本発明に従うガイドRNA配列は、センスまたはアンチセンス配列であり得る。ガイドRNA配列は、概してプロトスペーサー隣接モチーフ(PAM)を含む。PAMの配列は、使用されるCRISPRエンドヌクレアーゼの特異性要件に応じて変化し得る。S.pyogenes由来のCRISPR-Cas系において、標的DNAは、典型的には5’-NGGプロトスペーサー隣接モチーフ(PAM)の直前にある。したがって、S.pyogenes Cas9の場合、PAM配列は、AGG、TGG、CGG、またはGGGであり得る。ガイドRNAの特異的配列は変化し得るが、その配列に関係なく、有用なガイドRNA配列となるのは、オフターゲット効果を最小化すると同時に、高効率及びウイルス(例えば、インフルエンザウイルス)の完全な除去を達成する配列である。ガイドRNA配列の長さは、約20~約60またはそれ以上のヌクレオチド、例えば、約20、約21、約22、約23、約24、約25、約26、約27、約28、約29、約30、約31、約32、約33、約34、約35、約36、約37、約38、約39、約40、約45、約50、約55、約60またはそれ以上のヌクレオチドに変動し得る。
RNA誘導型エンドヌクレアーゼをコードする本開示の核酸と、ウイルスにおける標的配列に相補的なガイドRNAとを含む、医薬組成物が提供される。本開示の組成物を含む医薬組成物は、非経口(筋肉内、腹腔内、静脈内(IV)、または皮下注射)、経皮(受動的に、またはイオン導入もしくはエレクトロポレーションを用いて)、あるいは経粘膜(経鼻、経膣、経直腸、または舌下)の投与経路または生分解性挿入物の使用による投与向けとすることができ、また、各投与経路に適した剤形で製剤化することができる。1つの実施形態において、RNA誘導型エンドヌクレアーゼをコードする核酸及びガイドRNAは、同じ医薬組成物に含まれる。別の実施形態において、RNA誘導型エンドヌクレアーゼをコードする核酸及びガイドRNAは、別々の医薬組成物で製剤化される。

いくつかの実施形態において、組成物は経口送達用に製剤化される。経口用固体剤形は、Remington:The Science and Practice of Pharmacy,21st Ed.2006(Lippincott Williams and Wilkins,Baltimore,MD)のChapter 45で概説されている。固体剤形としては、錠剤、カプセル、丸薬、トローチもしくはロゼンジ、カシェ剤、ペレット、粉末、または顆粒、あるいはポリマー化合物(例えば、ポリ乳酸、ポリグリコール酸など)の粒子調製物への材料の組込み、またはリポソームへの材料の組込みが挙げられる。このような組成物は、本開示物の物理的状態、安定性、in vivo放出速度、in vivoクリアランス速度に影響を及ぼし得る。組成物は、液体形態で調製されてもよく、乾燥粉末(例えば、凍結乾燥)形態をとってもよい。リポソームまたはプロテイノイドカプセル化を使用して組成物を製剤化してもよい。リポソームカプセル化を使用することができ、リポソームを様々なポリマーで誘導体化することができる(例えば、米国特許第5,013,556号)。概して、製剤は、ペプチド(またはその化学修飾形態)と、胃環境でペプチドを保護し腸内で生物学的活性物質を放出する不活性成分とを含む。
本開示の組成物は、局所適用することができる。局所投与用の製剤としては、限定されるものではないが、エアロゾル送達、皮膚パッチ、局所用ゲルまたはローションが挙げられる。
本明細書で開示される組成物は、制御放出製剤で投与することもできる。制御放出ポリマーデバイスは、ポリマーデバイスの留置(ロッド、シリンダー、フィルム、ディスク)または注射(マイクロ粒子)の後、全身に長期間放出するように作製することができる。マトリックスは、ミクロスフェアなどの微粒子の形態をとることができ、この場合、薬剤は固体ポリマーマトリックスまたはマイクロカプセル内で分散し、コアはポリマーシェルとは異なる材料であり、ペプチドは、事実上液体でも固体でもよいコア内で分散または懸濁している。本明細書で特に定義されない限り、マイクロ粒子、ミクロスフェア、及びマイクロカプセルは互換的に使用される。代替的に、ポリマーは、ナノメートルから4センチメートルの範囲の薄いスラブもしくはフィルムとして、粉砕もしくは他の標準的技術によって産生した粉末として、またはヒドロゲルなどのゲルとしてであっても、流延することができる。
