Claims (13)

  1. -150,000 정도의 분자량을 가지며,
    -RAV 비루스의 엔벨로프 당단백질(envelope glucoprotein)과 함께 풀리펩티드 서열을 함유하는 폴리펩티드 골격을 가지며,
    -RAV 비루스로 인한 SIDA또는 LAS로 감염된 환자 혈청에 의해 또는 RAV 비루스의 비계통적인 담체의 혈청에 의해 인지되며,
    -35) _시스테인으로 라벨될 수 있으며,
    -콘카나발린 A와 함께 복합체를 이룰 수 있으며, 이 복합체는 O-메틸-알파-D-만노 피라노시드의 존재하에 해리될 수 있으며,
    -"LENTYLLECTINES"의 상표명으로 공지된 것과 같은 다른 렉틴과 함께 복합체를 이를 수 있으며,
    -엔도글리코시다제, 특히 엔도-베타-N-메틸글리코사미니다제-의 작용에 감수적이며, 이 작용은 약 95,000분자량의 단백질 형성에 수반되며,
    -[14C] -글루코사민에 의해 라벨될 수 있는 특징을 갖는 항원의 제조방법에 있어서, 그 표면에서 T4 분자를 표현하며, LAV 비루스로 감염될 수 있는 하이브리드 셀 라인을 감염시키며, 이 하이브리드 셀은 인체 임파구와 종양 임파 세포 특히 MOLT-4종류를 융합시키므로써 형성되었으며, 이 하이브리드 라인의 감염 세포의 배양, 감염된 하이브리드 셀의 회수 및 용해, 생산된 항원의 분리와 약 150,000 분자량의 글리코프로틴의 회수를 특징으로 하는 상기 항원의 제조방법.
  2. 분자량이 135,000이며,
    -LAV 바이러스의 글리코프로레인과 동일한 풀리펩타이드 서열을 갖는 풀리펩타이드 구조를 지니며,
    -LAV 바이러스에 의한 LAS나 또는 SIDA에 의해 감염된 환자의 혈청에 의한 것으로 인식되거나 또는 LAV 바이러스의 무증후성담체의 혈청에 의한 것으로 인식되며,
    -35, -시스테인이거나 라벨링 되며,
    -O -메틸-알파-D-만노피라노시드 존재하에 해리될 수 있는 콘카나발린 A를 지닌 복합체를 형성할 수 있으며 ;
    -"LENTYLLECTINES"라는 상표명으로 공지된 기타의 펙틴을 지닌 복합체를 형성할 수 있으며,
    -80,000 분자량인 프로테인의 형성을 부반하는 엔더글리코시다제, 특히 엔도-베타-N-메틸글히코사미니다제-H의 작용에 민감하며,
    -[14C] -글루코사민이라 라벨링될 수 있는 항원의 제조방법에 있어서, 이 방법은 셀 표면에서 T4분자를 표현하며 LAV 바이러스에 의한 감염에 민감한 하이브리드 셀 라인(인체 T4 임파구의 임파종양 셀, 특히 MOLT-4 타잎의 융합에 의해 혈성된것)의 감염과, 감염된 하이브리드 셀의 회수 및 용해와, 생성된 항원의 분리와, 135,000 분자량을 지닌 글리코프로테인의 회수 등을 포함하는 것을 특징으로 하는 항원의 제조방법.
  3. 제1항 또는 2항에 있어서, 감염된 하이브리드 셀의 용해는 살아있는 바이러스성 입자생성으로 종결되는 감열성 절차가 끝나기 전에 수행되는 것을 특징으로 하는 방법.
  4. 제1항에 있어서, 용해는 감염후, 3시간 동안 수행되며, 150,000의 분자량을 지닌 당단백질이 회수되는 것을 특징으로 하는 방법.
  5. 제2항에 있어서, 용해가 감염후 12시간 동안 수행되며, 135,000의 분자량을 갖는 당단백질이 회수되는 것을 특징으로 하는 방법.
  6. 제1항 내지 5항중 어느 한 항에 있어서, 항원의 분리는 항원을 함유하는 매체를 콘카나발린 A 또는 렌틸-렉틴 같은 항원과 친화력이 있는 렉틴(고체 지지체상에 고정되어 있다.)과 접촉시킨뒤, O-데틸-알파-D- 만노 피라노사이드 용액으로 항원을 용출시켜 수행되는 것을 특징으로 하는 방법.
  7. 제6항에 있어서, 정제는 항원에 대해 활성인 항체를 지닌 환자의 혈청에 의해 면역침강되거나 또는 사전에 형성되어 모노클로날 항체를 지니고 있는 하이브리도마에 의해 생성된 모노클로날 항체에 의해 면역침강된 뒤, 모노클로날 항체를 분비하고, 이것을 인식함으로서 수행되는데, 여기서 항원을 분자량이 150,000 또는 135,000인 것을 특징으로 하는 방법.
  8. 제1항 내지 7항중 어느 한항에 있어서, 하이브리드 셀은 MOLT-4 라인 타잎의 셀과 T4 임파구 사이에 형성된 하이브리드 셀라인에 속하는 바이러스에 의해 초기에 감염되며, 상기 하이브리드 셀 라인은 셀 표면에서 T4 분자를 표현할 수 있으며, LAV 바이러스 또는 이와 관련된 바이러스에 의한 감염에 민감한 것을 특징으로 하는 방법.
  9. 제8항에 있어서, 사용된 하이브리드 셀 라인은 1985년 4월 15일 No. I-435로 C.N.C.M에 기탁된 것을 특징으로 하는 방법.
  10. 제1항 내지 9항에 따른 방법에 의해 제조된 항원에 생리적 액체를 접촉시킨뒤 형성된 항원 항체 복합체를 탐지하는 것을 포함하며 혈청 또는 뇌척수액 같은 인체의 생리적 액체중의 항-LAV 항체 존재를 시험관내에서 탐지하는 방법.
  11. 제10항에 있어서, 항원은 감염고정된 셀 표면에 노출되어 있는 것을 특징으로 하는 방법.
  12. 제10항에 있어서, 항원은 고체 지지체 표면에 고정되어 있는 것을 특징으로 하는 방법.
  13. 제1항 내지 9항중 어느 한항에 의한 방법으로 제조된 항원의 규정량을 적정용 마이크로플레이트 컵에 침전시키고, 이 컵속으로 혈청을 유입시켜 측정하고, 마이크로플레이트를 배양하고, 마이크로플레이트를 조심스럽게 세척하고, 마이크로플레이트 컵속으로 라벨된 혈액의 면역글로블린 항체를 유입하는데 라벨링은 기질을 하이브리드화 시킬 수 있는 것들 중에서 선택된 효소에 의해 수행되므로 항체가 적어도 규정된 파동벤드의 조사흡수를 조절하도록 하고, 병이나 또는 병을 일으킬 위험을 측정하기 위해 기질의 가수분해되는 양을 대조물과 비교하여 탐지하는 것 등을 포함하여 혈청 또는 생리학적 매체중에서 LAV 항바이러스 항체 존재를 진단하는 방법.
