(5)動物又はヒトにおけるインスリン抵抗性の処置方法に使用するための組成物を製造するための、上記(2)に記載の(A)、(B)、(C)いずれか1つ以上の使用。
(6)次の工程

(肥満)
ヒトにおいてBMI(Body Mass Index)値が25以上の場合を指す。BMI値は、(体重(kg))/(身長(m))2で示される。
(インスリン抵抗性)
「インスリン抵抗性」又は「インスリン感受性の低下」とは、インスリンの作用についての「予想された」又は「正常な」値と比較して、インスリンの作用に対する体の組織の感受性が低下していることを指す。より具体的には、インスリン抵抗性は、外因性又は内因性インスリンのいずれかに対する生物学的応答が損なわれていることと定義される。インスリン抵抗性又はインスリン感受性の低下が存在すると、グルコース負荷試験において経時的な血糖値の低下が小さくなり、またインスリン負荷試験においてインスリン摂取に伴う血糖値の低下が起こらないため、これらをインスリン抵抗性の指標として使用しうる。
(インスリン抵抗性の治療又は予防)
インスリン抵抗性を、よりインスリン感受性へ変化させること、及び、インスリン抵抗性を、よりグルコース輸送活性へ変化させることが含まれ、具体的には、インスリン抵抗性におけるインスリンシグナル伝達阻害作用を制御することが含まれ、例えばインスリン抵抗性を伴う疾患を治療することがあげられる。
「FSTL3」は、アクチビンと結合する分泌糖タンパク質である(Tsuchidaら、J Biol Chem 275:40788-96(2000);Hillら、J Biol Chem 277:40735-41(2002))。このタンパク質は、シグナルペプチドならびにフォリスタチン様ドメイン(SMARTアクセッションSM00274)及びKazal様セリンプロテアーゼインヒビタードメイン(SMARTアクセッションSM00280)を含む2つのタンデムセグメントからなる。FSTL3は、主に胎盤、卵巣、子宮及び精巣において発現するが、皮膚、心臓、肺及び腎臓などの組織においても発現する。
「FSTL3阻害剤」とは、FSTL3が有する、肥満及び/又はインスリン抵抗性を惹起する機能・作用を阻害・遮断する物質を意味する。そのような物質の典型として、(A)FSTL3と特異的に結合してFSTL3の機能・作用を阻害又は抑制する物質、(B)FSTL 3の発現に対する阻害物質、(C)FSTL3との競合を通じてFSTL3の機能を阻害又は抑制する物質を挙げることができる。
上記(A)の具体例として、FSTL3結合タンパク質、FSTL3に対する抗体又はその抗原結合活性を有するフラグメント、(B)の具体例として、FSTL3のアンチセンスオリゴヌクレオチド、siRNA、それらの化学的修飾体、及びFSTL3発現阻害薬物、さらに(C)の具体例として、FSTL3が有する、肥満及び/又はインスリン抵抗性を惹起する機能・作用を欠損したFSTL3の断片やFSTL3ホモログがあげられる。
本発明に用いる「抗体」は、FSTL3に固有の相互作用しうる相手との結合活性、あるいはシグナル伝達阻害活性等の機能を阻害又は抑制する活性を示すものであり、そのような抗体はFSTL3の機能を選択的に阻害する、すなわちFSTL3によるインスリン抵抗性を改善することができる。本発明には、抗体はポリクローナル抗体、モノクローナル抗体のどちらでもよい。さらに、抗体又はその抗原結合活性を有するペプチドフラグメントであってもよいし、当業者に周知の抗原を投与する免疫法を用いた製造法以外にDNA免疫法や遺伝子組み換え技術を用いて製造されたものであってもよい。
以下に代表的な抗体の製造方法を例示する。
一般に、特定のタンパク質又はそのペプチドフラグメントに対するポリクローナル抗体を得るには、該タンパク質又はペプチドを抗原として動物を免疫する。免疫は哺乳動物(例えばラット、マウス、ウサギ、ヒトなど) に静脈内、皮下又は腹腔内に投与することにより行う。また、免疫の間隔は特に限定されず、数日から数週間間隔、好ましくは2~3週間間隔で、1~10回、好ましくは4~5回である。最終の免疫日から7~10日後に抗体価を測定し、最大の抗体価を示した日に採血し、抗血清を得る。抗体価の測定は、酵素免疫測定法(ELISA)、放射性免疫測定法(RIA)、免疫組織染色法等により行うことができる。抗血清から抗体の精製が必要とされる場合は、硫安塩析法、イオン交換クロマトグラフィー、ゲル濾過、アフィニティークロマトグラフィーなどの公知の方法を適宜選択して、又はこれらを組み合わせることにより精製することができる。
1)免疫
モノクローナル抗体を得るには、一般に、抗原タンパク質又はそのペプチドフラグメントを用いて動物を免疫する。免疫は、哺乳動物( 例えばラット、マウスなど) に静脈内、皮下又は腹腔内に投与することにより行う。抗原の1回の投与量は、マウスの場合1 匹当たり30μgである。また、免疫の間隔は特に限定されず、数日から数週間間隔、好ましくは2~3週間間隔で、最低4~5回行う。そして、最終免疫後、抗体産生細胞を採集する。抗体産生細胞としては、脾臓細胞が好ましい。
