Generelt katalyseres disse transmethyleringsreaksjoner av et utall transmethylaser som anvender AdoMet som et methyl-donorsubstrat ved methyleringen av et utall methyl-akseptorsubstrater slik som catecholer; norepinefrin; histamin; serotonin; tryptamin; membranfosfo-lipider; lysyl, arginyl, histidyl, aspartyl, glutamyl og carboxylgrupper av visse proteiner; tRNA og mRNA; og DNA. Disse forskjellige transmethylaser produserer S-adenosin-L-homocystein (AdoHcy) som et biprodukt etter overføringen av en methylgruppe fra AdoMet til det egnede methyl-akseptor-substrat.
AdoHcy er blitt vist å være en kraftig "feed-back"-inhibitor av de AdoMet-avhengige transmethyleringsreaksjoner. Denne "feed-back"-inhibering av transmethylasene reguleres av bionedbrytningen av AdoHcy med S-adenosyl-L-homocysteinhydrolase som tilveiebringer en homeostatisk kontroll på vev-nivå av AdoHcy. Aktiviteten av S-adenosyl-L-homocysteinhydrolase betraktes generelt av fagmannen å spille en viktig rolle ved regulering av vevnivåene av AdoHcy og derved regulering av aktiviteten av de AdoMet-avhengige transmethyl-eringsreaks joner .
Forbindelsene fremstilt ifølge foreliggende oppfinnelse er inhibitorer av S-adenosyl-L-homocysteinhydrolase. Disse forbindelser inhiberer derfor den naturlig forekommende bioned-brytning av AdoHcy og resulterer i forhøyede vevnivåer av AdoHcy. Forhøyede nivåer av AdoHcy tilveiebringer i sin tur en endogen "feed-back"-inhibering av forskjellige AdoMet-avhengige transmethyleringsreaksjoner som er forbundet med biologiske prosesser relatert til viral vekst og replikasjon, viral transformasjon av celler, vekst av ondartede celler,
og prosesser slik som kjemotaxis og sekresjon.
Forbindelsene er derfor anvendbare som inhibitorer av
disse biologiske prosesser og er anvendbare ved deres slutte-lige anvendelse som terapeutiske midler ved behandling av pasienter angrepet av forskjellige patologiske tilstander hvori disse prosesser er involvert, slik som virale infeksjoner og neoplastiske sykdomstilstander.
Oppfinnelsen angår således en analogifremgangsmåte for fremstilling av terapeutisk aktive forbindelser med formelen
hvori
X1 og X2 er hver hydrogen eller halogen, forutsatt at minst én av Xx og X2 er halogen.
Som anvendt her, angir uttrykket "halogen" et fluor-, klor-, brom- eller jodatom.
Analogifremgangsmåten ifølge oppfinnelsen er kjenne-tegnet ved at
(a) et 2'-fenylsulfonyl-halomethyliden-adenosin-derivat som bærer egnede blokkerende grupper av formel
omsettes med tributyl-tinnhydrid i nærvær av 2,2'-azobis-isobutylnitril under dannelse av det tilsvarende 2'-tributyl-tinn-halomethylidenderivat av formel (b) det således dannede 2'-tributyl-tinn-halomethylidenderivat av formel
omsettes med hexamethyldisilazan i nærvær av en katalytisk mengde av ammoniakk i formamid, og
(c) de blokkerende grupper fjernes.
En generell synteseprosedyre for fremstilling av forbindelser av formel (1) hvori én av Xx og X2 er hydrogen, er angitt i det etterfølgende reaksjonsskjerna. I det etterføl-gende reaksjonsskjema er alle substituenter med mindre annet er angitt, som tidligere definert. I tillegg angir uttrykket "(j)" en fenylgruppe; uttrykket "X,^" angir et halogena tom; uttrykket "LP=" angir en ylidgruppe [eksempelvis kan et difluormethylidenfosfonatylid ha formelen (<|>)2P(0)=C(F)2] .
I trinn a omsettes ketonderivatet (2), f.eks. 6-klor-9-[(3,5-0-tetraisopropyldisiloxan-l,3-diyl)-2-keto-f3-D-erythro-pentof uranosyl ]-purin i en Wittig-reaks jon med et fosforylid som generelt beskrevet for reaksjonsskjema A. Når en forbindelse av formel (1) ønskes hvori én av X, eller X2 er hydrogen, kan (2) omsettes med et fenylsulfonyl-halomethyliden-fosforylid under dannelse av det tilsvarende 2-fenylsulfonyl-halomethylidenderivat (6).
