Description
Bicykliske forbindelser.
Den foreliggende oppfinnelse vedrorer nye bicykliske forbindelser med formelen
hvori R er lavere-alkyl
R 2 hydrogen, lavere-alkyl, aralkyl eller en
rest -(CH2)2R<5>;
R^ lavere-alkoksykarbonyl eller aryloksykarbonyl, og n er 1 eller 2.
Uttrykket lavere-alkyl - alene eller som bestanddel av komplekse grupper som lavere-alkoksykarbonyl - omfatter alifatiske rettkjedete eller forgrenede hydrokarbonrester med 1 til 7, fortrinnsvis med 1 til 4 karbonatomer. Eksempler på lavere-alkyl-grupper er metyl, etyl, isopropyl, tert.butyl, heptyl. Uttrykket aryl refererer seg til fenyl og til med halogen, nitro, lavere-alkyl eller lavere-alkoksy substituert fenyl.\ Eksempler på aralkylgrupper er benzyl, m-metoksybenzyl, fenetyl og m-metoksyfenetyl. Uttrykket lavere-alkoksykarbonyl omfatter grupper som karbometoksy og karboetoksy. Eksempler på lavere-alkoksy-grupper er metoksy, etoksy, propoksy, butoksy.
Forbindelsene ifolge oppfinnelsen kan fremstilles ved at et 1,3-cykloalkandion med den generelle formel .
cykliseres i et aprotisk losningsmiddel i nærvær av en optisk aktiv forbindelse, som inneholder en eventuelt forestret karboksylgruppe og/eller en hydroksylgruppe sammen med en primær eller sekundær aminogruppe, eller i nærvær av en aldolase.
Et vesentlig særtrekk ved fremgangsmåten er det ytterligere nærvær av optisk aktive forbindelser i reaksjonsmediet,
da derigjennom reaksjonens steriske forlop påvirkes. I nærvær av disse optisk aktive forbindelsene er det nemlig mulig å oppnå overveiende eller nesten utelukkende en av de to optisk aktive komponentene til sluttproduktet og ikke en racemisk blanding av de to stereoisomere formene.
Som det fremgår av strukturformel II kan det nukleofile angrepet til den gj ennom den oc-stående karbonylgruppen aktiverte metylengruppen i sidekjeden finne sted på begge ring-karbonylgruppene. Det har nå overraskende vist seg at under ringslutningsbetingelser i nærvær av en optisk aktiv forbindelse, finner angrepet overveiende bare sted på en av de kjemisk ekvivalente ring-karbonylgruppene, og derfor dominerer også i sluttproduktet en av de to stereoisomerene.
De bicykliske forbindelsene med formel I , særlig i optisk
aktiv form, er nyttige mellomprodukter for fremstillingen av steroider, med kjente verdifulle farmakologiske egenskaper og kan overfores i disse ved kjente metoder.
Som optisk aktive forbindelser i hvis nærvær cykliseringen gjennomfores, kommer eksempelvis enzymer eller andre optisk aktive organiske forbindelser på tale, som inneholder minst en sur og/eller basisk funksjon.
Egnede optisk aktive forbindelser med basiske funksjoner er f. eks. optisk aktive primære og sekundære aminer som oc-metylbenzylamin eller lavere-alkylester av prolin.
Da uorganiske som organiske baser kan katalysere dannelsen
av uonskede cykliske ketoler ved kondensasjon av kjede-karbonylgruppen med én metylengruppe i a-stilling til en ring-karbonylgruppe, kan det være nodvendig ved anvendelse av en organisk base å tilsette en syre som kompensasjon. Eksempler på for dette formål egnede syrer er karbonsyrene,
som eddiksyre eller propionsyre,eller uorganiske syrer som fosforsyre.
