生分解性粒子
本発明は、生物学的活性物質および診断用試薬の制御された送達のための生分
解性ナノ粒子およびマイクロ粒子の分野に関する。
発明の背景
生物学的活性物質の非経口投与の分野における主な試みは、制御された送達用
デバイスの開発であり、このデバイスは静脈から投与し得るほど小さく、そして
長い循環半減期を有する。制御された方法で組織または血液に投与された生物学
的活性物質は、それが溶液の形態で注入された場合と比べて、有毒な副作用の低
減が期待され、そして血漿中の不安定な化合物の分解を減少し得る。
マイクロカプセル、マイクロ粒子、リポソーム、およびエマルションを含む数
多くの注入可能な薬剤送達システムが検討されてきた。これらの注入可能な薬剤
送達物質の使用において重要な障害は、網内系(RES)のマクロファージによる血
流からの物質の速やかなクリアランスである。例えば、直径60ナノメーターほど

欧州特許出願番号第0 520 888 A1号および同第0 520 889 A1号は、生物学的活
性物質の注入可能な制御された投与のためのポリ乳酸(polylactic acid)とポリ
(エチレングリコール)とのブロックコポリマーのナノ粒子を開示している。こ
れらの出願は、薬剤放出のプロフィールを変化させるためにコポリマーをどのよ
うに改変するかも、コポリマーの改変がインビボでの送達デバイスの分布および

に小さく、そして特定の細胞または器官を標的とするために、あるいは物質の送
達速度を操作するための必要性に応じて改変され得る、生物学的活性物質の制御
された送達用の粒子を得ることが望まれる。
従って、本発明の目的は、血流から速やかに浄化されない、生物学的活性物質
の制御された送達用の粒子を提供することである。
本発明の他の目的は、特定の細胞または器官を標的とするため、あるいは物質
の送達速度を操作するために必要に応じて改変され得る粒子を提供することであ
る。
本発明の他の目的は、診断用イメージングのために磁性物質を含有する生分解
性粒子を提供することである。
本発明のさらに他の目的は、物質の制御された放出用あるいは特定の器官また
は細胞に対して必要に応じて標的にされる診断用イメージングのためのマイクロ
粒子を提供することである。
発明の要旨
網内系のマクロファージにより血流から速やかに浄化されず、そして調整可能
な放出速度を達成するために、あるいは所望する特定の細胞または器官を標的に
とするために、必要に応じて改変され得る粒子が提供される。この粒子は、生物
学的活性物質および/またはイメージング用の造影剤を含有する生分解性固体の
コア、および表面にポリ(アルキレングリコール)部分(moiety)を有する。ポリ
(アルキレングリコール)の末端ヒドロキシル基は、生物学的活性分子を含む粒
子表面に共有的に結合するために用いられ得る。生物学的活性分子は、特定の細
胞または器官を標的とする抗体、あるいは粒子の電荷、親油性または親水性に影

図2は、モノメトキシポリ(エチレングリコール)の存在下でのラクチド(lac
tide)とグリコリド(glycolide)との重合反応における、30分後、1時間後、2時
間後、および4時間後の成分のゲルパーミエーションクロマトグラムである。ラ
クチドおよびグリコリドが消費されてポリマーとなることは、ピークDの減少で
表される。時間の経過によるより低い保持時間(より高分子量)へのピークPの
シフトは、ポリ(エチレングリコール)鎖のヒドロキシル末端基で付加反応が起

図3は、重量比1:2、1:3、1:4、および1:9のPEG-PLGAについて発
熱を伴う熱の流れ(任意の単位で表現)の温度の関数としてのサーモグラムであ
る。これはPerkin-Elmer示差走査熱量計で得た。試料の重量範囲は20〜25mgであ
った。インジウムを、温度およびエンタルピー較正用に用いた。各ポリマー試料
を、毎分10℃の速度の−60〜150℃の加熱−冷却−加熱サイクルに供した。

発明の詳細な説明
1つ以上の疎水性生浸食性(bioerodible)ブロックおよび1つ以上の親水性ブ
ロックを含む、ジブロック、トリブロックおよびマルチブロックコポリマーにつ
いて開示する。これらのブロックコポリマーから形成された粒子を開示する。こ
の粒子は、網内系のマクロファージにより血流から速やかに浄化されず、そして
種々の放出速度を達成しまたは所望の特定の細胞または器官を標的にするために
必要に応じて改変され得る。これらの粒子は、広範囲な目的に対して、生物学的
活性物質を制御された方法で投与するために有用であり得る。
I.ナノスフェアおよびマイクロスフェアを構築するためのコポリマー
ナノ粒子からの物質の放出時間および放出の動力学は、ナノ粒子を製造するた
めに選択されたコポリマーあるいはコポリマー混合物またはブレンドに応じて変
化する。本明細書に所与の開示によれば、当業者ならば所望の効果を達成するた
めの適切なポリマーまたはポリマーの組み合わせを選択し得る。
A.ポリマーの選択
親水性ポリマー
ポリ(アルキレングリコール)(これは、ポリマーがグリコールの代わりに
オキシドから調製された場合はポリ(アルキレンオキシド)ともいえる)が、1
ブロックコポリマーの1つまたは複数の末端親水性ブロックとして用いられる。
本明細書において、用語ポリ(アルキレングリコール)は、式HO-[(アルキル)O]y
-OHのポリマーを意味する。ここで、アルキルは、C1〜C4の直鎖または分岐鎖
アルキル部分を意味し、メチル、エチル、プロピル、イソプロピル、ブチル、お
よびイソブチルを含むがこれらには限定されない。Yは、4を超える整数であり
、典型的には8と500との間であり、より好ましくは40と500との間である。
インビボでの結果は、PEGの分子量(MW)が高くなればなるほど、血液中での
循環時間(半減期)は長くなることを示す(図7および8を参照のこと)。
ポリ(アルキレングリコール)の具体的な例は、ポリ(エチレングリコール)
(poly(ethylene glycol))、ポリプロピレン 1,2-グリコール ポリ(プロピレン
オキシド)(polypropylene 1,2-glycol poly(propylene oxide))、およびポリプ
ロピレン 1,3-グリコール(polypropylene 1,3-glycol)を包含する。好ましい親
水性ポリマー性部分は、分子量がおよそ20,000 DaのPEGである。ポリ(アルキレ
ングリコール)の代わりに用い得る他の親水性ポリマーは、ポリピロリドン(pol