本開示の組成物及び方法は、宿主細胞または対象からのウイルスの治療及び不活性化に使用することができる。本発明の方法は、宿主生物から、宿主の遺伝物質の完全性を妨害することなく、ウイルスまたはその他の外来遺伝物質を除去するのに使用することができる。ヌクレアーゼを使用してウイルス核酸をターゲティングし、それによってウイルス複製もしくは転写を妨害することができ、またはさらに宿主ゲノムからウイルス遺伝物質を切り取ることもできる。ヌクレアーゼは、ウイルス核酸が細胞内に粒子として存在するとき、またはウイルス核酸が宿主ゲノム内に組み込まれているときに、宿主物質に作用することなくウイルス核酸のみを除去するように特異的に標的とすることができる。ウイルス核酸のターゲティングは、ヌクレアーゼが破壊するウイルスゲノム物質を標的とし、宿主細胞ゲノムを標的としない配列特異的部分(例えば、ガイドRNA)を用いて行われ得る。いくつかの実施形態において、共にウイルスゲノム物質を標的とし、ウイルスゲノム物質を選択的に編集または破壊する、CRISPR/Casヌクレアーゼ及びガイドRNA(gRNA)が使用される。CRISPRは、ファージ感染から細菌を保護する細菌免疫系の天然の要素である。ガイドRNAは、CRISPR/Cas複合体をウイルス標的配列に局在化させる。複合体の結合によって、Casエンドヌクレアーゼがウイルスゲノムの標的配列に局在化され、ウイルスゲノムを切断する。その他のヌクレアーゼ系も使用することができ、例えば、ジンクフィンガーヌクレアーゼ、転写活性化因子様エフェクターヌクレアーゼ(TALEN)、メガヌクレアーゼ、または宿主の遺伝物質の通常機能を妨害することなくウイルス核酸を分解もしくは妨害するのに使用され得るその他の任意のヌクレアーゼ系がこれに含まれる。
1.ウイルス感染
本開示の組成物は、ウイルスに感染した対象におけるウイルス負荷を減少させるのに使用することができる。1つの実施形態において、本開示の組成物は、ウイルス負荷の減少に有効な量で、ウイルス感染を有する対象に投与される。RNA誘導型エンドヌクレアーゼをコードする核酸コンストラクト及びガイドRNAコンストラクトは、同じ医薬組成物で、または2つの別々の組成物で、同時に投与することができる。別の実施形態において、RNA誘導型エンドヌクレアーゼをコードする核酸コンストラクト及びガイドRNAコンストラクトは、2つの別々の組成物で順次投与される。1つの実施形態において、対象は、活性化可能なヌクレアーゼ組成物を1、2、3、4、5、6、7、8週間、または8週間超投与される。対象のウイルス負荷が、もはや対象が感染していないことを示す範囲内であることをウイルス力価が示すまで、対象に活性化可能なヌクレアーゼ組成物を投与することができる。
1つの実施形態は、その必要のある対象におけるウイルス感染を治療するまたは減少させる方法を提供する。1つの実施形態において、本開示の組成物は、RNAウイルスを治療するのに使用することができる。例示的な方法は、RNA誘導型エンドヌクレアーゼをコードする核酸コンストラクトと、RNAウイルス遺伝子を標的とする配列を有するガイドRNAとを含む組成物を、その必要のある対象に投与することを含む。ヒトにおける疾患を引き起こす例示的なRNAウイルスとしては、限定されるものではないが、西ナイルウイルス、エボラウイルス、ヒト免疫不全ウイルス(HIV)、デングウイルス、黄熱病ウイルス、インフルエンザウイルス、ラッサウイルス、ハンタウイルス、マールブルグウイルス、ヘンドラウイルス、ニパ(Nipha)ウイルス、チクングニアウイルス、アデノウイルス、ヒトサル痘ウイルス、C型肝炎ウイルス、リフトバレーウイルス、呼吸器合胞体ウイルス、及びエンテロウイルスが挙げられる。