次いで、脾臓細胞等の抗体産生細胞とミエローマ細胞との細胞融合を行う。ミエローマ細胞として、マウスなどの動物由来の細胞であって一般に入手可能な株化細胞を使用することができる。使用する細胞株として、薬剤選択性を有し、未融合の状態ではHAT選択培地(ヒポキサンチン、アミノプテリン及びチミジンを含む)で生存できず、抗体産生細胞と融合した状態でのみ生存できる性質を有するものが好ましい。例えば、ミエローマ細胞の具体例としてはP3X63-Ag、X63Ag8.653などのマウスミエローマ細胞株が挙げられる。血清を含まないDMEM、RPMI-1640培地などの動物細胞培養用培地中に、抗体産生細胞とミエローマ細胞とを所定の割合(例えば3:1)で混合し、ポリエチレングリコール等の細胞融合促進剤存在の下で、あるいは電気パルス処理(例えばエレクトロポレーション)により行う。
ハイブリドーマを選別する。例えば、ヒポキサンチン(100μM)、アミノプテリン(0.4μM) 及びチミジン(16μM) を含む培地を用いて培養し、生育する細胞をハイブリドーマとして得ることができる。次に、増殖したハイブリドーマの培養上清中に、目的とする抗体が存在するか否かをスクリーニングする。ハイブリドーマのスクリーニングは、通常の方法に従えばよく、特に限定されるものではない。例えば、ハイブリドーマとして生育したウェルに含まれる培養上清の一部を採集し、免疫染色法、酵素免疫測定法(ELISA)、放射性免疫測定法(RIA)等によってスクリーニングすることができる。
融合細胞のクローニングは、限界希釈法等により行い、最終的に単クローン抗体産生細胞であるハイブリドーマを樹立する。樹立したハイブリドーマからモノクローナル抗体を採取するには、通常の細胞培養法等を採用することができる。細胞培養法においては、ハイブリドーマを10%牛胎児血清含有RPMI-1640培地又はMEM培地等の動物細胞培養培地中、通常の培養条件(例えば37℃、5% CO2濃度) で 例えば14日間培養し、その培養上清からモノクローナル抗体を取得する。
抗体の精製が必要とされる場合は、硫安分画法、イオン交換クロマトグラフィー、アフィニティークロマトグラフィー、ゲルクロマトグラフィーなどの公知の方法を適宜に選択して、又はこれらの方法を組み合わせることにより精製することができる。
抗体は、ファージベクター又は類似のベクターからなる抗体ライブラリーからの選択によっても得ることができる( Huseら、(1989)「Generation of a Large Combinatorial Library of the Immunoglobulin Repertoire in PhageLambda」, Science 246: 1275-1281;及びWardら、(1989)Nature 341:544-546) 。
抗体は、キメラ抗体又はヒト化抗体等の形であってもよい。特にヒトに適用する場合は、免疫反応を最小にするためにヒト化することが好ましい。そのようなヒト化抗体の作成方法は当業者にとって既知である。また、目的に応じて適宜、放射性核種、酵素、基質、コファクター、インヒビター、蛍光性標識、化学発光性標識、磁気粒子など、検出可能な標識を用いて標識することができる。
前記(A)FSTL3と特異的に結合してFSTL3の機能・作用を阻害又は抑制する物質としては、FSTL3と特異的に結合し、FSTL3の機能を阻害又は抑制することができることを条件として、上記抗体以外のFSTL3特異的結合性物質を用いることができる。そのような物質としてアクチビン類、ポリマー又は化学的試薬等が挙げられる。
・db/dbマウスや高脂肪食負荷マウス、遺伝子操作等によってFSTL3遺伝子の発現量が増加しているマウス個体に候補化合物を投与してITTあるいはGTTを行う。
・リアルタイムPCRやノザンブロット等で、M1マクロファージのマーカーであるIL-6やTNF-α等の発現量の減少、あるいはM2マクロファージのマーカーであるアルギナーゼなどの発現量の増加を確認する。
・ウエスタンブロット等で肝臓あるいは骨格筋中のリン酸化されたAktを測定する。
これらのアッセイの具体的な手順は、本願明細書の実施例を参照した当業者には明らかであろう。
(i)FSTL3のアンチセンス
本明細書において、「アンチセンスオリゴヌクレオチド(アンチセンス)」とは、2本鎖DNAのアンチセンス鎖のDNA又はそのアンチセンス鎖のDNAに対応するRNAであって、DNAもしくはRNAに結合し、FSTL3の発現を調節するものを意味する。アンチセンスオリゴヌクレオチドは、例えばFSTL3タンパク質をコードする遺伝子の塩基配列を基にしてDNAとして製造するか、又はこのDNAをアンチセンスの向きに発現プラスミドに組み込むことでRNAとして製造することができる。このアンチセンスオリゴヌクレオチドはFSTL3 DNAのコーディング配列、5’ノンコーディング配列のいずれの部分のDNA断片と相補的な配列であってもよいが、好ましくは転写開始部位、翻訳開始部位、5’非翻訳領域、エクソンとイントロンとの境界領域もしくは5’CAP領域に相補的配列であることが望ましい。