I en særlig foretrukket utforelsesform av fremgangsmåten for fremstilling av forbindelser ifolge oppfinnelsen arbeides i nærvær av en optisk aktiv forbindelse som besitter minst to funksjonelle grupper som en karboksyl og/eller en hydroksygruppe og en primær eller sekundær aminogruppe. Eksempler på slike forbindelser er a- eller p-aminosyrer som alanin, serin, treonin, valin, leucin, isoleucin, fenylalanin, tyrosin, L-azetidin-2-karbonsyre eller prolin, særlig (S) - (-.) -prolin, . og (S) - (-) -4-trans-hydroksy-prolin,
samt a- eller. (3-aminoalkoholer som (-)-efedrin eller (S) -.(-)-2-hydroksymetyl-pyrrolidin. Som ganske særlig egnet forbindelsesklasse må dog betraktes de sekundære a-amino-syrene, hvortil f.eks. (S)-(-)-prolinet horer. Som eksempel på et i denne sammenheng egnet enzym kan aldolasen nevnes.
Eksempler på aprotiske løsningsmidler er benzen, tetrahydro-furan, acetonitril og dimetylformamid. Her er det for å
oppnå et så hoyt optisk utbytte som mulig atter onskelig at los-ningsmidlet besitter en relativt stor polaritet. Ved å anvende aprotiske løsningsmidler oppnås eykliske sluttprodukter som
til en overveiende del består av ketoler med formel I og bare for en liten del av enoner med formel I '.
Ketolene med formel I kan lett gjennom dehydratisering ifolge kjente fremgangsmåter overfores i de tilsvarende enoner med formel I ', eksempelvis ved behandling med p-toluensulfonsyre i et inert organisk løsningsmiddel som benzen under tilbakelop.
Denne reaksjonen gjennomfores på egnet måte ved en temperatur fra -5 til +loo°C, fortrinnsvis ved 18 - 85°C. Det blir såvidt mulig arbeidet under inertgass-atmosfære, eksempelvis under kvelstoff eller en edelgass som helium eller argon. Generelt utgjor reaksjonstiden 3 timer til 3 uker, fortrinnsvis 16 timer til 6 dager. En forlengelse av reaksjonstiden ut over det nevnte tidsrom er på grunn av uonskede bireak-sjoner ikke fordelaktig.
Ved anvendelse av sekundære a-aminoalkoholer som optisk aktive forbindelser kan oksazolidiner isoleres som mellomprodukter. Det er meget sannsynlig at reaksjonsmekanismen forloper over forbindelser av enamin-typen. Således erholdes f.eks. ved cykliseringen av et triketon med formel
i nærvær av (-)-efedrin med formel en blanding av oksazolidinderivater med formlene V og VI , hvori en av diastereoisomerene (i dette tilfelle forbindelsen med formel V) er til stede i overskudd. Den folgende meka-nismen ble postulert for reaksjonen:
Oksazolidinderivatene med formel V og VI blir lett overfort
i de tilsvarende bicykliske diketoner med formel 1-2 gjennom hydrolyse. Hydrolysen gjennomfores på egnet måte med en vandig syre, som en vandig mineralsyre, f.eks. saltsyre, ved en temperatur på o til loo°C, fortrinnsvis ved lo til 4o°C.
På analog måte er postuleringen av en overgangstilstand ved anvendelse av en sekundær a-aminosyre som optisk aktiv forbindelse mulig. For cykliseringen av et triketon med formel II i nærvær av (S)-(-)-prolin som optisk aktiv komponent ble folgende overgangstilstand postulert:
Det skal fastholdes at i foranstående avbildede overgangstilstand må ved reaksjonen av (S)-(-)-prolin med triketonene formel II begge de vicinale hydrogenatomene i pyrrolidin-ringen av (S)-(-)-prolin sitte på den samme siden av 5-ringen, for samtidig å muliggjore hydrogenbrodannelse og oksazolidinon-ringslutning. Den rommelige anordningen til det optisk aktive molekylet er av stor viktighet, da derigjennom en meget stiv overgangstilstand tilveiebringes, hvori den hindrende oksa-zolidinonringen og vinkelstående substituenten står på mot- • satte sider av molekylet. På denne måten kan det hoye optiske utbyttet ved reaksjonen forklares godt, fordi en deltakelse av den andre karbonylgruppen i cykloalkandion-ringén ville fore til en sterisk hindret, ugunstig overgangstilstand.