別の実施形態において、本開示の組成物は、DNAウイルスを治療または防止するのに使用することができる。例示的なDNAウイルスとしては、限定されるものではないが、ヒトパピローマウイルス(HPV)、B型肝炎、アデノウイルス、ヘルペスウイルス、ポックスウイルス、水痘帯状疱疹ウイルス、及びパルボウイルスが挙げられる。DNAウイルスは、カプシドと呼ばれるタンパク質を含む外殻によって保護された一連のDNA遺伝子から構成される。カプシドは、宿主内に侵入するとDNA遺伝子を保護する。感受性細胞に侵入すると、ビリオンは分解されてウイルスゲノムを細胞内に放出し、その時点でウイルスDNA内の遺伝子が転写され、ウイルスのメッセンジャーリボ核酸(mRNA)が産生される。mRNAは、宿主の正常な細胞機能の改変を担うタンパク質に翻訳される。1つの実施形態において、RNA誘導型エンドヌクレアーゼをコードする本開示の核酸コンストラクトは、タンパク質に翻訳される前のDNAウイルスのmRNAを標的とする。
材料及び方法:
Cas13aコンストラクトの設計及び合成:レプトトリキア・ブッカリス(Leptotrichia buccalis)配列由来のCas13aを、Addgene p2CT-His-MBP-Lbu_C2C2_WT(プラスミド番号83482)から得た。pMA7ベクター(Thermo Scientific,USA)において、野生型Cas13aをV5タグと共にクローニングし、マウスアルファグロビン(Genbankアクセッション番号NM_001083955)由来の3’UTRを付加した。また、V5タグをコードするgBlocks(Integrated DNA technologies)を用いて、Cas13aコンストラクトに核局在化配列(3’及び5’)ならびにV5タグを付加して、Cas13a-V5-NLSバージョンを作成した(図1Aの概略図)。
様々なcrRNA及びtrRNAに対するCas13a活性を評価するため、ウサギ網状赤血球ライセート系を製造業者(Promega,WI,USA)の指示に従って用いて、Cas13a mRNAをin vitro翻訳した。RNaseAlert(登録商標)-1基質を、A549細胞RNA(100ng)、20mM HEPES(pH6.8)、50mM KCl、5mM MgCl2、BSA(10μg/ml)、酵母tRNA(10μg/ml)、0.01% Igepal CA-630、及び5%グリセロール19からなるRNA処理緩衝液中のcrRNA(500nM)、trRNA(500nM)と混合した。この混合物を、96ウェルプレートウェル中の翻訳されたCas13aライセート(5μl)に低温で添加し、ピペットを用いてウェルを混合した。全ての試薬調製及び添加は氷上で行った。蛍光測定値(励起485±20nm/発光528±20nm)を室温で10分間隔で90分間記録した。次いで、RNA切断産物(最終産物)を15%ウレアTBEゲルで泳動した。ゲルをSYBRgoldで30分間染色し、UV照射下で可視化した。
Neon(登録商標)Transfection System(Invitrogen,Fisher Scientific)、Lipofectamine 3000(Thermos Fisher Scientific)、及びViromer Red(Lipocalyx GmbH)の形質移入法に対し、MDCK及びA549細胞株におけるCas13a mRNAの形質移入をアッセイした。Lipofectamine 3000及びViromer Red形質移入実験については、細胞をカバーガラス(12mm)上に一晩播種し(125,000細胞/ウェル)、翌日、各製造業者のプロトコルに従って1μgのCas13a mRNAを形質移入した。Neon(登録商標)transfection systemについては、Neon(登録商標)Transfection SystemのA549及びMDCK用の組込みプログラムを用いて細胞に1μgのCas13a mRNAをエレクトロポレーションし、培地を含むカバーガラス上に播種した。形質移入細胞を、形質移入から2、4、6、16、及び24時間後に4%パラホルムアルデヒドで固定した(10分)。細胞をトリトンX-100(0.1%)で5分間透過処理した後、5%のBSAで1時間ブロックした。ウサギ抗V5(Abcam ab9116)一次抗体及びロバ抗ウサギA488(A21206 Invitrogen)二次抗体を、それぞれ1時間インキュベートした。PerkinElmer Ultraview ERS Spinning Disk顕微鏡で細胞を可視化した。