FSTL3のmRNAに相同的な21~25塩基程度で両方の3’末端が突き出た短い二本鎖RNAである。siRNAは、FSTL3のcDNAを鋳型として合成することができる。本発明に用いられるsiRNAは、FSTL3の塩基配列に対して、上記アンチセンスオリゴヌクレオチドと同様の関係を有するものであってよい。
FSTL3アンチセンスオリゴヌクレオチドの化学的修飾体としては、DNA又はRNAの細胞内への移行性又は細胞内での安定性を高めるために修飾された誘導体であり、例えば、ホスホロチオエート、ホスホロジチオエート、アルキルホスホトリエステル、アルキルホスホナート、アルキルホスホアミデート等の誘導体("Antisense RNA and DNA" WILEY-LISS刊 1992 P.1-50)が挙げられる。この化学的修飾体は、同文献等に記載の方法に従って製造することができる。このアンチセンスオリゴヌクレオチド又はその化学的修飾体を用いて、FSTL3タンパク質をコードする遺伝子の発現を制御することにより、インスリン抵抗性を改善することができる。アンチセンスオリゴヌクレオチド又はその化学的修飾体をそのまま投与する場合は、このアンチセンスオリゴヌクレオチドの好ましい長さとしては、例えば5~200塩基のものが挙げられ、さらに好ましくは10~50塩基のものが挙げられ、特に好ましくは15~30塩基のものが挙げられる。
アンチセンスオリゴヌクレオチドを発現プラスミドに組み込む場合は、このアンチセンスオリゴヌクレオチドの好ましい長さとしては、1,000塩基以下、好ましくは500塩基以下、より好ましくは150塩基以下である。アンチセンスオリゴヌクレオチドを発現プラスミドに組み込んだ後、目的の細胞に常法に従って導入する。導入はリポソームや組換えウイルスなどを利用する方法で行うことができる。アンチセンスオリゴヌクレオチドの発現プラスミドは通常の発現ベクターを用いてプロモーターの下流、逆向きに、すなわちFSTL3遺伝子が3’から5’の向きに転写されるように、連結することにより作製できる。
siRNAは、細胞内に導入されると標的mRNAを配列特異的に認識して切断することにより標的遺伝子の発現を抑制する。siRNAは、直接細胞内に導入するか、siRNA発現ベクターに組み込んで導入することができる。(参考:「RNAi実験プロトコール」羊土社)
FSTL3の変動とインスリン抵抗性の関係において、FSTL3が相互作用しうる相手(例えばアクチビンなど)との間でFSTL3と競合が成立し、FSTL3阻害剤(C)が共存することにより、共存しない場合に起こりうるFSTL3の機能・作用を阻害できるものであれば、特に制限はない(ここで、当該FSTL3阻害剤(C)が、FSTL3が相互作用しうる相手が有するインスリン感受性(インスリンシグナル伝達経路の活性)の恒常性維持に関して、実質的に悪影響を与えないものであることは当然のことである)。具体的にはFSTL3が相互作用しうる相手への作用を欠損したFSTL3由来のペプチド断片(以下、単に「FSTL3のペプチド断片」ということがある)やFSTL3ホモログ、アクチビン分子におけるFSTL3結合部位と結合する抗アクチビン抗体などがそれに相当するが、FSTL3阻害剤(A)との相違は、FSTL3との間で結合を形成することまで必要としないことである。
「FSTL3阻害剤」の候補化合物が、「(A):FSTL3と特異的に結合してFSTL3の機能を阻害又は抑制する物質」であるか否かを判定するためには、例えば、まず(1)候補化合物とFSTL3との特異的な結合を確認し、次いで(2)候補化合物によるFSTL3の機能の阻害又は抑制を確認することで達成される。
(1)候補化合物とFSTL3との特異的な結合の確認
候補化合物とFSTL3との特異的な結合を確認する具体的な方法としては、ELISA法や表面プラズモン共鳴現象(SPR)を原理とするBIACORE(登録商標)法をあげることができる。
(2) 候補化合物によるFSTL3の機能の阻害又は抑制の確認
候補化合物によるFSTL3の機能の阻害又は抑制を確認する方法としては、FSTL3の発現を遺伝子レベル、あるいはタンパク質レベルで検出できる方法であればいずれも用いることができる。
すなわち、FSTL3によるシグナル伝達によって遺伝子発現が誘導されるIL-6又はTNF-αの遺伝子発現量が、候補化合物の存在によって発現量が減少することを、遺伝子レベルあるいはタンパク質レベルで検出することができればよい。
(i) FSTL3に特異的に結合する抗体(抗FSTL3抗体)