På grunn av det overordentlig hoye optiske utbyttet som oppnås ved anvendelsen av optisk aktive proliner, er denne optisk aktive forbindelsen det foretrukne reagens. Dertil oppnås ved anvendelse av (S)-(-)-prolin i sammenheng med ketoner med formel II i overveiende mengde optisk aktive forbindelser med en slik absolutt konfigurasjon som er nodvendig for mellomprodukter for fremstilling av de fleste naturstoffer.
Det ble videre fastslått at ved anvendelse av de andre enantiomere av den optisk aktive forbindelsen erholdtes et speilbilde-opp-bygget sluttprodukt. Således forer f.eks. cykliseringen av forbindelser med formel II i nærvær av (S)-(-)-prolin til forbindelser med formel I, som oppviser den samme absolutte konfigurasjon som ble funnet i de fleste naturstoffer, d.v.s.
R og hydroksylgruppen, befinner seg i (3-stilling. Ved anvendelse av (R)-(+)-prolin som optisk aktiv forbindelse oppnås derimot som sluttprodukt forbindelser med formel I,
hvori R og hydroksylgruppen står i oc-stilling. De sist-
nevnte nye forbindelsene, fremfor alt de med formel I, egner seg som mellomprodukter for fremstillingen av terpener med verdifulle luktestoffegenskaper, d.v.s. de kan finne anvendelse som aromastoffer. Overforingen av forbindelser med formel I
i terpener kan lett skje ifolge den av Piers et al., Chem. Comm., lo69 (1969) angitte stereoselektive fremgangsmåte.
Utgangsforbindelsene med den generelle formel II kan fremstilles ved omsetning av en forbindelse med formel
2 hvori R har den ovenfor nevnte betydning,
med en forbindelse med formel
hvori R"1" og n har de ovenfor angitte betydninger.
Denne omsetningen gjennomfores hensiktsmessig i noytralt eller svakt surt vandig medium med en pH.på ca. 4 til 7, fortrinnsvis i avsaltet vann ved temperaturer på ca. 5 - 6o°C, fortrinnsvis ved romtemperatur.
Det er særlig viktig at den foranstående beskrevne omsetning av vinylketoner til forbindelser med formel II skjer i fravær av en base, da enolisering av ketogruppene i ringen finner sted i nærvær av baser og brodannede ketoler dannes.
De folgende eksempler belyser oppfinnelsen.
EKSEMPEL 1
En blanding av 91o mg 2-metyl-2-(3-oxobutyl) -1,3-cyklopentan-dion og 17,25 mg S-(-)-prolin ble rort 12 dager under utelukkelse av lys og argon ved 2o°C. Blandingen ble tatt opp i lo ml acetonitril og filtrert. Filtratet ble inndampet i vakuum til 921,2 mg av en olje, som ble lost i 25 ml etylacetat og ble kromatografert over 2 g kiselgel med ytterligere loo ml etylacetat. Det ble oppnådd 87o>6 mg av en olje som på grunnlag av IR- og UV-spektrene besto av ( + )-3a(3-hydroksy-7a|3-metyl-perhydroindan-1,5-dion og 29,6% (+)-7ap-metyl-5,6,7, 7a-tetrahydroindan-1,5-dion. Blandingen ble oppvarmet i 15 ml av en o,olN losning av p-toluensulfonsyre i benzen 15 minutter under nitrogen og anvendelse av en■Dean-Stark-vannutskiller med Linde-molekylarsi 1 type 4A under tilbakelop. Løsningen, som var avkjolt til romtemperatur,ble rort 5 minutter med 0. 3 ml IN vandig natriumbikarbonat-^losning, torket med magnesiumsulfat og filtrert under vakuum. Filtratet ble inndampet i vakuum og gav 782,2 mg (+)-7a(3-metyl-5, 6, 7, 7a-tetrahydroindan-1, 5-dion, [°GD= +282,92 (c = l,o i benzen). UV-maksimum ved 232 nm ( £. = 8, 87o) . Ved en beregnet optisk enhet på
77,3% og en UV-spektroskopisk.renhet på 8o,3%, gav dette et optisk utbytte på. 96,3%.
