Verfahren zur Gewinnung eines sedativen Wirkstoffes Gegenstand der vorliegenden Erfindung ist ein Verfahren-zur Gewinnung eines seda tiven Wirkstoffes a. lls Pflanzen der Rauvolfia- Arten in reiner Form, insbesondere aus dem Wurzelnraterial von Rauwolfia serpentina Benth. Dieser Wirkstoff wird naehfolgend als Reserpin bezeiehnet.
Es ist bekannt, dass sieh aus Pflanzen von Rauwolfia-Arten Extrakte mit einer sedativen Wirkung, deren Träger bis heute noch nicht isoliez worden ist, gewinnen lassen.
Es kurde mu gefunden, dass man zu Re serpin in reiner Form gelangen kann, wenn man Pflanzenmaterial von Rauwolfia-Arten mit einem wässrigen, sauren Mittel unter Bildung einer sedativ wirksamen, das entspreehende Salz des Reserpins enthaltenden Lo sung extrahiert, hierauf das Reserpin in Salzform mit einem mit Wasser höchstens teilweise mischbaren organischen Losungsmittel behandelt und aus dem Extrakt das Reserpin in Forum. der Base isoliert. Da. bei kann man z.
B. die durci Extraktion ; der wässrigen sauren Losung erhaltene Reserpinsalzlosung direkt mit einem mit NVasser höchstens teilweise misch- baren organisehen Losungsmittel behandeln, oder man kann aneh ein aus der wässrig sauren Losung gewonnenes festes Salz des Reserpins mit dem organischen Losungsmittel hehandeln.
Reserpin bildet, aus Aceton in der Hitze umkristaliisiert, schwach gelbliche Kristalle vom F. = 262-263 (unter Zersetzung) ; [a] D-117 (Chloroform). Es lässt sieh auch ans einer Mischung von Chloroform und Äther Umkristallisieren. Reserpin ist eine schwache Base und bildet. ein sehwerlösliehes Hydrochlorid vom F. = 221 (unter Zersetzung) und ein Pikrat vom F. = 183-186".
Das leichtlösliche Acetat ist nur in übersehüs- siger verdünnter Essigsäure beständig. Ausserdem ist das Reserpin unlöslich in Tasser uns Petroläther, mässig loslich in Benzol, Äther, kaltem Alkohol und kaltem Aceton und leicht löslich z. B. in Chloroform, 10 /oiger Essig- säure oder verdünnter Phosphorsäure.
Das pharmakologischeWirkungsbild des Reserpins ist komplex. Es zeigt vorwiegend eine starke und langdauernde zentralsedative und blutdrucksenkende Wirkung. Niedere Dosen genügen, um die Tiere für mehrere Stunden in, einen ruhigen Schlaf zu versetzen, aus dem sie vorübergehend wieder aufgeweekt werden können. Es ist für Reserpin charakteristiseh, dab auch hohe Dosen keine narko- tische Wirkung besitzen, sondern dass die Tiere aufweckbar bleiben. Das Reserpin soll therapeutisch ; e Verwendung finden.
Als Ausgangsmaterial bedient man sieh z. B. des fein zerriebenen Pflanzenmaterials von Rauwolfia-Arten, insbesondere des Wur- zelmaterials von Rauwolfia serpentina Benth.
Zur Gewinnung des Wirkstoffes wird beispielsweise wie folgt vorgegangen : Das Aus- gangsmaterial wird mit einem wässrigen sauren Mittel, insbesondere der wässrigen Löslmg einer niederen Fettsäure, wie z. B. der Ameisensäure, Essigsäure, Propionsäure, oder einer Phosphorsäure oder eines sauren Salzes e-iner mehrbasisehen Säure, mehrmals extrahiert. Die so erhaltene sedativ wirksame saure Losung kann zur Weiterverarbeitung entweder auf ein kleineres Volumen eingeengt oder mit Wasser verdünnt oder unverändert belassen werden.
Dieser Auszug wird dann mit einem organisehen, mit Wasser höchstens teilweise mischbaren Losungsmittel, vorzugs- weise mit einem ha-logenierten Kohlenwasserstoff, wie Chloroform, Äthylenchlorid, Methy- lenehlorid oder Trichloräthylen, ferner Benzol oder Essigester, mehrmals extrahiert.
Zur Isolierung des Wirkstoffes kann man z. B. so vorgehe-n, dass man zunäehst die organisehe Losung neutral wäscht und zur Trockne eindampft ; anschliessend wird vorteilhaft der Eindampfrüekstand an einem Adsorptionsmittel, insbesondere einem mittelstarken Adsorptionsmittel, wie mässig aktivem Aluminium- oxyd, Kieselsäure, Hyflo Super Cel (Mar kenprodukt) oder einem andern Silikat ehro matographiert. Das Reserpin lässt sich dureh Petroläther-Benzol-Gemische, Benzol und Ben zol-Aceton-Gemische eluieren. Es kristallisiert in Gegenwart von wenig Aeeton spontan. Es lässt sich z.