前記した製造方法により得られた抗FSTL3抗体は、それを直接標識するか、又は該抗体を一次抗体とし、該一次抗体を特異的に認識する(抗体を作製した動物由来の抗体を認識する) 標識二次抗体と協働で検出に用いられる。
(ii)固相化プレートの作成
FSTL3を固相化する方法としては、例えば、96穴プレート(イムノプレート・マキシソープ(ヌンク社製)等) にFSTL3の希釈液(例えば、0.05%アジ化ナトリウムを含むリン酸緩衝生理食塩水(以下「PBS」という)で希釈したもの)を入れて4℃~室温で一晩、又は37 ℃で1 ~3時間静置して、ウェル底面にFSTL3を吸着させればよい。
試料を固相化させたプレートのウェル内底面への抗体の非特異的吸着を阻止するため、予め非特異的吸着を阻害する物質(スキムミルク、カゼイン、ウシ血清アルブミン、ゼラチン、ポリビニルピロリドン等) を含む緩衝液をウェルに分注し、一定時間静置しておく(ブロッキング)。ブロッキング溶液の組成は、例えば、5 %スキムミルク、0.05~0.1% Tween20(登録商標)を含むリン酸緩衝生理食塩水(PBS)又はトリス緩衝生理食塩水(TBS)が用いられる。スキムミルクの代わりに、ブロックエース(大日本製薬社製) 、1~10%のウシ血清アルブミン、0.5~3%のゼラチン又は1%のポリビニルピロリドン等を用いてもよい。
ここで、用いた抗体が標識されたものである場合は、ただちに検出操作を行うことができる。未標識の抗体を用いた場合には、引き続き二次抗体反応を行う。標識二次抗体は、例えば、市販のものを使用する場合は洗浄液で2000~20000倍に希釈して用いる(添付の指示書に好適な希釈倍率が記載されている場合は、その記載に従う) 。一次抗体を洗浄除去した後のウェルに二次抗体溶液を分注して室温で1~3時間インキュベーションし、洗浄液で洗浄してから、標識方法に合わせた検出操作を行う。
(iv)任意の発現プラスミドを用いてFSTL3を強制発現させた任意の培養細胞に、上記工程にて選別された抗体を添加し、FSTL3によって発現が誘導・増加するIL-6の発現が抑制・減少するかを、遺伝子レベル、あるいはタンパク質レベルで検出する。IL-6の発現の検出方法は、FSTL3に関して後述する方法が適用できることは当業者にとって明らかであろう。
「FSTL3阻害剤」の候補化合物が、「(B): FSTL3の発現に対する阻害物質」であるか否かを判定する方法としては、FSTL3の発現を(1)遺伝子レベル、あるいは(2)タンパク質レベルで検出できる方法であればいずれでもよい。
遺伝子レベルの発現を検出する方法としては、遺伝子チップ、cDNAアレイ、及びメンブレンフィルターから選ばれる固相化試料を用いた核酸ハイブリダイゼーション法、RT-PCR法、リアルタイムPCR法等をあげることができ、特にRT-PCR法、リアルタイムPCR法が好ましい。
FSTL3遺伝子の検出方法としては、例えば、脂肪細胞からまず全RNAを抽出し、該全RNA中におけるFSTL3遺伝子(mRNA)の発現量を検出する方法を挙げることができる。
(i)全RNAの抽出
全RNAの抽出は、公知の方法にしたがい、単離された血液又は細胞よりRNA 抽出用溶媒を用いて抽出する。該抽出溶媒としては、例えば、フェノール等のリボヌクレアーゼを不活性化する作用を有する成分を含むもの(例えば、TRIzol試薬: ギブコ・ビーアールエル社製等) が好ましい。RNAの抽出方法は特に限定されず、例えば、チオシアン酸グアニジン・塩化セシウム超遠心法、チオシアン酸グアニジン・ホットフェノール法、グアニジン塩酸法、酸性チオシアン酸グアニジン・フェノール・クロロホルム法(Chomczynski, P. and Sacchi, N., (1987) Anal. Biochem., 162, 156-159) 等を採用することができる。なかでも、酸性チオシアン酸グアニジン・フェノール・クロロホルム法が好適である。
次に、被験物質の投与又は非投与条件下における、全RNA中のFSTL3遺伝子の発現量を検出する。遺伝子の発現量は、得られた全RNAよりcRNA又はcDNAを調製し、これを適当な標識化合物でラベルすることにより、そのシグナル強度として検出することができる。以下、遺伝子の発現量の検出方法について、RT-PCR法(リアルタイムPCR法)について説明する。
RT-PCR法やその1つであるリアルタイムPCR(TaqMan PCR)法は微量なDNAを高感度かつ定量的に検出できるという点で本発明の評価方法に好適である。リアルタイムPCR(TaqMan PCR)法では、5’端を蛍光色素(レポーター)で、3’端を消光剤(クエンチャー)で標識した、目的遺伝子の特定領域にハイブリダイズするオリゴヌクレオチドプローブが使用される。該プローブは、通常の状態ではクエンチャーによってレポーターの蛍光が抑制されている。この蛍光プローブを目的遺伝子に完全にハイブリダイズさせた状態で、その外側からTaq DNAポリメラーゼを用いてPCRを行う。Taq DNAポリメラーゼによる伸長反応が進むと、そのエキソヌクレアーゼ活性により蛍光プローブが5’端から加水分解され、レポーター色素が遊離し、蛍光を発する。リアルタイムPCR法は、この蛍光強度をリアルタイムでモニタリングすることにより、鋳型DNAの初期量を正確に定量することができる。リアルタイムのモニタリングには、LightCycler(登録商標) 480システム(ロシュ・ダイアグノスティックス株式会社)などを用いることができる。