ばならない。ポリ乳酸とポリ(エチレングリコール)とのブロックコポリマーが

ている。しかし、乳酸とグリコール酸(glycolic acid)とのコポリマー、ならび
にポリ無水物(polyanhydride)、ポリオルトエステル(polyorthoester)、ポリフ
ォスファゼン(polyphosphazene)、ポリフォスフェート(polyphosphate)、乳酸の
ホモポリマー以外のポリヒドロキシ酸、特にポリヒドロキシ酪酸(polyhydroxybu
tyric acid)、ポリカプロラクトン(polycaprolactone)、あるいはこれらのポリ
マーのモノマーから調製されたコポリマーもまた、生物学的活性物質の送達用ナ
ノ粒子の調製に用いられ得ることが今や発見された。粒子を調製するために用い
られ得る物質の多様性は、放出速度の多様性およびインビボで果たされ得る放出
プロフィールを顕著に増大させる。
生分解性の疎水性ポリ無水物は、例えば米国特許第4,757,128号、同第4,857,3
11号、同第4,888,176号、および同第4,789,724号に開示されている。ポリヒドロ
キシブチレート(polyhydroxybutyrate)は、Agostini,S.,「Synthesis and Cha
racterization of PHB,」Ph.D.論文,Case Western University,U.S.A.(1971)
および米国特許第3,044,942号に開示されている。これらの刊行物の教示は本明
細書に参考文献として援用される。
好ましい実施態様においては、ポリ(乳酸−コ−グリコール酸)(poly(lactic
-co-glycolic)acid)(PLGA)のポリエステルが粒子のコアを形成する。これらのポ
リマーは、非経口投与に関してFDAにより承認されている。PLGAは加水分解を経
て分解するので、インビボ分解速度はインビトロデータから予測され得る。PLGA
は、身体内で普通に見出される物質である乳酸およびグリコール酸に分解する。
さらに、乳酸とグリコール酸のモル比およびコポリマーの分子量を操作すること
により、異なる分解パターンが得られ得る。
ナノ粒子を製造するために用いられるポリマーの分子量およびポリマーの化学
組成および立体化学的配置は、種々の有機溶媒へのポリマーの溶解性ならびにポ
リマーの結晶性に影響する。この点に関して、乳酸とグリコール酸とのコポリマ
ーは好ましい。
身体からの***を確実にするために、PEGはおよそ300と20,000ダルトンとの間
の分子量を有するべきである。PEGの分子量が350〜20,000 Daの範囲にあり、そ
してPLGAの分子量が350〜200,000 Daの範囲にあるいくつかのPEG-PLGAのジブロ
ックコポリマーが評価された。粒子の親水性および疎水性領域の分子量は、粒子
の水溶性、従って粒子の水溶液における安定性に影響することが決定された。例
として、PEG:PLGA 1:1(重量比)、(PEGの分子量=PLGAの分子量=5,000ダルトン
)は、水に溶けやすく、そしてQUELS(準弾性光散乱)を用いて決定された平均
直径約20nmのミセルの形成に用いられ得る。高密度のコアを有するより大きなナ
ノ粒子を形成するためには、このポリマーはPLGAのような疎水性ポリマーとブレ
ンドされるべきである。
水またはリン酸緩衝液(pH7.4)への溶解性は、PLGAのMWが5,000から20,000 D
aへ増加すると低下する。PLGA-PEG(20,000〜5,000 Da)を用いて、簡単な乳化
法(simple emulsion technique)を使用して平均直径120〜140nmの安定なナノス
フェアが得られた。PLGAコポリマーは、粒子における第2ポリマーとして好まし
い。なぜなら、これは酢酸エチル(ethyl acetate)およびアセトン(acetone)に可
溶であるからである。酢酸エチルまたはアセトンは、ジクロロメタン(dichlorom
ethane)およびクロロホルム(chloroform)よりも好ましい。なぜなら、それらは
インビボ適用に関して毒性が少ないからである。
ポリL-ラクチド(poly L-lactide)は、結晶度の高いポリマーである。ポリD,L-
ラクチドは結晶性が低く、そして有機溶媒により溶けやすい。D,L-ラクチドとグ
リコリドとの比が75:25のランダムコポリマーは、有機溶媒、特に酢酸エチルに
極めて溶けやすい。このコポリマーは完全に非晶質であり、このことはこのポリ
マーを制御された放出用のナノスフェアおよびマイクロスフェアの製造に有用な
ポリマーにする。
ポリ-L-ラクチドはインビトロで数ヶ月から数年の分解時間を有する。この長
い分解時間は、ポリマーを水の侵入から保護するそのより高い結晶性に起因する
。D,L-ラクチドは非晶質であるため、その分解時間は典型的には1ヶ月から数ヶ
月である。ポリグリコリドもまた結晶性構造を有し、そして1ヶ月から数ヶ月の
分解時間を有する。D,L-PLGAは非晶質であり、インビトロで数週間から数ヶ月の

B. ジブロックコポリマーの調製
ポリ(アルキレングリコール)(PAG)、好ましくはポリ(エチレングリコール)
を有するこれらの任意の疎水性ポリマーのジブロックコポリマーの調製は、多く
の方法で行われ得る。方法には、(i)ポリマーを、モノメトキシPEGまたは他の当
該技術分野で公知の酸素保護基で(1つの末端ヒドロキシル基を保護し、そして
もう一方はポリマーと反応しないように)保護したPEGのようなモノメトキシポ

マルチブロックポリマーは、D,L-ラクチドとPEGを170〜200℃で塊状共重合する
ことによって調製された(X.M.Dengら、J.of Polymer Science:Part C:Poly
mer Letters,28,411-416(1990)。3および4枝星状PEG-PLAコポリマーは、開
始剤オクチル酸スズ(stannous octoate)の存在下160℃で星状PEGにラクチドを重
合することによって作製された。K.J.Zhuら、J.Polym.Sci.,Polym.Lett.E
d.,24,331(1986)。PLA-PEG-PLAのトリブロックコポリマーは、溶媒を用いずに
2つの末端ヒドロキシル基を有するPEGの存在下で触媒としてオクチル酸スズを
用いてD,L-ラクチドから180〜190℃で開環重合することにより合成された。多分
散度(MwとMnとの比)は2〜3の範囲であった。
他の実施態様においては、ポリマーまたはモノマーは、アミノ基末端を有する
ポリ(アルキレングリコール)(Shearwater Polymers,Inc.から入手可能)と
反応してアミド結合を形成し得る。アミド結合は一般にエステル結合よりも強い
。アミド結合は、ナノ粒子表面上でポリ(アルキレングリコール)の保持時間を
長くし得る。
ポリ(アルキレングリコール)、特にポリ(エチレングリコール)を末端に有
するトリブロックまたは他のタイプのブロックコポリマーは、分岐状または他の
適切なポリ(アルキレングリコール)を用い、そして反応するべきでない末端基
を保護して、上記反応を用いて調製され得る。Shearwater Polymers,Inc.,は、
広範なポリ(アルキレングリコール)誘導体を提供する。例としては、トリブロ
ックPEG-PLGA-PEGおよび以下の構造のポリマー

PEG-PLGA-PEGのような直鎖状トリブロックコポリマーは、ラクチド、グリコリ

することによって調製され得る。トリブロックコポリマーはまた、CH3-(CH2CH2)n
-O-PLGA-OHをHO-PLGAと反応させることにより調製され得る。
ある実施態様においては、マルチブロックコポリマーは、PLAまたはPLGAのよ
うな疎水性ポリマー性部分の末端基を適切なポリカルボン酸モノマーと反応させ
ることによって調製される。ポリカルボン酸としては、1,3,5-ベンゼントリカル

