Description
本發明關於一種抗體藥物複合體及包含其之抗癌用組成物。
組織因子(Tissue factor,TF)會在大多數人體癌細胞表現出來,臨床研究證實了其表現與惡性度為正相關。另外,由本發明人等製作出了以組織因子為標的的抗體,以及包含抗組織因子抗體與細胞傷害劑的抗體藥物複合體(ADC)(專利文獻1及2,並且非專利文獻1)。

[發明所欲解決的課題]
本發明目的為提供一種抗組織因子抗體,其係在生物體內發揮出抗腫瘤效果;及包含細胞傷害劑之抗體藥物複合體。
[用於解決課題的手段]
本發明人等為了達成前述目的反覆鑽研,製作出了一種ADC(以下亦稱為「hu1084-PEG12-MMAE」),其係在抗組織因子抗體上透過由馬來醯亞胺-聚乙二醇12-纈胺酸-瓜胺酸-對胺基苄醇所形成的連接子結合了細胞傷害劑的單甲基奧瑞他汀E(MMAE)。
此外還製作出了一種ADC(以下亦稱為「hu1084-MMAE」),其係在抗組織因子抗體上透過由馬來醯亞胺己醯基-纈胺酸-瓜胺酸-對胺基苄醇所形成的連接子結合了細胞傷害劑的MMAE。
另外還製作出了一種ADC(以下亦稱為「hu1084-Dxd」),其係在抗組織因子抗體上透過由馬來醯亞胺己醯基-甘胺酸-甘胺酸-***酸-甘胺酸所形成的連接子結合了細胞傷害劑Dxd(關於連接子及Dxd可參考專利文獻3)。
而且,對於這些ADC解析其對人體胰臟腺癌細胞株(BxPC-3、PSN-1、HPAF-II)的殺細胞效果,結果在任一種人體胰臟腺癌細胞之中,hu1084-PEG12-MMAE或hu1084-MMAE都表現出比hu1084-Dxd還高的殺細胞效果。尤其,對於PSN-1,hu1084-Dxd完全沒有表現出殺細胞效果。
這樣的結果提示了對於人體胰臟癌而言,含有抗組織因子抗體及MMAE的ADC是有效的。然而,使用移植了人體胰臟癌組織的無毛小鼠(PDX模型)來進行評估時,結果驚訝地發現,與前述在活體外的結果相反,在活體內hu1084-Dxd會對來自人體的胰臟癌組織表現出顯著的腫瘤增殖抑制效果,甚至還可觀察到腫瘤退縮,而完成了本發明。
亦即,本發明是關於一種ADC,其係Dxd透過由馬來醯亞胺己醯基-甘胺酸-甘胺酸-***酸-甘胺酸所形成的連接子結合於抗組織因子抗體,較具體而言,提供以下各項。
<1>一種複合體,其係結合於組織因子的抗體與由下述式表示的連接子及藥物結合而成。
<2>如<1>之複合體,其中前述結合於組織因子的抗體之解離速率常數kd為1×10-4
(1/s)以上,且結合速率常數ka為1×104
(1/Ms)以上。
<3>如<1>或<2>之複合體,其中前述結合於組織因子的抗體為保有重鏈可變區域與輕鏈可變區域的抗體,
該重鏈可變區域作為CDR1~3分別包含:
序列識別號:1~3所記載的胺基酸序列;或在序列識別號:1~3所記載的胺基酸序列的至少任一者之中,一個或數個胺基酸被取代、缺失、附加及/或***之胺基酸序列,
該輕鏈可變區域作為CDR1~3分別包含:
序列識別號:5~7所記載的胺基酸序列;或在序列識別號:5~7所記載的胺基酸序列的至少任一者之中,一個或數個胺基酸被取代、缺失、附加及/或***之胺基酸序列。
<4>如<1>或<2>之複合體,其中前述結合於組織因子的抗體為保有重鏈可變區域與輕鏈可變區域的抗體,
該重鏈可變區域包含:
序列識別號:4所記載的胺基酸序列;對比於序列識別號:4所記載的胺基酸序列有80%以上的相同性之胺基酸序列;或在序列識別號:4所記載的胺基酸序列的至少任一者之中,一個或數個胺基酸被取代、缺失、附加及/或***之胺基酸序列,
該輕鏈可變區域包含:
序列識別號:8所記載的胺基酸序列;對比於序列識別號:8所記載的胺基酸序列有80%以上的相同性之胺基酸序列;或在序列識別號:8所記載的胺基酸序列的至少任一者之中,一個或數個胺基酸被取代、缺失、附加及/或***之胺基酸序列。
<5>如<1>~<4>中任一項之複合體,其中前述連接子及藥物之平均每個抗體的結合數為3~8個。

<7>一種抗胰臟癌用組成物,其中包含如<1>~<5>中任一項之複合體。
另外,本發明還關於一種前述複合體之使用,其係用於製造抗癌用組成物(抗胰臟癌用組成物等);前述複合體之使用,其係用於抗癌用途;一種癌症治療方法,其係將有效量的前述複合體投予至對象。
此外,在本發明中,「組織因子(Tissue factor,TF)」是指血液凝固因子的其中一個,是亦被稱為F3、CD142、TFA、凝血因子III(coagulation factor III)、促凝血酶原激酶、組織血栓形成質的蛋白質。組織因子是膜貫通型糖蛋白質,已知是外因型血液凝固反應的起始因子。組織因子宜為人類組織因子。人類組織因子的胺基酸序列,例如,如NCBI Reference Sequence:NP_001984所揭示的內容。然而,基因的DNA序列會因為該變異等,在自然界中(亦即非人工)發生變異,被編碼於其中的蛋白質的胺基酸序列也會隨著被改變。所以,本發明所關連的「組織因子」,並沒有被指定為前述典型的胺基酸序列所形成的蛋白質,這種天然的變異體也被包括在本發明內。
「結合速率常數」(ka)及「解離速率常數」(kd),是兩分子間的結合解離反應的速率常數。分別如後述實施例所揭示般,可藉由例如利用表面電漿共振法(SPR)的測定來決定。抗體與抗原間的結合的SPR測定已為所周知,只要是業界人士,即可根據周知技術求得抗體的結合速率常數ka及解離速率常數kd。在SPR測定中,由以一定濃度讓分析物(analyte)流過的相(結合相)中RU的變化量可求得結合速率常數,然後由運轉緩衝液(Running Buffer)流過的相(解離相)中RU的變化量可求得解離常數。測定可藉由single-cycle kinetics來進行。另外,解析可藉由bivalent analysis來進行。對於實測到的SPR感應圖的近似曲線的曲線擬合,可藉由kinetic titration 1:1 interaction model來進行。曲線擬合的細節可參考Karlsson, R., Katsamba, P.S., Nordin, H., Pol, E. and Myszka, D.G. (2006)."Analyzing a kinetic titration series using affinity biosensors.", Anal. Biochem. 349(1):136-47。