また、タンパク質レベルの発現を検出する方法としては、ウェスタンブロット法、ドットブロット法、スロットブロット法、ELISA法、及びRIA法等をあげることができ、特にウェスタンブロット法が好ましい。
(i)試料の調製
検体としては、脂肪細胞又はFSTL3やその遺伝子が高発現している細胞が好ましい。これらの細胞(細胞抽出液として使用する)は、必要に応じて高速遠心を行うことにより不溶性の物質を除去した後、以下のようにウエスタンブロット用試料として調製する。
ウエスタンブロット用(電気泳動用)試料は、例えば、細胞抽出液をそのまま使用するか、緩衝液で適宜希釈して、SDS-ポリアクリルアミドゲル電気泳動用の2-メルカトルエタノールを含むサンプル緩衝液(シグマ社製等)と混合したものを用いる。
上記方法では、まず、FSTL3が含まれる試料中のポリペプチドをメンブレンあるいは96穴プレートのウェル内底面等に固相化する。メンブレンに固相化する方法としては、試料のポリアクリルアミドゲル電気泳動を経てメンブレンにポリペプチドを転写する方法(ウエスタンブロット法)と、直接メンブレンに試料又はその希釈液を染み込ませる方法(ドットブロット法やスロットブロット法)を挙げることができる。用いられるメンブレンとしては、ニトロセルロースメンブレン(例えば、バイオラッド社製等)、ナイロンメンブレン(例えば、ハイボンド-ECL(アマシャム・ファルマシア社製)等)、コットンメンブレン(例えば、ブロットアブソーベントフィルター(バイオラッド社製)等)又はポリビニリデン・ジフルオリド(PVDF)メンブレン(例えば、バイオラッド社製等)等を挙げることができる。また、ブロッティング方法としては、ウエット式ブロッティング法(CURRENT PROTOCOLS IN IMMUNOLOGY volume 2 ed by J. E. Coligan, A. M. Kruisbeek, D. H. Margulies, E. M. Shevach,W. Strober)、セミドライ式ブロッティング法(上記CURRENT PROTOCOLS IN IMMUNOLOGY volume 2 参照) 等を挙げることができる。
本工程で用いられる抗FSTL3抗体は、前記した方法で作製することができる。
(iv) 測定操作
まず、抗体の非特異的吸着を阻止するため、予めメンブレンを非特異的吸着を阻害する物質(スキムミルク、カゼイン、ウシ血清アルブミン、ゼラチン、ポリビニルピロリドン等) を含む緩衝液中に一定時間浸しておく操作(ブロッキング) を行う。ブロッキングの条件等は、前記したELISA法と同様である。
FSTL3阻害剤の候補化合物が、「(C):FSTL3の競合物質」に該当するか否かを判定する方法としては、候補化合物が、特性(1):アクチビンに対しFSTL3と競合的に結合してアクチビン-FSTL3複合体の形成を阻害し、特性(2):アクチビンの活性を阻害しない、という二つの特性の両方を有することを確認できる方法であればいずれも使用できる。
特性(1)及び特性(2)の両方の特性を有する物質としては、例えば、アクチビンに結合するが、アクチビン活性を阻害しないFSTL3ホモログや、アクチビン分子中のFSTL3結合部位に対する抗体またはその断片であって、アクチビンのアクチビン受容体への結合を阻害しないもの等があげられる。
特性(1)を確認する方法としては、表面プラズモン共鳴現象(SPR)を原理とするBIACORE(登録商標)法等があげられる。特性(1)を有する化合物のうち、特性(2)を有する化合物は、上記したIL-6あるいはTNF-αの発現を遺伝子レベルあるいはタンパク質レベルで測定すれば特定することができる。
アクチビン分子は二量体であるため、アクチビンI型受容体結合部位、及びアクチビンII型受容体結合部位がアクチビン1分子中にそれぞれ2箇所存在するが、二量体のアクチビン1分子に対し2分子のFSTL3がアクチビン分子を取り囲むように結合することで、アクチビンが受容体と結合することを阻害している(J Biol Chem. 2008 Nov. 21; 283(47): 32831-8.)。FSTL3は、連続していない2箇所の接触点がアクチビンと結合していて、1つ目の接触点はN末ドメインで、この接触はアクチビンI型受容体結合部位をブロックし、2つ目の接触点はフォリスタチンドメイン(FSD)の1(FSD1)と2(FSD2)で、この接触はアクチビンII型受容体結合ドメインをブロックしている。