たは診断用薬剤は、薬剤の送達および/または放出のために粒子内にカプセル化

めまたは粒子の標的化送達のためにブロックコポリマーに結合される。粒子は、
両方の実施態様の要素を含み得る。例えば、送達される物質、および抗体または

本明細書に記載のようにして調製される粒子は、細胞混合物中の所定の細胞に

て特定の組織に対して標的化され得る。磁性粒子も導入された場合、粒子は、組
織特異的レセプターまたは組織特異的表面タンパク質に対する抗体のようなリガ

られる。
好ましい実施態様においては、粒子は、送達される物質、およびポリ(乳酸-
コ-グリコール酸)、ポリ(乳酸)、ポリ(グリコール酸)、またはポリ無水物
を有するポリ(アルキレングリコール)のコポリマーを含む。ここで、ポリ(ア
ルキレングリコール)は抗体または抗体フラグメントに共有結合する。粒子は、
抗ガン剤のような全身投与された場合に重大な副作用を引き起こす、活性かつ有
効な薬剤の長期間放出に用いられ得る。制御された放出は、一般に非カプセル化

はまた、短い生物学的半減期を有する薬剤に関して、血漿中での分解に対する保
護を提供する。
送達または診断目的のための粒子内への物質の導入
広い範囲の生物学的活性材料または薬物を、粒子形成時にポリマーに導入し得
る。導入しようとする物質は製造中または放出プロセス中にポリマーと化学的に
相互反応すべきではない。当業者に既知の通り、物質放出のプロフィールを変更
するために、無機塩、BSA(ウシ血清アルブミン)、および不活な有機化合物を使
用し得る。生物学的に不安定な物質、例えば細菌細胞、酵母細胞、あるいはヒト
細胞を含む哺乳動物細胞のような原核または真核細胞、または細胞壁のようなそ
れらの成分、または細胞接合体もまた、粒子内に含まれ得る。生物学的活性物質
という用語は、ペプチド、タンパク質、炭水化物、核酸、脂質、多糖、またはこ

非限定的な例としては、抗原、酵素、ホルモン、レセプター、およびペプチド

リドカイン(lidocaine)またはテトラカイン(tetracaine)のような疎水性薬物
は、粒子内に捕捉され得、そして数時間にわたって放出され得る。ナノ粒子内に
おいて(重量で)40%程度の装填(loading)が達成された。しかし、疎水性物質は
さらにカプセル化が困難であり、そして一般に、親水性物質より装填効率が減少
する。
1つの実施態様においては、抗体がナノ粒子に導入される。抗原という用語は
、抗体の形成を刺激し、または細胞介在性体液性反応を引き起こすあらゆる化学
構造を意味し、これはタンパク質、多糖、核タンパク質、リポタンパク質、合成
ポリペプチド、またはタンパク質担体に連結された小分子(ハプテン)を包含す

診断を目的としても、粒子内に物質を導入し得る。例としては、放射線標識、
空気またはバリウムのような放射線不透過性物質、蛍光化合物、および磁性化合
物が挙げられる。例えば、ローダミンのような疎水性蛍光化合物は、粒子のコア
に導入され得る。親水性蛍光化合物もまた導入され得るが、カプセル化の効率は
より小さい。なぜなら、親水性物質に対する疎水性コアの適合性の方がより低い
からである。親水性物質は、水中で別に溶解されなければならず、そして多重乳
化法(multiple emulsion technique)が粒子の製造のために使用される。
導入され得る造影剤としては、超音波イメージングに特に有用なガスが挙げら
れる。ヒドロゲル形成時に、適切なガスがポリマー性物質に導入され得る。例え
ば、空気、アルゴン、窒素、二酸化炭素、二酸化窒素、メタン、ヘリウム、ネオ
ン、酸素、およびパーフルオロカーボンである。滅菌空気または酸素は、インビ
ボでの適用のための好ましいイメージング造影剤である。
別の好ましい例は、インビボでの粒子の生体分布をモニターするために使用さ
れ得るγ線標識ナノ粒子である。薬学的に許容されるいかなるγ線放射部分も使
用され得、これにはインジウムまたはテクネチウムが含まれるが、限定はされな
い。本明細書に記載の通り、磁性粒子が調製され得、あるいは表面が修飾された
磁性ナノ粒子を含む磁性ナノ粒子が商業的に購入され得るが、その表面は親水性

