Abstract
Cette invention se rapporte à un procédé et à un appareil qui permettent d'encapsuler des substances biologiquement actives dans des globules rouges du sang et qui se caractérisent par un système d'électroporation autonome à flux continu éventuellement automatisé (5), qui permet de prélever du sang d'un patient, de séparer les globules rouges et, éventuellement, de recombiner le plasma et les globules rouges modifiés, afin de produire du sang ayant des caractéristiques biologiques modifiées. Cette invention est particulièrement adaptée pour encapsuler des effecteurs allostériques d'hémoglobine, afin de réduire l'affinité des hérythrocytes pour l'oxygène et afin d'améliorer la libération de l'oxygène contenue dans les hérythrocytes de tissus. Le sang est introduit dans le système (5) par une entrée (11) et mélangé à un anticoagulant (27). Le mélange de sang et d'anticoagulant est ensuite amené à passer à travers un filtre (18), jusqu'à atteindre un bol (44) de séparation et de lavage du sang. Le plasma et les globules blancs sont introduits dans le réservoir de plasma (89), tandis que les globules rouges sont retenus dans le bol de lavage (44). Les globules rouges séparés sont ensuite extraits par pompage et mélangés au niveau d'une jonction (67) avec une solution d'hexaphosphate d'inositol (HPI) provenant d'un réservoir (50). Ce mélange est refroidi par un serpentin de refroidissement (68) et ensuite pompé dans une chambre d'électroporation (72) pour y être traité.