抗體的結合速率常數ka及解離速率常數kd,另外還可例如使用由GE Healthcare公司販售的Biacore(註冊商標)等的SPR裝置,依照該製造商的使用手冊求得。較具體而言,SPR測定裝置中也搭載了計算ka及kd的程式,由SPR感應圖可計算出ka及kd。例如,使用Biacore T200 Evaluation Software來解析藉由Biacore(註冊商標)裝置所得到的SPR感應圖,採用Bivalent Analyte model作為Fitting model,導出Fitting curve,能夠以抗體或ADC的Kinetics parameter的形式計算出ka或kd、KD。
[發明之效果]
依據本發明,可提供一種抗組織因子抗體,其在生物體內發揮出抗腫瘤效果;及包含細胞傷害劑之抗體藥物複合體。
<抗體藥物複合體>
如後述實施例所揭示般,可知結合於組織因子的抗體與細胞傷害劑Dxd透過由馬來醯亞胺己醯基-甘胺酸-甘胺酸-***酸-甘胺酸所形成的連接子結合成的複合體(抗體藥物複合體、ADC),會在生物體內對於癌症表現出顯著的腫瘤增殖抑制效果,甚至表現出腫瘤退縮效果。所以,本發明提供了該抗體藥物複合體。
(結合於組織因子的抗體)
本發明中的「抗體」,包括免疫球蛋白所有的類型(IgG、IgE、IgM、IgD、IgA、IgY等)及亞型(IgG1、IgG2、IgG3、IgG4、IgA1、IgA2等)。「抗體」包括多株抗體、單株抗體,而且意思還包括抗體的功能片段型態。「多株抗體」是抗體調製物,含有對應不同的抗原決定區的不同抗體。「單株抗體」意指由實質上均勻的抗體的集合所得到的抗體(包括抗體片段)。與多株抗體相對地,單株抗體是辨識抗原上單一決定位的抗體。本發明之抗體宜為單株抗體。本發明之抗體,是由自然環境的成分分離出來及/或回收到的(亦即分離後的)抗體。
該抗體可藉由融合瘤法來製作、另外還可藉由重組DNA法來製作。
融合瘤法具代表性的例子,可列舉Kohler及Milstein的方法(Kohler&Milstein, Nature, 256:495(1975))。此方法中,細胞融合步驟所使用的抗體生產細胞,是經過抗原免疫(抗原為組織因子、其部分胜肽、於這些物質上融合了Fc蛋白質等的蛋白質、或表現出這些物質的細胞等)的動物(例如大鼠、小鼠、倉鼠、兔子、猴子、山羊)的脾臟細胞、淋巴節細胞、末梢血白血球等。由未經免疫的動物預先分離出上述細胞或淋巴球等,並在培養基中使其與抗原發生作用,藉此得到的抗體生產細胞亦可拿來使用。骨髓瘤細胞可使用周知的各種細胞株。抗體生產細胞及骨髓瘤細胞,只要這些細胞可融合,則可源自於不同動物,宜為源自於相同動物。融合瘤是藉由例如由經過抗原免疫的小鼠所得到的脾臟細胞與小鼠骨髓瘤細胞之間的細胞融合所產生,然後藉由篩選可得到能產生對組織因子有特異性的單株抗體的融合瘤。對應於組織因子的單株抗體,可藉由培養融合瘤,並且從投予了融合瘤的哺乳動物的腹水來取得。
重組DNA法,是將編碼了本發明之抗體的DNA由融合瘤或B細胞等來加以複製(Cloning),殖入適當的載體並導入宿主細胞(例如CHO細胞、HEK細胞等的哺乳類細胞株、大腸菌、酵母細胞、昆蟲細胞、植物細胞等),以重組抗體的形式來生產本發明之抗體的手段(例如P. J. Delves, Antibody Production:Essential Techniques, 1997 WILEY、P. Shepherd and C. Dean Monoclonal Antibodies, 2000 OXFORD UNIVERSITY PRESS、Vandamme A.M. et al., Eur. J. Biochem. 192:767-775(1990))。讓編碼了本發明之抗體的DNA表現出來方面,可分別將編碼重鏈或輕鏈的DNA殖入表現載體來讓宿主細胞轉形,或可將編碼重鏈及輕鏈的DNA殖入單一的表現載體來讓宿主細胞轉形(參考國際公開第94/11523號)。本發明之抗體,藉由培養上述宿主細胞,並由宿主細胞內或培養液分離純化,能夠以實質上純粹且均勻的形態取得。抗體的分離純化,可使用通常多肽純化所使用的方法。只要使用基因轉殖動物製作技術製作出殖入了抗體基因的基因轉殖動物(牛、山羊、綿羊、豬等),即可由該基因轉殖動物的乳汁大量取得來自抗體基因的單株抗體。
本發明之抗體只要能結合於組織因子即可,其來源、種類、形狀等並未受到特別限定。具體而言,包括了來自非人動物的抗體(例如大鼠抗體、小鼠抗體、駱駝抗體)、來自人類的抗體、嵌合體抗體、人源化抗體、及這些抗體的功能片段。在將本發明之抗體作為醫藥投予至人體的情況,從減少副作用的觀點看來,希望為嵌合體抗體或人源化抗體。
在本發明中「嵌合體抗體」,是指將某生物種的抗體的可變區域和不同於該生物種的抗體的恆定區域連結而成抗體。嵌合體抗體,可藉由例如從如上述般製作出的單株抗體的基因將能與抗原結合的抗體可變部(可變區域)切出,並與來自人體骨髓的抗體恆定部(恆定區域)基因結合,將其殖入表現載體,導入宿主並使其生產抗體而獲得(例如日本特開平8-280387號公報、美國專利第4816397號公報、美國專利第4816567號公報、美國專利第5807715號公報)。
嵌合體抗體的恆定區域,通常是使用來自人類的抗體的恆定區域。例如在重鏈方面,可利用Cγ1、Cγ2、Cγ3、Cγ4、Cμ、Cδ、Cα1、Cα2、及Cε作為恆定區域。另外,在輕鏈方面,可使用Cκ或Cλ作為恆定區域。這些恆定區域的胺基酸序列,以及將其編碼的鹼基序列已為所周知。另外,為了改善抗體本身的安定性或生產抗體的安定性,可讓來自人類的抗體的恆定區域中一個或數個胺基酸發生取代、缺失、附加及/或***。