EKSE MPEL 2

dager under argon og utelukkelse av lys ved 2o - 23°C. S-(-)-prolin ble frafiltrert og filtratet inndampet i vakuum, blandet med benzen og inndampet igjen. Resten ble lost i 3o ml etylacetat og filtrert over 4 g kiselgel. Filtratet gav etter inndamping i vakuum 1,77 g (97,3%) rått (+)-3ap-hydroksy-7a(3-metyl-perhydroindan-l, 5-dion; [od^ = +64, o°
(c = 1 i kloroform). På grunnlag av UV-spektret inneholdt
dette produktet 3,7% (+)-7ap-metyl-5,6,7,7a-tetrahydroindan-1,5-dion. Dette gav som faktisk dreiningsverdi for 3-hydroksyforbindelsen [a]^5 = +53,75° (89,6% optisk renhet). Totalutbytte av optisk rent materiale: 76,37%, beregnet på
det rå sluttproduktet. Optisk rent materiale ble oppnådd ved omkrystallisasjon fra eter: L<a>Q^<5>= +60,4° (c = l,o6% i kloroform) .
EKSEMPEL 3
En blanding av 1,82 g 2-metyl-2-(3-oxobutyl)-1,3-cyklopentan-dion, lo ml vannfritt N,N-dimetylformamid og 34,5 g S-(-)-prolin ble rort under argon 2o timer ved 23°C. Etter filtrering ble filtratet inndampet under forminsket trykk ved 22°C (utbytte 2,4 g). Det ble opptatt med lo ml etylacetat og filtrert gjennom 8 g kiselgel. Opparbeidingen av filtratet gav 1,995 g (+)-3a(3-hydroksy-7a|3-metyl-perhydroindan-l, 5-dion i form av en olje som krystalliserte etter poding med autentisk materiale. Etter fjerning av siste spor av N,N-dimetylformamid i hoyvakuum' ved 45 o C erholdtes 1,828 g av et gul-brunt pulver, Calo 2 5 = +56,06 (c = 1 i kloroform); optisk renhet 93,4%..
En losning av 1,79 g av det erholdte råproduktet i 15 ml
o,ol N benzenisk p-toluensulfonsyre ble oppvarmet under nitrogen og anvendelse av en Dean-Stark-vannutskiller samt Linde-molekylarsil type 4A 15 minutter under tilbakelop. Etter av-kjoling av blandingen ble den rort 5 minutter med o,3 ml IN vandig natriumbikarbonat-losning, torket med magnesiumsulfat og etter filtrering inndampet under redusert trykk.
Det erholdtes 1,6 g (99,4%) rått (+)-7a(3-metyl-5, 6, 7, 7a-tetrahydroindan-1,5-dion i form av en olje som krystalliserte

etter poding med autentisk materiale; • £a] = +321,93
(c = o,935 i benzen);/(<Me>Oh=23o nm ( £, = lo2oo) . Dette tilsvarer 87,7% optisk renhet, 92,4% spektroskopisk renhet
(UV) og dermed et optisk utbytte på 94,94%. Totalutbyttet, beregnet på utgangsmaterialet, utgjor 99,4%. Utbyttet av optisk rent produkt, beregnet på utgangsmaterialet, utgjor 87,2%.
Ved rensing av råproduktet med eter erholdtes fargelose krystaller i et utbytte på 7o,2%,beregnet på utgangsmaterialet;
smp. 64 - 65,5°C; [a] ^5 = 356, o7° (c = o,993 i benzen).
Dette tilsvarer en optisk renhet på 97%.
