Description
Foreliggende oppfinnelse vedrører kjemiluminiserende 1,2-dioksetan derivater som kan bli enzymatisk aktivert for å nedbryte og gjennom nedbrytning frigjøre lys. Dioksetaner er spesielt kjennetegnet ved tilstedeværelse av en aromatisk (fenyl eller naftyl) ring bundet til dioksetanet, i det ringen bærer en meta-substituert eller atskilt enzymatisk spaltbar gruppe som ved spalting forlater fenoksyanionet eller naftyloksyanionet til dioksan, og i fjerde eller femte posisjon når det gjelder fenyl, for eksempel, en elektron-donerende eller elektrontiltrekkende gruppe. Ved å velge identiteten på substituenten i fjerde eller femte posisjon (Z delen) kan spesielle aspekter av kjemiluminescens egenskapene til dioksetan, inkludert halveringstiden, kvantumutbytte, S/N forholdet osv bli endret.
1,2-dioksetan enzym substrater er vel etablert som meget effektive kjemiluminiserende reporter molekyler for anvendelse i enzym immunoanalyser og nukleinsyre probeanalyser av mange forskjellige typer. Disse analysene utgjør et foretrukket alternativ i forhold til konvensjonelle analyser som er basert på radioisotoper, fluoroforer, kompliserte farveskift, sekundære reaksjoner og lignende. Dioksetaner utviklet for dette formålet innbefatter de som er beskrevet i US-PS 4.978.614 samt US-PS 5.112.960. US-PS 4.978.614 beskriver blant andre 3-(2'-spiroadamantan)4-metoksy-4-(3''-fosforyloksy)fenyl-l,2-dioksetan, som har fått omfattende oppmerksomhet over hele verden og som er kommersielt tilgjengelig under varemerket AMPPD. US-PS 5.112.960 beskriver forbindelser hvor den adamantylstabiliserende ringen er substituert med enten brohodeposisjonen, med forskjellige substituenter, inkludert hydroksy, halogen og lignende, som omdanner den ellers statiske eller passive adamantyl stabiliserings gruppen til en aktiv gruppe som er involvert i kinetikken ved dekomponering av dioksetanringen. Forbindelsene av denne typen har likeledes mottatt inter-nasjonal oppmerksomhet og har gitt et raskere og sterkere signal enn AMPPD i mange anvendelser. CSPD er et spiroada-
mantylfenylfosfatdioksetan som bærer en klorsubstituent på adamtyl gruppen og som AMPPD er tilgjengelig fra Tropix, Inc. of Bedford, Mass.
Forbindelsene av denne typen er spesielt blitt utviklet for forsterket sensitivitet i analyser for tilstedeværelse av analyter i konsentrasjoner som er så lave som IO"<12> M og lavere. I visse anvendelser blir forbindelsene av denne typen anvendt sammen med forsterkere for å detektere analyter i konsentrasjon på 10_<12>M eller lavere. Disse forsterknings-mldlene, som innbefatter naturlige og syntetiske vann-oppløselige makromolekyler, er beskrevet i detalj i US-PS 5.145.772. Foretrukne forsterkningsmidler innbefatter vann-oppløselige polymeriske kvarternære ammoniumsalter, så som poly(vinylbenzyltrimetylammoniumklorid) (TMQ), poly(vinyl-benzyltributylammoniumklorid) (TMQ) og poly(vinylbenzyldi-metylbenzylammoniumklorid) (BDMQ).
Disse forsterkningsmidlene forbedrer kjemiluminiscenssignalet til dioksetan reportermolekylene ved å tilveiebringe et hydrofobt miljø hvor dioksetan er utskilt. Vann er uungåelig i de fleste analysene, og på grunn av anvendelse av kropps-fluider, er vann en naturlig "slukker" av dioksetankjemilumi-nescensen. Forsterkningsmolekylene ekskluderer sannsynligvis vann fra mikromiljøet hvori dioksetanmolekylene, eller der hvor i det minste utstrålingsartene i eksitert tilstand er, som resulterer i forsterket kjemiluminescens. Andre effekter assosiert med enhancer-dioksetaninteraksjonen kan også bidra med kjemiluminescens forsterkningen.
Ytterligere fordeler kan sikres ved anvendelse av valgte membraner, inkludert nylonmembraner og behandlet nitrocellulose, som tilveiebringer en lignende hydrofob overflate for membran-baserte analyser, og andre membraner belagt med de beskrevne forsterknings-type ("enhancer-type") polymerene som er beskrevet.
Det er fortsatt et generelt mål innen industrien å forbredre ytelsen til disse stabiliserte, kjemiluminescensdioksetan-. reportermolekylene for å forbedre maskinens avlesningsevne, sensitivitet og ytelsesaspektene til immunoanalysene avhengig av kjemiluminescenssignalet som blir frigjort av dioksetanene.
Som bakgrunn og som beskrevet i alle de refererte patentene blir enzymatisk-aktiverte dioksetaner anvendt som reportermolekyler, som substratet for enzymer som spalter den enzym-labile gruppen bundet til en aromatisk substituent på dioksetanringen. Enzymet, for eksempel alkalisk fosfatase er dermed tilstede alene eller er kovalent koblet eller på annen måte kompleksbundet med enten et antigen eller antistoff, i konvensjonelle antigen/antistoff ligand bindingsanalyser, eller en nukleinsyreprobe i nukleinsyre-analyser. Det enzym-bærende antigenet eller antistoffet, eller nukleinsyreproben, blir deretter blandet sammen med analyten som antas å inneholde målantigenet, eller nuklein-syresekvensen, under betingelser som muliggjør kompleks-binding eller hybridisering mellom antigen/antistoff eller probe/nukleinsyresekvensen. Etter bortvasking eller ut-separering av det totale ikke-kompleksbundede eller ikke-hybridiserte materialet blir dioksetansubstratet tilsatt. Dersom den antatte analyten er tilstede vil enzymet spalte den enzym-labile gruppen på den aromatiske substituenten på dioksetan, for eksempel fenyl eller naftyl, som tilveiebringer fenoksy eller naftyloksyanion mellomproduktet. Dette anionet nedbrytes, ved elektronoverføring gjennom den aromatiske ringen, spaltning av dioksetanringen og tilveiebringer to karbonyl-baserte produkter. Spaltnings-/nedbrytningsheneldsen er den lys-frigjørende heneldsen.