在本發明中「人源化抗體」,是指將來自非人類(大鼠等)的抗體的抗原結合部位(CDR)的基因序列移植至來自人類的抗體基因(CDR-grafting)而成的抗體,其製作方法為重疊延伸PCR等,已為所周知(例如歐洲專利申請公開第239400號說明書、歐洲專利申請公開第125023號說明書、國際公開第90/07861號、國際公開第96/02576號)。抗體的可變區域,通常是由被四個骨架區域(FR)夾住的三個CDR所構成。CDR是實質上決定抗體的結合特異性的區域。CDR的胺基酸序列富有多樣性,而另一方面,構成FR的胺基酸序列,在具有不同結合特異性的抗體之間仍表現出高相同性的情形很多。因此一般而言,藉由CDR的移植,可將某抗體的結合特異性移植至其他抗體。另外,從維持CDR功能的觀點看來,來自非人類的CDR移植至人類FR時,可選擇與來自該非人動物的FR相同性高的人類FR。也就是說,CDR內的胺基酸,不僅會辨識抗原,還會與CDR附近的FR的胺基酸配位,也和維持CDR的環狀構造有關,因此以利用由和欲移植的CDR鄰接的FR的胺基酸序列相同性高的胺基酸序列所形成的人類FR為佳。
與來自非人動物的FR相同性高的周知人類FR的檢索,可利用例如可在網際網路上使用的抗體特化檢索系統(http://www.bioinf.org.uk/abysis/)來進行。為了和以此方式得到的人類FR的序列一致,亦可在來自非人類的抗體的CDR以外的序列中導入變異。或者,在可取得編碼了由檢索所得到的人類FR的胺基酸序列的基因(cDNA)的情況,亦可在該序列中導入來自非人類的CDR。變異的導入等,可使用核酸合成、部位特異性的變異誘發等該領域周知的技術來進行。
藉由定性或定量地測定評估以此方式製作出的人源化抗體在抗原上的結合活性,可適當地選擇出在透過CDR連結時該CDR會形成良好的抗原結合部位之來自人類的抗體的FR。另外還可因應必要依照Sato, K. et al., Cancer Res, 1993, 53, 851-856等所記載的方法來取代FR的胺基酸殘基,讓人源化抗體的CDR形成適當的抗原結合部位,進一步藉由測定評估取代該胺基酸後的變異型抗體對於抗原的結合活性,可選擇出具有所希望的性質的變異FR序列。
在本發明中抗體的「功能片段」,意指被稱為抗原結合片段,是抗體的一部分(部分片段),能夠特異性地辨識組織因子。具體而言,可列舉Fab、Fab’、F(ab’)2、可變區域片段(Fv)、雙硫鍵Fv、單鏈Fv(scFv)、sc(Fv)2、雙價抗體、多特異性抗體,及其聚合物等。
此處「Fab」意指由一個輕鏈及重鏈的一部分所形成的免疫球蛋白的一價抗原結合片段。可藉由抗體的木瓜酶消化,還有重組抗體的方法來獲得。「Fab’」包含了抗體的鉸鏈區域的一個或更多的半胱胺酸,會因為在重鏈CH1區的羧基末端附加一些殘基,而與Fab有所不同。「F(ab’)2」意指由兩者的輕鏈與兩者的重鏈部分所形成的免疫球蛋白的二價抗原結合片段。
「可變區域片段(Fv)」是具有完整的抗原辨識及結合部位的最小抗體片段。Fv是重鏈可變區域及輕鏈可變區域藉由非共價鍵穩固地連結成的二聚物。「單鏈Fv(scFv)」包含了抗體的重鏈可變區域及輕鏈可變區域,這些區域會存在於單一多肽鏈中。「sc(Fv)2」是將兩個重鏈可變區域及兩個輕鏈可變區域以連接子等結合而形成單鏈。「雙價抗體」是指具有兩個抗原結合部位的小抗體片段,該片段包含了在同一個多肽鏈之中結合於輕鏈可變區域的重鏈可變區域,並且各區域會與其他鏈的互補區域成對。「多特異性抗體」是對於至少兩個不同抗原具有結合特異性的單株抗體。例如,可藉由兩個重鏈為具有不同特異性的兩個免疫球蛋白重鏈/輕鏈對的同時表現來調製。
本發明之抗體,包括了其胺基酸序列經過修飾且沒有降低所希望的活性(對於抗原的結合活性等)的抗體。這種胺基酸序列變異體,例如可藉由在編碼後述人源化1084抗體的抗體鏈的DNA上導入變異,或可藉由胜肽合成來製作。這種修飾,包括了例如抗體的胺基酸序列內殘基的取代、缺失、附加及/或***。抗體的胺基酸序列被改變的部位,只要具有與改變前的抗體同等的活性,可為抗體的重鏈或輕鏈的恆定區域,另外還可為可變區域(FR及CDR)。CDR以外的胺基酸的改變,被認為對於與抗原的結合活性的影響相對較少,而現在,改變CDR的胺基酸以提高在抗原上的結合活性的抗體,其篩選手段已為所周知(PNAS, 102:8466-8471(2005)、Protein Engineering, Design&Selection, 21:485-493(2008)、國際公開第2002/ 051870號、J. Biol. Chem., 280:24880-24887 (2005)、Protein Engineering, Design&Selection, 21:345-351(2008)、MAbs. Mar-Apr;6(2):437-45 (2014))。另外,現在還可藉由利用統合計算化學系統等(例如Molecular operating Enviroment,加拿大CCG公司製)來模擬在抗原上的結合活性提高後的抗體(參考例如http://www.rsi.co.jp/kagaku/ cs/ccg/products/application/protein.html)。此外,如Protein Eng Des Sel. 2010 Aug;23(8):643-51所記載般,得知了重鏈可變區域的CDR1及輕鏈可變區域的CDR3在與抗原的結合活性方面並無關聯的例子,另外同樣地,Molecular Immunology 44:1075-1084(2007)報告了在大部分的抗體之中,輕鏈可變區域的CDR2和在抗原上的結合活性無關。像這樣,在抗體在抗原上的結合活性方面,即使重鏈可變區域及輕鏈可變區域各自的CDR1~3全部都不要,也可發揮出同等的活性。實際上,在Biochem Biophys Res Commun. 2003 Jul 18;307(1):198-205、J Mol Biol. 2004 Jul 9;340(3):525-42、J Mol Biol. 2003 Aug 29;331(5):1109-20中,報告了藉由保有原本抗體的至少1個CDR來維持在抗原上的結合活性的例子。然而,只要是業界人士,則為了維持或提升該抗體在抗原上的結合活性的目的,不僅是FR,CDR的胺基酸序列也可改變。
