これらおよび他の必要性に応じるために、本明細書では、以前に開示されている製剤に比べて、熱安定性が向上し、撹拌ストレスに対する安定性も向上した、ポックスウイルスを含む組成物について記載する。前記組成物に含まれるポックスウイルスは、その効力および感染力を保持する。これは、約6.0~8.5の間の範囲のpHを有する緩衝液中のポックスウイルスに、硫酸塩、好ましくは、硫酸ナトリウムを約5mM~300mMの間の濃度で加えることにより実現された。こうした組成物は、たとえば、室温で、-20℃の貯蔵温度で、または加速ストレスに従って貯蔵したとき、示されるウイルスの凝集および立体構造変化がより少なく、貯蔵後のpH変化がより少なく、かつ/または濁り度の変化がより少ないことによって、当技術分野で公知の従来の組成物より改善されている。詳細には、本発明は、より長期間にわたり、凍結条件の外部で、25℃で数週間もしくは数カ月、または2℃~8℃の貯蔵寿命を有するという目標とする製品プロファイル特性を実現することが予想される。本発明では、0℃未満、詳細には-20℃で数週間もしくは数カ月またはより長期間の貯蔵寿命も実現される。
添付の図面は、明細書中に組み込まれ、その一部を構成するが、本発明のいくつかの実施形態を例証し、詳細な説明と共に本発明の原理を説明するのに役立つ。本発明は、図面に示す厳密な実施形態に限定されないと理解されるべきである。

本発明によるポックスウイルスとは、ヒトに感染可能なポックスウイルス属のいずれか(たとえば、オルソポックスウイルス、アビポックスウイルス、パラポックスウイルス、ヤタポックスウイルス、モルシポックスウイルス)を指す。
種々の実施形態において、本発明に適する組換え体の作製に使用されるMVAまたはMVAは、MVA-572、MVA-575、MVA-I721、ATCC(登録商標)VR-1508(商標)として寄託されているMVA、ATCC(登録商標)VR-1566(商標)として寄託されているMVA、ACAM3000 MVA、MVA-BN、または同様に弱毒化されたいずれかのMVA株である。好ましい実施形態では、組換え体の作製に使用されるMVAは、MVA-575、ATCC(登録商標)VR-1508(商標)として寄託されているMVA、ATCC(登録商標)VR-1566(商標)として寄託されているMVA、ACAM3000 MVA、およびMVA-BNである。組換え体の作製に使用されるMVAは、MVA-BNであることがより好ましい。

ある特定の実施形態では、硫酸塩は、一価カチオン塩である。本発明による一価カチオンは、たとえば、ナトリウムカチオン、カリウムカチオン、マグネシウムカチオン、またはアンモニウムカチオンである。
本発明による組成物は、約6.0~8.5の間、好ましくは、約6.5~8.5の間のpHを有する。
本発明の組成物は、医薬的に許容されるかつ/または承認されている1種または複数の担体、添加剤、抗生物質、保存剤、アジュバント、酸化防止剤、希釈剤、および/または安定剤をさらに含んでもよい。このような補助物質は、限定はしないが、1種または複数の糖、糖アルコール、ポリオール、界面活性剤(detergent)、1種または複数のアミノ酸、増粘剤(viscosity enhancer)、1種または複数の追加の塩、酸化防止剤、エタノール、EDTAなどである場合がある。
一部の実施形態では、本発明の組成物は、宿主に生理的に許容されるイオン強度を有する。組成物に塩を含める目的は、所望のイオン強度またはモル浸透圧濃度を実現して組成物を安定化させること、ならびに組成物をワクチンとして注射する部位における副作用または局所反応を軽減することである。一部の実施形態では、本発明の組成物は、約200mOsm/L~約1000mOsm/Lの範囲、約200mOsm/L~約900mOsm/L、約200mOsm/L~約800mOsm/L、約200mOsm/L~約700mOsm/L、約200mOsm/L~約600mOsm/L、または約200mOsm/L~約500mOsm/Lの範囲である合計モル浸透圧濃度(溶液中の分子の総数)を有する。一部の実施形態では、本発明の組成物は、約250mOsm/L~約450mOsm/Lの範囲であり、好ましくは、約300mOsm/Lの合計モル浸透圧濃度を有する。記載したとおりのモル浸透圧濃度によって、組成物が、非経口、特に、筋肉内または皮下注射に適するものになりながら、2℃~8℃またはより高い温度での長期の貯蔵が容易になる。イオン強度への寄与は、緩衝性化合物によって生じたイオンだけでなく、非緩衝性塩のイオンからももたらされる場合がある。溶液のモル浸透圧濃度およびイオン濃度は、ヒト身体のものと一致する(等張性である)ことが好ましい。