For å automatisere kliniske analyser og for å tilveiebringe vesentlig gjennomkjøring er kontinuerlig reduksjon i halveringstid, eller ^i/z"til dioksetan, samt en reduksjon i tidsmengden nødvendig for å nå maksimal emissjon av lys fra reporter molekylet, er ønskelig. På samme tid for å detektere analyter i ekstreme lave konsentrasjoner, under for eksempel omtrent 10_<12>M er det ønskelig å forbedre intensiteten til signalet fra dioksetan reportermolekylet og samtidig ønskelig å unngå og øke bakgrunnsstøyen forårsaket av ikke-enzymatisk-indusert lysfrigjøring for å forbedre den totale sensi-tiviteten til analysen. Ytterligere forbedringer i kjemiluminescens dioksetan reportermolekyler er ønskelig.
Ovennevnte mål og andre blir oppfylt av en ny klasse dioksetaner som er spesielt kjennetegnet ved en substituent på den aromatiske ringen bundet til dioksetan, i tillegg til den metasubstituerte enzym-labile gruppen. Det nye dioksetan ifølge denne oppfinnelsen har formelen
kjennetegnet ved at Y<1> og Y<2> uavhengig er H eller et halogen, hvor R er C1-C12 alkyl,
hvor X er en enzym-labil gruppe valgt fra gruppen bestående av en fosfat,
hvor Z er et halogen eller metoksy og okkuperer 4- eller 5-posisjonen på fenylringen.
Ved å velge den bestemte identiteten og beliggenheten til Z, som en elektron-tiltrekkende eller en elektron-donerénde gruppe, kan spesifikke karaktertrekk ved kjemiluminescens atferden til dioksetan, inkludert tl/2-kjemiluminescens halveringstid, tid til maksimal emissjon, maksimal emissjons-bølgelengde og kjemiluminescens signal intensitet bli påvirket.
Foreliggende oppfinnelse vedrører et kit for å utføre en analyse ved anvendelse av et kjemiluminescens dioksetan reportermolekyl, kjennetegnet ved at det omfatter dioksetan som tidligere nevnt, og et enzym med evne for spalting, i vandig oppløsning, del X til nevnte dioksetan.
Foreliggende oppfinnelse vedrører også en fremgangsmåte for å detektere tilstedeværelsen av et enzym i en prøve, kjennetegnet ved å kontakte nevnte prøve med et dioksetan som tidligere nevnte, og detektere frigjøring av lys forårsaket derved, idet nevnte enzym spalter nevnte enzym-labile gruppe X og hvor deteksjonen av lys som blir frigjort indikerer tilstedeværelse av nevnte enzym i en prøve.
Videre vedrører oppfinnenlsen en fremgangsmåte for å detektere et første medlem av et spesifikt bindingspar som har første og andre medlemmer, kjennetegnet ved å optisk detektere kjemiluminescens produsert ved omsetning av et dioksetan som nevnt ovenfor, med et enzym som spalter nevnte enzym-labile gruppe X til nevnte dioksetan, hvor nevnte enzym er kompleksbundet med et andre medlem av nevnte spesifikke bindingspar.
Kort beskrivelse av tegningene;
Figur 1 og 2 sammenligner ytelsen til dinatrium 3(4-metoksy-spiro[l,2-dioksetan-3,2'-(5'-klor)tricyklo[3.3.1.I<3>•<7>]dekan]-4-yl)fenylfosfatdioksetan (CSPC) med en forbindelse ifølge denne oppfinnelsen dinatrium-2-klor-5-(4-metoksyspiro[l,2-dioksetan-2 , 3 * ( 5 ' -klor - ) tr i cyklo-[3 . 3.1.13 •7]-dekan]-4yl )-fenylfosfat hvor fenyldelen bærer en klorsubstituent i 4 posisjonen (CDP-Star). Figur 2 reflekterer tilstedeværelse av en kjmiluminescensforsterker, polyvinylbenzyltributyl-ammoniumklorid. Figur 3 er en sammenligning mellom CSPD og CDP-Star på en nylonmembrananalyse for biotinylert pBR322-35mer.
Figurene 4 og 5 er reproduksjoner av Kodak XAR-5 film eksponeringer av western blot analyser utført på nylon og PVDF membraner som sammenligner CSPD og CDP-Star.
Figurene 6, 7 og 8 er reproduksjoner av røntgenfilmkontrast CSPD og CDP-Star inkubasjoner i 10 minutter, 70 minutter og 19 timer, til en analyse utført på nylonmembraner for gjaergenet RPB1. Figur 9 er en reproduksjon av røntgenfilm eksponeringer som reflekterer kjemiluminescens deteksjon av DNA sekvens stige utført på nylonmembraner contrasting CSPD og CDP-Star.
Figurene 10 og 11 er elektrofotografiske duplikasjoner av røntgenfilmbiIder av DNA sekvensering oppnådd ved anvendelse av dioksetaner i den krevde oppfinnelsen. Disse blir sammenlignet mot den kommersielle standarden, CSPD.