Det egnede fenylsulfonyl-halomethyliden-fosforylid kan fremstilles i henhold til prosedyrer som er velkjente innen faget. Hvor eksempelvis en forbindelse av formel (1) ønskes hvori én av X^ eller X2 er hydrogen, kan det egnede keton (2) omsettes med et fenylsulfonyl-halomethyliden-fosforylid fremstilt ved omsetning av et halogenfosfat (slik som diethylklorfosfat) med et halomethylfenylsulfon i nærvær av en base (slik som lithiumdiisopropylamid).
Når en forbindelse av formel (1) ønskes hvori én av
X^ eller X2 er fluor, kan nærmere bestemt det egnede keton (2) omsettes med et fenylsulfonylfluormethyliden-fosforylid fremstilt ved omsetning av et halogenfosfat (slik som diethylklorfosfat) med fluormethylfenylsulfon i nærvær av en base (slik som lithiumdiisopropylamid).
I trinn b omdannes 2-fenylsulfonyl-halomethyliden-derivatet (6) til det tilsvarende 2-tributyl-tinn-halomethylidenderivat (7). Eksempelvis kan denne reaksjon utføres ved omsetning av (6) med tributyl-tinnhydrid (HSnBu^) i nærvær av 2,2'-azobis-isobutylnitril (AIBN) i et egnet løsnings-middel slik som benzen. De geometriske isomerer av 2-tributyl-tinn-halomethylidenderivatet (7) kan eventuelt isoleres under anvendelse av prosedyrer og teknikker som er velkjente innen faget. Eksempelvis kan de geometriske isomerer av (7) separeres hensiktsmessig ved flashkromatografi (silicagel)
og eluering med 7% ethylacetat i hexan.
I trinn c fjernes tributyl-tinngruppen av (7) og erstattes med et hydrogenatom for å tilveiebringe det tilsvarende 2-halomethylidenderivat (8). Dette kan utføres ved omsetning av (7) med hexamethyldisilazan i nærvær av en katalytisk mengde ammoniakk i formamid. I tillegg vil fagmannen lett innse at under forløpet for fjerning av tributyl-tinngruppen av (7) vil også 6-klorgruppen av purinbasen av (7) bli erstattet med en aminogruppe.
I trinn d fjernes den tetraisopropyldisiloxan-blokkerende gruppe av (8) som beskrevet for reaksjonsskjema A (trinn b) under dannelse av det tilsvarende avblokkerte 2'-halomethyliden-adenosinderivat (9). Eksempelvis kan et fluoridsalt slik som cesiumfluorid, anvendes.
Som det klart fremgår for fagmannen, eksisterer 2'-halomethyliden-adenosinderivatene representert ved forbindelse (9) i reaksjonsskjema B som to geometriske isomerer som kan angis som (Z)- og (E)-isomerene. Disse isomerer kan separeres under anvendelse av konvensjonelle separasjonstek-nikker velkjente innen faget. Eksempelvis kan de geometriske isomerer oppløses ved kolonnekromatografi under anvendelse av en Dowex<®> 1-X2 (0H~ form) harpiks. Eventuelt kan 2-tributyl-tinn-halomethylidenderivatet (7) eller det tilsvarende 2-halomethylidenderivat (8) hensiktsmessig oppløses i dets geometriske isomerer under anvendelse av kjente teknikker. De tilsvarende geometriske isomerer av 2<1->halomethyliden-adenosinderivatene (9) vil bli dannet ved fortsettelse av syntesen som vist i reaksjonsskjema B under anvendelse av de individuelle isomerer av (7) eller (8).
Det etterfølgende eksempel illustrerer en typisk syntese som beskrevet i reaksjonsskjemaet.