MRIに対する適切な物質の例としては、ジエチレントリアミン五酢酸(diethyle
ne triamine pentaacetic acid)(DTPA)およびガトペントテートジメグルミン(Ga
topentotate dimeglumine)、並びに鉄、マグネシウム、マンガン、銅、およびク
ロムのような現在使用し得るガタリニウムキレート(gatalinium chelate)が挙げ
られる。これらは典型的には、70kgのヒトに対して、0.5M/リットル溶液の14ml
に等しい用量で投与される。
CTおよびX線に有用な物質の例としては、ジアトリゾエート(Diatrizoate)お
よびイオサラメート(iothalamate)によって代表されるイオン性モノマー(30mg/m
l溶液の2.2mlの用量で投与される)、イオパミドール(iopamidol)、イソヘキソル
(isohexol)、およびイオベルソル(ioversol)によって代表される非イオン性モノ
マー(150〜300mg/ml溶液の2.2mgの用量で投与される)、イオトロール(iotrol)お
よびイオジキサノール(iodixanol)によって代表される非イオン性ダイマー、お
よびイオン性ダイマー(例えばイオキサガルテ(ioxagalte))のような静脈内投与
用のヨウ素に基づく物質が挙げられる。他に有用な物質としては、経口用のバリ
ウムが挙げられる。
III.ナノ粒子の調製および特徴付け
粒子のサイズ
本明細書に記載の通り、製造し得る粒子の典型的なサイズは、80nmと10,000nm
との間であり、最も好ましくは80nmと700nmとの間である。実施例ではナノ粒子
の製造について記載しているが、得られる粒子の直径を増大させて、1ミクロン
以上の直径を有するマイクロ粒子を形成し得る。一般的な記載において本明細書
で言及することを容易にするために、マイクロ粒子およびナノ粒子の両方のこと
を、他に指定しない限り粒子という。
本明細書に記載の通り、ナノ粒子という用語は、10nm〜1000nmの範囲のサイズ
の固体粒子を意味する。特に好ましいナノ粒子は、生分解性、生体適合性であり
、200nm未満のサイズを有し、そして送達しようとする物質を内部に取り込んだ
剛性の生分解性のコアを有する。
本明細書で使用される、用語「マイクロ粒子」は、1ミクロンより大きく1000
ミクロンまでの範囲のサイズの粒子を意味する。本明細書に記載のいずれのナノ
粒子も、所望の用途に対してより適切であれば、マイクロ粒子として代替的に構
成され得る。
粒子の構造
図1は、本明細書に記載のように調製された粒子の断面の略図である。図示さ
れている通り、粒子は、生物学的活性物質を含む生分解性の固体コア、および表
面にポリ(アルキレングリコール)部分を有する。表面のポリ(アルキレングリコ
ール)部分は、粒子表面上でのタンパク質の吸着を抑制する水に対して、高い親
和性を有する。従って、網状内皮系(RES)による粒子の認識および取り込みは抑
制される。ポリ(アルキレングリコール)の末端のヒドロキシル基は、生物学的活
性分子、または粒子の電荷、親油性、または親水性に影響を与える分子を、粒子
表面上に共有結合するために使用され得る。
ナノスフェアは、形状が球形のナノ粒子を意味する。本明細書の手順または他
の既知の手順に従って調製されたナノ粒子の形状は、電子顕微鏡のスキャンによ
って容易に測定される。球形の形状をしたナノ粒子は、血流による循環のために
好ましい。所望であれば、本粒子は、既知の技術を用いて、特定の適用にさらに
有用な他の形状に構成され得る。
分解特性
生分解性または生浸食性という用語は、本明細書中では、所望の適用(通常、
インビボ治療)において許容可能な期間内に溶解または分解するポリマーを意味
する。この期間は、25℃と37℃との間の温度のpH6〜8の生理学的溶液に曝露し
たときに、通常5年未満、そして好ましくは1年未満である。好ましい実施態様
において、ナノ粒子は、適用に応じて1時間と数週間との間の期間で分解する。
粒子の組成
本明細書中には、粒子の、多くの特定の実施態様が記載される。第1の実施態
様においては、ポリ(アルキレングリコール)とポリ(乳酸−コ−グリコール酸
)のジブロック、トリブロック、またはマルチブロックのコポリマーから調製さ
れる粒子が提供される。第2の実施態様においては、ポリ(アルキレングリコー
ル)と、ポリ無水物、ポリホスフェート、ポリホスファゼン、ポリオルトエステ
ル、乳酸のホモポリマー以外のポリヒドロキシ酸(特にポリヒドロキシ酪酸を包
含する)、ポリカプロラクトン、またはこれらのポリマーのモノマーから調製さ
れるコポリマーとのコポリマー(ここで上記コポリマーはジブロック、トリブロ
ック、またはマルチブロック構造であり得る)から調製される粒子が提供される
。例えば、この粒子は、ポリ(アルキレングリコール)−[ポリ(乳酸−コ−グリコ
ール酸)またはポリ(乳酸)]−ポリ(アルキレングリコール)の形のコポリマーから
作製され得る。さらに他の実施態様においては、粒子は、ポリ(乳酸)またはポリ
(グリコール酸)と2つまたはそれ以上のポリ(アルキレングリコール)部分とのコ
ポリマーから作製され得る。あるいは、粒子は、ポリ(乳酸−コ−グリコール酸)
、ポリ(乳酸)、またはポリ(グリコール酸)と、ポリ(アルキレングリコール)との
コポリマーから作製され得、ここでこのコポリマーはポリ(乳酸−コ−グリコー
ル酸)とブレンドされる。
粒子の調製
粒子は、ブロックコポリマーを第1の溶媒に溶解し、次いで、コポリマーの疎
水性部分が粒子のコア中に存在し、そして親水性ポリ(アルキレングリコール)部
分が粒子の表面上に存在するような様式で、溶媒系からブロックコポリマーを沈
澱させることによって調製される。必要に応じて、送達されるべき物質をカプセ
ル化する、得られる粒子中に同時に該物質をカプセル化し得る。送達されるべき
物質は、そのコポリマーまたはコポリマーブレンドと、0より大きい値から99の
比で、そしてより好ましくは1から70の比で、粒子を形成する前に混合される。
例えば、酢酸エチルまたは塩化メチレン(methylene chloride)のような適切な
溶媒でのブロックコポリマー溶液を調製する。ポリ(アルキレングリコール)ホモ
ポリマーに関しては非溶媒であり、そしてブロックコポリマーの他の1つまたは
複数のユニットのホモポリマーに関しては溶媒である有機溶媒を選択すべきであ
る。この溶液に蒸留脱イオン水を加えることによってエマルションが形成される
。有機溶媒をゆっくりと蒸発させることによって、ポリマー鎖が小滴の内部およ
び表面上に再編成される。有機溶媒に溶解しないポリ(アルキレングリコール)は
、水相へと移動する傾向にあり、他方、水に溶解しないコポリマーの他のユニッ
トは、小滴内部に残り、そして溶媒が全部蒸発した後、粒子のコアを形成する。
コア内部にPEG鎖が存在することは避けるべきである。なぜならこれはコアによ
る水の吸収を誘導し得、次いでカプセル化された物質の加速され、かつ制御され
ない放出が起こるからである。
有機溶媒を除去した後、粒子は遠心分離によって水相から単離される。この粒
子は、後で容易に水中に再分散され得る。
他の実施態様においては、アセトン、メタノール(methanol)、またはエタノー
ル(ethanol)およびそれらの水溶液を、蒸留脱イオン水の代わりに用い得る。一
般に、水が好ましい。なぜなら、水は、粒子の表面へより高濃度のポリ(アルキ
レングリコール)を持ってくる力があるからである。しかし、水に対して鋭敏な
コポリマーまたは第2のポリマー(例えば、ポリ無水物)の沈澱溶媒として、ア
セトンを使用し得る。
他の実施態様において、粒子を形作る前にブロックコポリマーを第2のポリマ
ーとブレンドし(例えば、PLGA-PEGをPLGAまたはPLAと混合する)、粒子に別の
特性を与える(例えば、インビボにおける半減期の変更)。PLGA PEGを他のポリ
マーに加えることで、インビボにおける半減期が増大する。
典型的な実施態様において、第2のポリマーは、ブロックコポリマーと、0よ
り大きな値から100までの比で混合される。
光散乱による研究によれば、得られる粒子のサイズは、有機相の粘度、有機相
と水相との比、および超音波処理の力および時間によって決定されることが示さ
れている:粘度が高くなると、より大きな粒子が生じ、そして水相の容積が有機
相に比べて高いと、より小さな粒子が生じる。超音波処理の力および時間の効果
は以下の通りである:25mgのポリマー/2mlのCH2Cl2を30mlの0.3%ポリビニルア
ルコール(polyvinyl alcohol)溶液に加える。この混合物にボルテックスを最大