Cas13a mRNA及びcrRNA(RSV_1.1及びRSV_m1.1)ならびにRSVで処置した細胞からの上清を14000xgで5分間遠心分離した。上清を収集し、24ウェルプレート内のVero細胞単層上に50μl/ウェルを1時間(15分毎に断続的に揺動)置いた。単層に、2% FBSならびに100U mL-1ペニシリン及び100mg mL-1ストレプトマイシン(Life Technologies)を伴った2×DMEM中2.4% Avicel(FMC Biopolymer RC-481)を1:1の比で含むAvicel-DMEMオーバーレイ培地を添加し、6日間インキュベートした。培地を除去し、1×PBSで洗浄した後、細胞を4%パラホルムアルデヒドで10分間固定した。細胞を37℃で5% BSAで30分間ブロックし、続いて37℃で抗RSV(ヤギポリクローナル)一次抗体で染色した。30分後、細胞を洗浄し、HRP結合体化二次抗体(ロバ抗ヤギ)を37℃で30分間インキュベートした。TrueBlue(商標)ペルオキシダーゼ基質を用いて室温で10分間、プラークを展開した。TrueBlueを除去し、細胞を水ですすぎ、プラークを風乾し、カウントした。
ウサギ網状赤血球ライセートを用いて、Cas13a及びCas13a-NLS(核局在配列を有するバージョン)mRNAをin vitroで翻訳し、これを使用して、IVA crRNA及びtrRNAと連携した発現Cas13aタンパク質のRNA切断活性を評価した(図1A)。crRNA及びtrRNAは、IVAのゲノム分節に由来した(表1)。RNaseAlert(商標)基質蛍光をRNA切断の出力とした。Cas13a及びCas13a-NLSによるRNA切断によって生じた蛍光は、それぞれ最初の10分間及び20分間に最大まで増加し、次いで、標的RNAが枯渇したために経時的に徐々に減少した。RNA切断の全体的な傾向は、Cas13a(図1B)及びCas13a-NLS(図1C)で共に類似した。RNA切断は、15% TBE-ウレアゲルを用いてゲル電気泳動によってライセートを調べたときにも観察された(図1D及び1E)。これらの結果は、in vitro翻訳Cas13a媒介性のRNA切断が特異的であり、crRNA及びtrRNA両方が存在する場合にのみ生じたことを示している。crRNA、NT-CR、またはCas13a mRNAは、単独では標的RNAを切断しない。これらの結果は、精製したCas13aタンパク質を使用したときの特異的活性化という過去の知見(East-Seletsky,A.,et al.,Nature,538:270-273(2016))を確証するものである。Cas13a活性をアッセイするための切断緩衝液中に酵母tRNAまたは細胞RNAが存在した場合、蛍光または切断されたRNA産物が生じず、これにより、標的RNAに対するCas13a:crRNAの特異性が示された。これらの設計されたcrRNAは、対応するtrRNAが存在する場合にのみ、in vitro翻訳されたCas13a及びCas13a-NLSによるRNA切断を示した(図2A~2K及び3A~3H)。
結果:
次いで、インフルエンザ感染の細胞モデルでCas13a:crRNA系を評価した(図4A)。MDCK及びA549細胞株において、形質移入剤、Viromer(登録商標)Red、及びLipofectamine 3000を用いたmRNA送達、ならびにNeon(登録商標)transfection systemを用いたエレクトロポレーションによるmRNA送達を評価した。Cas13a発現を2、4、6、16、及び24時間時点で評価した(図5A~5II)。Cas13a発現は、全ての形質移入剤及び細胞タイプにおいて、形質移入から2時間後という早期に観察された。しかし、形質移入から16時間後、Cas13a発現は、Neon及びLipofectamine 3000で形質移入した全ての細胞株で減少した。Viromer(登録商標)Red形質移入細胞は、24時間時にも発現を示した。この結果に基づき、以後の実験にはViromer(登録商標)Red及びA549細胞株を選択した。免疫蛍光法を用いると、Cas13aが主に細胞質内に局在していたのに対し、Cas13a-NLSは細胞質(Cas13aがまだ合成中であるため)と、48時間時には核内と、両方に存在することが観察された(図4B~4K)。