Eksempel
( Z)- og ( E)- 2'- deoxy- 2'- fluormethyliden- adenosin
Trinn a; 6- klor- 9-[ ( 3, 5- 0- tetraisopropyldisiloxan- l, 3- diyl)-[ 3- D- erythro- pentofuran- 2- ( 2- f luor- 2- fenylsulfonylmethyliden) - osyl]- purin
Fremstill diethylfluormethylfenylsulfonylfosfonat som følger: Tilsett til en løsning av 2,02 g (11,6 mmol) fluormethylfenylsulfon i 23 ml tørt THF som er blitt avkjølt til ca. -78°C i en tørr 3-halset kolbe på 100 ml utstyrt med rørestav, argon innløpsventil, termometer og gummimembran, 4.02 g (3,37 ml, 11,6 mmol) diethylklorfosfat via en sprøyte. Tilsett til denne blanding en løsning av 17,7 ml (23 mmol)
1.3 M lithiumdiisopropylamid i cyclohexan via en sprøyte og følg dannelsen av diethylfluormethylfenylsulfonylfosfonat ved gasskromatografi (GC).
Tilsett til den ovenfor angitte diethylfluormethyl-fenylsulfonylfosfonatløsning som er blitt oppvarmet til 0°C, en løsning av 6,11 g (11,6 mmol) 6-klor-9-[(3,5-0-tetraisopropyldisiloxan-l , 3-diyl) -2-keto-(3-D-erythro-pentofuranosyl ] -purin i 10 ml tørt THF. Tillat reaksjonsblandingen å oppvarmes til romtemperatur og omrør under en argonatmosfære i 4 timer.
Hell blandingen over i en mettet, iskald løsning av ammonium-klorid og ekstraher blandingen med ethylacetat (3 ganger, 75 ml hver gang). Kombiner de organiske lag, tørt med vann-fritt magnesiumsulfat, og fordamp til tørrhet. Kromatografer residuet på en silicagel flashkolonne og eluer med ethylacetat/hexan (1/3, v/v). Fordamp løsningsmidlet til tørrhet under redusert trykk under dannelse av tittelforbindelsen som et hvitt skum. Triturer med hexan, avkjøl over natten, filtrer og tørk bunnfallet under dannelse av 4,9 g av tittelforbindelsen.
Trinn b: 6- klor- 9-[( 3, 5- 0- tetraisopropyldisiloxan- l, 3- diyl)-( 3- D- erythro- pentofuran- 2- ( 2- f luor- 2- tributyl- tinn- methyliden) - osyl]- purin
Tilsett 769 mg (0,71 ml, 2,6 mmol) tributyl-tinnhydrid og 0,6 g (0,88 mmol) 6-klor-9-[(3,5-0-tetraisopropyldisiloxan-l , 3-diyl) -p-D-erythro-pentofuran-2-(2-fluor-2-fenyl-sulf onylmethyliden) -osyl] -purin til 8,8 ml benzen og oppvarm til tilbakeløpskokning. Tilsett 35 mg (0,26 mmol) 2,2<*->azobis-isobutylnitril i 0,5 ml benzen i løpet av 1 time. Separer isomerene ved flashkromatografi (silicagel) og eluer med 7% ethylacetat i hexan under dannelse av 0,31 g av den først eluerende isomer (hurtigere isomer) og 0,38 g av den andre eluerende isomer (langsommere isomer) av tittelforbindelsen.
Trinn c: ( Z)- og ( E)- 6- amino- 9-[( 3, 5- 0- tetraisopropyldisiloxan-1, 3- diyl) -( 3- D- erythro- pentofuran- 2- ( 2- f luormethyliden) - osyl]-purin
Hurtigere isomer: Kombiner 0,7 g (0,8 mmol) av den hurtigere isomer av 6-klor-9-[(3,5-0-tetraisopropyldisiloxan-1,3-diyl)-p-D-erythro-pentofuran-2-(2-fluor-2-tributyl-tinn-methyliden)-osyl]-purin og 3,5 ml hexamethyldisilazan i 1,75 ml formamid som er blitt mettet med ammoniakk. Oppvarm blandingen til 100°C i 3 timer. Fordamp løsningsmidlet ved redusert trykk og rens residuet ved flashkromatografi (silicagel) og eluer med ethylacetat/hexan (1/1, v/v). Fordamp løsningsmidlet under dannelse av 0,165 g (0,31 mmol)
av tittelforbindelsen.