Figurene 12-21 er elektrofotografiske duplikasjoner av dot blot analyseresultatene på membraner som vist ved anvendelse av dioksetaner ifølge den krevde oppfinnelsen, dioksetaner utenfor rammen av den krevde oppfinnelsen og de kommersielle standardene til CSPD og AMPPD. Membranen som disse analysene ble utført på er angitt i figurene.
Dioksetan ifølge denne oppfinnelsen er kritisk karakterisert av substituentene på den aromatiske ringen koblet til dioksetanene, i det ringen bestemmer elektronoverføringen i aryloksyanionet som fører til nedbrytning og kjemiluminescens. Fenyldioksetanet ifølge oppfinnelsen har dermed følgende og generelle struktur (I).
Adamantyl-stabiliserte dioksetaner ifølge den krevde oppfinnelsen bærer dermed to substituenter på fenylringen i tillegg til koblingspunktet til dioksetan, samt 0, 1 eller 2 ikke-hydrogen substituenter på adamantylringen. Disse substituentene karakteriserer kritisk de elektroniske karaktertrekkene til dioksetan, oksyanionet og dekompo-neringsatferden. Identitetene til hver substituent er angitt nedenfor.
R er Cl-C12-alkyl. R er fortrinnsvis C1-C4 alkyl. Identiteten til R kan bli optimalisert med hensyn på oppløselighet hvor uvanlige analyter eller buffere kan gi bestemte problemer. Ever Y<1> og Y<2> representerer individuelt og uavhengig hydrogen eller et halogen. Foretrukne identiteter til en av Y<1> og Y<2 >er klor, hvor den andre er hydrogen.
X er en enzym-spaltbar del. Ved riktig kontakt med et egnet enzym blir X spaltet fra molekylet og etterlater oksygenet koblet til fenylringen og dermed fenoksyanionet. X er fosfat. Det er viktig å bemerke at når substituert på fenylringen er OX meta med hensyn på koblingspunktet til dioksetanringen, dvs. okkuperer tre posisjonen.
Z kan enten okkupere fire eller fem posisjon. Z er en elektron-aktiv substituent, karakteren til de elektron-aktive artene (elektron-donerende eller elektron-tiltrekkende), som optimaliserer forskjellige aspekter av dioksetandelen. Som et eksempel kan en elektron-donerende gruppe, så som en metoksygruppe forsterke dioksetanfenoksyanion dekomponerings-prosessen ved å lette overførbarheten til de fri ektronene fra den aromatiske ring 0~ donorgruppen, til dioksetanringen. I kontrast til dette kan en elektron-tiltrekkende gruppe redusere eller ødelegge evnen til å overføre frie elektroner til dioksetan og dermed gjøre dekomponeringsreaksjonen og lysemissjonen langsommere til tross for at det dermed gir et lyssignal med høyere intensitet. Dette bør stå i kontrast til innvirkningen av den elektron-tiltrekkende (eng. "with-drawing") substituenten på adamantyl gruppen, så som klor, som vesentlig aksellererer lysemissjonen og sterkt reduserer "tl/2- Av overraskende betydning er det faktumet at substitu-sjonen i seks posisjonen er spesielt lite ønsket. Slike seks-substituerte fenyldioksetaner utviser ekstraordinær hurtig dekomponeringskinetikk og nesten ikke noe lysemissjon. Uten å være begrenset til denne teorien antas det at denne atferden er forårsaket av steriske betraktninger, dvs. orto substituenten "dreier" fenylringen slik at det destabiliserer dioksetanringen (destabilisering gjennom steriske krefter, ikke elektronoverføring) og en substituent i seks-posisjonen for eksempel metoksy, deltar ikke i elektronoverføringen. Som diskutert nedenfor gir eksperimenter som involverer 6-substituerte fenyldioksetaner vesentlig ikke noe signal.
Som angitt ovenfor kan Z være et halogen eller metoksy. Foretrukne elektron-aktive substituenter innbefatter klor, metyl eller -OCH3.
Dioksetan med type beskrevet ovenfor, uten innbefattelse av Z substituenten, som tidligere angitt, er beskrevet i patenter som hører hertil. Patenter vedrørende dioksetan av denne typen uten innbefatning av Y og Z substituentene er også blitt gitt til Wayne State University, så som 4.962.192. Substitusjon av Z substituenten på dioksetaner krevde utvikling av syntese av tri-substituerte fenylfosfonater som er beskrevet nedenfor, under tittelen nye tri-substituerte fenyl 1,2-dioksetanfosfater. Den samme generelle synteseveien kan bli anvendt for naftyldioksetaner innbefattet heri tatt i betraktning av substitusjonsmønstrene som er nød-vendige som beskrevet ovenfor. Syntese av disse forbindelsene ved hjelp av veien beskrevet nedenfor innbefatter frem-stilling av nye tri-substituerte benzener. Som beskrevet nedenfor innbefatter en eksempelvis forbindelse involvert i syntese av dioksetaner fra denne klassen 3-klor-5-metoksy-benzaldehyd. Disse tri-substituerte forbindelsene utgjøre viktige mellomprodukter i forskjellige syntetiske reak-sjonsveier, i det 1,3,5-substitusjonsmønsterert er et generelt foretrukket og meget anvendbart mønster. Disse mellomproduktene er aldri tidligere blitt fremstilt og er i synteseveien beskrevet nedenfor markert med en stjerne.
Blant andre forbindelser innenfor strukturen ifølge formel I har en forbindelse følgende struktur:
Navnet på denne forbindelsen er dinatirum 2-klor-5-(4-metoksy-spiro[1.2-dioksetan-3,2'-(5'klor-)tricyklo-[3.3.1.l<3-7>]dekan]-4-yl)-l-fenylfosfat.
Denne forbindelsen blir generelt referert til som CDP-Star. Den kan bli fremstilt som vist i følgende reaksjonsskjema.