EKSEMPEL 4
En blanding av 4,9 g 2-etyl-2-(3-oxobutyl)-1,3-cyklopentandion, o,86 g S-(-)-prolin og 25 ml vannfritt N/N-dimetylformamid ble rort 2o timer under argon ved 2o - 22°C, filtrert og inndampet under redusert trykk. Resten ble opptatt med lo ml etylacetat. Losningen ble filtrert gjennom 2o g kiselgel og gav etter inndamping under redusert trykk 4,83 g (98,6%)
rått ( + )-7a(3-etyl-3a(3-hydroksy-perhydroindan-l, 5-dion,
[a]D= +18,o9° (c = 1,15 i kloroform). Ved gjentatt rensing over 2o g kiselgel og krystallisasjon fra eter erholdtes 3,47 g (7o,95%) perhydroindandion med et smeltepunkt på
111,5 - 112,5°C. IR-maksimum: 362o, 335o - 355o, 1745 og 1725 cm \ ■(+)-7a|3-etyl-3a|3-hydroksy-perhydroindan-l, 5-dion ble, som beskrevet i eksempel 6, dehydratisert til (+)-7a(3-etyl-5, 6, 7, 7a-tetrahydroindan-l, 5-dion, F. 56 - 58,5°, [a]^<5>= +26o,29° (c = l,o2 i benzen). UV-maksimum i.metanol: 233 -^234 nm (£ = 1157o); IR-maksima: 1755 og 168o cm<-1>.
Utgangsmaterialet erholdtes som folger:
En blanding av 6,3 g 2-etyl-l,3-cyklopentandion, 8,3 ml metylvinylketon og 12 ml avsaltet vann ble rort 7 dager ved 2o°C under nitrogen og så filtrert. Filtratet ble ristet med 4o ml benzen og den dannede emulsjonen ble blandet til metning av den vandige fasen med fast natriumklorid. Etter atskillelse av den organiske fasen ble det vandige sjiktet ekstrahert to ganger med 2o ml benzen. Benzen-fåsene ble vasket med vann og mettet saltlosning, torket over magnesiumsulfat, filtrert og inndampet under redusert trykk. Det erholdtes 9,91 g av en olje, som ble destillert under redusert trykk og gav 8,o4 g rent 2-etyl-2-(3-oxobutyl)-1,3-cyklopentan-dion, kp. o,o35 99 - lol°,
EKSEMPEL 5
I analogi til eksempel 3 erholdtes ut fra 2-metyl-2-(3-oxo-pentyl)-1,3-cyklopentan-dion under anvendelse av 6 mol-%
S-(-)-prolin og N,N-dimetylformamid som løsningsmiddel ved 6o°C (+) -3a|3-hydroksy-4a, 7a(3-dimetyl-perhydroindan-l, 5-dion, F.
16o - 161°, [o]q5 +34, 6o° (c = l,o i kloroform).
Utgangsmaterialet erholdtes som folger:
En blanding av lo ml etylvinylketon, 88 mg hydrokinon-monometyl-eter, 11,2 g 2-metyl-l,3-cyklopentandion,0,08 ml iseddik og 26 ml avsaltet vann ble rort 15 dager under argon ved romtemperatur, etter at på 4. dag 9 ml og på 9. dag enda en gang 9,5 ml etylvinylketon var blitt tilsatt. Reaksjonsblandingen ble ristet med 5o ml benzen og den dannede emulsjonen ble blandet til metning av den vandige"fasen med fast natriumklorid. Den vandige fasen ble ekstrahert med 5o ml benzen.
De samlede benzenekstraktene ble behandlet med en blanding av natriumsulfat, magnesiumsulfat og aktivt kull og inndampet under redusert trykk. Det erholdtes 21,1 g av en olje, som ble fraksjonert destillert under et trykk på o,o5 mm Hg.
Den ved lo5 - 118°C kokende fraksjonen (17,47 g = 89%) ble
nok en gang destillert under et trykk på o,o25 mm Hg og gav 16 g (83,5%) 2-metyl-2-(3-oxopentyl)-1,3-cyklopentandion,
kp. o,o25 97 - 115°C.
