吉林人参低聚肽在制备改善和增强性功能的食品或保健食品中的用途 技术领域
本发明涉及吉林人参低聚肽的一种新用途,具体来说是吉林人参低聚肽在改善和增强性功能的食品或保健食品中的用途。
背景技术
随着社会和经济的不断发展,人类对生活质量的要求越来越高,现代社会的快节奏引起男性的身心疲劳、劳累过度、烟酒过量等诱因使得男性性功能障碍的发病率不断升高。据估计,全球范围内患不同程度性功能障碍的男子至少有数亿人。我国上海对1582名40岁以上的城市男性进行调查,发现***功能障碍发病率高达73.1%。性功能障碍直接影响男性的自尊心和心理健康,损害夫妻感情及家庭稳定,且与其它的涉及男性健康的疾病如心血管疾病、糖尿病、肝肾疾病、***疾病以及精神方面的疾病等密切相关,因此寻找安全而有效的天然化学物来增强性功能和改善性功能障碍显得尤为迫切。
生物活性肽是指对生物机体的生命活动有益或具有生理作用的肽类化合物。肽在分子结构上处于氨基酸和蛋白质的中间位置,但它具有氨基酸和蛋白质所不能替代的功能。低聚肽,又称为活性小分子肽,一般由10个或10个以下氨基酸组成。研究发现低聚肽比单个氨基酸的吸收更有效,且能直接参与蛋白质的合成,因此提高了机体对蛋白质的利用率。在氨基酸运输***出现障碍的情况下,摄入低聚肽却能获得良好的吸收效果。另外,生物活性肽类具有广泛的生物活性,如免疫调节、降胆固醇、降血压、抗血栓、抗癌、抗病毒、抗氧化和清除自由基作用、促进生长等。目前生物活性肽以其高效、安全的特点异军突起,逐渐显示出其在临床营养中的重要作用和广泛的应用前景。
人参为名贵中药材,有关人参的药用已经有四千多年的历史。目前世界上的人参主要有四种:即我国的“吉林人参”、朝鲜的“高丽参”、日本的“东洋参”和加拿大、美国的“西洋参”。我国的人参为五加科植物人参的干燥根及根茎,主要产于吉林省。2012年9月4日,中国***批准人参成为新资源食品,人参的应用将由单一的中药材及保健产品拓展到食品、饮料等领域,范围大幅扩大,这为我国的人参产业发展提供了新的契机。人参具有大补元气、生津止渴、安神等功效。研究发现,人参可加速性成熟过程,对于性机能及性器官发育均有良好促进作用,对应激所致性行为低下亦有较好改善作用。对人参提取物人参皂甙的研究也发现,人参皂苷Rb1可显著改善小鼠性功能。而其中的低聚肽类作为人参中重要的成分对性功能的影响目前未见报道。人参低聚肽在人体不需消化可直接吸收,因而可避免活性物质的 浪费,大大提高其生物活性,可作为一种新型的具有改善和增强性功能作用的食品或保健食品的配方。
发明内容
本发明的目的在于提供一种低粘度、易吸收且吸收快的吉林人参低聚肽在制备改善和增强性功能食品或保健食品中的用途,为通过饮食方式来改善和增强性功能提供一种新途径。
其中所述的食品为粉剂、液体饮料、奶粉及奶制品或烘焙制品。
其中所述的保健食品为粉剂、片剂、软/硬胶囊或口服液。
本发明发现了现有物质—吉林人参低聚肽具有增强性功能的功能,发现了其在制备改善和增强性功能的食品或保健食品中的用途。动物实验发现吉林人参低聚肽能够有效的提高雄性小鼠的性功能。
具体实施方式
下面结合具体实施例对本发明做进一步说明,这些实例应被理解为仅是举例说明,而非以任何方式限制本发明的范围。
实施例1、对雄性小鼠性功能影响的实验研究
一、材料和方法
样品:吉林人参低聚肽,由吉林肽谷生物工程有限责任公司生产,淡黄色固体粉末,主要成分为分子量小于1000的小分子低聚肽,含量为97.4%。
实验动物:健康清洁级成年ICR雄性小鼠168只,体重18~22g,成年ICR雌性小鼠48只,体重14~19g,由北京大学医学部实验动物中心提供。实验动物分笼饲养,每笼6只,自由饮食、饮水。动物饲养实验室符合国标清洁级,温度范围25℃±1℃,相对湿度50%~60%,室内照明控制在12h/12h光暗周期节律。
二、实验方法
剂量及分组:将雄性小鼠按体重随机分为7组,每组24只。设立空白对照组、乳清蛋白组(0.50g/kg·bw)及5个吉林人参低聚肽剂量组(0.125g/kg·bw 0.25g/kg·bw、0.50g/kg·bw、1.00g/kg·bw、2.00g/kg·bw,分别用吉林人参低聚肽一、二、三、四、五组来表示)。各组动物再分为2个亚组,每组12只。空白对照组小鼠灌胃蒸馏水,其余各组灌胃相应浓度的乳清蛋白及吉林人参低聚肽的水溶液。各组小鼠均给以普通饲料,自由进食和饮水。每天灌胃1次,连续灌胃30d后进行各项实验。
1)血清中NO和睾酮检测及性器官称重
取各组第1亚组12只小鼠,末次给样30min后称量小鼠体重。眼球取血,将小鼠血样置4℃冰箱约3h,2000r/min离心15min分离血清,分别检测血清中NO和睾酮含量。血清睾酮含量采用西安核仪器厂γ放射免疫计数器XH-6080测定,试剂为北京北方生物技术研究所试剂。血清中NO含量按一氧化氮检测试剂盒说明书进行,试剂盒购自碧云天生物技术研究所。采完血后处死动物,分别取睾丸、附睾、精囊和***于电子天平上精确称重,计算脏器指数(脏器重量/体重×100)。
2)雄性小鼠交配实验方法
取各组第2亚组12只小鼠,末次给药60min后进行。雌性小鼠于实验前3d连续给予乙烯雌酚,每天按1mg/kg皮下注射给药,使之进入动情期。小鼠交配试验开始时现将单只雄性小鼠放入30cm×15cm×15cm的交配箱中适应5min,然后放入2只雌性小鼠。记录合笼至第一次交配的时间(即交配潜伏期)及20min内交配的次数。观察时间为20:00~23:00,交配试验在弱光、安静条件下进行。
统计方法:所有实验数据均以