結果:
IVAのポリメラーゼ(PB1)、ヘマグルチニン(HA)、ノイラミニダーゼ(NA)、核タンパク質(NP)、ならびに核外輸送タンパク質(NS1及びNS2)、ならびにM2タンパク質を含めたIVAの様々なゲノム分節を標的とするcrRNAを設計した(図6A~6C)。Cas13aまたはCas13a-NLSのmRNA及び各crRNAをA549細胞に同時形質移入し、次に当該細胞をインフルエンザウイルスに感染させた(感染多重度MOI 0.01)。ポリメラーゼがIVA複製に不可欠なタンパク質であることを考慮し、ポリメラーゼ構成成分PB1を標的とするいくつかのcrRNAを設計した。最初に、これらのcrRNAをCas13a及びCas13a-NLSの両方で試験した。試験したcrRNAのうち、gPB1が、PB1遺伝子コピー減少に最も効果的であった。他のIVA遺伝子を標的とするcrRNAを試験したところ、PB1遺伝子コピーの有意な減少が観察され、このことからウイルス感染性に対する全体的な効果が示唆された。IVAは核内に局在するため、Cas13aを核局在化シグナル(NLS)ありまたはなしで試験した。
結果:
スクリーニングした全てのcrRNAの中で、PB1ゲノム分節を標的とするcrRNA(gPB1)は、ウイルスRNAコピー数を最も効率的に減少させ(図6B及び6E)、他のウイルスタンパク質に影響を及ぼすことが分かった。このことをin situで実証するため、Cas13a、IVA M2タンパク質(ウイルス集合または分解部位24を示す)に対する免疫蛍光法を実施し、次にIVAゲノム可視化のために蛍光in situハイブリダイゼーション(FISH)を実施した。Cas13aまたはCas13a-NLS mRNAのいずれかと、PB1部位を標的とするcrRNA(gPB1、及びPB1 mRNAを標的とするmPB1)とを形質移入した細胞は、IVA感染の減少を示すM2タンパク質の減少(図7A~7H)と、サイトゾル内のウイルス感染部位へのCas13aの局在化とを示した。このデータから、ポリメラーゼ遺伝子を標的とすることにより、他のウイルスタンパク質を阻害し、そのためより高い阻害効果を達成できたことが明らかである。
結果:
Cas13a:crRNAの用量応答アッセイ
細胞におけるIVA阻害に要するRNA用量を決定するため、Cas13a mRNA及びgPB1(crRNA)の濃度を試験した。Cas13a mRNA及びgPB1の濃度を増加して形質移入した細胞は、IVA感染の逆比例を示した。用量依存的IVA阻害は200nMのgPB1まで観察され、crRNAをさらに増加してもIVA感染は減少しなかった。最も低い組合せ(Cas13a mRNA(0.5μg)及びgPB1(25nM))ならびに最も高い組合せ(Cas13a mRNA(3μg)及びgPB1(300nM))において、IVA感染は、それぞれ0.35log(55%)及び0.48log(66%)PB1コピー減少した(図8A~8B)。最も大きな阻害は、2μgのCas13a及び100nMのgPB1で見られた(76.78%)。しかし、適度でありその上効率的なCas13a mRNA(1μg)及びgPB1(100nM)の用量を、以後の全ての細胞におけるIVA実験に使用した。
次に、Cas13a:crRNA系の性能をキャラクタリゼーションするため、多くの実験条件を調べた。MOIの効果を評価し、PB1を標的とするCas13a:crRNAは、形質移入から24時間後にインフルエンザウイルス遺伝子コピーを0.77~1.26log(21%~72.4%)減少できることが示された(図8B)。MOI 0.01(図8C)及び0.1(図8D)(いずれも生理的に適切なMOIである)でgPB1を試験することにより、最適なウイルス力価を評価した。MOI 0.01における阻害は、gPB1、mPB1、及びgPB1+mPB1についてそれぞれ85.8%、86.6%、及び92.5%であった。同様に、MOI 0.1におけるPB1コピー数の減少は、gPB1、mPB1、及びgPB1+mPB1についてそれぞれ86.7%、83.1%、及び94.5%であった。
