎成80-100目的细粉,备用;
3.取山药,粉碎至80-100目,加入6-8倍量水,60℃温浸3次,每次2小时,过滤,合并滤液,于60℃减压浓缩,60℃减压干燥,粉碎成80-100目的细粉,备用;
4.取葛根,粉碎成粗粉,加50%的乙醇6-8倍量,加热回流提取2次,每次2小时,过滤,合并滤液,回收乙醇,得葛根提取液;
5.取沙棘、黄精、桔梗、栀子、枸杞子、罗汉果、苦瓜干片、番石榴干片、桑叶,加水8-10倍量,加热提取3次,第一次2小时,其余1小时/次,过滤,合并滤液,加入步骤4得到的葛根提取液,浓缩至60℃相对密度为1.10-1.30的清膏,加入麦芽糊精、可溶性淀粉,混匀,制粒,干燥,得混合物颗粒;
6.在混合物颗粒中加入步骤2中得到的阿胶粉及步骤3中得到的山药粉,混匀,分装,即得。
本发明的制备方法是经过筛选优化得到的,筛选优化过程主要分为提取工艺和制剂工艺,主要筛选情况如下:
葛根的提取工艺:葛根素为葛根的主要有效成分,具有降糖、改善胰岛素、降血脂、扩血管等作用。试验中以葛根素及制剂溶化性为考察指标,对药材粉碎粒度、提取溶剂(水、30%乙醇、50%乙醇、75%乙醇)、溶剂量、提取次数、提取时间进行筛选,试验结果表明:取葛根粗粉,加50%的乙醇6-8倍量,加热回流提取2次,每次2小时,得到的葛根素含量较高,且颗粒溶化性好。
山药的提取工艺:山药含淀粉、粘液蛋白等多糖,粘液蛋白具有降血糖作用。试验中以粘液蛋白为考察指标,对药材粉碎粒度、溶剂量、提取温度、提取次数、提取时间进行筛选,试验结果表明:山药粉碎至80-100目,加入6-8倍量水,60℃温浸3次,每次2小时,过滤,合并滤液,于60℃减压浓缩,60℃减压干燥,得到的粘液蛋白含量最高。
其余成分提取工艺:本发明其余药味采用水提,试验中对加水量,提取次数、时间等进行筛选,结果表明“加水8-10倍量,加热提取3次,第一次2小时,其余1小时/次”最好,以下是对本发明加水量的考察,以正丁醇浸出物为考核指标,详见表1:

提取液浓缩、制粒工艺:以葛根素收得率及溶化性为考核指标,确定药材水提取液浓缩方法、清膏相对密度、加入辅料的成分(对比了淀粉、可溶性淀粉、糊精、麦芽糊精、乳糖),试验结果表明:“浓缩至60℃相对密度为1.10-1.30的清膏,加入麦芽糊精、可溶性淀粉,混匀,制粒,干燥”,且阿胶、山药提取物不加入湿法制粒效果最好;制剂过程中使用的辅料更少,颗粒含生药量多,服用量减少,颗粒粒度均匀,在水中溶化性更好,能在3分钟内完全溶解,溶液透明澄清,没有焦屑及沉淀现象。
本发明的有益效果是:
(1)原料药均取自“药食同源”品种及食用蔬菜水果,使用安全,可长期服用。
(2)本发明配方科学,具有具有降糖降脂、生津止渴、滋阴补肾等功效,可明显改善高血糖伴随高血脂、多饮、多尿、多食、消瘦、体倦无力、脾胃虚弱患者的症状,修复胰岛功能,同时还可以补充人体所需的营养。
(3)含有丰富的营养物质及改善人体健康和的治疗疾病有效成分,方中有罗汉果,口感好,不含蔗糖,也不需要加入甜味剂,适合糖尿病患者服用,还可以补充人体所需的营养。
(4)本发明颗粒剂,根据各原料药的有效成分,采用现代先进工艺制备而成,提取纯化方法科学,稳定性好,辅料少,服用量少,吸收快;不易吸湿受潮,开水冲服,颗粒易溶解,溶液透明澄清,没有焦屑及沉淀现象。
具体实施方式
下面结合具体实施例对本发明作进一步描述,但不限制本发明的保护范围和应用范围:
一、降糖颗粒的制备
实施例1
降糖颗粒的制备方法,包括以下步骤:
1.分别称取沙棘25kg、葛根25kg、黄精20kg、阿胶15kg、桔梗15kg、栀子10kg、山药20kg、枸杞子10kg、罗汉果20kg、苦瓜干片15kg、番石榴干片15kg、桑叶10kg;
2.将阿胶粉碎成80目的细粉,备用;
3.取山药,粉碎至100目,加入7倍量水,60℃温浸3次,每次2小时,过滤,合并滤液,于60℃减压浓缩,60℃减压干燥,粉碎成100目的细粉,备用;
4.取葛根,粉碎成粗粉,加50%的乙醇8倍量,加热回流提取2次,每次2小时,过滤,合并滤液,回收乙醇,得葛根提取液;
5.取沙棘、黄精、桔梗、栀子、枸杞子、罗汉果、苦瓜干片、番石榴干片、桑叶,加水8倍量,加热提取3次,第一次2小时,其余1小时/次,过滤,合并滤液,加入步骤4得到的葛根提取液,浓缩至60℃相对密度为1.26的清膏,加入麦芽糊精、可溶性淀粉,混匀,制粒,干燥,得混合物颗粒;
6.在混合物颗粒中加入步骤2中得到的阿胶粉及步骤3中得到的山药粉,混匀,分装,即得。
实施例2
降糖颗粒的制备方法,包括以下步骤:
1.分别称取沙棘20kg、葛根20kg、黄精18kg、阿胶13kg、桔梗13kg、栀子8kg、山药18kg、枸杞子8kg、罗汉果18kg、苦瓜干片13kg、番石榴干片13kg、桑叶8kg;
2.将阿胶粉碎成100目的细粉,备用;
3.取山药,粉碎至80目,加入6倍量水,60℃温浸3次,每次2小时,过滤,合并滤液,于60℃减压浓缩,60℃减压干燥,粉碎成80目的细粉,备用;
4.取葛根,粉碎成粗粉,加50%的乙醇6倍量,加热回流提取2次,每次2小时,过滤,合并滤液,回收乙醇,得葛根提取液;
5.取沙棘、黄精、桔梗、栀子、枸杞子、罗汉果、苦瓜干片、番石榴干片、桑叶,加水9倍量,加热提取3次,第一次2小时,其余1小时/次,过滤,合并滤液,加入步骤4得到的葛根提取液,浓缩至60℃相对密度为1.12的清膏,加入麦芽糊精、可溶性淀粉,混匀,制粒,干燥,得混合物颗粒;
6.在混合物颗粒中加入步骤2中得到的阿胶粉及步骤3中得到的山药粉,混匀,分装,即得。
实施例3
降糖颗粒的制备方法,包括以下步骤:
1.分别称取沙棘30kg、葛根30kg、黄精22kg、阿胶17kg、桔梗17kg、栀子12kg、山药22kg、枸杞子12kg、罗汉果22kg、苦瓜干片17kg、番石榴干片17kg、桑叶12kg;
2.将阿胶粉碎成100目的细粉,备用;
3.取山药,粉碎至100目,加入8倍量水,60℃温浸3次,每次2小时,过滤,合并滤液,于60℃减压浓缩,60℃减压干燥,粉碎成80目的细粉,备用;
4.取葛根,粉碎成粗粉,加50%的乙醇7倍量,加热回流提取2次,每次2小时,过滤,合并滤液,回收乙醇,得葛根提取液;
5.取沙棘、黄精、桔梗、栀子、枸杞子、罗汉果、苦瓜干片、番石榴干片、桑叶,加水10倍量,加热提取3次,第一次2小时,其余1小时/次,过滤,合并滤液,加入步骤4得到的葛根提取液,浓缩至60℃相对密度为1.30的清膏,加入麦芽糊精、可溶性淀粉,混匀,制粒,干燥,得混合物颗粒;
6.在混合物颗粒中加入步骤2中得到的阿胶粉及步骤3中得到的山药粉,混匀,分装,即得。
二、实施例产品试用