B. durch Umkristallisieren aus heissem Aceton, Alkohol oder aus einer Chlo roform-Ather-Misehung weiter reinigen.
Beispiel 1
2000 Gewiehtsteile Rindenpulver aus der Wurzel von Rauwolfia serpentina Benth. werden in 12 000 Volumteilen 15"Anger Essig- säure aufgeschlämmt und während 15 Stuc- den intensiv gerührt. Dann wird die saure Extraktlosung vorteilhafterweise mittels einer Korbzentrifuge vom Rückstand abgetrennt.
Man erhält auf diese Weise etwa 8000 Volum- teile einer dunkelbraunen Losung (Extrakt A).
Die Extraktion wird in g-leicher Weise noch zweimal wiederholt, indem je 9000 Volumteile 15 /oiger wässriger Essigsäure verwendet werden, die jeweils 8000-9000 Volumteile Ex traktlosung' (Extrakt B und C) ergeben. Jede der drei Extraktlosungen von etwa 8000 Volumteilen wird mit 4000 Volumteilen Wasser verdünnt und vorteilhafterweise getrennt mit etwa 8000 Volumteilen Chloroform, aufgeteilt in 3-4 Fraktionen, ausgeschüttelt.
Die zur Extraktion verwendete Menge Chloroform kann bedeutend reduziert werden, wenn z. B. der Chloroformauszug des Extraktes A noch zum erstmaligen Ausziehen des Extraktes B, oder der dritte Chloroformauszug des Extrak- tes A zum zweiten Ausziehen des Extraktes B und anschliessend zum erstmaligen Ausziehen des Extraktes C verwendet wird.
Die Chloro formauszüge werden mit Pottasche oder Soda losung und Wasser gewaschen mit Natrumsulfat getrocknet und im Vacuum zur Trockne eingedampft. Der 45-50 Gewichts- teile betragende Rückstand wird zur weiteren Reinigung an 700--800 Gewiehtsteilen Alu Miniumoxyd chromatographiert. Man erhalt dahei etwa 2, 5 Gewichtsteile kristallisiertes Reserpin.
Beispiel 2
1000 Gewichtsteile gemahlene Wurzeln von Rauwolfia serpentina Benth. werden in der in Beispiel 1 angegebenen Weise mit 1000 Vo lumteilen 15 /oiger Essigsäure estrahiert.
8500 Volumteile der essigsauren Extrakt- losung werden unter gutem Rühren zuerst mit 60 Gewichtsteilen Hyflo Super CeI ttmd anschliessend nacheinander portionenweise mit 70 Gewichtsteilen Natriumnitrat und 700 Gewichtsteilen Kochsalz versetzt. 160 Gewichts teile gebrannter Kalk werden vorsichtig mit 160 Volumteilen Wasser gelöscht und in kleinen Portionen zur Losung gegeben.
Zum Schluss wird noeh eine Stunde weiter gerührt und dann das Gemisch durch ein Filtertuell abgenutseht. Der abgepresste Filterkuchen wird mit 2-300 Vohimteilen gesättigter Kochsalzlösung und 100 Volumteilen Wasser gewaschen und im Vakuum über Phosphor- pentoxd getrocknet.
160 Gewichtsteile des trockenen Rückstan- des werden mit einem Gemiseh aus 350 Volumteilen Chloroform und 150 Volumteiilen Methanol unter Rühren extrahiert, abgenutse-ht und der Rückstand mit 400 Volumteilen desselben Gemisches ausgewaschen. Darauf wird der Rüekstand ein zweites Mal mit 400 Vo lumteilen des Gemisches extrahiert und mit 100 Volumteilen gewaschen. Auf je 500 Vo lamteile der Extraktlösung werden 300 Volumteile Wasser und so viel gesättigte Na triumbiearbonatlösung, dass ein pg von 8,5-9 erreieht wird, zugesetzt, gesehüttelt und die zwei Phasen getrennt.
Die kombinierten untern Phasen (Chloroform) werden mit 200 Volumteilen 2-3 /oiger Kochsalzlosung noci- mals gewaschen. Die kombiniertee obern Phasen (Wasser/Methanol) werden mit 100 Vo lumteilen Chloroform naehextrahiert, worauf diese zweite Chloroformphase die obgenannte 2-3 /aige Kochsalzlösung ebenfalls passiert.
Die vereinigten Chloroformextrakte werden mit Natriumsulfat t getrocknet und im Vakuum znr Trockne eingedampft. Der Rüekstand wird durch Erwärmen in der ein-bis zweifachen Menge Methanol und langes Ste lienlassen kristallisiert. Das Rohreserpin kann über das Hydroehlorid in die Reinbase über geführt werden.