EKSEMPEL 6
En blanding av 182 mg 2-metyl-2-,(3-oxobutyl)-1,3-cyklopentan-dion og 1 ml av en iskold suspensjon av aldolase fra kanin-muskel i 2,o ml ammoniumsulfatlosning ble rort 3 dager under argon. Etter tilsetning av 1 ml aldolase-suspensjon ble rort ytterligere 5 dager. Den erholdte losningen ble fortynnet med metanol og filtrert. Filtratet ble inndampet under redusert trykk og den olje-aktige resten renset tynn-sj ikt-kromatograf isk. Det erholdtes (+)-3a(3-hydroksy-7af3-metyl-perhydroindan-1,5-dion i en optisk renhet på 1,44%.
Materialet viste seg i IR-spektrum og tynnsjikt-kromatografisk opptreden identisk med en autentisk prove. '
EKSEMPEL 7
En blanding av 182 mg 2-mety1-2-(3-oxobutyl)-1,3-cyklopentan-dion, 165 mg (S)-(-)-fenylalanin og 1 ml 2-propanol ble rort 7 dager under argon ved 2o - 22°C og så filtrert. Filtratet ble inndampet under redusert trykk til 187,5 mg av en olje
som ble renset ved preparativ tynnsjikt-kromatografi på kiselgel med en blanding av benzen og etylacetat (1:1). Etter omkrystallisasjon fra eter erholdtes (+)-3a(3-hydroksy-7a(3-metyl-perhydroindan-1,5-dion i 36,9% utbytte; F. lo7,5 - lo9°;
[.ct<]>^ = +ll,6o° (c = 1,12 i kloroform), hvilket tilsvarer en optisk renhet på 19,33%.
EKSEMPEL 8
En blanding av 182 mg 2-metyl-2-(3-oxobutyl)-1,3-cyklopentan-dion, (2S)-(+)-2-hydroksymetylpyrrolidin og l,o ml acetonitril ble rort 72 timer under argon ved +2o°C og så inndampet under redusert trykk. Resten ble tatt opp i kloroform og losningen filtrert gjennom 2 g kiselgel. Etter behandling med aktivt kull og inndamping under redusert trykk erholdtes lo9 mg rått (+)-3ap-hydroksy-7a(3-metyl-perhydroindan-l, 5-dion,
F. 98 - lo2,5°, [a]^<5>= +lo,4° (c = l,o i kloroform), hvilket tilsvarer en optisk renhet på 13,45%.
EKSEMPEL 9
En blanding av 182 mg 2-metyl-2-(3-oxobutyl)-1,3-cyklopentan-dion, 131 mg (2S)-(-)-trans-4-hydroksy-prolin og 1 ml 2-propanol ble rort 26 dager under nitrogen ved 2o° og så filtrert. Inndamping av filtratet til torrhet og preparativ tynnsjikt-kromatografi gav (+)-3a|3-hydroksy-7a|3-metyl-perhydroindan-1,5-dion i 12,1% utbytte og med en optisk renhet på 73,1%.
EKSEMPEL lo
En blanding av 182 mg 2-metyl-2-(3-oxobutyl)-1,3-cyklopentan-dion, 1,15 g (R)-(+)-prolin og lo ml acetonitril ble rort under kvelstoff-atmosfære og utelukkelse av lys 14o timer ved romtemperatur, avkjolt i et isbad og filtrert. Filtratet ble inndampet ved 4o°C, og den etter tilsetning av lo ml benzen erholdte losningen inndampet nok en gang. Resten ble lost i 5o ml etylacetat, losningen filtrert over 2,o g kiselgel, filtratet behandlet med l,o g aktivt kull og inndampet under redusert trykk ved 4o°C. Det erholdtes 1,73 g (-)-3aa-hydroksy-7aa-metyl-perhydroindan-l,5-dion i form av en lysegul olje; WD = -.57,46 (c =l,o i kloroform). 1,73 g av det erholdte råproduktet ble lost i 15 ml benzen. Losningen ble etter tilsetning av 5o mg p-toluensulfonsyre oppvarmet 1 1/2 time i en Dean-Stark-vannutskiller under tilbakelop, avkjolt og så ekstrahert med 15 ml mettet natriumbikarbonat-losning, samt to ganger med 15 ml mettet natriumklorid-losning. Etter to gangers vask av de vandige fasene med hver gang lo ml benzen, ble den organiske fasen torket over magnesiumsulfat og etter filtrering inndampet under redusert trykk. Utbytte: 1,2 g av en gylden olje. Ut fra denne oljen erholdtes1 etter tilsetning av 6 ml eter og 2 ml heksan ved utfrysning 58o mg (35%) (-)-7aa-metyl-5,6,7,7a-tetrahydroindan-l,5-dion i form av hvite krystaller; F. 63,2 o ; [alD 2 5 = -37o,26 o (c o, 5 i benzen).