強度で30秒間かけ、次いで、プローブ超音波処理器で出力7で30秒間、超音波処
理する。この条件で、15nmの粒子が再現性良く生じる。このパラメーターは狭い
単分散(unimodal)サイズ分布で平均サイズが約150nmのナノスフェアを得るとこ
ろまで最適化され得る。
粒子の表面特性の改変
他の実施態様において、ポリ(アルキレングリコール)を、粒子の電荷、親油性
または親水性に影響を与える化合物に結合させ得る。例えば、ポリ(アルキレン
グリコール)以外のポリマーを表面親水性コーティングとして使用する。任意の
生体適合性親水性ポリマーをこの目的のために使用し得、このポリマーにはポリ
(ビニルアルコール)が包含されるが、これには限定されない。粒子はまた、デキ
ストランによってコーティングされ得る。デキストランは一般にポリ(アルキレ
ングリコール)よりも親水性が高いが可撓性は低い。デキストランでコーティン
グされたナノ粒子は、磁気共鳴イメージング法(magnetic resonance imaging(MR
I))のために特に好適である。
生物学的活性分子に共有結合したポリマーからの粒子の調製
さらに、他の実施態様において、生物学的活性分子(例えば、抗体またはFab
またはFab2のような抗体フラグメント)または標的分子に共有結合したブロック
コポリマーから粒子を作製する。ここでこの粒子は、生物学的活性分子が粒子の
表面の外側に存在するように調製される。生物学的活性分子は、タンパク質、炭
水化物または多糖類、核酸、脂質、これらの組合せ、または合成分子であり得、
上記のような有機材料または無機材料を包含する。
IV.検出可能な試薬を含む粒子の検出
この粒子はインビボでの安定性が高いので、肝疾患および腎疾患、ファロピー
オ管疾患における脈管のイメージング、腫瘍塊および組織の検出および特徴付け
、および末梢血速度の測定、ならびにもっと標準的な適用のために有用である。
患者の内部の粒子中の気泡を検出することによるイメージングのための方法は
、超音波音響学的エネルギーの実例であり、所定の振動特性を有するパルスを発
生させる振動子を使用する。第1のパルスは時間と共に振動数が増加し、そして
第2のパルスは時間と共に振動数が減少する。イメージング装置は、第1および
第2のパルスへの曝露の後、検体内の領域のイメージを生じさせる。
血流中の気泡の存在を測定するための通常の手法は、血液によって反射された
超音波音響学的エネルギーの振動数におけるドップラーシフトを利用する。ドッ
プラー気泡シグナルは、気泡の半径が増大するにつれてほぼ比例的に増大する。
ヒトの聴覚機構は、気泡シグナルが存在するか存在しないかを認識するための最
も正確なプロセッサーであると考えられる。この理由から、ドップラー血流モニ
ター装置を用いて満足すべき結果を得るためには、熟練したオペレーターがいる
ことが好ましい。
空気充填粒子がインビボイメージングのために有用であるかどうかを決定する
ために、以下のインビトロ法を用い得る。
上記方法によって調製された粒子を、ふたをした組織培養チューブ中で懸濁さ
せる。超音波イメージングのために、振動子(transducer)上方のカップリング媒
体で覆われたパッドの上部に、このチューブを置く。振動子は、いかなる人工的
な動きをも最小にするために、およそ入射角90°の位置に維持される。振動子は
送信器として働き、そしてまたチューブから散乱して戻ってくる超音波照射を受
信する。B−モードおよびドップラーイメージがポリマー性マイクロカプセルで
満たされたチューブについて確立され、そして得られたイメージを、緩衝液のみ
を入れたチューブからのイメージからなるコントロールと比較する。ディスプレ
イのBモードは、スキャンされたチューブのスライスの二次元イメージを与える
。この結果は、ドップラーイメージング手法(以下で、述べる)によって示され
るように、インビボでの結果と良好に相関する。インビトロおよびインビボのデ
ータは、実施例において高度に相関するので、この試験はマイクロ粒子のインビ
ボ安定性を合理的に予想させる。
検出手段の別の例にとしては、PET(陽電子放射断層法(positoron emission to
mograph))、CAT(コンピューター断層撮影法(computer assisted tomograph))、
X線、蛍光板透視法(fluoroscopy)、およびMRI(磁気共鳴イメージング法)が包含
される。これらは、標準的な手法および装置を用いて、他の市販の造影剤を用い
るのと同様に行われる。
超音波、磁気共鳴イメージング法(MRI)、コンピューター断層法(CT)、X線の
ような、より一般的な手法を用いるイメージングにおいて有用な粒子と同様の粒
子が、より一般的ではない陽電子放射断層法(PET)およびシングルフォトン放射
コンピューター断層法(single photon emission computerized tomograpy(PET))
においても有用である。
本明細書で記載されるようにして調製されるナノスフェアは、ヒトにおけるガ
ンマ−シンチグラフィ試験に使用されているインジウム111で標識され得る。イ
ンジウム111は半減期が短いため(6時間)、廃棄および汚染の問題を低減する
。インジウム111はまた、他の標識に比べてエネルギー放射が低い。これらの特
性が、インジウム111を、インビボでの研究のためにナノスフェアを標識する化
合物として興味深いものとしている。
放射活性標識は、まずInとジエチルトリアミノペンタ酢酸(diethyltriaminope
ntaacetic acid(DTPA))とを、両化合物を水性エタノール中で混合することによ
って錯形成させ、次に、EDCによって、この錯体の1級アミノ基を粒子表面のカ
ルボキシ基(表面上のPLGAのエステル基は、遊離のカルボン酸を生成するために
部分的に加水分解されるべきである)と反応させることによって、すでに調製さ
れたナノスフェアの表面に付着され得、表面標識された粒子を提供し得る。ある
いは、標識は、製造の際にコア中に導入される。標識の脱離は、ガンマ−シンチ
グラフィによってインビボでの器官分布を評価するためのデバイス中に十分な放
射活性が保持されるに十分ゆっくりであるべきである。
V.粒子の薬学的投与
本明細書に記載されている粒子は、種々の経路(例えば、経口、非経口、静脈
内、皮内、皮下、または局所)によって、液体、クリーム、ゲル、または固体形
態で、患者に投与され得る。
粒子は凍結乾燥され、そして次に、使用前に、マイクログラム/mlから100mg/m
lまでの範囲で水性懸濁液に処方される。あるいは、粒子は、ペースト、軟膏、
クリーム、またはゲル、あるいは経皮パッチに処方される。
薬剤送達のためには、粒子は、送達すべき物質を、患者に治療的に有効量の化
合物を送達するに十分であり、かつ患者に重篤な毒性効果を引き起こすことのな
い量で含むべきである。粒子中の活性化合物の所望濃度は、薬剤の吸収、失活、
および排出速度、ならびに粒子からの該化合物の送達速度に依存する。投薬量は
また、軽減すべき状態の重篤度に応じて変化することもまた留意すべきである。
いかなる特定の被験体についても、特定の投薬養生法(dosage regimen)は、個体
の要求、およびこの組成物を投与する人物または投与を監督する人物の専門的な
判断に従って、経時的に調節すべきであることもまた理解されるべきである。
粒子の放出速度および所望の投薬量に依存して、粒子は、1回で投与され得、
あるいは種々の時間間隔で投与されるべき、より少量の複数の投薬量に分割され
得る。
本明細書に記載されるようにして調製される制御送達デバイスはまた、眼への
長期の薬剤放出のための眼内挿入物(ocular insert)としても使用され得る。
VI.移植可能な(implantable)デバイスのコーティング
本明細書に記載されているように調製されるポリマーはまた、ステント、カテ
ーテル、人工脈管移植片、およびペースメーカーのような移植可能なデバイスを
コーティングするために使用され得る。このデバイスは、粒子の凍結乾燥粉末に
よってコーティングされ得、あるいは当該分野で公知の別の方法によってコーテ
ィングされ得る。このコーティングはまた、抗生物質、抗炎症薬、または抗凝固
剤を所定の速度で放出し、移植されたデバイスに関連する合併症を予防するため
に、使用され得る。
粒子の非薬学的用途
この粒子の非薬学的用途には、食品添加物(安定剤および分散剤または他の粘
度調節剤を包含する)の送達、農薬、除草剤、殺虫剤、肥料、およびフェロモン
の制御されかつ選択的な送達、ならびに印刷およびインク工業における着色剤お
よびインク処方での使用が包含される。
実施例