一実施形態では、本発明は、a)ポックスウイルスと、b)緩衝液と、c)約5mM~約300mMの間の濃度の硫酸塩とを含む、約6.0~約8.5のpHを有する組成物であって、ポックスウイルスは、MVAであることが好ましく、ポックスウイルスは、フィロウイルス抗原を発現するMVAであることがより好ましい、組成物を提供する。
原体を含有する液体ポックスウイルスの調製
以下の実施例で使用するようなMVA-BN-FILO(MVA-mBN266)は、詳細には、国際公開第2016/034678号パンフレットに記載されている。MVAは、3種のフィロウイルス糖タンパク質、すなわち、エボラウイルスSudan(GP-SEBOV)、エボラウイルスザイール-メインガ(GP-ZEBOV-メインガ)、およびマールブルクウイルス・ムソーク(GP-MARV-ムソーク)の糖タンパク質、ならびにエボラウイルスアイボリーコースト(NP-EBOV-CdI)の核タンパク質をコードする。GP-ZEBOV-メインガおよびGP-MARV-ムソーク用の発現カセットを、遺伝子間領域IGR148/149に挿入した。GP-SEBOVおよびNP-EBOV-CdI用の発現カセットは、国際公開第2016/034678号パンフレットに記載されているとおりに、ポックスウイルスプロモーター下の遺伝子間領域IGR88/89に挿入した。組換えウイルスは、当業者公知の標準の方法を使用して精製した。ウイルス濃度が5×108InfU/mLであるMVA-BN-FILO原体(BDS)を、40mMのNaClを含有し、pH7.7である10mMのトリス中で貯蔵した。製品の2℃~8℃での安定性を向上させるために、様々な塩、糖添加剤、アミノ酸、および緩衝液を使用して、200種を超える異なる処方を分析した。最良の結果は、硫酸ナトリウム(Na2SO4)で得られた。
pH、緩衝液、および硫酸塩の影響
MilliQグレード水への限外濾過/ダイアフィルトレーションを使用して、MVA-BN-FILO BDSの緩衝液交換を行った。その後、特定の体積の(異なるpHの)濃縮トリス緩衝液および硫酸ナトリウム(Na2SO4)溶液を加えて、最終処方を得た。この処方、すなわち処方Bは、10mMのトリス緩衝液および100mMのNa2SO4からなっており、それぞれpH7.5、8、8.5、および9である処方B1、B2、B3、およびB4に分けた。
異方性測定の結果は、図1に示す。加速ストレスを適用した後、すべての異方性走査値が変化したことが示された。すべての処方について、ストレス後に異方性走査値が低下し、MVAのタンパク質中の励起された基がより柔軟になったことが示唆された。柔軟性のこの増大は、こうしたタンパク質の特定の基のアンフォールディングおよび/またはタンパク質構築物の解離のせいであるとすることができる。しかし、この変化の程度は、溶液中に硫酸塩が存在したとき、それほど顕著でなかった。加えて、硫酸塩含有処方では、異なるpHでも変化が同様であり、硫酸塩の堅固な安定化効果が示唆された。異方性がタンパク質濃度および光散乱と無関係であるという事実を踏まえると、これにより、この方法で得られた結果が非常に信頼できることが示唆される。
実験の設計および方法
MVAウイルスの安定性に対する硫酸塩の効果をさらに調べるために、本発明者らは、異なる塩を含むいくつかの追加の処方を試験した。それらの追加処方の概要を表2に示す。
この実験で得られたすべての結果の概要を、図3に示している。各グラフは、異なるpHの処方ごとに得られた結果を表す。グラフの各縦列は、別個のアッセイで得られた測定値を示す。
実験の設計および方法