4- klor- 3- metoksybenzaldehyddimetylacetal 3
En heterogen blanding av metanol (2 ml), CH2C12 (3 ml), 4-klor-3-metoksybenzaldehyd (2 g, 11.7 mmol); fremstilt vesentlig som beskrevet av R.M. Riggs et al., J. Med. Chem., 30 1887, 1987), trimetylorthoformat (1.7 ml, 15.5 mmol) og en stor krystall av p-toluensulfonsyre ble omrørt ved romtemperatur i 1 time. Ytterligere MeOH (1 ml) og en krystall av p-toluensulfonsyre ble tilsatt og oppløsningen ble varmet helt til den var homogen. Ved endt reaksjon ble oppløsningen omrørt i 5 min. med overskudd fast NaHC03 og rotoravdampet for å fjerne oppløsningsmidlene. Pastaen ble løst opp i 40 ml EtOAc, fordelt mot fortynnet NaHC03 oppløsning og avdampet for å tilveiebringe en lysebrun olje. Reaksjonen ble gjentatt med ytterligere 2 g 4-klor-3-metoksybenzaldehyd og begge produktoljene ble kombinert for å tilveiebringe 4.37 g (86$) dimetylacetal 3.

Dietyl l- metoksy- l-( 4- klor- 3- metoksyfenyl) metanfosfonat 4
En oppløsning av dimetylacetal 3 (4.3 g, 20 mmol) sikt-tørket CH2C12 (20 ml) og trietylfosfit (4.1 ml, 24 mmol) ble omrørt under argon ved -78°C. Bortrifluoridetereat (2.95 ml, 24 mmol) ble dråpevis tilsatt ved -78°C, oppløsningen ble omrørt i 5 min. og lagret over natt ved -20° C. Neste dag ble reaksjonen varmet til romtemperatur og omrørt i 5 timer for å fullføre fosfonatdannelsen. Med omfattende omrøring ble reaksjonen stoppet med fast NaHCC>3 etterfulgt av 40 ml mettet NaHCOs oppløsning. Etter at gassutviklingen var opphørt ble 40 ml CH2Cl2 og 20 ml H20 tilsatt, den bifasiske blandingen ble fordelt og CH2C12 fasen ble isolert, tørket over Na2S04 og avdampet. Etter pumping i vakuum ble oljen renset på en silika gelplugg, eluerende med CH2C1<2> for å tilveiebringe fosfonat 4 som en lysegul olje (6.01 g., 99%).

Fosfonat 4 (3.2 g, 10 mmol) ble løst opp i 30 ml tørr THF under argon og avkjølt til -78°C. Dråpevis tilsetning av nBuLi (2.3M, 4.4 ml, 10.1 mmol) dannet et gul-orange fosfonatylid. Etter omrøring av ylidoppløsningen i 10 min. ble 5-klor-2-adamantanon (1.75 g, 9.5 mmol) løst opp i 8 ml THF tilsatt dråpevis til ylidet ved -78°C. Reaksjonen ble sakte varmet til romtemperatur over 45 min. og tilbake-strømmet i 2 timer. Ved avkjøling ble THF strippet i vakuum og produktet ble fordelt mellom EtOAc/heksaner (1:1) og fortynnet NaHCC^. Det organiske laget ble tørket over NaHCC^. Det organiske laget ble tørket over Na2S04, strippet for oppløsningsmiddel og renset på silikagel (2-4$ EtOAc/- heksaner) for å tilveiebringe 3.3 g (96$) enoleter 5 som en farveløs, viskøs gummi.

Demetylering til enoleterfenol 6 forløp med oppvarming av enoleter 5 (3.3 g, 9.3 mmol) i 22 ml DMF ved 135°C ved 175°C i nærvær av natrium etanthiolat (14 mmol) i 1.5 time. Reaksjonen ble avkjølt og fordelt mellom 50 ml EtOAc, 100 ml IM NH4CI og 10 ml mettet NaHC03 oppløsning. Den organiske fasen ble isolert og grundig vasket med vann mens den vandige fasen ble vasket en gang med EtOAc. EtOAc lagene ble kombinert, vasket med saltvann, tørket over Na2S04 og strippet for oppløsningsmiddel i vakuum. Råoljen ble renset på en silika gelkolonne, eluerende med 50$ C^C^/heksaner, for å tilveiebringe 3.6 g fenol 6 som en råolje. Ytterligere rensning ved to krystalliseringer frå en avkjølt 15$ CH2Cl2/heksaner oppløsning ga fenol 6 som et fast stoff (2.18 g, bO%). Natrium 2- cyanoetyl 2- klor- 5-( metoksy-( 5- klor) tricvklo-r3. 3. i. l3-=- Z-| dek_ 2_ vlldenmetvl )- i- fenylfosfat 7
Til en oppløsning av fenol 6 (0.75 g, 2.2 mmol), trietylamin (400 ul, 2.86 mmol) og vannfri TEF (8 ml) ble tilsatt til 2-klor-2-okso-l,3,2-dioksafosfolan (Fluka, 240 ul, 2.6 mmol) ved romtemperatur under argon. Reaksjonen ble omrørt i 3 timer og trietylammoniumhydroklorid presipiterte ut. Reaksjonsoppløsningen ble pipettert av presipitatet med en sprøyte med bomullstopp under sterk strømning av argon. Presipitatet ble skylt flere ganger med eter og den kombi-nerte oppløsningen og skyllingene ble avdampet i vakuum for å tilveiebringe et skum som ble beskyttet fra eksponering for fuktighet.