FSTL3とアクチビンそれぞれの遺伝子配列及びアミノ酸配列は既に公知であり、FSTL3とアクチビンの結合部位、及びアクチビンとアクチビン受容体結合部位のアミノ酸あるいは塩基配列、立体構造、電荷などの情報を基に、特性(1)及び特性(2)の両方の特性を有するペプチドの塩基配列あるいはアミノ酸配列を設計し、任意の遺伝子発現システムやペプチド合成機等を用いてFSTL3ホモログを得ることができる。
また、本発明で用いられるFSTL3ホモログの一例としては、配列番号3で表されるアミノ酸配列と実質的に同一のアミノ酸配列を含有するペプチドが好ましく用いられる。改変を加える部位としては、例えば配列番号3の110番目のアミノ酸(S:セリン)から131番目のアミノ酸(D:アスパラギン酸)があげられ、少なくとも1以上のアミノ酸を欠失、置換、挿入する変異を加えることができる。
また、本発明で用いられるFSTL3ホモログの一例としては、配列番号3で表されるアミノ酸配列と実質的に同一のアミノ酸配列を含有するペプチドが好ましく用いられる。改変を加える部位としては、例えば配列番号3の156番目のアミノ酸(V:バリン)から169番目のアミノ酸(R:アルギニン)または、194番目のアミノ酸(S:セリン)から210番目のアミノ酸(V:バリン)があげられ、それぞれ少なくとも1以上のアミノ酸を欠失、置換、挿入する変異を加えることができる。
なお、第1番目から第71番目までのアミノ酸を改変した場合には、第72番目から第236番目までのアミノ酸に改変を加えないことが好ましく、また、第72番目から第236番目までのアミノ酸を改変した場合には、第1番目から第71番目までのアミノ酸に改変を加えないことが好ましいが、アクチビン活性を指標として上記以外の組合せで改変を加えることもできる。
このようにして設計されたFSTL3ホモログは、本発明のスクリーニング方法の試験化合物(候補化合物)として好適に使用することができる。
「(C):FSTL3の競合物質」は、アクチビンの分子構造のうちFSTL3結合部位と結合し、かつアクチビンがアクチビン受容体に結合することを妨げない抗アクチビン抗体又は該抗アクチビン抗体の機能性断片でもよい。例えば、アクチビンの受容体結合部位のみを抗原として、公知の方法で抗アクチビン抗体を作製する。該抗体の機能性断片も分解酵素処理など公知の方法で得ることができる。
本発明のFSTL3阻害剤を用いてFSTL3の機能を阻害・抑制するには、FSTL3阻害剤の有効量を、単独で、あるいは別の薬物と組み合わせて、適当な経路で、治療等が必要な患者に投与する。
本発明者により、FSTL3がインスリン抵抗性に重要な役割を担っていることが明らかになった。従って、前記したスクリーニング法に加え、該タンパク質を発現している細胞を用いてFSTL3を高発現している個体を治療するための化合物(FSTL3阻害剤)をスクリーニングすることができる。そのようなスクリーニングは、(1)試験化合物をFSTL3発現細胞に接触させる工程及び(2)試験化合物がFSTL3の機能を阻害するか否かを確認する工程を含む。FSTL3発現細胞は、2型糖尿病モデル動物及び該動物由来細胞、あるいはFSTL3遺伝子を適当な発現ベクターに組込み、宿主細胞に導入し、FSTL3を発現させた細胞を用いることもできる。宿主細胞としては、FSTL3を発現しうる任意の原核性、真核性等の既知の宿主細胞を使用できる。発現ベクターの構築や宿主細胞の形質転換法は当業者に既知である。また、FSTL3の機能を阻害するか否かは、試験化合物の存在下と非存在下でのインスリンシグナル伝達の状態(例えばAktのリン酸化)等を比較する方法等を利用して実施することができる。

(1) 試験化合物をFSTL3発現細胞に接触させる工程
(2) 試験化合物がFSTL3によるシグナル伝達阻害を改善するか否かを確認する工程
を含む、肥満あるいはインスリン抵抗性を予防又は治療するための化合物をスクリーニングする方法。」
1. FSTL3組換えアデノウイルス投与によるFSTL3過剰発現の糖代謝への影響
全長マウスFSTL3 cDNAは、pcDNA3.1/V5-HisA(インビトロジェン製)を用い、推奨された方法に従って増幅された。増幅産物をHindIII及びEcoRV処理し、組換えアデノウイルス作製に用いた。組換えアデノウイルスは、Takara Adenovirus Expression Vector Kit(Takara製)を用いて、推奨された方法に従って作製した。ネガティブコントロールとして、キットに添付されているβ-ガラクトシダーゼ遺伝子含有ウイルスを用いた。HEK293細胞へのアデノウイルスのトランスフェクションはCellPhect(登録商標) Transfection Kit(GE Healthcare製)を用いてリン酸カルシウム法で行った。7週齢の雄性B6マウス(普通食摂取)に、作製したウイルスを、5.0x 1010pfu/匹になるようPBS 150μLに溶解し、週に1度、2週間投与した。