本发明曾用于观察92例高血糖患者。将上述制备好的3种实施例样品,分别用于高血糖伴随高血脂、多饮、多尿、多食、消瘦、体倦无力或脾胃虚弱患者(男48例,女44例,平均年龄53岁)。结果详见表2。

结果表明:采用本发明降糖颗粒,使用安全、有效,具有降糖降脂、生津止渴、滋阴补肾等功效,可明显改善高血糖伴随高血脂、多饮、多尿、多食、消瘦、体倦无力、脾胃虚弱患者的症状,修复胰岛功能,同时还可以补充人体所需的营养。以下为几个典型病例:
病例1:高××,女,55岁,高血糖伴高血脂、多饮、多尿、多食、消瘦症状,空腹血糖15.6mmol/l,餐后2小时血糖28.5mmol/l。服用本发明实施例1,每次1袋(5g/袋),每日3次,连服30日,血糖下降到正常水平(空腹5.8mmol/l,餐后6.5mmol/l),血脂也恢复到正常水平,多饮、多尿、多食症状消除,饮食恢复正常,体重增加,痊愈。
病例2:蒋××,男,60岁,高血糖伴多饮、多尿、多食、消瘦、体倦无力,空腹血糖14.3mmol/l,餐后2小时血糖26.2mmol/l。服用本发明实施例2,每次1袋(5g/袋),每日3次,连服60日,血糖下降到正常水平(空腹5.8mmol/l,餐后6.1mmol/l),体重增加,精力充沛,痊愈。
病例3:张××,女,48岁,高血糖伴消瘦、体倦无力、脾胃虚弱,空腹血糖12.5mmol/l,餐后2小时血糖23.7mmol/l。服用本发明实施例3,每次1袋(5g/袋),每日3次,连服50日,血糖下降到正常水平(空腹5.5mmol/l,餐后6.0mmol/l),食欲增加,体重增加,脸色红润,精力充沛,痊愈。
Claims (3)
1.一种降糖颗粒,其特征在于,由以下重量份比的原料药制成:沙棘200-300份、葛根200-300份、黄精180-220份、阿胶130-170份、桔梗130-170份、栀子80-120份、山药180-220份、枸杞子80-120份、罗汉果180-220份、苦瓜干片130-170份、番石榴干片130-170份、桑叶80-120份。
2.根据权利要求1所述的降糖颗粒,其特征在于,由以下重量份比的原料药制成:沙棘250份、葛根250份、黄精200份、阿胶150份、桔梗150份、栀子100份、山药200份、枸杞子100份、罗汉果200份、苦瓜干片150份、番石榴干片150份、桑叶100份。
3.一种如权利要求1或2所述降糖颗粒的制备方法,其特征在于,包括以下步骤:
(1)按重量份比分别称取沙棘、葛根、黄精、阿胶、桔梗、栀子、山药、枸杞子、罗汉果、苦瓜干片、番石榴干片、桑叶;
(2)将阿胶粉碎成80-100目的细粉,备用;
(3)取山药,粉碎至80-100目,加入6-8倍量水,60℃温浸3次,每次2小时,过滤,合并滤液,于60℃减压浓缩,60℃减压干燥,粉碎成80-100目的细粉,备用;
(4)取葛根,粉碎成粗粉,加50%的乙醇6-8倍量,加热回流提取2次,每次2小时,过滤,合并滤液,回收乙醇,得葛根提取液;
(5)取沙棘、葛根、黄精、桔梗、栀子、枸杞子、罗汉果、苦瓜干片、番石榴干片、桑叶,加水8-10倍量,加热提取3次,第一次2小时,其余1小时/次,过滤,合并滤液,加入步骤(4)得到的葛根提取液,浓缩至60℃相对密度为1.10-1.30的清膏,加入麦芽糊精、可溶性淀粉,混匀,制粒,干燥,得混合物颗粒;
(6)在混合物颗粒中加入步骤(2)中得到的阿胶粉及步骤(3)中得到的山药粉,混匀,分装,即得。