關於本發明之抗體,被改變的胺基酸數目宜為10個胺基酸以內(例如9個胺基酸以內、8個胺基酸以內、7個胺基酸以內、6個胺基酸以內),較佳為5個胺基酸以內(例如4個胺基酸以內),更佳為3個胺基酸以內(例如2個胺基酸以內、1個胺基酸)。胺基酸的改變宜為保守性取代。「保守性取代」,意指以具有化學上同樣的側鏈的其他胺基酸殘基來取代。具有化學上同樣的胺基酸側鏈的胺基酸殘基的群組,在本發明之所屬技術領域已為所周知。例如,能夠以酸性胺基酸(天門冬胺酸・麩胺酸)、鹼性胺基酸(離胺酸・精胺酸・組胺酸)、中性胺基酸來分類,中性胺基酸當中又分成具有烴鏈的胺基酸(甘胺酸・丙胺酸・纈胺酸・白胺酸・異白胺酸・脯胺酸)、具有羥基的胺基酸(絲胺酸・蘇胺酸)、含硫的胺基酸(半胱胺酸・甲硫胺酸)、具有醯胺基的胺基酸(天門冬醯胺・麩醯胺酸)、具有亞胺基的胺基酸(脯胺酸)、具有芳香族基的胺基酸(***酸・酪胺酸・色胺酸)。
另外,具有由改變後的胺基酸序列對比於後述人源化1084抗體的抗體鏈在胺基酸序列等級上有80%以上的相同性的胺基酸序列所形成的抗體鏈的抗體,只要具有與改變前的抗體同等的活性,則也被包括在本發明之抗體內。該相同性只要至少80%即可,宜為85%以上,較佳為90%以上,更佳為95%以上(例如96%以上、97%以上、98%以上、99%以上)。另外,序列的相同性,可利用BLASTP(amino acid level)的程式(Altschul et al. J. Mol. Biol., 215:403-410, 1990)來決定。該程式是以由Karlin及Altschul所開發的演算法BLAST(Proc. Natl. Acad. Sci. USA, 87:2264-2268, 1990, Proc. Natl. Acad. Sci. USA, 90:5873-5877, 1993)為基礎。在藉由BLASTP解析胺基酸序列的情況,參數定為例如score=50,word length=3。另外,在使用Gapped BLAST程式來解析胺基酸序列的情況,可如Altschul等(Nucleic Acids Res. 25:3389-3402, 1997)所記載的內容來進行。在使用BLAST與Gapped BLAST程式的情況,是使用各程式的預設參數。這些解析方法的具體手段已為所周知。另外,「具有同等的活性」,意指在抗原上的結合活性與對象抗體(代表性地來說,為人源化1084抗體)同等(例如70%以上,宜為80%以上,較佳為90%以上)。
另外,本發明之抗體的改變,可為例如改變了醣基化部位的數目或位置等的抗體在轉譯後程序的改變。藉此可提升例如抗體的ADCC活性。抗體的醣基化,典型來說為N-連接或O-連接。抗體的醣基化,會大幅依存於讓抗體表現出來所使用的宿主細胞。醣基化模式的改變,可藉由導入或去除和糖生產相關的特定酵素等的周知方法來進行(日本特開2008-113663號公報、美國專利第5047335號、美國專利第5510261號、美國專利第5278299號、國際公開第99/54342號)。此外,在本發明中,為了增加抗體的安定性等目的,可藉由將脫醯胺化的胺基酸或與脫醯胺化的胺基酸鄰接的胺基酸取代為其他胺基酸來抑制脫醯胺化。另外還可將麩胺酸取代成其他胺基酸來增加抗體的安定性。本發明也提供像這樣安定化的抗體。
另外,本發明之抗體可具有內化能力(可參考專利文獻1)。「內化」意指抗體與細胞表面的抗原形成免疫複合體之後,被吸收進到細胞內的現象。抗體是否具有內化能力,可藉由例如使結合了標識物質的抗體與表面表現出組織因子的細胞接觸,確認該標識物質是否轉移至細胞內的方法;使結合了具有細胞毒性的物質的抗體與表面表現出組織因子的細胞接觸後,確認細胞是否死亡或被誘導出細胞增殖阻礙的方法等來判斷。
本發明之抗體,另外還可具有選自ADCC活性及CDC活性的至少1種細胞障礙活性。「ADCC活性(抗體依存性細胞障礙活性)」,意指在抗體結合於標的細胞的細胞表面抗原時,藉由效應細胞(NK細胞、單核球等的免疫細胞)進一步結合於該抗體的Fc區域,該細胞會被活性化,伴隨於此,釋放出因子而將標的細胞殺傷的活性。另一方面,「CDC活性(補體依存性細胞障礙活性)」,意指抗體結合於標的細胞時,補體系統活性化,結果將標的細胞溶解的活性。
從一旦結合於組織因子之後迅速解離,成為ADC時,滲透至固體腫瘤內部的能力高,還有從抗腫瘤效果高的傾向的觀點看來(可參考專利文獻2),本發明所關連的抗體的解離速率常數kd以1×10-4

此外,相同性或胺基酸的改變個數如上述般,只要是業界人士,就能夠以在組織因子上的結合活性等作為指標來適當地調整。另外,可改變的部位也如上述般,只要是業界人士即可適當地選擇,理想的情況,可列舉大鼠1084抗體的CDRs與人源化1084抗體的CDRs當中胺基酸不同的部位,較具體而言,可列舉選自重鏈可變區域CDR2的N末端算起的第11~13、15及16位、輕鏈可變區域CDR1的N末端算起的第4位,以及輕鏈可變區域CDR2的N末端算起的第4位之中至少1個部位(例如1、2、3、4、5、6或7個部位)。另外,在這些部位上的胺基酸可為任意胺基酸(所謂Xaa)。
(連接子及藥物)
在本發明之ADC之中,上述抗體與由下述式表示的連接子及藥物會結合(CAS編號:1599440-13-7)。
本發明所關連的連接子,如上述般,是由馬來醯亞胺己醯基(MC,maleimidocaproyl)-甘胺酸(Gly)-甘胺酸(Gly)-***酸(Phe)-甘胺酸(Gly)所形成。另外,藥物是由下述式表示的抗腫瘤性化合物(Dxd,CAS編號:1599440-33-1)。
此外,已知Dxd具有喜樹鹼構造,因此在酸性的水性溶媒中(例如pH3左右),平衡會偏向形成內酯環的構造(閉環體),另一方面,在鹼性的水性溶媒中(例如pH10左右),平衡會偏向內酯環開環的構造(開環體)。導入了對應於這種閉環構造及開環構造的依沙替康(exatecan)殘基的ADC,也可期待同等的抗腫瘤效果,而應理解任一者皆被包含在本發明之範圍。
該連接子及藥物,只要是業界人士,就會使用周知的方法適當地合成。另外還能夠以市售品(例如MedChemExpress公司,製品型號:HY-13631E,商品名:Deruxtecan)的形式購買到。