Langsommere isomer: Kombiner 0,3 g (0,36 mmol) av den langsommere isomer av 6-klor-9-[(3,5-0-tetraisopropyldisiloxan-1, 3-diyl) -|3-D-erythro-pentofuran-2- (2-f luor-2-tributyl-tinn-methyliden)-osyl]-purin og 1,5 ml hexamethyldisilazan i 0,75 ml formamid som er blitt mettet med ammoniakk. Oppvarm blandingen til 100°C i 3 timer. Fordamp løsnings-midlet ved redusert trykk og rens residuet ved flashkromatografi (silicagel) og eluer med ethylacetat/hexan (1/1, v/v). Fordamp løsningsmidlet under dannelse av 0,065 g (0,12 mmol)
av tittelforbindelsen.
Trinn d: ( Z)- og ( E)- 2'- deoxy- 2'- fluormethyliden- adenosin
Hurtigere isomer: Kombiner 0,114 g (0,22 mmol) 6-amino-9- [ (3 , 5-0-tetraisopropyldisiloxan-l, 3-diyl) -|3-D-erythro-pento-furan-2-(2-fluormethyliden)-osyl]-purin (hurtigere isomer) og 1,32 g (8,7 mmol) cesiumfluorid i 30 ml ethanol og omrør ved omgivende temperatur i 24 timer. Fordamp løsningsmidlet i nærvær av silicagel, anbring silicagelresiduet på en flash-kromatograf ikolonne og eluer med ethylacetat etterfulgt av 10% ethanol i ethylacetat. Fordamp løsningsmidlet under dannelse av 47 mg (0,16 mmol) av tittelforbindelsen i et 7,0% totalt utbytte. Smp. 114-118°C.
Langsommere isomer: Kombiner 0,206 g (0,4 mmol) 6-amino-9-[ (3 ,5-0-tetraisopropyldisiloxan-l, 3-diyl) -|3-D-erythro-pentofuran-2-(2-fluormethyliden)-osyl]-purin (langsommere isomer) og 2,28 g (15 mmol) cesiumfluorid i 60 ml ethanol og omrør ved omgivende temperatur i 24 timer. Fordamp løsnings-midlet i nærvær av silicagel, anbring silicagelresiduet på en flashkromatografikolonne og eluer med ethylacetat etterfulgt av 10% methanol i ethylacetat. Fordamp løsningsmidlet under dannelse av 81 mg (0,28 mmol) av tittelforbindelsen i et totalt utbytte på 5,9%. Smp. 121-122°C.
Utgangsmaterialer for anvendelse i den generelle synteseprosedyre vist i reaksjonsskjemaet er lett til-gjengelige under anvendelse av syntesemetoder og prosedyrer som er velkjente innen faget. Eksempelvis kan 6-amino-9-[ (3 ,5-0-tetraisopropyldisiloxan-l, 3-diyl) -2-keto-(3-D-erythro--pentofuranosyl]-purin anvendes som utgangsmateriale for mange av forbindelsene av formel (1) og kan fremstilles fra adenosin i henhold til prosedyren beskrevet av Usui og Ueda [Chem. Pharm. Bull. _3_4, 15 (1986)]. 6-klor-9-[ (3 , 5-0-tetraisopropyldisiloxan-l, 3-diyl) -2-keto-|3-D-erythro-pentofuranosyl]-purin kan fremstilles fra 6-klor-9 ((3-D-ribo-furanosyl)-purin ved en prosedyre analog med den som er beskrevet av Usui og Ueda.
Forbindelsene av formel (1) er antatt å utøve sin inhiberende effekt på AdoMet-avhengig transmethylering ved inhibering av AdoHcy-hydrolase og derved tilveiebringe en økning i vevnivåer av AdoHcy som i sin tur tilveiebringer feedback-inhibering av AdoMet-avhengig transmethylering. Virale infeksjoner som er karakterisert ved en overdreven AdoMet-avhengig transmethyleringsaktivitet og for hvilke behandling med en forbindelse av formel (1) vil være særlig anvendbar, innbefatter: retrovirus slik som HTLV-I, HTLV-II, humane immunsvekkende virus, HTLV-III (AIDS-virus) og lignende; RNA-virus slik som influensa type A, B og C, kusma, meslinger, rhinovirus, denguefeber, rubella, rabies, hepatitis-virus A, encefalitis-virus og lignende; DNA-virus slik som herpes, vaccinia, papillomavirus (vorter), hepatitis-virus B og. lignende.