この詳細な説明によって、当業者は、ナノスフェアの製造に適切な広範囲な種
類のマルチブロックコポリマーの製造方法、コポリマーへの抗体および抗体フラ
グメントの共有結合方法、粒子からのナノスフェアの調製方法、および生物学的
物質および診断薬のナノスフェアへの導入方法を理解する。
材料および方法
低毒性のオクチル酸スズをICNから購入した。D,L-ラクチドをAldrich Chemica
l Companyから購入し、グリコリドをPolysciences,Inc.から購入した。これら
の化合物は、使用前に酢酸エチルから再結晶化した。分子量5,000、12,000およ
び20,000の高純度モノメトキシPEG(M-PEG)をShearwater Polymers,Inc.から
購入した。ポリマーの数平均分子量を、Phenomenexから入手した5μmの粒子を
充填した混合床Phenogelカラムを備えた、LC-25屈折率検出器を有するPerkin-El
mer GPCシステムを用いて決定した。クロロホルムを毎分0.9mlの流量で溶離剤と
して用いた。分子量を、Polysciencesからの狭い分子量のポリスチレンおよびポ
リ(エチレングリコール)標準と比較して決定した。
熱転移データをPerkin-Elmer DSC-7(Newton Center,MA)で採集した。試料の
重量は、20mg〜25mgの範囲であった。インジウムを温度およびエンタルピーの較
正に用いた。各試料を、毎分10℃の速度で-60℃〜150℃の加熱−冷却−加熱サイ
クルにかけた。広角X線回折スペクトルを、ニッケルフィルターを用いたCu Kα
源を用いて、S=0.05で、Rigaku Corporation(Danvers,MA)から入手したRigak
u Rotaflex Diffractometerを用いて得た。データをMicro Vax IIコンピュータ
で分析した。IRスペクトルを、塩化ナトリウム結晶上で融解して薄膜としたポリ
マー粉末を用いて、Nicolet 500スペクトロメータで記録した。重クロロホルム
中に溶解した試料について、Nicolet NT-360スペクトロメータを用いて13C NMR
の研究を行った。VGデータシステムを用いてピークフィッティングを行った。
実施例1:PLGAおよびPEGを連結することによるPLGA-PEGの調製
モノメトキシ−PEG(M-PEG)のヒドロキシル末端基をPLGAのカルボキシル末端
基と反応させることによってジブロックコポリマーPLGA-PEGを形成した。PLGA(
0.1mmol)およびM-PEG(1.2mmol)を塩化メチレンとジメチルホルムアミドとの
5:2混合物中に溶解した。1.2mmolの1-エチル-3-(3'-ジメチルアミノプロピル
)カルボジイミド(1-ethyl-3-(3'-dimethylaminopropyl)carbodiimide)(EDC)
およびDMAPの当量を添加した。室温で24時間反応させた後、ポリマーを水で沈澱
させ、未反応PEGおよびカップリング試薬を除去した。遠心分離によって固体生
成物を収集し、水で3回洗浄した。GPCによって検出されたように、約10%のジ
ブロックコポリマーが形成された。
実施例2:乳酸およびグリコール酸のPEGへの重合によるPLGA-PEGの調製
欧州特許出願第0 520 888A1号および第0 520 889 A1A号は、PLA-PEGジブロッ
クコポリマーが、溶媒としてトルエンを用い、触媒としてオクチル酸スズを用い
ることによって114℃でPEG乳酸から調製され得ることを開示している。この方法
はPLGA-PEGの一連のジブロックコポリマーを合成するために用いられ、種々の鎖
長のPEGで開始し、反応混合物中のPEGの初期量を減少させることによってPLGAの
鎖長を連続的に増加させた。
シリコン化ガラス製品(siliconized glassware)製品全体を130℃で一晩加熱
し、使用前に真空下で冷却した。ラクチド(4g)およびグリコリド(1g)を、
重合反応の前日に乾燥酢酸エチルから再結晶化し、真空下で一晩乾燥させた。モ
ノマーを重合フラスコに添加し、種々の量(0.5gから5g)のPEGと共にアルゴン
下で乾燥トルエン(10ml)に溶解させた。混合物を114℃に加熱し、次に、トル
エン中に溶解したオクチル酸スズ(10mg)を混合物に添加した。
試料を定期的に収集し、GPCで分析し、反応の進行を測定した。図2は、モノ
メトキシポリ(エチレングリコール)存在下での、ラクチドおよびグリコリドの
重合反応における成分の30分後、1時間後、2時間後および4時間後のゲルパー
ミエーションクロマトグラムである。ラクチドおよびグリコリドが消費されてポ
リマーとなることは、ピークDの減少によって示される。時間と共にピークPが
低保持時間(高分子量)に移行することは、ポリ(エチレングリコール)鎖のヒ
ドロキシル末端基で生じている付加反応を示す。
反応溶液中のPEGの割合が高いほど、反応が速いことが測定された。例えば、P
EG(NW5000)がラクチドおよびグリコリドとPEG対モノマーが50:50の重量比で反応
するとき、反応は20分未満で完了する。PEG(MW5000)がラクチドおよびグリコリ
ドとPEG対モノマーが10:90の重量比で反応するとき、反応は、約5時間で完了す
る。
開始物質が消費された後、反応混合物を0℃に冷却し、トルエンを除去するこ
とによって反応を停止させた。ポリマー生成物を塩化メチレン中に再溶解させ、
ヘキサンで沈澱させることにより精製した。3回の再沈澱の後、ポリマーを水で
洗浄した。
図3は、Perkin-Elmer示差走査熱量計で収集した、重量比1:2、1:3、1
:4および1:9のPEG-PLGAについての温度(℃)変化を示すグラフである。試料
重量は、20mgから25mgの範囲であった。インジウムを温度およびエンタルピーの
較正に用いた。各サイクルを、毎分10℃の速度で、-60℃から150℃の加熱−冷却
−加熱サイクルにかけた。ランダム乳酸−エチレンオキシドコポリマーは、2つ
の異なるTgを示し、このことは、ポリマー内での相分離を示唆する。この場合に
観察される単一なTgは、容易に相分離され得ないポリマー中の長いPEGおよびPLG
A鎖のからみ合い効果のためであり得る。
ポリマーを特徴付ける核磁気共鳴により、生成物が純粋なジブロックPEG-PLGA
であることが確認された。このポリマーを非経口投与用のナノスフェアを形成す
るために用いた。
実施例3:抗体で末端封止したブロックコポリマーの調製
PLGAにおける末端-COOH基を、CH2Cl2中のEDCおよびN-ヒドロキシスクシンイミ
ドを用いて、N-ヒドロキシスクシンイミドエステルで活性化する。活性化された
物質を単離後、水に入れた抗体をDMF、CH3OH、またはDMSO水溶液中のPLGA-ヒド
ロキシスクシンイミドエステルの溶液に添加する。24時間後、反応混合物をHPLC
(イオン交換、ゲル濾過、または逆相)に供し、所望の結合体を分画した。
実施例4:PLGA-PEGのナノスフェアの調製
立体的に安定化された粒子を、ジブロックPLGA-PEGコポリマーあるいはPLGAと
PLGA-PEGとのブレンドから調製した。これらのポリマーを通常の溶媒(酢酸エチ
ルあるいは塩化メチレン)に溶解した。混合物を30秒間ボルテックスし、音波処
理することによって水中油エマルションを形成した。次いで有機溶媒を、2時間
静かに撹拌することによって室温でゆっくりと蒸発させた。溶媒をゆっくりと除
去することによって、小滴の内部および小滴表面上にポリマー鎖が再編成された
。この場合、より親水性の高いPEG鎖が水の界面に向かって移動し、より疎水性
の高いPLGA鎖が小滴内部に残り、ナノスフェアの核を形成した。
有機溶媒を除去した後、ナノスフェアを遠心分離により水相から単離した。こ
のナノスフェアは、後で水中に容易に再分散され得る。
PEGがナノスフェアの表面に存在していることを確認するために、凍結乾燥さ
せた粒子の表面組成をX線光電子分光測定(XPS)によって決定した。図4は、3
00Wの電力のMgK-αX線を用いた、PLGA-PEGの凍結乾燥粒子の表面組成のX線光
電子スペクトルである。ナノスフェア粉末を分析すると、炭素1sのエンベロー
プが観察された。1つの顕著な炭素環境がPEGについて観察され、これはエーテ
ル炭素C-Oに対応する。PLGAポリマーのスペクトルは、3つの顕著な炭素環境を
示している。これらのピークはすべてPEG-PLGAナノスフェアのXPSスペクトルに
おいて明白であるが、PEGピークが顕著である。XPSから得られる情報はナノス
フェアの表面層(深さ約5nm)に対応するので、このスペクトルはPEGがナノスフ
ェア粉末の表面に集中していることを示している。