表示。采用SPSS13.0软件进行单因素方差分析,p<0.05有统计学意义。
三、实验结果
1)吉林人参低聚肽对雄性小鼠性器官脏器指数的影响
如表1所示,见表1,各组小鼠睾丸的脏器指数相比较均无显著性差异(p﹥0.05),与乳清蛋白组相比,吉林人参低聚肽剂量组小鼠的附性器官(精囊+***)指数显著升高(p﹤0.05)。

2)吉林人参低聚肽对小鼠血清NO和睾酮含量的影响
如表2所示,空白对照组与乳清蛋白组小鼠血清NO和睾酮含量均无显著性差异(p﹥0.05);与乳清蛋白组相比,吉林人参低聚肽各剂量组小鼠血清NO和睾酮含量均有所增高,其中吉林人参低聚肽二、三、四剂量组小鼠血清NO浓度增高最为显著(p﹤0.01),吉林人参低聚肽一、二、三剂量组小鼠血清睾酮浓度增高最为显著(p﹤0.01)。

3)吉林人参低聚肽对小鼠交配潜伏期及20min内交配次数的影响
如表3所示,空白对照组与乳清蛋白组小鼠交配潜伏期及20min内交配次数均无显著性差异(p﹥0.05);与乳清蛋白组相比,吉林人参低聚肽各剂量组小鼠交配潜伏期均缩短,其中以二、三剂量组最为显著(p﹤0.05),吉林人参低聚肽二、三、四剂量组小鼠20min内交配次数显著增多(p﹤0.01)。

四、实验结论
本研究通过设立空白对照组和乳清蛋白组作为对照组,探讨吉林人参低聚肽对雄性小鼠性功能的影响。结果表明,吉林人参低聚肽可以显著增加雄性小鼠附性器官指数,提高血清中的NO和睾酮浓度,缩短小鼠交配潜伏期并增加交配次数,从而有效的增强小鼠性功能,具备作为一种新型增强性功能制剂的潜力,其适宜浓度范围为每天0.125~1g/kg·bw。