結果:
感染中におけるウイルスRNAの時間依存的分解を調べた(図10A)。gPB1またはmPB1のいずれかを標的とすることにより、Cas13a(図10B)及びCas13a-NLS(図10C)の両方において、ウイルスコピーは24時間時に0.96~1.21log(89.2%~93.9%)、48時間時に1.88~2.6log(98.6%~99%)減少したが、さらに72時間時にウイルスRNAコピーは3.53~3.8log(99.97%~99.98%)減少した。Cas13aの持続的効果はIVA複製を妨げ、ウイルスRNAコピーを最大72時間減少させるため、このデータは注目に値する。
結果:
Cas13a:crRNA系が、シミュレートされた「予防的方式」でIVA感染を軽減することから、既にIVAに感染した細胞の感染を減少させるCas13a:crRNA系の能力を評価した。IVA(MOI 0.01)に4時間感染させた細胞に、その後、Cas13a/Cas13a-NLS mRNA及びcrRNA(gPB1及びmPB1)を形質移入し(図10D)、IVAに対する効果を72時間モニターした。形質移入から48時間後のCas13a(図10E)またはCas13a-NLS(図10F)でIVAコピー数の最大0.74log(83.3%)の減少が観察され、72時間後には1.47log(93.9%)の減少が観察された。IVA感染に対するCas13a-crRNAの効率は、細胞にCas13a mRNA及びcrRNAを24時間形質移入し(Cas13aの発現減少が予測される)(図10Gに基づく)、次いでインフルエンザウイルスに8時間感染させることによって評価した(図10H)。この条件下では、依然として最大0.3log(63%)のIVAの減少が観察され、したがって、Cas13a媒介性IVA標的指向性のロバストネスを強調するものであった。同様に、持続的感染がCas13a媒介性RNA標的指向性に及ぼす効果を評価した。ここでは、細胞を24時間感染させ、次に形質移入し、形質移入から8時間後のウイルスコピー数を評価した(図10I)。この場合も、ウイルス力価における最大49%の有意な減少が観察された(図10J)。したがって、これらの実験から、Cas13aが予防的治療及び感染後治療の両方においてIVA阻害に有効であることが示された。また、正常なヒト初代気管支/気管上皮細胞モデルにおいて、Cas13a:crRNA系がIVAを最大62%有効に阻害することも観察された(図9)。形質移入の効率が細胞タイプによって異なることから、Cas13a:crRNAによるIVAの阻害も細胞タイプによって異なる。一次細胞は通常は形質移入が困難であるが、ここでは、このアプローチがヒト一次細胞に適用できるという概念実証が示される。
結果:
Cas13a-crRNA系は、特異的crRNAの設計を通じて様々なウイルス性病原体を標的とする可能性をもたらす。これを実証するため、ヒト呼吸器合胞体ウイルス(RSV)のゲノム(gRSV)及びM2 mRNA(RSV-M2)の両方に対してcrRNAを設計した。Cas13a mRNA及び両方のcrRNAを形質移入した細胞は、予防(図11A~11B)及び感染後(図11C~11D)の両方でRSV力価の減少を示した。Cas13a:crRNAを感染前に投与したときには、最大0.55log(71%)の減少が認められた(図10B)。しかし、形質移入前にRSVに感染させた場合、Cas13a:crRNAは、一緒にもたらされたgRSV及びm1.1におけるRSV力価を0.3~0.44log(63.5%)減少させ、一方、個別にもたらされたゲノム(gRSV)及びRSV mRNA(RSV-M2)を標的とするcrRNAの場合、ウイルス力価は、それぞれ51.3%及び49.7%であった(図10D)。hRSVにおいて予防による約71%のノックダウンが観察されたが、これらのデータはM2/L遺伝子末端配列のみに基づく第一世代ガイドで得られたことに注意すべきである。これらの結果は、天然の感染よりも著しく高いMOI 1において阻害が見られるため、有望である。加えて、Cas13a:crRNAを投与した細胞では、プラークの数が減少しただけでなく、プラークのサイズも減少した。このことは、細胞間の拡散が少ないことと、より多くの干渉性欠陥(DI)粒子が生成される可能性とを示すものである。全体としては、hRSVゲノム及びmRNAに対する将来的なバイアスなしのガイドスクリーニングはノックダウンの改善をもたらす可能性があるため、この結果単独でも極めて有望である。これらの知見は、いくつかのウイルス性病原体に適用可能な見込みのある、抗ウイルスアプローチとしてのCas13a:crRNAシステムの可能性を示すものである。