さらに、蒸留水中で24時間インキュベーションした後、PEGはナノ粒子の表面
に残ることを確認した。ナノスフェアを遠心分離および凍結乾燥によって回収し
た。XPS分析は、時間が経つにつれてPEGピークがPLGAに対応するピークと比較し
て低減していることを示しており、このことはPEGの全体ではなく一部が除去さ
れたことを意味している。
図5は、リン酸緩衝液中で37℃で24時間のインキュベーションを行っている間
にナノスフェアの表面から除去されるポリ(エチレングリコール)の画分のグラ
フである。このインビトロデータは、これらの状況下でPEG含有量全体の4%未
満がナノスフェアの表面から除去されているというXPSによる観察を支持してい
る。
実施例5:活性成分を含有するPLA-PEGナノスフェアの調製
2mlの塩化メチレン、25mgのリドカインおよび25mgの実施例2のジブロックポ
リマーの溶液を調製した。溶液を撹拌することによって塩化メチレンを除去した
後、ナノスフェアを遠心分離によって単離し、それによってリドカイン装填され
た直径15nmの粒子を得た。
実施例6:薬剤放出特性
リドカインおよびプレドニゾロン(prednisolone)(Sigma)をカプセル化のため
に選択した。なぜならそれらは、低水溶性(水中で5mg/mL未満と定義される)で
あり、有機溶媒中で高溶解性(塩素化炭化水素、テトラヒドロフラン(tetrahydro
furan)、ジメチルホルムアミド(dimethyl formamide)またはジオキサン(dioxine
)などの有機溶媒中で20mg/mLより高いと定義される)であり、そしてUV分光測光
法により検出容易だからである。
放出試験は、異なる量(20%wt)33%wt)のリドカインを装填したナノスフェア
を用いて、リン酸緩衝液(PBS、pH7.4)中、37℃で行った。透析膜(50,000カット
オフ)を凍結乾燥ナノスフェアの懸濁液(10mg/5mlPBS)で満たし、そして次いで2
5mlのPBS中に置いた。試料を外側の溶液から取り、次いでその度に新しい溶液で
置き換えた。薬剤放出は、240nmの分光測定で検出した。
PEGでコーティングされたナノスフェアの放出特性、特にナノスフェア表面上
のPEGの存在の影響、および薬剤放出動力学に関するナノスフェアコア組成(ポリ
マーおよび薬剤の特性、薬剤装填)の影響を調査するためにインビトロ研究を行
った。ナノスフェアの懸濁液は、凍結乾燥粒子をさらなる添加物を用いずにボル
テックスにより水溶液中に再分散することにより容易に得られた。リドカインを
モデル薬剤として用いた。リドカインの放出は、直鎖状PEG-PGLAコポリマーから
作製された粒子において試験された。
これらの粒子は、数時間にわたってインビトロで連続放出を示す。分子量は、
PEG-PLGAナノスフェアの放出パターンに影響を与えない。なぜなら、薬剤は、分
子量5、12、20KDaのPEGとのコポリマーを用いて約10時間以内で完全に放出され
るからである。ナノスフェア表面上のPEGの存在は、薬剤放出を改変することが
期待されない。10時間以内で、リドカインの90%以上がPLAナノスフェアから放
出された。
PEG-ε-ポリカプロラクトン(PEG-ε-polycaprolactone)から作製されるナノス
フェアからの薬剤放出は、二相性(biphasic)である。
ポリマー浸食のため、ポリ無水物から作られるコアがより速い薬剤放出を導き
得ることが通常期待される。しかし、最初の2時間における初期の速い放出の後
、薬剤放出はプラトーに到達したが、次の8時間の間は薬剤は一定速度で放出さ
れた。
ポリマー分解の動力学もまたインビトロで調査した。PEG-PLGAおよびPEG-PCL
では、ポリマーは数週間後分解を開始する。ポリ無水物から作製されるナノスフ
ェアコアは即座に分解を開始する。前者の場合では、薬剤放出は、拡散メカニズ
ムに支配される。なぜなら、薬剤は、ポリマー分解が起こる前に完全に放出され
得るからである。ポリ無水物では、ポリマー浸食が薬剤放出に影響し、そして薬
剤の特性は、放出の動力学においてより重要な役割を有する。粒子の小さなサイ
ズと大きな表面領域は、スラブ(slab)のような他の薬剤送達系に比べてポリマー
浸食の速度を増加し、そしてその後、薬剤溶解度が溶解動力学を支配する。
薬剤装填量は、放出動力学に強い影響を有し得る。33%wtのリドカインを含む
PEG-PLGAナノスフェアは、12時間にわたり薬剤を放出し得る。驚くべきことに、
10%の薬剤を装填した粒子は、6時間以内で完全な薬剤の放出を示し得る。薬剤
装填が増加すると、DSCにより示されるように、装填薬剤の一部のコア中での再
結晶化を起こし得る。リドカインのような疎水性薬剤の結晶の存在は、放出の動
力学を遅延し得る。薬剤装填ナノスフェアについて行われたESCAによる研究によ
って、薬剤結晶がナノスフェア表面上に位置しなかったことが確認された。ポリ
マー組成物もまた改変され、そして薬剤装填は45%wtまで増加した。
実施例7:111In標識されたナノ粒子のインビボにおける生体分布の評価
錯体形成によりPLGAおよびPEGポリマー鎖にインジウム111を直接付着させた。
Inおよびジエチルトリアミノペンタ酢酸(diethyltriaminopentaacetic acid;D
TPA)をステアリルアミン(stearylamine)と反応させる。得られた化合物であ
るIn-DTPA-ステアリルアミドは、PLGA核と相互反応するに十分疎水性である。こ
の場合、PLGAおよびPEGのMwは、相互反応にほとんど影響を及ぼさない。PBSまた
はウマ血清中において37℃で24時間より長くインキュベートした後、遠心分離後
の上清溶液の放射活性を測定することから評価されるように、標識のロスは全く
生じなかった。従って、この標識方法は、血液および/または異なる器官中の放
射活性の、ガンマシンチグラフィーまたは直接測定による、インビボ研究にとっ
て有用である。
111In標識された、コーティングされていないPLGAナノスフェアおよびPEGコー
ティングされたPLGAナノスフェアをBALB/Cマウス(18〜20g)の尾の静脈に注入す
ることにより、予備生体分布実験を行った。コーティングされていないPLGAナノ
スフェアの注入から5分後に、ナノスフェアに関連する111In放射活性の40%が肝
臓において、そして約15%が血液中において見い出された。PEGコーティングされ
たナノスフェアの場合、結果は逆であった。すなわち、注入された放射活性のう
ち15%が肝臓において、60%が血液中において見い出された。4時間後、ナノスフ
ェアの30%は、まだ血液中で循環していたが、コーティングされていないものは
、血液中から完全になくなっていた。
図6は、肝臓および血液中のPLGAおよびPEG-PLGAの生体分布を、注入された用
量に対するパーセンテージの関数として分単位で示すグラフである(白四角:肝
臓内の12kDa PEGを有するPEG-PLGA;黒菱形:肝臓内のコーティングされていな
いPLGA;黒四角:血液中の12 k Da PEG-PLGA;および白菱形:血液中のコーティ
ングされていないPLGA)。図示されるように、PEGコーティングされたPLGAナノ
スフェアは、長時間血液中に残留する一方、コーティングされていないPLGAナノ
スフェアは、肝臓に集中し、そこで分解され、浄化した。
図7は、BALB-Cマウス内のコーティングされていないPLGAおよびPEG-PLGAナノ
スフェアのクリアランスを、組織内における注入された用量に対するパーセンテ
ージの関数として分単位で示される時間に対して示すグラフである(白四角:PL
GA;黒四角:12 k Da PEGを有するPLGA-PEG;黒三角:5 k Da PEGを有するPLGA-
PEG;および白丸:20 k Da PEGを有するPLGA-PEG)。図示されるように、コーテ
ィングされていないナノスフェアは、すばやく浄化し、一般に、PEGの分子量が
大きいほど循環時間は長い。
図8は、BALB-CマウスにおけるコーティングされていないPLGAおよびPEG-PLGA
ナノスフェアの肝臓内での蓄積を、組織内における注入された用量に対するパー
センテージの関数として分単位で示される時間に対して示すグラフである(白四
角:PLGA;黒四角:12 k Da PEGを有するPEG-PLGA;黒三角:5 k Da PEGを有す
るPLGA-PEG;および白丸:20 kDa PEGを有するPLGA-PEG)。コーティングされて
いないPLGAナノスフェアは、肝臓内において、コーティングされていない粒子よ
りもすばやく蓄積した。
本発明を、好適な実施態様を参照して記載してきた。本発明の改変および変更