本發明所關連的抗組織因子抗體與上述連接子及藥物的「結合」,如下述式所表示般,意指透過由該抗體中的硫醇基(SH基、硫氫基)與連接子末端的馬來醯亞胺基發生反應所形成的硫醚基的結合。此外,於下述式中,「mAb」表示本發明所關連的抗組織因子抗體,「Dxd」表示本發明所關連的藥物。
本發明之ADC,其每一抗體分子上的藥物結合數,需考慮其有效性、安全性,讓該數到達一定數目來設定反應的原料、試藥的使用量等的反應條件而實施。然而,與低分子化合物的化學反應不同,通常會以不同結合數的藥物組合而成的混合物的形式獲得。藥物在一個抗體分子上的結合數為平均值,亦即,以平均藥物結合數來指定以及表記。所以,在本發明說明書中,只要沒有特別註明,藥物的結合數也意指平均值。
每一抗體分子上的藥物結合數,如後述實施例所揭示般,可藉由調整所使用的還原劑的種類及其濃度、連接子及藥物相對於抗體的濃度(莫耳過剩率等),以及結合反應的溫度或時間來控制。「還原劑」,只要是可使抗體中的雙硫鍵還原性地裂解並發生上述透過硫醇基的結合,則並無特別限制,可列舉例如二硫蘇糖醇(DTT、DL-DTT)、2-巰基乙胺、巰基乙醇、TCEP。
在本發明中,每一抗體上的藥物平均結合數,宜為3~8個,較佳為4~8個,更佳為5~8個,較佳為6~8個,更佳為7~8個,特佳為8個。此外,每一抗體分子上的藥物結合數,如後述實施例所揭示般,只要是業界人士,即可使用DTNB等的以比色來定量硫醇基的試藥求得。
另外,本發明之ADC,會因為放置在大氣中或者進行再結晶時,吸收水分,而有帶著吸附的水或者變成水合物的情形,這種含水的化合物及鹽也被包括在本發明內。
構成本發明之ADC的原子當中的1種以上,也能夠以非天然的比例含有原子同位素。原子同位素可列舉例如2
H、3
H、125
I、14
C。另外,為了在生物影像等技術領域中進行偵測,本發明之ADC亦可與標識物質結合。標識物質可依照所使用的偵測方法等的種類適當地選擇,可列舉例如放射性標識物質、螢光性標識物質、順磁性標識物質、超順磁性標識物質、酵素標識物質。另外,這種標識物質與ADC的結合可為直接或間接。間接結合,可列舉透過結合有標識物質的二次抗體、結合有標識物質的聚合物(蛋白質A、蛋白質B等)的結合。
較具體而言,可利用正子發射核種作為標識ADC的放射性同位素。例如,可藉由如64
Cu、18
F、55
Co、66
Ga、68
Ga、76
Br、89
Zr及124
I般的正子發射核種來標識抗體。在以這些正子發射核種來標識ADC時,可利用周知的方法(Nucl Med Biol. 1999;26(8):943-50.、J Nucl Med. 2013;54(11):1869-75.等)。另外,被正子發射核種標識的ADC,在被投予至對象之後,藉由PET(正子斷層攝影裝置),由體外測量由該放射性核種發出的放射線,以電腦斷層掃描的手段轉換成影像。以此方式可追蹤投予至對象的ADC在腫瘤組織中的滲透等。
<抗癌用組成物>
如後述實施例所揭示般,本發明之ADC會在生物體內表現出顯著的腫瘤增殖抑制效果,甚至還可發揮出腫瘤退縮效果。所以,本發明提供一種用來治療癌症的組成物(抗癌用組成物),其係包含本發明之ADC。
本發明對象的「癌」,可列舉例如固體腫瘤,較具體而言,可列舉胰臟癌、淋巴腫、肺癌、頭頸部癌、***癌、膀胱癌、乳癌、食道癌、胃癌、大腸癌、子宮癌、卵巢癌、皮膚癌、甲狀腺癌、胸腺癌、腎臟癌、***癌、陰莖癌、肝臟癌、膽道癌、腦腫瘤、骨軟部腫瘤、後腹膜腫瘤、血管・***肉腫。另外,此癌可為原發性或轉移性。
本發明之組成物中,除了ADC之外,還可包含藥理學上容許的其他成分。這種其他成分,可列舉例如擔體、乳化劑、濕潤劑、pH緩衝化劑、賦形劑、崩壞劑、緩衝劑、等張化劑、懸浮劑、可溶化劑、無痛劑、安定劑、保存劑、防腐劑。較具體而言,藥理學上容許的其他成分,在注射劑等的液狀製劑的情況,可列舉水溶液(生理食鹽水、注射用水、磷酸鹽緩衝液、葡萄糖水溶液、甘油水溶液等)、氫氧化鋁等作為例子。另外,在冷凍乾燥製劑的情況,可列舉糖(甘露醇、乳糖、蔗糖等)、蛋白素等作為例子,然而並不受這些例子限定。此外,本發明之組成物可為在投予前可混合上述成分的套組態樣。另外,在作為注射劑使用的情況,可為被裝入注射筒的形態。
本發明之組成物,可僅包含本發明之ADC作為有效成分,或可包含該ADC及至少一個其他癌治療劑。本發明之ADC也可與其他癌症治療劑一起投予,藉由這種方式可增強抗癌效果。以此目的使用的其他抗癌劑,可分開或者連續地與本發明之ADC同時投予至對象,或可改變各投予間隔來投予。這種癌治療劑,可列舉例如免疫檢查點阻礙藥(抗PD-1抗體等)、吉西他濱(Gemcitabine)、S-1、厄洛替尼(Erlotinib)、5-FU、甲醯四氫葉酸(Leucovorin)、愛萊諾迪肯(Irinotecan)(CPT-11)、奧沙利鉑(Oxaliplatin)、亞伯杉(Abraxane)、卡鉑(Carboplatin)、紫杉醇(Paclitaxel)、培美曲塞(Pemetrexed)、索拉非尼(Sorafenib)、長春花鹼、LH-RH類似物(亮丙瑞林(Leuprorelin)、戈舍瑞林(Goserelin)等)、雌莫司汀磷酸酯(Estramustine phosphate)、***擷抗劑(泰莫西芬(Tamoxifen)、雷洛昔芬(Raloxifene)等)、芳香化酶阻礙劑(阿那曲唑(Anastrozole)、來曲唑(Letrozole)、依西美坦(Exemestane)等)、國際公開第WO2003/038043號所記載的藥劑,只要是具有抗腫瘤活性的藥劑則不受限定。
<癌症的治療方法>
本發明另外還提供一種治療癌症的方法,其係將有效量的本發明之ADC或包含該ADC的組成物投予至對象。
在該方法之中,所投予的ADC及組成物如上述般。被投予這些的「對象」,只要是以該治療為必要的人,則特別沒有限制,可為罹患上述癌症的患者,或有罹患顧慮的人,另外還可為有癌症復發顧慮的人。另外,對象可為人類或非人類哺乳動物。非人類哺乳動物並無特別限制,可列舉家畜、寵物、實驗用動物,較具體而言,可列舉猴子、小鼠、大鼠、倉鼠、豚鼠、牛、豬、馬、兔子、綿羊、山羊、貓、狗。