EKSEMPEL 11
3,o mg L(-)-azetidin-2-karbonsyre ble satt til en losning av 182 mg 2-mety1-2-(3-oxobutyl)-1,3-cyklopentandion i 1 ml acetonitril. Den ble gasset med argon og rort 6 dager ved 2o - 23°C. Etter filtrering fikk filtratet lope gjennom o,4 g kiselgel. Det ble ettervasket med 3o ml etylacetat, inndampet under redusert trykk, og 186 mg av et råprodukt erholdtes med [a]^ = +19,61° (c = l,o34% i kloroform). Tynnsjikt-kromatografisk rensing over kiselgel med 1:1 blanding av benzen-etylacetat gav 93 mg (51%) krystallinsk
r i 2 5 (+)-3a|3-hydroksy-7a(3-metyl-perhydroindan-l, 5-dion, LaJ^ = +38/55° (c = l,o36% i kloroform), hvilket tilsvarer en optisk renhet på 63,9%.
EKSEMPEL 12
En losning av 143 mg (S)-(-)-prollnetylester i o,5 ml acetonitril ble satt til en losning av 182 mg 2-metyl-2-(3-oxobutyl)-1,3-cyklopentandion i o,5 ml acetonitril.
Blandingen ble rort under argon 2o timer ved 2o - 23°C.
Ved direkte påfolgende tynnsjikt-kromatografi på kiselgel erholdtes 68 mg (41,5%) av en blanding av (+)-7a(3-metyl-5,6,7,7a-tetrahydroindan-l,5-dion og utgangsmateriale som olje; ta];;<5>= +13,6° (c = o, 994% i benzen) ; j<Me0H>= 232 nm
D max
(£ = 6,77o). Ved UV-spektroskopi ble et innhold av enon på 61% fastslått, hvilket tilsvarer et optisk utbytte på 6,o7%.

EKSEMPEL 14
I en til o°C avkjolt losning av 19,6 g 2-metyl-2-(3-oxobutyl)-1,3-cykloheksandion i loo ml vannfri N,N-dimetylformamid ble tilsatt 115 mg (S)-prolin i lopet av 3o minutter. Blandingen ble latt stå under nitrogen og utelukkelse av
lys ved 2o - 23°C. Etter 24 timer ble ytterligere 115 mg (S)-prolin tilsatt og en ytterligere tilsetning av 115 mg (S)-prolin fant sted etter 78 timer. Hoyvakuum-destillasjon gav etter en reaksjonstid på tilsammen 72 timer en rest
som ble lost i 4oo ml dietyleter. Etter behandling med 5,o g aktivt kull ble losningen filtrert gjennom 5,o g kiselgel.. Ut fra filtratet erholdtes etter 16 timer ved o°C
3,4 g (17,3%) rått, krystallinsk (-)-4a(3-hydroksy-8a|3-metyl-perhydro-naf talin-1,6-dion, [a] 25 = -19,83 o (c = 1,22% i kloroform), F. 131,5 - 141,5°C. Gjennom opparbeiding av moderluten erholdtes videre råprodukt. Totalutbytte: lo,24 g (52,1%), [<q>Q^<5>= -16'33°-Ved omkrystallisasjon fra eter erholdtes forbindelsen i optisk ren form, F. 134,5 - 135,5°C, [a]o^ = -21/97° (c = l,lol3%i kloroform), IR-maksima i kloroform ved 3625, 345o og 1725 cm<-1>. Det erholdte produktet ble dehydratisert etter den i eksempel 3 beskrevne metode til (+)-8a-metyl-l,2,3,4,6,7,8,8a-oktahydronaftalin-1,6-
dion, 1 al^5 = +75° (c = l,oo% i benzen),XMe0H = 24o nm
' L JD ' max ( £. = 946o). Dette tilsvarer en optisk renhet på 75% og en spektroskopisk renhet på 75,2% og dermed et optisk utbytte på 99,8%.