本研究では、実施例3と同じ塩の効果を、異なるpHのリン酸緩衝液中で試験した。それらの追加処方の概要を表3に示す。
この実施例で得られたすべての結果の概要を、図4に示している。各グラフは、異なるpHの処方ごとに得られた結果を表す。グラフの各縦列は、別個のアッセイで得られた測定値を示す。
実験の設計および方法
この実験では、これまでの研究からの有望な処方ならびに新規処方および硫酸塩を含有しない対照処方を調製した。それらの処方の概要を表4に示す。
加速ストレス条件の結果としての効力の低下の概要を、表5に、試験した各処方について示す。
実験の設計および方法
この実験では、これまでの研究からの有望な硫酸塩含有処方(実施例5、表5:処方1、3、および7)ならびに硫酸塩なしの対照処方を調製した。限外濾過/ダイアフィルトレーションを使用して、MVA-BN-FILO原体の緩衝液交換を行った。所望の薬物製品濃度前後で、処方物をガラスバイアルに充填し、栓をし、蓋を被せた。処方の概要を表6に示す。各処方を次のストレス条件下に置いた。i)5℃で3カ月、ii)-20℃で3カ月、iii)20回の凍結解凍サイクル、iv)200rpmで1日の撹拌。示したストレス条件の1つの前後に、種々の分光学的方法によってサンプルの測定を行った(たとえば、異方性、自家蛍光、90度光散乱、燐光、動的光散乱、および濁り度)。加えて、サンプルの効力も測定した。
ストレス条件を適用する前後の処方物の効力を、表7および8に示す。図5、6、7、および8には、それぞれ、調製後、5℃で3カ月、-20℃で3カ月、20回の凍結解凍サイクル、および200rpmで1日の撹拌という、受けたストレス条件後のMVA処方物についての異方性、蛍光発光、および90度光散乱スペクトルを収載する。図9には、試験したサンプルの濁り度、Z平均直径、およびPDIを収載する。処方3は、-20℃で3カ月後、20回の凍結解凍サイクル後、および1日の撹拌後に効力低下を示さなかった(表7および8)。測定されたばらつきは、アッセイ変動値の範囲内であった。処方7は、1日の撹拌後には効力低下を示さなかったが、-20℃で3カ月または20回の凍結乾燥サイクル後には顕著な低下があった(表7および8)。
自家蛍光アッセイ
MVAタンパク質は、励起後に光を再び発する芳香族アミノ酸(フルオロフォア)、特に、トリプトファン、ならびに、より程度は低いが、チロシンおよびフェニルアラニンを含んでいる。トリプトファンの発光極大および量子収量は、その環境の極性に大きく左右される。極性の水性環境(たとえば、球状タンパク質の表面、または解離および遊離状態)では、量子収量が比較的低いが、極性環境(たとえば、凝集体の内側、または親油性のウイルスエンベロープ内)では、量子収量が増大する。この特色により、トリプトファン蛍光は、タンパク質立体構造変化、凝集、分解、解離、および分子間相互作用を研究するのに有用なツールとなる。
定常状態蛍光異方性によって、フルオロフォアが偏光を回転させる能力が測定される。励起波長は、フルオロフォアによって発光されると、一方向に偏極され、引き続いて、回転する。フィルターを使用して、励起された波長と同じ平面にある発光波長を測定するが、これは、励起波長と同じ平面にある光だけが検出されることを意味する。偏光が回転するほど、検出器におけるシグナルは弱い。光を回転させる能力は、個々のフルオロフォアの平均柔軟性に応じて決まり、フルオロフォアの柔軟性が高いほど、光は回転し、検出器におけるシグナルは弱くなる。このため、異方性は、タンパク質立体構造変化、凝集、分解、解離、および分子間相互作用を研究するのに有用なツールとなる。
キュベットによる90度光散乱測定を、Fluoromax-4分光蛍光光度計(Jobin Yvon Horiba、英国)で実施した。測定は、サーモスタットで調温されたキュベットホルダーを使用して25℃で行った。Hellma石英キュベットを使用した。最適化されたバンドパスの組合せおよび0.01秒の積分時間を使用して励起波長で発光をモニターすることにより、角度90度での光散乱を、500~750nmの間の波長で測定した。光散乱スペクトルからは、溶液中の粒子の濃度、屈折率、および大きさに関する情報が得られる。この技術は、生物学的製剤の凝集状態の小さな変化に高感度であることが示されている。たとえば、光散乱の減少は、粒子の濃度の低下もしくは同数の粒子の大きさの縮小のいずれかによって、またはこれらを組み合わせて説明をつけることができる。光散乱の増加は、多くの場合、粒子の数の増加もしくは同数の粒子の大きさの増大またはこれらの組合せによる凝集と関連付けられる。