(2)インスリン負荷試験(ITT)及びグルコース負荷試験(GTT)
最後の投与から5日後にITT、7日後にGTTを行った。試験は、各群5~6匹で行った。
(2)‐1
インスリン負荷試験(ITT)
ITTは以下のように行った。絶食させずに随時マウスの腹腔にインスリン3.0mU/g-BW(生理的食塩水で希釈)を投与した。採取した全血からグルテストセンサー(三和化学)を用いてグルコース濃度を測定した。
(2)‐2
グルコース負荷試験(GTT)
GTTは以下のように行った。試験前日より16時間絶食させたマウスの腹腔にグルコース1.0mg/g-BW(生理的食塩水で希釈)を投与した。採取した全血から血清を分離し、グルコーステストワコー(和光純薬工業社製)にてグルコース濃度を測定した。
(3)結果
ITTにおいてFSTL3遺伝子を有するウイルスを投与したマウスでは、コントロールマウスと比較して、明らかに血糖値の低下が少なかった。また、グルコース負荷試験においてFSTL3遺伝子を有するウイルスを投与したマウスでは、コントロールマウスと比較して、有意な血糖値の上昇が確認された。この結果から、FSTL3はインスリン抵抗性を惹起することがわかった。(図1-1参照)
(1) 組換えアデノウイルス作製とマウスへの投与
組換えアデノウイルスの作製及びマウスへの投与は上述の方法で行った。
(2) RNAの調製
RNeasy Mini Kit (250) (Qiagen製, Cat. number 74106)を用いて、内臓脂肪よりRNAを抽出した。
(3) アディポカインの測定
(2)で抽出したRNAを用い、High Capacity cDNA Reverse Transcription Kit with RNaseInhibitor (ABI製)のプロトコールに則って逆転写反応を行って、TaqMan PCRを行った。M2マクロファージマーカーとしてアディポネクチン、M1マクロファージマーカーとしてIL-6、TNF-αを選び、それぞれTaqManプローブを作成した。
(4) 結果
M2マクロファージのマーカーであるアディポネクチンの発現に有意な変化が見られなかったのに対し、M1マクロファージのマーカーであるIL-6及びTNF-αの発現はコントロールと比較して有意に上昇した。FSTL3が脂肪細胞における炎症性サイトカイン類の産生誘導を通じてインスリン抵抗性の惹起に関与していることが示唆された。(図1-2参照)
(1) アンチセンスオリゴヌクレオチドの投与
7週齢の雄性db/dbマウスを1週間馴化した後、下記配列のアンチセンスオリゴヌクレオチドを週に2回ずつ、合計8回腹腔内投与(計80mg/body)した。db/dbマウスは遺伝性肥満のモデルマウスであり、インスリン抵抗性を示すことが知られている。コントロールマウスと比較して、脂肪細胞においてFSTL3を高発現していることをノーザンブロット解析により確認した(図1-3参照)。

(2) リン酸化シグナル測定
アンチセンスオリゴヌクレオチド投与から31日後に解剖して肝臓及び骨格筋を摘出し、各部位におけるAktリン酸化の測定を行った。
(3) Aktのリン酸化測定
各々200mgの肝臓と骨格筋を1% Nonident-P 40を含むLiver Buffer(組成は表1)にてホモジナイズした。Liver Bufferを、肝臓は7mL、骨格筋は4mL使用した。3,000rpm、10分、4℃にて遠心を行い、上清をチューブに取り分けた。分取した上清を55,000rpm、1時間、4℃にて超遠心して、上清をピペットで吸ってチューブに取り分けた。タンパク定量したのち、10% ゲルでSDS-PAGEを行った。常法によりゲルから膜に転写したのちブロッキングを30分行い、2,000分の1に希釈した一次抗体(Phospho-Akt(Ser473)(cellsignaling, #92715))を室温にて一晩反応させた。5,000分の1に希釈した2次抗体(goat antiRabbit IgG HRP(SantaCruze, sc-2054))を室温で2時間反応させた。
(4)結果
FSTL3アンチセンスオリゴヌクレオチドを投与されたマウスにおいてはインスリン刺激により肝臓、骨格筋ともにコントロールを上回るAktのリン酸化が確認された。FSTL3はPI3K-Akt系におけるインスリンシグナル伝達の阻害を通じてインスリン抵抗性の惹起に関与していることが示唆された。(図1-4参照)
1.FSTL3の発現抑制による耐糖能の改善
(1)アンチセンスオリゴヌクレオチドの投与
アンチセンスオリゴヌクレオチドの投与は、上述の方法と同様に行った。
(2)グルコース負荷試験(GTT)
投与初日から28日後にGTTを行った。各試験とも上述の方法と同様に行った。
(3)結果