在本發明中「治療」,意指完全康復、治癒、緩解、改善症狀或狀態、降低進展(惡化)速度。另外,「有效量」意指讓該治療有效的藥劑量。
本發明之ADC或組成物的投予方法,會依照投予對象的年齡、體重、性別、健康狀態等而不同,能夠以非口服投予(例如靜脈內投予、動脈內投予、皮內投予、肌肉內投予、腹腔內投予、局部投予)、口服投予的任一種投予途徑來投予。合適的投予方法為非口服投予,較佳為靜脈內投予。投予可藉由例如注入或彈丸注射。
本發明之ADC或組成物的投予量,會依照對象的年齡、體重、性別、健康狀態、症狀發展程度及所投予的組成物的成分而變動,一般而言,在靜脈內投予的情況,以所含有的藥物量來換算,成人每1kg體重1天0.001~1000mg,宜為0.01~100mg。投予次數可為例如1次、或以1天~1年1次的間隔(例如每1週、每10~30天、每1月、每3~6月、每半年、每1年)投予多次。
[實施例]
以下基於實施例較具體地說明本發明,然而本發明並不受以下的實施例限定。
[抗組織因子抗體的調製]
在本實施例之中,由專利文獻2所記載的來自大鼠的融合瘤No.1084株來產生單株抗體(大鼠1084抗體),使其人源化,將其作為抗組織因子抗體來使用。
具體而言,首先依照Kabat的定義來決定大鼠1084抗體的各CDR區域的序列。所定出的大鼠1084抗體的各序列如以下所述。

接下來,依照Human Monoclonal Antibody (Springer Protocols)的Chapter 7(123-138)記載的方法,將大鼠1084抗體的骨架區域等替換為來自人類的骨架區域。人源化的1084抗體的各胺基酸序列如以下所述。
重鏈可變區域CDR1~3的胺基酸序列…序列識別號:1~3
重鏈可變區域的胺基酸序列…序列識別號:4
輕鏈可變區域CDR1~3的胺基酸序列…序列識別號:5~7

此外,人源化1084抗體與大鼠1084抗體會有一部分的CDR區域的序列不同。這是因為,該部分由於並未露出分子表面,因此並非與抗原辨識部位,而是與人源化的骨架區域發生交互作用,有助於構造安定化,所以採用了人類的序列,而並非大鼠的序列。
另外,前述可變區域以外的序列(恆定區域),使用了利妥昔單抗(由抗人類CD20人類・小鼠嵌合體抗體所形成的單株抗體,CAS登錄編號:174722-31-7,DrugBank:DB00073)的序列。分別將具有來自利妥昔單抗序列的人源化1084抗體的重鏈及輕鏈的胺基酸序列揭示於序列識別號:17及18。
然後,依照常法製作出編碼人源化1084抗體的載體,使用CHO細胞(製品名;ExpiCHO expression system(thermofisher公司製)),使該抗體一時性地表現出來,進行純化。
[抗體藥物複合體(ADC)的調製]
使用前述所製作出的人源化1084抗體,依照以下所揭示的方法調製出下述表1所揭示的4種ADC。
(hu1084-Dxd3的調製)
首先進行還原處理時使用的緩衝液的製作。調製出含5mM EDTA(pH8.0)的杜氏磷酸緩衝生理食鹽水(DPBS)作為緩衝液A。另外,調製出100mM磷酸二氫鈉、150mM氯化鈉、5mM EDTA(pH8.0)作為緩衝液B。
接下來,在含抗體的DPBS溶液中添加0.5M EDTA(pH8.0),並調整成最終EDTA濃度為5mM。然後,將最終抗體濃度成為1mg/mL的液量定為反應液總量,將其中1/6量的緩衝液B與抗體溶液混合,最後,以緩衝液A將最終抗體濃度調整成1mg/mL。
將該還原反應用混合液,使用恆溫槽在37℃下預保溫15分鐘。然後添加混合液總量的1/100的還原劑(2-巰乙基胺-HCl溶液,Sigma-aldrich公司製,終濃度20mM),使用恆溫槽在37℃下保溫30分鐘。
反應結束後,迅速將樣品移至冰上,使用VIVASPIN TURBO 15(Sartorius公司製,Gottingen, Germany),以緩衝液A進行緩衝液置換,進行還原劑的除去。緩衝液置換可重覆至將原本的反應液稀釋100萬倍以上。
接下來,將抗體溶液最終濃度成為0.5mg/mL的液量定為反應液總量,將其中1/6量的緩衝液B、含有被還原的抗體的抗體溶液、由變成自由態的硫醇基作計算的必要量的3倍莫耳量的連接子Dxd(MedChemExpress公司,製品型號:HY-13631E,商品名:Deruxtecan(曲妥珠單抗))混合,並藉由緩衝液A調整成抗體最終濃度為0.5mg/mL。將混合液藉由顛倒混合來和緩地攪拌,然後在4℃下靜置18小時。
此外,連接子Dxd是由下述式表示的化合物。
最後,再度使用VIVASPIN TURBO 15,以DPBS進行緩衝液置換。在此緩衝液置換的過程中,也可重覆至將原本的反應液稀釋100萬倍以上。
以此方式調製出的ADC,在無菌條件下使用PVDF膜0.22μm過濾單元來進行過濾處理,然後以滅菌後的DPBS調整成抗體濃度為2mg/mL,並進行分注,到使用時為止冷凍保存於-80℃。冷凍只有這一次,解凍後迅速使用於各實驗。
(hu1084-Dxd8的製作)
除了將還原反應用混合液的預保溫溫度定為26℃,將添加至該混合液的還原劑定為DL-DTT溶液(Sigma-aldrich公司製,終濃度35mM),並將該添加後的保溫溫度定為26℃之外,與前述(hu1084-Dxd3的調製)所記載的方法同樣地調製出ADC。
[hu1084-PEG12-MMAE3的製作]
除了使用下述連接子PEG12-MMAE(Mal-PEG12-vc-PABC-MMAE,日本國立研究開發法人理化學研究所的眞鍋史乃博士等人所提供)來代替連接子Dxd之外,與前述(hu1084-Dxd3的調製)所記載的方法同樣地調製出ADC。此外,連接子PEG12-MMAE是由下述式表示的化合物。
[hu1084-PEG12-MMAE8的製作]
除了使用前述連接子PEG12-MMAE來代替連接子Dxd之外,與前述(hu1084-Dxd8的製作)所記載的方法同樣地調製出ADC。
[hu1084-MMAE3的製作]