Skummet ble løst opp i vannfri DMF (4 ml) og omrørt med tørr NaCN (140 mg, 2.8 mmol) ved romtemperatur i 24 timer for å danne p<->cyanoetylfosfatdiester. Reaksjonsblandingen ble pumpet ved høyt vakuum ved 55 °C for å fjerne DMF og ga fosfatdiester 7 som en gummi. Rå fosfatdiester ble fotooksy-genert uten ytterligere rensning.
Syn- og anti- dinatrium 2- klor- 5-( 4- metoksyspirori. 2- dioksetan - 3. 2'-( 5>klor- ) tricyklor3 . 3. 1. l-3-^-7-! - dekanl - 4- vi )- l-fenylfosfat 1
Fosfatdiester 7 ble løst opp i 20 ml 10$ MeOE/CECl3 hvor det ble tilsatt 5 ,10 ,15 ,20-tetrafenyl-21E,23H-porfin (TPP, 0.8 ml av en 2 mg/ml CECI3 oppløsning). Reaksjonsblandingen ble mettet med oksygen og bestrålt med en 250W, natriumdamplampe med høyt trykk omhyllet av Kapton film ved 5°C mens oksygen ble sendt gjennom oppløsningen. Analytisk revers fase EPLC analyse viste fullstendig dioksetandannelse ved bestrålning i 20 min. Oppløsningsmidlene ble strippet i vakuum ved romtemperatur, resten ble pumpet til en gummi under høyt vakuum og lagret ved -20°C.
Rå cyanoetylfosfatdiesterdioksetan 8, løst opp i 11 ml MeOH, ble avspaltet med natriummetoksid (0,5 ml 25$ vekt NaOMe i MeOH) ved romtemperatur i 30 min. Ved fullført p-eliminering av cyanoetylgruppen ble 2 ml mettet NaHC03 oppløsning tilsatt og meOH ble roterende avdampet. HPLC kvalitetvann (15 ml) ble tilsatt og den brune oppløsningen ble sendt gjennom et 0.45u nylonfilter. Oppløsningsvolumet ble justert til 40 ml med HPLC kvalitetvann og renset ved preparativ HPLC ved anvendelse av en HC3CN/H2O gradient gjennom en polystyren kolonne (PLRP-S, Polymer laboratories). Fryse-tørkede fraksjoner ga 0.81 g (74$ fra fenol 6) av dioksetan 1. Analytisk revers fase HPLC på en polystyrenkolonne ved anvendelse av en gradient av acetonitril og 0.1$ NaHC03 og UV-deteksjon ved 270 nm viste en topp med en front som beveger seg som skulder og som representerer en blanding av syn og antiisomerer. En prøve av produktet, som en isomer-blanding, ble løst opp i en 0,1 M dietanolaminbuffer (1 mM MgCl2) ved pH 10. Etter å ha blitt behandlet med alkalisk fosfatase ble lys emittert som ventet og bekreftet dermed at produktet er et 1,2-dioksetan av tittelstrukturen.
EKSEMPLER
Dioksetan innenfor rammen av denne oppfinnelsen er blitt dannet og testet for essensielle egenskaper. Innbefattet som fremstilte og undersøkte dioksetaner er de hvor R er metyl, X er fosfat og Y<2> er klor, og Z er i 4 eller 5 posisjonen på en fenylring. I forsøkene nedenfor blir dioksetanene sammenlignet mot de kommersielle standardene CSPD og AMPPD.
K. jemiluminescensdeteks. ion av alkalisk fosfatase i oppløsning Alkalisk fosfatase (5.25 X IO-<17> mol) ble inkubert i 0.5 ml 0.1 M dietanolamin, 1 mM MgCl2, pH 10, inneholdende 0.4 mM dioksetan ved romtemperatur. Kjemiluminescens (5 sekund integral) ble målt i et Berthold LB952T luminometer ved 5, 10, 20, 30, 44, 50 og 60 minutter. Figurene 1 og 2 sammenligner ytelsen til CSPD og CDP-Star. Figur 1 viser sammenligning av CSPD og CDP-Star plottet som relative lysenheter (RLU) vs tid. Gjennomsnittet av tre replikater er plottet.
Figur 2 viser resultatene oppnådd fra et annet sett av prøver inneholdende 1 mg/ml kjemiluminescens forsterkende polymer-polyvinylbenzyltributylammoniumklorid.
Chemiluminescensdeteks. ion av biotinylert pBR322- 35mer på nylonmembran
Biotinylert pBR322 35-mer (13.1 pg i 1 ul) ble applisert på et lite stykke positivt ladet nylonmembran (Tropilon-pluss). Membranen ble fullstendig tørket og DNA ble fiksert til membranen ved UV kryss-binding (129 mJ/cm<2>). Membranen ble fuktet med PBS og deretter inkubert i blokkeringsbuffer (0.2$ 1-Block, 0.5$ SDS i PBS) i 10 minutter, i streptavidin-alkalisk fosfatase konjugat (Avidex-AP, Tropix; fortynnet 1:5000 i blokkeringsbuffer) i 20 minutter og vasket en gang i 5 minutter med blokkeringsbuffer og tre ganger i 5 minutter med vaskebuffer (0.5$ SDS i PBS). Membranene ble deretter vasket to ganger i fem minutter med analysebuffer (0.1 M DEA, 1 mM MgClg, pH 10.0). Til slutt ble membranene trimmet og forseglet i en liten firkant av varme-forskjærbar plast med 40 ul 0.25 mM CSPD eller CDP-Star (fortynnet i analysebuffer). Det forseglede stykket av membranen ble øyeblikkelig festet til et rør (pre-inkubert ved 22° C) og plassert i et Turner Model 20e luminometer (Turner Designs, Inc. Mountain view, CA). Lysemissjonen ble registrert hvert 5. minutt i en periode på 24 timer ved 22° C. Figur 3 er et plot av kjemiluminescens intensiteten (RLU) vs tid for SCPD og CDP-Star.
