Abstract
Description
また、本発明は、AMPK活性化作用、運動機能向上作用、筋持久力向上作用および筋萎縮抑制作用において優れた作用を有する天然物由来化合物を配合した、AMPK活性化用途、運動機能向上用途、筋持久力向上用途または筋萎縮抑制用途に用いられる経口組成物を提供することを目的とする。
本実施形態に係るAMPK活性化剤、運動機能向上剤、筋持久力向上剤および筋萎縮抑制剤は、下記式(I)で表される化合物を有効成分とするものである。また、本実施形態に係るAMPK活性化用経口組成物、運動機能向上用経口組成物、筋持久力向上用経口組成物および筋萎縮抑制用経口組成物は、下記式(I)で表される化合物が配合されるものである。
例えば、水と低級脂肪族アルコールとの混合液を抽出溶媒として使用する場合には、水と低級脂肪族アルコールとの混合比(容量比)を9:1以上とすることができ、さらには7:3以上とすることができ、あるいは水と低級脂肪族アルコールとの混合比を1:9以下、さらには2:8以下とすることができる。また、水と多価アルコールとの混合液を使用する場合には、水と多価アルコールとの混合比を8:2以上、あるいは1:9以下とすることができ、水と低級脂肪族ケトンとの混合液を使用する場合には、水と低級脂肪族ケトンとの混合比を9:1以上、あるいは2:8以下とすることができる。
以上のようにして得られる化合物(I)は、優れたAMPK活性化作用、運動機能向上作用、筋持久力向上作用および筋萎縮抑制作用を有しているため、AMPK活性化剤、運動機能向上剤、筋持久力向上剤および筋萎縮抑制剤の有効成分として用いることができる。
言い換えると、AMPK活性化剤、運動機能向上剤、筋持久力向上剤または筋萎縮抑制剤を製造するために、化合物(I)を使用することができる。
本実施形態のAMPK活性化剤、運動機能向上剤、筋持久力向上剤および筋萎縮抑制剤は、医薬品、医薬部外品、化粧品等の幅広い用途に使用することができる。
言い換えると、化合物(I)は、PGC-1α mRNA発現促進剤、GLUT4 mRNA発現促進剤、またはIL-6 mRNA発現促進剤の有効成分として用いることもできる。
言い換えると、化合物(I)は、筋肉量増加剤、MuRF-1 mRNA発現抑制剤、Atrogin-1 mRNA発現抑制剤、またはAkt活性化剤の有効成分として用いることもできる。
例えば、本実施形態のAMPK活性化剤は、そのAMPK活性化作用により、糖代謝や脂質代謝を活性化することができるため、糖代謝異常に起因する疾患、例えば、糖尿病、糖尿病関連疾患、糖尿病合併症など;脂質代謝異常に起因する疾患、例えば、高脂血症、高コレステロール血症、肥満、脂肪肝、動脈硬化症など;の予防、治療または改善用途に用いることができる。
化合物(I)は、AMPK活性化作用、運動機能向上作用、筋持久力向上作用および筋萎縮抑制作用において優れた作用を有しているため、経口組成物に配合するのに好適である。この場合、化合物(I)または化合物(I)を含有する組成物をそのまま配合してもよいし、化合物(I)から製剤化した、AMPK活性化剤、運動機能向上剤、筋持久力向上剤または筋萎縮抑制剤を配合してもよい。
化合物(I)(試料1)を用い、以下のようにして、ラット筋萎縮モデルに対する筋萎縮抑制作用を試験した。
また、第2群には、表1に示す用量の化合物(I)を投与した。化合物(I)(試料1)を注射用水に溶解し、経口ゾンデを取り付けた注射筒を用い、強制経口投与した。群分け日を初日(投与1日)として、1日1回、14日間投与し、投与14日の翌日(投与15日)を飼育期間最終日とした。
結果を表2に示す。
以上より、化合物(I)の投与により筋肉量が増加したものと認められた。
上記試験例1で摘出した腓腹筋について、リアルタイムPCRによる遺伝子発現解析試験を実施した。
結果を表3に示す。
一方、筋肉増加に寄与する遺伝子であるPGC-1α(同Ppargc1a)、GLUT4(同Slc2a4)、IL-6(同Il6)は、第2群(投与群)で第1群(対照群)と比較して遺伝子発現が増加傾向にあると認められた。
そのため、化合物(I)の投与によるこれらの遺伝子の発現減少または増加は、筋肉量増加に寄与したものと考えられる。
上記試験例1で摘出した腓腹筋について、筋肉量増加に寄与するキナーゼであるAMPKおよびAktのリン酸化状態を、Luminex測定およびELISA測定により評価した。
Aktは、MILLIPLEX(R) MAP Phospho/Total Akt 2-Plex Magnetic Bead Kit 96-well Plate Assay(EMD Millipore社製,カタログ番号:48-618MAG)を用い、総Akt量(t-Akt)とリン酸化Akt(p-Akt,Ser473)とを1反応系にて一括して測定した。
なお、内部標準として、GAPDHを、Total GAPDH Magnetic Bead MAPmateTM(EMD Millipore社製,カタログ番号:46-667MAG)を用いて測定し、上記AMPKおよびAktの測定値を標準化した。
結果を表4に示す。
そのため、化合物(I)の投与によるこれらのキナーゼの活性化は、筋肉量増加に寄与したものと考えられる。
常法により、以下の組成を有するカプセル剤を製造した。なお、カプセルとしては、2号ハードゼラチンカプセルを使用した。
<1カプセル(1錠200mg)中の組成>
化合物(I) 10.0mg
コーンスターチ 70.0mg
乳糖 100.0mg
乳酸カルシウム 10.0mg
ヒドロキシプロピルセルロース(HPC-L) 10.0mg
常法により、以下の組成を有する経口液状製剤を製造した。
<1アンプル(1本100mL)中の組成>
化合物(I) 0.3質量%
ソルビット 12.0質量%
安息香酸ナトリウム 0.1質量%
香料 1.0質量%
硫酸カルシウム 0.5質量%
精製水 残部(100質量%)