FSTL3アンチセンスオリゴヌクレオチドを投与したdb/dbマウスではコントロールdb/dbマウスと比較してグルコース負荷試験における血糖値上昇が顕著に抑制され、耐糖能が改善された。FSTL3の発現を抑制することでインスリン抵抗性を改善できることが示された。(図2-1参照)
(1)組換えアデノウイルス作製とマウスへの投与
全長マウスインヒビンβB cDNAを用いた以外は、組換えアデノウイルスの作製は上述の方法で行った。
(2)db/dbマウスへの組換えアデノウイルス投与
7週齢の雄性db/dbマウスを1週間馴化した後、5.0X1011 pfu/mLのアデノウイルスをPBS 150μLに溶解し、週に1度、2週間、尾静脈より投与した。2回目の投与後4日目にGTTを行った。
(3)12週間高脂肪食B6マウスへの組換えアデノウイルス投与
5週齢の雄性B6マウスを普通食下で1週間馴化した後、12週間高脂肪食を負荷した。5.0X1011pfu/mLのアデノウイルスをPBS 150μLに溶解し、18週目に入ったマウスに週に1度、2週間、尾静脈より投与した。2回目の投与後7日目にGTTを行った。高脂肪食負荷マウスは、肥満のモデルマウスであり、インスリン抵抗性を示すことが知られている。コントロールマウスと比較して、脂肪細胞においてFSTL3を高発現していることがノーザンブロット解析で確認された(図2-2参照) 。
(4)グルコース負荷試験(GTT)
両マウスともGTTは以下のように行った。試験前日より16時間絶食させたマウスの腹腔にグルコース1.0mg/g-BW(生理的食塩水で希釈)を投与した。採取した全血から血清を分離し、グルコーステストワコー(和光純薬工業社製)にてグルコース濃度を測定した。
(5)結果
インヒビンβB遺伝子を有するアデノウイルスを投与されたマウスではコントロールマウスと比較してグルコース投与直後の血糖値が有意に低かった。さらにコントロールマウスがグルコース投与120分後においても血糖値の降下はわずかであったのに対し、インヒビンβB遺伝子を有するアデノウイルスを投与されたマウスではグルコース投与後の最大血糖値の半分以下となった。FSTL3結合タンパク質であるアクチビンBを投与することで、インスリン抵抗性を改善できることが示された。(図2-3参照)
(1) 抗体投与
6週齢の雄性db/dbマウスを1週間馴化した後、抗FSTL3抗体あるいはコントロール抗体を30μg/匹、隔日、計5回腹腔内投与した。用いた抗体は下記のとおりである。
・抗FSTL3抗体
Anti FLAG, Mouse(Goat) (R&D System社)
・コントロール抗体
Goat IgG Control (R&D System社)
(2)インスリン負荷性試験(ITT)
抗体投与後、絶食せずに随時マウスの腹腔にインスリン3.0mU/g-BW(生理的食塩水で希釈)を投与した。採取した全血からグルテストセンサー(三和化学)を用いてグルコース濃度を測定した。
(3)結果
抗FSTL3抗体を投与されたマウスはコントロールマウスと比較してインスリン投与による血糖値が有意に低下した(図2-4参照、縦軸は血糖値(mg/dL)、横軸は時間(分)を表す)。抗FSTL3抗体投与によってインスリン抵抗性を改善することができることが示された。
Claims (10)
- FSTL3阻害剤を有効成分として含むインスリン抵抗性改善薬。
- FSTL3阻害剤が
(A) FSTL3と特異的に結合してFSTL3の機能を阻害又は抑制する物質、
(B) FSTL 3の発現に対する阻害物質、
(C) FSTL3の競合物質
のいずれかである、請求項1に記載のインスリン抵抗性改善薬。 - 請求項2に記載の(A)、(B)又は(C)、及び薬学的に許容される希釈剤又は担体を含む、インスリン抵抗性改善薬。
- FSTL3阻害用薬剤を製造するための、請求項2 に記載の(A)、(B)、(C)いずれか1つ以上の使用。
- 動物又はヒトにおけるインスリン抵抗性の処置方法に使用するための組成物を製造するための、請求項2に記載の(A)、(B)、(C)いずれか1つ以上の使用。
- 次の工程
(1) 試験化合物をFSTL3発現細胞に接触させる工程
(2) 試験化合物がFSTL3によるシグナル伝達阻害を改善するか否かを確認する工程
を含む、肥満あるいはインスリン抵抗性を予防又は治療するための化合物をスクリーニングする方法。 - 前記(A)FSTL3と特異的に結合してFSTL3の機能を阻害又は抑制する物質が抗体である、請求項2に記載のインスリン抵抗性改善薬。
- 前記(A)FSTL3と特異的に結合してFSTL3の機能を阻害又は抑制する物質がアクチビンA、アクチビンB又はアクチビンABである、請求項2に記載のインスリン抵抗性改善薬。
- 前記(B)FSTL3の発現に対する阻害物質がアンチセンスオリゴヌクレオチドである、請求項2に記載のインスリン抵抗性改善薬。
- FSTL3阻害剤を投与する工程を含む、インスリン抵抗性改善方法。