除了使用下述連接子MMAE(Mal-vc-PABC-MMAE,商品名:vcMMAE,製造商:MedChemExpress,型號:HY-15575)來代替連接子Dxd之外,與前述(hu1084-Dxd3的調製)所記載的方法同樣地調製出ADC。此外,連接子MMAE是由下述式表示的化合物。
[hu1084-MMAE8的製作]
除了使用前述連接子MMAE來代替連接子Dxd之外,與前述(hu1084-Dxd8的製作)所記載的方法同樣地調製出ADC。
此外,這些ADC中的連接子及藥物之平均每個抗體的結合數(DAR),是使用比色定量硫醇基的試藥DTNB(5,5’-dithiobis(2-nitrobenzoic acid))來進行。亦即,分別在利用還原劑進行抗體還原之後以及附加連接子化合物之後添加該試藥。然後,藉由測定吸光度(λmax=412nm)來定量自由態硫醇基。然後,根據前述還原後與附加後的自由態硫醇基數目計算出DAR。
[殺細胞效果]
對於上述調製出的ADC,依照以下方法來評估在活體外對人體胰臟腺癌細胞的殺細胞效果。
培養液是使用添加了10%牛胎兒血清及青黴素-鏈黴素-胺苄青黴素B(×100)懸浮液的RPMI-1640。將人體胰臟腺癌BxPC-3、PSN-1及HPAF-II分別調整成5000細胞/mL、2000細胞/mL及5000細胞/mL,以100μL/well接種至96孔盤,在CO2
保溫箱內(37℃)靜置一晩。
以培養液將各ADC、Dxd單劑及MMAE單劑調整成終濃度(MMAE及Dxd換算)0.1、0.3、1、3、10、30、100nM等,以100μL/well添加至各接種了人體胰臟腺癌的96孔盤後,在CO2
保溫箱內(37℃)靜置6天。然後,以抽吸器將含有藥劑的培養液除去,藉由Cell Counting Kit-8(同仁化學公司製),依照附屬的實驗手冊進行活細胞數的測定。試藥的反應時間定為3小時,吸光度的測定是藉由SpectraMax Paradigm(Molecular Devices公司製)來進行。將殺細胞效果的結果表示於圖1A~1E。
由圖1A所示的結果可知,MMAE、hu1084-PEG12-MMAE3、hu1084-PEG12-MMAE8、Dxd、hu1084-Dxd3、及hu1084-Dxd8對BxPC-3的IC50值分別為0.62±0.04 nM、0.05±0.00nM、0.05±0.00nM、2.58±0.26nM、0.27± 0.02nM、及0.20±0.01nM。
由圖1B所示的結果可知,MMAE、hu1084-PEG12-MMAE3、hu1084-PEG12-MMAE8、Dxd、hu1084-Dxd3、及hu1084-Dxd8對PSN-1的IC50值分別為0.73± 0.04nM、6.29±1.04nM、0.35±0.12nM、7.92±0.84nM、N.D.、及N.D.。
另外,由圖1C~1F所示的結果可知,在三種人體胰臟腺癌細胞的細胞株之中,游離態MMAE的IC50值比游離態Dxd低30~50倍。亦即,MMAE對這些人體胰臟腺癌細胞株的殺細胞效果顯著較高。另外,包含這些藥劑的各ADC之中,也是MMAE的殺細胞效果高了數倍。
如以上所述,在任一種人體胰臟腺癌細胞之中,由MMAE連接子(連接子PEG12-MMAE或連接子MMAE)及抗組織因子抗體所形成的複合體,皆表現出比由連接子Dxd及抗組織因子抗體所形成的複合體還高的殺細胞效果,尤其在hu1084-Dxd3及hu1084-Dxd8的情況,對於PSN-1求得IC50都沒有達成(皆為N.D.)。
[抗腫瘤效果]
(患者腫瘤組織移植(PDX)模型的製作)
接下來,對於各ADC,依照以下的方法評估在活體內對人體胰臟癌細胞的抗腫瘤效果。
在所有的實驗中,使用了馴化1週的5週齡無毛小鼠(BALB/c nu/nu,雌性,日本Charles River)。將3~5mm見方的人體胰臟腫瘤組織片(型號:PAN-02-JCK、PAN-04-JCK、PAN-08-JCK、PAN-14-JCK(實驗動物中央研究所))移植至各無毛小鼠的皮下,並將所形成的腫瘤定為Passage1(P1)。此外,各人體胰臟腫瘤組織來源不同(各罹患胰臟腫瘤的人)。將P1腫瘤切碎,修整成3~5mm見方,並移植至小鼠皮下,得到P2腫瘤。重覆此操作,進行腫瘤的繼代。抗腫瘤效果的檢討使用了P4~P6。
(免疫組織化學染色)
藉由過度麻醉讓小鼠安樂死之後,迅速將腫瘤組織摘出,包埋於冰凍切片包埋劑(OCT Compound)中,並在-80℃下冷凍保存。使用冷凍切片機將6μm的腫瘤組織薄切,貼附於載玻片上,並且風乾30分鐘。藉由4%多聚甲醛(PFA)-DPBS溶液在室溫下進行固定處理15分鐘,以DPBS洗淨後,以3%雙氧水甲醇溶液在室溫下處理30分鐘,進行內在性過氧化酶的去活化。然後以DPBS洗淨,並藉由5%脫脂牛乳-DPBS溶液在室溫下進行阻斷處理1小時。同樣地以DPBS洗淨,將來自山羊的抗組織因子多株抗體(R&D systems公司)以5%脫脂牛乳溶液進行1/500稀釋,並使其在室溫下反應1小時。以DPBS洗淨後,將HRP標識抗山羊多株抗體(MBL公司製)以5%脫脂牛乳溶液進行1/500稀釋,並使其在室溫下反應1小時。以DPBS洗淨後,使二胺基聯苯胺(DAB)在室溫下反應3分鐘,再度以DPBS洗淨,在室溫下利用蘇木精溶液進行核染色10分鐘。以流水洗滌10分鐘讓顏色分明,並利用乙醇及二甲苯來進行脫水、透明處理。最後,將蓋玻片蓋上,並使用Mount-Quick(封片膠)來進行組織的封入。觀察是使用了Virtual Slide System(Olympus公司製)來進行。將染色的結果表示於圖2A~2C、圖3D及3E。
圖2A~2C所示般,在胰臟癌PDX模型的三例(no,2、4及8)之中,確認了組織因子的表現。任一個模型皆觀察到一部分組織因子陰性的癌細胞集團,關於組織因子的表現,為不均勻的腫瘤。另外,如圖3E所示般,no,14也與前述三例同樣地,關於組織因子的表現,為不均勻的腫瘤。尤其胰臟癌PDX模型no,8及no,14,與no,2及4相比,組織因子的表現程度較低,而且其表現為較不均勻的腫瘤,並未表現出組織因子的癌在全體當中存在50%或其以上。
(使用PDX模型的治療實驗)
在PDX模型的平均腫瘤大小成為200~300mm3