K. iemiluminescens deteksjon av Western blottet human transferrin
Renset human transferrin (Boehringer/Mannheim cat#1317 415) ble fortynnet i serie med IX SDS-PAGE ampi iseringsbuffer
(0.06 M Tris-HCl, pH 6.8, 2.25$ glycerol, 0. 5% e-merkapto-etanol, 2% SDS, 0.05$ bromfenol blå). Fortynningene ble oppvarmet ved 95 "C i 5 minutter og 5 pl fortynnet prøve ble applisert pr. gelkolonne. Prøvene ble elektroforetisk separert ved SDS-PAGE på 10$ polyakrylamid minigeler ved anvendelse av en Hoefer SE250 minigel apparatur. Hvert blot inneholder 10, 3.3, 1.1, 0.37, 0.12 og 0.04 ng mengder transferrin. Etter elektroforese ble geler og membraner ekvilibrert med transf erbuf f er (5 mM MP0S pH 7.5, 2 mM natriumacetat, 20$ MeOH) i 15 minutter. Proteinet ble overført til PVDF (Tropifluor) og positivt ladet nylon (Tropilon-Plus) membran i 1 time ved 90V ved 4°C. Blottene ble inkubert i blottebuffer (BB-1 [0.2$ 1-Block, 0.1$ Tween-20 i PBS] ble anvendt for PVDF og BB-2[3# 1-Block, 0.1$ Tween-20 i PBS] ble anvendt for nylon) i 30 mnutter. Blottene ble deretter inkubert med kaninpolyklonalt antihumant transferrin (Boehringer/Mannheim eat # 615 015; fortynnet 1:5000 i hensiktsmessig blokkeringsbuffer) i 30 minutter, og deretter vasket to ganger i 5 minutter (PVDF med BB-1, og nylon med vaskebuffer [0.1$ Tween-20 i PBS]). Deretter ble blottene inkubert med geiteanti-kanin IgG alkalisk fosfatasekonjugat (tropix; fortynnet 1:10000 i hensiktsmessig blokkeringsbuffer) i 30 minutter og deretter vasket to ganger i 5 minutter som ovenfor. Blottene ble deretter vasket to ganger i 5 minutter i analysebuf f er (0.1 M DEA, 1 mM MgClg» pH 10.0). Til slutt ble blottene inkubert med 0.25 mM CSPD eller CDP-Star (fortynnet i analysebuffer) i 5 minutter. BLottene ble drenert for overskuddsubstratoppløsning, plassert i plastreporter lag og eksponert for Kodak XAR-5 film. Resultatene oppnådd på nylon og på PVDF membraner er vist i figurene 4-5.
K. jemi luminescens deteksjonen av et enkelt kopi gjærgen på nylonmembran
Totalt genomisk DNA fra gjær Saccharomyces cerevisiae ble spaltet med EcoRI og Bgl 11 restriksjonsendonukleaser. 5.0, 0.5 og 0.05 jjg mengder av hver DNA spaltning ble separert ved elektroforese i en horisontal 0. 8% agarose IX TBE gel. Etter elektroforese ble gelen nedsenket i 1.5 M naCl, 0.5 M BaOH for 45 minutter for å denaturere DNA og deretter inkubert i nøytraliseringsbuffer (IM Tris, 1.5 M NaCl, pH 7.4) i 45 minutter. DNA ble overført til positivt ladede nylonmembraner (Tropilon-Plus) ved overnatt kapillær overføring ved anvendelse av 20X SSC. Membranene ble lufttørket og DNA ble TJV-kryssbundet til membranene ved 120 mJ/cm<2>. Et 1 kb Bgl 11 fragment, spaltet fra gjærgenet RPB 1, ble renset på gel og biotinylert ved en tilfeldig timingsreaksjon som inn-korporerer biotin-14-dCTP. Membranene ble prehybridisert i 30 minutter ved 68°C i hybridiseringsoppløsning { 7% SDS, 0.25 M Na2P04, 1.0 mM EDTA) hybridisert over natt ved 68°C med 5 ml frisk hybridiseringsbuffer inneholdende 5 ng/ml denaturert probe og deretter fjernet fra hybridiseringsoppløsningen og vasket som følger: to ganger i 5 min, med 2X SCC, 1% SDS ved romtemperatur; to ganger i 15 minutter i 0.1XSSC, 1% SDS ved 68° C; og to ganger i 5 minutter i 1XSSC ved romtemperatur. Membranene ble deretter inkubert i 10 minutter i blokkerings-buf f er ( 0. 2% cassein, 0.5$ SDS, PBS) og 20 min. med 1:5000 fortynning AVIDx-AP i blokkeringsbuffer. De ble deretter vasket i blokkeringsbuffer i 5 min., 3 ganger i 10 min. i vaskebuffer ( 0. 5% SDS, PBS) og to ganger i 2 min. i analysebuf f er (0.1 M dietanolamin, 1.0 mM MgCl2, pH 10.0) etterfulgt av inkubasjon i 5 minutter i 0.25 mM dioksetan i analysebuf fer. Etter drenering av overskuddsubstratoppløsning ble membranene omhyllet av plast og eksponert for røntgenfilm. 60 minutter utsettelse tatt 10 minutter, 70 minutter og 19 timer etter substratinkubasjon er reflektert i følgende eksponeringer. Sammenligninger av CSDP og CDP-Star er vist i figurene 6, 7 og 8.