FORSØKSRAPPORT
Antiretroviral aktivitet ble bestemt i XC-plakk-bestemmelsen under anvendelse av Moloney-leukemivirus (MoLV) som en modell for human immunosvekkelsesvirus (HIV), hoved-sakelig som beskrevet av Bowlin og Proffitt [J. Interferon Res., 1983, 3, 19]. Forbindelsen [(Z)-2'-deoxy-2'-fluormethyliden-adenosin] inhiberte den cytopatiske effekt av Moloney-leukemivirus til 27,4 % ved 1 ug/ml og 76,8 % ved
10 ug/ml.
I tillegg ble antiviral aktivitet overfor herpes simplex I-virus bestemt i en plakkreduksjonsbestemmelse under anvendelse av Vero-apenyreceller infisert med herpesvirus Hominis type I. Forbindelsen [(Z)-2'-deoxy-2'-fluormethyliden-adenosin] inhiberte den cytopatiske effekt av HSV-I til 54 % ved 1 ug/ml og 84,2 % ved 10 ug/ml. Forbindelsen [(E)-2'-deoxy-2' -f luormethyliden-adenosin] inhiberte den cytopatiske effekt av HSV-I til 57,8 % ved 10 rø/ml.
En terapeutisk effektiv mengde av en forbindelse av formel (1) forventes å variere fra 0,1 mg pr. kg kroppsvekt pr. dag (mg/kg/dag) til 100 mg/kg/dag. Foretrukne mengder er forventet å variere fra 0,5 til 10 mg/kg/dag.
ADA deaminerer forbindelsene av formel (1), og ned-bryter derved de aktive forbindelser til relativt inaktive metabolitter. Når en forbindelse av formel (1) og en ADA-inhibitor administreres i ledsagende behandling, vil dosen være mindre i mengde eller frekvens enn hva som er nødvendig når forbindelsen av formel (1) administreres alene.
Forskjellige farmasøytisk akseptable ikke-toksiske ADA-inhioitorer kan anvendes, innbefattende deoxycoformycin. En terapeutisk effektiv inhiberende mengde av ADA-inhibitoren vil variere fra 0,05 mg/kg/dag til 0,5 mg/kg/dag og vil for-trinnsvis være fra 0,1 mg/kg/dag til 0,3 mg/kg/dag. Deoxycoformycin er den foretrukne ADA-inhibitor for anvendelse i ledsagende behandling med forbindelsene av formel (1).
Forbindelsene kan administreres alene eller i form av
et farmasøytisk preparat i kombinasjon med farmasøytisk akseptable bærere eller eksipienter, hvor mengden og arten av disse bestemmes av løseligheten og de kjemiske egenskaper av den valgte forbindelse, den valgte administreringsmåte og standard farmasøytisk praksis. I tillegg kan forbindelsene administreres som ovenfor angitt i ytterligere kombinasjon med en ADA-inhibitor. Selv om forbindelsene fremstilt ifølge oppfinnelsen er effektive i seg selv, kan de formuleres og administreres i form av deres farmasøytisk akseptable syre-addisjonssalter av stabilitetsformål, hensiktsmessig krystal-lisering, økt løselighet og lignende.
Claims (2)
1- Analogifremgangsmåte for fremstilling av terapeutisk aktive forbindelser av formelen
hvori X2 og X2 er hver hydrogen eller halogen, forutsatt at minst én av Xx og X2 er halogen,karakterisert ved at (a) et 2'-fenylsulfonyl-halomethyliden-adenosinderi- vat som bærer egnede blokkerende grupper av formel omsettes med tributyl-tinnhydrid i nærvær av 2,2'-azobis-isobutylnitril under dannelse av det tilsvarende 2<*->tributyl- tinn-halomethylidenderivat av formel (b) det således dannede 2'-tributyl-tinn-halomethylidenderivat av formel
omsettes med hexamethyldisilazan i nærvær av en katalytisk mengde av ammoniakk i formamid, og (c) de blokkerende grupper fjernes.
2. Fremgangsmåte ifølge krav 1 for fremstilling av (Z)-2'-deoxy-2'-fluormethyliden-adenosin eller (E)-2'-deoxy-2'-fluormethyliden-adenosin,
karakterisert ved at tilsvarende utgangsmaterialer anvendes.