動的光散乱(DLS)は、懸濁液または溶液中の粒子の粒度分布を測定するのに使用されている技術である。偏極レーザー光をサンプルに侵入させ、サンプル中に存在する粒子によって散乱させる。その後、散乱光を光電子増倍管によって収集する。ここで、経時的な強度の変動を分析する。小さい粒子は、大きい粒子より溶液中を速く動き回るため、その高度なブラウン運動により、小さい粒子の方が、強度の変動が大きい。MVAは、粒子であり、偏極レーザー光を散乱させるので、DLSを使用して、MVA粒子の大きさの増大(たとえば、凝集)および/または縮小(たとえば、解離)を見極めることができ、このため、DLSは、凝集または解離を検出するのに有用な技術となる。
濁り測定では、サンプル中の分子が光を吸収しない波長(たとえば、タンパク質が主な発色団であるサンプルでは350nm)で検出される、サンプル中の粒子の散乱による透過光の強度低下(見かけの吸光度)が測定される。分子が凝集し、または超分子複合体を形成するとき、別々の粒子から生じるときはランダムであった光散乱が、このとき干渉性になり、それによって、測定される強度が増大する。このため、光散乱および濁り測定は、凝集および複合体形成または解離を検出するのに有用な技術となる。
蛍光が、励起された一重項状態からの発光を指すのに対し、燐光は、励起された三重項状態からの発光を指す。燐光は、基本的には、蛍光(nsスケール)と同様であるがより長い時間スケール(ms~ms)での放射線の放出であるため、励起が止んだ後も発光が持続する。ヤブロンスキー図によれば、フルオロフォアは、光を吸収し、基底状態から一重項状態へと進んだ後、三重項状態への項間交差へと進む。三重項状態からの発光(燐光)は、蛍光に比べて低エネルギーであり、したがって、長波長である。燐光シグナルは、活性医薬成分(API)の特定の性質である場合があり、減少の場合では、立体構造変化、分解、または解離の徴候となる。燐光発光の増加は、より大きく厳格な構造が形成されるとき、たとえば、タンパク質凝集の場合に起こりうる。
蛍光活性化細胞選別機(FACS)アッセイ
組換えまたは非組換えMVAの力価(InfU/mL)を、蛍光活性化細胞選別機(FACS)アッセイによって求めた。MVAに感染させたベビーハムスター腎臓細胞21(BHK-21)細胞を、ワクシニアウイルス(VACV)に特異的な蛍光色素結合抗体で免疫染色し、引き続いて、定量的な細胞計数のためにBD Flow Sensorを備えたFACSVerse(商標)(BD Bioscience)機器を使用してこれを収集し、定量化した。
TCID50は、国際公開第03/053463号パンフレットの実施例2に記載されているように、96ウェルフォーマットにおいて10倍希釈物を使用して、MVAの感染力を滴定する方法である。MVAの滴定は、96ウェルフォーマットにおいて10倍希釈物を使用する、TCID50に基づくアッセイにおいて実施した。アッセイの終点において、抗ワクシニアウイルス抗体および適切な染色溶液を使用して、感染した細胞を可視化した。2~3日齢の初代CEF(ニワトリ胚線維芽細胞)細胞を7%RPMI中に希釈して1×105細胞/mLとした。1希釈物あたりの反復試験を8回として、10倍希釈を行った。希釈した後、96ウェルプレートのウェルあたり100μLを播いた。次いで、細胞を37℃、5%CO2で終夜インキュベートした。

- 10mM~100mMの間の濃度の塩化ナトリウムを含む、請求項14に記載の組成物。

a)前記ポックスウイルスと、b)前記緩衝液と、c)約5mM~約300mMの濃度の前記硫酸塩とを組成物において合わせることを含む、方法。 - 請求項1~21のいずれか一項に記載の組成物を調製することを含む、ポックスウイルス組成物を安定化させる方法。