Kjemiluminescens deteksjon av DNA sekvensstiger
DNA sekvenseringsreaksjoner ble utført med Tropix SEO-Light sett ved anvendelse av biotinylert (-20) universal primer og enkelttrådet M13 mpl8 templat DNA. Reaksjonsproduktene ble separert på en b% polyakrylamid 8 M ureagel, overført til Tropilon-Plus nylonmembraner ved kapillær virkning og UV-kryssbinding til membranen (total bestrålning - 120 mJ/cm<2>). Kjemiluminescens deteksjon ble utført ved inkubering av membranen i 10 minutter i blokkeringsbuffer (0.2$ i-Block, 0.5$ SDS, PBS) i 20 minutter i konjugatoppløsning (1/5000 fortynning av Avidex-AP streptavidin alkaliskfosfatase konjugat i blokkeringsbuffer), deretter vasket 1X5 minutter med blokkeringsbuffer, 3X5 minutter med vaskebuffer (0.5$ SDS, PBS) og 2X2 minutter med analysebuf f er (0.1 M dietanolamin, 1 mM MgCl2» pH 10). Hver membranstrimmel ble inkubert i 5 minutter med enten 0.25 mM CSPD eller CDP-Star i analysebuf fer. Alle trinnene ble utført ved romtemperatur i en stor varmeforseglet plastpose med moderat risting (140-170 rpm). Sammenligning av CSPD og CDP-Star ved tre tidspunkter er tilveiebragt. Tid etter inkubasjon med substrat og eks-poneringstid for Kodak XAR-5 røntgenfilm er vist på figur 9.
Ytterligere testing reflekterer verdier så som kvantumutbytte (utført av et uavhengig laboratorium ifølge prosedyren angitt nedenfor), ^ ±/ 2 °S emissjonsbølgelengde maksimum. Disse dioksetanene er identifisert med tall og i tabellene som følger etter tallene er identiteten til substituenten på adamantylringen, om noen etterfulgt av identiteten til Z substituenten, gitt. I de testede forbindelsene er X fosfat. Verdier for kvantumutbytte og T^/g blir oppnådd både for dioksetan alene i 0.1 molar DEA og i nærvær av et forsterkningsmiddel, Sapphire II.
Protokoll for kvantumutbyttebestemmelse
500 ul 3.2 x IO"<4> M oppløsning av et dioksetan i 0.IM DEA, pH 10.0 ble plassert i et 12 x 75 mm rør ved 20"C. Oppløsningen ble ekvilibrert til 20°C i et avkjølt vannbad i 10 minutter.
2 ul alkalisk fosfatase suspensjon ble tilsatt til røret inneholdende dioksetan og øyeblikkelig vortex behandlet i 1 sekund og plassert i et 20 °C vannbad. Røret ble deretter plassert i MGM Optocomp I luminometer og lyssignalet ble målt ved 1 sek. integrasjonstider. Etter at lyssignalet var målt ble røret plassert tilbake inn i 20° C vannbad og målingen ble gjentatt. Totale tellinger for dioksetan ble bestemt ut fra intensitetsdataene. Totale tellinger observert for en gitt konsentrasjon av dioksetan er produktet av Photon deteksjonseffektiviteten (PDE) til luminometeret, kvantumutbytte av dioksetan og antall molekyler som kan emittere lys (konsentrasjon til defosforylerte dioksetaner). PDE for MGM Optocomp I luminometer ble bestemt å være 2.56 x IO-<3> målt med en Biolink absolutt standard og anvendelse av kjent spektral respons av luminometerets PMT og det kjente emissjonsspekteret til dioksetanene. Kvantumutbyttet blir beregnet ved å dele totale tellinger målt ved PDE og konsentrasjonen av dioksetan.
Beregning av halveringsliv eller halveringstid mot likevekt lvsemissjon
Fra avlesningen til Turner luminometeret ble det maksimale signalet målt. Maksimalt signal minus Turner lys enhet avlesningene ved 30, 150, 300 eller 600 sekund intervaller ble beregnet og oppstilt grafisk mot tid i sekunder. Ut fra grafene ble en eksponentiell ligning beregnet for å bestemme halveringslivet.
Halveringslivene til dioksetaner ble også bestemt direkte ut fra Turner luminometer utprintninger.
Emiss. i on smak sima
Til 2 ml av en pH 10 oppløsning av 0.4 mM dioksetan, 0.1M dietanolamin, 1 mM MgCl2 ble det tilsatt 9.9 x 10_<11>M alkalisk fosfatase. Oppløsningen ble ekvilibrert 5 minutter i et Spex Fluorolog Fluorimeter og deretter scannet 5 ganger ved 0.5 sek/nm for kjemiluminescens emissjon. Kjemiluminescens emissjons bølgelengde maksimum ble registrert.
K. jemiluminescens DNA sekvensering
DNA sekvensering med kjemiluminescens deteksjon ble utført som beskrevet i Tropix SEQ-Light protokollen. Kort fortalt
ble DNA sekvenseringsreaksjonene initiert med biotinylerte primere ved anvendelse av M13 enkelttrådet fag DNA som et templat. Reaksjonene ble separert på 8 M urea deanturerende PAGE, overført horisontalt til Tropilon-Plus nylonmembran ved kapillær virkning og kryss-bundet til membranen ved eksponering for UV-lys ved anvendelse av en Spectronics SpectroLinker XL-1500 ved 200 mJ/cm<2>. Membranene ble inkubert med blokkeringsbuffer (0.2$ I-Block, 0. 5% natrium, dodecyl-sulfat/SDS i fosfatbuffret saltvann/PBS [20 mM natriumfosfat, pH 7.2, 150 mM NaCl]) i 10 minutter, inkubert med en 1/5000 fortynning av Avidex-AP streptavidin-alkalisk fosfatase i blokkeringsbuffer i 20 minutter, vasket i 5 minutter i blokkeringsbuffer, vasket 3X5 minutter med vaskebuffer ( 0. 5% SDS, PBS), vasket <2> X 5 minutter med analysebuffer (0.1 M dietanolamin, 1 mM MgCl2 pH 10) og deretter inkubert med dioksetanoppløsning (enten CSPD, 140-17 eller 128-87 fortynnet til 0.25 mM i analysebuf fer) i 5 minutter. Membranene ble drenert, forseglet i en plastfolder og eksponert for Kodak XAR-5 røntgenfilm. For dioksetan 128-87 var eksponeringstiden 70 minutter og for 140-17, 80 minutter, begge 65 minutter etter substrattilsetning. For sammenligning av dioksetan 128-87 mot CSPD var membraneksponeringstiden 5 minutter etter en 24 timer lang inkubasjon med substrat.