EKSEMPEL 15
En losning av 7,84 g 2-mety1-2-(3-oxobutyl)-1,3-cykloheksan-dion i 4o ml vannfritt N,N-dimetylformamid ble blandet med 7,84 g aktivt kull og rort under nitrogen. Etter tilsetning av 1,4 g (S)-prolin ble det rort ytterligere 8 dager ved 2o - 23°C. Blandingen ble så filtrert, filtratet ble med eter brakt til ca. 5oo ml og den eteriske losningen vasket tre ganger med fortynnet saltsyre.Eter-ekstraktet ble torket over vannfritt natriumsulfat, filtrert og inndampet under redusert trykk til 7,o9 g (9o,5%) av en olje som krystalliserte og var identisk med det konjugerte ketonet fra eksempel 14, I>]^<5>= +63,7o° (c = 1,185% i benzen), }<E>^°<H>- 243 - 244 nm (£ = lo,97o), IR-maksima i kloroform: 1715, 1665 og 162o cm" . Dette tilsvarer en optisk renhet på 63,7%, en spektroskopisk renhet på 87,2% og ifolge dette et optisk utbytte på 73,2%.
EKSEMPEL 16
En blanding av 177 mg (1)-2-metyl-2-(3-oxobutyl)-cyklopentanon, 12,5 mg (S)-prolin og l,o ml vannfritt N,N-dimetylformamid ble rort under argon 24 timer ved 2o - 23°C. Reaksjonsblandingen ble filtrert gjennom 1,6 g kiselgel. Det ble ettervasket med 5o ml etylacetat og filtratet inndampet under redusert trykk til 784 mg.
Produktet, som ennå inneholdt N,N-dimetylformamid, ble renset ved tynnsjikt-kromatografi på kiselgel. Det erholdtes 25,1 mg (3o% av teoretisk utbytte) (-)-7ap-metyl-3ap-hydroksy-perhydroindan-5-on som olje, [aj^<5>= -8,53° (c = 1,255% i kloroform), IR-maksima i kloroform ved 365o, 355o - 34oo og 171o cm\Ved siden av erholdtes lo6,2 mg (6o%) av en olje med La3D~+5,o8° (c = l,o62% i kloroform) ,. hvis Rf-verdi var identisk med utgangsmaterialets. Behandlingen av denne oljen i nærvær av (R)-prolin under de ovenfor beskrevne reaksjonsbetingelser og rensing ved tynnsjikt-kromatografi på kiselgel gav 19,9 g (38,4% av teoretisk utbytte) (+)-7aa-metyl-3aa-hydroksy-perhydroindan-5-on i form av en olje, ^cc].D 25 = +lo, 55o (c = o,995% i kloroform), IR-maksima i kloroform ved 3625, 3 5oo - 3 4oo og 17o8 cm-<1>.
EKSE MPEL 17
En blanding av 34,5 mg (S)-prolin, 316 mg 2-metyl-2-[6-(m-metoksyfenyl)-3-oxo-heksyl]-1,3-cyklopentandion og l,o ml vannfritt N,N-dimetylformamid ble rort under nitrogen 38 timer ved 6o°C. Blandingen ble filtrert, bg filtratet inndampet ved 25°C og et trykk på o,o5 mm Hg. Etter tynnsjikt-kromatografisk rensing på kiselgel erholdtes 51,2 mg (16,2%)