Claims (1)
- 【特許請求の範囲】 1.コアと表面とを有する粒子であって、 ポリ(アルキレングリコール)と、ポリ無水物、ポリオルトエステル、乳酸ホ モポリマー以外のポリヒドロキシ酸、ポリシロキサン、ポリカプロラクトン、お よびこれらのポリマーのモノマーから調製されるコポリマーからなる群から選択 される生分解性ポリマーとのブロックコポリマーを含有し、 該コポリマーの生分解性部分が得られる粒子のコア中に存在し、そしてポリ( アルキレングリコール)部分が、該得られる粒子の表面部分に存在する、粒子。 15.コアと表面とを有する粒子の製造方法であって、以下の工程を包含する 、方法: (i)ポリ(アルキレングリコール)と、ポリ無水物、乳酸ホモポリマー以外の ポリヒドロキシ酸、ポリオルトエステル、ポリシロキサン、ポリカプロラクトン 、およびこれらのポリマーのモノマーから調製されるコポリマーからなる群から 選択される生分解性ポリマーとのブロックコポリマーの溶液を調製する工程;お よび、 (ii)該コポリマーの生分解性部分が得られる粒子のコア中に存在し、そしてポ リ(アルキレングリコール)部分が得られる粒子の表面部分に存在するように、 溶媒系からポリマーを沈殿させる工程。 22.患者に物質を送達する方法であって、 それを必要とする患者に送達されるべき治療的な量の物質;ならびに、ポリ( アルキレングリコール)と、ポリ無水物、ポリオルトエステル、乳酸ホモポリマ ー以外のポリヒドロキシ酸、ポリシロキサン、ポリカプロラクトン、およびこれ らのポリマーのモノマーから調製されるコポリマーからなる群から選択されるポ リマーとのコポリマー;を含有する粒子を含む組成物を患者に投与する工程を包 含し、 該コポリマーの生分解性部分が得られる粒子のコア中に存在し、そしてポリ( アルキレングリコール)部分が得られる粒子の表面部分に存在する、方法。
