Abstract

본 발명은 발효산물의 연속적 제조 방법, 보다 구체적으로는 발효에 의해 알콜을 비롯한 균주 대사산물을 제조하는데 있어서, 발효시에 응집성 균주 및 유동식 필터를 사용함으로써 발효산물을 높은 세포농도로 경제적 및 연속적으로 제조하는 방법에 관한 것이다.
본 발명에 따르면, 목적 발효산물, 특히 알콜의 발효에 의한 생산성이 이론치에 가까운 수율로서 반응기 체적 1리터당 1시간동안 70그람 이상(종래 방식의 70배 이상)에 달할 뿐만 아니라 발효산물의 균주 세포 농도도 반응기 1리터당 130그람 이상으로 유지되며, 발효 반응기를 수개월 이상 동안 연속 운전할 수가 있다는 잇점이 있다. 따라서, 본 발명에 의하면 원료 가격에 있어서 전체 생산원가 기준으로 30% 이상 절약할 수 있고, 시설비, 공장부지 비용, 인건비, 동력비, 폐수처리비용 등에 있어서 많은 생산 원가 절감을 할 수 있다.

본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 본 발명에 따른 발효에 의한 균주 대사산물의 연속적 제조 공정의 한 태양을 도시하기 위한 개략도이다.

Claims (20)

  1. 발효에 의해 균주 대사산물을 제조하는데 있어서, 발효시에 응집성 균주 및 유동식 필터를 사용하는 것을 특징으로 하는 연속적 제조 방법.
  2. 제1항에 있어서, 상기 균주 대사산물이 바이오 에너지, 의약품 또는 식품인 것을 특징으로 하는 방법.
  3. 제1항에 있어서, 상기 균주 대사산물이 에탄올인 것을 특징으로 하는 방법.
  4. 제1항에 있어서, 상기 발효를 위한 탄소 공급원이 공업용 포도당, 타피오카, 옥수수대, 목질계 물질, 고구마 또는 곡물인 것을 특징으로 하는 방법.
  5. 제1항에 있어서, 탄소 공급원을 분쇄, 액화 및 당화시켜 액당화 혼합물을 수득하는 단계; 상기 액당화된 혼합물을 유동식 필터가 장착된 발효조에 도입하여 응집성 세포 균주의 존재하에 발효시키는 단계; 및 상기 발효후의 발효 생성물 중의 상등액을 수거 및 분류하는 단계를 포함하는 것을 특징으로 하는 방법.
  6. 제5항에 있어서, 상기 액당화된 혼합물이 당 농도가 14 내지 21%이고 10 내지 1,000㎛ 입경의 고형분을 30 내지 50% 함유하는 방법.
  7. 제5항에 있어서, 상기 액당화된 혼합물을 발효조에 도입하기 전에 고형분/액체 분리기에 통과시키는 방법.
  8. 제5항에 있어서, 상기 응집성 세포균주가 사카로마이세스 우바룸(Saccharomyces uvarum), 사카로마이세스 세레비지에(Saccharomyces cerevisiae) 또는 사카로마이세스 다이아스타티쿠스(Saccharomyces diastaticus)인 방법.
  9. 제8항에 있어서, 상기 응집성 세포 균주가 사카로마이세스 우바룸인 방법.
  10. 제5항에 있어서, 상기 발효 단계에서 세포 균주의 농도를 90 내지 160g/L로 유지하는 방법.
  11. 제5항에 있어서, 상기 발효단계를 4 내지 40℃ 범위의 온도, 3.5 내지 9 범위의 pH 및 1000rpm 이하의 필터 회전 속도의 조건에서 0 내지 40g/L의 농도의 배지성분의 존재하에 수행하는 방법.
  12. 제11항에 있어서, 상기 발효 온도가 30 내지 35℃ 범위이고, pH가 4 내지 8 범위이며, 필터 회전 속도가 500rpm 이하인 방법.
  13. 제11항에 있어서, 상기 배지 성분이 제1 또는 제2인산 암모늄, 황산 암모늄, 초산 암모늄, 요소, 효모액기스, 금속이온 봉쇄제, 인산 제1 또는 제2칼륨, 염화 칼슘 및 황산 마르네슘으로 이루어진 군 중에서 선택되는 방법.
  14. 제5항에 있어서, 상기 발효 단계가 희석률 0.5 내지 2(hr-1)에서 연속적으로 수행되는 방법.
  15. 제1 또는 제5항에 있어서, 상기 유동식 필터가 50 내지 2,000메쉬 범위의 구멍 크기를 가진 스크린 또는 멤브레인 형태로 교반기를 갖추고 있는 것인 방법.
  16. 제5항에 있어서, 상기 발효 단계에서 수득된 발효 생성물을 침강조로 이송하여 침강물과 상등액을 분리 하는 방법.
  17. 제16항에 있어서, 상기 침강물을 발효조로 재순환시켜 추가로 발효시키는 방법.
  18. 제16항에 있어서, 상기 침강물을 추가적으로 설치된 2차 발효조에서 추가로 발효시키는 방법.
  19. 제1항 또는 제5항에 있어서, 상기 발효에 사용되는 발효조가 일련 또는 다단으로 구성되고, 액당화된 혼합물이 다중으로 공급되는 방법.
  20. 제5항에 있어서, 상기 분류를 증류 또는 흡착에 의해 수행되는 방법.
