Description
この実験では、哺乳動物細胞培養ブロスを凝集化させるための、ジアリルジメチルアンモニウムクロリド(PDADMAC)の様々な分子量の製剤を比較し、それらの沈降時間を比較する。
この実験では、二倍体細胞のような小さな細胞株および四倍体細胞株のような大きな細胞株から、組み換え抗体を発現する哺乳動物細胞培養ブロスを凝集化するために必要なPDADMACの量を比較する。
この実験では、高いラクテート濃度を生じる、かつ/または、高い細胞密度を有する細胞培養のためのPDADMAC凝集化の影響を見る。
この実験では、PDADMACと組み合わせて使用された様々な希釈剤の沈降時間を比較する。
この実験では、高い細胞密度を有する細胞培養のための沈降速度に対する、PDADMACおよびPEG3,000の組み合わせの影響を見る。
この実験では、沈降速度に対する、PDADMACとのPEG添加のタイミングの影響を見る。
この実験では、PDADMACおよびPEGと一緒に界面活性剤を加えることによる、沈降時間への影響を見る。
Claims (20)
- 哺乳動物細胞培養回収方法であって、
組み換えタンパク質を発現する哺乳動物細胞を、所定の時間、又は、所望する圧縮細胞体積が達成されるまで、細胞培養用培地で培養すること、
ジアリルジメチルアンモニウムクロリドのポリマーであるカチオン性ポリマー、並びにポリエチレングリコール、ベタイン及びデキストラン/ベタインからなる群から選択される非イオン性ポリマーを前記細胞培養用培地に加えて凝集化を開始させること、
前記細胞培養用培地を凝集化の間混ぜること、
凝集物を第1沈降のために沈降させること、及び
第1の清澄化された上澄みを回収すること
を含む、方法。 - カチオン性ポリマーが、ポリジアリルジメチルアンモニウムクロリドである、請求項1に記載の哺乳動物細胞培養回収方法。
- 非イオン性ポリマーが、PEG3,000及びPEG6,000から選択される、請求項1に記載の哺乳動物細胞培養回収方法。
- PEG3,000の濃度が、3%から4.5%である、請求項3に記載の方法。
- PEG6,000の濃度が、2.5%から3.5%である、請求項3に記載の方法。
- 非イオン性界面活性剤を細胞培養用培地に加えることをさらに含む、請求項1に記載の哺乳動物細胞培養回収方法。
- 非イオン性界面活性剤が、サポニン又はトリトン(登録商標)X-100である、請求項6に記載の方法。
- 非イオン性界面活性剤が、トリトン(登録商標)X-100である、請求項6に記載の哺乳動物細胞培養回収方法。
- トリトン(登録商標)X100の濃度が、0.05%(w/v)である、請求項8に記載の方法。
- 第1の沈降凝集物を洗浄すること、
洗浄した凝集物を第2沈降のために沈降させること、及び
第2の清澄化された上澄みを回収すること
をさらに含む、請求項1に記載の哺乳動物細胞培養回収方法。 - 第1沈降からの凝集物を9%スクロース溶液中で洗浄する、請求項10に記載の方法。
- カチオン性ポリマー及び非イオン性ポリマーを同時に加える、請求項1に記載の方法。
- カチオン性ポリマーを先ず加え、少なくとも30秒間混ぜた後に、非イオン性ポリマーを加える、請求項1に記載の方法。
- カチオン性ポリマー、非イオン性ポリマー及び非イオン性界面活性剤を同時に加える、請求項1に記載の方法。
- カチオン性ポリマーを先ず加え、少なくとも30秒間混ぜた後に、非イオン性ポリマー及び非イオン性界面活性剤を加える、請求項1に記載の方法。
- ポリジアリルジメチルアンモニウムクロリドを、全細胞密度当り20~90pgの濃度で加える、請求項1に記載の方法。
- ポリジアリルジメチルアンモニウムクロリドを、全細胞密度当り25pgの濃度で加え、哺乳動物細胞は二倍体細胞株に由来する、請求項1に記載の方法。
- ポリジアリルジメチルアンモニウムクロリドを、全細胞密度当り43pgから全細胞密度当り57pgの間で加え、哺乳動物細胞は四倍体細胞株に由来する、請求項1に記載の方法。
- 哺乳動物細胞培養用培地が、36℃から20℃の間である、請求項1に記載の方法。
- 哺乳動物細胞培養用培地が、20℃であるか20℃を超える、請求項1に記載の方法。