的時間點,隨機進行小鼠的分組,在各投予組藉由尾靜脈投予開始治療實驗。將各ADC以10mg/kg每週投予1次,合計3次,並且每週進行1次腫瘤大小與體重的測定。腫瘤大小是使用電動游標尺來測定皮下腫瘤的長徑與短徑,並由「腫瘤的長徑×短徑×短徑×1/2」來計算。在腫瘤大小超過2000mm3
的時間點,或在腫瘤部觀察到潰瘍等的時間點,以人道終點來考量,則利用異氟醚的過度麻醉來進行小鼠的安樂死。N數定為3~4。另外還製作出投予DPBS來代替ADC的小鼠以作為陰性對照組。
如圖3A~C所示般,在任一個PDX模型之中,皆與上述在活體外的結果相反,在投予由連接子Dxd及抗組織因子抗體所形成的複合體的情況,觀察到腫瘤退縮或腫瘤增殖抑制效果。
另一方面,在投予由MMAE連接子(連接子PEG12-MMAE或連接子MMAE)及抗組織因子抗體所形成的複合體的情況,如圖3C所示,對移植了人體胰臟腫瘤組織片PAN-08-JCK的PDX模型no.8投予hu1084-PEG12-MMAE8時,觀察到腫瘤增殖抑制效果。另外,如圖3F所示,對移植了人體胰臟腫瘤組織片PAN-14-JCK的PDX模型no.14投予hu1084-MMAE3時,觀察到腫瘤增殖抑制效果。然而,其效果的程度,與投予由連接子Dxd及抗組織因子抗體所形成的複合體的情況相比為較差。
像這樣,判明了由抗組織因子抗體及連接子Dxd所形成的ADC,會在活體內對於癌症發揮出高腫瘤退縮或腫瘤增殖抑制效果。尤其令人驚訝地觀察到了一部分組織因子陰性的癌細胞集團,即使關於組織因子表現為不均勻的腫瘤,前述ADC也會表現出高腫瘤增殖抑制效果等。
發揮出這樣的效果的理由未必確定,而本發明人等推測如下。亦即,Dxd的疏水性高,在以ADC的形式攻擊了組織因子陽性的癌細胞之後,還會以游離狀態被釋放到細胞外,並通過鄰接的組織因子陰性癌細胞的細胞膜。然後,容易侵入細胞內,推測還可殺死該癌細胞(旁觀者效應)。在體內形成的固體癌幾乎在組織因子的表現方面都是異質的癌組織,由此強烈暗示了由抗組織因子抗體及連接子Dxd所形成的ADC的有效性。
此外,雖然圖中並未表示,在使用連接子MMAE的情況,與圖3A~3C所示的連接子PEG12-MMAE及連接子Dxd不同、DAR為3的情況,腫瘤增殖抑制效果比DAR為8的情況還高。另外,在所有的治療實驗中,並未觀察到ADC投予組有顯著的體重減少。
[表面電漿共振解析]
藉由表面電漿共振(SPR、製品名:BiaCore T200(GE healthcare公司製))來解析人源化1084抗體的解離常數及結合常數。
具體而言,首先將1mg/mL的重組人類組織因子抗原添加至10mM醋酸鈉液(pH5.0)中,並調整成最終濃度為20μg/mL。接下來,將contact time設定在120秒,在CM5感應晶片(GE healthcare公司製)上藉由胺偶合法使前述抗原固相化。另一方面,將人源化1084抗體以HBS-N溶液(GE healthcare公司製)進行溶液置換,將抗體濃度調整成320nM、160nM、80nM、40nM及20nM。然後,使用這些稀釋抗體,藉由single-cycle kinetics測定抗體的binding kinetics。此外,抗體的contact time與解離測定時間分別設定為120秒與600秒。感應圖的解析是藉由1:1 binding來進行。將所得到的結果揭示於表2。
如以上說明般,依據本發明,可提供一種抗組織因子抗體,其在生物體內發揮出抗腫瘤效果;及包含細胞傷害劑之抗體藥物複合體。所以,本發明在抗癌劑的開發及癌症的治療等方面是有用的。
[圖1A]為表示抗體藥物複合體及藥物單劑對人體胰臟腺癌細胞株(BxPC-3)的殺細胞效果的解析結果圖。圖中,縱軸代表人體胰臟腺癌細胞株的存活率,橫軸代表藥物在細胞株培養基的添加濃度(各藥物換算)(關於圖的縱軸及橫軸的表記,在以下的圖1B~1E中也同樣)。
[圖1B]為表示抗體藥物複合體及藥物單劑對人體胰臟腺癌細胞株(PSN-1)的殺細胞效果的解析結果圖。

[圖3E]為表示移植了人體胰臟癌組織(人體胰臟腫瘤組織片,型號:PAN-14-JCK(實驗動物中央研究所))的無毛小鼠(no,14)身上的抗體藥物複合體的抗腫瘤效果的解析結果圖。另外還一併表示了藉由使用抗組織因子抗體的免疫染色對前述無毛小鼠(no,14)身上形成的腫瘤組織解析的結果。該顯微鏡照片中的比例尺表示200μm。
Claims (7)
- 如請求項1之複合體,其中前述結合於組織因子的抗體之解離速率常數kd為1×10-4 (1/s)以上,且結合速率常數Ka為1×104 (1/Ms)以上。
- 如請求項1或2之複合體,其中 前述結合於組織因子的抗體為保有重鏈可變區域與輕鏈可變區域的抗體, 該重鏈可變區域作為CDR1~3分別包含: 序列識別號:1~3所記載的胺基酸序列;或在序列識別號:1~3所記載的胺基酸序列的至少任一者之中,一個或數個胺基酸被取代、缺失、附加及/或***之胺基酸序列, 該輕鏈可變區域作為CDR1~3分別包含: 序列識別號:5~7所記載的胺基酸序列;或在序列識別號:5~7所記載的胺基酸序列的至少任一者之中,一個或數個胺基酸被取代、缺失、附加及/或***之胺基酸序列。
- 如請求項1或2之複合體,其中 前述結合於組織因子的抗體為保有重鏈可變區域與輕鏈可變區域的抗體, 該重鏈可變區域包含: 序列識別號:4所記載的胺基酸序列;對比於序列識別號:4所記載的胺基酸序列有80%以上的相同性之胺基酸序列;或在序列識別號:4所記載的胺基酸序列的至少任一者之中,一個或數個胺基酸被取代、缺失、附加及/或***之胺基酸序列, 該輕鏈可變區域包含: 序列識別號:8所記載的胺基酸序列;對比於序列識別號:8所記載的胺基酸序列有80%以上的相同性之胺基酸序列;或在序列識別號:8所記載的胺基酸序列的至少任一者之中,一個或數個胺基酸被取代、缺失、附加及/或***之胺基酸序列。
- 如請求項1~4中任一項之複合體,其中前述連接子及藥物之平均每個抗體的結合數為3~8個。
- 一種抗癌用組成物,其中包含如請求項1~5中任一項之複合體。
- 一種抗胰臟癌用組成物,其中包含如請求項1~5中任一項之複合體。