For å demonstrere positivt interaksjonen av dioksetan, eller i det minste eksitert-tilstand emitterer, med forsterkningsmidler av en type som er kjent for anvendelse i sammenheng med dioksetaner, ble bølgelengden for emissjonsmaksimum detektert i fravær av forsterkningsmiddel, i nærvær av BDMQ og på en nylonmembran. Data er angitt i følgende tabell.
DOT BLOT ANALYSER
Som angitt ovenfor er dioksetanene ifølge denne oppfinnelsen egnede for anvendelse i Dot blot analyser. Dioksetaner syntetisert ifølge synteseveien beskrevet ovenfor ble anvendt i dot blot analysene. Ved bekreftelse på fravær av kjemiluminescens til dioksetaner som bærer en Z substituent i seks posisjonen er det å bemerke at forbindelse 140-62 ga et vedvarende fravær av signal eller, under optimale betingelser, et nesten ikke detekterbart signal. Dioksetan med metoksysubstituent ved seks posisjonen med en klor substituent på adamtylringen, 140-73, ga ikke noe signal i dot blot analysen, som igjen bekrefter mangel på kjemiluminescens aktivitet i seks-substituerte metafosfatfenyldioksetaner.
Figurene 12-21.
Nitrocellulose og nylonmembraner ble belagt i flekker med en biotinylert 35 base oligonukleotidprobe. Proben ble fortynnet i IX SSC for å tilveiebringe en begynnende fortynning på 210 pg. Suksessive 1:2 fortynninger av utgangsfortynningen ble flekkvis applisert på membranene, 12 flekker totalt. Membranene ble tørket, utsatt for optimal UV-kryssbinding (120 mH/cm<2>), blokkert i 30 minutter i blokkeringsbuffer (nitrocellulose: 0.2$ I-Block, 0. 1% Tween-20, IX PBS; nylon: 0. 2% I-Block, 0. 5% SDS, IX PBS), inkubert i 20 minutter i en 1/5000 fortynning av streptavidin-alkalisk fosfatasekonjugat fortynnet i blokkeringsbuffer og vasket som følger: 1x5 minutter i blokkeringsbuffer; 3x5 minutter i IX PBS, 0. 3% Tween-20 (nitrocellulose) eller 3x5 minutter i IX PBS, 0.5$ SDS (nylon); 2x5 minutter i substratbuffer (0.1M dietanolamin, O.lmM MgCl2, pH 10); 1x5 minutter i en 1/20 fortynning av nitro-blokk (Tropix, Inc. Bedford, MA) fortynnet i substratbuffer (bare nitrocellulose eksperiment); og 2 x 5 minutter i substratbuffer (bare i nitrocellulose-eksperimentet). Membranene ble inkubert med 0.25 mM dioksetan fortynnet i substratbuffer i 5 minutter. Flere membraner i både nitrocellulose og nyloneksperimentene ble inkubert med 0.25 mg/ml Calfax DB-45, Calfax 10L-45 eller Calsoft T-60 (Pilot Chemical Company, Los Angeles, CA), 1.0 mg/ml Tween-20, 1.0 mg/ml Nitro-Block og 0.25 mM dioksetan fortynnet i substratbuffer i 5 minutter. Disse membranene ble ikke utsatt for en 1/20 fortynning av Nitro-Block. Membranene ble deretter eksponert for røntgenfilm og utviklet.
Som det fremgår fra ovennevnte resultater endrer de elektron-fratrekkende gruppene tilsatt til den aromatiske ringen av dioksetan kinetikkene til lysemissjonene, men øker kjemiluminescens signalet. I kontrast til dette aksellerer de elektron-donerende gruppene T ^/g ved å. lette elektronover-føring fra oksygenet gjennom den aromatiske gruppen til dioksetan. Ved riktig seleksjon av naturen og evnen til den elektron-donerende eller elektron-tiltrekkende Z substituenten, og samtidig seleksjon av hensiktsmessig substituent for adamantylringen kan dioksaner med spesifikke karaktertrekk, inkludert optimalisert signalintensitet, optimalisert hastighet, spesifikk emissjonsbølgelengde og lignende bli oppnådd.
Disse deoksytanene kan bli anvendt for analyser av alle typer hvor et enzym som kan spalte dioksetan er til stede alene eller som kan bli koblet til et element av komplekset som analyten, om til stede, vil danne. Konvensjonelle analyse-formater er kjente for fagfolk innenfor dette området og er beskrevet i patentene angitt ovenfor. Eksempelvise be-skrivelser av egnede analyser fremkommer i US-PS 5.112.960 og disse er innkorporert heri som referanse. Analyseformatet, per se, unntatt den forsterkede ytelsen deri til dioksetanene ifølge denne oppfinnelsen, utgjør ikke et aspekt ifølge oppf innelsen.
Dioksetanene ifølge denne oppfinnelsen, samt mellomprodukter derav, er blitt beskrevet med referanse til både generisk beskrivelse og spesifikk utførelse. I tillegg er dioksetan-yteevnen blitt beskrevet generelt og eksemplifisert.