結果:
Cas13a活性は、IVA RNAの切断において特異的であり、いかなる検出可能なオフターゲットRNA切断ももたらさなかった。送達から8及び24時間にCas13a mRNA及びcrRNA(PB1)を形質移入した細胞のトランスクリプトームプロファイルは、IVA感染の有無にかかわらず、mRNAレベルにおける内在性遺伝子発現に有意な変化を示さなかった(図12A~12L)。同様に、オフターゲット活性なしの特異的な内在性mRNAノックダウンは、過去にLeptotrichia wadei Cas13a(Abudayyeh,O.O.et al.,Nature,550:280-284(2107))で報告されている。進行中の転写及びCas13a媒介性分解は、遺伝子発現の有意な変化を推測するのに互いを打ち消す可能性があるため、Cas13aにおける任意の考えられるオフターゲット活性をRNA-Seqによって決定するのは困難であり得るという議論も考えられる。しかし、Lbu Cas13aは、RNA結合(crRNA及び標的RNA)ならびにRNase活性という別個の活性を有する。Cas13aはミスマッチのcrRNA-標的に結合することができるが、RNA切断はもたらさない(Tambe,A.,et al.,Cell Rep,24:1025-1036(2018))。RNase活性は、crRNA及び標的RNAが相補的であるときにのみ活性化されるため、Cas13aのオフターゲット活性は除外され得る。

- RNAウイルスをin vitroまたはin vivoで不活性化するための組成物であって、
RNA誘導型エンドヌクレアーゼをコードする単離核酸コンストラクトと、
RNAウイルスゲノム内の標的核酸配列に相補的な少なくとも1つのガイドRNA(gRNA)コンストラクトと
を含み、
該単離核酸コンストラクトがSEQ ID NO:1に記載の配列を有する、組成物。 - RNAウイルスをin vitroまたはin vivoで不活性化するための組成物であって、
RNA誘導型エンドヌクレアーゼをコードする単離核酸コンストラクトと、
RNAウイルスゲノム内の標的核酸配列に相補的な少なくとも1つのガイドRNA(gRNA)コンストラクトと
を含み、
該単離核酸コンストラクトがSEQ ID NO:2に記載の配列を有する、組成物。 - 前記単離核酸コンストラクトが核局在化配列をさらに含む、請求項1または請求項2に記載の組成物。
- 前記RNAウイルスがインフルエンザウイルスまたは呼吸器合胞体ウイルスである、請求項1~3のいずれか1項に記載の組成物。
- 前記RNAウイルスがマイナス鎖RNAウイルスである、請求項1または2に記載の組成物。
- 前記RNAウイルスがプラス鎖RNAウイルスである、請求項1または2に記載の組成物。
- 請求項1~6のいずれか一項に記載の組成物と、医薬的に許容される担体とを含む、医薬組成物。
- 少なくとも、RNA誘導型エンドヌクレアーゼをコードする単離核酸コンストラクトと、RNAウイルスゲノム内の標的核酸配列に相補的な少なくとも1つのガイドRNA(gRNA)コンストラクトとを含む、その必要のある対象におけるウイルス感染症を治療するための医薬組成物であって、
該単離核酸コンストラクトがSEQ ID NO:1に記載の配列を有する、医薬組成物。 - 少なくとも、RNA誘導型エンドヌクレアーゼをコードする単離核酸コンストラクトと、RNAウイルスゲノム内の標的核酸配列に相補的な少なくとも1つのガイドRNA(gRNA)コンストラクトとを含む、その必要のある対象におけるウイルス感染症を治療するための医薬組成物であって、
